RESUMO

Nanostructured metallic glass films (NMGF) can exhibit surface and intrinsic effects that give rise to unique physical and chemical properties. Here, a facile synthesis and electrochemical, structural, and morphologic characterization of Pd-Au-Si based MGs of approximately 50 nm thickness supported on Si/SiO2 is reported. Impressively, the maximum total hydrogen charge stored in the Pd-Au-Si nanofilm is equal to that in polycrystalline Pd films with 1 µm thickness in 0.1 m H2 SO4 electrolyte. The same NMGF has a volumetric desorption charge that is more than eight times and 25 % higher than that of polycrystalline PdNF and Pd-Cu-Si NMGF with the same thickness supported on Si/SiO2 , respectively. A significant number of nanovoids originating from PdHx crystals, and an increase in the average interatomic spacing is detected in Pd-Au-Si NMGF by high-resolution TEM. Such a high amount of hydrogen sorption is linked to the unique density fluctuations without any chemical segregation exclusively observed for this NMGF.

BACKGROUND: We assessed the clinical utility of ß-amyloid (Aß) imaging with (18)F-florbetaben (FBB) in mild cognitive impairment (MCI) by evaluating its prognostic accuracy for progression to Alzheimer's disease (AD), comparing semiquantitative with visual scan assessment, and exploring the relationships among Aß, hippocampal volume (HV) and memory over time. METHODS: 45 MCI underwent FBB positron emission tomography, MRI and neuropsychological assessment at baseline and 2 years and clinical follow-up at 4 years. Positive FBB (FBB+), defined by a cortical to cerebellar cortex standardised uptake value ratio (SUVR) ≥ 1.45, was compared with visual assessment by five readers. Amnestic MCI (aMCI) was defined by a composite episodic memory (EM) Z-score of <-1.5. RESULTS: At baseline, 24 (53%) MCI were FBB+. Majority reads agreed with SUVR classification (κ 0.96). In 2 years, 18 (75%) FBB+ progressed to AD compared with 2 (9.5%) FBB-, yielding a predictive accuracy of 83% (95% CI 61% to 94%). Four FBB- developed non-AD dementia. Predictive accuracies of HV (58% (95% CI 42% to 73%)) and aMCI status (73% (95% CI 58% to 81%)) were lower. Combinations did not improve accuracy. By 4 years, 21 (87.5%) FBB+ had AD whereas 5 (24%) FBB- had non-AD dementia yielding a predictive accuracy of 94% (95% CI 74% to 99%). While the strong baseline association between FBB SUVR and EM declined over 2 years, the association between EM and HV became stronger. FBB SUVR increased 2.2%/year in FBB+ with no change in FBB-. CONCLUSIONS: (18)F-florbetaben Aß imaging facilitates accurate detection of prodromal AD. As neurodegeneration progresses, and in contrast with the early stages of the disease, hippocampal atrophy and not Aß, seems to drive memory decline. TRIAL REGISTRATION NUMBER: NCT01138111.

Thin metal films on polymer substrates are of interest for flexible electronic applications and often utilize a thin interlayer to improve adhesion of metal films on flexible substrates. This work investigates the effect of a 10 nm Cr interlayer on the electro-mechanical properties of 50 nm Au films on polyimide substrates. Ex situ and in situ fragmentation experiments reveal the Cr interlayer causes brittle electro-mechanical behaviour, and thin Au films without an interlayer can support strains up to 15% without significantly degrading electrical conductivity.

Nanocrystalline and nanotwinned materials achieve exceptional strengths through small grain sizes. Due to large areas of crystal interfaces, they are highly susceptible to grain growth and creep deformation, even at ambient temperatures. Here, ultrahigh strength nanotwinned copper microstructures have been stabilized against high temperature exposure while largely retaining electrical conductivity. By incorporating less than 1 vol% insoluble tungsten nanoparticles by a novel hybrid deposition method, both the ease of formation and the high temperature stability of nanotwins are dramatically enhanced up to at least 400 °C. By avoiding grain coarsening, improved high temperature creep properties arise as the coherent twin boundaries are poor diffusion paths, while some size-based nanotwin strengthening is retained. Such microstructures hold promise for more robust microchip interconnects and stronger electric motor components.

Advanced in vitro models called "organ-on-a-chip" can mimic the specific cellular environment found in various tissues. Many of these models include a thin, sometimes flexible, membrane aimed at mimicking the extracellular matrix (ECM) scaffold of in vivo barriers. These membranes are often made of polydimethylsiloxane (PDMS), a silicone rubber that poorly mimics the chemical and physical properties of the basal membrane. However, the ECM and its mechanical properties play a key role in the homeostasis of a tissue. Here, we report about biological membranes with a composition and mechanical properties similar to those found in vivo. Two types of collagen-elastin (CE) membranes were produced: vitrified and nonvitrified (called "hydrogel membrane"). Their mechanical properties were characterized using the bulge test method. The results were compared using atomic force microscopy (AFM), a standard technique used to evaluate the Young's modulus of soft materials at the nanoscale. Our results show that CE membranes with stiffnesses ranging from several hundred of kPa down to 1 kPa can be produced by tuning the CE ratio, the production mode (vitrified or not), and/or certain parameters such as temperature. The Young's modulus can easily be determined using the bulge test. This method is a robust and reproducible to determine membrane stiffness, even for soft membranes, which are more difficult to assess by AFM. Assessment of the impact of substrate stiffness on the spread of human fibroblasts on these surfaces showed that cell spread is lower on softer surfaces than on stiffer surfaces.

Contrary to the electrochemical energy storage in Pd nanofilms challenged by diffusion limitations, extensive metal-hydrogen interactions in Pd-based metallic glasses result from their grain-free structure and presence of free volume. This contribution investigates the kinetics of hydrogen-metal interactions in gold-containing Pd-based metallic glass (MG) and crystalline Pd nanofilms for two different pore architectures and nonporous substrates. Fully amorphous MGs obtained by physical vapor deposition (PVD) co-sputtering are electrochemically hydrogenated by chronoamperometry. High-resolution (scanning) transmission electron microscopy and corresponding energy-dispersive X-ray analysis after hydrogenation corroborate the existence of several nanometer-sized crystals homogeneously dispersed throughout the matrix. These nanocrystals are induced by PdHx formation, which was confirmed by depth-resolved X-ray photoelectron spectroscopy, indicating an oxide-free inner layer of the nanofilm. With a larger pore diameter and spacing in the substrate (Pore40), the MG attains a frequency-independent impedance at low frequencies (∼500 Hz) with very high Bode magnitude stability accounting for enhanced ionic diffusion. On the contrary, on a substrate with a smaller pore diameter and spacing (Pore25), the MG shows a larger low-frequency (0.1 Hz) capacitance, linked to enhanced ionic transfer in the near-DC region. Hence, the nanoporosity of amorphous and crystalline metallic materials can be systematically adjusted depending on AC- and DC-type applications.

Compared to their conventional polycrystalline Pd counterparts, Pd79Au9Si12 (at%) - metallic glass (MG) nanofilm (NF) electrocatalysts offer better methanol oxidation reaction (MOR) in alkaline medium, CO poisoning tolerance and catalyst stability even at high scan rates or high methanol concentrations owing to their amorphous structure without grain boundaries. This study evaluates the influence of scan rate and methanol concentration by cyclic voltammetry, frequency-dependent electrochemical impedance spectroscopy and a related equivalent circuit model at different potentials in Pd-Au-Si amorphous NFs. Structural and compositional differences for the NFs are assessed by high-angle annular dark-field scanning transmission electron microscopy (HAADF-STEM), energy dispersive X-ray (EDX) mapping and X-ray diffraction (XRD). The ratio of the forward to reverse peak current density ipf/ipb for the MG NFs is ∼2.2 times higher than for polycrystalline Pd NFs, evidencing better oxidation of methanol to carbon dioxide in the forward scan and less poisoning of the electrocatalysts by carbonaceous (e.g. CO, HCO) species. Moreover, the electrochemical circuit model obtained from EIS measurements reveals that the MOR occurring around -100 mV increases the capacitance without any significant change in oxidation resistance, whereas CO2 formation towards lower potentials results in a sharp increase in the capacitance of the Faradaic MOR at the catalyst interface and a slight decrease in the corresponding resistance. These results, together with the high ipf/ipb = 3.37 yielding the minimum amount of carbonaceous species deposited on the thin film during cyclic voltammetry and stability in the alkaline environment, can potentially make these amorphous thin films potential candidates for fuel-cell applications.

Pliable and lightweight thin-film magnets performing at room temperature are indispensable ingredients of the next-generation flexible electronics. However, conventional inorganic magnets based on f-block metals are rigid and heavy, whereas the emerging organic/molecular magnets are inferior regarding their magnetic characteristics. Here we fuse the best features of the two worlds, by tailoring ε-Fe2O3-terephthalate superlattice thin films with inbuilt flexibility due to the thin organic layers intimately embedded within the ferrimagnetic ε-Fe2O3 matrix; these films are also sustainable as they do not contain rare heavy metals. The films are grown with sub-nanometer-scale accuracy from gaseous precursors using the atomic/molecular layer deposition (ALD/MLD) technique. Tensile tests confirm the expected increased flexibility with increasing organic content reaching a 3-fold decrease in critical bending radius (2.4 ± 0.3 mm) as compared to ε-Fe2O3 thin film (7.7 ± 0.3 mm). Most remarkably, these hybrid ε-Fe2O3-terephthalate films do not compromise the exceptional intrinsic magnetic characteristics of the ε-Fe2O3 phase, in particular the ultrahigh coercive force (∼2 kOe) even at room temperature.

The influence of irradiation on mechanical properties of polymer foils used in spacecraft applications has widely been studied via macroscopic tensile samples. An increase in the local resolution of this investigation can be achieved by reducing the sample's dimensions. A femtosecond laser enables a fast fabrication of micro-samples with dimensions from tens of µ m to the mm range, with ideally no influence on the material. Tensile experiments using such micro-tensile samples were conducted on FEP, Upilex-S and PET foils. The influence of the laser processing on the polymer foils was evaluated. Additionally an investigation of degradation due to electron irradiation was performed. Furthermore an outlook to extend this technique to depth-resolved measurements by preparing samples from locally thinned foils is presented. The study demonstrates the feasibility of femtosecond laser processing for rapid fabrication of micro-samples, enabling insights into the effect of electron irradiation on local mechanical properties of polymers.

RESUMO

The study of electromigration (EM) in metallisations for flexible thin film systems has not been a major concern due to low applied current densities in today's flexible electronic devices. However, the trend towards smaller and more powerful devices demands increasing current densities for future applications, making EM a reliability matter. This work investigates EM in 50 nm Au thin films with a 10 nm Cr adhesion layer on a flexible polyimide substrate at high current densities. Results indicate that EM does occur and could be used as a self-healing mechanism for flexible electronics.

OBJECTIVES: The aim of this study was to evaluate the diagnostic efficacy of gadobutrol enhanced preoperative breast magnetic resonance imaging (MRI) in 2 prospective studies. MATERIALS AND METHODS: Approval of ethics committees and informed consent from patients were obtained. Both Gadobutrol-Enhanced MR Mammography (GEMMA) trials followed a standardized protocol using 1.5 T scanners. After unenhanced scans, patients received 0.1 mmol/kg of gadobutrol for the dynamic study. Six independent blinded readers, 3 for GEMMA1 and 3 for GEMMA2, assessed unenhanced images and, 2 or more weeks apart, contrast-enhanced plus unenhanced breast MRI images (CE-BMRI), using a standard 5-region scheme. Another 6 independent readers (3 for each study) evaluated mammograms alone. Sensitivity was calculated taking into account the identification of regions harboring malignancies (within-patient sensitivity), whereas specificity was based on cancer-free breasts. The first patient from each center was used for site qualification and blinded reader training and excluded from the efficacy analyses. Reference standard was pathology for regions harboring malignancy and a combination of negative pathology, mammography, and ultrasound for cancer-free regions. RESULTS: Of 906 breast cancer patients enrolled in 13 countries in the 2 studies, 865 received gadobutrol and 787 were evaluated for diagnostic performance (390 in GEMMA1 and 397 in GEMMA2). Within-patient sensitivity, that is, the detection rate of malignant disease extent per patient, ranged from 80% to 89% for CE-BMRI and was significantly superior to unenhanced breast MRI alone (37%-73%) and to mammography alone (68%-73%) for all readers in both trials. Specifity of the CE-BMRI ranged from 83% to 95%. CONCLUSION: In a very large multicenter preoperative setting, gadobutrol-enhanced breast MRI demonstrated high levels of sensitivity and specificity, consistent with published data on breast MRI.

OBJECTIVE: To investigate brain ß-amyloid binding in subjects with Down syndrome (DS) using [(18)F]florbetaben PET imaging. METHODS: Thirty-nine subjects with DS (46.3 ± 4.7 years) were assessed with [(18)F]florbetaben PET imaging. Three blinded independent readers assessed the scans to provide a visual analysis. The primary quantitative imaging outcome was a standardized uptake value ratio (SUVR) obtained for 6 brain regions. Cognitive status was evaluated using the Dementia Screening Questionnaire for Individuals with Intellectual Disabilities (DSQIID). RESULTS: [(18)F]Florbetaben uptake was correlated with age (p < 0.0001, R(2) = 0.39); 90% of scans in subjects with DS aged 50 years or older (SUVR = 1.62 ± 0.26), 53% in those aged 45 to 49 years (SUVR = 1.43 ± 0.16), and 7% in those aged 40 to 45 years (SUVR = 1.27 ± 0.11) were visually assessed as positive. Visual and quantitative assessments were highly related (χ(2) = 11.3823, p = 0.0007; Cohen κ = 0.58). Only 2 of 34 participants were considered to have dementia by the DSQIID. CONCLUSIONS: Brain ß-amyloid binding, as measured by [(18)F]florbetaben, increases with age in DS. Subjects with DS who have no evidence of dementia demonstrate brain ß-amyloid binding in vivo, suggesting that [(18)F]florbetaben PET imaging may detect ß-amyloid in this at-risk population.

INTRODUCTION: (18)F-florbetaben and positron emission tomography were used to examine the relationships between ß-amyloid (Aß) deposition, cognition, hippocampal volume, and white matter hyperintensities in mild cognitive impairment (MCI). METHODS: Forty-five MCI participants were evaluated. A neocortical standardized uptake value ratio threshold ≥ 1.45 was used to discriminate high from low Aß burden. Correlations were adjusted for age, gender and years of education. RESULTS: High Aß burden was found in 53% of MCI. Regression analyses showed standardized uptake value ratio (r = -0.51, P = 0.0015) and hippocampal volume (r = 0.60, P = 0.024) both contributing to episodic memory impairment in independent fashion. White matter hyperintensities correlated with nonmemory cognition, and this correlation was particularly associated with Aß burden. CONCLUSION: Higher Aß deposition in MCI is associated with more severe memory impairment and is contributing to early amnestic symptoms independent of hippocampal atrophy.

UNLABELLED: Amyloid imaging with (18)F-labeled radiotracers will allow widespread use, facilitating research, diagnosis, and therapeutic development for Alzheimer disease. The purpose of the study program was to compare cortical amyloid deposition using (18)F-florbetaben and PET in controls and subjects with mild cognitive impairment (MCI), frontotemporal lobar degeneration (FTLD), dementia with Lewy bodies (DLB), vascular dementia (VaD), Parkinson disease (PD), and Alzheimer disease (AD). METHODS: One hundred nine subjects in 3 clinical studies at Austin Health were reviewed: 32 controls, 20 subjects with MCI, and 30 patients with AD, 11 with FTLD, 7 with DLB, 5 with PD, and 4 with VaD underwent PET after intravenous injection of 300 MBq of (18)F-florbetaben. Standardized uptake value ratios (SUVR) using the cerebellar cortex as a reference region were calculated between 90 and 110 min after injection. RESULTS: When compared with the other groups, AD patients demonstrated significantly higher SUVRs (P < 0.0001) in neocortical areas. Most AD patients (96%) and 60% of MCI subjects showed diffuse cortical (18)F-florbetaben retention. In contrast, only 9% of FTLD, 25% of VaD, 29% of DLB, and no PD patients and 16% of controls showed cortical binding. Although there was a correlation between Mini Mental State Examination and ß-amyloid burden in the MCI group, no correlation was observed in controls, FTLD or AD. CONCLUSION: (18)F-florbetaben had high sensitivity for AD, clearly distinguished patients with FTLD from AD, and provided results comparable to those reported with (11)C-Pittsburgh Compound B in a variety of neurodegenerative diseases.

PURPOSE The randomized First-Line Indolent Trial (FIT) was conducted in patients with advanced follicular lymphoma (FL), to evaluate the safety and efficacy of yttrium-90 ((90)Y) ibritumomab tiuxetan given as consolidation of complete or partial remission. This study of minimal residual disease was undertaken in parallel, to determine the rate of conversion from bcl-2 polymerase chain reaction (PCR) -detectable to -undetectable status and the corresponding effect on progression-free survival (PFS). PATIENTS AND METHODS Blood samples from 414 patients ((90)Y-ibritumomab, n = 208; control, n = 206) were evaluated using real-time quantitative polymerase chain reaction (RQ-PCR); 186 were found to have the bcl-2 rearrangement and were thus eligible for inclusion in the RQ-PCR analysis. Results Overall, 90% of treated patients converted from bcl-2 PCR-detectable to -undetectable disease status, compared with 36% in the control group. Treatment significantly prolonged median PFS in patients converting to bcl-2 PCR-undetectable status (40.8 v 24.0 months in the control group; P < .01, hazard ratio [HR], 0.399). In patients who had bcl-2 PCR-detectable disease at random assignment, treatment significantly prolonged median PFS (38.4 v 8.2 months in the control group; P < .01, HR, 0.293). CONCLUSION Eradication of PCR-detectable disease occurred more frequently after treatment with (90)Y-ibritumomab tiuxetan and was associated with prolongation of PFS.
