RESUMO

Amino acid (AA) metabolism plays a vital role in the central metabolism of plants. In addition to protein biosynthesis, AAs are involved in secondary metabolite biosynthesis, signal transduction, stress response, defense against pathogens, flavor formation, and so on. Besides these functions, AAs can be degraded into precursors or intermediates of the tricarboxylic acid cycle to substitute respiratory substrates and restore energy homeostasis, as well as directly acting as signal molecules or be involved in the regulation of plant signals to delay senescence of postharvest horticultural products (PHPs). AA metabolism and its role in plants growth have been clarified; however, only a few studies about their roles exist concerning the postharvest preservation of fruit and vegetables. This study reviews the potential functions of various AAs by comparing the difference in AA metabolism at the postharvest stage and then discusses the crosstalk of AA metabolism and energy metabolism, the target of rapamycin/sucrose nonfermenting-related kinase 1 signaling and secondary metabolism. Finally, the roles and effect mechanism of several exogenous AAs in the preservation of PHPs are highlighted. This review provides a comprehensive insight into the AA metabolism network in PHPs. © 2023 Society of Chemical Industry.

RESUMO

Objective To explore the clinical effect of clinical observation of supplemented wendan decoction combined with Western medicine in treating insomnia accompanied by anxiety and depression in phlegm-heat internal disturbance syndrome.Methods A total of 120 cases of insomnia with anxiety and depression comorbiditis with phlegm heat disturbance syndrome were randomly divided into control group and treatment group,60 cases in each group.The control group was given escitalopram oxalate tablet combined with dexzopiclonone tablet,and the observation group was given added Wendan decoction on the basis of the control group.Both groups were treated continuously for 6 weeks.Polysomnography monitoring parameters and heart rate variability were compared between the two groups during baseline period and visit 2(baseline period+3 months).Scale scores of the two groups were compared during baseline period,visit 1(baseline period+6 weeks)and visit 2.The content of heart rate variability includes:time domain analysis(standard deviation of normal interval(SDNN),square root of the square sum of the mean of the difference between adjacent normal interval(RMSSD)and frequency domain analysis(LF,HF,LF/HF).The scale scores included the Pittsburgh Sleep Quality Index(PSQI)and Insomnia Severity Index(ISI)to assess sleep status,and the Hamilton Depression Scale(HAMD),Hamilton Anxiety Scale(HAMA),Self-assessment Scale for Depression(PHQ-9)and Generalized Anxiety Disorder Scale(GAD-7)to assess anxiety and depression status.Results(1)Polysomnography monitoring:the wake time of observation group was significantly shorter than that of control group,the number of awakenings was significantly less than that of control group,and the percentage of N3 and REM was significantly higher than these of control group(P<0.05).(2)Heart rate variability:RMSSD and HF values in the observation group were significantly higher than those in the control group,and LF/HF values were significantly lower than those in the control group(P<0.05).(3)In terms of sleep:during the interview,PSQI total score,sleep quality,hypnotic drugs and daytime dysfunction in the observation group were significantly lower than those in the control group(P<0.05);At the 3 months,the sleep quality,hypnotic drugs and daytime dysfunction in the observation group were significantly lower than those in the control group(P<0.05).In terms of emotion:HAMA,HAMD and GAD-7 scores were significantly lower than those of control group at 6 weeks(P<0.05);At the 3 months,HAMA and GAD-7 scores were significantly lower than those of control group(P<0.05).Conclusion Supplemented Wendan decoction combined with western medicine can obviously optimize the sleep structure of insomnia patients with anxiety and depressionof phlegm-heat disturbance syndrome,improve sleep continuity and deepen sleep depth,and improve parasympathetic functional activities,contribute to sympathetic parasympathetic balance,can improve insomniaand depression symptoms recently,and significantly improve anxiety symptoms in the short term,with good safety.

Objective: Nonalcoholic fatty liver disease (NAFLD) has become a common chronic liver disease that is harmful to human health. Moreover, there is currently no FDA-approved first-line drug for the treatment of nonalcoholic steatohepatitis (NASH) or NAFLD. Traditional Chinese medicine (TCM) is widely used to ameliorate liver diseases, such as the traditional ancient recipe called Three Flower Tea (TFT), which consists of double rose (Rosa rugosa), white chrysanthemum (Chrysanthemum morifolium), and Daidaihua (Citrus aurantium). However, the mechanisms of the action of TFT are not clear. Therefore, this study aimed to elucidate the mechanisms of TFT against NAFLD in high-fat diet (HFD)-induced rats. Methods: This study utilized bioinformatics and network pharmacology to establish the active and potential ingredient-target networks of TFT. Furthermore, a protein–protein interaction (PPI) network was constructed, and enrichment analysis was performed to determine the key targets of TFT against NAFLD. Furthermore, an animal experiment was conducted to evaluate the therapeutic effect and confirm the key targets of TFT against NAFLD. Results: A total of 576 NAFLD-related genes were searched in GeneCards, and under the screening criteria of oral bioavailability (OB) ≥30% and drug-likeness (DL) ≥0.18, a total of 19 active ingredients and 210 targets were identified in TFT. Network pharmacology analysis suggested that 55 matching targets in PPIs were closely associated with roles for NAFLD treatment. Through the evaluation of network topology parameters, four key central genes, PPARγ, SREBP, AKT, and RELA, were identified. Furthermore, animal experiments indicated that TFT could reduce plasma lipid profiles, hepatic lipid profiles and hepatic fat accumulation, improve liver function, suppress inflammatory factors, and reduce oxidative stress. Through immunoblotting and immunofluorescence analysis, PPARγ, SREBP, AKT, and RELA were confirmed as targets of TFT in HFD-induced rats. Conclusion: In summary, our results indicate that TFT can prevent and treat NAFLD via multiple targets, including lipid accumulation, antioxidation, insulin sensitivity, and inflammation.

Objective To establish the fingerprint spectrum and assay three active components (hesperidin, salvianolic acid B and chrysophanol) in Shenshuaining granule by HPLC method. Methods The chromatographic separation was achieved on SunFireTM C18 column with acetonitrile-0.1% formic acid aqueous solution as mobile phase. Gradient elution program was applied with flow rate of 1.0 ml/min, detection wavelength at 254 nm and the column temperature at 25 ℃. The fingerprint spectrum was established and three active components in Shenshuaining granule were assayed. Results There were 22 common peaks on the fingerprints after analyzing chromatograms from 10 batches of Shenshuaining granules. Good fingerprint similarities (≥0.9) between different batches and the control chromatogram were found. This method has great repeatability, stability and precision, which meets all the assay requirements. Conclusion A simple and reliable HPLC method was developed, which is suitable for the fingerprint establishment of Shenshuaining granules. It provides a method for the quality control of Shenshuaining granules.

OBJECTIVE:To identi fy chemical components of Actinidia chinensis root rapidly ,and to provide reference for further material basis and quality control study of the crude medicine. METHODS :UHPLC-Q-TOF-MS/MS technique was used to detect chemical components of A. chinensis root. The separation was performed on Waters XSelect HSS T 3 column with mobile phase consisted of 0.1% formic acid acetonitrile solution- 0.1% formic acid water solution (gradient elution )at the flow rate of 0.3 mL/min. The column temperature was set at 40 ℃,and sample size was 3 μL. Electrospray ion source was adopted,the data was collected under negative ion mode ;the scanning range was m/z 50-1 500;the drying gas temperature was 350 ℃,the atomizing air pressure was 45 psi,the capillary voltage was 3 500 V,and sheath gas temperature was 350 ℃. According to the information of excimer ion and secondary fragment ion ,the chemical components were identified by combining with the relevant literature ,the retention time of the reference substance and the law of mass spectrometry cracking. RESULTS & CONCLUSIONS :Totally 58 chemical components was identified ,which included 16 pentacyclic triterpenes (such as hydroxyasiatic acid ,asiatic acid ,maslinic acid,corosolic acid ,oleanic acid ,ursolic acid ,etc.),12 flavonoids(such as rutin ,quercitrin,cynaroside,astragalin,etc.),17 organic acids (such as cryptochlorogenic acid ,chlorogenic acid ,isochlorogenic acid A ,isochlorogenicacid C ,etc.). There were 9 components(such as procydanidin B 1,B2 and luteolin ,etc.)identified for the first time in A. chinensis root. UHPLC-Q-TOF-MS/ MS technique can be used for the rapid identification of chemical components in A. chinensis root.

RESUMO

ObjectiveTo evaluate the therapeutic effect of phacoemulsifcation and posterior chamber intraocular lens(IOL) implantation combined with trabeculectomy in patients with cataract and glaucoma.Methods26 cases(26 eyes)were theated by phacoemulsification and IOL implantation combined with trabeculectomy.Follow-up period was for 2~16 months.ResultsThe average intraocular pressure(IOP)was reducded from 24.82 mmHg preoperatively to 16.52 mmHg postoperatively.IOP was less than 21 mmHg in 88.46% eyes.Visual acuity was 0.3 or better in 80.77% of eyes during following-up.61.54% of eyes were found with functional filtering blebs.ConclusionTriple procedure is safe and suitable method for most patiants with glaucoma and cateract
