RESUMO

PURPOSE: To develop and evaluate a novel method for computationally efficient reconstruction from noisy MR spectroscopic imaging (MRSI) data. METHODS: The proposed method features (a) a novel strategy that jointly learns a nonlinear low-dimensional representation of high-dimensional spectroscopic signals and a neural-network-based projector to recover the low-dimensional embeddings from noisy/limited data; (b) a formulation that integrates the forward encoding model, a regularizer exploiting the learned representation, and a complementary spatial constraint; and (c) a highly efficient algorithm enabled by the learned projector within an alternating direction method of multipliers (ADMM) framework, circumventing the computationally expensive network inversion subproblem. RESULTS: The proposed method has been evaluated using simulations as well as in vivo 1 $$ {}^1 $$ H and 31 $$ {}^{31} $$ P MRSI data, demonstrating improved performance over state-of-the-art methods, with about 6 × $$ \times $$ fewer averages needed than standard Fourier reconstruction for similar metabolite estimation variances and up to 100 × $$ \times $$ reduction in processing time compared to a prior neural network constrained reconstruction method. Computational and theoretical analyses were performed to offer further insights into the effectiveness of the proposed method. CONCLUSION: A novel method was developed for fast, high-SNR spatiospectral reconstruction from noisy MRSI data. We expect our method to be useful for enhancing the quality of MRSI or other high-dimensional spatiospectral imaging data.

The lasso peptide microcin Y (MccY) effectively inhibits various serotypes of Salmonella in vitro, but the antibacterial effect against S. Pullorum in poultry is still unclear. This study was the first to evaluate the safety and anti-S. Pullorum infection of MccY in specific pathogen-free (SPF) chicks. The safety test showed that the body weight, IgA and IgM levels of serum, and cecal microbiota structure of 3 groups of chicks orally administrated with different doses of MccY (5 mg/kg, 10 mg/kg, 20 mg/kg) for 14 days were not significantly different from those of the control group. Then, the chicks were randomized into 3 groups for the experiment of anti-S. Pullorum infection: (I) negative control group (NC), (II) S. Pullorum-challenged group (SP, 5 × 108 CFU/bird), (III) MccY-treated group (MccY, 20 mg/kg). The results indicated that compared to the SP group, treatment of MccY increased body weight and average daily gain (P < 0.05), reduced S. Pullorum burden in feces, liver, and cecum (P < 0.05), enhanced the thymus, and decreased the spleen and liver index (P < 0.05). Additionally, MccY increased the jejunal villus height, lowered the jejunal and ileal crypt depth (P < 0.05), and upregulated the expression of IL-4, IL-10, ZO-1 in the jejunum and ileum, as well as CLDN-1 in the jejunum (P < 0.05) compared to the SP group. Furthermore, MccY increased probiotic flora (Barnesiella, etc.), while decreasing (P < 0.05) the relative abundance of pathogenic flora (Escherichia and Salmonella, etc.) compared to the SP group.

RESUMO

Type I photosensitizers offer an advantage in photodynamic therapy (PDT) due to their diminished reliance on oxygen levels, thus circumventing the challenge of hypoxia commonly encountered in PDT. In this study, we present the synthesis and comprehensive characterization of a novel type I photosensitizer derived from a cyclometalated Ir(III)-rhodamine complex. Remarkably, the complex exhibits a shift in absorption and fluorescence, transitioning from "off" to "on" states in aprotic and protic solvents, respectively, contrary to initial expectations. Upon exposure to light, the complex demonstrates the effective generation of O2- and ·OH radicals via the type I mechanism. Additionally, it exhibits notable photodynamic antibacterial activity against both Gram-positive and Gram-negative bacteria, demonstrated through in vitro and in vivo experiments. This research offers valuable insights for the development of novel type I photosensitizers.

Objective: The present study was performed to evaluate the effect of ultrasound-guided erector spinae plane block (ESPB) on pain after laparoscopic transabdominal preperitoneal (TAPP) repair. Therefore, improved postoperative pain management is crucial for enhancing the overall patient experience and recovery. Methods: This prospective, double-blind, randomized controlled trial enrolled 40 male patients with a unilateral inguinal hernia at Xi'an Aerospace General Hospital from November 1, 2020, to February 1, 2021. Participants were assigned through a random number table at a 1:1 ratio to receive either ESPB with 20 ml 0.5% ropivacaine in the experimental group (Group E) or ESPB with 20 ml normal saline in the control group (Group C), with 20 cases in each group. The primary outcome was assessed using visual analogue scale (VAS) scores for exercise pain at 2h, 6h, 12h, 18h, and 24h postoperatively. Secondary outcomes included time lapses before patient-controlled intravenous analgesia (PCIA) use, intraoperative remifentanil usage, additional sufentanil, postoperative nalbuphine consumption, analgesic remedies at 24h postoperatively, and incidence of postoperative adverse events. Results: Group E provided more pain mitigation for patients than Group C, as evidenced by the significantly lower VAS scores during exercise pain at 2h (Group C: 1.95±1.19; Group E:4.00±1.38), 6h (Group C: 2.00±1.12; Group E:3.90±1.37), and 12h (Group C: 2.05±1.05; Group E:3.55±1.36) postoperatively (P < .05), and the pain mitigation for Group C was significant only at 18h and 24h postoperatively compared to at 2h postoperatively (P < .05). Group E resulted in significantly reduced intraoperative use of remifentanil and, additional sufentanil and postoperative nalbuphine consumption versus Group C (P < .05). Group E exhibited a better pain tolerance than Group C, as demonstrated by the longer time lapse before the use of PCIA (RR value=5.709, t=8.446, P < .05). Group C required more analgesic remedies within 24 h after surgery than Group E (P < .05). Group E did not increase the risk of postoperative adverse events, given the absence of statistical significance in the intergroup comparison (P > .05). Conclusion: Ultrasound-guided ESPB demonstrates notable benefits by decreasing intraoperative and postoperative anesthetic drug requirements, enhancing pain management, and elevating postoperative comfort and quality of life for patients. While acknowledging the study's limitations, it is crucial to highlight the potential clinical implications of these findings. The incorporation of ESPB with ropivacaine into postoperative pain management protocols could represent a significant advancement in clinical practice. The observed improvements in pain management and reduced reliance on anesthetic drugs may lead to more tailored and efficient postoperative care, potentially enhancing patient recovery experiences. Further research and practical implementation studies are warranted to fully elucidate the specific impact and optimal integration of ESPB with ropivacaine within broader clinical settings.

Block compressed sensing (BCS) is a promising method for resource-constrained image/video coding applications. However, the quantization of BCS measurements has posed a challenge, leading to significant quantization errors and encoding redundancy. In this paper, we propose a quantization method for BCS measurements using convolutional neural networks (CNN). The quantization process maps measurements to quantized data that follow a uniform distribution based on the measurements' distribution, which aims to maximize the amount of information carried by the quantized data. The dequantization process restores the quantized data to data that conform to the measurements' distribution. The restored data are then modified by the correlation information of the measurements drawn from the quantized data, with the goal of minimizing the quantization errors. The proposed method uses CNNs to construct quantization and dequantization processes, and the networks are trained jointly. The distribution parameters of each block are used as side information, which is quantized with 1 bit by the same method. Extensive experiments on four public datasets showed that, compared with uniform quantization and entropy coding, the proposed method can improve the PSNR by an average of 0.48 dB without using entropy coding when the compression bit rate is 0.1 bpp.

We report the numerical and experimental study of probe pulse deformation in a forward-pumped distributed Raman amplifier on a 40-km standard single mode fiber. Distributed Raman amplification can improve the range of OTDR-based sensing systems, but it could result in pulse deformation. A smaller Raman gain coefficient can be used to mitigate pulse deformation. The sensing performance can still be maintained by compensating for the decrease in the Raman gain coefficient by increasing the pump power. The tunability of the Raman gain coefficient and pump power levels are predicted while keeping the probe power below the modulation instability limit.

BACKGROUND: This investigation aimed to evaluate the impact of continuous pericapsular nerve group (PENG) block and continuous fascia iliac compartment block (FICB) on postoperative pain following total hip arthroplasty (THA). METHODS: This prospective, randomized, and controlled trial recruited a cohort of fifty-seven patients with unilateral femoral neck fractures from Xi'an Aerospace General Hospital in northwest China between July 2020 and November 2021. These patients were randomly assigned to two groups: the continuous PENG block group (PENG group, n = 29) and the continuous FICB group (FICB group, n = 28). Under ultrasound guidance, PENG block and FICB procedures were performed prior to spinal anesthesia, utilizing 20 ml of 0.25% ropivacaine for PENG block and 30 ml of 0.25% ropivacaine for FICB. Subsequently, a catheter was inserted. All study participants received a standardized postoperative multimodal analgesic regimen, including intravenous administration of 30 mg Ketorolac tromethamine every eight hours and patient-controlled neural analgesia (PCNA) after surgery. Numerical rating scale (NRS) scores at rest and during exercise were recorded at various time points: prior to block (T0), 30 min post-blockade (T1), and 6 h (T2), 12 h (T3), 24 h (T4), and 48 h (T5) postoperatively. Additional data collected encompassed postoperative quadriceps muscle strength, the time of initial ambulation after surgery, the number of effective PCNA activations, rescue analgesia requirements, and occurrences of adverse events (such as nausea and vomiting, hematoma, infection, catheter detachment, or displacement) within 48 h following surgery. RESULTS: In the PENG group, the resting NRS pain scores exhibited lower values at T1, T4, and T5 than those at T0. Furthermore, exercise NRS pain scores at T1-T5 were lower in the PENG group than in the FICB group. Similarly, during the same postoperative period, the PENG group demonstrated enhanced quadriceps strength on the affected side compared to the FICB group. Additionally, the PENG group displayed earlier postoperative ambulation and reduced occurrences of effective PCNA activations and rescue analgesia requirements compared to the FICB group. CONCLUSION: Continuous PENG block exhibited superior analgesic efficacy after THA compared to continuous FICB, promoting recovery of quadriceps strength on the affected side and facilitating early postoperative ambulation. TRIAL REGISTRATION: This clinical trial was registered in the China Clinical Trials Center ( http://www.chictr.org.cn ) on 20/07/2020, with the registration number ChiCTR2000034821.

To explore the function of phosphatidylinositol 4-phosphate 5-kinase (PIP5K) in tomatoes, members of the tomato PIP5K family were identified and characterized using bioinformatic methods, and their expression patterns were also analyzed under salt stress and in different tissues. Twenty-one PIP5K members-namely, SlPIP5K1-SlPIP5K21-were identified from ten chromosomes, and these were divided into three groups according to a phylogenetic analysis. Further bioinformatic analysis showed four pairs of collinear relationships and fragment replication events among the SlPIP5K family members. To understand the possible roles of the SlPIP5Ks, a cis-acting element analysis was conducted, which indicated that tomato PIP5Ks could be associated with plant growth, hormones, and stress responses. We further validated the results of the in silico analysis by integrating RNA-seq and qRT-PCR techniques for salt- and hormone-treated tomato plants. Our results showed that SlPIP5K genes exhibited tissue- and treatment-specific patterns, and some of the SlPIP5Ks exhibited significantly altered expressions after our treatments, suggesting that they might be involved in these stresses. We selected one of the SlPIP5Ks that responded to our treatments, SlPIP5K2, to further understand its subcellular localization. Our results showed that SlPIP5K2 was located on the membrane. This study lays a foundation for the analysis of the biological functions of the tomato SlPIP5K genes and can also provide a theoretical basis for the selection and breeding of new tomato varieties and germplasm innovation, especially under salt stress.

PURPOSE: To develop a novel method to achieve fast, high-resolution, 3D multi-TE 1 H-MRSI of the brain. METHODS: A new multi-TE MRSI acquisition strategy was developed that integrates slab selective excitation with adiabatic refocusing for better volume coverage, rapid spatiospectral encoding, sparse multi-TE sampling, and interleaved water navigators for field mapping and calibration. Special data processing strategies were developed to interpolate the sparsely sampled data, remove nuisance signals, and reconstruct multi-TE spatiospectral distributions with high SNR. Phantom and in vivo experiments have been carried out to demonstrate the capability of the proposed method. RESULTS: The proposed acquisition can produce multi-TE 1 H-MRSI data with three TEs at a nominal spatial resolution of 3.4 × 3.4 × 5.3 mm3 in around 20 min. High-SNR brain metabolite spatiospectral reconstructions can be obtained from both a metabolite phantom and in vivo experiments by the proposed method. CONCLUSION: High-resolution, 3D multi-TE 1 H-MRSI of the brain can be achieved within clinically feasible time. This capability, with further optimizations, could be translated to clinical applications and neuroscience studies where simultaneously mapping metabolites and neurotransmitters and TE-dependent molecular spectral changes are of interest.

RESUMO

Higher-order mode converters that work over a broad wavelength range are needed for various applications. A new, to the best of our knowledge, simple and cost-effective LP02 mode converter is fabricated by tapering a bundle of single-mode fibers. The device excites the LP02 mode in a four-mode step index fiber with a mode purity higher than 10 dB. The polarization-dependent cross talk of the device is measured using the S2 technique. The LP02 mode selectivity of the device is measured over the entire C and L bands by selectively launching different modes into the device using a spatial light modulator.

This research aimed to analyze the regulatory effect of chicken telomerase reverse transcriptase (chTERT) on the Wnt/ß-catenin signaling pathway and its effect on the tumorigenicity of avian leukosis virus subgroup J (ALV-J) through in vivo experiments. The chTERT eukaryotic expression plasmid and its recombinant lentivirus particles were constructed for in vivo transfection of chTERT to analyze the effect of chTERT continuously overexpressed in chickens on the tumorigenicity of ALV-J. During 156 days of the artificial ALV-J tumor-inducing process, 7 solid tumors developed in 3 chickens in the chTERT-overexpression group (n = 26*2) and no tumors developed in the control group (n = 26*2). Another 18 tumors induced by ALV-J were confirmed and collected from breeding poultry farms. And we confirmed that chTERT was significantly highly expressed in ALV-J tumors. The ELISA data suggested that the protein levels of ß-catenin and c-Myc in the chicken plasma of the chTERT-overexpressing group with ALV-J infected were consistently and significantly higher than those of the control group. Compared with that of the tumor-adjacent tissues, the activity of the Wnt/ß-catenin signaling pathway and expression of the c-Myc was significantly increased in ALV-J tumors. And the percentage of apoptosis in ALV-J tumors significantly lower than that in tumor-adjacent tissues. Immunohistochemistry, Western blot and RT-qPCR suggested that the replication level of ALV-J in tumors was significantly higher than that in tumor-adjacent tissues. This study suggests that chTERT plays a critical role in the tumorigenicity of ALV-J by enhancing the Wnt/ß-catenin signaling pathway, which will contribute to further elucidating the tumor-inducing mechanism of ALV-J.

BACKGROUND: The restraint water immersion stress (RWIS) model includes both psychological and physical stimulation, which may lead to gastrointestinal disorders and cause gastric mucosal damage. The ventrolateral periaqueductal gray (VLPAG) contributes to gastrointestinal function, but whether it is involved in RWIS-induced gastric mucosal damage has not yet been reported. METHODS: The expression of glial fibrillary acidic protein, neuronal c-Fos, and phosphorylated extracellular signal regulated kinase 1/2 in the VLPAG after RWIS was assessed using western blotting and immunocytochemical staining methods. Lateral ventricle injection of astrocytic toxin L-a-aminoadipate and treatment with extracellular signal-regulated kinase (ERK)1/2 signaling pathway inhibitor PD98059 were further used to study protein expression and distribution in the VLPAG after RWIS. RESULTS: The expression of c-Fos, glial fibrillary acidic protein, and phosphorylated extracellular signal regulated kinase 1/2 in the VLPAG significantly increased following RWIS and peaked at 1 hour after RWIS. Lateral ventricle injection of the astrocytic toxin L-a-aminoadipate significantly alleviated gastric mucosal injury and decreased the activation of neurons and astrocytes. Treatment with the ERK1/2 signaling pathway inhibitor PD98059 obviously suppressed gastric mucosal damage as well as the RWIS-induced activation of neurons and astrocytes in the VLPAG. CONCLUSIONS: These results suggested that activation of VLPAG neurons and astrocytes induced by RWIS through the ERK1/2 signaling pathway may play a critical role in RWIS-induced gastric mucosa damage.

Although cocrystallization has provided a promising platform to develop new organic optoelectronic materials, it is still a big challenge to purposely design and achieve specific optoelectronic properties. Herein, a series of mixed-stacking cocrystals (TMFA, TMCA, and TMTQ) were designed and synthesized, and the regulatory effects of the acceptors on the co-assembly behavior, charge-transfer nature, energy-level structures, and optoelectronic characteristics were systematically investigated. The results demonstrate that it is feasible to achieve effective charge-transport tuning and photoresponse switching by carefully regulating the intermolecular charge transfer and energy orbitals. The inherent mechanisms underlying the change in these optoelectronic behaviors were analyzed in depth and elucidated to provide clear guidelines for future development of new optoelectronic materials. In addition, due to the excellent photoresponsive characteristics of TMCA, TMCA-based phototransistors were investigated with varying light wavelength and optical power, and TMCA shows the best performance among all reported cocrystals under UV illumination.

Molecular dynamics simulation (MD) technology can be used to simulate and study the physicochemical properties of polymer materials on the basis of material data obtained in traditional experiments. In this study, we use MD to construct models of crosslinked carbon nanotubes/epichlorohydrin rubber composites with different carbon nanotube diameters and study the effect of CNT tube diameter on crosslinked ECO. The results show that with the increase in CNT tube diameter, the contact area between the CNTs and rubber matrix increases, the interaction force is enhanced, the free volume fraction of rubber matrix decreases by 21.36%, the glass transition temperature increases by 6.5%, the mean square displacement decreases, the radius of gyration decreases by 3.18 Å, and the radial distribution function of the C-atom group and the H-atom group in the system gradually decreases. The binding energy between the two is elevated, which in turn verifies the enhancement of the interaction force.

Lung adenocarcinoma, a chronic non-small cell lung cancer, needs to be detected early. Tumor gene expression data analysis is effective for early detection, yet its challenges lie in a small sample size, high dimensionality, and multi-noise characteristics. In this study, we propose a lung adenocarcinoma convolutional neural network (LATCNN), a deep learning model tailored for accurate lung adenocarcinoma prediction and identification of key genes. During the feature selection stage, we introduce a hybrid algorithm. Initially, the fast correlation-based filter (FCBF) algorithm swiftly filters out irrelevant features, followed by applying the k-means-synthetic minority over-sampling technique (k-means-SMOTE) method to address category imbalance. Subsequently, we enhance the particle swarm optimization (PSO) algorithm by incorporating fast-decay dynamic inertia weights and utilizing the classification and regression tree (CART) as the fitness function for the second stage of feature selection, aiming to further eliminate redundant features. In the classifier construction stage, we present an attention convolutional neural network (atCNN) that incorporates an attention mechanism. This improved model conducts feature selection post lung adenocarcinoma gene expression data analysis for classification and prediction. The results show that LATCNN effectively reduces the feature dimensions and accurately identifies 12 key genes with accuracy, recall, F1 score, and MCC of 99.70%, 99.33%, 99.98%, and 98.67%, respectively. These performance metrics surpass those of other comparative models, highlighting the significance of this research for advancing lung adenocarcinoma treatment.

Solid hygroscopic materials are extensively utilized in diverse fields, including adsorption heat transfer, adsorption heat storage, atmospheric water harvesting (AWH), and air conditioning dehumidification. The efficacy and energy efficiency of these materials in practical applications are significantly influenced by their adsorption and desorption properties. Yet, the introduction of inorganic salts to boost adsorption performance can result in issues like salt leakage. In this research, we prepared a polyacrylamide hydrogel through free radical polymerization, and its water-absorbing capabilities were improved by incorporating the hygroscopic salt lithium chloride. We compared it to a salt-based porous adsorbent, AlFum-LiCl, which also exhibited strong water adsorption properties and the potential for large-scale production. While AlFum-LiCl suffered from limited pores and salt leakage during high water uptake, the optimized PAM-LiCl displayed superior water sorption capabilities, showing no salt leakage even at water uptake of up to 3.5 g/g. At 25 °C, PAM-LiCl achieved equilibrium water uptake of 1.26 g/g at 30% RH and 3.15 g/g at 75% RH. In this context, utilizing 20 g of PAM-LiCl for the AWH experiment yielded daily water outputs of 8.34 L/kg at 30% RH and 16.86 L/kg at 75% RH. The salt-optimized PAM-LiCl hydrogel offers the benefit of application in higher relative humidity environments without the risk of deliquescence, underscoring its promise for atmospheric water harvesting.

OBJECTIVE: To develop a novel multi-TE MR spectroscopic imaging (MRSI) approach to enable label-free, simultaneous, high-resolution mapping of several molecules and their biophysical parameters in the brain. METHODS: The proposed method uniquely integrated an augmented molecular-component-specific subspace model for multi-TE 1H-MRSI signals, an estimation-theoretic experiment optimization (nonuniform TE selection) for molecule separation and parameter estimation, a physics-driven subspace learning strategy for spatiospectral reconstruction and molecular quantification, and a new accelerated multi-TE MRSI acquisition for generating high-resolution data in clinically relevant times. Numerical studies, phantom and in vivo experiments were conducted to validate the optimized experiment design and demonstrate the imaging capability offered by the proposed method. RESULTS: The proposed TE optimization improved estimation of metabolites, neurotransmitters and their T2's over conventional TE choices, e.g., reducing variances of neurotransmitter concentration by  âˆ¼  40% and metabolite T2 by  âˆ¼  60%. Simultaneous metabolite and neurotransmitter mapping of the brain can be achieved at a nominal resolution of 3.4 × 3.4 × 6.4 mm 3. High-resolution, 3D metabolite T2 mapping was made possible for the first time. The translational potential of the proposed method was demonstrated by mapping biochemical abnormality in a post-traumatic epilepsy (PTE) patient. CONCLUSION: The feasibility for high-resolution mapping of metabolites/neurotransmitters and metabolite T2's within clinically relevant time was demonstrated. We expect our method to offer richer information for revealing and understanding metabolic alterations in neurological diseases. SIGNIFICANCE: A novel multi-TE MRSI approach was presented that enhanced the technological capability of multi-parametric molecular imaging of the brain. The proposed method presents new technology development and application opportunities for providing richer molecular level information to uncover and comprehend metabolic changes relevant in various neurological applications.

Background: Emerging evidence has demonstrated the impact of psychological stress on intestinal microbiota, however, the precise mechanisms are not fully understood. Enteric glia, a unique type of peripheral glia found within the enteric nervous system (ENS), play an active role in enteric neural circuits and have profound effects on gut functions. In the present study, we tested the hypothesis that enteric glia are involved in the alterations in the intestinal microflora and barrier induced by chronic water-avoidance stress (WAS) in the gut. Methods and results: Western blotting and immunohistochemical (IHC) staining were used to examine the expression of glial fibrillary acidic protein (GFAP), nitric oxide synthetase (NOS) and choline acety1transferase (ChAT) in colon tissues. 16S rDNA sequencing was performed to analyse the composition of the intestinal microbiota in rats. Changes in the tight junction proteins Occludin, Claudin1 and proliferating cell nuclear antigen (PCNA) in the colon tissues were detected after WAS. The abundance of Firmicutes, Proteobacteria, Lactobacillus and Lachnospiraceae_NK4A136 decreased significantly, whereas the abundance of Actinobacteria, Ruminococcaceae_UCG-005 and Christensenellaceae-R-7 increased significantly in stressed rats. Meanwhile, the expression of Occludin, Claudin1 and PCNA significantly decreased after WAS. Treatment with L-A-aminohexanedioic acid (L-AA), a gliotoxin that blunts astrocytic function, obviously decreased the abundance of Actinobacteria, Ruminococcaceae_UCG-005 and Christensenel-laceae_R-7 in stressed rats and significantly increased the abundance of Proteobacteria, Lactobacillus and Lachnospiraceae_NK4A136. In addition, the protein expression of colon Occludin, Claudin1, and PCNA increased after intraperitoneal injection of L-AA. Furthermore, the expression level of NOS in colon tissues was significantly decreased, whereas that of ChAT was significantly increased following L-AA treatment. Conclusions: Our results showed that enteric glial cells may contribute to WAS-induced changes in the intestinal microbiota and barrier function by modulating the activity of NOS and cholinergic neurones in the ENS.

Previous researches seldom studied the selection of buffer distance between geological hazards (positive samples) and non-geological hazards (negative samples), and its reasonable selection plays a very important role in improving the accuracy of susceptibility zoning, protecting the environment and reducing the cost of hazard management. Based on GIS technology and random forest (RF) and frequency-ratio random forest (FR-RF) models, this study innovatively explored the influence of randomly selected non-geological hazard samples outside different buffer distances on the susceptibility evaluation results, with buffer distances of 100 m, 500 m, 1000 m and 2000 m in sequence. The results show that through the confusion matrix and ROC curve test, the accuracy of the model increases first and then decreases with the increase of buffer distance. Both RF and FR-RF models have the highest accuracy when the buffer distance is 1000 m, and the accuracy of the RF model is generally higher than that of the FR-RF model under the same buffer distance. Similar attribute values of positive samples and randomly selected negative samples or "extreme" attribute values of negative samples are the main reasons for the differences in evaluation results of different buffer distances. According to the weight analysis of causative factors, the distance from road, the distance from river and the normalized vegetation index (NDVI) are the main factors affecting the occurrence of hazards. The high and very high susceptibility areas in the study area are mainly distributed on both sides of roads and water systems, which are the key areas for hazard prevention and reduction. The HMC of RF-1000m decreased by 3.55% on average compared with other models. The results of this study improve the accuracy of geological hazard susceptibility assessment, maintain the safety of ecological environment, and provide a scientific basis for the selection of buffer distance index in local and surrounding areas in the future.

Lasso peptides, natural biological microcins composed of small molecules, have demonstrated efficient bactericidal activity. However, a single lasso peptide is characterized by a narrow and targeted bactericidal spectrum. In this study, a chitosan (CN) derivative-based polymer nanomaterial incorporating three lasso peptides (MccY, MccJ25, and Klebsidin) was designed and synthesized to broaden its antimicrobial spectrum. To enhance resistance to acid and alkali conditions, arginine was appended to the terminus of conjugates, resulting in Chitosan-Lasso-Peptides-Arg (CN-LPs-Arg), and the nanomaterial biocompatibility and bactericidal activity were characterized. Chemical stability test results demonstrate that CN-LPs-Arg effectively buffered the acid-base effect of the compound. Notably, CN-LPs-Arg extended the antimicrobial spectrum of Gram-negative and Gram-positive strains including Klebsiella, Salmonella, and Staphylococcus (MIC = 0.01-1.0 µM). CN-LPs-Arg exerts its destructive effects on bacteria via a series of mechanisms; it adheres to and then penetrates the membrane, causes rupture, and leads to bacterial death. Transcriptomic data revealed that CN-LPs-Arg produced a distinct inhibitory effect on ribosomal protein subunits synthesis pathways and membrane metabolic inhibition. Furthermore, CN-LPs-Arg was nontoxic to cells and exhibited excellent biocompatibility. CN-LPs-Arg reduced bacterial burden in organs and the levels of inflammatory factors IL-6, IL-8, and TNF-α in tissues of mice with acute bacterial infections. Furthermore, it promoted the recovery of Klebsiella-infected C57BL/6 mice, demonstrating a favorable therapeutic effect in vivo. The multilasso peptide-based synergistic nanocomposite of CN-LPs-Arg exhibited high stability as a broad-spectrum antimicrobial agent with potential for combined antibacterial therapy and utilization in the fields of food, biomedicine, and public health.
