OBJECTIVES: While inorganic mercury is being gradually withdrawn from industry, environmental exposure to mercury is recognized as one of the greatest present toxicological problems. The aim of this study was to evaluate the effect of polyunsaturated fatty acids (PUFAs) supplementation on selected cardiovascular risk factors and the urinary mercury (Hg-U) concentration in workers occupationally exposed to mercury vapor. MATERIAL AND METHODS: Overall, 38 workers of an electrolyzer hall (Hg-U: 46.6±35.7 µg/g creatinine) and a control group of 60 employees not exposed to Hg (Hg-U: 4.3±15.5 µg/g creatinine) were included in a clinical cross-over study. Clinical and laboratory tests were carried out 4 times: before and after a 3-month period of PUFAs supplementation (1000 mg daily), then after a 3-month break, and then after another 3-month period of PUFAs supplementation. RESULTS: The baseline heart rate (HR) and serum triglyceride levels were higher in the Hg-exposed workers than in the controls, whereas systolic blood pressure (SBP) and cholesterol (C) levels exceeded normal values in both groups. There was a positive correlation between high-density lipoprotein 3 cholesterol (HDL3-C) and Hg-U levels. The PUFAs use was associated with a decrease in both HR and SBP. After the first stage of supplementation, a decrease in the Hg-U concentration was observed. In a multivariate logistic regression model, decreases in Hg-U were associated only with exposure to mercury; ORΔHg = 0.562 (95% CI: 0.323-0.979), p < 0.042. After the second 3-month period of PUFAs supplementation, a significant association between HDL3-C and a Hg-U decrease was shown: ORHDL3 = 1.222 (95% CI: 1.01-1.46), p < 0.033. CONCLUSIONS: In the workers exposed to mercury vapor, PUFAs supplementation led to some beneficial effects on HR and SBP. The first stage of supplementation was associated with a decrease in Hg-U in which HDL3 metabolism probably plays an important role. Int J Occup Med Environ Health. 2021;34(4):551-64.

The purpose of this work was to assess the influence of selected CNR1, MC4R, LEP, FTO and VDR FOKI gene polymorphisms on blood and urine concentration markers of lead, cadmium and arsenic in a population directly exposed to these metals. Eighty-five people exposed to lead, arsenic and cadmium were qualified to take part in the study. Standard urine samples and 25mL of venous blood from each worker were collected to assay basic laboratory and toxicological markers as well as selected single nucleotide polymorphisms (SNPs) within CNR1-cannabinoid receptor 1 gene (rs806368, rs806381, rs1049353, rs12720071), MC4R-melanocortin 4 receptor gene (rs17782313), LEP-leptin promoter gene (rs7799039), FTO-alpha-ketoglutarate-dependent dioxygenase gene (rs9939609) and VDR-vitamin D receptor (rs10735810) genes. It appeared that, except for the MC4R SNP, all the other polymorphisms were found to be associated with various laboratory parameters. Arsenic concentration in urine was associated with all four CNR1 and LEP SNPs, while cadmium concentration in blood was affected by the VDR polymorphism. Moreover, some significant relationships were also observed between CNR1 rs1049353 and FTO rs9939609 gene variants and markers of lead exposure. These results imply SNPs within genes coding for proteins involved in development of metabolic syndrome may be of prognostic value for persons directly exposed to lead, cadmium and arsenic.

BACKGROUND: Exposure to arsenic, lead, and cadmium poses the risk of cancer. Cathepsins (CP) and their natural inhibitors (CPI) in blood serum are markers of carcinogensis. The aim of this work was to verify the existence of relationships between the exposure to the airesaid elements and the activity of CP and CPL. MATERIALS AND METHODS: The levels of Pb and Cd in blood, Mn, Cu, Zn, Ca, Mg, Fe, Se CP as well as free and bound CPI in serum, FEP in erythrocytes, and As in urine were measured in 186 production workers of Legnica Copper Smelter (study group) and 56 administrative employees (control group) and then analyzed. RESULTS: CF levels were significantly elevated in both groups and CPI levels were elevated in the study group vs. the control group. There were no statistically significant relationships between CF and CPI levels and the remaining parameters in the study group. whereas they were found for CPI in the control group. The results indicate that occupational and environmental exposures to toxic metals (arsenic, lead, cadmium) may lead to the elevated levels of CF and CPI. CONCLUSIONS: The obtained results support the assumptions of the study.

Some dietary factors could inhibit lead toxicity. The aim of this study was to evaluate the effect of dietary compounds rich in unsaturated fatty acids (FA) on blood lead level, lipid metabolism, and vascular reactivity in rats. Serum metallothionein and organs' lead level were evaluated with the aim of assessing the possible mechanism of unsaturated FA impact on blood lead level. For three months, male Wistar rats that were receiving drinking water with (100 ppm Pb) or without lead acetate were supplemented per os daily with virgin olive oil or linseed oil (0.2 mL/kg b.w.) or egg derived lecithin fraction: "super lecithin" (50 g/kg b.w.). Mesenteric artery was stimulated ex vivo by norepinephrine (NE) administered at six different doses. Lecithin supplementation slightly reduced pressor responses of artery to NE. Lead administered to rats attenuated the beneficial effect of unsaturated FA on lipid metabolism and vascular reactivity to adrenergic stimulation. On the other hand, the super lecithin and linseed oil that were characterized by low omega-6 to omega-3 ratio (about 1) reduced the blood lead concentration. This effect was observed in lead poisoned rats (p < 0.0001) and also in rats nonpoisoned with lead (p < 0.05).

UNLABELLED: The aim of this study was to find a relationship between polymorphisms of ALAD rs1805313, rs222808, rs1139488, VDR FokI and HFE C282Y and H63D and basic toxicological parameters (lead and ZnPP blood concentration) in people occupationally exposed to lead. We collected data of 101 workers (age 25-63 years) directly exposed to lead. The toxicological lab tests included blood lead, cadmium and ZnPP concentration measurement and arsenic urine concentration measurement. Workers were genotyped for ALAD (rs1805313, rs222808, rs1139488), HFE (C282Y, H63D) and VDR (FokI). Individuals with the lead exposure and coexisting F allel in the locus Fok-I of VDR gene are suspected of higher zinc protoporphyrins concentrations. Workers exposed to the lead with the Y allel in the locus C282Y of the HFE gene are predisposed to lower ZnPP levels and individuals with coexisting H allel in the locus H63D HFE gene are predisposed to lower Pb-B levels. The T allel in the locus rs1805313 of the ALAD gene determines lower Pb-B and ZnPP levels in lead-exposed individuals. The heterozigosity of the locus rs2228083 of the ALAD gene has a strong predilection to higher Pb-B levels. The carriage of the C allel in the locus rs1139488 of the ALAD gene might determine higher Pb-B levels and the heterozigosity of the locus rs1139488 of the ALAD gene might result in higher ZnPP levels. CONCLUSION: The study revealed relationship between VDR, HFE and ALAD genes polymorphism and basic toxicological parameters in occupationally exposed workers.
