The question how the brain distinguishes between information about self and others is of fundamental interest to both philosophy and neuroscience. In this functional magnetic resonance imaging (fMRI) study, we sought to distinguish the neural substrates of representing a full-body movement as one's movement and as someone else's movement. Participants performed a delayed match-to-sample working memory task where a retained full-body movement (displayed using point-light walkers) was arbitrarily labeled as one's own movement or as performed by someone else. By using arbitrary associations we aimed to address a limitation of previous studies, namely that our own movements are more familiar to us than movements of other people. A searchlight multivariate decoding analysis was used to test where information about types of movement and about self-association was coded. Movement specific activation patterns were found in a network of regions also involved in perceptual processing of movement stimuli, however not in early sensory regions. Information about whether a memorized movement was associated with the self or with another person was found to be coded by activity in the left middle frontal gyrus (MFG), left inferior frontal gyrus (IFG), bilateral supplementary motor area, and (at reduced threshold) in the left temporoparietal junction (TPJ). These areas are frequently reported as involved in action understanding (IFG, MFG) and domain-general self/other distinction (TPJ). Finally, in univariate analysis we found that selecting a self-associated movement for retention was related to increased activity in the ventral medial prefrontal cortex.

We conducted three experiments to test the effect of assumed task-relevance of self-association on the self-prioritization effect (SPE). Participants were first performing the standard matching task, and then a pseudo-word matching task, in which familiar labels from the standard task were replaced with pseudo-words. In the pseudo-words task, the association between stimuli and the self was, in fact, task-irrelevant. Learning instructions were varied to make participants believe that the self-association was task-relevant (Experiments 1 and 3) or task-irrelevant (Experiment 2). In Experiment 1, we told participants that geometrical shapes represent specific identities (self, friend, none) and pseudo-words' meanings reflect each of these identities to make participants believe that semantic associations of pseudo-words were task-relevant. In this experiment, a SPE was present in the pseudo-words task. Experiment 2 was identical to Experiment 1, but participants were told to form association pairs between pseudo-words and shapes, and not pseudo-words and identities. Thus, because the matching task presented only shapes and pseudo-words, participants were led to correctly believe that associations were task-irrelevant. Under these conditions, we did not observe a SPE in the pseudo-words task. Finally, in Experiment 3, participants were told that pseudo-words are "paired with" identities, making their relations with identities weaker than in Experiment 1, but still task-relevant. The SPE in the pseudo-word task reappeared. Together, the results suggest that a SPE can be observed in the absence of any stimuli with established self-associations, but only if self-associations are represented as task-relevant.

Alcohol consumption during pregnancy constitutes one of the leading preventable causes of birth defects and neurodevelopmental disorders in the exposed children. Fatty acid ethyl esters (FAEEs), ethyl glucuronide (EtG) and ethyl sulfate (EtS) have been studied as potential biomarkers of alcohol consumption. However, most analytical approaches proposed for their analysis in meconium samples consist of separated extraction procedures requiring the use of two meconium aliquots, which is costly in terms of both time and materials. Therefore, the aim of this study was to develop and validate a method for the simultaneous extraction of 9 FAEEs, EtG and EtS from one meconium aliquot. The sample was homogenized using methanol, and then FAEEs were extracted with hexane while EtG and EtS were isolated using acetonitrile. Then, extracts were applied to solid-phase extraction columns and analysed by gas chromatography mass spectrometry (FAEEs) and liquid chromatography tandem mass spectrometry (EtG and EtS). Calibration curves were linear with r values greater than 0.99. The LODs ranged from 0.8 to 7.5 ng/g for FAEEs and were 0.2 ng/g and 0.8 ng/g for EtS and EtG, respectively. LOQs ranged from 5 to 25 ng/g for FAEEs and were 1 ng/g and 2.5 ng/g for EtS and EtG, respectively. Accuracies and precisions were between 93.8 and 107% and between 3.5 and 9.7%, respectively. The recovery values ranged from 89.1 to 109%. The method proved to be sensitive, specific, simple and fast and allowed for the reduction of the amount of organic solvent used for extraction compared to other published data while higher recoveries were obtained. The method was used for analysis of meconium samples in two cases of mothers who were consuming alcohol during pregnancy.

Ibuprofen is widely used in human and veterinary medicine for the treatment of chronic pain as well as rheumatic and musculoskeletal disorders. However, the analgesic and anti-inflammatory properties of Ibuprofen have contributed to frequent drug abuse in equestrian sports. A sensitive and rapid gas chromatography with tandem mass spectrometry based method with a simple liquid-liquid extraction and derivatization requiring 200 µL volume of sample and 2 mL of extraction solvent for the simultaneous determination of ibuprofen and its metabolites was developed. The proposed procedure was optimized and validated according to the principles for bioanalytical methods. The assay achieved satisfactory validation parameters, namely, recovery (92.2-105%), interday accuracy (92.5-106%), and precision (0.3-4.4%) for all investigated compounds as well as limits of quantification of 50 ng/mL for ibuprofen, 2-hydroxyibuprofen, and carboxyibuprofen, 25 ng/mL for 1-hydroxyibuprofen and 100 ng/mL for 3-hydroxyibuprofen. The applicability of the method was evaluated by the analysis of five real urine samples collected from different horses after drug administration. In view of the low limits of quantification, high selectivity, repeatability, and recovery, the procedure can be utilized for laboratory applications, including the control of ibuprofen abuse in equestrian sports for anti-doping purposes and drug/pharmaceutical mentality investigations.

We investigated whether people take into account an interaction partner's attentional focus and whether they represent in advance their partner's part of the task when planning to engage in a synchronous joint action. The experiment involved two participants planning and performing joint actions (i.e., synchronously lifting and clinking glasses), unimanual individual actions (i.e., lifting and moving a glass as if clinking with another person), and bimanual individual actions. EEG was recorded from one of the participants. We employed a choice reaction paradigm where a visual cue indicated the type of action to be planned, followed 1.5 sec later by a visual go stimulus, prompting the participants to act. We studied attention allocation processes by examining two lateralized EEG components, namely the anterior directing attention negativity and the late directing attention positivity. Action planning processes were examined using the late contingent negative variation and the movement-related potential. The results show that early stages of joint action planning involve dividing attention between locations in space relevant for one's own part of the joint action and locations relevant for one's partner's part of the joint action. At later stages of joint action planning, participants represented in advance their partner's upcoming action in addition to their own action, although not at an effector-specific level. Our study provides electrophysiological evidence supporting the operation of attention sharing processes and predictive self/other action representation during the planning phase of a synchronous joint task.

Depersonalization is a common and distressing experience characterized by a feeling of estrangement from one's self, body, and the world. In order to examine the relationship between depersonalization and selfhood we conducted an experimental study comparing processing of three types of self-related information between nonclinical groups of people experiencing high and low levels of depersonalization. Using a sequential matching task, we compared three types of biases for processing of self-related information: prioritization of one's name, of self-associated abstract stimuli (geometrical shapes), and of self-associated bodily stimuli (avatar faces). We found that both groups demonstrated the standard pattern of results for self-prioritization of one's name and geometrical shapes, but they differed with regard to avatar faces. While people with low depersonalization showed the standard prioritization of avatar faces, people with high depersonalization showed overall better response accuracy with avatar faces, and faster response times for stranger-associated avatar faces. These results were complemented by the additional finding that people with high depersonalization reported being more likely to use avatars of a different gender to their own outside of the experimental context. Finally, in this large sample (N = 180) we investigated the relationships between different measures of self-related processing and self-identification, finding no correlation between explicit reports of self-identification with self-associated avatar faces and geometrical shapes, self-prioritization of these stimuli, and prioritization of one's name. Benzodiazepines (BZDs) and Z-drugs are among the most commonly prescribed pharmaceuticals in the world and are considered standard care for various mental illnesses and for the treatment of sleeping and anxiety disorders, alcohol withdrawal, muscle spasms and epilepsy. Some BZDs are not allowed as pharmaceuticals in many countries, and they are used as designer benzodiazepines (DBZDs). All these compounds are typically screened in routine toxicological analyses for forensic purposes. Knowledge of time-dependent decreases in drug concentrations during storage or transport of samples is of considerable significance and allows forensic toxicologists to achieve reliable data, proper interpretation and high-quality results. The aim of this study was to evaluate changes in the amounts of selected BZDs, DBZDs and Z-drugs in blood samples stored at various temperatures. The study involved BZDs (19), DBZDs (3) and Z-drugs (2) spiked into blank blood. Subsequently, the blood samples were stored at various temperatures (room temperature, 4°C, -20°C and -80°C) for up to 6 months. Analyses were performed at 1- to 2-week intervals using liquid chromatography-tandem mass spectrometry. The stability of compounds was evaluated under four temperature conditions over a 6-month period. Some BZDs were stable at all temperatures tested (e.g., diazepam, oxazepam, nordazepam and prazepam) with a degradation rate of only 0-10%. The highest instability was observed for analyte samples kept at room temperature, and the losses in content for some compounds, e.g., lorazepam and chlordiazepoxide, were almost 100%. For other compounds, the stability was clearly different at each tested temperature. To the best of our knowledge, this is one of the first such comprehensive study of the long-term stability of BZDs covering a wide range of different storage temperatures.

Preterm birth may result from overlapping causes including maternal age, health, previous obstetric history and a variety of social factors. We aimed to identify factors contributing to preterm birth in respect to new social and environmental changes in the reproductive patterns. Our cross-sectional study included 495 mother-infant pairs and was based on maternal self-reporting in an originally developed questionnaire. Neonates were divided into two groups: 72 premature babies (study group) and 423 full-term babies (control group). We analyzed maternal, sociodemographic and economic characteristics, habits, chronic diseases, previous obstetric history and pregnancy complications. For statistical analysis, Pearson's Chi-squared independence test was used with a statistical significance level of 0.05. Preterm births were more common among mothers living in villages (p < 0.001) and with lower education level (p = 0.01). Premature births were also positively associated with mothers who were running their own businesses (p = 0.031). Mothers with a history of previous miscarriages gave birth at a significantly older age (p < 0.001). The most frequent pregnancy complications were hypothyroidism (41.4%), pregestational and gestational diabetes mellitus (DM; 17.8%) and hypertension (8.1%). Pregestational DM significantly influenced the occurrence of prematurity (p < 0.05). Pregestational DM, being professionally active, a lower education level and living outside cities are important risk factors of prematurity.

Making one's actions predictable and communicating what one intends to do are two strategies to achieve interpersonal coordination. It is less clear whether these two strategies are mutually exclusive or whether they can be used in parallel. Here, we asked how the availability of communication channels affects the use of strategy to make one's actions predictable. In three experiments, we investigated how people reach joint decisions if they are not allowed to communicate at all (Experiment 1), allowed minimal reciprocal communication (Experiment 2), or allowed to use the full range of conventional communication (Experiment 3). We found that when participants were not allowed to communicate, coordination was achieved by increasing action predictability. When conventional communication was allowed, there were no attempts to increase action predictability. In the minimal reciprocal communication condition, successful pairs both increased action predictability and established a communication system. Overall, this study demonstrates that people are able to flexibly adapt to coordination challenges during joint decision making and that communication reduces behavioural constraints on joint action coordination.

In the current study, we separately tested whether coordinated decision-making increases altruism and whether it increases trust. To this end, we implemented a paradigm in which participants repeatedly perform a coordinated decision-making task either with the same partner on every trial, or with a different partner on each trial. When both players coordinate on the same option, both are rewarded. In Experiment 1 (N = 52), participants were sometimes presented with tempting opportunities to defect. In Experiment 2 (N = 97), participants sometimes had to decide whether or not to trust that their partners had resisted such tempting opportunities. The results show that repeatedly coordinating with the same partner increased participants' resistance to temptation (Experiment 1) but did not increase trust (Experiment 2). These findings support the hypothesis that coordinating with a partner increases altruistic motivation towards that partner; they do not support the hypothesis that coordinating boosts trust.

Depersonalisation is a common dissociative experience characterised by distressing feelings of being detached or 'estranged' from one's self and body and/or the world. The COVID-19 pandemic forcing millions of people to socially distance themselves from others and to change their lifestyle habits. We have conducted an online study of 622 participants worldwide to investigate the relationship between digital media-based activities, distal social interactions and peoples' sense of self during the lockdown as contrasted with before the pandemic. We found that increased use of digital media-based activities and online social e-meetings correlated with higher feelings of depersonalisation. We also found that the participants reporting higher experiences of depersonalisation, also reported enhanced vividness of negative emotions (as opposed to positive emotions). Finally, participants who reported that lockdown influenced their life to a greater extent had higher occurrences of depersonalisation experiences. Our findings may help to address key questions regarding well-being during a lockdown, in the general population. Our study points to potential risks related to overly sedentary, and hyper-digitalised lifestyle habits that may induce feelings of living in one's 'head' (mind), disconnected from one's body, self and the world.

Pentoxifylline is a xanthine derivative used in vascular disorders that is recognized as a safe drug for patients. The paper describes a rare case of fatal and suicidal pentoxifylline poisoning in an 82-year-old man with multiple preexisting diseases (arterial hypertension, coronary artery disease, type 2 diabetes, and intermittent claudication). The patient was admitted to the clinical toxicology unit approximately 2 h after the overdose and died 36 h after the admission despite intensive care. Multiple arterial blood gas analyses and other laboratory tests were performed during the hospitalization and are reported in the paper. Postmortem examination of the biological material was carried out with the use of histopathological techniques. The toxicological studies using chromatographic techniques coupled with mass spectrometry showed that postmortem blood levels of pentoxifylline have been found in the range which is described in the available literature to be toxic and lethal. The analysis of test results and clinical data showed that the patient died as a result of increasing circulatory and respiratory failure, complicated by disorders of the acid-base and electrolyte balance (respiratory alkalosis, concomitant lactic acidosis, and hypokalemia), hyperglycemia, and coagulation disorders.

N-Ethylhexedrone [2-(ethyloamino)-1-phenylhexan-1-one; α-ethylaminohexanophenone (NEH)] is one of the most recent synthetic cathinones that appeared on the illegal market in late 2015. The majority of information concerning the model of consumption of NEH and its impact on the body originates only from self-reports from gray literature websites and drug forums. There are only limited data associated with the concentrations of NEH in blood samples available in the literature. This article presents a case of fatal NEH intoxication and a method for the determination of this substance in whole blood. A 21-year-old man without any diagnosed diseases was admitted to the hospital due to disorientation, aggression and finally loss of consciousness. Hyperthermia (>41°C), tachycardia (>160 beats per minute), tachypnea (20 breaths per minute), blood pressure (110/60 mmHg) and acute kidney failure were diagnosed. After a few hours of hospitalization, the patient died. A plastic bag with a white powder was found in his underwear. Analysis of the powder by another laboratory revealed cocaine hydrochloride; however, no cocaine or its metabolites were found in the biological material upon testing in our laboratory. Therefore, re-analysis of the powder was performed, and NEH was identified. Liquid-liquid extraction followed by liquid chromatography-triple quadrupole-mass spectrometry (LC-MS/MS) analysis were used for the determination of NEH in blood. The validation parameters were as follows: calibration range 1-250 ng/mL, accuracy 106.5-109.9%, precision 3.5-6.3%, recovery 90.1-96.9%, limit of detection 0.07 ng/mL and limit of quantification 1 ng/mL. NEH was quantified in the blood at a concentration of 145 ng/mL. Additionally, amphetamine at low concentrations and 11-nor-9-karboksy-Δ9-tetrahydrokannabinol (THC-COOH) were detected. Our study provided information on the possible lethal concentration and toxidrome that clinicians can observe for NEH-intoxicated patients and can be helpful during the preparation of toxicology analysis reports for a court of law for proper data interpretation.

Recent studies suggest that we rapidly and effortlessly associate neutral information with the self, leading to subsequent prioritization of this information in perception. However, the exact underlying processes behind these effects are not fully known. Here, we focus specifically on top-down and bottom-up processes involved in self-prioritization, and report results from three experiments involving face detection, using a sequential match-non-match task. Across the three experiments we asked participants to associate an unfamiliar face with the self (Experiment 1), to associate one's face with a stranger's name (Experiment 2), and to establish both associations simultaneously (Experiment 3). We found that while participants showed evidence of bottom-up prioritization of their real faces, they did not show such an effect for self-associated strangers' faces. However, the participants showed a robust self-related top-down effect; when presented with a self-related cue, they were later faster at classifying both subsequent correct and incorrect targets. Together, our results suggest that self-prioritization is underpinned by distinct top-down and bottom-up processes. We discuss our findings in the context of the proposal that the self acts as an "integrative glue", and suggest an interpretation of our results within the framework of predictive coding.

Benzodiazepines (BZDs) and Z-drugs have been particularly important treatments for sleeping and anxiety disorders for many years. However, recently, a number of new benzodiazepines (named designer benzodiazepines, DBZDs) were synthesised, but some of them have never been used in the clinic; they reached the black drug market as new psychoactive substances and are used for recreational purposes. The abuse of these substances has led to many crimes and even deaths. Therefore, it is necessary to develop new methods for their quantification for forensic and clinical toxicology. A liquid chromatography-tandem mass spectrometry-based method was developed for the simultaneous determination of 20 classical BZDS, 4 DBZDs and 3 Z-hypnotic drugs in human whole blood. As a sample preparation step, liquid-liquid extraction requiring the use of only 0.5 mL of blood sample and 1 mL of extraction solvent was applied. The selectivity, linearity, carry-over effects, limits of detection (LOD) and quantification (LOQ), precision, accuracy (both intra- and inter-day assays) and recovery were evaluated for validation. Calibration curves were linear with r values > 0.98. The LODs ranged from 0.01 to 0.33, and the LOQs were assumed to be 1 ng/mL. Inter-day precisions and accuracies were in the ranges of 87.8% - 108.5% and 1.8% - 11.2%, respectively. The recovery values ranged from 81.0% to 106.7%. The developed method proved to be sensitive, specific, simple, and fast and can be quickly modified and expanded for new compounds by the optimization of MRM. The method was applied for analysis of blood samples in 145 toxicological cases over a three-year study (2017 - 2019), which allowed us to obtain information on the prevalence of the use of these substances. The most frequently determined compounds were nordazepam (87 cases; 60%), diazepam (81 cases; 55.9%), temazepam (72 cases; 49.7%), oxazepam (56 cases; 38.7%), and midazolam (36 cases; 24.8%). The ranges of concentrations were wide and are presented as box plots. The results were used for the preparation of medico-legal opinions, which proved the utility of the method for routine toxicology analyses.

Recently, Sui and colleagues introduced an experimental task to investigate prioritization of arbitrary stimuli associated with the self. They demonstrated that after being told to associate three identities (self, friend, stranger) with three arbitrary stimuli (geometrical shapes), participants were faster in a perceptual matching task to recognise matching pairs of self-associated shape with self-label, than respective friend or stranger-related pairings. They interpreted this as evidence that a brief self-association is sufficient to facilitate processing of previously neutral stimuli. However, in the matching trials of the self-prioritization task, participants are processing not only self-associated arbitrary stimuli but also familiar verbal labels with an established meaning. Therefore, the self-advantage may be caused by familiarity of the labels, rather than self-association of the shapes. To test whether self-prioritization can be elicited in a task employing exclusively neutral stimuli, we asked participants to associate avatar faces with three identities (self, name of best friend, and stranger) and replaced labels with unfamiliar abstract symbols that were associated to the words (you, friend, stranger) before the actual experiment started. The results presented the usual pattern of self-prioritization showing that this effect does not critically depend on the presence of familiar labels and that it can be elicited in the absence of any familiar stimuli.

BACKGROUND: Ethyl alcohol is the most popular legal drug, but its excessive consumption causes social problems. Despite many public campaigns against alcohol use, car accidents, instances of aggressive behaviour, sexual assaults and deterioration in labor productivity caused by inebriated people is still commonplace. Fast and easy diagnosis of alcohol consumption is required in order to introduce proper and effective therapy, and is crucial in forensic toxicology analysis. The easiest method to prove alcohol intake is determination of ethanol in body fluids or in breath. However, since ethanol is rapidly metabolized in the human organism, only recent consumption can be detected using this method. Because of that, the determination of alcohol biomarkers was introduced for monitoring alcohol consumption over a wider range of time. OBJECTIVE: The objective of this study was to review published studies focusing on the sample preparation methods and chromatographic or biochemical techniques for the determination of alcohol biomarkers in whole blood, plasma, serum and urine. METHODS: An electronic literature search was performed to discuss possibilities and limitations of application of alcohol biomarkers in toxicological analysis. RESULTS: Authors described the markers of alcohol consumption such as: ethanol, its nonoxidative metabolites (ethyl glucuronide, ethyl sulfate, phosphatidylethanol, ethyl phosphate, fatty acid ethyl esters) and oxidative metabolites (acetaldehyde and acetaldehyde adducts). We also discussed issues concerning the detection window of these biomarkers, and possibilities and limitations of their use in routine analytical toxicology for monitoring alcohol consumption or sobriety during alcohol therapy.

Do people engaged in joint action form action plans that specify joint outcomes at the group level? EEG was recorded from pairs of participants who performed coordinated actions that could result in different postural configurations. To isolate individual and joint action planning processes, a pre-cue specified in advance the individual actions and/or the joint configuration. Participants had 1200 ms to prepare their actions. Then a Go cue specified all action parameters and participants performed a synchronized action as quickly as possible. Action onsets were shorter when the pre-cue specified the joint configuration, regardless of whether individual action was also specified. EEG analyses showed that specifying joint action parameters in advance reduced ambiguity in a structured joint action plan (reflected in the decrease of the amplitude of the P600) and helped with representing action goals and interpersonal coordination patterns in sensorimotor brain areas (reflected in increased alpha/mu suppression and CNV amplitudes). These results provide clear evidence that joint action is driven not only by action plans that specify individual contributions, but also by action plans that specify joint action outcomes at the group level.

James (1890) distinguished two understandings of the self, the self as "Me" and the self as "I". This distinction has recently regained popularity in cognitive science, especially in the context of experimental studies on the underpinnings of the phenomenal self. The goal of this paper is to take a step back from cognitive science and attempt to precisely distinguish between "Me" and "I" in the context of consciousness. This distinction was originally based on the idea that the former ("Me") corresponds to the self as an object of experience (self as object), while the latter ("I") reflects the self as a subject of experience (self as subject). I will argue that in most of the cases (arguably all) this distinction maps onto the distinction between the phenomenal self (reflecting self-related content of consciousness) and the metaphysical self (representing the problem of subjectivity of all conscious experience), and as such these two issues should be investigated separately using fundamentally different methodologies. Moreover, by referring to Metzinger's (2018) theory of phenomenal self-models, I will argue that what is usually investigated as the phenomenal-"I" [following understanding of self-as-subject introduced by Wittgenstein (1958)] can be interpreted as object, rather than subject of experience, and as such can be understood as an element of the hierarchical structure of the phenomenal self-model. This understanding relates to recent predictive coding and free energy theories of the self and bodily self discussed in cognitive neuroscience and philosophy.

Behavioral and neuroimaging studies have demonstrated that people process preferentially self-related information such as an image of their own face. Furthermore, people rapidly incorporate stimuli into their self-representation even if these stimuli do not have an intrinsic relation to self. In the present study, we investigated the time course of the processes involved in preferential processing of self-related information. In two EEG experiments three unfamiliar faces were identified with verbal labels as either the participant, a friend, or a stranger. Afterwards, participants judged whether two stimuli presented in succession (ISI = 1500ms) matched. In experiment 1, faces were followed by verbal labels and in experiment 2, labels were followed by faces. Both experiments showed the same pattern of behavioral and electrophysiological results. If the first stimulus (face or label) was associated with self, reaction times were faster and the late frontal positivity following the first stimulus was more pronounced. The self-association of the second stimulus (label or face) did not affect response times. However, the central-parietal P3 following presentation of the second stimulus was more pronounced when the second stimulus was preceded by self-related first stimulus. These results indicate that even unfamiliar faces that are associated to self can activate a self-representation. Once the self-representation has been activated the processing of ensuing stimuli is facilitated, irrespective of whether they are associated with the self.
