RESUMO

In experiments with mature Wistar male rats under irradiation by dose of 5 Gy the effect of emoxypine, citomedine and echinacea purpurea on the content of liposoluble vitamin A, carotene, vitamin E and its metabolites (quinone and oxidized tocopherol) in blood plasma, spleen, liver and testes was studied. It was shown the drugs under study mobilized the internal reserves of these vitamins and promoted effective functioning of vitamin E redox system. Mechanisms of their action are different. The drugs might be used as radioprotectors, but they exhaust the reserves of the liposoluble vitamins. Therefore they should be used in a combination with vitamin preparations.

RESUMO

It has been shown in experiments on dogs that the constant electric current (tension 20 V, exposition time 30 sec) transmitted through an animal organism induces the development of hypercoagulation and hyperfibrinolysis. In the course of these changes in blood coagulation and fibrinolysis an important role is played by tissue hemocoagulating compounds liberated from arteria, veins, muscles and nerves. It is concluded that the ejection of blood coagulation factors from tissues results from the changes in the functional state of cellular membranes, and their permeability in particular.

RESUMO

It is shown that atheroarteriosclerosis develops with attendant hypercoagulation changes in the blood of animals kept on an antioxidant-free diet for 100 days. The changes in blood coagulation are caused by disturbances in the procoagulant properties of the tissue (heart and aorta) cell membranes and those of the red cells. The role of cell membranes in the development of hypercoagulation due to changes in their cell thromboplastic properties is discussed.

RESUMO

It was shown in dog experiments that intravenous infusion of 30 and 70 U/kg of thrombin leads to the development of a short-term hypercoagulation phase which is replaced by long-term hypocoagulation and marked stimulation of fibrinolysis. An electrocardiogram provides evidence that after the infusion of 70 U/kg of thrombin acute ischemic disorders of coronary circulation develop in the dogs and remain for 3 days. The authors contend that the appearance of high doses of thrombin in the vascular channel is not a physiological reaction.

RESUMO

In dogs, i.v. administration of thrombin leads to hypocoagulation, consumption of fibrinogen, increasing fibrinolysis, and to a slight raise of antithrombin. This has no connection with appearance of natural anticoagulants and fibrinolytic agents in the lymph stream. After the heterotransfusion shock, apart from hypocoagulation and hyperfibrinolysis in the blood, the incoagulability of lymph developes due to a sharp increase of free heparin in it. The hypocoagulation after the heterotransfusion shock seems to depend on appearance of anticoagulants in the tissue fluid and lymph. Common and different features of thrombin and heterogenic blood effects on the mechanism of hypocoagulation and hyperfibrinolysis, are discussed.

RESUMO

In a group of volunteers of different age, engaged in health-improving running for various time, a level of antioxidants and a capacity of vascular wall to synthesize the antiaggregating substances were increased as well as blood coagulation parameters were decreased. These alterations were especially important for men because of the favourable effect of the running on blood rheology and microcirculation as well as on metabolism of cell and tissues.

Examinations carried out on intact animals and healthy people have revealed a paired and multiple correlation with exists between thromboplastic properties of erythrocytes, the level of malondialdehyde accumulation in their membranes and the superoxide dismutase activity.

RESUMO

The treatment of erythrocytes with different enzymes (neuraminidase, trypsin and papain) has been studied for its effect on the lipid peroxidation in their membranes, activity of antioxidant enzymes and on the ability of such erythrocytes to interact with neutrophilic leucocytes. It is shown that desialization of the membrane erythrocyte structures causes the intensification of lipid peroxidation in the membranes, decrease of their stability. Simultaneously, the interaction of neutrophils increases. The treatment with proteases causes the opposite changes. It is shown possible to split out the end molecules of sialic acids with exposure of the subterminal residues of B-galactose as a result of the effect of active oxygen forms. An assumption is advanced concerning the functional conjugation of the free-radical oxidation and proteolysis in provision of stability of the cell membranes.

It is stated that fluoride intoxication promotes a sharp intensification of the peroxidation processes in the parodontium tissues. It is caused by a respirator explosion of neutrophils, a decrease in activity of antioxidant system enzymes, thus leading to disturbances of microcirculation and blood coagulation, ischemia development. The last factor can retard the enzymatic oxidation in a cell due to hypoxia, injures lysosomal membranes promoting partial autolysis of parodontium tissues, cell structures, intercellular substance. The interaction of blood with destructive elements of cells and collagen favours the development of trombohemorrhagic reactions in the parodontium tissues. A generalized damage of parodontium arises which intensifies under the effect of any other pathogenetic factors.

Polypeptide cytomedine, isolated from renal tissues has been studied for its regulatory effect on hemocoagulation and lipid peroxidation with fluoric intoxication (FI). FI was caused by inoculation of laboratory animals (guinea pigs) with sodium fluoride (100 mg/kg) for 14 days. Following it polypeptide (0.1 mg/kg) was introduced intramuscularly for 7 days. The development of FI was expressed by hypercoagulation delay of fibrinolysis with para-coagulation products appearing in blood decrease of antiaggregation activity of the renal tissue. These phenomena were estimated as manifestations of the first phase of disseminated intravascular blood coagulability (DIC-syndrome). The above reactions proceeded simultaneously with lipid peroxidation activation decrease of the antioxidant protection. The necrotic-dystrophic processes developed in renal and hepatic parenchyma. The renal peptide-cytomedine induced the normalization of lipid peroxidation in blood and renal tissues and fibrinolysis, the decrease in the concentration of para-coagulation products. The pathological changes decreased both in the renal and hepatic tissues. It is possibly, a result of the normalization of secretion and reabsorption in the kidneys. Thus, cytomedine of the kidney exerts a pronounced regulatory and protective effect in the case of acute renal pathology. These results correspond to the conception of the peptidergic organism regulation.

RESUMO

Theoretical grounds of the existence (in the organism) of physiological process of the neutrophil granulocyte-erythrocyte interaction are presented. This process underlies damage and destruction of old erythrocytes. A conclusion is made concerning the peroxide mechanism of this interaction. This process possibly takes place under physiological conditions and becomes more pronounced after splenectomy .

RESUMO

The effect of 3- and 5-day fasting on coagulation blood properties, antioxidant system activity, and the content of oxidized hemoglobin was studied in 2 series of observations over 22 normal volunteers. Fasting was discovered to lead to the reduction of blood plasma and red cell coagulation, to the deterioration of platelet aggregation, rise in the oxidized hemoglobin content, and to the increase in red cell resistance to peroxide hemolysis. The changes revealed were shown to favour the maintenance of the blood in a liquid state, to lower the risk of intravascular coagulation and thrombus formation.

Periodontitis induction causes a drastic elevation of free-radical lipid peroxidation, of ceruloplasmin level, and a reduction of antioxidant enzyme activities. Blood analysis shows hypercoagulation, appearance of paracoagulation products, reduced fibrinolysis, enhanced proaggregation activity of the periodontium, elevated levels of circulating immune complexes; this may by regarded as adaptation failure manifesting by these reactions. Morphologic examinations show foci of destructive changes. Administration of periodontal cytomedin reduced the level of free-radical oxidation, enhanced the activity of antioxidant enzymes, normalized antiaggregation activity of the periodontium and hemostasis and fibrinolysis parameters, and reduced the level of circulating immune complexes. The inflammatory reaction disappears completely, that is seen from reduced ceruloplasmin level and clinically. Morphologic study shows disappearance of foci of destruction. Thus, cytomedin of periodontal tissues may be regarded as an effective agent for the therapy of experimental periodontitis.

RESUMO

The effect of renal cytomedin on coagulative and microcirculatory hemostasis and lipid peroxidation was studied in 30 Wistar rats with induced Heymann's nephritis. The course of nephritis caused activation of lipid peroxidation, development of the syndrome of disseminated intravascular coagulation of blood in the animals, and diminution of antiaggregation properties of the kidney. Administration of the polypeptide normalized these processes. Thus, the discussed cytomedin produces a marked positive regulatory effect in experimental renal pathology.

Rats with spontaneous periodontitis develop lipid peroxidation in the blood and periodontal tissues, that is associated with hypercoagulation and fibrinolysis inhibition, reduced antiaggregation activity of the periodontal tissues. Administration of periodontal cytomedin results in normalization of lipid peroxidation, blood coagulation, and euglobulin lysis, as well as in increase in the activities of periodontal antioxidant enzymes and antiaggregation properties. Cytomedin effect on lipid peroxidation and antioxidant enzymes was more manifest in periodontal tissues than in the blood. Visual examination has shown an improvement of the periodontal tissue status, thus indicating good prospects for such therapy.

RESUMO

Effects of polypeptide bioregulators isolated from periodontal tissue on free-radical oxidation of lipids and the microcirculatory hemostasis during the development of chronic stress were under study. Increased reactions of lipid free-radical oxidation and blood hydrocortisone levels, denudation of dental necks were observed in animals during stress, which was explained by disordered reaction of microcirculatory hemostasis assessed from the antiaggregation activity of the periodontium. Administration of a polypeptide bioregulator led to a reduction of free-radical oxidation of lipids, to an increase in the activities of antioxidative enzymes, and hence, to a reduction of the injury to the cellular structures of the periodontium due to decrease of disorders of the microcirculatory hemostasis and ischemia in it.

During a 5-year follow-up of rescue party members exposed to radioactive effects of the Chernobyl accident no material changes were observed versus healthy donors in the content of calcium, cholesterol, atherogenic lipoproteins, dienic conjugates, insulin, thyroxin, triiodothyronine, thyroglobulin, IgA, IgG and fibrinogen. Inhibition of peroxidation and emergence of hypocoagulation are attributed to a reduced respiratory splash of the neutrophils. Concentrations of hydrocortisone, testosterone, IgM rose, while the activity of some antioxidant enzymes remained stable.
