RESUMO

Obesity is an established risk factor for many diseases including intestinal cancer. One of the responsible mechanisms is the chronic inflammation driven by obesity. However, it remains to be defined whether diet-induced obesity exacerbates the intestinal inflammatory status by cytokines produced in adipose tissue or the high fat diet first alters the gut microbiota and then drives intestinal inflammation. To address this question, we fed C57BL/6 mice with a high fat diet (HF, 60%) and sacrificed them sequentially after 8, 12, and 16 weeks, and then compositions of gut microbiota and expressions of antimicrobial peptides were determined. The compositions of gut microbiota were altered at 8 wk HF feeding, followed with reduced Paneth antimicrobial peptides lysozyme and Reg IIIγ after 12 and 16 wk HF feeding (p < 0.05), whereas elevations of circulating inflammatory cytokines IFNγ and TNF-α were observed until feeding a HF diet for 16 weeks (p < 0.05). These results indicated that high fat diet may stimulate intestinal inflammation via altering gut microbiota, and it occurs prior to the potential influence by circulating inflammatory cytokines. These findings emphasized the importance of microbiota, in addition to adipose tissue per se, in driving intestinal inflammation, which may thereafter promote intestinal tumorigenesis.

Akirin is a recently discovered nuclear factor that plays an important role in innate immune responses. Beyond its role in innate immune responses, Akirin has recently been shown to play an important role in skeletal myogenesis. In this article, we will briefly review the structure and tissue distribution of Akirin and discuss recent advances in our understanding of its role and signal pathway in skeletal myogenesis.

The anatase/rutile mixed crystal TiO2 was prepared and modified with Ag decoration and SnO2 coupling to construct a Ag@SnO2/anatase/rutile composite photocatalytic material. The crystal structure, morphology, element valence, optical properties and surface area were characterized, and the effects of Ag decoration and SnO2 coupling on the structure and photocatalytic properties of TiO2 were studied. Ag decoration and SnO2 coupling are beneficial to reduce the recombination of photogenerated electrons and holes. When the two modification are combined, a synergistic effect is produced in suppressing the photogenerated charge recombination, making Ag@SnO2/TiO2 exhibits the highest quantum utilization. After 30 min of illumination, the degradation degree of tetracycline hydrochloride (TC) by pure TiO2 increased from 63.3% to 83.1% with Ag@SnO2/TiO2.

Using butyl titanate and absolute ethanol as raw materials, TiO2 was prepared by a hydrothermal method with different hydrothermal times, and the influences of hydrothermal time on the structure and photocatalytic performance of TiO2 were investigated. The obtained samples were characterized by XRD, SEM, TEM, BET, PL and DRS, separately. The results show that TiO2 forms anatase when the hydrothermal time is 12 h, forms a mixed crystal composed of anatase and rutile when the hydrothermal time is 24 h, and forms rutile when the hydrothermal time is 36 h. With the extension of hydrothermal time, anatase gradually transforms into rutile and the surface area decreases. Although TiO2-24 h and TiO2-36 h show lower photoinduced charge recombination and higher light source utilization, TiO2-12 h exhibits the highest photocatalytic activity owing to its largest surface area (145.3 m2/g). The degradation degree of rhodamine B and tetracycline hydrochloride reach 99.6% and 90.0% after 45 min.

The Ascaris roundworms (Ascaridida: Nematoda), one of the commonest soil-transmitted helminths (STHs), can cause ascariasis with significant socioeconomic and public health impact. In this study, the mitochondrial genome of Ascaris ovis, a representative of this genus from the sheep in Southwest China was determined using Illumina sequencing technology. The assembled genome was 14,205 bp in size and encoded 36 genes, including 12 protein-coding genes, 22 transfer RNA genes, and two ribosomal RNA genes. Phylogenetic analysis showed that A. ovis grouped with the congeneric Ascaris lumbricoides of humans, Ascaris spp. of non-human primates and Ascaris suum of pigs and together formed a monophyletic group relationship with either species of Baylisascaris/Toxascaris/Parascaris, species of Toxocara, species of Anisakis/Pseudoterranova or species of Ascaridia/Heterakis in the order Ascaridida, supporting its genetic similarity with A. lumbricoides, A. suum, and other congeneric species. The cumulative mitochondrial DNA data of this genus should contribute to a better understanding of the phylogenetic relationships among these roundworms.

The main issue remains finding a nitrite alternative able to provide its multiple functions. Flos Sophorae exerts antioxidant and prebiotic actions, chili pepper has potent coloring capacity, thus this study investigated whether combination of Flos Sophorae and chili pepper could address the multiple activities of nitrite in Chinese sausages. Dry-fermented sausages were prepared: control and four treatments added with 150 mg/kg sodium nitrite (Nit), 0.2% Flos Sophorae (FS), 1% chili pepper (CP), and combination of 0.2% Flos Sophorae and 1% chili pepper (FS + CP). Results indicated that FS, CP and FS + CP had higher moisture, antioxidant activity and numbers of beneficial Staphylococcus and yeasts Candida, and lower numbers of Escherichia coli and harmful fungi, while FS had lower redness and harder texture than control. Their combination inhibited the declines of capsanthin and antioxidant capacity with ripening time, further improved microbiological communities compared with CP, and resulted in higher redness, similar color score and bacterial community, less lipid oxidation and softer texture compared with Nit. These results suggested that Flos Sophorae in combination with chili pepper could replace the nitrite's contribution to red curing color and microbiological communities, and effectively hinder lipid oxidation in Chinese sausages.

Decabrominated diphenyl ether (BDE-209) is widely used as a flame retardant and is detected at high levels in the environment. Its toxicities have been reported and have attracted attention. In the present study, broilers were used to determine the response in growth performance, carcass traits, meat quality, blood profiles and antioxidant system to BDE-209 exposure at doses of 0, 0.02, 0.4 and 4 mg/kg. The results showed that BDE-209 exposure at levels of 0.02 or 0.4 mg/kg increased feed intake and decreased feed efficiency. BDE-209 altered the blood profiles, such as reducing the numbers of white blood cells, lymphocytes and neutrophilic granulocytes. As compared with the control, BDE-209 exposure significantly increased abdominal fat percentages of broilers at 64.9-159.5% and adversely affected the selected biochemical indicators, including alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatine (CRE), which indicated its toxicity to liver and kidney functions. Moreover, BDE-209 exposure significantly increased plasma malondialdehyde (MDA) concentrations and decreased the activities of glutathione peroxidase (GSH-px) and superoxide dismutase (SOD), which implied aggravating oxidant stress and decline of antioxidant capacity in broilers. In conclusion, our data demonstrated that the environmental pollutant BDE-209 adversely influenced growth performance, increased the deposition of abdominal fat, impaired antioxidant capacity and the immune system and had potential toxicity to the liver and kidney of broilers.

BACKGROUND: Rabbit is a good model for genetic and medical studies in other livestock species. The rabbit shows low adipose tissue deposition, and the phenomena indicates that there is some specificity of adipose deposition during the rabbit growth. However, little is known about genes that regulate the growth of adipose tissue in rabbits. MATERIALS AND METHODS: Deep RNA-seq and comprehensive bioinformatics analyses were used to characterize the genes of rabbit visceral adipose tissue (VAT) at 35, 85 and 120 days after birth. Differentially expressed genes (DEGs) were identified at the three growth stages by DESeq. To explore the function of the candidate genes, Gene Ontology (GO) enrichment and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses were performed. Six DEGs were randomly selected, and their expression profiles were validated by q-PCR. RESULTS: A total of 20,303 known transcripts and 99,199 new transcripts from 8 RNA sequencing libraries were identified, and 34 differentially expressed genes (DEGs) were screened. GO enrichment and KEGG pathway analyses revealed that the DEGs were mainly involved in lipid metabolism regulation including acylglycerol metabolic process and mobilization, and decomposition of lipids to generate ATP in adipocytes and fatty acid metabolism, included LOC100342322 and LOC100342572. In addition, 133 protein-coding genes that play a role in adipose growth and development were screened, including acyl-CoA synthetase long-chain family member 5 (ACSL5) and fatty acid-binding protein 2 (FABP2). The validation results of six DEGs by q-PCR showed similar trends with the results of RNA-seq. CONCLUSION: In summary, this study provides the first report of the coding genes profiles of rabbit adipose tissue during different growth stages. These data allow for the identification of candidate genes for subsequent studies on rabbit genetics and regulation of adipose cells, and provide an animal model for studying obesity in humans.

Punicalagin and ellagic acid are the major polyphenols present in pomegranate peels. The contents of α-punicalagin, ß-punicalagin, and ellagic acid in the pomegranate peels were approximately 75, 72, and 20 µM, respectively. The reactions of polyphenols in pomegranate peels with sodium nitrite under simulated stomach conditions were studied. The reactions decreased the polyphenolic contents of the pomegranate peels and accompanied the formation of nitroso compounds. The oxidation rates followed the order ellagic acid <α-punicalagin ≈ ß-punicalagin. The results suggested that the reactions can occur in the stomach after a meal, while the pH changes from 2 to 4.5.

RESUMO

We demonstrate phase-sensitive amplification of multiple wavelength-division-multiplexed continuous-wave (CW) signals by frequency nondegenerate four-wave-mixing process in optical fiber. By fine-tuning the optical wavelengths of the CW signals, simultaneous phase-sensitive in-line amplification of three signal channels is realized. This indicates the possibility of amplifying multiple data channels by an in-line phase-sensitive fiber parametric amplifier. We also discuss a potential system architecture employing such amplifiers.

Firmicutes and Bacteroidetes are closely related to body fat in humans and mice, which are the two dominant bacterial divisions of gut microbiota in mammals. Here real-time PCR analysis indicated that Meishan pigs had a 34% reduction in percentage Bacteroidetes (P = 0.008) and a significantly lower proportion of Bacteroides (P = 0.013) than Landrace pigs. The percentage of Bacteroidetes or Bacteroides had a negative correlation with body fat (R2 was 0.63 for Bacteroidetes and 0.57 for Bacteroides, P < 0.05). There was a trend that the percentage of Firmicutes in Meishan pigs was higher in numerical value than in Landrace pigs, although this difference was not statistically significant (P = 0.290) between the two breeds. These suggested that body fat correlated with the percentage of Bacteroidetes division of the gut microbiota in the common pig breeds, and the differences of gut microbial ecology in obese versus lean animals may be analogous.

The pure, tin (Sn)-doped, lanthanum (La)-doped and Sn/La co-doped titanium dioxide (TiO2) nanomaterials were synthesized using sol-gel method followed by calcination at the temperature of 360 °C, 450 °C and 600 °C, respectively. The structures of the nanomaterials were characterized by X-ray diffraction (XRD), Thermogravimetric (TG), Differential Thermal Analysis (DTA), Scanning Electron Microscopy (SEM), Energy Dispersive Spectrum (EDS), Transmission Electron Microscopy (TEM), X-ray Photoelectron Spectrum (XPS), Diffuse Reflectance Spectrum (DRS), Photoluminescence Spectrum (PL), Brunauer-Emmett-Teller Measurements (BET), respectively. The photocatalytic property of the photocatalysts under UV light was evaluated through the degradation of Rhodamine B (RhB). The results show that the anatase-rutile phase transition is promoted by Sn-doping while La-doping retards the phase transition. However, La doping plays a major role in the process of phase transformation. The photocatalytic activity of pure TiO2 is affected by annealing temperature remarkably and the optimal annealing temperature is 450 °C. The photocatalytic activity of TiO2 is enhanced significantly by Sn and La doping at three different temperatures. Sn/La-TiO2 exhibits the highest degradation rates and the fastest reaction rates probably owing to the synergistic effect of Sn4+ and La3+ ions in inhibiting the recombination of photogenerated electron-hole pairs. The formation of extra surface hydroxyl groups and additional surface area are also beneficial for the photocatalytic activity.

Gut dysbiosis induced by high fat diet (HF) or obesity is a predisposing factor to develop diverse inflammatory diseases. Polyphenols and fibers, often eaten together, have been reported to have prebiotic actions, but their health promoting benefits still need to be further characterized and defined. This study attempted to understand how polyphenol rutin and polysaccharide inulin influence intestinal health in mouse model fed a HF (60 kcal%) diet. A total of 48 C57BL/6J mice were divided into four groups fed with a low fat (10% kcal%) control diet (LC), a high fat control diet (HC), a high-fat diet supplemented with rutin (HR), or a high-fat diet supplemented rutin and inulin (HRI) for 20 weeks. Rutin supplementation reduced the HF diet-induced increase of Firmicutes/Bacteroidetes (F/B) ratio, Deferribacteraceae population and plasma lipopolysaccharide (LPS) (p < 0.05); ameliorated inflammation as indicated by the decreased circulating inflammatory cytokines (p < 0.05) and the reduced expressions of intestinal inflammatory mediators (p < 0.05); and attenuated the endoplasmic reticulum (ER) stress in Paneth cells as indicated by the decreased expressions of the ER markers (p < 0.05). Compared to the rutin supplementation alone, the co-administration of rutin with inulin improved the utilization of rutin as indicated by its decreased excretion, suppressed a number of harmful bacteria including Deferribacteraceae and Desulfovibrionaceae (p < 0.05), and further reduced the expression of the key inflammatory cytokine TNF-α and increased the production of butyrate, despite the supplementation of inulin reversed the decrease of body weight induced by rutin supplementation due to an increased food intake. Taken together, our data demonstrated that rutin supplementation ameliorated the inflammatory status and ER stress in Paneth cells under a HF-induced obese state, and its co-administration with inulin further mitigated the inflammatory status, indicating the potential to combine polyphenol rutin and the polysaccharide inulin as a dietary strategy to ameliorate gut dysbiosis, to improve inflammatory status and thereby to reduce medical disorders associated with HF-induced obesity.

We experimentally demonstrate, for the first time to our knowledge, a phase-sensitive amplifier based on frequency nondegenerate parametric amplification in optical fiber, where the input signal-idler pair is prepared all-optically. Using two fiber-optic parametric amplifier sections separated by a fiber-based wavelength-dependent phase shifter, we observe and investigate phase-sensitive gain profile in the 1550 nm region both experimentally and theoretically. The realized scheme automatically generates gain-defining phase that is environmentally stable, making it advantageous for building phase-sensitive transmission links.

RESUMO

We demonstrate a novel, self-starting optoelectronic oscillator based on an electro-absorption modulator in a fiber-extended cavity for generating an optical pulse stream with high-rate and ultra-low jitter capabilities. Optical pulses at 10GHz repetition rate are demonstrated with >90dBc/Hz side-mode suppression and the lowest timing jitter (42fs in the 100Hz-1MHz range) reported to date for a self-starting source. Along with the optical pulse stream, the oscillator also generates a 10GHz electrical signal with ultra-low phase noise.

The study was designed to investigate the effects of nutritional level (control diet (CD), 14.19% crude protein, 13.81MJ of DE/kg; low nutritional level diet (LND), 11.08% crude protein, 12.55MJ of DE/kg) on pork quality and gene expression of mu-calpain and calpastatin in muscle of finishing pigs. The LND treatment increased drip loss (P<0.05), had a trend to increase intramuscular fat (IMF) content (P=0.09), decreased Warner-Bratzler shear force (WBSF) of pork (P<0.05), improved mRNA level of mu-calpain (P<0.05) in skeletal muscle, but had no effect on gene expression of calpastatin, compared with the CD treatment. These data suggest that a moderately reduced energy and protein diet increased pork tenderness and intramuscular fat. The increase in tenderness by LND treatment may be partly due to increased gene expression of mu-calpain in muscle.

The objective of this study was to investigate the effects of supplementing swine finishing diets with two levels of magnesium aspartate (MgAsp) and short-term transportation stress on blood parameters, pork quality and the mRNA abundance of p-calpain and calpastatin in muscles of finishing pigs. Thirty-six crossbred finishing pigs (mean BW 90 kg) were assigned randomly to 0, 1000, or 2000 mg supplemental Mg from MgAsp per kg of diet for five days before slaughter. Then six pigs from each dietary treatment were subjected either to no transportation stress (NTS) or 2 h of transportation stress (TS). Transportation stress resulted in higher concentrations (p < 0.01) of serum calcium, glucose and cortisol, lower pH (p < 0.01), higher Warner-Bratzler shear force (WBSF) (p < 0.05) and higher calpastatin mRNA abundance (p = 0.05) of longissimus muscle (LM) compared with NTS treatments. Supplementation of MgAsp in TS treatments increased serum Mg concentration (p < 0.05) at 2000 mg of Mg/kg, reduced drip loss (p < 0.05) and improved pork quality colour (p < 0.05) at 2000 mg of Mg/kg, and decreased 1-day and 3-day WBSF (p < 0.05) at 1000 mg of Mg/kg compared with TS treatments fed the control diet. It is concluded that supplementation of MgAsp improves water-holding capacity and pork colour, and alleviates the negative effects of transportation stress on meat tenderness.

We demonstrate a regeneratively mode-locked fiber-optical parametric oscillator that utilizes intracavity dispersion compensation to generate pulses at a 10-GHz repetition rate in both soliton and nonsoliton regimes. At the threshold pump power the generated pulses are close to fundamental solitons. At higher pump powers we found a significant deviation of the pulses from the sech2 shape. In addition, the use of an ultralow-jitter self-starting pump-pulse source in a regenerative feedback loop allows for a significant reduction of the signal's timing jitter and amplitude noise.

We report measurement of the noise statistics of spontaneous parametric fluorescence in a fiber parametric amplifier with single-mode, single-photon resolution. We employ optical homodyne tomography for this purpose, which also provides a self-calibrating measurement of the noise figure of the amplifier. The measured photon statistics agree with quantum-mechanical predictions, and the amplifier's noise figure is found to be almost quantum limited.

RESUMO

Recent theoretical work predicts that the quantum-limited noise figure of a x(3)-based fiber-optical parametric amplifier operating as a phase-insensitive in-line amplifier or as a wavelength converter exceeds the standard 3-dB limit at high gain. The degradation of the noise figure is caused by the excess noise added by the unavoidable Raman gain and loss occurring at the signal and the converted wavelengths. We present detailed experimental evidence in support of this theory through measurements of the gain and noise-figure spectra for phase-insensitive parametric amplification and wavelength conversion in a continuous-wave amplifier made from 4.4 km of dispersion-shifted fiber. The theory is also extended to include the effect of distributed linear loss on the noise figure of such a long-length parametric amplifier and wavelength converter.
