Dolphins have become famous for their ability to perform a wide variety of athletic and acrobatic behaviors including high-speed swimming, maneuverability, porpoising and tail stands. Tail stands are a behavior where part of the body is held vertically above the water's surface, achieved through thrust produced by horizontal tail fluke oscillations. Strong, efficient propulsors are needed to generate the force required to support the dolphin's body weight, exhibiting chordwise and spanwise flexibility throughout the stroke cycle. To determine how thrust production, fluke flexibility and tail stroke kinematics vary with effort, six adult bottlenose dolphins (Tursiops truncatus) were tested at three different levels based on the position of the center of mass (COM) relative to the water's surface: low (COM below surface), medium (COM at surface) and high (COM above surface) effort. Additionally, fluke flexibility was measured as a flex index (FI=chord length/camber length) at four points in the stroke cycle: center stroke up (CU), extreme top of stroke (ET), center stroke down (CD) and extreme bottom of stroke (EB). Video recordings were analyzed to determine the weight supported above the water (thrust production), peak-to-peak amplitude, stroke frequency and FI. Force production increased with low, medium and high efforts, respectively. Stroke frequency also increased with increased effort. Amplitude remained constant with a mean 33.8% of body length. Significant differences were seen in the FI during the stroke cycle. Changes in FI and stroke frequency allowed for increased force production with effort, and the peak-to-peak amplitude was higher compared with that for horizontal swimming.

Direct reading instruments (DRIs) for aerosols have been used in industrial hygiene practice for many years, but their potential has not been fully realized by many occupational health and safety professionals. Although some DRIs quantify other metrics, this article will primarily focus on DRIs that measure aerosol number, size, or mass. This review addresses three applications of aerosol DRIs that occupational health and safety professionals can use to discern, characterize, and document exposure conditions and resolve aerosol-related problems in the workplace. The most common application of aerosol DRIs is the evaluation of engineering controls. Examples are provided for many types of workplaces and situations including construction, agriculture, mining, conventional manufacturing, advanced manufacturing (nanoparticle technology and additive manufacturing), and non-industrial sites. Aerosol DRIs can help identify the effectiveness of existing controls and, as needed, develop new strategies to reduce potential aerosol exposures. Aerosol concentration mapping (ACM) using DRI data can focus attention on emission sources in the workplace spatially illustrate the effectiveness of controls and constructively convey concerns to management and workers. Examples and good practices of ACM are included. Video Exposure Monitoring (VEM) is another useful technique in which video photography is synced with the concentration output of an aerosol DRI. This combination allows the occupational health and safety professional to see what tasks, environmental situations, and/or worker actions contribute to aerosol concentration and potential exposure. VEM can help identify factors responsible for temporal variations in concentration. VEM can assist with training, engage workers, convince managers about necessary remedial actions, and provide for continuous improvement of the workplace environment. Although using DRIs for control evaluation, ACM and VEM can be time-consuming, the resulting information can provide useful data to prompt needed action by employers and employees. Other barriers to adoption include privacy and security issues in some worksites. This review seeks to provide information so occupational health and safety professionals can better understand and effectively use these powerful applications of aerosol DRIs.

With advances in technology, there are an increasing number of direct-reading instruments available to occupational health and safety professionals to evaluate occupational aerosol exposures. Despite the wide array of direct-reading instruments available to professionals, the adoption of direct-reading technology to monitor workplace exposures has been limited, partly due to a lack of knowledge on how the instruments operate, how to select an appropriate instrument, and challenges in data analysis techniques. This paper presents a review of direct-reading aerosol instruments available to occupational health and safety professionals, describes the principles of operation, guides instrument selection based on the workplace and exposure, and discusses data analysis techniques to overcome these barriers to adoption. This paper does not cover all direct-reading instruments for aerosols but only those that an occupational health and safety professional could use in a workplace to evaluate exposures. Therefore, this paper focuses on instruments that have the most potential for workplace use due to their robustness, past workplace use, and price with regard to return on investment. The instruments covered in this paper include those that measure aerosol number concentration, mass concentration, and aerosol size distributions.

Two prototype cyclones were the subjects of a comparative research campaign with a diesel particulate matter sampler (DPMS) that consists of a respirable cyclone combined with a downstream impactor. The DPMS is currently used in mining environments to separate dust from the diesel particulate matter and to avoid interferences in the analysis of integrated samples and direct-reading monitoring in occupational environments. The sampling characteristics of all three devices were compared using ammonium fluorescein, diesel, and coal dust aerosols. With solid spherical test aerosols at low particle loadings, the aerodynamic size-selection characteristics of all three devices were found to be similar, with 50% penetration efficiencies (d 50) close to the design value of 0.8 µm, as required by the US Mine Safety and Health Administration for monitoring occupational exposure to diesel particulate matter in US mining operations. The prototype cyclones were shown to have 'sharp cut' size-selection characteristics that equaled or exceeded the sharpness of the DPMS. The penetration of diesel aerosols was optimal for all three samplers, while the results of the tests with coal dust induced the exclusion of one of the prototypes from subsequent testing. The sampling characteristics of the remaining prototype sharp cut cyclone (SCC) and the DPMS were tested with different loading of coal dust. While the characteristics of the SCC remained constant, the deposited respirable coal dust particles altered the size-selection performance of the currently used sampler. This study demonstrates that the SCC performed better overall than the DPMS.

Dogs have been bred for different sizes and functions, which can affect their locomotor biomechanics. As quadrupeds, dogs must distribute their mass between fore and hind legs when standing. The mass distribution in dogs was studied to determine if the proportion of supported mass on each limb couplet is dependent on body size. A total of 552 dogs from 123 breeds ranging in size from Chihuahua to Mastiff were examined. Each dog was weighed on a digital scale while standing, alternating foreleg, and hind leg support. The overall "grand" mean proportion of mass on the forelegs to the total mass was 60.4% (range: 47.6-74.4%). The data set indicated no significant change in the ratio with total mass but there was a significant difference by sex. When separated into American Kennel Club categories, no group was notably different from the grand mean or from each other, but when sex was also considered, there was a significant difference that was not specifically discerned by post hoc analysis. The mean for female Hounds was notably below the grand mean. For clades based on genetics, the mean for European origin mastiffs was notably greater than the grand mean and significantly different from UK origin herders and coursers. The mass of the head, chest, and musculature for propulsion could explain the mass support differential. Mass distribution and terrestrial locomotion in dogs shows substantial variation among breeds.

Esophageal squamous cell carcinoma (ESCC) is an aggressive cancer with late-stage detection and poor prognosis. This emphasizes the need to identify new markers for early diagnosis and treatment. Altered mitochondrial genome (mtDNA) content in primary tumors correlates with poor patient prognosis. Here we used three-dimensional (3D) organoids of esophageal epithelial cells (EECs) from the MPV17-/- mouse model of mtDNA depletion to investigate the contribution of reduced mtDNA content in ESCC oncogenicity. To test if mtDNA defects are a contributing factor in ESCC, we used oncogenic stimuli such as ESCC carcinogen 4-nitroquinoline oxide (4-NQO) treatment, or expressing p53R175H oncogenic driver mutation. We observed that EECs and 3D-organoids with mtDNA depletion had cellular, morphological and genetic alterations typical of an oncogenic transition. Furthermore, mitochondrial dysfunction induced cellular transformation is accompanied by elevated mitochondrial fission protein, DRP1 and pharmacologic inhibition of mitochondrial fission by mDivi-1 in the MPV17-/- organoids reversed the phenotype to that of normal EEC organoids. Our studies show that mtDNA copy number depletion, activates a mitochondrial retrograde response, potentiates telomere defects, and increases the oncogenic susceptibility towards ESCC. Furthermore, mtDNA depletion driven cellular plasticity is mediated via altered mitochondrial fission-fusion dynamics.

Osteosarcoma (OSA) is an aggressive mesenchymal tumor of the bone that affects children and occurs spontaneously in dogs. Human and canine OSA share similar clinical, biological and genetic features, which make dogs an excellent comparative model to investigate the etiology and pathogenesis of OSA. Mitochondrial (mt) defects have been reported in many different cancers including OSA, although it is not known whether these defects contribute to OSA progression and metastasis. Taking a comparative approach using canine OSA cell lines and tumor tissues we investigated the effects of mtDNA content and dysfunction on OSA biology. OSA tumor tissues had low mtDNA contents compared to the matched non-tumor tissues. We observed mitochondrial heterogeneity among the OSA cell lines and the most invasive cells expressing increased levels of OSA metastasis genes contained the highest amount of mitochondrial defects (reduced mtDNA copies, mt respiration, and expression of electron transport chain proteins). While mitochondria maintain a filamentous network in healthy cells, the mitochondrial morphology in OSA cells were mostly "donut shaped", typical of "stressed" mitochondria. Moreover the expression levels of mitochondrial retrograde signaling proteins Akt1, IGF1R, hnRNPA2 and NFkB correlated with the invasiveness of the OSA cells. Furthermore, we demonstrate the causal role of mitochondrial defects in inducing the invasive phenotype by Ethidium Bromide induced-mtDNA depletion in OSA cells. Our data suggest that defects in mitochondrial genome and function are prevalent in OSA and that lower mtDNA content is associated with higher tumor cell invasiveness. We propose that mt defects in OSA might serve as a prognostic biomarker and a target for therapeutic intervention in OSA patients.

All known orthopoxviruses, including ectromelia virus (ECTV), contain a gene in the E3L family. The protein product of this gene, E3, is a double-stranded RNA-binding protein. It can impact host range and is used by orthopoxviruses to combat cellular defense pathways, such as PKR and RNase L. In this work, we constructed an ECTV mutant with a targeted disruption of the E3L open reading frame (ECTVΔE3L). Infection with this virus resulted in an abortive replication cycle in all cell lines tested. We detected limited transcription of late genes but no significant translation of these mRNAs. Notably, the replication defects of ECTVΔE3L were rescued in human and mouse cells lacking PKR. ECTVΔE3L was nonpathogenic in BALB/c mice, a strain susceptible to lethal mousepox disease. However, infection with ECTVΔE3L induced protective immunity upon subsequent challenge with wild-type virus. In summary, E3L is an essential gene for ECTV.

A four-stage personal diffusion battery (pDB) was designed and constructed to measure submicron particle size distributions. The pDB consisted of a screen-type diffusion battery, solenoid valve system, and electronic controller. A data inversion spreadsheet was created to solve for the number median diameter (NMD), geometric standard deviation (GSD), and particle number concentration of unimodal aerosols using stage number concentrations from the pDB combined with a handheld condensation particle counter (pDB+CPC). The inversion spreadsheet included particle entry losses, theoretical penetrations across screens, the detection efficiency of the CPC, and constraints so the spreadsheet solved to values within the pDB range. Size distribution parameters (NMD, GSD, and number concentration) measured with the pDB+CPC with inversion spreadsheet were within 25% of those measured with a scanning mobility particle sizer (SMPS) for 5 of 12 polydisperse combustion aerosols. For three tests conducted with propylene torch exhaust, the pDB+CPC with inversion spreadsheet successfully identified that the NMD was smaller than the constraint value of 16 nm. The ratio of the nanoparticle portion of the aerosol compared to the reference (Rnano) was calculated to determine the ability of pDB+CPC with inversion spreadsheet to measure the nanoparticle portion of the aerosols. The Rnano ranged from 0.87 to 1.01 when the inversion solved and from 0.06 to 2.01 when the inversion solved to a constraint. The pDB combined with CPC has limited use as a personal monitor but combining the pDB with a different detector would allow for the pDB to be used as a personal monitor.

Three studies were performed to assess the effectiveness of various techniques to control metalworking fluid (MWF) mist. The studies consisted of a detailed main study that determined the effect of degree of enclosure on personal exposures and area concentrations of MWF mist on two machining transfer lines. One ancillary study was conducted to determine the effect of shutting off MWF delivery during down time; the second ancillary study investigated the effectiveness of improved retrofitted enclosure. In the main study, the two operations were identical except for degree of enclosure. Personal and area sampling results for the new line were about half those found in the old line. Measurements at the new operation exhibited significantly less variability. Personal exposures and area concentrations were significantly less at the new operation than at the older, less enclosed operation, demonstrating that the total enclosure in the new operation provides better and more consistent control of the mist. The first ancillary study was conducted to determine if shutting off MWF delivery to the parts being machined reduced area MWF mist concentration during downtime at a partially enclosed transfer machining line. A significant reduction in concentration of 80% was measured with machining off/MWF off. Mist concentrations measured with machining off/MWF on were not significantly different from mist levels measured during machining on/MWF on. The second ancillary study investigated the reduction of mist concentration achieved through improved enclosure of an existing set of machines. Area mist concentrations were measured at a machining operation before and after the installation of an improved enclosure. Area mist concentrations were reduced by 87% with the improved enclosure.
