RESUMO

The patterns of supravital staining with euchrysine, a fluorescent dye thought to bind preferentially to membranes of lysosomes and other structures involved in endocytosis, were evaluated sequentially in PHA-stimulated cultures of human unseparated, T and non-T lymphocytes under conditions of a reversible, amethopterine-imposed DNA synthesis block. It was found that type I cells (with a single conglomerate of fluorescent granules, constituting approx. 50% of resting T cells) were almost completely replaced by type III cells (with large conglomerates of coarse granules, typical of lymphocytes undergoing blast transformation) up to the end of second day of culture. Type II cells (with very fine fluorescent granules scattered over the cytoplasms, present in about 50% of T and all non-T lymphocytes) did not initially change their number but later were replaced by type III cells, usually starting from the third day of culture. A hypothesis is discussed according to which only type I cells are primarily responsive to PHA, whereas type II cells require "help" from the former, prior to transition into the proliferative stage.

RESUMO

The distribution of the fluorescent euchrysine-binding grains in the peripheral and bonmarrow stem cells was estimated in 28 cases of different types of acute leukemia verified according to Lòffler's cytochemical classification. The relationship between the pattern of fluorescent euchrysine-binding grains of human stem cells and the cytochemical type of acute leukemias has been established. Consequently, the authors suggest that the euchrysine-binding grains pattern of human stem cells may have a diagnostic value in distinguishing between various types of acute leukemia.

The literature data indicating nonspecific pathologic symptoms of the circulatory system in those occupationally exposed to caprolactam and diphenyl inspired the authors to evaluate the circulatory system in a group of 495 workers of a Chemical Plant producing polyamide fibres. The anamnestic data, family inquiry and subjective changes indicate that any hints of the significant effects of the technological process on circulatory diseases in this professional group are unwarranted. The percentage values of the latent circulatory failure (1-2% of subjects), coronary insufficiency (6.6% of subjects), arterial hypertension (5.7% of subjects) did not show any significant differences, as compared to the control group and normal Polish population. The obtained results of ECG test evaluated according to the Minnesota-Code criterion did not significantly differ from 100 controls composed of those in whom no changes in the circulatory system were found, whereas clear differences were found related to 50 persons with documented heart ischemia.
