RESUMO

Animal manure is an important raw material for Agaricus bisporus production; however, it is also a reservoir for antibiotic residues, antibiotic resistance genes (ARGs), and antibiotic-resistant bacteria. Little is known about the influence of the commercial cultivation of A. bisporus on the dynamics of ARGs and the underlying mechanisms that cause their variations. In this study, we investigated the fate of 285 ARGs, 10 mobile genetic elements, and seven major categories of antibiotic residues in substrate and mushroom samples at different production phases. The results showed that commercial substrate preparation, particularly the pasteurization phase, was highly efficient in removing ARGs from the substrate. We further found that mycelium proliferation of A. bisporus contributed significantly to the removal of ARGs from the substrate and casing soil. The bacterial community is the key driver of changes in ARGs during the commercial cultivation of A. bisporus, which explained 46.67% of the variation in ARGs. Our results indicate that, despite the addition of animal manure, the risk of ARG dissemination to fruiting bodies and the environment is low. We propose that bioremediation by specific edible fungi might be a novel and promising method for scavenging antimicrobial resistance contamination from soil environment.

RESUMO

The analysis of multi-subject functional magnetic resonance imaging (fMRI) data and the extraction of accurate brain functional networks (FNs) are of great importance. However, traditional independent component analysis (ICA) methods perform analysis on multi-subject fMRI data under the condition of known or assumed classes of subjects, which may decrease its ability to extract accurate individual brain FNs. Although a previous method named clusterwise ICA (C-ICA) clusters subjects and obtains shared FNs in group-level for each class, its clustering performance on complex data is not ideal. To address the issues, we propose a novel method called independent component analysis with clustering embedded (ICA-CE) that can achieve both the estimation of individual FNs and the clustering of subjects in an unsupervised or semi-supervised manner. Using the simulated data with different properties, ICA-CE achieved better clustering performance than group ICA followed by K-means and C-ICA, and the mean accuracy of extracted individual FNs obtained by ICA-CE was greater than 90%. Using the task-related fMRI data from Human Connectome Project (HCP), our method also achieved higher clustering accuracy, while extracting task-related class-specific FNs. In summary, ICA-CE is effective in estimating accurate brain FNs while achieving the clustering of multiple subjects.Clinical Relevance- Our method is promising in estimating accurate brain functional networks for patients with brain disorders and outputting related class label for each subject.

Ofloxacin (OFL) with high biological activity and antimicrobial degradation is a kind of the typical high concentration and environmental risk antibiotics in rural sewage. In this paper, a combined rural sewage treatment facility based on anaerobic baffled reactor and integrated constructed wetlands was built and the removal performance, pathway and mechanism for OFL and conventional pollutants were evaluated. Results showed that the OFL and TN removal efficiency achieved 91.78 ± 3.93 % and 91.44 ± 4.15 %, respectively. Sludge adsorption was the primary removal pathway of OFL. Metagenomics analysis revealed that Proteobacteria was crucial in OFL removal. baca was the dominated antibiotic resistance genes (ARGs). Moreover, carbon metabolism with a high abundance was conductive to detoxify OFL to enhance system stability and performance. Co-occurrence network analysis further elucidated that mutualism was the main survival mode of microorganisms. Denitrifers Microbacterium, Geobacter and Ignavibacterium, were the host of ARGs and participated in OFL biodegradation.

Avian hepatitis E virus (HEV) is the main etiologic pathogen of chicken big liver and spleen disease which is widely prevalent in China in recent years. However, due to the lack of a highly effective culture system in vitro, a genetically engineered subunit vaccine is the main direction of vaccine development. In this study, ORF3 genes of VaHEV strain from laying hens and YT-aHEV strain from broilers were amplified, respectively, and ORF3 protein was successfully expressed by Escherichia coli prokaryotic expression system. The serum samples were collected periodically to detect avian HEV antibodies by indirect immunofluorescence after specific pathogen free chickens immunized with the two proteins and their mixed proteins, the results showed that all serum samples were positive for antibodies to avian HEV. The antibody-positive chickens were artificially challenged with the cell-adapted strain YT-aHEV strain. The chickens from the immunized control group were infected successfully; no fecal detoxification was detected in the immunized group. In this study, two representative strains of ORF3 subunit vaccines of laying hens and broilers were prepared by prokaryotic expression, the immune effects of different proteins of these were evaluated through immunization and challenge studies in vivo, which provided a new technical possibility for prevention and control of avian HEV.

Here we first present a novel cathode material of α-Ag2S for rechargeable Mg batteries. Due to the unique cation displacement reaction mechanism and the in situ generated uniformly distributed silver matrix, α-Ag2S nanostructures deliver remarkably improved magnesiation kinetics compared to conventional transition metal sulfides.

The concentrations of methoxylated polychlorinated biphenyls (MeO-PCBs) and hydroxylated polychlorinated biphenyls (OH-PCBs) were measured in the sewage sludge samples collected from twelve wastewater treatment plants in China. Two MeO-PCB congeners, including 3'-MeO-CB-65 and 4'-MeO-CB-101, were detected in three sludge with mean concentrations of 0.58 and 0.52 ng/g dry weight, respectively. OH-PCBs were detected in eight sludge samples, with an average total concentration of 4.2 ng/g dry weight. Furthermore, laboratory exposure was conducted to determine the possible source of OH-PCBs and MeO-PCBs in the sewage sludge, and their metabolism by the microbes. Both 4'-OH-CB-101 and 4'-MeO-CB-101 were detected as metabolites of CB-101 at a limited conversion rate after 5 days. Importantly, microbial interconversion between OH-PCBs and MeO-PCBs was observed in sewage sludge. Demethylation of MeO-PCBs was favored over methylation of OH-PCBs. The abundant and diverse microbes in sludge play a key role in the transformation processes of the PCB analogues. To our knowledge, this is the first report on MeO-PCBs in environmental matrices and on OH-PCBs in sewage sludge. The findings are important to understand the environmental fate of PCBs.
