Long noncoding RNAs (LncRNAs), a class of noncoding RNAs greater than 200 bases in length, are widely involved in the initiation, progression and glycolytic processes of many tumors, and can act as competitive endogenous RNA sponges to absorb miRNAs. LncRNAs can also inhibit miRNA expression, thereby regulate the glycolysis of tumor cells, affects cell proliferation, invasion and other biological activities. This review explores the roles of LncRNAs and glycolysis in digestive system tumors (DST), a representative group of malignant tumors. Extending the LncRNA role in the diagnosis, treatment and prognosis of other tumors, we conclude that LncRNAs have the potential to be new candidate genes for tumorigenesis and serve as tumor biomarkers, which provides new insight into morbidity and mortality decrease of DST and other tumors.

A simulating method for dripping process of Ginkgo biloba leaf dripping pills based on computational fluid dynamics was constructed. Ginkgo biloba leaf dripping pills was explored as the experimental subject to simulate the dripping process based on FLOW-3D software. The dripping process was simulated through the derivation of the governing equations, the selection of the models, and simulation parameters. Firstly, the droplet morphologies and drop speeds under different liquid viscosity were simulated. It was found that with the increase of the liquid viscosity, the drop speed decreased and the difficulty of droplet preparation gradually increased. The simulation results were consistent with the experiment results. Secondly, the droplet morphologies at different drop speeds were investigated and verified by experiments. It was found that the simulation results had a good correlation with the experiment results. The results shown that the viscosity of the liquid was the critical material attribute, and the drop speed was the critical process parameter, according to the droplet morphology. The establishment of the simulation method can deepen the understanding of the dripping process and provide a reference for the selection of raw materials and process parameters.

ObjectiveThe aim of this study is to investigate the role of salidroside in regulating the miR-1343-3p/MAP3K6 (mitogen-activated protein kinase kinase kinase 6)/MMP24 (membrane-type matrix metalloproteinase 24) signaling pathway to inhibit gastric cancer cell proliferation and migration. MethodsHuman gastric cancer cells (MGC-803) were divided into several groups based on different salidroside concentrations: a control group (0 μmol/mL), a low-dose group (6 μmol/mL), a medium-dose group (12 μmol/mL), and a high-dose group (24 μmol/mL). The anti proliferative effects of salidroside on human gastric cancer cells were evaluated by CCK-8 assay. Clonogenic assay was used to examine the effects of salidroside drugs on the clonogenic ability of human gastric cancer cells. Transwell assay was performed to detect the effect of salidroside on the invasive ability of human gastric cancer cells. Cell scratch assay was performed to detect the effect of salidroside on the migration ability of human gastric cancer cells. The miRNA expression profile was analyzed by using RNA-seq in cancer cells for 24 h after salidroside treatment. The differentially expressed miRNAs were clustered and their target genes were predicted. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) were used to analyze and predict the functions of these target genes, and the interaction networks were established. Immunocytofluorescence was used to detect the expression of target proteins, and the transcription of candidate genes was detected by q-PCR. ResultsCCK-8 cytotoxicity experiments showed that salidroside inhibited the proliferation of MGC-803 cells (P < 0.01). Cell cloning experiments showed that salidroside reduced the clonal formation capacity of MGC-803 cells (P < 0.000 1). Cell invasion experiments showed that salidroside reduced the MGC-803 cell invasion capacity (P < 0.000 1). Cell scratch experiments showed that salidroside reduced the cell migration capacity (P < 0.000 1). RNA-seq findings showed that the expression of 44 miRNAs changed significantly after salidroside treatment in cancer cells (P < 0.05). Bioinformatic analysis showed that there were 1 384 target mRNAs corresponding to the differentially expressed miRNAs, and the expression of the tumor suppressor miR-1343-3p was significantly upregulated after salidroside treatment (P < 0.01),and resulted in down-regulated transcription of MAP3K6 and MMP24 genes which are related to the proliferation and migration of cancer cells (P < 0.05). Immunofluorescence experiments demonstrated that salidroside reduced protein expression levels in MAP3K6 and MMP24 genes (P < 0.000 1). q-PCR experiments showed that salidroside reduced the mRNA expression level of MAP3K6 and MMP24 genes (P < 0.000 1), while miRNA expression in miR-1343-3p gene was upregulated (P < 0.000 1). ConclusionSalidroside regulates the miRNA-1343-3p/MAP3K6/MMP24 signaling molecules to inhibit proliferation and invasion of gastric cancer cells.

ObjectiveTo explore the mechanism of Tongbi Jiangu prescription (TBJG) in the treatment of knee osteoarthritis (KOA) based on network pharmacology and molecular docking,and further verify it by cell experiments. MethodThe active components and the corresponding targets of TBJG were screened out according to the traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP). The targets of KOA were obtained from GeneCards,online mendelian inheritance in man(OMIM), and DrugBank. The common targets of active components of TBJG and KOA were the targets of TBJG against KOA. The active component-target network and protein-protein interaction (PPI) network were constructed by Cytoscape 3.7.2. STRING was used for PPI network analysis. DAVID was used for gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) enrichment analyses. Key targets and core active components were selected for molecular docking by AutoDock. The results of network pharmacology were verified by cell experiments and the pharmacodynamic responses were observed. ResultThe prediction of network pharmacology showed that there were 111 active components of TBJG in the treatment of KOA. The core active components were quercetin,kaempferol, and β-sitosterol,and the key targets were interleukin-1β(IL-1β), matrix metalloproteinase-3 (MMP-3),and tumor necrosis factor-α (TNF-α). Biological processes (BP) in GO analysis mainly involved inflammatory response,response to lipopolysaccharide,apoptosis signaling pathway,and regulation of DNA activity in binding transcription factor. Cellular components (CC) included plasma membrane protein complex,RNA polymerase Ⅱ transcription factor complex,membrane raft,and serine/threonine protein kinase complex. Molecular functions (MF) were mainly enriched in cytokine receptor binding,nuclear receptor activity,protein domain specific binding,serine hydrolase activity,chemokine receptor binding,and activity of nitric oxide synthase regulator. As revealed by the KEGG analysis, the relevant signaling pathways were nuclear factor(NF)-κB, Janus kinase(JAK)/signal transducer and activator of transcription(STAT), and Wnt signaling pathways. Molecular docking results showed that the core active components had good binding activities with key targets. The experimental results showed that TBJG could down-regulate IL-1β, MMP-3,TNF-α, and NF-κB p65 expression levels (P<0.05),and up-regulated NF-κB inhibitor(IκB)-α(P<0.05). ConclusionThe mechanism of TBJG in the treatment of KOA lies in the application of active components such as quercetin,kaempferol, and β-sitosterol with IL-1β,MMP-3, and TNF-α as key targets through the NF-κB,JAK/STAT, and Wnt signaling pathways.

ObjectiveTo study the correlation between the apparent color, comprehensive sweetness and the content of main components in the preparation of Rehmanniae Radix Praeparata processed with Amomi Fructus and Citri Reticulatae Pericarpium, so as to lay a foundation for revealing the processing principle of Rehmanniae Radix. MethodThe color of Rehmanniae Radix Praeparata sample powder was measured by automatic colorimeter, the contents of 14 active components in samples with different heating time points were determined by high performance liquid chromatography, including 7 glycosides of catalpol, rehmannia glycoside D, leonurine glycoside, 5-hydroxymethyl furfural, verbascoside, isoacteoside and hesperidin, and 7 carbohydrates of D-fructose, glucose, sucrose, melibiose, raffinose, manninotriose and stachyose), and the mobile phase was acetonitrile-water for gradient elution. The comprehensive sweetness difference of sample was calculated by the sweetness of saccharides, SPSS 21.0 was used to analyze the relationship between the color, comprehensive sweetness and the main component contents in the processing of Rehmanniae Radix Praeparata processed with Amomi Fructus and Citri Reticulatae Pericarpium, the quality comprehensive evaluation index of Rehmanniae Radix Praeparata by triangular area method was established. ResultDuring the processing, the color value of the powder increased, and the apparent color of the sample became darker. the content determination results showed that the content of glycosides decreased, monosaccharides and comprehensive sweetness increased with the increase of heating time. The results of correlation analysis showed that chromaticity value, comprehensive sweetness were significant negatively correlated with the content of iridoid glycosides (P<0.01), the chromaticity value was significant positively correlated with the contents of furaldehyde derivatives, phenylethanoid glycosides, flavonoids and comprehensive sweetness was significant positively correlated with the contents of furaldehyde derivatives, phenylethanoid glycosides (P<0.01), and the comprehensive sweetness was positively correlated with the content of flavonoids (P<0.05). After 52 h of processing, the comprehensive evaluation index of samples reached 0.99. ConclusionThe overall trend of cluster analysis of powder chromaticity value of Rehmanniae Radix Praeparata is basically consistent with that of naked eyes, the comprehensive quality evaluation of Rehmanniae Radix Praeparata processed with Amomi Fructus and Citri Reticulatae Pericarpium can be carried out by combining the three indexes of powder chromaticity value, comprehensive sweetness and glycosides content.

The incidence of malignant melanoma, a highly fatal skin tumor, is on the rise worldwide. Melanomas are highly aggressive and have strong metastatic capability that leads to lethality. Recurrence occurs in patients with distant metastases, even with the latest treatments, and median survival is only a few months. At present, the prevention and treatment of melanoma includes surgical resection, chemotherapy, immunotherapy and targeted therapy. However, these strategies can lead to drug resistance and adverse effects. In recent years, an increasing number of studies have found that natural products have effective anti-melanoma activities, including inhibition of tumor growth, induction of cell apoptosis, inhibition of angiogenesis and metastasis and toxicity to tumor stem cells. In addition, several studies have reported that the combination of natural products and traditional anti-melanoma drugs can enhance the therapeutic efficacy. In this review we summarize the prevention and treatment of melanoma with natural products.

Objective:To investigate constituents containing phenolic hydroxyl or carboxylic acid (excluding diarylheptanoids) from Curcumae Rhizoma and Sparganii Rhizoma herbal pair and single herb. Method:Multiple chromatographic separation techniques, including silica gel,MCI gel and Sephadex LH-20 gel, were employed to isolate and purify the compounds. Their structures were identified by means of the nuclear magnetic resonance (NMR),mass spectrometry (MS) and physicochemical properties. Constituents were quickly analyzed by UPLC-LTQ-Orbitrap-MSn,and scanned in positive and negative ion modes. Result:These compounds were determined as trans-p-hydroxycinnamic acid(1),vanillic acid(2),protocatechuic acid(3),fumalic acid(4),succinic acid(5),succinic acid monomethyl ester(6),docosanoic acid(7),azelaic acid(8) and p-hydroxybenzaldehyde(9). Forty compounds were speculated by comparing mass spectrometry data,retention times of some compounds and reference materials,including 14 phenols and 26 organic acids. Among the compounds of herbal pair,8 phenols and 22 organic acids in Sparganii Rhizoma as well as 11 phenols and 13 organic acids in Curcumae Rhizoma were identified. Cleavage pathways of main compounds were described. Conclusion:There are abundant phenols and organic acids in Curcumae Rhizoma and Sparganii Rhizoma herbal pair and single herb. The results enrich pharmacodynamic material basis of Curcumae Rhizoma and Sparganii Rhizoma herbal pair.

Objective To discuss the related factors influencing the initiation time of forensic psychiatric assessment by analysis of the initiation time of forensic psychiatric assessment of criminal cases in Hunan Province. Methods Related data in assessment files of criminal cases accepted by 8 forensic psychiatric assessment institutions in Hunan Province from January 1, 2011 to December 31, 2016 were extracted. The Logistic regression analysis was used to explore the factors influencing the initiation time of forensic psychiatric assessment. After using property score matching (PSM) to control the influence of confounding factors, the efficiency of public security organs to initiate assessments of suspects with (without) mental disorders and with (without) responsibilities were compared. Results A total of 4 346 cases were included. The Logistic regression analysis suggested that the factors independently related to the initiation time of assessment include: cause of assessment, nationality of the assessed, history of diagnosis and treatment of mental illnesses, history of crimes, history of drug abuse, and status of alcohol consumption before the crime (all P<0.05). The initiation time of assessment of suspects diagnosed with mental disorder was shorter than those with none (P<0.05); the initiation time of assessment of suspects without criminal responsibility was shorter than those with responsibility (P<0.05). After using PSM to control confounding factors, the differences above still existed. Conclusion The cause of assessment, nationality of the assessed, history of diagnosis and treatment of mental illnesses, history of crimes, history of drug abuse, and status of alcohol consumption before the crime are factors that influence the efficiency of public security organs to initiate forensic psychiatric assessments. Under the current assessment initiation mode, forensic psychiatric assessment of suspects who have mental disorders, especially those with no responsibility may be given priority to initiate.

Objective:To optimize the entropy TOPSIS model to evaluate the quality of Hemerocallis Flava from different regions,in order to provide a new evaluation method for the quality control of traditional Chinese medicine. Method:The entropy weight TOPSIS model optimized by analytic hierarchy process(AHP) method was used to analyze the quality of Hemerocallis Flava from 14 different regions, and a comprehensive evaluation index system for the quality of Hemerocallis Flava, which covered 3 layers (target layer,decision layer and index layer),and 10 indexes (corolla of Hemerocallis Flavathe's bud,pistils,stamens,peduncle length,extract,total ash,quercetin,β-rhamnocitrin,kaempferol,sitosterin) was established. Result:Qingyang showed the best quality of Hemerocallis Flava,which was followed by Weinan,and the lowest quality was found in Datong, Shanxi, and Xiaowan village, Sichuan. The results were consistent with the evaluation results of traditional empirical identification,suggesting the successful modeling. The contents of β-rhamnocitrin and kaempferol in Qingyang were 1.72 times and 2.74 times of those of Xiaowan village, Sichuan. There was no significant difference in other active ingredients from different regions. It was suggested that quercetin and kaempferol could be used as the identification and quality evaluation indexes of cauliflower. Conclusion:The entropy TOPSIS model based on the AHP method is clear,simple to use and easy to calculate, with distinct evaluation indexes. It is a practical,quick and effective comprehensive evaluation method for multi-objective decision analysis.

Traditional Chinese medicine is planted in mountainous areas with suitable natural conditions. The planting area is complex in terrain,and the planting plots are mostly irregularly shaped. It is difficult to accurately calculate the planting area by traditional survey methods. The method of extracting Chinese herbal medicine planting area combined with remote sensing and GIS technology is of great significance for the rational development and utilization of traditional Chinese medicine resources. Taking Bletilla striata planting in Ningshan county of Shaanxi province as an example,the extraction method of planting area of traditional Chinese medicine in county was studied. High-resolution ZY-3 and GF-1 multi-spectral multi-temporal remote sensing images were used as data sources. Through field sampling,samples such as B. striata,cultivated land,forest land,water body,artificial surface,alpine meadow,etc. are collected. The spectral features,texture features and shape features of remotely identifiable objects in different planting areas and cultivated land,vegetable sheds were analyzed,confusing ground objects were eliminated and interpretation marks were establish. The method of visual interpretation is used to realize the extraction of B. striata planting areas,and the B. striata planting area are calculated by combining GIS technology. The results showed that the method of visual interpretation,using high-resolution ZY-3 and GF-1 multi-spectral multi-temporal remote sensing image data extracted the planting area of 403.05 mu. It can effectively extract the B. striata planting area in research region.

Objective:To establish a high performance liquid chromatography (HPLC) method for determination of four active constituents, namely MSTG-A,MSTG-B,gaultherin and chlorogenic acid in the anti-inflammatory and analgesic active fraction (ARF) of the ethnic medicine Gaultheria leucocarpa var. yunnanensis, in order to provide a methodological basis for the in-depth study and quality control of G. leucocarpa var. yunnanensis,and lay a foundation for later preparation and clinical application. Method:The determination was performed on COSMOSIL 5C18-PAQ (4.6 mm×250 mm,5 μm) column with methanol-0.2% glacial acetic acid (gradient elution) as the mobile phase at a flow rate of 1 mL·min-1. The column temperature was 25℃. The detection wavelength was set at 294 nm. Result:The linear range of MSTG-B,MSTG-A,gaultherin and chlorogenic acid were 0.014 06-0.450 00,0.007 81-0.250 00,0.003 13-0.100 00,0.000 94-0.030 00 g·L-1 (r ≥ 0.999 7),respectively,with a good precision,repeatability and stability. And the average recoveries were 100.81%,98.99%,96.12% and 102.56%,respectively. RSDs were 1.4%,0.7%,0.7%,2.4%,respectively. The contents of MSTG-B,MSTG-A,gaultherin and chlorogenic acid in ARF fraction of G. leucocarpa var. yunnanensis were 23.608,41.973,8.282,2.673 mg·g-1,respectively. Conclusion:The established method was simple and accurate, with a high repeatability. It can be used for determination of four active constituents in ARF fraction of G. leucocarpa var. yunnanensis,so as to provide a reference for the in-depth research,quality control and comprehensive evaluation of G. leucocarpa var. yunnanensis and lay a solid foundation for preparation and clinical application.

Objective To observe the effects of three effective constituents of Min Angelicae Sinensis Radix on hyperhomocysteinemia (HHcy) induced rabbit atherosclerotic lesions (AS).Methods Forty eight Japanese big ear rabbits were randomly divided into normal group, model group, Angelicae Sinensis Radix volatile oil group, Angelicae Sinensis Radix polysaccharide group, ferulic acid group and folic acid group, with 8 rabbits in each group. The HHcy rabbit model induced AS was duplicated by injected DL-methionine. After 8 weeks, each medication group was given relevant medicine for gavage. After 12 weeks, Hcy, TC, TG, HDL-C and LDL-C level in serum were detected, and abdominal aorta vascular ultrasound were observed.Results The model group was higher than the normal group in the level of Hcy (P<0.05). Compared with the model group, the levels of Hcy in Angelicae Sinensis Radix volatileoil group, the ferulic acid group and the folic acid group decreased (P<0.05). Compared with the model group, the levels of TC, TG and LDL-C in AngelicaeSinensis Radix volatile oil group, ferulic acid group and folic acid group decreased (P<0.05). Abdominal aorta ultrasonography showed that the degree of AS was significantly attenuated in folic acid group, Angelicae Sinensis Radix volatile oil group and ferulic acid group.Conclusion Angelicae Sinensis Radix volatile oil and ferulic acid in Min Angelicae Sinensis Radix can reduce the harm to endothelium and can prevent and treat HHcy caused by rabbit AS.

Parkinson's disease (PD) is a chronic and progressive degenerative disorder of brain commonly seen among the elderly. As conventionally medical therapy is of limited relief and potential side effects, complementary and alternative medicine (CAM) has attracted growing public and professional attention. Therapies such as acupuncture, musical/rhythmic therapy and deep brain stimulation have been gradually proved positively in clinic. In this review, we retrospected the scientifific or evidence-based-medicine advances of application and research for modern treatment of PD by CAM, especially traditional Chinese medicine in categories.

Based on the principle of particle design, the powder of Xiaojin Pills was prepared, and the quality uniformity was investigated by means of powder characterizations and content uniformity. By studying the mixed crushing rules of the classified materials and the design principle of the powder particles of Chinese medicine, the powder of the Xiaojin Pills was prepared. At the same time, the manmade mixed powder and the control powder prepared by pharmacopoeia were prepared. The mixed homogeneity of the three powders was evaluated by particle size distribution and color difference. The GC-MS and LC-MS/MS were used to study the homogeneity of their contents. The best preparation process of particle design powder is:materials easily crushed are smashed for 50 min in the vibrating ultrafine mill with -15℃, then add the materials difficultly crushed into the mill and let them crushed together for 3 min. The particle size range of manmade mixed powder was the largest with the particle size difference being more than 100 microns, the RSD value being 26.07%. The particle size range was more than 50 microns in the powder prepared by pharmacopoeia, and the RSD was nearly 15%. The difference in particle size was only around 4 μm and the RSD value was 3.18%. The color difference test showed that the composite chromatism (dE*) value of the powder prepared by pharmacopoeia was the largest for the RSD was 84.56%. The RSD of manmade mixed powder and the powder prepared by Pharmacopeia were 53.83% and 32.83%, respectively. The RSD value of the particle designed powder's muscone content is about 50% of the other two kinds of powders. The contents of 10 components in powders were determined by LC-MS/MS. The RSD values of the particle designed powder were much smaller than other two kinds of powders. Results indicate that the uniformity of the particle designed powder is better than other two kinds of powders. Chinese medicine particle design technology can effectively improve the uniformity of traditional Chinese medicine powder.

Increasing number of endovenous techniques are available for the treatment of saphenous vein reflux and endovenous laser ablation (EVLA) is a frequently used method. A newly developed alternative, based on thermal therapy, is endovenous microwave ablation (EMA). This study evaluated the effect of the two procedures, in terms of coagulation and histological changes, in occluding lateral veins in goats. Twelve animals were randomized into two group, with 6 treated with EMA (EMA group), and the rest 6 with EVLA (EVLA group). Results of coagulation, including coagulation, fibrinolysis and platelet activation, were assessed at three or four different time points: before, immediately after, 24 h (and 48 h) after ablation. The diameter change, a measure of efficacy, was ultrasonographically measured before and 1 month after the ablation. Histological changes were grossly and microscopically evaluated immediately, 1 and 3 month(s) after the ablation. The length of the ablated vein and preoperative average diameter were comparable between the two groups. In both EMA and EVLA groups, several coagulation parameters, fibrinolysis and platelet activation parameters only underwent slight changes. Ultrasound imaging displayed that the diameter reduction of the veins treated by EMA was significantly larger than by EVLA, in consistent with the results of macroscopic examination. Microscopic examination revealed necrosis and thickening of the vein wall, and occlusion of the lumen within 3 months after ablation in both EMA and EVLA groups. It is concluded that EMA is a minimally invasive therapy, which appears to be safe and effective for treatment of lateral veins in goats.

AIM:To explore and compare effect of coaxial micro-incision phacoemulsification and standard coaxial phacoemulsification in the treatment of cataract. METHODS:A total of 88 patients(88 eyes) with senile cataract who underwent selective cataract ultrasonic emulsification resorption combined with intraocular lens implantation in the hospital from Aug. 2013 and Aug. 2015 were selected, they were divided randomly into the control group ( 44 cases, 38 cases completed, 6 cases dropped out ) and the observation group ( 44 cases, 40 cases completed, 4 cases dropped out ) . The control group received standard coaxial 3. 0mm small incision phacoemulsification, the observation group received coaxial 1. 8mm micro-incision phacoemulsification. All patients were combined with intraocular lens implantation. Intraoperative effective ultrasonic time, cumulative release energy, best corrected visual acuity before and after surgery in the two groups were recorded, corneal endothelium cell population, incision corneal thickness, percentage of cornea hexagon cell, corneal astigmatism changes before the operation, 1d, 7d, 30d after the operation were compared. RESULTS: There was no significant difference in intraoperative effective ultrasonic time, cumulative release energy in the two groups(P>0. 05). There was no significant difference in postoperative best corrected visual acuity between the two groups(P>0. 05). Corneal endothelium cell population, percentage of cornea hexagon cell 1d, 7d, 30d after the operation in the two groups were significantly decreased compared with those before the treatment ( P 0. 05 ). Incision corneal thickness 30d after the operation in the observation group was significantly lower than the control group(PCONCLUSION: Coaxial micro - incision phacoemulsification and standard coaxial phacoemulsification can improve visual acuity of cataract patients, but the former can reduce degree of corneal astigmatism.

Twelve animals were randomized into two group, with 6 treated with EMA (EMA group), and the rest 6 with EVLA (EVLA group). Results of coagulation, including coagulation, fibrinolysis and platelet activation, were assessed at three or four different time points: before, immediately after, 24 h (and 48 h) after ablation. The diameter change, a measure of efficacy, was ultrasonographically measured before and 1 month after the ablation. Histological changes were grossly and microscopically evaluated immediately, 1 and 3 month(s) after the ablation. The length of the ablated vein and preoperative average diameter were comparable between the two groups. In both EMA and EVLA groups, several coagulation parameters, fibrinolysis and platelet activation parameters only underwent slight changes. Ultrasound imaging displayed that the diameter reduction of the veins treated by EMA was significantly larger than by EVLA, in consistent with the results of macroscopic examination. Microscopic examination revealed necrosis and thickening of the vein wall, and occlusion of the lumen within 3 months after ablation in both EMA and EVLA groups. It is concluded that EMA is a minimally invasive therapy, which appears to be safe and effective for treatment of lateral veins in goats.

<p><b>OBJECTIVE</b>To explore the effect of a self-made guiding needle of steel wire in guiding the wire through the tibial tunnel for the treatment of avulsion fractures of tibial posterior cruciate ligament with open reduction and wire fixation.</p><p><b>METHODS</b>From February 2011 to June 2014, a total of 22 patients with avulsion fractures of tibial posterior cruciate ligament underwent surgical treatments were analyzed, including 14 males and 8 females with an average age of 35.6 years old (ranged, 17 to 63 years old). According to Meyers classification, 9 patients were classified as type II, 13 patients were classified as type III. All the patients underwent open reduction and wire fixation with medial knee "L" shape approach. A wire guiding needle was used to guide the wire through the tibial tunnel during operation.</p><p><b>RESULTS</b>With the assistance of wire guidance needles, wires passed through the tibial tunnel rapidly during the operation in all the 22 patients. All the patients were followed up, X-ray imagings 6 months after operation showed the fractures healed well. The average follow-up time in all patients was 6 months (ranged, 6 to 12 months). The averaged Lysholm knee score in 22 knee was 92.7 +/- 3.4. All patients' posterior drawer test were negative.</p><p><b>CONCLUSION</b>Self-made wire guiding needle can simplify the operation procedures in which the wires pass through the tibial tunnel, shorten the operation time, reduce the surgical trauma and complications, and be worthy of clinical application.</p>

<p><b>OBJECTIVE</b>The objective of this study was to review the research on clinical genetics of Wilson's disease (WD).</p><p><b>DATA SOURCES</b>We searched documents from PubMed and Wanfang databases both in English and Chinese up to 2014 using the keywords WD in combination with genetic, ATP7B gene, gene mutation, genotype, phenotype.</p><p><b>STUDY SELECTION</b>Publications about the ATP7B gene and protein function associated with clinical features were selected.</p><p><b>RESULTS</b>Wilson's disease, also named hepatolenticular degeneration, is an autosomal recessive genetic disorder characterized by abnormal copper metabolism caused by mutations to the copper-transporting gene ATP7B. Decreased biliary copper excretion and reduced incorporation of copper into apoceruloplasmin caused by defunctionalization of ATP7B protein lead to accumulation of copper in many tissues and organs, including liver, brain, and cornea, finally resulting in liver disease and extrapyramidal symptoms. It is the most common genetic neurological disorder in the onset of adolescents, second to muscular dystrophy in China. Early diagnosis and medical therapy are of great significance for improving the prognosis of WD patients. However, diagnosis of this disease is usually difficult because of its complicated phenotypes. In the last 10 years, an increasing number of clinical studies have used molecular genetics techniques. Improved diagnosis and prediction of the progression of this disease at the molecular level will aid in the development of more individualized and effective interventions, which is a key to transition from molecular genetic research to the clinical study.</p><p><b>CONCLUSIONS</b>Clinical genetics studies are necessary to understand the mechanism underlying WD at the molecular level from the genotype to the phenotype. Clinical genetics research benefits newly emerging medical treatments including stem cell transplantation and gene therapy for WD patients.</p>

OBJECTIVE: To analyze the sociodemographic and clinical factors related to anxiety in patients with major depressive disorder (MDD). METHODS: This study involved a secondary analysis of data obtained from the Diagnostic Assessment Service for People with Bipolar Disorders in China (DASP), which was initiated by the Chinese Society of Psychiatry (CSP) and conducted from September 1, 2010 to February 28, 2011. Based on the presence or absence of anxiety-related characteristics, 1,178 MDD patients were classified as suffering from anxious depression (n=915) or non-anxious depression (n=263), respectively. RESULTS: Compared with the non-anxious group, the anxious-depression group had an older age at onset (t=-4.39, p<0.001), were older (t=-4.69, p<0.001), reported more lifetime depressive episodes (z=-3.24, p=0.001), were more likely to experience seasonal depressive episodes (chi2=6.896, p=0.009) and depressive episodes following stressful life events (chi2=59.350, p<0.001), and were more likely to have a family history of psychiatric disorders (chi2=6.091, p=0.014). Their positive and total scores on the Mood Disorder Questionnaire (MDQ) and the 32-item Hypomania Checklist (HCL-32) (p<0.05) were also lower. The logistic regression analysis indicated that age (odds ratio [OR]=1.03, p<0.001), a lower total MDQ score (OR=0.94, p=0.011), depressive episodes following stressful life events (OR=3.04, p<0.001), and seasonal depressive episodes (OR=1.75, p=0.039) were significantly associated with anxious depression. CONCLUSION: These findings indicate that older age, fewer subclinical bipolar features, an increased number of depressive episodes following stressful life events, and seasonal depressive episodes may be risk factors for anxiety-related characteristics in patients with MDD.
