RESUMO

Limb expression 1-like protein (LIX1L) might be an RNA-binding protein involved in post-transcriptional regulation. However, little is known regarding the biological function and mechanism of LIX1L in cancer cells. Here we demonstrate a clear correlation between LIX1L expression and epithelial-mesenchymal transition (EMT) markers in 81 non-small cell lung cancer (NSCLC) tissues and The Cancer Genome Atlas database, suggesting that LIX1L is a mesenchymal marker. Besides, LIX1L expression is obviously elevated in TGFß1-induced EMT NSCLC cells and enhances cell migration, invasion, anoikis resistance, epidermal growth factor receptor-tyrosine kinase inhibitor (EGFR-TKI) resistance, and proliferation. Interestingly, the increased LIX1L expression prominently localizes to the nucleoli, where it physically interacts with the key ribosome biogenesis regulator NCL protein, inducing ribosomal RNA (rRNA) synthesis in EMT NSCLC cells. NCL knockdown or inhibition of rRNA synthesis reverses the enhanced EMT functions and proliferation ability caused by LIX1L overexpression in NSCLC cells, indicating that NCL expression and rRNA synthesis participates in LIX1L-mediated biological functions during EMT. Collectively, our findings suggest that the LIX1L-NCL-rRNA synthesis axis is a novel EMT-activated mechanism. Targeting the pathway might be a therapeutic option for EMT and EGFR-TKI resistance in NSCLC.

RESUMO

High-purity, monodisperse, and low-oxygen submicron copper powder particles with particle sizes in the range of 100-600 nm were synthesized under alkaline conditions using ascorbic acid (C6H8O6) as a reductant and copper chloride (CuCl2·2H2O) as a copper source. The redox potential of the Cu-Cl-H2O system was obtained by calculations and plotted on pH-E diagrams, and a one-step secondary reduction process (Cu(ii) → CuCl(i) → Cu2O(i) → Cu(0)) was proposed to slow down the reaction rate. The commonalities and differences in the nucleation and growth process of copper powders under methionine (Met), hexadecyl trimethyl ammonium bromide (CTAB), and sodium citrate dihydrate (SSC) as protectants and without the addition of protectants are compared, and the reaction mechanism is discussed. Among them, methionine (Met) showed excellent properties and the Cu2O(i) → Cu(0) process was further observed by in situ XRD. The synthesized copper powder particles have higher particle size controllability, dispersibility, antioxidant properties, and stability, and can be decomposed at lower temperatures (<280 °C). The resistivity can reach 21.4 µΩ cm when sintered at a temperature of 325 °C for 30 min. This green and simple synthesis process facilitates industrialization and storage, and the performance meets the requirements of electronic pastes.

RESUMO

Cognitive theories suggest that patients with major depressive disorder (MDD) constantly negatively evaluate their self-referential information. Unlike Westerners with an independent self, self-representation is strongly influenced by cultural differences; the Chinese self may include others (interdependent self). This study uses a self-referential effect task combined with event-related potentials (ERP), and 34 patients with MDD and 54 healthy controls (HC) were asked to judge whether an adjective was suitable for describing the self, mother, or a public person, followed by an unexpected recognition task. They were required to judge whether a word was presented during the encoding phase. The results reveal that during the encoding phase, for both self- and mother-referential adjectives, patients with MDD endorsed fewer positive adjectives and more negative adjectives than the HCs. During the recognition phase, both groups showed a typical task effect (self = mother > other), while patients recognized more self-referential adjectives than the HCs. ERP data reveal that patients with MDD show larger P2 amplitudes during the encoding stage than healthy individuals. During the recognition phase, negative adjectives evoked larger P2 amplitudes in patients than in HCs under the self-referential condition. These findings shed important light on the information processes that may contribute to our understanding of depression and may offer implications for clinical interventions.

RESUMO

Three-dimensional (3D) printing of Cu items is a new way to build up the structured Cu materials, but 3D printing of Cu items is usually a challenge because of the high melting point, high thermal conductivity, and high light reflection rate of Cu material. In this study, the composite of Cu microspheres powder and Cu nanoparticles (micro/nano Cu powder) is used to realize the 3D printing of Cu items with the selective laser melting technology. The sintering temperature and the thermal conductivity of micro/nano Cu powder are evidently decreased due to Cu nanoparticles' addition in the micron Cu powder. The results reveal that the 3D printing of 50%/50% micro/nano Cu powder needs laser power range of 100-240 W, which is in contrast to 200-340 W for 3D printing of 100% Cu microspheres powder. Furthermore, the conductivity, mechanical strength, and density of 3D-printed Cu items are improved with the addition of Cu nanoparticles into the micron Cu powder. The increasement of 34% on electrical conductivity and 17% on tensile strength are reached by the addition of 50% Cu nanoparticles with the laser power of 240 W.

RESUMO

In this study, doxorubicin (DOX)-loaded long circulating liposomes combined with curcumin (CUR) (DOX-CUR-LCLs) were successfully prepared as a novel formulation for cancer treatment. The particle size and distribution, zeta potential, drug loading capacity, and entrapment efficiency (EE) of the preparation were characterized. The in vitro anti-tumor activities of DOX-CUR-LCLs and DOX-LCLs against A549 cells were then evaluated and compared with that of free DOX. Cytotoxicity evaluation showed that DOX-CUR-LCLs had a significantly higher antitumor activity than other DOX preparations. These results suggest that novel DOX-CUR-LCLs, combination of DOX and CUR administered in long-circulating liposomes, could improve antitumor activity.

RESUMO

Composite porous foam NiZn alloy electrodes with nano pore structure were prepared by the combination of eletrodeposition, heat treatment and HCl etching. The morphology of the electrodes was examined by scanning electron microscopy (SEM). And the component of the electrodes was analyzed by Energy Dispersive Spectrum (EDS). The specific surface area and pore size of the electrode were investigated by nitrogen adsorption. The phase constituents were analyzed by X ray diffraction (XRD), and the electrocatalytic characteristics for hydrogen evolution reaction of the electrodes in 30% (mass fraction) KOH solution were investigated by cathode polarization curve. The experimental results showed that the pores were formed on surface of the foam NiZn alloy electrodes after heat treatment at 600 °C, and with the etching by 10% HCl, nano layered structure was formed on the surface of the porous skeleton. Compared with the nickel foam, the surface area of the NiZn foam alloy electrode became larger, and the nano pore structure had good catalytic activity. At current density of 200 mA·dm-2, the hydrogen evolution overpotential of the NiZn foam alloy electrodes were reduced by 222 mV and 276 mV, respectively, through heat treatment of 600 °C and etching in 10% HCl solution, which indicated that the hydrogen evolution overpotential was effectively reduced because of the composite nano porous structure, while the activity of hydrogen evolution of the electrodes was obviously improved.

RESUMO

Pear black spot (PBS) disease, which is caused by Alternaria alternata (Aa), is one of the most serious diseases affecting sand pear (Pyrus pyrifolia Nakai) cultivation worldwide. To investigate the defense mechanisms of sand pear in response to Aa, the transcriptome of a sand pear germplasm with differential resistance to Aa was analyzed using Illumina paired-end sequencing. Four libraries derived from PBS-resistant and PBS-susceptible sand pear leaves were characterized through inoculation or mock-inoculation. In total, 20.5 Gbp of sequence data and 101,632,565 reads were generated, representing 44717 genes. Approximately 66% of the genes or sequenced reads could be aligned to the pear reference genome. A large number (5213) of differentially expressed genes related to PBS resistance were obtained; 34 microsatellites were detected in these genes, and 28 genes were found to be closely related to PBS resistance. Using a transcriptome analysis in response to PBS inoculation and comparison analysis to the PHI database, 4 genes (Pbr039001, Pbr001627, Pbr025080 and Pbr023112) were considered to be promising candidates for sand pear resistance to PBS. This study provides insight into changes in the transcriptome of sand pear in response to PBS infection, and the findings have improved our understanding of the resistance mechanism of sand pear to PBS and will facilitate future gene discovery and functional genome studies of sand pear.
