This study aimed to evaluate the effect of chemical modifications of the structure of active compounds on the skin permeation and accumulation of ibuprofen [IBU] from the acrylic pressure-sensitive adhesive used as a drug-in-adhesives matrix type transdermal patch. The active substances tested were ibuprofen salts obtained by pairing the ibuprofen anion with organic cations, such as amino acid isopropyl esters. The structural modification of ibuprofen tested were Ibuprofen sodium salt, [GlyOiPr][IBU], [AlaOiPr][IBU], [ValOiPr][IBU], [SerOiPr][IBU], [ThrOiPr][IBU], [(AspOiPr)2][IBU], [LysOiPr][IBU], [LysOiPr][IBU]2, [PheOiPr][IBU], and [ProOiPr][IBU]. For comparison, the penetration of unmodified ibuprofen and commercially available patches was also investigated. Thus, twelve transdermal patches with new drug modifications have been developed whose adhesive carrier is an acrylate copolymer. The obtained patches were characterized for their adhesive properties and tested for permeability of the active substance. Our results show that the obtained ibuprofen patches demonstrate similar permeability to commercial patches compared to those with structural modifications of ibuprofen. However, these modified patches show an increased drug permeability of 2.3 to even 6.4 times greater than unmodified ibuprofen. Increasing the permeability of the active substance and properties such as adhesion, cohesion, and tack make the obtained patches an excellent alternative to commercial patches containing ibuprofen.

RESUMO

The measurement of nitric oxide (NO) in exhaled air is used in diagnostics and monitoring the pathologies not only in the respiratory system but also in the oral cavity. It has shown a huge increase in its level in asthma and diseases of the oral cavity. It seems reasonable to undertake research on the impact of inflammation on the level of NO in exhaled air. The aim of the study is to make an evidence-based review of the application of NO levels in exhaled air in the diagnosis of inflammation and treatment monitoring on the basis of selected measuring devices. METHODS AND RESULTS: This paper presents an example of the application of NO measurement in exhaled air in individual human systems. Selected measuring devices, their non-invasiveness, and their advantages are described. DISCUSSION: The usefulness of this diagnostic method in pathologies of the oral cavity was noted. CONCLUSIONS: Measuring the level of NO in exhaled air seems to be a useful diagnostic method.

This study aimed to investigate the potential of photoreactive acrylate patches as systems for transdermal drug delivery, in particular, using more renewable alternatives and more environmentally friendly synthesis routes of transdermal patches. Therefore, the aim of this study was to develop a transdermal patch containing ibuprofen and investigate its performance in vitro through the pigskin. Transparent patches were prepared using four acrylate copolymers with an incorporated photoinitiator. Two types of transdermal patches based on the photocrosslinking acrylic prepolymers with isobornyl methacrylate as biocomponent and monomer increasing Tg ("hard") were manufactured. The obtained patches were characterized for their adhesive properties and tested for permeability of the active substance. It turns out that patches whose adhesive matrix is photoreactive polyacrylate copolymers have a higher cohesion than patches from commercial adhesives, while the modification of the copolymers with isobornyl methacrylate resulted in an improvement in adhesion and tack. This study demonstrates the feasibility of developing photoreactive acrylic-based transdermal patches that contain biocomponents that can deliver a therapeutically relevant dose of ibuprofen.

The concentration of fungal spores in the air is traditionally considered as a proxy of allergen exposure. However, in vitro experiments have shown that the allergenicity of Alternaria spores varies depending on ecophysiological and developmental factors. Despite the potential clinical significance of these findings, it has never been verified in outdoor environments. This study, therefore, aims to investigate variability in the amount of the major allergen (Alt a 1) released from Alternaria spores in outdoor air. During the 3-year monitoring study (2014-2016), the median seasonal allergenicity of Alternaria spores exceeded 8.6 × 10-3 pg Alt a 1/spore. The most allergenic spores were collected during the driest and the most polluted season (with respect to seasonal concentrations of ozone, sulphur dioxide, and particulate matter). Within the season, daily spore allergenicity ranged from 2.4 to 34.7 × 10-3 pg Alt a 1/spore (5th-95th percentile). No repeatable effects of weather and pollution on short-term variations in Alternaria spore allergenicity were found. However, during the episodes when high-potency spores were recorded, the air masses arrived from eastern directions. Contrary, the spores with the lowest allergenicity were related to western winds. This suggests that factors such as source area (habitat types) and species diversity could be responsible for the varying exposure to Alternaria allergens. Our findings show that high and low-potency spores are recorded in the air; therefore, the airborne concentrations of fungal spores alone may not be sufficient to provide allergy sufferers and healthcare professionals with information about allergen exposure.

Iron-sulfur (FeS) clusters are prosthetic groups critical for the function of many proteins in all domains of life. FeS proteins function in processes ranging from oxidative phosphorylation and cofactor biosyntheses to DNA/RNA metabolism and regulation of gene expression. In eukaryotic cells, mitochondria play a central role in the process of FeS biogenesis and support maturation of FeS proteins localized within mitochondria and in other cellular compartments. In humans, defects in mitochondrial FeS cluster biogenesis lead to numerous pathologies, which are often fatal. The generation of FeS clusters in mitochondria is a complex process. The [2Fe-2S] cluster is first assembled on a dedicated scaffold protein (Isu1) by the action of protein factors that interact with Isu1 to form the "assembly complex". Next, the FeS cluster is transferred onto a recipient apo-protein. Genetic and biochemical evidence implicates participation of a specialized J-protein co-chaperone Jac1 and its mitochondrial (mt)Hsp70 chaperone partner, and the glutaredoxin Grx5 in the FeS cluster transfer process. Finally, various specialized ISC components assist in the generation of [4Fe-4S] clusters and cluster insertion into specific target apoproteins. Although a framework of protein components that are involved in the mitochondrial FeS cluster biogenesis has been established based on genetic and biochemical studies, detailed molecular mechanisms involved in this important and medically relevant process are not well understood. This review summarizes our molecular knowledge on chaperone proteins' functions during the FeS protein biogenesis.

The spores of Cladosporium Link. are often present in the air in high quantities and produce many allergenic proteins, which may lead to asthma. An aerobiological spore monitoring program can inform patients about the current spore concentration in air and help their physicians determine the spore dose that is harmful for a given individual. This makes it possible to develop optimized responses and propose personalized therapy for a particular sensitive patient. The aim of this study was to assess the extent of the human health hazard posed by the fungal genus Cladosporium. For the first time, we have determined the number of days on which air samples in Poland exceeded the concentrations linked to allergic responses of sensitive patients, according to thresholds established by three different groups (2800/3000/4000 spores per 1m3 of the air). The survey was conducted over three consecutive growing seasons (April-September, 2010-2012) in three cities located in different climate zones of Poland (Poznan, Lublin and Rzeszow). The average number of days exceeding 2800 spores per cubic meter (the lowest threshold) ranged from 61 (2010) through 76 (2011) to 93 (2012), though there was significant variation between cities. In each year the highest concentration of spores in the air was detected in either Poznan or Lublin, both located on large plains with intensive agriculture. We have proposed that an effective, science-based software platform to support policy-making on air quality should incorporate biological air pollutant data, such as allergenic fungal spores and pollen grains.

BACKGROUND: Back pain is common among people working in the sitting position. Quality of the sitting position depends among others on the standard of work station. Dental units are built on the basis of contemporary ergonomic workplace concepts, while medical assistant workstations are typically computer office workstations. The purpose of the study was pain estimation in association with the identification of the spine arrangements during sitting work with better and worse ergonomic adjustment. MATERIAL AND METHODS: The study involved 80 women working in the sitting position suffering from pain - 40 dentists working on dental units with a higher ergonomic potential and 40 medical assistants working on stations with lower ergonomic potential. The study included interviews, pain assessment and working spine arrangements analysis using the ultrasonographic measurement system. RESULTS: There were exceeds of work parameters in particular segments of the spine observed in all examined persons. The relationship between pain intensity and work parameters as frontal bending amplitude of lumbar spine segment, transversal amplitude of cervical spine segment and transversal amplitude of lumbar spine segment were observed in the group of dentists. There was observed the relationship between pain severity and the values of sagittal bending amplitude of cervical spine segment and thoracic spine segment in the group of medical assistants. Other determinants of pain also have been identified. CONCLUSIONS: Exceeded values of work parameters were observed amongst subjects working both at the higher and the lower ergonomic potential work stations. The working postures of people working in the sitting position are similar only to their external characteristics. Pain depends on working position. The pain intensity is determined by other factors: length of professional experience, duration of the pain, the number of working hours per week, body mass index (BMI). Med Pr 2018;69(5):509-522.

INTRODUCTION: The role of long-distance transported (LDT) Ambrosia pollen in inducing new sensitization and affecting sensitization rates in Artemisia-sensitized patients is unclear. AIM: The aim of this study was to estimate the degree of cross-sensitization to Ambrosia/Artemisia allergens in citizens of Poznan (Western Poland). This area is covered by extensive Artemisia populations but does not currently have local Ambrosia populations. MATERIAL AND METHODS: Sera of 119 patients were tested by fluoroenzyme immunoassay (CAP-FEIA system) against pollen allergen extracts of Artemisia vulgaris and Ambrosia artemisiifolia, an allergenic component of A. vulgaris (nArt v 1), and an allergenic component of A. artemisiifolia (nAmb a 1). Skin prick tests (SPTs, n = 86) were performed with pollen allergen extracts of A. vulgaris and A. artemisiifolia. Artemisia and Ambrosia pollen in ambient air was collected (1996-2013) by a Hirst type volumetric trap sited at roof level (33 m). RESULTS: The SPT showed that the prevalence of sensitization to Ambrosia and Artemisia pollen exceeded 3.5%, and 10.5%, respectively. The measurements of IgE in blood serum (CAP-FEIA) revealed that among Ambrosia-sensitized patients 90.1% (20/22 patients) were concomitantly sensitized to Artemisia. 59.1% (13/22) of these patients reacted to nArt v 1, suggesting primary sensitization to Artemisia pollen. Only 2 (9.1%) patients were mono-sensitized to Ambrosia pollen extract, but surprisingly not to nAmb a 1. CONCLUSIONS: The LDT Ambrosia pollen had a negligible effect on the rate of sensitization to Ambrosia allergens in Poznan and did not increase the prevalence of sensitization to Artemisia pollen in this region. However, the majority of patients showing hypersensitization to Artemisia pollen might also present symptoms during elevated episodes of LDT of Ambrosia pollen.

Temperature is the environmental factor that systematically changes for decades and, as in plants and animals, can significantly affect the growth and development of fungi, including the abundance of their sporulation. During the time of study (2010-2012), a rapid increase in air temperature was observed in Poland, which coincided with the substantial decrease in rainfall. The increase in annual mean temperatures at three monitoring sites of this study was 0.9 °C in Lublin and Rzeszow (east Poland) and 2.0 °C in Poznan (west Poland). Such warming of air masses was comparable to the average global air temperature rise in the period of 1880-2012 accounting for 0.85 °C, as reported by the Intergovernmental Panel on Climate Change. Moreover, there was a substantial decrease in rainfall, ranging from 32.7 % (Poznan) to 43.0 % (Rzeszow). We have demonstrated that under such conditions the mean and median values of total Cladosporium spore counts significantly increased and the spore seasons were greatly accelerated. Moreover, earlier start and later end of the season caused its extension, lasting from over 20 days in Rzeszow to around 60 days in Lublin and Poznan, when the cumulative amount of 5-95 % of spores was considered. The time of reaching the cumulative amount of 50 % of spores was up to 25 days earlier (difference in Poznan between 2010 and 2012). There was also a striking acceleration of the date of the maximal Cladosporium spore concentration per cubic metre of air (26 days for Lublin, 43 for Poznan and 56 for Rzeszow).

A significant increase in summer temperatures has been observed for the period 1996-2011 in Poznan, Poland. The phenological response of four weed taxa, widely represented by anemophilous species (Artemisia spp., Rumex spp. and Poaceae and Urticaceae species) to this recent climate warming has been analysed in Poznan by examining the variations in the course of airborne pollen seasons. Pollen data were collected by 7-day Hirst-type volumetric trap. Trends in pollen seasons were determined using Mann-Kendall test and Sen's slope estimator, whereas the relationships between meteorological and aerobiological data were established by Spearman's rank correlation coefficient. Significant trends in pollen data were detected. The duration of pollen seasons of all analysed taxa increased (from +2.0 days/year for Urticaceae to +3.8 days/year for Rumex), which can be attributed to a delay in pollen season end dates rather than earlier start dates. In addition, the intensity of Artemisia pollen seasons significantly decreased and correlates with mean July-September daily minimum temperatures (r = -0.644, p < 0.01). In contrast, no significant correlations were found between temperature and characteristics of Rumex pollen seasons. The results of this study show that observed shifts in weed pollen seasons in Poznan, i.e. longer duration and later end dates, might be caused by the recorded increase in summer temperature. This influence was the strongest in relation to Artemisia, which is the taxon that flowers latest in the year. The general lack of significant correlations between Rumex and Urticaceae pollen seasons and spring and/or summer temperature suggests that other factors, e.g. land use practices, could also be partially responsible for the observed shifts in pollen seasons.

C-reactive protein is the widely used marker in a clinical practice but also a mediator of the inflammatory process. In the introduction the authors describe the structure and function of the C-reactive protein in relation with inflammatory response to different morbidity stimulations. After this, there have been seen the oscillations of C-reactive protein plasma level of healthy people and in inflammatory, neoplastic and immunological diseases with an explanation of the clinical utilities. The current knowledge about presence of C-reactive protein in a different type of circulatory diseases, in heart failure and arrhythmia has been discussed. The importance of the role of C-reactive protein, especially high-sensivity C-reactive protein, in the prediction of circulatory diseases of the healthy members of the population and the prognostic value of C-reactive protein has been underlined.

BACKGROUND: Exhaled nitric oxide is helpful in the diagnosis of the inflammation process. The study aimed to analyze the impact of the COVID-19 disease on the oral bacterial flora of patients using complete dentures with a diagnostic device that measures the level of NO in exhaled air. MATERIALS AND METHODS: The study included patients using upper and lower acrylic complete dentures. All patients participating in the study were vaccinated against COVID-19. The patients were divided into two groups. A dental examination was conducted in each group. The NO concentration was measured using the Vivatmo Pro device. An oral microbiological examination was performed by taking a swab from the bottom of the mouth. RESULTS: There were no statistically significant differences in the distribution of NO in relation to the number of bacteria from isolated families in the study and control groups and no statistically significant correlations between the level of NO and the number of bacteria from all families in the control and study group. Significantly higher NO values were present in the vaccinated and COVID-19-positive history population compared to the vaccinated and with no COVID-19 history population (patients with no clinical symptoms of infection or unaware they had COVID-19). CONCLUSIONS: There are statistically significant differences in NO distribution in the considered populations: vaccinated and sick, and vaccinated and with a negative history of COVID-19. The measurement of NO in exhaled air can be a complementary, non-invasive diagnostic and inflammation monitoring method.

Hoard finds from the Bronze Age have appeared all over Europe, prompting questions about their functions (either as raw materials for recycling or votive objects). The hoard trove of raw materials from Przybyslaw in Greater Poland is an interesting example of a discovery that is related to the foundry activities of Late Bronze Age and Early Iron Age communities (c. 600 BC). The deposit consists of fragments of raw materials that were damaged end products intended for smelting. The research included the characterisation of the material in terms of the variety of the raw materials that were used. The individual elements of the hoard were characterised in terms of their chemical compositions, microstructures, and properties. A range of modern instrumental research methods were used: metallographic macroscopic and microscopic observations by optical microscopy (OM), scanning electron microscopy (SEM), chemical-composition analysis by X-ray fluorescence spectroscopy (ED-XRF), X-ray microanalysis (EDS), and detailed crystallisation analysis by electron microscopy with an emphasis on electron backscatter diffraction (EBSD). As part of this study, model alloys were also prepared for two of the selected chemical compositions, (i.e., CuPbSn and CuPb). These alloys were analysed for their mechanical and technological properties. This research of the hoard from Przybyslaw (Jarocin district, Greater Poland) has contributed to the recognition and interpretation of the function and nature of the hoard by using modern research and modelling methods as a cultic raw material deposit.

This study assesses the value of the CXCR3 ligands CXCL9/MIG, CXCL10/IP-10 and CXCL11/I-TAC when used to supplement the standard infection markers C-reactive protein (CRP) and procalcitonin (PCT) in the diagnostic algorithm of neutropenic fever in children with cancer. The concentration of CRP, PCT and chemokines was determined during the first hour of fever and 12-24 h afterwards in pediatric oncology patients with neutropenia. Among 100 consecutive febrile episodes in neutropenic patients, 34 cases demonstrated fever of unknown origin (FUO) (group A), 47 demonstrated mild clinically or microbiologically proven infection (Group B) and 19 severe infection (Group C). Significantly higher PCT-1 levels were found in group C (0.24 ng/mL) vs. group A (0.16 ng/mL), and PCT-2 in group C (1.2 ng/mL) vs. A (0.17 ng/mL), and in C vs. B (0.2 ng/mL). Chemokine concentrations (I-TAC-1, IP-10-1, IP-10-2) were significantly lower in Group A vs. B+C; I-TAC 1: 48.64 vs. 70.99 pg/mL, p = 0.03; IP-10 1: 59.95 vs. 96.84 pg/mL, p = 0.04; and IP-10 2: 102.40 vs. 149.39 pg/mL, p = 0.05. The selected pro-inflammatory chemokines I-TAC and IP10 might help to distinguish cancer patients with febrile neutropenia with the highest risk of infection. Although procalcitonin could serve as a marker of a high risk of infection, its delayed response diminishes its usefulness.

This study aimed to evaluate the effect of introducing structural modification of ibuprofen in the form of an ion pair on the permeability of ibuprofen through the skin and the properties of the adhesive layer of the medical patch produced. The active substances tested were the salts of ibuprofen obtained by pairing the anion of ibuprofen with organic cations such as propyl esters of amino acids such as tyrosine, tryptophan, histidine, or phenylalanine. For comparison, the penetration of unmodified ibuprofen and commercially available patches was also tested. Acrylate copolymers based on isobornyl methacrylate as a biocomponent and a monomer increasing the T g ("hard") were used to produce the adhesive layer of transdermal patches. The obtained patches were characterized in terms of adhesive properties and tested for the permeability of the active ingredient and the permeability of the active ingredient through the skin. This study demonstrates the possibility of developing acrylic-based photoreactive transdermal patches that contain biocomponents that can deliver a therapeutically appropriate dose of ibuprofen.

In this paper, the results from studies regarding near-eutectic Al-Si alloys with Sn as an alloying addition are presented. In most Al-Si alloys, tin is regarded as a contaminant; thus, its amount is limited to up to 0.3 wt.%. The few studies that can be found in the literature regarding the behaviour of tin in aluminium alloys suggest the beneficial effect of this element on selected properties. However, these results were obtained for hypereutectic Al-Si alloys or wrought aluminium alloys. In our studies, the influence of tin contents of up to 1.7 wt.% was determined on the AlSi10 alloy. Thermal analysis, measurements of the mechanical properties of the cast and heat-treated alloy, metallographic observations (light microscopy, scanning electron microscopy), and EDS (X-ray energy dispersive spectrometry) measurement allowed us to fully describe the effect of tin on the aluminium alloy. The results of the thermal analysis showed changes in the range of the α-Al solution crystallisation and the α+ß eutectic through a decrease in the alloy's solidification start point and eutectic solidification point. As a result, the elongation of the alloy was more than double in the AlSi10Sn1.7 alloy, with an A5 value of 8.1% and a tensile strength that was above 200 MPa.

BACKGROUND: The most common cause of implant loss and deteriorating restoration aesthetics is infection and chronic inflammation of the tissues around the implants. Inflammation in the oral cavity, confirmed by clinical and histopathological examination and determination of exhaled nitric oxide, is a situation which may cause the complications on the whole human body. Elimination of the patology in the oral cavity in some cases is the only resonable treatment. The aims and objectives of our work is to present a gradual treatment of advanced infalmmation and present huge reduction stamp of inflammation measured with marker nitric oxide (NO) in exhaled air. MATERIALS AND METHODS: Simple treatment containing elimantion of pathology in the oral cavity was conducted. Patient that came to the dental practice suffered from the inflammation caused by lack of proper hygiene. First aid in this situation was to eliminate the inflammation which may affect negatively for general health. At first visit full hygienization was performed, at the second visit roots of abutment teeth and implants were removed under local anesthesia along with cystic changes. RESULTS: The hygiene precedures and extraction of the unsteady inflammationprosthetic restorations significantly decreased the level of NO in exhaled air. CONCLUSIONS: During the examination of the patient coming to the dental practice great attention should be paid to the coexistence of pathologies related to the oral cavity. Omission of a dental examination and possible elimination of odontogenic foci may affect the implication of the results of general diagnostics and subsequent treatment. Measuring the level of NO on exhaled air seems to be useful diagnostic method.

Research related to the development of a dental infiltrant for minimally invasive treatment of initial caries of hard dental tissues is presented. The formulation of the developed infiltrant material includes typical methacrylate monomers used in dentistry, an author's adhesion monomer containing metronidazole, a photoinitiating system and yttrium trifluoride (YF3). The main objective of the study was to evaluate penetration into decalcified root cementum using scanning electron microscope of an experimental preparation with the characteristics of a dental infiltrant compared to a commercial preparation with the addition of YF3 as a contrast agent. Microscopic observations showed that YF3 particles virtually did not penetrate deep into the root cementum-this was mainly due to the particle size of YF3. Observations of cementum and root dentin tissue infiltration: resin tissue infiltration was visible to a depth of about 80 to 120 µm without the use of a tracer, which, due to agglomeration and particle size, remained on the cementum surface or in the resin used for inlaying. There were no differences between the degree of penetration of an experimental preparation with the characteristics of a dental infiltrant, as compared to a commercial preparation.

The tapeworm species Cloacotaenia megalops (Nitzsch in Creplin, 1829) is characterized by a very specific morphology. A particularly distinct feature is a large almost square scolex in which four fleshy suction cups are embedded and a rudimentary rostellum. During standard studies of wild duck cestode fauna in northwestern Poland, some morphological differences were observed among specimens, mainly in the shape of scolices and hermaphroditic proglottids of C. megalops isolated from different bird species. This paper attempts to demonstrate the impact of the host species on morphology of the parasite. The study material consisted of 39 individuals of C. megalops (19 from Aythya fuligula, 10 from Anas platyrhynchos, 4 from Bucephala clangula, 4 from Aythya marila and 2 from A. ferina). The isolated parasites were fixed and stored in 70% ethanol and solid preparations were made. Selected external and internal structures of the parasite were measured and photographed using a Zeiss microscope, a microscope camera Opta Tech 2.1 and Opta View 6.0.2.2. It was stated that the sizes of the most important elements of the parasite morphology are generally consistent with those presented in the available literature, nevertheless some differences were observed in both morphology and sizes of some anatomical structures, especially in the form of scolices and hermaphroditic proglottids between specimens isolated from hosts representing three different eco-tribes (Aythyini, Anatini and Mergini).

Cloacotaenia megalops (Nitzsch in Creplin, 1892) is a polyxenic and cosmopolitan tapeworm from the family Hymenolepididae. Its generic name derives from their typical location (cloaca), and the typical final hosts which are birds typically associated with water and marsh environments: Anseriformes, Galliformes and Gruiformes. In Poland, the presence of C. megalops has been observed so far in 16 species of ducks from the Baltic coast, the Mazurian Lake District, Wielkopolsko-Kujawska Lowland, Mazovian Lowland, and Podlasie Lowland. In Western Pomerania, quantitative structure analyses were only carried out on Anas platyrhynchos, and therefore the aim of this study was the detailed analysis of environmental populations of C. megalops in wild ducks. The examined tapeworms were isolated from the digestive tract of 1005 wild ducks representing 17 species belonging to three different eco-tribes: Anatini (n=225), Aythyini (n=413) and Mergini (n=367), from northwestern Poland. During the Study 187 C. megalops were found in 89 birds (8.8% of examined ducks) belonging to 7 species: Anas crecca (common teal), A. querquedula (garganey), A. platyrhynchos (mallard) (Anatini); Aythyaferina (pochard), A. fuligula (tufted duck), A. marila (greater scaup) (Aythyini) and Bucephala clangula (goldeneye) (Mergini). The results show the differences in the quantitative structure of C. megalops among the examined species of ducks. The highest prevalence was found in mallard (18.6%) and the lowest in greater scaup (3.2%). The highest mean intensity was observed in greater scaup (4.0), and the lowest in garganey and common teal (1.0). Relative density was at a similar level in the tested birds. Based on the ratio of dominance, it was found that C. megalops is a rare species in the cestodofauna in the examined birds.
