RESUMO

PURPOSE: This study aimed to develop and psychometrically evaluate a patient-reported outcome measure (PROM), SAlivary, LAcrimal, NaSal (SALANS), to document patients' symptoms after radioactive iodine (RAI) treatment for differentiated thyroid cancer (DTC). METHODS: We generated and iteratively revised SALANS items based on expert input, focus group discussions and feedback from cognitive testing (n = 17). We administered an initial SALANS measure with 39 items to patients diagnosed with DTC in the past two years (n = 105). Exploratory factor analysis (EFA) examined the factor structure of the SALANS items. We assessed the consistency reliability and related the total and subscale scores of the final SALANS to existing PROMs to assess validity. RESULTS: The final SALANS consisted of 33 items and six subscales (sialadenitis, taste, xerostomia, dry eyes, epiphora, and nasal) with six factors extracted by EFA. The six subscales demonstrated good internal reliability (α range = 0.87-0.92). The SALANS total score showed good convergent validity with the Xerostomia Inventory (r = 0.86) and good discriminant validity with a measure of spirituality (r = - 0.05). The mean SALANS total score was significantly higher (d = 0.5, p < 0.04) among patients who had RAI compared to those who did not have RAI. CONCLUSION: Preliminary evidence suggests that SALANS is a novel and reliable PROM to assess the type and frequency all symptoms experienced after RAI treatment for DTC. Future work is needed to further validate and develop the scale.

BACKGROUND: The AMPA-type ionotropic glutamate receptor mediates fast excitatory neurotransmission in the brain. A variety of auxiliary subunits regulate its gating properties, assembly, and trafficking, but it is unknown if the binding of these auxiliary subunits to the receptor core is dynamically regulated. Here we investigate the interplay of the two auxiliary subunits γ-2 and GSG1L when binding to the AMPA receptor composed of four GluA1 subunits. METHODS: We use a three-color single-molecule imaging approach in living cells, which allows the direct observation of the receptors and both auxiliary subunits. Colocalization of different colors can be interpreted as interaction of the respective receptor subunits. RESULTS: Depending on the relative expression levels of γ-2 and GSG1L, the occupancy of binding sites shifts from one auxiliary subunit to the other, supporting the idea that they compete for binding to the receptor. Based on a model where each of the four binding sites at the receptor core can be either occupied by γ-2 or GSG1L, our experiments yield apparent dissociation constants for γ-2 and GSG1L in the range of 2.0-2.5/µm2. CONCLUSIONS: The result that both binding affinities are in the same range is a prerequisite for dynamic changes of receptor composition under native conditions.

Activated biochar is a promising porous carbonaceous adsorbent material for organic pollutant removal, but it remains challenging to obtain high porosity and aromaticity through a simple and low-cost synthetic method. The common adsorption mechanisms of organic dyes on activated biochar should be further investigated in order to guide the synthesis of high-efficiency adsorbent materials. Here, we proposed a high-yield (up to 40 wt %) synthetic method of phosphoric acid-activated biochar from pomelo peel (PPC) with a high specific area of 877.3 m2/g through a facile thermal treatment at a relatively low temperature (250 °C). The specific activation mechanism of H3PO4 in the preparation of the adsorbent was investigated by a range of experiments and characterizations. The kinetic and isotherm experiments are also conducted to evaluate its dye adsorption behavior. According to the adsorption experiment results, PPC exhibits high saturated adsorption capacities for methyl orange (MO, 239.1 mg/g), rhodamine B (RhB, 2821.8 mg/g), methylene blue (MB, 580.5 mg/g), and crystal violate (CV, 396.6 mg/g) according to the Langmuir model. The maximum initial concentration of each dye solution for acquiring 90% removal efficiency is estimated to be 234.55 ppm (MO), 2943.8 ppm (RhB), 633.8 ppm (MB), and 423.6 ppm (CV) at 298 K with an adsorbent dosage of 1 g/L. The characterization results also indicate PPC has a complex synergetic mechanism for ionic dye adsorption behavior. This provides perspectives regarding PPC as a promising biochar adsorbent from biomass waste, which is probably useful for high-efficiency dye removal in water treatment.

Experimental investigations of cell size control in fission yeast Schizosaccharomyces pombe have illustrated that the cell cycle features 'sizer' and 'timer' phases which are distinguished by a growth rate changing point. Based on current biological knowledge of fission yeast size control, we propose here a model of ordinary differential equations (ODEs) for a possible explanation of the facts and control mechanism which is coupled with the cell cycle. Simulation results of the ODE model are demonstrated to agree with experimental data for the wild type and the cdc2-33 mutant. We show that the coupling of cell growth to cell division by translational control may account for observed properties of size control in fission yeast. As the translational control in the expression of cycle proteins Cdc13 and Cdc25 constructs positive feedback loops, the dynamical activities of the key components undergoes a rapid rising after a preliminary stage of slow increase. The coupling of this dynamical behavior to the elongation of the cell naturally gives rise to a rate change point and to 'sizer' and 'timer' phases, which characterize the cell cycle of fission yeast.
