Galectins (Gal) are a family of glycan-binding proteins characterized by their affinity for ß-galactosides. Galectin-1 (Gal-1), a dimeric lectin with two galactoside-binding sites, regulates cancer progression and immune responses. Coordination chemistry has been engaged to develop versatile multivalent neoglycoconjugates for binding Gal-1. In this study we report a fast and original method to synthesize hybrid gold nanoparticles in which a hydrochloride lactose-modified chitosan, named CTL, is mixed with dicarboxylic acid-terminated polyethylene glycol (PEG), leading to shell-like hybrid polymer-sugar-metal nanoparticles (CTL-PEG-AuNPs). The aim of this paper is to preliminarily study the interaction of the CTL-PEG-AuNPs with a target protein, namely, Gal-1, under specific conditions. The molecular interaction has been measured by Transmission Electron Microscopy (TEM), UV-vis, and Raman Spectroscopy on a large range of Gal-1 concentrations (from 0 to 10-12 M). We observed that the interaction was strongly dependent on the Gal-1 concentration at the surface of the gold nanoparticles.

We investigate the characteristics, fate and photocatalytic activity of spheroid- and rod-shaped TiO2 nano-crystals in aqueous solutions to better understand their behaviour in media of biological and environmental interest. For this purpose, the potential of a solvothermal method in synthesizing highly crystalline nanoparticles and tuning their sizes/shapes is explored. Spheroid- and rod-shaped nanoparticles are successfully obtained with different aspect ratios, while keeping their structures as well as their cross-sectional areas identical. The aggregation/agglomeration of these nanostructures in aqueous solutions shows an obvious shape effect, revealing critical coagulation concentrations (CCCs) significantly lower for the rods compared to the spheroids (aspect ratio â¼ 2-3). This trend is observed in both NaCl and CaCl2 electrolytes at pH values above and below the pHPZC of TiO2 nanoparticles. The photocatalytic activity of the spheroids is unexpectedly superior to that of the rods at NaCl and CaCl2 concentrations over a range of 2 to 100 and 1 to 50 mM, respectively. Our results show that an increase in the chloride concentration leads to an inhibition of the photocatalytic activity rate, with a more pronounced impact for the rods. In contrast, the size of aggregates/agglomerates has only a little effect on the photocatalytic properties of both nano-crystals.

HydrophobinVmh2 is a small amphiphilic protein, which self-assembles on different surfaces and naturally interacts with glucose. Here, we report on the synthesis of a nanobiocomplex made of polyethylene glycol, Vmh2 and gold nanoparticles by a one-step process and on its ability to recognise glucose in an aqueous solution at 0.3-0.6-1.2 mg ml(-1) concentrations. Even though the Vmh2 proteins are intrinsically bonded to the gold core, effective glucose interaction monitoring was demonstrated by using dynamic light scattering, ultraviolet-visible, polarization-modulated infrared reflection-absorption and x-ray photoelectron spectroscopies. Experimental results highlighted an affinity constant of 7.3 ± 0.3 mg ml(-1) between the nanobiosystem and the sugar, and a detection sensitivity of 0.13 ± 0.06 a.u./mg ml(-1).

Phytochelatins are small peptides that can be found in several organisms, which use these oligopeptides to handle heavy metal elements. Here, we report a method for monitoring interactions between lead(ii) ions in aqueous solutions and phytochelatin 6 oligopeptide bioconjugated onto pegylated gold nanorods (PEG-AuNrs). This study is the first step towards a high sensitive label free optical biosensor to quantify heavy metal pollution in water.

The thermophilic bacterium Thermus thermophilus HB27 encodes chromosomal arsenate reductase (TtArsC), the enzyme responsible for resistance to the harmful effects of arsenic. We report on adsorption of TtArsC onto gold nanoparticles for naked-eye monitoring of biomolecular interaction between the enzyme and arsenic species. Synthesis of hybrid biological-metallic nanoparticles has been characterized by transmission electron microscopy (TEM), ultraviolet-visible (UV-vis), dynamic light scattering (DLS) and phase modulated infrared reflection absorption (PM-IRRAS) spectroscopies. Molecular interactions have been monitored by UV-vis and Fourier transform-surface plasmon resonance (FT-SPR). Due to the nanoparticles' aggregation on exposure to metal salts, pentavalent and trivalent arsenic solutions can be clearly distinguished by naked-eye assay, even at 85 µM concentration. Moreover, the assay shows partial selectivity against other heavy metals.

Strategies employed to interface biomolecules with nanomaterials have considerably advanced in recent years and found practical applications in many different research fields. The construction of nucleic acid modified interfaces together with the label-free detection of hybridization events has been one of the major research focuses in science and technology. In this paper, we demonstrate the high interest of graphene-on-metal surface plasmon resonance (SPR) interfaces for the detection of DNA hybridization events in the attomolar concentration range. The strategy consists on the noncovalent functionalization of graphene-coated SPR interfaces with gold nanostars carrying single-stranded DNA (ssDNA). Upon hybridization with its complementary DNA, desorption of the nanostructures takes place and thus enables the sensitive detection of the DNA hybridization event. The DNA sensor exhibits a detection limit of ≈500 aM for complementary DNA with a linear dynamic range up to 10(-8) M. This label-free DNA detection platform should spur off new interest toward the use of commercially available graphene-coated SPR interfaces.

We show in this article that doxorubicin-modified gold nanoparticles (Au NP-DOX) can be used for the post-amplification of the wavelength shift of localized surface plasmon resonance (LSPR) signals after DNA hybridization events. We take advantage of the intercalation properties of DOX with guanine-rich oligonucleotides and the plasmon coupling between surface-linked gold nanostructures and Au NP-DOX in solution to detect in a sensitive manner DNA hybridisation events. Post-treatment of double-stranded DNA with Au NP-DOX resulted in a detection limit of ≈600 pM, several times lower than that without post-incubation (LOD ≈ 40 nM).

A one-step method is reported to synthesize hybrid gold nanoparticles (AuNPs) by reduction of HAuCl4 in acetic solution in the presence of collagen (Col), dicarboxylic acid-terminated polyethylene glycol (PEG), and cetyltetrammonium bromide (CTAB) mixed with hydoxyapatite (HAP) as surfactants. Such formation process of AuNPs was shown to be responsible for purple stains naturally formed on Egyptianizing archaeological gilded ivories from 8th BC Syria. The understanding of this formation mechanism, which most likely involves a step with hybrid AuNPs, allows the establishing of an authenticity marker of ancient gold-plated ivories.

Introduction: The aim of this study is focused on the development of theranostic hybrid nanovectors based on gold-doxorubicin (DOX)-gemcitabine (GEM) complexes and their active targeting with Galectin-1 (Gal-1) as a promising therapeutic and prognostic marker in cancer. Methods: For this purpose, a gold salt (HAuCl4) interacts with antitumor drugs (DOX; GEM) by chelation and then stabilizes with dicarboxylic acid-terminated polyethylene glycol (PEG) as a biocompatible surfactant. The proposed methodology is fast and reproducible, and leads to the formation of a hybrid nanovector named GEM@DOX IN PEG-AuNPs, in which the chemo-biological stability was improved. All synthetic chemical products were evaluated using various spectroscopic techniques (Raman and UV-Vis spectroscopy) and transmission electron microscopy (TEM). Results: To conceive a therapeutic application, our hybrid nanovector (GEM@DOX IN PEG-AuNPs) was conjugated with the Galectin-1 protein (Gal-1) at different concentrations to predict and specifically recognize cancer cells. Gal-1 interacts with GEM@DOX in PEG-AuNPs, as shown by SPR and Raman measurements. We observed both dynamic variation in the plasmon position (SPR) and Raman band with Gal-1 concentration. Discussion: We identified that GEM grafted electrostatically onto DOX IN PEG-AuNPs assumes a better chemical conformation, in which the amino group (NH3+) reacts with the carboxylic (COO-) group of PEG diacide, whereas the ciclopenthanol group at position C-5' reacts with NH3+ of DOX. Conclusion: This study opens further way in order to built "smart nanomedical devices" that could have a dual application as therapeutic and diagnostic in the field of nanomedicine and preclinical studies associated.

In this article, the detection of DNA hybridization taking advantage of the plasmonic properties of gold nanostructures is described. The approach is based on the amplification of the wavelength shift of a multilayered localized surface plasmon resonance (LSPR) sensor interface upon hybridization with gold nanorods and nanostars-labeled DNA. The amplification results in a significant decrease of the limit of detection from ≈40 nM as observed for unlabeled DNA to 0.2 nM for labeled DNA molecules. Furthermore, the plasmonic band, characteristic of the labeled DNA, is different from that of the LSPR interface. Indeed, next to the plasmon band at around 550 nm, being in resonance with the plasmon band of the LSPR interface, additional plasmonic peaks at 439 nm for gold nanostar-labeled DNA and 797 nm for gold nanorod-labeled DNA are observed, which were used as plasmonic signatures for successful hybridization.

The mechanism of interaction of Zn porphyrin (ZnPP) with TiO2 surfaces is investigated with a view to optimizing the synthesis of hybrid nanomaterials. The strategy consists of studying the adsorption of ZnPP on TiO2 flat surfaces by taking advantage of complementary surface characterization techniques. Combining a detailed X-ray photoelectron spectroscopic analysis with AFM imaging allows ZnPP-surface and ZnPP intermolecular interactions to be discriminated. Probing the adsorption of ZnPP on TiO2 nanoparticles (NPs) reveals the dominant role of ZnPP-mediated interactions, which are associated with the formation of ZnPP multilayers and/or with the state of aggregation of NPs. These preliminary investigations provide a guideline to synthesizing a novel ZnPP-TiO2 hybrid nanomaterial in a one-step protocol. In this material, ZnPP molecules are presumably involved in the TiO2 lattice rather than on the NP surface. Furthermore, ZnPP molecules preserve their electronic properties within the TiO2 NPs, and this makes the ZnPP-TiO2 hybrid nanomaterial an excellent candidate for nanomedicine and related applications, such as localization of nanoparticles in cells and tissues or in photodynamic therapy.

For the first, time a strong selectivity is evidenced in inorganic peptide synthesis. When an equimolar mixture of Ala and Arg monomers is added to the synthesis medium of TiO2 nanoparticles from Ti(IV) isopropoxide in benzyl alcohol, the Ala-Arg dipeptide is observed by ¹³C NMR in the resulting solid, at the exclusion of other dipeptides or higher peptides.

This study promotes an innovative synthesis of a nanotheragnostic scaffold capable of targeting and destroying pancreatic cancer cells (PDAC) using the Biotinylated NFL-TBS.40-63 peptide (BIOT-NFL), known to enter various glioblastoma cancer cells (GBM) where it specifically destroys their microtubule network. This recently proposed methodology (P7391FR00-50481 LIV) applied to other peptides VIM (Vimentin) and TAT (Twin-Arginine Translocation) (CPP peptides) has many advantages, such as targeted selective internalization and high stability under experimental conditions, modulated by steric and chemical configurations of peptides. The successful interaction of peptides on gold surfaces has been confirmed by UV-visible, dynamic light scattering (DLS), Zeta potential (ZP) and Raman spectroscopy analyses. The cellular internalization in pancreatic ductal adenocarcinoma (PDAC; MIA PACA-2) and GBM (F98) cells was monitored by transmission electron microscopy (TEM) and showed a better cellular internalization in the presence of peptides with gold nanoparticles. In this work, we also evaluated the power of these hybrid peptide-nanoparticles as photothermal agents after cancer cell internalization. These findings envisage novel perspectives for the development of high peptide-nanotheragnostics.
