RESUMO

The course of bronchial asthma can be accompanied by cognitive impairments. However, the relationship between cognitive dysfunction and asthma has not been fully revealed, nor has it been fully established what causes cognitive impairments in patients with asthma. There is an opinion that transient hypoxia and persistent systemic inflammation with insufficient control of bronchial asthma can be accompanied by neurotoxicity in relation to the hippocampus and indirectly lead to deterioration of cognitive functions. Comorbid conditions, such as obesity, allergic rhinitis, and depressive states can increase cognitive dysfunction in asthmatics. The review considers the pathophysiology of cognitive dysfunction in patients with bronchial asthma, as well as the impact of comorbid conditions on the cognitive status. This information will allow systematizing the available knowledge about the state of cognitive functions in asthma for timely detection and correction of their impairments and, ultimately, optimization of the management of these patients.

We performed a comparative study of the parameters of chemiluminescence of blood neutrophils in patients with different severity of chronic obstructive pulmonary disease in its different periods. The maximum values of induced and spontaneous chemiluminescence were recorded at moderate severity of the disease during exacerbation. Low levels of chemiluminescence indicators were found in severe chronic obstructive pulmonary disease in the stable phase. The values of the induction period of the chemiluminescent response in patients with moderate chronic obstructive pulmonary disease were higher than in the control group. Correlations between the values of induced chemiluminescence of neutrophils and the respiration function parameter FEV1 were established, which may indicate the influence of multidirectional changes in the functional activity of systemic neutrophils on the development and worsening of airway obstruction in chronic obstructive pulmonary disease patients.

RESUMO

In patients with chronic obstructive pulmonary disease, the levels of cytokines IL-1ß, IL-4, IL-8, TNFα, and IFNγ depended on the degree of bronchial obstruction and severity and period of the disease. The maximum levels of IL-4, IL-8, and TNFα were observed in severe chronic obstructive pulmonary disease during exacerbation. The highest concentration of IL-1ß and IFNγ were recorded during activation of inflammation in patients with moderate bronchial obstruction. The revealed correlations between the tested cytokines and spirometry parameters make it possible to consider the levels of these proteins as quantitative markers of systemic inflammation progression.

RESUMO

The article presents the results of study of in-family (child-parents) relationships in families with children suffering with diagnosed diabetes mellitus type I. The purpose of study is to investigate the disorders of family relationships in families of such kind. The established disorders allow to develop psychotherapeutic measures both for children and their parents. Materials and methods. The study covered 30 families with children of primary school age (7-10 years old) with diabetes mellitus. The families with children with severe concomitant diseases and broken families were excluded of the sampling. The control group consisted of 30 relevant families with healthy children. The in-family relationships were explored using a number of psychological tests applied to children and their mothers. The results. In mothers of the studied families prevailed authoritarian type of upbringing, which was expressed in tendency to hyper-protect and to over-control. The phobia of losing child and the tendency to reject it were also established. The children suffered from high level of anxiety. They had more positive attitude to mother than to father. The conclusion. The established disorders of in-family relationships in families with children with diabetes mellitus can serve as targets for psychotherapeutic activities both with children and their parents.

RESUMO

The article considers various mental reactions of psychiatrists and narcologists against the background of the SARS-CoV-2 pandemic, based on a brief scientific analysis of publications and experimental psychological research methods. The purpose of study was to identify impact of the COVID-19 pandemic on mental health of psychiatrists and psychiatrists-narcologists. The results of survey of psychiatrists and narcologists are presented. The study was carried out using such test techniques as life quality scale SF-36 (Health Status Survey), Lazarus questionnaire "Coping Behaviors" adjusted to conditions of Russia and Hamilton scales to assess depression and anxiety (HDRS, HARS). The results.The low level of psychological well-being, moderate degree of preference for all coping strategies were established. The anxiety was determine in 34.8% of respondents and depression in 8.7%. It was established that actually, the impact of the COVID-19 pandemic on medical workers is conditioned by complex of mental reactions (from anosognosia to expressed anxiety and depressive disorders) and is accompanied by increasing general anxiety.

RESUMO

The article presents the results of pilot study implemented to determine cognitive disorders in patients taking "Spice". The purpose of research is to identify targets of psychological correction interventions to prevent cognitive deficiency in patients addicted to synthetic cannabinoids. The pilot sampling consisted of 30 patients taking "Spice". The control group consisted of 30 healthy individuals. Along with social consequences, one of the important focuses of the study was investigation of cognitive sphere of patients. A number of pathopsychologic tests was applied. The study established that patients demonstrated disorders of various functions of memory, attention, mental pace and thinking. In particular, disorders of short-term, long-term and mediate memory and mental tempo were revealed. Also, reduced activity of attention took place. The distortion and decreasing of generalization and poor prehension occurred reliably more often as compared with control group. The established cognitive disorders in patients taking "Spice" can be considered as targets for psychological correction interventions to prevent cognitive deficiency in patients addicted to synthetic cannabinoids.

A type-VII clathrate with a Eu2+ guest embedded into a Ni-P covalent framework, EuNi2P4, was synthesized by a standard two-stage ampoule synthesis and confirmed to crystallize in the orthorhombic space group Fddd with unit cell parameters a = 5.1829(1) Å, b = 9.4765(1) Å, and c = 18.9900(1) Å. A general technique for studying the lattice and magnetic properties of REE containing compounds is proposed. The temperature and field dependences of electrical resistivity ρ(T,H), magnetization M(T,H), magnetic susceptibility χ(T,H), heat capacity Cp(T), and unit cell parameters a(T), b(T), c(T), and volume V(T) were experimentally studied and analyzed at different pressures in the temperature range of 2-300 K. A cascade of anomalies in the studied dependences was identified and attributed to the magnetic phase transformation and peculiar lattice contributions at temperatures below 20 K. As a result of comparison with an isostructural clathrate SrNi2P4, the parameters of the magnetic and lattice contributions were determined. It is characteristic that the phase transition from the paramagnetic to the magnetically ordered state is not reflected in the temperature changes of the lattice parameters due to weak bonds between guest europium atoms and the Ni-P host matrix. We have constructed a tentative H-T phase diagram based on the M(T) and M(H) data, which includes 6 different phases. It is established that the anomalous lattice contribution to the clathrate heat capacity CTLS(T) appears due to the effect of two-level systems (TLS) in the Eu2+ subsystem on the thermodynamic properties of EuNi2P4. The values of TLS parameters as well as the parameters of the magnetic subsystem of the clathrate were determined.

We studied changes in the transcription of genes encoding cytokines (TNF, IL-6, IL-10, and IL-32), cell activation markers (ICAM1, CD38, Fas, and FCGRIII), ROS production catalyst (NOX2), autophagy (Beclin 1, LC3B, and p62) and apoptosis (BAX, BCL2) regulators in peripheral blood mononuclear cells upon contact with quantum dots CdSe/ZnS-MPA and CdSe/CdSeZnS/ZnS-PTVP. Up-regulation of TNF, ICAM1, Fas, p62 mRNA and down-regulation of the FCGRIII and NOX2 mRNA in response to the presence of quantum dots were revealed. The formation of serum protein corona on the surface of quantum dots abolished this effect.

A sample of lutetium carboboride LuB2C was synthesized from a mixture of lutetium hydride, boron and carbon by annealing in argon. The temperature dependence of the heat capacity Cp(T) (2-300 K) and lattice parameters a(T), b(T), and c(T) (5-300 K) of the carboboride was experimentally determined. The experimental values of the heat capacity were fitted with the approximation Cp(T) = aT + ΣCD + CE + CTLS(T). Here the first term is the electronic contribution, the second is the sum of the Debye components, the third is the Einstein contribution, and the fourth is the contribution to the heat capacity due to the vibrations of the two-level systems which are formed in the Lu-subsystem due to the asymmetry of the B-C atomic arrangement around the Lu3+-ions and, as a consequence, the possible transition of the lutetium atoms between spatially close, but energetically non-equivalent positions. A strong anisotropy of the thermal expansion of the carboboride was revealed. Along the c axis the coefficient of thermal expansion monotonically increases; in the basal plane, the expansion is practically not observed. The temperature dependence of the unit cell volume Vu(T) has been analyzed in the Debye-Einstein approximation taking into account the electronic contribution and effect of two-level systems. The values of the Gruneisen parameters corresponding to different modes of the phonon spectrum of the carboboride have been determined. The frequencies of the lattice vibrations, determined in a Raman scattering experiment, are in satisfactory agreement with the parameters obtained from Cp(T) using the Debye-Einstein approximation. Using ab initio band theory methods and an exchange-correlation functional in the PBE form in the VASP package, it was established that the total energies of these two crystal structures differ by no more than 0.01 eV f.u.-1. Calculations of the thermodynamic properties of LuB2C yielded similar results for orthorhombic and tetragonal phases of the carboboride.

We studied the influence of magnetite nanoparticles (FeO•Fe2O3) and quantum dots (CdSe/ZnS coated with mercaptopropionic acid) on the expression of 5 common reference genes (BA, B2M, PPIA, UBC, and YWHAZ) in peripheral blood cells from 20 volunteers by reverse transcription PCR method. The stability of the expression of reference genes varied depending of the cells type and chemical structure of nanoparticles. The level of YWHAZ mRNA after exposure by nanoparticles demonstrated highest stability in lymphocytes, neutrophils, and monocytes. Stability of YWHAZ expression was confirmed by Western blotting. Our findings suggest that YWHAZ is the most suitable as the reference gene.

The temperature dependencies of heat capacity, CP(T), and cubic unit cell parameter, a(T), were experimentally obtained in the range of 2-300 K for the compounds Sn24P19.2I8, Sn20Zn4P20.8I8, and Sn17Zn7P22I8, which belong to a family of type-I clathrates. The experimental data were analyzed in the frames of the Debye-Einstein approximation, further accounting for the contributions of positional disorder in the clathrate frameworks as well as those of defect modes arising from the distribution of guest atoms over unequal in energy but close in space positions inside the framework cages. By fitting the experimental data, the Debye and Einstein characteristic temperatures describing the dynamics of the framework and guest atoms, respectively, were obtained. Their analysis revealed peculiar dependencies of the characteristic temperatures upon the number of substituted zinc atoms and the concentration of vacancies in the framework, which are discussed in this paper.

The expression of cancer-testis (CT) genes varies with tumor type. There are tumors with high, low, and intermediate gene expressions. Tumor cells of different origin are characterized by ST gene co-expression. The expression of ST genes increases in later stages of tumor development in the presence of metastases. In colon cancer, the tumor samples showed most frequently MAGE-A and SSX mRNA. The peripheral blood samples displayed most commonly XAGE, MAGE-C, and SSX mRNA. In patients with colon cancer, the expression of TSP50, MAGE-A(1-6), and SSX1,2,4 genes was associated with a poor prognosis, that of MAGE-C1 and XAGE1 was related to a favorable prognosis.

The hypo-glycated mucin I (MUC I) frequently is redundantly expressed in cells of breast carcinoma that is considered as an unfavorable prognostic marker for patients with breast cancer. The purpose of study is to evaluate in tumor areas of patients with breast cancer the interrelationships between expression of MUC I and genes participating in implementation of immune reactions. The technique of polymerase chain reaction with reverse transcription in real-time was applied to investigate expression of genes of MUC I, Fas, ICAMI, FcyR3A, FcyR3B, FoxP3, IL2Ra, IL32 and TNF in tumor areas of 40 patients with breast cancer. The level of mRNA of each of analyzed genes was evaluated relatively to three genes of household. The rate of detection of mRNA MUC I amounted to 80%, mRNA Fas - 95%, mRNA ICAM I - 97.5%, mRNA FcyR3A - 95%, mRNA FcyR3B - 32.5%, mRNA FoxP3 - 85%, mRNA IL2Ra - 87.5%, mRNA IL32 - 75%, mRNA TNF - 95%. The direct correlation was established between levels of mRNA MUC I, ICAM I, IL32, FcyR3A and FoxP3. The most strong relationship was registered between levels of expression of genes MUC I and IL32 (r-0.72; p=0.0001). In comparison with MUC I negative breast carcinoma, MUC I positive breast carcinoma characterized by higher level of mRNA of pro-inflammatory cytokines IL32 and ЕТА, molecules of adhesion ICAM I and molecular marker of T-regulative cells FoxP3. The given profile of expression of genes in tumors of patients with breast cancer demonstrates relationship of expression of MUC I with inflammation and its modulation by T-regulative cells. The effect of expression of analyzed genes on phenotype of cancer cells and course of disease are discussed.

Single-nucleotide polymorphism rs5498 E469K (A/G) in ICAM-1 gene modulates functional activity of this protein and is associated with the risk of some diseases. Tumor cell rs5498 E469K (A/G) genotypes were compared with genotypes of patients with colorectal and breast cancer, residents of the Nizhniy Novgorod region. Specimens of peripheral blood and tumor foci from patients with colorectal and breast cancer and peripheral blood specimens from normal subjects were analyzed. The frequency of genotypes of single-nucleotide rs5498 E469K (A/G) sequence in the Nizhny Novgorod population coincide with the frequency of genotypes characteristic of residents of Germany. Comparison of genotypes of peripheral blood and tumor cells from patients with colorectal and breast cancer showed loss of cancer cell heterozygosity. The frequency of heterozygosity loss increases with colorectal cancer progress from one stage to another.

Mucin 1 (MUC1) is a multistructural and multifunctional protein that is involved in regulating diverse cellular activities. This strongly glycosylated transmembrane protein forms a mucous gel on the surface of epithelial cells that protects the cells from injury. MUC1 acts as a signaling molecule and transcription factor modulating metabolism and resistance to bacterial-induced inflammation. This article presents a review of the relationship between structural and functional changes of the MUC1 and the characteristics of cancer cells. The alteration in MUC1 expression level, a number of structural forms, protein glycosylation and localization occurs in cancer cells. These alterations lead to metabolic reprogramming associated with proliferation, resistance to hypoxia and angiogenesis which affects the survival of cancer cells. Furthermore, cancer cells can take advantage of MUC1 interaction with adhesion molecules for invasion and metastasis. Thus, MUC1 plays a key role both in the homeostasis of epithelial cells and in cancer progression. Understanding the role of MUC1 expression in tumor cells survival is important for the development of new monitoring and therapeutic approaches for the treatment MUC1 positive maligancies.

AIM: To investigate the efficacy and safety of nebulized budesonide and systemic glucocorticosteroids (GCS) (SGCS) in the treatment of an exacerbation of chronic obstructive pulmonary disease (COPD) and their effects on the serum concentration of soluble leukocyte differentiation antigens. MATERIALS AND METHODS: Seventy-eight hospitalized patients with an acute exacerbation of COPD were randomized into two groups: 1) 37 patients took nebulized budesonide 4 mg/day; 2) 41 patients received intravenous prednisolone. The symptoms of COPD, forced expiratory volume in one second (FEV1) and other spirometric indicators, peripheral blood oxygen saturation (SpO2), and adverse events were studied. The serum levels of the soluble adhesion molecules CD50 (sCD50) and CD54 (sCD54) and the lymphocyte activation molecules CD38 (sCD38) and CD25 (sCD25) were investigated by an enzyme immunoassay. RESULTS: There was a significant resolution of the symptoms of COPD, FEV1, and SpO2 in both groups after treatment. The incidence of hyperglycemia episodes was lower in the budesonide group than in the sGCS group. GCSs caused a decrease in the serum level of soluble interleukin-2 receptor (sCD25) in both groups. A prednisolone cycle, unlike a budesonide one, was found to reduce the concentrations of sCD54, sCD50, and sCD38. CONCLUSION: Nebulized budesonide is an effective and safe alternative to SGCS in treating an exacerbation of COPD. Inhaled GCSs, unlike SGCSs, exhibit anti-inflammatory activity, but exert no immunosuppressive activity.

Fas-induced apoptosis plays an important role in the mechanisms of tissue injury in myocardial infarction. The level of membrane Fas mRNA was elevated during the postinfarction period in the blood of patients and did not change in response to levocarnitine. The mRNA level of soluble Fas, inhibiting Fas-dependent apoptosis, remained normal during the first 7 days, but increased 14 days after myocardial infarction, which corresponded to previously detected increase of serum level of soluble Fas molecules. Addition of levocarnitine, inhibiting Fas-dependent apoptosis, to therapy caused no changes in the level of soluble Fas mRNA, presumably because of similar effects of soluble Fas and levocarnitine on the apoptotic processes in myocardial infarction.

RESUMO

It is shown that the exposure of heparinized venous human blood diluted in phosphate buffer saline to extremely weak alternating magnetic fields of the ultralow-frequency (1 Hz, 600 nT; 4.4 Hz, 100 nT; 16.5 Hz, 160 nT) in combination with a collinear static magnetic field of 42 microT at physiological temperatures, causes a sharp 3-4 fold increase in its chemiluminescence after addition of luminol.

The immune-enzyme analysis was applied using anti-CD38 monoclonal antibodies to detect serum content of total (sCD38) and oligomeric fractions of soluble molecules CD38 (ol.sCD38) in patients with cervix cancer and malignant neoplasms of uterine body. The three- and four-fold increasing of serum level of sCD38 is demonstrated both under cervix cancer and malignant neoplasms of uterine body. The level of ol.sCD38 decreased in patients with cervix cancer and increased in case of patients with malignant neoplasms of uterine body. The serum content of ol.sCD38 was increased under adenocarcinoma of uterus and glandular epidermoid cancer. The content of sCD38 increased under adenocarcinoma of uterus but decreased under sarcoma of uterus. The highest level of soluble molecules of CD38 was established in patients with highly differentiated tumors and under invasion into endometrium but not into miometrium. On the whole, alterations in content of soluble molecules CD38 are associated with characteristics of tumor process that indicates at their monitoring significance under malignant neoplasms of uterus.

It is shown that an exposure of pupae of the mealworm beetle Tenebrio molitor to the combined static (42 µT) and very weak alternating (250 nT) magnetic fields exerts different influence, depending on the frequency of the alternating magnetic field, on duration of metamorphosis processes in these insects. For instance, an exposure of pupae to weak combined magnetic fields, adjusted to the frequency of ion cyclotron resonance for glutaminic acid (4,4 Hz), stimulates metamorphosis process--a transitional stage from pupae to imago lasts shorter. An inhibiting effect was observed when adjusted to the frequency of ion cyclotron resonance for Ca2 (32,2 Hz). At some frequencies this effect is not seen. For instance, an exposure at a frequency of ion cyclotron resonance for K+ (16,5 Hz) exerts no noticeable effect on the duration of the pupal metamorphosis stage.
