RESUMO

Wearing surgical masks remains the most effective protective measure against COVID-19 before mass vaccination, but insufficient comfortability and low antibacterial/antiviral activities accelerate the replacement frequency of surgical masks, resulting in large amounts of medical waste. To solve this problem, we report new nanofiber membrane masks with outstanding comfortability and anti-pathogen functionality prepared using fluorinated carbon nanofibers/carbon fiber (F-CNFs/CF). This was used to replace commercial polypropylene (PP) nonwovens as the core layer of face masks. The through-plane and in-plane thermal conductivity of commercial PP nonwovens were only 0.12 and 0.20 W/m K, but the F-CNFs/CF nanofiber membranes reached 0.62 and 5.23 W/m K, which represent enhancements of 380% and 2523%, respectively. The surface temperature of the PP surgical masks was 23.9 ℃ when the wearing time was 15 min, while the F-CNFs/CF nanocomposite fibrous masks reached 27.3 ℃, displaying stronger heat dissipation. Moreover, the F-CNFs/CF nanofiber membranes displayed excellent electrical conductivity and produced a high-temperature layer that killed viruses and bacteria in the masks. The surface temperature of the F-CNFs/CF nanocomposite fibrous masks reached 69.2 ℃ after being connected to a portable power source for 60 s. Their antibacterial rates were 97.9% and 98.6% against E. coli and S. aureus, respectively, after being connected to a portable power source for 30 min.

Listeria monocytogenes is a deadly foodborne pathogen, and infections can result in meningoencephalitis and sepsis with mortality rates of up to 30%. In this study, we performed comparative whole-genome analysis of 30 clinical isolates sequenced together with 32 previously sequenced clinical and food isolates from China. The data indicate that L. monocytogenes isolates belonging to the clonal complexes (CC) -1, -8, -9, -87, -121, and -155 are present in human clinical cases. The majority of isolates are from CC-87, 9, and 8 and overlap with those CCs previously reported on the basis of multilocus sequence typing for isolates from Chinese food products. Detailed genome analysis of isolates, representative of CCs in clinical and food products, revealed strong similarities both in their core- and accessory genomes indicating that they are highly related. When compared to genome sequences of isolates of a given CC worldwide, clinical isolates of China were distinct and clustered in unified clades. Our data indicate that epidemic clones of L. monocytogenes (CC-87, 9, and 8) with unusually high occurrence of plasmids are unique to China and suggest that common populations of L. monocytogenes clones are present in both clinical and food products in China.

BACKGROUND: Phosphorus (Pi) deficiency induces root morphological remodeling in plants. The primary root length of rice increased under Pi deficiency stress; however, the underlying mechanism is not well understood. In this study, transcriptome analysis (RNA-seq) and Real-time quantitative PCR (qRT-PCR) techniques were combined with the determination of physiological and biochemical indexes to research the regulation mechanisms of iron (Fe) accumulation and callose deposition in rice roots, to illuminate the relationship between Fe accumulation and primary root growth under Pi deficient conditions. RESULTS: Induced expression of LPR1 genes was observed under low Pi, which also caused Fe accumulation, resulting in iron plaque formation on the root surface in rice; however, in contrast to Arabidopsis, low Pi promoted primary root lengthening in rice. This might be due to Fe accumulation and callose deposition being still appropriately regulated under low Pi. The down-regulated expression of Fe-uptake-related key genes (including IRT, NAS, NAAT, YSLs, OsNRAMP1, ZIPs, ARF, and Rabs) inhibited iron uptake pathways I, II, and III in rice roots under low Pi conditions. In contrast, due to the up-regulated expression of the VITs gene, Fe was increasingly stored in both root vacuoles and cell walls. Furthermore, due to induced expression and increased activity of ß-1-3 glucanase, callose deposition was more controlled in low Pi treated rice roots. In addition, low Pi and low Fe treatment still caused primary root lengthening. CONCLUSIONS: The obtained results indicate that Low phosphorus induces iron and callose homeostatic regulation in rice roots. Because of the Fe homeostatic regulation, Fe plays a small role in rice root morphological remodeling under low Pi.

The responses of sulfur (S) uptake assimilation-related genes' expression in roots of two rice cultivars to cadmium (Cd), bensulfuron-methyl (BSM) and their co-contamination (Cd+BSM) were investigated by gene-chip microarray analysis and quantitative real-time PCR (QRT-PCR) technology. Treatments of Cd and Cd+BSM induced expression of sulfate transporter and permease genes, and promoted sulfate uptake in rice roots. Cd+BSM could alleviate Cd toxicity to cv. Fengmeizhan seedlings, probably due to Cd+BSM promoting greater S absorption by seedlings. Cd and Cd+BSM induced expression of sulfate assimilation-related genes, and thus activated the sulfur assimilation pathway. Cd and Cd+BSM induced expression of phytochelatin synthase and metallothionein genes, and induced expression of glutathione S-transferases (GSTs), glutathione synthase (GS) and S-containing antioxidation enzyme genes, which detoxified Cd(2+). It is suggested that (to cope with the toxicity of Cd, BSM and their co-contamination) the S uptake and assimilation pathway was activated in rice roots by increased expression of related genes, thus enhancing the supply of organic S for synthesis of Cd or BSM resistance-related substances.

Wearing surgical masks is one of the best protective measures to protect humans from viral invasion during the 2019 coronavirus (COVID-19) outbreak. However, wearing surgical masks for extended periods will cause uncomfortable sweltering sense to users and are easy to breed bacteria. Here, we reported a novel fibrous membrane with outstanding comfortability and antibacterial activity prepared by PP ultrafine fiber nonwovens and antibacterial functionalized h-BN nanoparticles (QAC/h-BN). The thermal conductivity of commercial PP nonwovens was only 0.13 W m-1 K-1, but that of the QAC/h-BN/PP nanocomposite fibrous membranes can reach 0.88 W m-1 K-1, an enhancement of 706.5% than commercial PP nonwovens. The surface temperature of commercial PP surgical masks was 31.8 °C when the wearing time was 60 min. In contrast, QAC/h-BN/PP surgical masks can reach 33.6 °C at the same tested time, exhibiting stronger heat dissipation than commercial PP surgical masks. Besides, the antibacterial rates of QAC/h-BN/PP nanocomposite fibrous membranes were 99.3% for E. coli and 96.1% for S. aureus, and their antibacterial mechanism was based on "contact killing" without the release of unfavorable biocides. We think that the QAC/h-BN/PP nanocomposite fibrous membranes could provide better protection to people.

The molecular response of wheat (Triticum aestivum L., cv. Yangmai 13) seedlings to heavy metal (Cd, Hg) and 1,2,4-trichlorobenzene (TCB) stresses were examined by two-dimensional gel electrophoresis, image analysis, and peptide mass fingerprinting. The results showed inhibitions of root and shoot growth by Cd, Hg, and TCB. These stresses led to water deficit and lipid phosphorylation in the seedling which also promoted protein phophorylation in the leaves. Hg stress inhibited protein synthesis while Cd and TCB stresses induced or up-regulated more proteins in the leaves. Most of these induced proteins played important roles in the biochemical reactions involved in tolerance of wheat to Cd and TCB stresses. The primary functions of Cd- and TCB-induced proteins included methionine metabolism, Rubisco modification, protein phosphorylation regulation, protein configuration protection, H+ transmembrane transportation and also the synthesis of ethylene, defense substances and cell wall compounds.

The proteomic analysis of rice (Oryza sativa L.) roots and leaves responding to 1,2,4-trichlorobenzene (TCB) stress was carried out by two dimensional gel electrophoresis, mass spectrometric (MS), and protein database analysis. The results showed that 5 mg/L TCB stress had a significant effect on global proteome in rice roots and leaves. The analysis of the category and function of TCB stress inducible proteins showed that different kinds of responses were produced in rice roots and leaves, when rice seedlings were exposed to 5 mg/L TCB stress. Most responses are essential for rice defending the damage of TCB stress. These responses include detoxication of toxic substances, expression of pathogenesis-related proteins, synthesis of cell wall substances and secondary compounds, regulation of protein and amino acid metabolism, activation of methionine salvage pathway, and also include osmotic regulation and phytohormone metabolism. Comparing the TCB stress inducible proteins between the two cultivars, the beta-glucosidase and pathogenesis-related protein family 10 proteins were particularly induced by TCB stress in the roots of rice cultivar (Oryza sativa L.) Aizaizhan, and the glutathione S-transferase and aci-reductone dioxygenase 4 were induced in the roots of rice cultivar Shanyou 63. This may be one of the important mechanisms for Shanyou 63 having higher tolerance to TCB stress than Aizaizhan.

Abscisic acid (ABA) is an important phytohormone controlling responses to abiotic stresses and is sensed by proteins from the PYR/PYL/RCAR family. To explore the genetic contribution of PYLs toward ABA-dependent and ABA-independent processes, we generated and characterized high-order Arabidopsis mutants with mutations in the PYL family. We obtained a pyl quattuordecuple mutant and found that it was severely impaired in growth and failed to produce seeds. Thus, we carried out a detailed characterization of a pyl duodecuple mutant, pyr1pyl1/2/3/4/5/7/8/9/10/11/12. The duodecuple mutant was extremely insensitive to ABA effects on seed germination, seedling growth, stomatal closure, leaf senescence, and gene expression. The activation of SnRK2 protein kinases by ABA was blocked in the duodecuple mutant, but, unexpectedly, osmotic stress activation of SnRK2s was enhanced. Our results demonstrate an important role of basal ABA signaling in growth, senescence, and abscission and reveal that PYLs antagonize ABA-independent activation of SnRK2s by osmotic stress.

This study is the first to comprehensively characterize m6A patterns in the Arabidopsis chloroplast and mitochondria transcriptomes based on our open accessible data deposited in NCBI's Gene Expression Omnibus with GEO Series accession number of GSE72706. Over 86% of the transcripts were methylated by m6A in the two organelles. Over 550 and 350 m6A sites were mapped, with ~5.6 to ~5.8 and ~4.6 to ~4.9 m6A sites per transcript, to the chloroplast and mitochondria genome, respectively. The overall m6A methylation extent in the two organelles was greatly higher than that in the nucleus. The m6A motif sequences in the transcriptome of two organelles were similar to the nuclear motifs, suggesting that selection of the m6A motifs for RNA methylation was conserved between the nucleus and organelle transcriptomes. The m6A patterns of rRNAs and tRNAs in the organelle were similar to those in the nucleus. However, the m6A patterns in coding RNAs were distinct between the nucleus and the organelle, suggesting that that regulation of the m6A methylation patterns may be different between the nuclei and the organelles. The extensively methylated transcripts in the two organelles were mainly associated with rRNA, ribosomal proteins, photosystem reaction proteins, tRNA, NADH dehydrogenase and redox. On average, 64% and 79% of the transcripts in the two organelles showed differential m6A methylation across three organs of the leaves, flowers and roots. The m6A methylation extent in the chloroplast was higher than that in the mitochondria. This study provides deep insights into the m6A methylation topology and differentiation in the plant organelle transcriptomes.

BACKGROUND: m(6)A is a ubiquitous RNA modification in eukaryotes. Transcriptome-wide m(6)A patterns in Arabidopsis have been assayed recently. However, differential m(6)A patterns between organs have not been well characterized. RESULTS: Over two-third of the transcripts in Arabidopsis are modified by m(6)A. In contrast to a recent observation of m(6)A enrichment in 5' mRNA, we find that m(6)A is distributed predominantly near stop codons. Interestingly, 85 % of the modified transcripts show high m(6)A methylation extent compared to their transcript level. The 290 highly methylated transcripts are mainly associated with transporters, stress responses, redox, regulation factors, and some non-coding RNAs. On average, the proportion of transcripts showing differential methylation between two plant organs is higher than that showing differential transcript levels. The transcripts with extensively higher m(6)A methylation in an organ are associated with the unique biological processes of this organ, suggesting that m(6)A may be another important contributor to organ differentiation in Arabidopsis. Highly expressed genes are relatively less methylated and vice versa, and different RNAs have distinct m(6)A patterns, which hint at mRNA fate. Intriguingly, most of the transposable element transcripts maintained a fragmented form with a relatively low transcript level and high m(6)A methylation in the cells. CONCLUSIONS: This is the first study to comprehensively analyze m(6)A patterns in a variety of RNAs, the relationship between transcript level and m(6)A methylation extent, and differential m(6)A patterns across organs in Arabidopsis.
