RESUMO

Soybean is a typical short-day crop, and most commercial soybean cultivars are restricted to a relatively narrow range of latitudes due to photoperiod sensitivity. Photoperiod sensitivity hinders the utilization of soybean germplasms across geographical regions. When grown in temperate regions, tropical soybean responds to prolonged day length by increasing the vegetative growth phase and delaying flowering and maturity, which often pushes the harvest window past the first frost date. In this study, we used CRISPR/LbCas12a to edit a North American subtropical soybean cultivar named 06KG218440 that belongs to maturity group 5.5. By designing one gRNA to edit the nuclear localization signal (NLS) regions of both E1 and E1Lb, we created a series of new germplasms with shortened flowering time and time to maturity and determined their favourable latitudinal zone for cultivation. The novel partial function alleles successfully achieve yield and early maturity trade-offs and exhibit good agronomic traits and high yields in temperate regions. This work offers a straightforward editing strategy to modify subtropical and tropical soybean cultivars for temperate growing regions, a strategy that could be used to enrich genetic diversity in temperate breeding programmes and facilitate the introduction of important crop traits such as disease tolerance or high yield.

Premature Ovarian Insufficiency (POI) is a highly heterogeneous condition characterized by ovarian dysfunction in women occurring before the age of 40, representing a significant cause of female infertility. It manifests through primary or secondary amenorrhea. While more than half of POI cases are idiopathic, genetic factors play a pivotal role in all instances with known causes, contributing to approximately 20-25% of cases. This article comprehensively reviews the genetic factors associated with POI, delineating the primary candidate genes. The discussion delves into the intricate relationship between these genes and ovarian development, elucidating the functional consequences of diverse mutations to underscore the fundamental impact of genetic effects on POI. The identified genetic factors, encompassing gene mutations and chromosomal abnormalities, are systematically classified based on whether the resulting POI is syndromic or non-syndromic. Furthermore, this paper explores the genetic interplay between mitochondrial genes, such as Required for Meiotic Nuclear Division 1 homolog Gene (RMND1), Mitochondrial Ribosomal Protein S22 Gene (MRPS22), Leucine-rich Pentapeptide Repeat Gene (LRPPRC), and non-coding RNAs, including both microRNAs and Long non-coding RNAs, with POI. The insights provided serve to consolidate and enhance our understanding of the etiology of POI, contributing to establishing a theoretical foundation for diagnosing and treating POI patients, as well as for exploring the mechanisms underlying the disease.

Osteoarthritis has become a major disease threatening human health. The mechanism of injury under fluid involvement can be studied by finite element method. However, most models only model the articular cartilage to study the subchondral bone structure, which is too simplistic. In this study, a complete osteochondral unit was modeled and provided with a poroelastic material, and as osteoarthritis develops and the size, thickness, and shape of the osteochondral unit defect varies, the fluid flow behavior is altered, which may have functional consequences that feed back into the progression of the injury. The results of the study showed that interstitial fluid pressure and velocity decreased in defective osteochondral units. This trend was exacerbated as the size and thickness of the defect in the osteochondral unit increased. When the defect reached the trabeculae, pressure around the cartilage defect in the osteochondral unit was greatest, flow velocity in the subchondral cortical bone was greatest, and pressure and flow velocity around the trabecular defect were lowest. As osteoarthritis develops, the osteochondral unit becomes more permeable, and the pressure of the interstitial fluid decreases while the flow rate increases, resulting in severe nutrient loss. This may be the fluid flow mechanism behind osteochondral defects and osteoarthritis.

Problems, such as broken screws, broken rods, and cage subsidence after clinical spinal fusion surgery affect the success rate of fusion surgery and the fixation effect of fusion segments, and these problems still affect the treatment and postoperative recovery of patients. In this study, we used the biomechanical finite element analysis method to analyze and study the fixation effect of three kinds of spinal internal fixation systems on L4-L5 lumbar spine segments in percutaneous endoscopic posterior lumbar interbody fusion (PE-PLIF). The three different fixation systems compared in this study include bilateral pedicle screw fixation (M1); bilateral pedicle screw with cross-link fixation (M2); bilateral pedicle screws with double bent rods fixation (M3). The internal fixation systems with different structures were analyzed with the help of Hypermesh, and Abaqus. It was found that the internal fixation system with double bent rods reduced screw stresses by 23.8 and 22.2% in right and left axial rotation than the traditional bilateral pedicle screw system, while titanium rod stresses were reduced by 9.6, 3.7, 9.6, and 2.9% in flexion, left and right lateral bending, and right axial rotation, respectively, and L5 upper endplate stresses were reduced by 35.5, 18.9, 38.4, 10.2, and 48.3% in flexion, left and right lateral bending, and left and right axial rotation, respectively. The spinal range of motion (ROM) of the M3 internal fixation system was less than that of the M1 and M2 internal fixation systems in left lateral bending, left lateral rotation, and right axial rotation, and the intact vertebral ROM was reduced by 93.7, 94.9, and 90.9%, respectively. The double bent rod structure of the spinal internal fixation system has better biomechanical properties, which can effectively reduce the risk of screw breakage, loosening, cage subsidence, and endplate collapse after fusion surgery.

Both ammonium sulfite slurry (ASS) from ammonia-based desulfurization and lignite are waste materials with low value. In this work, an innovative method was developed by applying ASS in lignite activation to produce water-soluble humic substances (WHSs) with a high bioactivity and economic value. The optimal activation method was to mix lignite and ASS at a 4:1-liquid-solid ratio by vortex blender and then oscillate it for 30 min at 25 °C. Compared with that of the unactivated lignite (UAL), the yield of WHSs from activated lignite (AL) increased by 42.72%. WHSs from AL consisted of a large number of aliphatic carbons with low molecular weight and functional groups such as amides, amines, sulfonic acid groups, C-O, and so forth. Moreover, WHSs from AL at lower concentrations (2 mg/L) has a more obvious root-elongation-promoting effect than WHSs from UAL (10 mg/L). Activation experiment with the lignite-related model compounds revealed that ASS caused the breakage of Caliph-O, Caliph-Caliph, and Carom-Caliph linkages between aromatic rings. These findings provide a theoretical basis for the development of green and sustainable technologies for the beneficial reuse of ASS and lignite in agriculture.

Due to the ecological toxicity and environmental residues, how to remove the persistent organic pollutants (POPs), especially of polycyclic-aromatic-hydrocarbons (PAHs) and dichloro-diphenyl-trichloroethanes (DDTs), from agricultural soil has captured the attention of scholars for a long time. To develop an effective and low-cost in situ co-remediation technique, five independent but complementary treatments were used on an over-standard PAHs-DDTs co-contaminated soil in an agricultural greenhouse. Experimental results identified that the combination of microbe (Bacillus methylotrophicus) - plant (Brassica rapa) could remove rhamnolipid activated PAHs and DDTs effectively after enhanced by Staphylococcus pasteuri. Also, the Benzoapyrene and total DDTs residue in Brassica rapa was up to the standard of National (China) food safety. The lignin enhanced the removal of high-rings PAHs and p-p' DDE but reduced soil microbial biomass carbon and soil enzymes activity (polyphenol oxidase, invertase and acid phosphatase). Pearson correlation analysis showed that polyphenol oxidase activity was significantly related to the PAHs/DDTs dissipation rate. Our research suggested a new amendment that could remediate PAHs/DDTs co-contaminated agricultural soil without interrupting crop production, and the polyphenol oxidase activity should be considered as a micro-ecological indicator in this process.

Se evaluó la eficiencia de la trampa Mosquito Magnet Liberty PlusTM (MMLP) cebada con los atrayentes químicos 1-octen-3-ol (octenol) y Lurex 3TM (L-ácido láctico) junto con dióxido de carbono (CO2) para recolectar anofelinos en la localidad Ye'kwana de Boca de Nichare, municipio Sucre, estado Bolívar, Venezuela. La trampa fue operada entre las 18:00 y 06:00 horas, durante 10 noches por mes, durante cuatro meses (Julio ­ Octubre, 2015), alternándose cada noche el atrayente. A fin de caracterizar la actividad nocturna de los anofelinos, cada cuatro horas se cambiaba la jaulita donde eran atrapados los mosquitos. Se capturaron cinco especies de Anopheles y cuatro de Culicinae, siendo las más abundantes, Anopheles (Nyssorhynchus) darlingi Root y Anopheles (Nyssorhynchus) oswaldoi (Peryassú) sensu lato. El ANOVA factorial no paramétrico de Transformación de Rangos Alineados para la variable atrayente y sus interacciones con especie e intervalo horario no fueron estadísticamente significativas (p > 0.05). Estas especies muestran comportamientos diferentes: An. darlingi tiene el pico de actividad nocturna entre las 22:00 y 02:00 horas, mientras que en An. oswaldoi s.l. se observa un incremento de la actividad nocturna progresivamente que se mantiene durante el intervalo 02:00-06:00 horas. Los resultados permiten concluir que la trampa MMLP cebada con octenol o Lurex es un método alternativo de recolección de mosquitos para la vigilancia entomológica en áreas maláricas remotas con población indígena. La actividad horaria de los vectores An. darlingi y An. oswaldoi s.l. sugiere que el uso de mosquiteros tratados con insecticidas puede resultar un método efectivo para el control de la transmisión de malaria en esta zona del país(AU)


The efficiency of the Mosquito Magnet Liberty PlusTM (MMLP) trap was evaluated baited with the chemical attractants 1-octen-3-ol (octenol) and Lurex3M (L-lactic acid) together with carbon dioxide (CO2) to collect anophelines in the Amerindian village Boca de Nichare, Sucre municipality, Bolívar State, Venezuela. The trap was operated between 18:00 and 06: hours, 10 nights per month for four months (July-October 2015), the attractants were alternated nightly. To determine the host seeking behavior of anophelines, the trap cage was changed every 4 hours. Five species of Anopheles and four of Culicinae were collected; Anopheles (Nyssorhynchus) darlingi Root and Anopheles oswaldoi (Peryassú) sensu lato were the most abundant species collected. The Align Rank Transform (ART) test for nonparametric ANOVA for the variable attractant and its interactions with species and hour interval were not significant (p > 0.05). An. darlingi and An. oswaldoi s.l. showed contrasting host seeking activity: An. darlingi showed a peak between 22:00 and 02:00 hours, while in An. oswaldoi s.l. there was a steady increase in activity up to the interval 02:00-06:00 hours. The MMLP trap baited either with octenol or Lurex is an alternative method for entomological surveillance in remote Amerindian malaria endemic areas. The biting activity of the vectors An. darlingi and An. oswaldoi s.l. suggests that the use of long-lasting insecticide treated nets could be an effective method to control the transmission of malaria in this area(AU)
