RESUMO

The re-examination of nematodes collected decades ago from a spiny rat, Proechimys roberti (Echimyidae) from Pará State, revealed the presence of Acanthostrongylus acanthostrongylus Travassos, 1937, and two coparasitic species of Pudica (Heligmonellidae: Pudicinae), one of them new to science. Pudica minima n. sp. mostly resembles Pudica tenua Durette-Desset, 1970, described from Proechimys semispinosus in Colombia. Nevertheless, it differs from P. tenua by having a synlophe with 12 ridges with poorly developed careen, bursal pattern of type 2-2-1 on the right lobe, bursal rays 2 and 3 similar in length, and spicules longer with spoon-shaped tips. Pudica evandroi (Travassos, 1937) was already described from Pr. roberti but its synlophe remained undescribed and the original description lacked some details. A detailed study of the synlophe is provided and the range of morphometric data for males and females is enlarged. The study of the synlophe allowed ratifying the placement of this species within Pudica, as early suggested by previous authors. The species list and identification key to species of Pudica are updated with the inclusion of three species. The status of Heligmosomum alpha Travassos, 1918, which was transferred to Pudica in 1990, is emended and the species is considered a Nippostrongylinae incertae sedis.

Hassalstrongylus dollfusi (Díaz-Ungría, 1963) Durette-Desset, 1971 was described in a wild house mouse, Mus musculus, from Venezuela and, since then, has never been reported again in the type host or in any other host. In this work, specimens assignable to H. dollfusi were found at 10 localities in Northeast Argentina, in five species of sigmodontine rodents. The nematodes were attributed to H. dollfusi based on diagnostic characters such as: synlophe with 22-31 subequal ridges; in males, hypertrophy of right ray 4 of the male bursa, thickening of the dorsal ray and bases of rays 8, distal tip of the spicules bent and spoon shaped; and, in females, presence of subventral postvulvar alae supported by hypertrophied struts. The new host recorded are: Oligoryzomys fornesi, O. flavescens, O. nigripes, Holochilus chacarius and Akodon azarae. The parasite showed a strong preference for host species of Oligoryzomys, which appear to act as primary hosts. The parasite could be present, parasitizing different species of Oligoryzomys, in a geographic area from the type locality in Venezuela southward to north Corrientes in Argentina. It has not been reported from populations of Oligoryzomys spp. of the Argentinean and Brazilian Atlantic Forest, nor south of 28° S, which may be explained by constraints in the environmental conditions required by the free-living stages of the parasite. This study provides the first identification and redescription of H. dollfusi in southern South America, from autochthonous hosts, six decades after its description.

ABSTRACT: Hassalstrongylus dollfusi (Díaz-Ungría, 1963) Durette-Desset, 1971 a été décrit chez une souris grise sauvage, Mus musculus, au Venezuela et, depuis lors, n'a plus jamais été signalé chez l'hôte-type ni chez aucun autre hôte. Dans ce travail, des spécimens attribuables à H. dollfusi ont été trouvés dans dix localités du nord-est de l'Argentine, chez cinq espèces de rongeurs Sigmodontinae. Les nématodes ont été attribués à H. dollfusi sur la base de caractères diagnostiques tels que : synlophe avec 22­31 crêtes subégales; chez le mâle, hypertrophie de la côte 4 droite de la bourse, épaississement de la côte dorsale et des bases des côtes 8, extrémité distale des spicules recourbée et en forme de cuillère; et, chez les femelles, présence d'ailes postvulvaires subventrales à fort support cuticulaire. Les nouveaux hôtes signalés sont : Oligoryzomys fornesi, O. flavescens, O. nigripes, Holochilus chacarius et Akodon azarae. Le parasite a montré une forte préférence pour les espèces hôtes du genre Oligoryzomys, qui semblent agir comme hôtes primaires. Le parasite pourrait être présent, parasitant différentes espèces d'Oligoryzomys, dans une zone géographique allant de la localité-type au Vénézuéla vers le sud jusqu'au nord de Corrientes en Argentine. Il n'a pas été signalé dans les populations d'Oligoryzomys spp. de la Forêt Atlantique argentine et brésilienne, ni au sud de 28° S, ce qui peut s'expliquer par des contraintes dans les conditions environnementales requises par les stades libres du parasite. Cette étude fournit la première identification et redescription d'H. dollfusi dans le sud de l'Amérique du Sud, à partir d'hôtes autochtones, six décennies après sa description.

In recent decades, packaged fruit has gained market ground over loose fruit, and fruit containers have become a source of information for consumers. This study approaches three objectives related to consumer information needs for decision making when purchasing fruit: (1) Determine if consumers' choice of packaged fruit rather than loose fruit is motivated by their interest in the information provided on packaging; (2) identify information gaps on fruit packaging labels; (3) identify those sensory attributes that consumers consider to be of major interest to be included in sensory labels of different fruit types. The study was based on an online questionnaire answered by 394 fruit consumers. Ninety percent of the participants stated having an interest in receiving information when purchasing fruit, but their choice between packaged or loose fruit was not conditioned by their information needs. Moreover, a gap between information interest and information use was detected as their final purchase decisions were not always based on the provided information. 'Harvest date', 'production method', 'percentage of the price received by the farmer', 'applied treatments', 'sensory characteristics', and 'environmental information' were identified as the major information gaps, as these labels were unavailable for a high percentage of consumers, who stated their interest in them. According to consumers, sensory labels should include information about 'sweetness' and 'flavor intensity' irrespectively of fruit type. 'Sourness' and 'juiciness' attributes were particularly interesting for citrus, as 'sourness' and 'firmness' were for kiwi. Information about texture properties was required for pome and stone fruit. Other attributes, such as easiness to peel, were important only for citrus fruit.

The present study was conducted to characterise rabbit sperm proteins focusing on the influence of the genetic origin. Six samples were recovered during two months from five males from genotype A (New Zealand White origin) and five from genotype R (California origin). Sperm proteins were extracted and subjected to in-gel digestion nano LC-MS/MS and bioinformatics analysis. The resulting library included 487 identified proteins validated with ≥95% Confidence (unused Score ≥ 1.3). All the identified proteins belonged to Oryctolagus cuniculus taxonomy. These data are available via ProteomeXchange with identifier PXD007989. Only 7 proteins were specifically implicated in reproductive processes according to Gene Ontology annotation. Regarding the comparison of the sperm proteins abundance between genotypes, forty proteins were differentially expressed. Among them, 25 proteins were over-expressed in genotype A, while 15 proteins were over-expressed in genotype R. In conclusion, this study characterizes for the first time rabbit sperm proteins and provides evidence that genotype is related to a specific abundance of spermatozoa proteins.

The present study was conducted to characterise rabbit seminal plasma proteins (SP proteins) focusing on the influence of the genetic origin and seasonality. In addition, ß-NGF protein quantity in SP was determined. Semen samples were recovered from January to December 2014 using 6 males belonging to genotype A and six from genotype R. For each genotype, one pooled sample at the beginning, middle and end of each season was selected to develop the experiment. A total of 24 pools (3 for each season and genetic line) were analysed. SP proteins of the two experimental groups were recovered and subjected to in-solution digestion nano LC-MS/MS and bioinformatics analysis. The resulting library included 402 identified proteins validated with ≥95% Confidence (unused Score ≥ 1.3). These data are available via ProteomeXchange with identifier PXD006308. Only 6 proteins were specifically implicated in reproductive processes according to Gene Ontology annotation. Twenty-three proteins were differentially expressed between genotypes, 11 over-expressed in genotype A and 12 in genotype R. Regarding the effect of season on rabbit SP proteome, results showed that there is no clear pattern of protein variation throughout the year. Similar ß-NGF relative quantity was observed between seasons and genotypes. In conclusion, this study generates the largest library of SP proteins reported to date in rabbits and provides evidence that genotype is related to a specific abundance of SP proteins.

We report the finding of 2 species of Pudica (Nematoda: Heligmonellidae: Pudicinae) in 2 rodents endemic to Chile, the common degu Octodon degus (Octodontidae) and the Bennett's chinchilla rat Abrocoma bennettii (Abrocomidae). Pudica degusi ( Babero and Cattan, 1975 ) n. comb., originally described as a species of Longistriata (Heligmosomidae), was found in the common degu; through the study of its synlophe, the species is reassigned to the Heligmonellidae: Pudicinae and the genus Pudica, and it is revalidated through comparison with the remaining species of the genus. Pudica cattani n. sp. is described from both O. degus and A. bennettii. It is characterized by its large body size, bursal pattern of type 1-3-1 on right lobe, 1-3-1 tending to 1-4 on left lobe, synlophe with 11 ridges including a careen, dorsal ray of the bursa dividing proximally and bursal rays 9 and 10 relatively short. Pudica degusi n. comb. and Pudica cattani n. sp. were found in the same host species but not as coparasitic in the same individuals. The common degu is confirmed as the sole and primary host of Pudica degusi n. comb. It is unlikely that it is the primary host for Pudica cattani n. sp., whose host affinities are less clear mainly due to the scarcity of data. Pudica cattani n. sp. is the first helminth reported from the Bennett's chinchilla rat. Both findings enlarge the host range of the Pudicinae to the families Octodontidae and Abrocomidae, i.e., 9 out of the 11 extant families of caviomorphs, thereby establishing the presence of this nematode subfamily as typical parasites of the Neotropical Hystricognathi.

A new genus and species of Viannaiidae (Trichostrongylina, Heligmosomoidea), Ischilinema baldoi n. gen. et sp. is described parasitizing two species of tuco-tucos, Ctenomys bergi and Ctenomys rosendopascuali (Rodentia, Hystricomorpha, Ctenomyidae) from Córdoba province, Central Argentina. No helminths were previously known from these two host species. The new genus is defined by the following characters: synlophe with 15 continuous ridges subequal in size, presence of left cuticular dilatation, a gap between ridges 1' and 2', bursa asymmetrical with dorsal ray hypertrophied and displaced to the right, and spicules not twisted. This is the first record of Viannaiidae from the Ctenomyidae, enlarging the host range of these parasites to five out of the 12 extant families of caviomorphs.

Although uterine prolapse and carcinoma of the uterine cervix are not rare events, their association is very uncommon. The treatment of cervical cancer has been protocolled, but the management of uterovaginal prolapse associated with carcinoma of the cervix is not standardised and therapy strategies vary considerably among authors. Our case reports a 74-year-old patient, admitted to the emergency department with an ulcerated prolapsed uterus. Biopsy of the cervical lesion confirmed a squamous-cell carcinoma. The patient underwent vaginal hysterectomy plus open bilateral iliopelvic lymphadenectomy complemented with radiotherapy with quimiosensibilisation. With this aggressive treatment approach, there was progression of the disease. The authors believe that this case typiaddition to the few published reports.

The effect of dietary inclusion of distillers dried grains with solubles (DDGS) on carcass and meat quality of longissimus muscle was studied in 100 growing rabbits from 28 to 59days old. Diets with no DDGS (C), barley (Db20), wheat (Dw20) and corn (Dc20) DDGS at 20% and corn (Dc40) DDGS at 40% were formulated. No effects on most of the carcass traits, texture and water holding capacity were found. Barley and corn DDGS led to a higher dissectible fat percentage. Meat redness was higher with Dw20 and pH was higher with Dw20 and Db20 than with Dc20. Protein and saturated fatty acids concentration declined as corn DDGS level increased. Dc40 led to the lowest saturated/unsaturated fatty acid ratio, atherogenic index and thrombogenic index. In conclusion, dietary inclusion of these DDGS at 20% did not affect most of the carcass and meat quality traits in rabbits.

Picornavirus RNAs initiate translation using an internal ribosome entry site (IRES)-dependent mechanism. The IRES element of foot-and-mouth disease virus (FMDV) is organized in domains, being different from each other in RNA structure and RNA-protein interaction. Wild-type transcripts provided in trans rescue defective FMDV IRES mutants. Complementation, however, was partial since translation efficiency of the mutant RNAs was up to 10% of the wild type IRES. We report here that mutations diminishing the RNA-RNA interaction capacity induced a decrease in IRES rescue. On the other hand, IRES transcripts bearing mutations that reorganize the RNA structure of the apical region of central domain, although weakly, complement defective IRES that are unable to interact with the initiation factor eIF4G in a separate domain. Together, these results suggest that IRES rescue may involve RNA-mediated contacts between defective elements, each carrying a defect in a separate domain but having the complementing one with the appropriate structural orientation and/or ribonucleoprotein composition. Our observations further support the essential role of the central domain of the FMDV IRES during protein synthesis and demonstrate that there is a division of functions between the IRES domains.

A distinctive feature of positive-strand RNA viruses is the presence of high-order structural elements at the untranslated regions (UTR) of the genome that are essential for viral RNA replication. The RNA of all members of the family Picornaviridae initiate translation internally, via an internal ribosome entry site (IRES) element present in the 5' UTR. IRES elements consist of cis-acting RNA structures that usually require specific RNA-binding proteins for translational machinery recruitment. This specialized mechanism of translation initiation is shared with other viral RNAs, e.g. from hepatitis C virus and pestivirus, and represents an alternative to the cap-dependent mechanism. In cells infected with many picornaviruses, proteolysis or changes in phosphorylation of key host factors induces shut off of cellular protein synthesis. This event occurs simultaneously with the synthesis of viral gene products since IRES activity is resistant to the modifications of the host factors. Viral gene expression and RNA replication in positive-strand viruses is further stimulated by viral RNA circularization, involving direct RNA-RNA contacts between the 5' and 3' ends as well as RNA-binding protein bridges. In this review, we discuss novel insights into the mechanisms that control picornavirus gene expression and compare them to those operating in other positive-strand RNA viruses.

Translation initiation driven by internal ribosome entry site (IRES) elements is dependent on the structural organization of the IRES region. Picornavirus IRES are organized in structural domains, in which the terminal stem-loops participate in functional RNA-protein interactions. However, the mechanistic role performed by the central domain during internal initiation has not been elucidated yet. Here we show that the foot-and-mouth-disease virus IRES contains a structural motif that serves in vitro as substrate for the Synechocystis sp. RNase P ribozyme, a structure-dependent endonuclease that participates in tRNA precursor processing. Recognition of the IRES substrate was dose dependent, required high magnesium concentration, and resulted in the formation of cleavage products with 5' phosphate and 3' hydroxyl ends. Mapping of the core recognition motif indicated that it overlapped with the apical region of the central domain. Two IRES constructs containing nucleotide substitutions in the apical region of the central domain that reorganized RNA structure displayed an altered pattern of cleavage by the cyanobacterial ribozyme generating new cleavage events in nearby residues. From these data it is inferred that the central domain of the IRES region has evolved a tRNA structural mimicry that renders it a substrate for RNase P ribozyme reaction. Recognition of this motif was affected in defective IRES mutants with a local RNA structure reorganization, suggesting that its structural preservation is required for IRES activity.

Foot-and-mouth disease virus (FMDV) infection induces major changes in the host cell including the shutoff of cellular protein synthesis. Here, protein extracts from FMDV-infected cells have been used to monitor changes in the profile of RNA-binding factors interacting with regulatory regions of the viral RNA. Relevant differences have been detected in the pattern of interaction with proteins prepared from either infected or uninfected cells with RNA probes encompassing the internal ribosome entry site (IRES), the 5' and 3'end regions. The binding patterns obtained for two divergent FMDV isolates showed differences depending on the viral isolate used. The identity of the host proteins giving a shifted binding pattern to RNA regulatory regions has been inferred by immunoblotting. Our results show that polypyrimidine tract-binding protein (PTB) and two subunits of translation initiation factor eIF3 interacting with the IRES undergo proteolytic processing during FMDV infection. In addition, poly(A)-binding protein (PABP), interacting with the 3'end of the viral RNA is partially processed. Proteolysis of eIF3a, eIF3b, PABP and PTB correlated with the extent of cytopathic effect induced by FMDV in infected cells.

The untranslated regions (UTRs) of the foot-and-mouth disease virus (FMDV) genome contain multiple functional elements. In the 5' UTR, the internal ribosome entry site (IRES) element governs cap-independent translation initiation, whereas the S region is presumably involved in RNA replication. The 3' UTR, composed of two stem-loops and a poly(A) tract, is required for viral infectivity and stimulates IRES activity. Here, it was found that the 3' end established two distinct strand-specific, long-range RNA-RNA interactions, one with the S region and another with the IRES element. These interactions were not observed with the 3' UTR of a different picornavirus. Several results indicated that different 3' UTR motifs participated in IRES or S region interactions. Firstly, a high-order structure adopted by both the entire IRES and the 3' UTR was essential for RNA interaction. In contrast, the S region interacted with each of the stem-loops. Secondly, S-3' UTR interaction but not IRES-3' UTR interaction was dependent on a poly(A)-dependent conformation. However, no other complexes were observed in mixtures containing the three transcripts, suggesting that these regions did not interact simultaneously with the 3' UTR probe. Cellular proteins have been found to bind the S region and one of these also binds to the 3' UTR in a competitive manner. Our data suggest that 5'-3'-end bridging through both direct RNA-RNA contacts and RNA-protein interactions may play an essential role in the FMDV replication cycle.
