RESUMO

Lung adenocarcinoma (LUAD) is one of the most common forms of lung cancer, with a very high mortality rate. Although the treatments available for LUAD have become more effective in recent years, significant improvement is still needed. Advances in sequencing technologies and bioinformatics analysis have enabled new approaches to be developed for identifying drug targets. In this work we utilized data from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) databases to identify hub genes related to LUAD through Weighted Gene Correlation Network Analysis (WGCNA) and other bioinformatics methods, with the goal of identifying new drug targets for cancer treatment.

Hydrogen evolution reaction (HER) catalyzed by molybdenum sulfide quantum dots (MoS2 QDs) has attracted extensive attention in the energy field. Monitoring HER catalyzed by MoS2 QDs based on a glass nanopore with an electrochemically confined effect was proposed for the first time. MoS2 QDs inside the glass nanopore is driven toward the orifice of the nanopore and bonded with the Ag nanoparticles (Ag NPs) to form a single nanocomposite. When enough voltage is applied across the orifice, the single Ag NP acts as a single nanoparticle electrode to conduct the electrochemically bipolar reaction on its two extremities. In the process, HER is catalyzed by MoS2 QDs, and Ag NPs are oxidized at the same time. The appearance of blockages on the elevated ionic current is attributed to the generation of a H2 bubble. Furthermore, by analyzing the modulations in the ionic current oscillation, the frequency of hydrogen bubble generation that is related to the catalytic efficiency of MoS2 QDs could be estimated. The results reveal the capability of the glass nanopore for the real-time monitoring electrocatalytic behavior, which makes the glass nanopore an ideal candidate to further reveal the heterogeneity of catalytic capability at the single particle level.

Tissue engineering technology is applicable for study of nerve regeneration after spinal cord injury. Many natural and artificial scaffold are not applicable because of poor mechanical properties and cell compatibility. Polypeptides with fine three-dimensional structure and cell compatibility and are widely used in tissue engineering research. The purpose of this study was to verify the neuronal differentiation of neural stem cells by using self-polymerize dendritic polypeptide for spinal cord tissue engineering. Neural stem cells were isolated from cerebral cortex of neonatal SD rats.Conventional media was triggered the 1wt% nano peptide solution self polymerizated to formed a nano gel. The gel was tested by scanning electron microscope and transmission electron microscope. Neural stem cells were inoculated onto gel or on Polylysine-coated slides with fetal bovine serum or not. SD rats were randomized divided into four groups. neural stem cells and self-polymerized peptide were transplanted into spinal cord injury models. Then we test the Density of NF-positive axons in the spinal cord injury area at 8 weeks after surgery and MS score of the locomotive function of hind limbs among mice of four groups. Neural stem cells were showed anti Nestin (+), anti NSE (+), anti GFAP (+). The gel tested by scanning electron microscope was showed thick wall structure, another one tested by transmission electron microscope was showed self-polymerized dendritic nanofibers, which contains several spacings. The cells in serum group were differentiate into neurons, but non serum group were not. These results suggest that the self-assembling peptide nanofiber scaffold(SAPNS) were cytocompatible to neural stem cells which were differentiated into neurons. A large number of axonal regeneration and recovery of joint function of hind limb were appeared. The self-polymerized Peptide maybe used as practical tissue engineering materials as future.

A new natural compound, dehydrophyllodulcin (1) was isolated from the tubers of Scirpus yagara, together with 11 known compounds. Among them, compounds 2, 5-8, and 10-12 were isolated from this plant for the first time. (1)H NMR, (13)C NMR, and 2D NMR data of compound 1 are first reported in this article, though it was synthesized in 1996. The structures of all compounds were determined by comprehensive analyses of their spectroscopic data and compared with literature information. Moreover, the anti-inflammatory effects of compounds 1, 3, 4, 6, and 9 against inflammatory cytokines production in Lipopolysaccharide - or Pam3csk4-stimulated macrophage RAW264.7 cells were evaluated by Enzyme-linked immunosorbent assay. And these compounds significantly inhibited the tumor necrosis factor (TNF)-α, interleukin (IL)-6 productions in RAW264.7 cells, with IC50 values less than 20 µM.

Objectives: N7-methylguanosine (m7G) plays a crucial role in mRNA metabolism and other biological processes. However, its regulators' function in Primary Sjögren's Syndrome (PSS) remains enigmatic. Methods: We screened five key m7G-related genes across multiple datasets, leveraging statistical and machine learning computations. Based on these genes, we developed a prediction model employing the extreme gradient boosting decision tree (XGBoost) method to assess PSS risk. Immune infiltration in PSS samples was analyzed using the ssGSEA method, revealing the immune landscape of PSS patients. Results: The XGBoost model exhibited high accuracy, AUC, sensitivity, and specificity in both training, test sets and extra-test set. The decision curve confirmed its clinical utility. Our findings suggest that m7G methylation might contribute to PSS pathogenesis through immune modulation. Conclusions: m7G regulators play an important role in the development of PSS. Our study of m7G-realted genes may inform future immunotherapy strategies for PSS.

BACKGROUND: To date, no one-phase survey of childhood depression has been performed in China that involves both urban and rural community children. The objective of this study was to determine the prevalence, correlates, and mental health service utilization of depressive disorders (DDs) in a community-based sample of 6-14-year-old children in south-central China. METHODS: Children (3,582) were approached through multistage sampling and interviewed using a Chinese version of the Mini International Neuropsychiatric Interview for Children and Adolescents 5.0, which is a structured interview that is administered by trained psychiatrists to obtain information from children and their guardians. RESULTS: The overall prevalence of all current DDs was found to be 2.8% (95%CI: 1.5-3.9). The risk factors for depression included being 9-14-year old, not attending school, having unmarried parents, living in a non-nuclear family (single parent or parentless family), being taken care of by people other than two parents (single parent, grandparent(s), other relatives, or others) during the past year, and not being breastfed prior to 1 year of age. Only 5.8% of the depressed children had received professional help prior to the interview. CONCLUSIONS: The prevalence of DDs among children in this part of China is relatively high compared with most figures reported in other countries. Depression in this age group has been a major public health concern, but it is often underrecognized. There is an urgent need to develop efficacious interventions aimed at the prevention and early recognition of childhood depression.

The combination of fenretinide and selenite on ovarian cancer cells was investigated to assess its effects on proliferation and ability to induce apoptosis. Our results showed that fenretinide and selenite in combination significantly suppress the proliferation of ovarian cancer cells and induced apoptosis (including reactive oxygen species generation, and the loss of mitochondrial membrane potential) compared with either drug used alone. The caspase3/9-dependent pathway was triggered significantly in combination treatment, and moreover, the AMPK pathway also mediated the apoptosis induction in fenretinide and selenite combination. Fenretinide and selenite combination treatment was demonstrated to suppress tumor growth in vivo, this drug combination has been thus found to have an enhanced anti-tumor effect on ovarian cancers cells.

OBJECTIVE: To investigate the effect of intra-articular berberine injection on the structural remodeling of subchondral bone plate and osteoprotegerin/receptor activator of nuclear factor kappa-B ligand(OPG/RANKL) system expression in rabbits with osteoarthritis(OA). METHODS: Forty 12-month-old male rabbits with an average of(2.73±0.18) kg of body weight, underwent left anterior cruciate ligament transection(ACLT), and were divided into berberine group and placebo groups after operation, 20 rabbits in each group. The berberine group received intra-articular injection of 100 µmol/L berberine 0.3 ml every week for 6 weeks. In placebo group, the same dose of 0.9% sodium chloride injection was injected into the left knee joint cavity every week for 6 weeks. Another 20 12-month-old male rabbits, weighing (2.68±0.18) kg, underwent sham operation on the left knee joint without intra-articular injection intervention (sham operation group). On the last day of the sixth week after operation, three groups of animals were sacrificed to obtain knee joint specimens. The femoral medial condyle samples were obtained for histological evaluation of cartilage and subchondral bone, Mankin scoring system was used to evaluate articular cartilage structure. Image-Pro Plus(IPP) software was used to evaluate subchondral bone plate bone volume(BV), bone volume/total volume(BV/TV), trabecular circumference(TC), mean trabecular thickness (Tb.Th). Real-time quantitative reverse transcription polymerization Enzyme chain reaction(reverse transcription-polymerase chain reaction, RT-PCR) was used to detect the mRNA expression levels of OPG and RANKL in subchondral bone tissue at 6 weeks after operation. RESULTS: The cartilage structure evaluation showed that the surface of cartilage tissue in the sham operation group was smooth and flat, and the safranin coloration was full in the full thickness of the cartilage;the cartilage tissue in the berberine group showed uneven surface layer, and the staining of safranin O was mildly decreased;the surface layer fibrosis was seen in placebo group, Safranin O faded significantly. The Mankin score in the berberine group was lower than that in placebo group(P<0.01), but higher than that in sham operation group(P<0.01). The structural evaluation of subchondral bone plate showed that the trabecular bone in sham-operated group was densely arranged;after berberine intervention, the trabeculae were closely arranged;the subchondral bone trabeculae in placebo group were relatively sparse, and the distance between trabeculae was wider. Subchondral bone plate IPP software evaluation showed that BV, BV/TV, TC, Tb.Th in berberine group were higher than those in placebo group(P<0.01), BV, BV/TV, TC, Tb.Th in berberine group were higher than those in placebo group(P<0.01), while lower than the sham operation group (P<0.01). PCR test results showed that the expression of OPG mRNA in the berberine group was significantly higher than that in placebo group(P<0.01), and OPG mRNA in the berberine group was lower than that in sham operation group (P<0.01). There was no significant difference in mRNA expression of RANKL among three groups(P>0.05);the ratio of OPG/RANKL in berberine group was higher than that in placebo group(P<0.01), but lower than that in sham operation group(P<0.01). CONCLUSION: Intra-articular injection of berberine can effectively inhibit the resorption of subchondral bone in the early stage of OA and delay the development of the disease. The specific mechanism may be that berberine maintains the balance of OPG/RANKL system by up-regulating the expression of OPG gene in subchondral bone.

Pancreatic adenocarcinoma (PAAD) is a highly dangerous malignant tumor of the digestive tract, and difficult to diagnose, treat, and predict the prognosis. As we all know, tumor and inflammation can affect each other, and thus the inflammatory response in the microenvironment can be used to affect the prognosis. So far, the prognostic value of inflammatory response-related genes in PAAD is still unclear. Therefore, this study aimed to explore the inflammatory response-related genes for predicting the prognosis of PAAD. In this study, the mRNA expression profiles of PAAD patients and the corresponding clinical characteristics data of PAAD patients were downloaded from the public database. The least absolute shrinkage and selection operator (LASSO) Cox analysis model was used to identify and construct the prognostic gene signature in The Cancer Genome Atlas (TCGA) cohort. The PAAD patients used for verification are from the International Cancer Genome Consortium (ICGC) cohort. The Kaplan-Meier method was used to compare the overall survival (OS) between the high- and low-risk groups. Univariate and multivariate Cox analyses were performed to identify the independent predictors of OS. Gene set enrichment analysis (GSEA) was performed to obtain gene ontology (GO) terms and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways, and the correlation between gene expression and immune infiltrates was investigated via single sample gene set enrichment analysis (ssGSEA). The GEPIA database was performed to examine prognostic genes in PAAD. LASSO Cox regression analysis was used to construct a model of inflammatory response-related gene signature. Compared with the low-risk group, patients in the high-risk group had significantly lower OS. The receiver operating characteristic curve (ROC) analysis confirmed the signature's predictive capacity. Multivariate Cox analysis showed that risk score is an independent predictor of OS. Functional analysis shows that the immune status between the two risk groups is significantly different, and the cancer-related pathways were abundant in the high-risk group. Moreover, the risk score is significantly related to tumor grade, stage, and immune infiltration types. It was also obtained that the expression level of prognostic genes was significantly correlated with the sensitivity of cancer cells to anti-tumor drugs. In addition, there are significant differences in the expression of PAAD tissues and adjacent non-tumor tissues. The novel signature constructed from five inflammatory response-related genes can be used to predict prognosis and affect the immune status of PAAD. In addition, suppressing these genes may be a treatment option.

OBJECTIVES: To automate image delineation of tissues and organs in oncological radiotherapy by combining the deep learning methods of fully convolutional network (FCN) and atrous convolution (AC). METHODS: A total of 120 sets of chest CT images of patients were selected, on which radiologists had outlined the structures of normal organs. Of these 120 sets of images, 70 sets (8,512 axial slice images) were used as the training set, 30 sets (5,525 axial slice images) as the validation set, and 20 sets (3,602 axial slice images) as the test set. We selected 5 published FCN models and 1 published Unet model, and then combined FCN with AC algorithms to generate 3 improved deep convolutional networks, namely, dilation fully convolutional networks (D-FCN). The images in the training set were used to fine-tune and train the above 8 networks, respectively. The images in the validation set were used to validate the 8 networks in terms of the automated identification and delineation of organs, in order to obtain the optimal segmentation model of each network. Finally, the images of the test set were used to test the optimal segmentation models, and thus we evaluated the capability of each model of image segmentation by comparing their Dice coefficients between automated and physician delineation. RESULTS: After being fully tuned and trained with the images in the training set, all the networks in this study performed well in automated image segmentation. Among them, the improved D-FCN 4s network model yielded the best performance in automated segmentation in the testing experiment, with an global Dice of 87.11%, and a Dice of 87.11%, 97.22%, 97.16%, 89.92%, and 70.51% for left lung, right lung, pericardium, trachea, and esophagus, respectively. CONCLUSION: We proposed an improved D-FCN. Our results showed that this network model might effectively improve the accuracy of automated segmentation of the images in thoracic radiotherapy, and simultaneously perform automated segmentation of multiple targets.

BACKGROUND: Immunotherapy has significantly changed the treatment prospects of non-small cell lung cancer (NSCLC). However, there is no report based on immune score to predict the overall survival (OS) of lung adenocarcinoma (LUAD) in the stages I, II, and III. Therefore, this study aimed to investigate the immune score and the prognosis-related factors of LUAD and construct a nomogram to predict the prognosis. METHODS: A total of 390 cases with lung adenocarcinoma in the stages I, II, and III were included in the study. The clinicopathological characteristics and immune scores of LUAD patients were downloaded from the TCGA database. Cox proportional hazards regression model was used to estimate hazard ratio (HR) and 95% confidence interval (CI). A Nomogram was composed of the Cox model and internally validated using 1000 bootstrap. The concordance index (c-index) and the calibration curves were used to evaluate the model. The decision curve analysis (DCA) was performed to evaluate the clinical practical value of the model. RESULTS: According to the immune score, the patients were divided into low-, medium-, and high-score groups. This study showed that compared with patients with low and medium immune scores, only patients with high immune scores had significantly improved OS (HR and 95% confidence interval (CI): 0.489 [0.324-0.737]). The C-index for OS prediction was 0.691 (95% CI, 0.646-0.736). The calibration curves for nomogram-predicted probabilities of 3- and 5-year survival have good ability for the calibration and discrimination. CONCLUSION: The high immune score was significantly correlated with better OS of patients with LUAD in the stages I, II, and III. Moreover, the nomogram of predicting prognosis may help assess the survival of LUAD patients.

OBJECTIVE: The aim of this study was to explore the role of the AI system which was designed and developed based on the characteristics of COVID-19 CT images in the screening and evaluation of COVID-19. METHODS: The research team adopted an improved U-shaped neural network to segment lungs and pneumonia lesions in CT images through multilayer convolution iterations. Then the appropriate 159 cases were selected to establish and train the model, and Dice loss function and Adam optimizer were used for network training with the initial learning rate of 0.001. Finally, 39 cases (29 positive and 10 negative) were selected for the comparative test. Experimental group: an attending physician a and an associate chief physician a read the CT images to diagnose COVID-19 with the help of the AI system. Control group: an attending physician b and an associate chief physician b did the diagnosis only by their experience, without the help of the AI system. The time spent by each doctor in the diagnosis and their diagnostic results were recorded. Paired t-test, univariate ANOVA, chi-squared test, receiver operating characteristic curves, and logistic regression analysis were used for the statistical analysis. RESULTS: There was statistical significance in the time spent in the diagnosis of different groups (P<0.05). For the group with the optimal diagnostic results, univariate and multivariate analyses both suggested no significant correlation for all variables, and thus it might be the assistance of the AI system, the epidemiological history and other factors that played an important role. CONCLUSION: The AI system developed by us, which was created due to COVID-19, had certain clinical practicability and was worth popularizing.

Arabinoxylan (BIF-60) was isolated from barley water-insoluble fiber (BIF) by ethanol precipitation at 60 % (v/v). BIF-60 was composed of xylose (48.5 %) and arabinose (30.3 %). Its average molecular weight was 1360 kDa. Methylation and 1D/2D NMR analysis showed that BIF-60 possessed ß-(l→4)-xylan as backbone, comprised of un-substituted (1,4-linked ß-Xylp, 56.9 %), mono-substituted (1,2,4-linked and 1,3,4-linked ß-Xylp, 22.1 %) and di-substituted (1,2,3,4-lin4ked ß-Xylp, 18.4 %) xylose units, as well as other residues (T-Araf-(1→, T-Xylp-(1→, →5)-Araf-(1→, →2)-Araf-(1→, →3)-Araf-(1→ and →4)-Glcp-(1→). BIF-60 exhibited shear-thinning behaviour, low gel stability and weak gelling ability at high concentrations. This work provides a theoretical and experimental basis for molecular structure and properties of the alkali-extracted arabinoxylan from barley kernel, which could guide further functional research and application of barley-derived arabinoxylan.

OBJECTIVE: To study and compare the clinical effects of Rehmannia Decoction and alendronate sodium for the treatment of primary osteoporosis. METHODS: From January 2016 to December 2017, 72 patients with primary osteoporosis who took Dihuang Decoction(DHD) orally and alendronate regularly for more than one year were randomly divided into 2 groups:experimental group and control group. The experimental group consisted of 14 males and 22 females, with an average age of(63.97±3.70) years old. The patients in the experimental group took Chinese medicine DHD, one dose each time, one time in the morning and one time in the evening, twice a week. The control group consisted of 16 males and 20 females with an average age of(63.36±3.07) years old. Patients in the control group were given alendronate 70 mg orally once a week. The basic treatment for osteoporosis remained unchanged in both groups(600 mg of calcium carbonate D3 and 0.5 µg of calcitriol capsules were taken daily). Bone mineral density (BMD) of femoral neck and lumbar vertebrae was measured by dual energy X-ray absorptiometry before and after treatment for one year. The levels of serum collagen type I C-terminal peptide (beta-CTX) and serum osteoclast (SOST) were measured before and after treatment for two groups. RESULTS: The age, bone mineral density, SOST and beta-CTX baseline values between the two groups before and after anti-osteoporosis treatment were compared. The difference was not statistically significant(P>0.05). Compared with the two groups, the BMD of femoral neck and lumbar vertebrae were increased after 1 year of anti-osteoporosis treatment. The differences were statistically significant (P<0.001). The value of serum beta-CTX was significantly lower than before. The t values were 52.002 and 50.071 respectively. The value of serum SOST was increased than that before treatment. The t values were -29.242 and -30.807 respectively. The differences were statistically significant (P<0.001). BMD of the femoral neck and lumbar spine was compared between the two groups after treatment. The P values were 0.294 and 0.478 respectively. The difference was not statistically significant (P>0.05). The serum beta-CTX values were compared between the two groups after treatment. The P value was 0.908. The serum SOST values were compared between the two groups after treatment. The P value was 0.888. The difference was not statistically significant (P>0.05). CONCLUSIONS: In this study, traditional Chinese medicine DHD is used to treat osteoporosis. It is found that DHD and alendronate have a good effect. The DHD can be used as a choice of Chinese medicine in the treatment of primary osteoporosis.

Simultaneous anterior and posterior traumatic dislocations of both hips are very rare. Only 33 cases have been previously reported in the English language literature. Although they were all due to high-energy injuries, they were hemodynamically stable and had a stable pelvic ring. We report a unique case of asymmetrical hip dislocations with an unstable pelvic ring and hemodynamic instability. A 40-year-old man was injured in a high-energy motor vehicle accident. He was hemodynamically unstable when he presented in the emergency department. Radiolographs showed asymmetrical dislocations of both hips with an unstable pelvic ring. Under general anesthesia, he had closed reduction of the dislocations of both hips, followed by temporary stabilization with an external fixator. Transcatheter arterial embolization was performed to stop active pelvic bleeding. Delayed open reduction and internal fixation was performed 12 days later with anterior and posterior plates. The patient recovered well with an uneventful post-operative course. Asymmetrical bilateral hip dislocations with pelvic ring instability caused by trauma, as presented in this case, is very rare and potentially life threatening. Prompt treatment can give a good outcome.

A novel and unique nickel-cobalt hydroxyfluorides (NiCo-HF) nanowires material is fabricated by one-pot solvothermal synthesis method for asymmetric supercapacitor. The synthesis mechanism and factors that influence the formation of the NiCo-HF nanowires have been further discussed. The as-prepared NiCo-HF electrode exhibits a high specific capacitance of 3,372.6 F g-1, and the capacitance retention of 94.3% can be achieved at a high current density of 20 A g-1 after 10,000 cycles. The outstanding electrochemical performance of the electrode can be attributed to the synergistic effect of the nanowires morphology and complicated redox process of active material. Furthermore, an asymmetric supercapacitor assembled with NiCo-HF nanowires as positive electrode and activated carbon as the negative electrode shows an ultrahigh energy density of 83.6 Wh kg-1 at a power density of 379.4 W kg-1 and an excellent cycling stability with 86.3% capacitance retention after 10,000 cycles, indicating that this novel material has great promise for potential application in energy storage device.

Although the function of the anterolateral stabilizing structures of the knee in the anterior cruciate ligament (ACL) injuries has been recognized since many years, most of orthopedic surgeons do not take the anterolateral structure into consideration when performing an ACL reconstruction. Anatomic ACL reconstruction will improve knee stability, but a small subset of patients may experience some residual anteroposterior and rotational instability. For this reason, some researchers have paid attention to the anterolateral aspects of the knee, especially the anterolateral ligament. We don't know the best time to perform ACL and ALL reconstruction. And we lack the evidence to prove which technique is the best one. So we look forward to more random controlled trial.

AIM: To observe the expression of nuclear factor-kappaB (NF-kappaB) and glucose transporter-4 (GLUT-4) in uteru of rats with gestational diabetes mellitus (GDM) and to explore the relationship between inhibition of NF-kappaB and insulin resistance. METHODS: Thirty Sprague-Dawley rats were randomly divided into 3 groups: normal pregnant control (NC) group, GDM group and PDTC group, 10 rats each group. GDM rat model was established by intraperitoneal (ip) injection of streptozotocin (STZ). PDTC (40 mg/kg) was ip injected in PDTC group.The rats were sacrificed on the 20-day of pregnancy before delivery. The uterus were taken to observe the pathological changes. The expressions of GLUT-4 and NF-kappaB in uteru tissue were detected by immunohistochemical staining and Western blot. RESULTS: The renal NF-kappaB activity in GDM group was higher significantly than that in NC group (P<0.01); and NF-kappaB activity in PDTC group was lower than that in GDM group (P<0.01). The expression of GLUT-4 in GDM group was significantly lower than that in NC group (P<0.01). However, compared with GDM group, the expression of GLUT-4 in PDTC group was increased (P<0.01). CONCLUSION: The inhibition of NF-kappaB activity can increase the expression of GLUT-4 in uterus of GDM rats, indicating that the occurrence of GDM in rats with uteru insulin resistance may be related with the intervention of NF-kappaB activity on the down regulation of the expression of GLUT-4.

OBJECTIVE: To determine the prevalence, correlates and recognition of depression among inpatients of general hospitals in Wuhan, China. METHOD: A total of 513 patients were randomly selected from 1923 inpatients from three general hospitals and evaluated with a Chinese version of the Structured Clinical Interview for Diagnostic and Statistical Manual-IV Axis I disorders by eight psychiatrists. Logistic regression was used to identify factors that were associated with depression. RESULTS: The prevalence (95% confidence interval) of all current depressive disorders and major depressive disorder (MDD) was found to be 16.2% (13.0-19.4%) and 9.4% (6.8-11.9%), respectively. The correlates for depression include higher hospital class, divorce/being widowed/separation, low family income, chronic diseases, lack of medical insurance, dwelling in rural area, suffering from severe illness and multiple hospitalization history. None of the patients with current MDD were detected, treated or referred to psychiatric consultation. CONCLUSIONS: The prevalence of depression among inpatients of general hospitals in Wuhan, China, was high. None of the depressive patients were recognized or treated for depression, indicating a serious neglect of depression in general hospitals. Our studies suggest an urgent need to improve clinicians' ability to detect and treat depression.
