RESUMO

Mice are the most commonly used model animals for itch research and for development of anti-itch drugs. Most laboratories manually quantify mouse scratching behavior to assess itch intensity. This process is labor-intensive and limits large-scale genetic or drug screenings. In this study, we developed a new system, Scratch-AID (Automatic Itch Detection), which could automatically identify and quantify mouse scratching behavior with high accuracy. Our system included a custom-designed videotaping box to ensure high-quality and replicable mouse behavior recording and a convolutional recurrent neural network trained with frame-labeled mouse scratching behavior videos, induced by nape injection of chloroquine. The best trained network achieved 97.6% recall and 96.9% precision on previously unseen test videos. Remarkably, Scratch-AID could reliably identify scratching behavior in other major mouse itch models, including the acute cheek model, the histaminergic model, and a chronic itch model. Moreover, our system detected significant differences in scratching behavior between control and mice treated with an anti-itch drug. Taken together, we have established a novel deep learning-based system that could replace manual quantification for mouse scratching behavior in different itch models and for drug screening.

RESUMO

Wetlands constitute very important ecological areas. The aim of this study was to quantify the soil losses due to fluvial erosion from 2006 to 2008 in two riverbanks under three types of vegetal coverage dominated by Haematoxylum campechianum, Dalbergia brownei and Brachiaria mutica, in the Pantanos de Centla Biosphere Reserve, SE Mexico. The relationship between the texture, organic matter and pH of soils and soil losses was evaluated. We used erosion sticks to estimate soil losses in 18 plots (three plots per type, three vegetation types, two riverbanks). Soil loss decreased in this order: H. campechianum>B. mutica>D. brownei indicating that D. brownei scrubland has the most potential to retain soil. The higher erosive impact within H. campechianum sites can be related with the low density of these trees in the study areas, as well as the lack of association with other types of vegetation that could reinforce the rooting of the soil profile. Furthermore, soil losses in H. campechianum sites were dependent on soil texture. The soils under this type of vegetal coverage were mainly sandy, which are more vulnerable to the erosive action in comparison with fine textured soils or soils with higher clay content, like the ones found in D. brownei and B. mutica sites. Soil losses of 100 % in the second year (B. mutica plots) can be attributed to the distribution of roots in the upper soil layer and also to livestock management along riverbanks. This study recognizes the importance of D. brownei scrublands in riverbank soil retention. Nevertheless it is necessary to consider the role of an entire vegetal community in future research.

RESUMO

INTRODUCTION: Primary breast lymphoma by definition is localized to one or both the breasts with or without involvement of regional lymph nodes with no other extra mammary site involvement. Role of surgery in the treatment armamentarium is less well defined. PRESENTATION OF CASE: A 62year old post-menopausal lady was found to have 1.2×1.2×0.6cm mass in the left breast on diagnostic mammography. Core needle biopsy reveals marginal zone lymphoma. Complete staging work-up was performed which was negative for regional spread and metastatic foci. She was diagnosed to have a localized stage IE extra nodal low grade marginal zone lymphoma of the MALT type involving breast. She received radiotherapy to the breast and is in remission at follow up after one year. DISCUSSION: Accurate diagnosis requires adequate tissue biopsy either by a core needle or a surgical biopsy for histopathologic evaluation and immunophenotyping. Immunohistochemically the tumors are positive for pan B-cell markers (CD 19, CD 20, CD22, and CD 79a) and lack T cell markers. For stage IE Marginal zone lymphoma radiotherapy alone and for stage IIE combined chemo-radiation offers the best possible control of the disease. Progression free survival for MZL is better than diffuse large B-cell lymphoma. CONCLUSION: At present the best evidence support radiotherapy in localized disease and combined chemo-radiation in advanced disease. Role of surgery is minimal and is seen in case of diagnostic dilemma. Rarity of the diagnosis precludes large scale prospective studies making therapeutic decisions difficult and challenging.

RESUMO

Despite the increasing impact of heavy metal pollution in southern Mexico due to urban growth and agricultural and petroleum activities, few studies have focused on the behavior and relationships of these pollutants in the biotic and abiotic components of aquatic environments. Here, we studied the bioaccumulation of heavy metals (Cd, Cr, Ni, Pb, V, Zn) in suspended load, sediment, primary producers, mollusks, crustaceans, and fish, in a deltaic lagoon habitat in the Tabasco coast, with the aim to assess the potential ecological risk in that important wetland. Zn showed the highest concentrations, e.g., in suspended load (mean of 159.58 mg kg(-1)) and aquatic consumers (15.43-171.71 mg kg(-1)), particularly Brachyura larvae and ichthyoplankton (112.22-171.71 mg kg(-1)), followed by omnivore Callinectes sp. crabs (113.81-128.07 mg kg(-1)). The highest bioconcentration factors (BCF) of Zn were observed for planktivore and omnivore crustaceans (3.06-3.08). Zn showed a pattern of distribution in the food web through two pathways: the pelagic (where the higher concentrations were found), and the benthic (marsh plants, sediment, mollusk, fish). The other heavy metals had lower occurrences in the food web. Nevertheless, high concentrations of Ni and Cr were found in phytoplankton and sediment (37.62-119.97 mg kg(-1)), and V in epiphytes (68.64 mg kg(-1)). Ni, Cr, and Cd concentrations in sediments surpassed international and national threshold values, and Cd entailed a "considerable" potential risk. These heavy metals are most likely transferred into the food web up to fishes through the benthic pathway. Most of the collected fishes are residents in this type of habitat and have commercial importance. Our results show that the total potential ecological risk in the area can be considered as "moderate". Nevertheless, heavy metal values were similar or surpassed the values from other highly industrialized tropical coastal regions.
