Functional transcranial Doppler sonography (fTCD) is a time- and cost-effective, non-invasive approach to determining real time hemispheric lateralization and is well-suited for repetitive study designs comprising multiple days. To date, no study has examined the reproducibility of the direction and degree (strength) of lateralization during word fluency (WF) over multiple, consecutive sessions within a single person, although there are many studies of lateralization during language processing. Moreover, there is conflicting evidence as to whether there is a relationship between the degree of laterality and the word fluency performance. In this study, one right-handed male (aged 24 years) completed a total of seven examination sessions in the time span of 10 days. Each session comprised multiple phonological and semantic WF tasks. The maximum difference of relative cerebral blood flow velocity (CBFV) changes between the left and right middle cerebral artery (MCA) during WF was defined as the Lateralization Index (LI). The word-fluency performance and the LIs were used in a linear regression model to detect relative changes in the direction and degree of lateralization during repetitive WF tasks. The reproducibility of the direction of language-related lateralization is very stable over multiple sessions within this single person and the processed LIs were left-lateralized in every session for both WF tasks. In addition, performance during phonological WF could significantly predict the variability in the degree of lateralization. This result could not be confirmed for the semantic WF task. The results of this pilot study support the usage of fTCD as a reliable method for examining lateralization patterns, especially in longitudinal study designs. They also provide evidence for the notion that performance in WF tasks can be related to the degree of lateralization, at least intra-individually.

BACKGROUND: The goal of this study was to investigate whether the combination of semantic feature analysis (SFA) and transcranial direct current stimulation (tDCS) is effective in treating word retrieval in the semantic variant of primary progressive aphasia (svPPA) and how long the potential effects last. METHODS: A 56-year-old woman diagnosed with frontotemporal dementia (FTD) and svPPA participated in this longitudinal single-subject design. A total of four 2-week stimulation phases were conducted over a 14-month period, each of which was started depending on the participant's language performance. Follow-up testing was conducted shortly after the stimulation period, approximately 2 weeks, and approximately 4 weeks thereafter. RESULTS: Significant improvement in word retrieval occurred after SFA and tDCS therapy. Two weeks after the end of each stimulation phase, approx. 80% of the trained words could be named correctly. For the untrained words, also significantly more words were correctly named at follow-ups compared to the baseline. Furthermore, the Boston Naming Test (BNT) demonstrated a significant increase in naming performance and showed that phonological cues facilitated word retrieval compared to semantic cues. CONCLUSION: The combination of SFA and tDCS was able to counteract the expected language deterioration of a participant with svPPA. This effect increased until approximately 2 weeks after each intervention. In addition, a generalization of the effect to untrained words was shown.

The diversity of cellular actin functions is attained by the activation of actin nucleator complexes, which initiate the polymerization of actin monomers into a helical double-stranded filament at defined subcellular compartments. Next to actin functions at the cell membrane, including different forms of membrane protrusions and invaginations, actin dynamics at intracellular membranes has recently become a research focus. Experiments addressing the vesicle-associated Spir WH2 domain containing actin nucleators have provided novel mechanistic insights into the function of actin dynamics at intracellular membranes. Spir proteins are targeted by a modified FYVE zinc finger motif toward endosomal and vesicle membranes, where they interact and cooperate with the distinct nucleators of the FMN subfamily of formins in the nucleation of actin filaments. The function of the Spir/formin actin nucleator complex is closely related to the Rab11 small G protein, which is a key regulator of recycling and exocytic transport processes. Together with the actin motor protein and Rab11 effector myosin Vb, Spir/formin nucleated actin filaments mediate actin-dependent vesicle transport processes. Drosophila and mouse genetic studies as well as cell biology experiments point toward an important role of the Spir/formin complex in oocyte maturation and in the structure and signaling of the nervous system.

Progressive supranuclear palsy (PSP) is an atypical Parkinsonian disorder which results in deterioration of motor and cognitive skills, including language disorders such as impaired word retrieval. While there is evidence of successful use of tDCS to improve word fluency in PSP, little is known about the effectiveness of brain stimulation for word retrieval in sentence context. Therefore, we investigated whether tDCS reduces sentence completion time in PSP patients. In this sham-controlled, triple-blinded crossover study, anodal tDCS (atDCS) was applied over the left Broca's area at 2 mA for 20 min (n = 23). In contrast to patients with multiple system atrophy (MSA), also an atypical Parkinsonian disorder, and healthy elderlies, sentence completion improved in PSP patients when tDCS was applied. The improvement in word fluency reported in previous studies using other electrode positions was not replicated. By using atDCS of the left Broca's area, we were able to demonstrate a difference between the two movement disorders. The obtained insight could be helpful to improve language therapy of these disorders.

Introduction: In addition to age-related changes in language, hemispheric lateralization of language functions steadily declines with age. Also, performance on word fluency tasks declines and is sensitive to the expression of dementia-related changes. The aim of this study is to evaluate the effect of anodal tDCS combined with a word fluency training on language lateralization and word fluency performance in healthy elderly subjects and in persons with mild cognitive impairment (MCI). Methods: The effect of anodal tDCS over the left inferio frontal gyrus (IFG) was measured in a group of healthy elderly up to the age of 67 years (YG, Ø = 63.9 ± 3.02), a group of healthy elderly aged 68 years and older (OG, Ø = 78.1, ± 4.85), and a group of patients with MCI (Ø = 81.18, ± 7.35) by comparing performance in phonological and semantic word fluency tasks before and after 3 days of tDCS. Half of the experimental participants received sham stimulation. In addition, language lateralization was determined using a lateralization index (LI) measured with functional transcranial Doppler sonography (fTCD) before and after the stimulation period. Results: Anodal tDCS was associated with significantly higher scores in phonological but not semantic word fluency in both YG and OG. In MCI patients, no difference was measured between the tDCS and sham groups in either word fluency task. fTCD showed significantly increased left lateralization in all three groups after the training phase. However, this effect was independent of tDCS and the degree of lateralization could not be predicted by word fluency performance in any of the groups. Discussion: Phonological word fluency can be increased with atDCS in healthy elderly people by stimulating the IFG in a 3-day training. When cognitive decline has reached a certain stage, as is the case with MCI, this paradigm does not seem to be effective enough.

In this study, heat-treated and multisurface engineered DIN 1.2367 tool steel was subjected to room and elevated temperature wear tests, and the effect of nitriding on its tribological behavior was investigated. CrN, AlTiN, and CrN/AlTiN coatings with a total thickness of 2 µm were obtained by arc cathodic physical vapor deposition on conventional heat-treated and gas-nitrided steels. The white layer formed during nitriding was removed, and a diffusion layer (100 µm) was achieved in the cross section of the steel having a tempered martensitic matrix. The highest surface hardness was attained with an integral coating (CrN/AlTiN), and surface hardness increased even more after nitriding due to the formation of a multicomponent ceramic layer on top of the diffusion layer. The room temperature wear tests performed against an alumina counterpart revealed that (i) CrN/AlTiN-coated steel had the highest friction coefficient of 0.26, which further increased to 0.33 by nitriding due to the increase in shear strength, and that (ii) with increasing surface hardness, the specific wear rates (W) of the heat-treated and coated steels could be ranked as follows: WCrN/AlTiN < WAlTiN < WCrN. The wear rates decreased when nitriding was carried out prior to coating. In order to simulate the aluminum extrusion conditions, hot wear behavior of the surfaces against AA6080 alloy at 450 °C was investigated. The hot wear tests revealed that (i) high friction coefficients were reached due to the adhesive characteristic of aluminum to the surfaces, (ii) the nitrided and CrN/AlTiN-coated sample exhibited the lowest wear rate among all studied surfaces, and (iii) the film damage on the worn surfaces mostly occurred in the form of droplet delamination.

For over two decades, vascular stents have been widely used to treat clogged vessels, serving as a scaffold to enlarge the narrowed lumen and recover the arterial flow area. High-purity oligocrystalline austenitic steel is usually applied for the production of stents. Despite the popularity and benefit of stenting, it still may cause serious clinical adverse issues, such as in-stent restenosis and stent fracture. Therefore, the study of the mechanical properties of stents and in particular the prediction of their life cycles are in the focus of materials research. In our contribution, within the finite element method, a two-scale model of crack initiation in the microstructure of stents is elaborated. The approach is developed on the basis of the physically based Tanaka-Mura model (TMM), considering the evolution of shear bands during the crack initiation phase. The model allows for the analysis of the microstructure with respect to the life cycles of real materials. The effects of different loading conditions, grain orientation, and thickness of the specimen on Wöhler curves were analysed. It was found that the microstructural features of oligocrystals are very sensitive to different loading conditions with respect to their fatigue behaviour and play a major role in fatigue crack initiation. Different grain-orientation distributions result in qualitative and quantitative differences in stress distribution and in the number of cycles for crack initiation. It was found that presence of a neutral zone in the cut-out of the microstructure under three-point-bending loading conditions changes the qualitative and quantitative patterns of stress distribution and affects the number of cycles for crack initiation. It was found that under both tensile and bending loading conditions, thicker specimens require more cycles for crack initiation. The Wöhler curves for crack initiation in oligocrystalline microstructures of stents could be compared with the ones in the experiment, taking into account that for high cyclic fatigue (HCF), typically, more than 70% of the cycles refer to crack initiation. The developed numerical tools could be used for the material design of stents.

Repulsive guidance molecule A (RGMa) is an inhibitor of neuronal growth and survival which is upregulated in the damaged central nervous system following acute spinal cord injury (SCI), traumatic brain injury, acute ischemic stroke (AIS), and other neuropathological conditions. Neutralization of RGMa is neuroprotective and promotes neuroplasticity in several preclinical models of neurodegeneration and injury including multiple sclerosis, AIS, and SCI. Given the limitations of current treatments for AIS due to narrow time windows to intervention (TTI), and restrictive patient selection criteria, there is significant unmet need for therapeutic agents that enable tissue survival and repair following acute ischemic damage for a broader population of stroke patients. In this preclinical study, we evaluated whether elezanumab, a human anti-RGMa monoclonal antibody, could improve neuromotor function and modulate neuroinflammatory cell activation following AIS with delayed intervention times up to 24 h using a rabbit embolic permanent middle cerebral artery occlusion model (pMCAO). In two replicate 28-day pMCAO studies, weekly intravenous infusions of elezanumab, over a range of doses and TTIs of 6 and 24 h after stroke, significantly improved neuromotor function in both pMCAO studies when first administered 6 h after stroke. All elezanumab treatment groups, including the 24 h TTI group, had significantly less neuroinflammation as assessed by microglial and astrocyte activation. The novel mechanism of action and potential for expanding TTI in human AIS make elezanumab distinct from current acute reperfusion therapies, and support evaluation in clinical trials of acute CNS damage to determine optimal dose and TTI in humans. Healthy elderly people often experience a subjective loss of daily memory performance whereas an objective decrease in memory performance is often observed in patients with memory complaints. In this paper, we investigate the influence of a single session of "anodal" transcranial direct current stimulation (a-tDCS) on auditory word recognition performance in a decision time experiment. Three groups of participants (>64 years of age) with and without memory complaints underwent a word recognition task, in which they had to recognize words previously encoded among several distractors (semantically or phonologically related words) via a button press. In this double-blinded study, the participants completed two sessions (sham/a-tDCS), counterbalanced between subjects with a washout period of at least 10 days. Twenty minutes of 1.5 mA a-tDCS was applied over the left temporal cortex during the memorizing and decision phases. Overall, our results demonstrated that the participants, independent of their memory performance, were faster in word recognition during a-tDCS. As expected, older participants with memory complaints recognized significantly less words correctly compared to other participants. However, tDCS did not have a beneficial effect on the extent of successful word recognition. These results suggest a general effect of a single session of a-tDCS over the left temporal cortex, with participants becoming faster in their word recognition, thus having easier access to encoded words.

Regarding the embodiment of language processing in adults, there is evidence of a close connection between sensorimotor brain areas and brain areas relevant to the processing of action verbs. This thesis is hotly debated and has therefore been thoroughly studied in adults. However, there are still questions concerning its development in children. The present study deals with the processing of action verbs in concrete and abstract sentences in 60 eleven-year-olds using a decision time paradigm. Sixty-five children mirrored arm movements or sat still and rated the semantic plausibility of sentences. The data of the current study suggest that eleven-year-olds are likely to misunderstand the meaning of action verbs in abstract contexts. Their decision times were faster and their error rates for action verbs in concrete sentences were lower. However, the gender of the children had a significant influence on the decision time and the number of errors, especially when processing abstract sentences. Females were more likely to benefit from an arm movement before the decision, while males were better if they sat still beforehand. Overall, children made quite a few errors when assessing the plausibility of sentences, but the female participants more often gave plausibility assessments that deviated from our expectations, especially when processing abstract sentences. It can be assumed that the embodiment of language processing plays some role in 11-year-old children, but is not yet as mature as it is in adults. Especially with regard to the processing of abstract language, the embodied system still has to change and mature in the course of child development.

Impulse Friction Stir Welding (IFSW) was utilized to join 6082-T6 alloy plates at various impulse frequencies. A distinctive feature of IFSW is the generation of mechanical impulses that enhances the forging action of the tool, and thereby, alters the weld microstructure. The microstructural evolution in the Stir Zone (SZ) with special focus on the strengthening precipitation behavior, and overall mechanical properties of the IFSW joints have been investigated. It was demonstrated that the strengthening ß″ precipitates reprecipitated in the SZ of the IFSW joints during natural aging. In contrast, no precipitates were found in the SZ of the Friction Stir Welding (FSW) weld. Partial reversion of ß″ after IFSW is supposed to occur due to more developed subgrain network and higher dislocation density introduced by impulses that accelerated precipitation kinetics. Dynamic recrystallisation was facilitated by impulses resulting in a fine, homogeneous structure. There was no significant difference between the microhardness in the SZ, tensile and yield strength of the FSW and IFSW joints. However, the application of impulses demonstrated the smoothing of the hardness reduction in the transition region at the advancing side. The shift of the fracture location from the Heat-Affected Zone (HAZ) by FSW to the SZ as well as higher elongation of the joints by IFSW of lower frequencies could be related to the grain refinement and the change of the grain orientation.

This paper describes the microstructure and properties of titanium-based composites obtained as a result of a reactive spark plasma sintering of a mixture of titanium and nanostructured (Ti,Mo)C-type carbide in a carbon shell. Composites with different ceramic addition mass percentage (10 and 20 wt %) were produced. Effect of content of elemental carbon covering nc-(Ti,Mo)C reinforcing phase particles on the microstructure, mechanical, tribological, and corrosion properties of the titanium-based composites was investigated. The microstructural evolution, mechanical properties, and tribological behavior of the Ti + (Ti,Mo)C/C composites were evaluated using X-ray diffraction (XRD), scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDX), electron backscatter diffraction analysis (EBSD), X-ray photoelectron spectroscopy (XPS), 3D confocal laser scanning microscopy, nanoindentation, and ball-on-disk wear test. Moreover, corrosion resistance in a 3.5 wt % NaCl solution at RT were also investigated. It was found that the carbon content affected the tested properties. With the increase of carbon content from ca. 3 to 40 wt % in the (Ti,Mo)C/C reinforcing phase, an increase in the Young's modulus, hardness, and fracture toughness of spark plasma sintered composites was observed. The results of abrasive and corrosive resistance tests were presented and compared with experimental data obtained for cp-Ti and Ti-6Al-4V alloy without the reinforcing phase. Moreover, it was found that an increase in the percentage of carbon increased the resistance to abrasive wear and to electrochemical corrosion of composites, measured by the relatively lower values of the friction coefficient and volume of wear and higher values of resistance polarization. This resistance results from the fact that a stable of TiO2 layer doped with MoO3 is formed on the surface of the composites. The results of experimental studies on the composites were compared with those obtained for cp-Ti and Ti-6Al-4V alloy without the reinforcing phase.

OBJECTIVE: Impaired insulin metabolism has been implicated in migraine. However, to date only some putative effects, especially regarding the involvement of adipocytokines and glucagon-like peptides (GLPs), have been described. The aim of the present study was to investigate adipocytokines and GLPs in non-obese female migraineurs. METHODS: Various parameters of the insulin metabolism and body measurements were determined in 84 non-obese female subjects. RESULTS: We found highly significantly increased insulin levels with an odds ratio of 10.62 for migraine. Leptin and GLP-2 levels were also increased and correlated with insulin. Logistic regression analysis of leptin and GLP-2 revealed odds ratios of 3.79 and 4.26 for migraine, respectively, when comparing the lowest with the highest quartile of the test variable in the complete study cohort. DISCUSSION: We show that non-obese female migraineurs suffer from hyperinsulinemia, which is associated with elevated leptin and GLP-2 levels. Increased leptin and GLP-2 are risk factors for migraine. Our data suggest that migraine is associated with a higher risk for insulin resistance and its clinical consequences.

The microstructural and functional behavior of TiNi-based wires with a silver content of 0-1.5 at.% was evaluated. The concentration range for Ag doping determined for the TiNi wires with potential for the medical industry was 0-0.2 at.%. Microstructure analysis of TiNi wires with different silver contents at room temperature indicated a multiphase structural state. Various internal structures with tangled grain boundaries were formed by intense plastic deformation. The nanocrystalline structure and phase state of wire with the minimum silver content (0.1 at.% Ag) provide full shape recovery, the greatest reversible strain, and optimal strength and ductility. TiNi ingots with a high Ag content (0.5-1.5 at.%) cracked under minimum load due to excess silver that crystallized along the grain boundaries and broke cohesion bonds between the TiNi grains.

Porous TiNi alloys fabricated by self-propagating high-temperature synthesis (SHS) are biomaterials designed for medical application in substituting tissue lesions and they were clinically deployed more than 30 years ago. The SHS process, as a very fast and economically justified route of powder metallurgy, has distinctive features which impart special attributes to the resultant implant, facilitating its integration in terms of bio-mechanical/chemical compatibility. On the phenomenological level, the fact of high biocompatibility of porous SHS TiNi (PTN) material in vivo has been recognized and is not in dispute presently, but the rationale is somewhat disputable. The features of the SHS TiNi process led to a multifarious intermetallic Ti4Ni2(O,N,C)-based constituents in the amorphous-nanocrystalline superficial layer which entirely conceals the matrix and enhances the corrosion resistance of the unwrought alloy. In the current article, we briefly explore issues of the high biocompatibility level on which additional studies could be carried out, as well as recent progress and key fields of clinical application, yet allowing innovative solutions.

According to the Standards of the World Marrow Donor Association (WMDA), unrelated stem cell donor registries and donor centers are responsible for compliance of their collection and apheresis centers with these Standards. To ensure high stem cell product quality and high standards for safety and satisfaction of voluntary unrelated stem cell donors, we here present guidelines for audits of collection and apheresis centers that can be used by new and established donor registries, as well as by collection centers in preparation of audits. We define the general requirements and recommendations for collaboration with the collection and apheresis centers and define critical procedures for the collection of the stem cell product, such as information session, medical assessment, product collection, quality controls, product handover for transportation, and donor follow-up. The specific guidelines are accompanied by detailed checklists and forms that can be found in Supplementary Information and may be used during an initial or follow-up on-site or paper-based audit.

Frequency analyses of EEG data yield large data sets, which are high-dimensional and have to be evaluated statistically without a large number of false positive statements. There exist several methods to deal with this problem in multiple comparisons. Knowing the number of true hypotheses increases the power of some multiple test procedures, however the number of true hypotheses is unknown, in general, and must be estimated. In this paper, we derive two new multiple test procedures by using an upper bound for the number of true hypotheses. Our first procedure controls the generalized family-wise error rate, and thus is an improvement of the step-down procedure of Hommel and Hoffmann [Hommel G., Hoffmann T. Controlled uncertainty. In: Bauer P. Hommel G. Sonnemann E., editors. Multiple Hypotheses Testing, Heidelberg: Springer 1987;ISBN 3540505598:p. 154-61]. The second new procedure controls the false discovery proportion and improves upon the approach of Lehmann and Romano [Lehmann E.L., Romano J.P. Generalizations of the familywise error rate. Ann. Stat. 2005;33:1138-54]. By Monte-Carlo simulations, we show how the gain in power depends upon the accuracy of the estimate of the number of true hypotheses. The gain in power of our procedures is demonstrated in an example using EEG data on the processing of memorized lexical items.

The current trend shows an increasing demand for composites due to their high stiffness to weight ratio and the recent progress in manufacturing and cost reduction of composites. To combine high strength and stiffness in a cost-effective way, composites are often joined with steel or aluminum. However, joining of thermoset composite materials is challenging because circular holes are often used to join them with their metal counterparts. These design based circular holes induce high stress concentration around the hole. The purpose of this paper is to focus on layup configuration and its impact on notch stress distribution. To ensure high quality and uniformity, the holes were machined by a 5 kW continuous wave (cw) CO2 laser. The stress distribution was evaluated and compared by using finite element analysis and Lekhnitskii's equations. For further understanding, the notch strength of the laminates was compared and strain distributions were analyzed using the digital image correlation technique.

Brain oscillations in the α- and ß-range become suppressed during motor processing and motor imagery. It has recently been discussed that such power changes also occur during action language processing. In our study, we compared ß2-oscillations (16-25Hz) during the observation of prototypical arm movements (revealed via motion tracking) as well as during semantic processing of concrete and abstract sentences containing arm-related action verbs. Whereas we did find a strong desynchronization in the ß2-range during action observation, the processing of action sentences evoked a rather weak desynchronization. However, this desynchronization occurred for action verbs in both concrete and abstract contexts. These results might indicate a tendency for abstract action language to be processed similar to concrete action language rather than abstract sentences. The oscillation patterns reflect the close relationship between language comprehension and motor functions - one of the core claims of current theories on embodied cognition.

In a behavioral study we analyzed the influence of visual action primes on abstract action sentence processing. We thereby aimed at investigating mental motor involvement during processes of meaning constitution of action verbs in abstract contexts. In the first experiment, participants executed either congruous or incongruous movements parallel to a video prime. In the second experiment, we added a no-movement condition. After the execution of the movement, participants rendered a sensibility judgment on action sentence targets. It was expected that congruous movements would facilitate both concrete and abstract action sentence comprehension in comparison to the incongruous and the no-movement condition. Results in Experiment 1 showed a concreteness effect but no effect of motor priming. Experiment 2 revealed a concreteness effect as well as an interaction effect of the sentence and the movement condition. The findings indicate an involvement of motor processes in abstract action language processing on a behavioral level.
