RESUMO

Effects of selenoproteins on many renal diseases have been reported. However, their role in renal ischemia-reperfusion (I/R) injury is unclear. The present study was performed to investigate the impact of ebselen and renal I/R injury on the expression of selenoproteins. Sprague-Dawley rats were pretreated with or without ebselen (10 mg/kg) through a daily single oral administration from 3 days before renal I/R surgery. RT-qPCR (real-time quantitative PCR) was performed to determine the mRNA expression of 25 selenoprotein genes in the renal tissues. The expression levels of two selenoproteins, including GPX3 (glutathione peroxidase 3) and DIO1 (iodothyronine deiodinase 1), were evaluated by Western blot or/and IHF (immunohistofluorescence) assays. Furthermore, renal function, renal damage, oxidative stress, and apoptosis were assessed. The results showed that in renal I/R injury, the mRNA levels of 15 selenoprotein genes (GPX1, GPX3, GPX4, DIO1, DIO2, TXNRD2, TXNRD3, SEPHS2, MSRB1, SELENOF, SELENOK, SELENOO, SELENOP, SELENOS, and SELENOT) were decreased, whereas those of eight selenoprotein genes (GPX2, GPX6, DIO3, TXNRD1, SELENOH, SELENOM, SELENOV, and SELENOW) were increased. I/R also induced a reduction in the expression levels of GPX3 and DIO1 proteins. In addition, our results indicated that ebselen reversed the changes in those selenoprotein genes, excluding SELENOH, SELENOM, SELENOP, and SELENOT, in renal I/R injury and alleviated I/R-induced renal dysfunction, tissue damage, oxidative stress, and apoptosis. To our knowledge, this is the first study to investigate the changes of 25 mammalian selenoprotein genes in renal I/R injury kidneys. The present study also provided more evidence for the roles of ebselen against renal I/R injury.

Test fixtures with high power capacity and an impedance matching network are generally chosen for measurements of high-power gallium nitride high electron mobility transistors. To make interconnection of the test fixtures and devices under test, wire bonding is an effective assembly method. Bonding wires become dominant parasitic elements, especially in the S parameter measurement and loadpull measurement, which should be taken into account and accurately de-embedded for device measurements and modeling. In this paper, test fixtures and through-reflect-line calibration kits are designed to achieve the S parameter measurements of the bonding wire with a vector network analyzer. Equivalent inductances of the bonding wire can be obtained with the electromagnetic simulation model and compact circuit model proposed based on the test fixture. The good agreement of the equivalent inductances extracted with the test fixture and models verified that the way to characterize bonding wire interconnection is effective and accurate. Cree's CGHV1J006D is chosen to take the S parameter measurement for proving the accurate model of the bonding wire. Finally, the equivalent inductance of the 2 mm gate-width transistor is obtained with the electromagnetic model. The drain impedance is accurately calculated after the loadpull measurement with bonding wire effects de-embedded, which matches the loadline theory.

Measurement for modeling of the high-power transistors is difficult due to its high-power and low-impedance characteristics. In this paper, novel methods and devices were designed and applied to achieve precise measurements of the high-power transistors. Fixtures capable of withstanding high voltage and current were designed to replace traditional radio frequency (RF) probes for higher power capacity. To reduce the impact of capacitive and inductive components of traditional bias tees on the rising/falling edge, two wideband 90° hybrid couplers that were connected back-to-back were designed for pulsed measurements. The measurement system of stable S-parameters with the Vector Network Analyzer (VNA) was reported, which could protect the devices and laboratory equipment from damage of self-oscillation. Application of several innovative approaches enabled accurate I-V characteristic and S-parameters measurements of high-power transistors in DC or pulsed mode. Experimental results of a 30 W gallium nitride high-electron-mobility transistor verified the validity.

Objective To analyze the efficacy and safety of transarterial chemoembolization (TACE) with chemotherapy regimen containing platinum in the treatment of primary liver cancer. Methods 86 patients with advanced hepatocellular carcinoma treated in our hospital from December 2015 to December 2016 were selected and randomly divided into two groups with 43 cases each group. Lipiodol chemotherapy was performed with Lobaplatin, epirubicin hydrochloride and mitomycin in the study group, control group while Lipiodol chemotherapy was performed with oxaliplatin, epirubicin hydrochloride and mitomycin in the control group. The gelatin sponge was used to block tumor blood vessels and the treatment lasted for 2 courses. According to the curative effect evaluation standard of solid tumor (RECIST), the curative effect was evaluated and the adverse reactions were compared between the two groups. Results The total effective rates of the two groups were not significantly different , but the effective rate of the study group (20.93%) was significantly higher than that of the control group (6.98%), and the difference was statistically significant (P<0.05). The incidence of hematologic toxicity in the study group (30.23%) was slightly higher than that in the control group (20.93%), the incidence of gastrointestinal reactions (44.19%) and liver and kidney damage (13.95%) were lower than that of the control group respectively by 60.47% and 25.58%, but there was no significant difference . The incidence of neurotoxicity in the study group (2.33%) was significantly lower than that in the control group (27.91%), and the difference was statistically significant (P<0.05). Conclusion For patients with advanced hepatocellular carcinoma, there was no significant difference between the efficacy and safety of transarterial chemoembolization with chemotherapy regimen containing platinum and oxaliplatin chemotherapy regimen, and adverse reactions are similar, but the incidence of neurotoxicity is low.

Objective@#To evaluate the role of the serum testosterone level as an independent predictor of bone metastasis of prostate cancer.@*METHODS@#This study included 165 male patients with prostate cancer confirmed by biopsy. The patients were aged 58-78 (66.6±5.3) years and none had received androgen-deprivation therapy, chemotherapy or radiotherapy previously. We obtained the baseline clinical data from the patients, including prostate biopsy Gleason scores and the levels of serum prostate-specific antigen (PSA), total testosterone (TT), luteinizing hormone (LH), follicle-stimulating hormone (FSH), estradiol (E2), alkaline phosphatase (ALP) and prolactin. According to the results of bone scanning, we divided the patients into a bone metastasis and a non-bone metastasis group and screened out the differential factors by univariate analysis and the independent predictor of bone metastasis using the multivariate non-conditional logistic regression model.@*RESULTS@#Univariate analysis showed no statistically significant differences between the bone metastasis and non-bone metastasis groups in age (P = 0.126) or the levels of serum LH (P = 0.930), FSH (P = 0.763) and E2 (P = 0.256), but that the former had remarkably higher Gleason scores (P < 0.01), total PSA (P <0.01) and ALP (P <0.01) but a lower TT level than the latter (P = 0.013). According to the results of multivariate logistic regression analysis, serum ALP (P <0.01, OR = 1.018 [1.011-1.026]) and total PSA (P <0.01, OR = 1.029 [1.015-1.044]) could be regarded as independent predictors of bone metastasis of prostate cancer but not low serum TT (P = 0.531, OR = 0.999 [0.996-1.002]) or biopsy Gleason score (P = 0.898, OR = 0.787 [0.412-1.9559]).@*CONCLUSIONS@#The low level of serum testosterone is closely associated with but not an independent predictor of bone metastasis of prostate cancer.
