In this work, we extracted chitosan from marine amphipods associated with aquaculture facilities and tested its use in crop protection. The obtained chitosan was 2.5 ± 0.3% of initial ground amphipod dry weight. The chemical nature of chitosan from amphipod extracts was confirmed via Raman scattering spectroscopy and Fourier transform infrared spectroscopy (FTIR). This chitosan showed an 85.7-84.3% deacetylation degree. Chitosan from biofouling amphipods at 1 mg·mL-1 virtually arrested conidia germination (ca. sixfold reduction from controls) of the banana wilt pathogenic fungus Fusarium oxysporum f. sp cubense Tropical Race 4 (FocTR4). This concentration reduced (ca. twofold) the conidia germination of the biocontrol fungus Pochonia chlamydosporia (Pc123). Chitosan from amphipods at low concentrations (0.01 mg·mL-1) still reduced FocTR4 germination but did not affect Pc123. This is the first time that chitosan is obtained from biofouling amphipods. This new chitosan valorizes aquaculture residues and has potential for biomanaging the diseases of food security crops such as bananas.

Aquaculture sustainability is restricted by environmental drawbacks such as the pollution derived from the released organic waste. Integrated multi-trophic aquaculture (IMTA) aims to lower the input of this waste by culturing other species of low trophic level which feed on them. Despite the appealing idea of IMTA, its implementation is very limited in marine ecosystems. Focusing on marine fish farming, in general terms, fish farm waste is not expected to constitute a relevant food source for low-trophic level organisms cultured in the water column. We propose Regional Integrated Multitrophic Aquaculture (RIMTA) as a shift of paradigm in the way IMTA is used to sequester the dissolved exported waste and derived primary production generated by high trophic level cultures. RIMTA advocates for independent allocation of cultures of low and high trophic level species within the same water body. RIMTA implementation should be economically supported through tax benefits or nutrient quota trading schemes. Moving from IMTA to RIMTA should not only foster aquaculture sustainability but also the circular economy and the ecosystem services that the low trophic level cultures provide.

Pollution is one of the most significant issues that is currently impeding the development of fish farming. Integrated multi-trophic aquaculture (IMTA) has the potential to reduce the accumulation of organic wastes in the environment by using taxa of lower trophic levels such as filter feeders. However, the capacity of filter feeders to assimilate significant quantities of fish farm wastes has not yet been fully tested in situ. We analyzed the stable isotopes δ13C and δ15N in mussels from six fish farms and from six other areas that were not influenced by fish farming, at two water strata (surface and mid-water) across a marked gradient of eutrophication along more than 900 km of coastline in the Western Mediterranean. We found that the mussels did not directly assimilate fish farming wastes. Consequently, fish farming wastes did not constitute a major component of mussel diet, irrespective of local productivity and depth in the water column. These outcomes do not necessarily mean that IMTA is not suitable in other cases, but rather that there should be a shift in the rationale of IMTA by modifying the concept of direct assimilation of wastes to a more general approach of IMTA based on regional budgets of nutrients.

Effective ecosystem-based management requires understanding ecosystem responses to multiple human threats, rather than focusing on single threats. To understand ecosystem responses to anthropogenic threats holistically, it is necessary to know how threats affect different components within ecosystems and ultimately alter ecosystem functioning. We used a case study of a Mediterranean seagrass (Posidonia oceanica) food web and expert knowledge elicitation in an application of the initial steps of a framework for assessment of cumulative human impacts on food webs. We produced a conceptual seagrass food web model, determined the main trophic relationships, identified the main threats to the food web components, and assessed the components' vulnerability to those threats. Some threats had high (e.g., coastal infrastructure) or low impacts (e.g., agricultural runoff) on all food web components, whereas others (e.g., introduced carnivores) had very different impacts on each component. Partitioning the ecosystem into its components enabled us to identify threats previously overlooked and to reevaluate the importance of threats commonly perceived as major. By incorporating this understanding of system vulnerability with data on changes in the state of each threat (e.g., decreasing domestic pollution and increasing fishing) into a food web model, managers may be better able to estimate and predict cumulative human impacts on ecosystems and to prioritize conservation actions.


Hacia un Marco de Trabajo para la Evaluación y el Manejo de los Impactos Humanos Acumulativos sobre las Redes Alimenticias Marinas Resumen El manejo efectivo con base en los ecosistemas requiere entender la respuesta de los ecosistemas a múltiples amenazas humanas en lugar de enfocarse en amenazas individuales. Para entender holísticamente la respuesta de los ecosistemas a las múltiples amenazas antropogénicas es necesario saber cómo estas amenazas afectan a los diferentes componentes dentro de los ecosistemas y cómo alteran finalmente el funcionamiento de los ecosistemas. Usamos el estudio de caso de la red alimenticia del pasto marino del Mediterráneo (Posidonia oceanica) y la obtención de conocimiento de expertos en una aplicación de los pasos iniciales de un método para la evaluación de los impactos humanos acumulativos sobre las redes alimenticias. Produjimos un modelo de red alimenticia de pastos marinos, determinamos las principales relaciones tróficas, identificamos a las principales amenazas para los componentes de la red y evaluamos la vulnerabilidad de los componentes a esas amenazas. Algunas amenazas tuvieron impactos altos (p. ej.: infraestructura costera) o bajos (p. ej.: escorrentía agrícola) sobre todos los componentes de la red, mientras que otros (p. ej.: carnívoros introducidos) tuvieron impactos muy diferentes sobre cada componente. Partir al ecosistema en sus componentes nos permitió identificar amenazas no vistas previamente y reevaluar la importancia de las amenazas percibidas comúnmente como mayores. Al incorporar este entendimiento de la vulnerabilidad del sistema con datos sobre los cambios en el estado de cada amenaza (p. ej.: disminución de la contaminación doméstica e incremento de la pesca) al modelo de red alimenticia, los manejadores pueden ser capaces de estimar y predecir de mejor manera los impactos humanos acumulativos sobre los ecosistemas y priorizar las acciones de conservación.

During adverse atmospheric events, enormous damage can occur at marine aquaculture facilities, as was the case during Storm Gloria in the southeastern Spanish Mediterranean in January 2020, with massive fish escapes. Fishes that escape were caught by professional fishermen. The objective of this study was to identify biomarkers in fish that enable differentiation among wild fish, escaped farm-raised fish, and farm-raised fish kept in aquaculture facilities until their slaughter. We focused on gilthead sea bream (Sparus aurata). We used nuclear magnetic resonance to search for possible biomarkers. We found that wild gilthead sea bream showed higher levels of taurine and trimethylamine-N-oxide (TMAO) in their muscle and higher levels of ω-3 fatty acids, whereas farm-escaped and farmed gilthead sea bream raised until slaughter exhibit higher levels of ω-6 fatty acids. From choline, carnitine, creatinine, betaine, or lecithin, trimethylamine (TMA) is synthesized in the intestine by the action of bacterial microflora. In the liver, TMA is oxidized to TMAO and transported to muscle cells. The identified biomarkers will improve the traceability of gilthead sea bream by distinguishing wild specimens from those raised in aquaculture.

Environmental genomics is a promising field for monitoring biodiversity in a timely fashion. Efforts have increasingly been dedicated to the use of bacteria DNA derived data to develop biotic indices for benthic monitoring. However, a substantial debate exists about whether bacteria-derived data using DNA metabarcoding should follow, for example, a taxonomy-based or a taxonomy-free approach to marine bioassessments. Here, we showcase the value of DNA-based monitoring using the impact of fish farming as an example of anthropogenic disturbances in coastal areas and compare the performance of taxonomy-based and taxonomy-free approaches in detecting environmental alterations. We analysed samples collected near to the farm cages and along distance gradients from two aquaculture installations, and at control sites, to evaluate the effect of this activity on bacterial assemblages. Using the putative response of bacterial taxa to stress we calculated the taxonomy-based biotic index microgAMBI. The distribution of individual amplicon sequence variants (ASVs), as a function of a gradient in sediment acid volatile sulphides, was then used to derive a taxonomy-free bacterial biotic index specific for this data set using a de novo approach based on quantile regression splines. Our results show that microgAMBI revealed a organically enriched environment along the gradient. However, the de novo biotic index outperformed microgAMBI by providing a higher discriminatory power in detecting changes in abiotic factors directly related to fish production, whilst allowing the identification of new ASVs bioindicators. The de novo strategy applied here represents a robust method to define new bioindicators in regions or habitats where no previous information about the response of bacteria to environmental stressors exists.

Marine sediments are a major sink of organic matter, playing a crucial role in the global cycling of major elements. Macrofauna, through the reworking of particles and movement of solutes (bioturbation), enhances oxic conditions and the sediment metabolic capacity. Increases in the inputs of organic matter can lead to profound changes in the seabed and impact benthic ecological functions. Through a microcosm experiment, the effect of bioturbation of the polychaete Lumbrineris latreilli on biogeochemical fluxes under scenarios of increasing loads of organic matter was quantified. We found that bioturbation can buffer the negative consequences of anoxic conditions produced by organic enrichment, preventing the build-up of toxic by-products derived from anaerobic metabolic pathways by maintaining oxic conditions. However, the maintenance of oxic conditions by bioturbation is at the expense of limiting the sediment metabolic capacity. The maintenance of oxic conditions may limit anaerobic metabolic pathways, and consequently, the metabolic capacity of sediment. Thus, under organic matter pollution conditions, bioturbation may lessen the metabolic capacity of the sediment.

In this study, we assessed plastic accumulation in marine sediments due to finfish aquaculture using floating net-pens. We studied plastic concentrations around three fish farms located at the Mediterranean coastline of Spain. The macroplastic categories and abundances were determined by video monitoring, detecting the majority of elements (78%), including ropes, nets and fibres, a basket trap and a cable tie, close to the facilities, which were not exclusively linked to fish farming but also to fishing activities. Concentrations of microplastics (<5 mm) ranged from 0 to 213 particles/kg dry weight sediment with higher values in sites directly under the influence of the fish farms. Most particles (27.8%) were within the size fraction from 1.1 to 2.0 mm and fibre was the most common shape with 62.2%. The Infrared spectroscopy analysis showed that PE and PP were the predominant types of polymers analysed. In addition, changes in the enthalpy of melting (ΔHm (J/g)) and the degree of crystallinity indicate degradation of the microplastics analysed. This study shows that, in the studied fish farms, levels of microplastic pollution can be one order of magnitude lower compared to other areas suffering other anthropogenic pressures from the same or similar regions. Nevertheless, more research effort is needed to get concluding results.

Marine protected areas (MPAs) could be useful as fisheries management tools for the exportation of pelagic eggs, larvae and adult fish. A decreasing gradient of fish biomass across MPAs boundary may indicate export. We determine whether gradients of decreasing biomass of fish assemblage occurred in Tabarca Marine Reserve over two habitats with different continuity across the boundaries, to test if the patchy nature of the marine environment might act as a barrier for the fish export. In general, significant decreasing gradients in total fish biomass and biomass of some species were observed on P. oceanica and rocky substrates, independently of their different continuity through the reserve boundaries. Changes in the multivariate structure of the fish assemblage were correlated with the distance from integral reserve. All of these results support the hypothesis that the exportation of adult fish from Tabarca Marine Reserve occurs, and this process may influence the surrounding fished areas.

Net-cage fish farms attract a great number of wild fishes, altering their behaviour and possibly their physiology. Wild Mediterranean horse mackerel (Trachurus mediterraneus), sampled from populations aggregated around two Mediterranean fish farms and from two natural control populations, were analyzed for differences in body condition, stomach content and fatty acid composition. Pellets used to feed caged fish in both farms were also analyzed to identify their relationship with the fatty acid composition of tissue of wild fish. T. mediterraneus aggregated around the farms throughout the year although large seasonal changes in abundance and biomass occurred. Wild fish aggregated at farms mainly ate food pellets while control fish fed principally on juvenile fish and cephalopods. Wild fish that fed around the cages had a significantly higher body fat content than the control fish (7.30+/-1.8% and 2.36+/-0.7%, respectively). The fatty acid composition also differed between farm-associated and control fish, principally because of the significantly increased levels of linoleic (C18:2omega6) and oleic (C18:1omega9) acids and decreased docosahexaenoic acid (C22:6omega3) in farm-associated fish. The increased condition of wild fish associated with farms could increase the spawning ability of coastal fish populations, if wild fish are protected from fishing while they are present at farms. The fatty acids compositions could also serve as biomarkers to infer the influence of a fish farm on the local fish community, helping to better describe the environmental impact of fish farming.

A metabolomics approach was used to analyze effects of salmon farming on wild saithe (Pollachius virens) populations. Saithe fish were captured at two salmon farms and at two control locations around the island of Hitra, Norway. Changes in diet seem to drive changes in metabolic status of fishes. The liver and muscle tissues, from the fishes captured around the farm, showed higher levels of lactate and certain amino acids (glutamine, glutamate, and alanine) and lower levels of glucose and choline than the fishes captured in the control locations, far from the farm locations. The higher levels of lactate and amino acids could be related to the facility of obtaining food around the farm and the deficit in choline to the deficit of this nutrient in the salmon feed. At each location the fish were captured with either benthic gill nets and automatic jigging machines, and this feature showed also variations in different metabolites.

We investigated the changes on the mean trophic level of fish assemblages across different spatiotemporal scales, before and after a massive escape event occurred off La Palma (Canary Islands), which resulted in the release of 1.5 million fish (mostly Dicentrarchus labrax) into the wild. The presence of escaped fish altered significantly the mean trophic level of fish assemblages in shallow coastal waters. This alteration was exacerbated by the massive escape. A nearby marine protected area buffered the changes in mean trophic level but exhibited the same temporal patterns as highly fished areas. Moreover, escaped fish exploited natural resources according to their total length and possibly, time since escapement. New concerns arise as a "farming up" process is detected in shallow coastal fish assemblages where marine aquaculture is established.

Groupers species are extremely vulnerable to overfishing and many species are threatened worldwide. In recent decades, Mediterranean groupers experienced dramatic population declines. Marine protected areas (MPAs) can protect populations inside their boundaries and provide individuals to adjacent fishing areas through the process of spillover and larval export. This study aims to evaluate the effectiveness of six marine reserves in the Western Mediterranean Sea to protect the populations of three species of grouper, Epinephelus marginatus, Epinephelus costae and Mycteroperca rubra, and to understand in which circumstances MPAs are able to export biomass to neighbouring areas. All the studied MPAs, except one where no grouper was observed, were able to maintain high abundance, biomass and mean weight of groupers. Size classes were more evenly distributed inside than outside MPAs. In two reserves, biomass gradients could be detected through the boundaries of the reserve as an indication of spillover. In some cases, habitat structure appeared to exert a great influence on grouper abundance, biomass and mean individual weight, influencing the gradient shape. Because groupers are generally sedentary animals with a small home range, we suggest that biomass gradients could only occur where groupers attain sufficient abundance inside MPA limits, indicating a strongly density-dependent process.

Marine fish farms could cause environmental disturbances on the sediment due to uneaten food and fish faeces that impact the marine benthos. Polychaete assemblages are considered good indicators of environmental perturbations. The present study aimed to establish groups of polychaetes as potential indicators of fish farm pollution. This study was carried out in ten fish farms along the Spanish coast. Changes in polychaete assemblage were analyzed with meta-analysis and multivariate techniques. Abundance, richness and diversity showed significant decreases under fish farm conditions. Distribution patterns of polychaetes responded to combinations of physicochemical variables. The main ones are sulfide concentration, silt and clays percentage, and stable nitrogen isotope ratio. The results showed that some families are tolerant, Capitellidae, Dorvilleidae, Glyceridae, Nereididae, Oweniidae and Spionidae; while others are sensitive to fish farm pollution, Magelonidae, Maldanidae, Nephtyidae, Onuphidae, Paralacydoniidae, Paraonide, Sabellidae and also Cirratulidae in spite of being reported as a tolerant family.

Interannual variability (2003-2008) of meiofaunal assemblages were analyzed in sediments beneath fish cages (Impact group) and in areas not affected by aquaculture activities (Control group). Organisms responded with spatial and seasonal variation in meiofauna assemblages, with an abrupt increase of abundances in locations beneath fish cages throughout the study period. This increase was greater during the last sampling year (2008) and mainly due to high abundances of nematodes. Univariate analyses showed differences between control and impacted sites at both sites, however, only significant variations were found in Los Gigantes, which are consistent with seasonal meiofauna variations throughout the study period. These results are partially explained by differences in current velocity between both sampling areas. The Ne/Co index showed the same trend and it seems to be a reliable index in sediment slightly affected by aquaculture wastes. This index is especially recommended in oligotrophic areas (e.g. Canary Islands) where meiofaunal assemblages are poorly represented in terms of abundances.

BACKGROUND: Ecological traps form when artificial structures are added to natural habitats and induce mismatches between habitat preferences and fitness consequences. Their existence in terrestrial systems has been documented, yet little evidence suggests they occur in marine environments. Coastal fish farms are widespread artificial structures in coastal ecosystems and are highly attractive to wild fish. METHODOLOGY/PRINCIPAL FINDINGS: To investigate if coastal salmon farms act as ecological traps for wild Atlantic cod (Gadus morhua) and saithe (Pollachius virens), we compared proxy measures of fitness between farm-associated fish and control fish caught distant from farms in nine locations throughout coastal Norway, the largest coastal fish farming industry in the world. Farms modified wild fish diets in both quality and quantity, thereby providing farm-associated wild fish with a strong trophic subsidy. This translated to greater somatic (saithe: 1.06-1.12 times; cod: 1.06-1.11 times) and liver condition indices (saithe: 1.4-1.8 times; cod: 2.0-2.8 times) than control fish caught distant from farms. Parasite loads of farm-associated wild fish were modified from control fish, with increased external and decreased internal parasites, however the strong effect of the trophic subsidy overrode any effects of altered loads upon condition. CONCLUSIONS AND SIGNIFICANCE: Proxy measures of fitness provided no evidence that salmon farms function as ecological traps for wild fish. We suggest fish farms may act as population sources for wild fish, provided they are protected from fishing while resident at farms to allow their increased condition to manifest as greater reproductive output.

Two south-west Mediterranean fish farms were monitored over a period of 22 months to test if sea-cage fish farms act as settlement habitats for juvenile fish. Twenty juvenile fish species were found to settle at farms throughout the year. Fish assemblage composition varied markedly over time and was dependent on the spawning period for each species. The most abundant species were Obladamelanura, Atherina sp., Diplodussargus, Boopsboops and Lizaaurata. Up to 3783+/-1730 individuals/cage were found closely associated with the cages. Highest densities were observed during the warmer summer and autumn months. Zooplankton sampling and stomach content analyses of the most abundant species were done to assess prey availability, selectivity and diet overlap among species. Copepods were the main prey item for all juvenile fish species, irrespective of fish size. Ivlev's Index indicated that food was not a limiting factor for juvenile fish at farms. Furthermore, food pellets from the farm affected the food chain by modifying the fatty acid profiles of farm-associated zooplankton and juveniles of L. aurata and O. melanura. These results show that aquaculture can directly influence the body composition of juvenile fish that recruit to sea-cage fish farms.
