Objective: To investigate the difference in the expression of Ras-associated protein 1 (Rap1) in necrotic and healthy areas of non-traumatic osteonecrosis of femoral head (NONFH) patients. Methods: Femoral head tissue samples from 30 cases of NONFH and 30 cases of traumatic osteonecrosis of the femoral head (TONFH) were collected after hip replacement surgery, respectively. No significant difference of Association Research Circulation Osseous (ARCO) staging was found between the NONFH and the TONFH groups ( Z=-0.769, P=0.442). In the NONFH group, 8 patients were ARCO stage IIIb, 10 were stage IV, and 12 were stage V, while in the TONFH ground, 11 patients were ARCO stage IIIb, 9 were stage IV, and 10 were stage V. There were 19 males and 11 females in the NONFH group, with an average age of 49.6 yr. (26-69 yr.), and 16 males and 14 females in the TONFH group, with an average age of 54.2 yr. (37-68 yr.). There was no significant difference in gender or age between the two groups ( P>0.05). Specimens were collected from different bone areas, including those from the necrotic areas (area A) and the healthy areas (area B) of the NONFH group, and those from the healthy areas (area B') of the TONFH group, i.e., the control group. Western blot and quantitative real-time reverse transcription PCR (qRT-PCR) were used to analyze the different expression of Rap1, vascular endothelial growth factor (VEGF) protein, phosphoinositide 3-kinase (PI3K), and Akt protein and their corresponding mRNA in the three areas of bone tissue. HE staining and immunohistochemisty staining were done in order to observe the morphological changes of each area. Results: Western blot results indicated that there was no statistical difference in the relative expression of Rap1, VEGF, PI3K, and Akt proteins ( P>0.05). The relative expressions of Rap1, VEGF, PI3K, and Akt proteins in the area A were lower than those in the area B and the difference was statistically significant ( P<0.05). qRT-PCR results showed that the relative expressions of Rap1, VEGF, PI3 K and Akt mRNA in area A were lower than those of area B, and a statistical difference was found ( P<0.05). The relative expression of the mRNA of Rap1, VEGF , PI3 K and Akt in area B and area B' were not significantly different ( P>0.05). HE staining and immunohistochemisty staining showed that chondrocytes decreased in the necrotic area (area A) of NONFH, chondrocytes nucleus disappeared, subchondral bone trabeculae were broken, bone trabeculae thickened, and empty bone lacunae appeared. Granulation tissues composed of new capillaries and fibrous cells have proliferated and crawled around the necrotic area. Positive expressions of the Rap1, VEGF, PI3K and Akt proteins in area A were weaker than those of the normal area. In addition, there were positive expressions of Rap1, PI3K and Akt on the trabecular bone of both area A and area B at similar intensity of expression. There were strong positive expressions of Rap1, VEGF, PI3K and Akt on the intima of arterioles and venules, and on the peripheral stromal cell membrane, but the positive expression in area A was significantly lower than that in area B. However, the positive expression positions and intensity of all indicators were similar in area B and area B'. Conclusion: The necrosis in NONFH may be related to vascular endothelial damages caused by the inhibition of the Rap1-PI3K/Akt signaling pathways and the subsequent decline in the protein expression.

The biotransformation of sulfamonomethoxine (SMM) was studied in an aerobic granular sludge (AGS) system to understand the role of sorption by microbial cells and extracellular polymeric substances (EPS) and the role of functional microbe/enzyme biodegradation. Biodegradation played a more important role than adsorption, while microbial cells covered with tightly bound EPS (TB-EPS) showed higher adsorption capacity than microbial cells themselves or microbial cells covered with both loosely bound EPS (LB-EPS) and TB-EPS. The binding tests between EPS and SMM and the spectroscopic analyses (3D-EEM, UV-Vis, and FTIR) were performed to obtain more information about the adsorption process. The data showed that SMM could interact with EPS by combining with aromatic protein compounds, fulvic acid-like substances, protein amide II, and nucleic acids. Batch tests with various substances showed that SMM removal rates were in an order of NH2OH (60.43 ± 2.21 µg/g SS) > NH4Cl (52.96 ± 0.30 µg/g SS) > NaNO3 (31.88 ± 1.20 µg/g SS) > NaNO2 (21.80 ± 0.42 µg/g SS). Hydroxylamine and hydroxylamine oxidoreductase (HAO) favored SMM biotransformation and the hydroxylamine-mediated biotransformation of SMM was more effective than others. In addition, both ammonia monooxygenase (AMO) and CYP450 were able to co-metabolize SMM. Analysis of UPLC-QTOF-MS indicated the biotransformation mechanisms, revealing that acetylation of arylamine, glucuronidation of sulfonamide, deamination, SO2 extrusion, and δ cleavage were the five major transformation pathways. The detection of TP202 in the hydroxylamine-fed Group C indicated a new biotransformation pathway through HAO. This study contributes to a better understanding of the biotransformation of SMM.

Atmospheric volatile organic compounds (VOCs) were collected from different functional zones of Lianyungang during the summer and autumn of 2018. One-hundred-seven VOCs species were measured by cryogenic pre-concentration and gas chromatography-mass spectrometry (GC/MS). The ozone generation potential (OFP) of VOCs was estimated by maximum incremental reactivity (MIR). Results showed that the average volume fraction of VOCs was (22.1±13.1)×10-9. Alkanes and alkenes from C2-C4 as well as acetone and ethyl acetate were the predominant species, accounting for 59.8%-75.8% of TVOCs. The mean volume fraction in the industrial zone was the largest[(28.4±13.5)×10-9], followed by the scenic zone[(21.7±4.4)×10-9] and the traffic and residential mixed zone[(20.8±7.2)×10-9]. The concentration of VOCs in autumn was significantly higher than that in summer. The industrial zone was the site with the highest volume fraction in autumn (35.4×10-9), while the scenic zone had the highest volume fraction in summer (21.5×10-9). Alkane, sulfur, or oxygen containing compounds and halogenated hydrocarbons were the predominant components of VOCs, accounting for 35.3%, 26.9%, and 15.6% of the total amount, respectively. Due to industrial emissions, the content of sulfur or oxygen containing compounds in the industrial zone was significantly higher than that in scenic zone and the traffic and residential mixed zone. The average ratio of T/B (toluene/benzene) indicated that vehicle and solvent use were the main sources of VOCs in the urban area. The highest OFP was found in the industrial zone, followed by the traffic and residential mixed zone and the scenic zone. Alkenes and aromatics were the largest contributors to OFP.

Online gas chromatography with flame ionization detection (GC-FID) systems were employed to observe 56 species of volatile organic compounds (VOCs) in the environmental atmosphere in Nanjing in 2015. The results showed that the annual volume fraction of VOCs was (17.49±11.35)×10-9. The highest concentration of VOCs was in April (22.21×10-9), and the lowest in July (12.39×10-9). Diurnal values of VOCs concentration had no obvious variations. Alkanes, alkenes, aromatics, and acetylene accounted for 56.51%, 11.06%, 24.62%, and 7.81%, respectively. Propane (15.26%), ethane (14.14%), acetylene (7.81%), toluene (6.97%), n-butane (6.23%), ethylene (5.23%), isobutane (4.34%), propylene (4.13%), isoprene (4.12%), and m/p-xylene (4.06%) were the most abundant VOCs species. The average ratio of toluene/benzene (T/B) was 2.02, indicating that the atmospheric VOCs were mainly affected by emissions of motor vehicles, especially gasoline vehicles. The average ratio of ethane/ethyne (E/E) was 2.24, showing that the plume had aged. The hydroxyl radical (·OH) loss rate(L·OH) and ozone formation potential(OFP)were applied to assess the chemical reactivity of VOCs. The atmospheric activity of VOCs was higher in the winter and spring than in the summer and autumn. Aromatics and alkenes were the most important contributors to L·OH and OFP. Their contributions to L·OH and OFP were 46.96% and 41.58%, and 33.73% and 39.86%, respectively. Propylene, ethylene, and xylene were the most active species.

Based on statistical activity data, emission factors, and source profiles, an emission inventory of anthropogenic-speciated VOCs in Jiangsu province in 2015 was calculated. The ozone formation potential (OFP) of VOCs was estimated by the maximum incremental reactivity (MIR). The result showed that the total anthropogenic emission amount of VOCs in Jiangsu Province was 1927.8 kt in 2015. Fossil fuel combustion, industrial processes, solvent utilization, biomass burning, vehicles, and storage and transport contributed 7.38%, 27.93%, 39.56%, 3.55%, 16.18%, and 5.39%, respectively. Suzhou, Nanjing, and Xuzhou were the three cities with the highest amount, contributing more than 200 kt of VOCs. The total amount of OFP from 56 ozone-precursor VOCs was 5429.5 kt. The contribution to OFP from the equipment manufacturing, vehicles manufacturing and building decoration was 30%-60% more than their contribution to the emissions. The coating VOC reduction would be more efficient for alleviating O3 pollution. The 10 VOC species with the largest OFP contribution were m/p-xylene, ethylene, propylene, 1,3-butadiene, toluene, o-xylene, 1-butene, ethylbenzene, 1,2,4-trimethylbenzene, and p-ethyltoluene, which contributed 75.63% of the total OFP.

An hourly vehicular emission inventory with 1 km×1 km resolution was developed for Jiangsu Province in 2015, using the COPERT model and ArcGIS technology. The improved "standard road length" method, as well as road network information and monthly, weekly, and daily variations of the congestion delay index were used to increase temporal-spatial resolution. The contributions of different vehicle types, emission standards, and road types to total vehicular emissions were analyzed. Results showed that vehicular emissions of NOx, HC, CO, PM2.5, SO2, OM, and BC were 490.9, 166.3, 1614.8, 16.9, 1.9, 3.6, and 6.7 thousand tons, respectively. On average, Suzhou and Xuzhou contributed 34%-45% of the total. Evaporative emissions of HC were 20.2 thousand tons, accounting for 12% of total HC emissions. Small passenger cars and motorcycles were the major HC and CO contributors, accounting for over 30% of total emissions. Heavy-duty diesel trucks contributed 36%-54% to NOx, PM2.5, SO2, OM, and BC emissions, representing much higher emissions than other vehicles. Heavy-duty and medium-duty diesel trucks in Suzhou and Xuzhou are the main sources of NOx and PM2.5emissions. China â ¢ diesel vehicles account for the largest share (42%-55%) of NOx, PM2.5, SO2, and BC emissions. China â ¢ heavy-duty diesel trucks and China 0 medium-duty diesel trucks are the primary and secondary contributors of NOx, PM2.5, OM, and BC emissions, with a total share of 40%-56%. China 0 motorcycles contributed about 16% to total HC and CO.

BACKGROUND: To explore the prevalence of lymphocyte subgroups CD3+ CD4+ and CD3+ CD8+ and their surface receptors NKG2D and NKG2A in patients with non-small cell lung cancer (NSCLC). MATERIALS AND METHODS: A total of 40 patients with NSCLC were divided into different groups according to different clinical factors (TNM staging, pathological patterns and genders) for assessment of relations with CD3+ CD4+ and CD3+ CD8+ and the surface receptors NKG2D and NKG2A of T lymphocytes in peripheral blood by flow cytometry. RESULTS: Patients in the advanced group had evidently lower levels of CD3+ CD4+ but markedly higher levels of CD3+ CD8+ in peripheral blood than those with early lesions (p<0.05). In addition, NSCLC patients in the advanced group had obviously higher CD3+ CD4+ NKG2D and CD3+ CD8+ NKG2A expression rates but lower CD3+ CD4+ NKG2A and CD3+ CD8+ NKG2D expression rates (p<0.05). However, there were no significant differences between NSCLC patients with different genders and pathological patterns in expression levels of lymphocyte subgroups CD3+ CD4+ and CD3+ CD8+ and their surface receptors NKG2D and NKG2A. CONCLUSIONS: Unbalanced expression of surface receptors NKG2D and NKG2A in CD3+ CD4+ and CD3+ CD8+ lymphocytes may be associated with a poor prognosis, greater malignancy and immunological evasion by advanced cancers, related to progression of lung cancer.

Oxidative stress is one of the major mechanisms implicated in carbon ion irradiation. Curcumin is a natural phenolic compound with impressive antioxidant properties. What's more, curcumin is recently proved to exert its effects partly radioprotection. In vivo, we investigated the protective effects of curcumin against (12)C(6+)radiation-induced cerebral injury. Our results showed that 4Gy heavy ion radiation-induced spatial strategy and memory decline and reduction of brain superoxide dismutase (SOD) activity levels were all consistently improved by curcumin, and the augmentation of cerebral malonaldehyde (MDA) was lowered by curcumin. Furthermore, both the cerebral cells nuclear erythroid 2-related factor 2 (Nrf2) protein and three typically recognized Nrf2 downstream genes, NAD(P)H quinine oxidoreductase 1 (NQO1), heme oxygenase-1 (HO-1), and γ-glutamyl cysteine synthetase (γ-GCS) were consistently up-regulated in curcumin-pretreated mice. Our study confirmed the antagonistic roles of curcumin to counteract radiation-induced cerebral injury in vivo and suggested that the potent Nrf2 activation capability might be valuable for the protective effects of curcumin against radiation. This provides a potential useful radioprotection dietary component for human populations.

OBJECTIVE: To explore the feasibility of pulmonary lobectomy combined with pulmonary arterioplasty by complete video-assisted thoracic surgery (VATS) in patients with lung cancer, and summarize its surgical methods. MATERIALS AND METHODS: Twenty-one patients with lung cancer in Beijing Chest Hospital Affiliated to Capital Medical University from Feb., 2010 to Jun., 2013 were selected, males and females accounting for 15 and 6 cases, respectively. Ten underwent right upper lobectomy, 5 right lower lobectomy, 4 left upper lobectomy (in which left upper sleeve lobectomy was conducted for 2) and 2 left lower lobectomy. At the same time, local resection of pulmonary arterioplasty was performed for 12 patients, and sleeve resection of pulmonary arterioplasty for 9. RESULTS: Twenty-one patients recovered well after surgery. Thoracic drainage tube was maintained for 3-8 days, with an average of 4.9 days, and hospital stays were 8-15 days, with an average of 11 days. There were no deaths in the perioperative period, and the complications like pulmonary embolism, bronchopleural fistula, chest infection and pulmonary atelectasis did not occur after surgery. CONCLUSIONS: Performance of pulmonary lobectomy and pulmonary arterioplasty together by complete VATS is a safe and effective surgical method, which can expand the indications of patients with lung cancer undergoing thoracoscopic pulmonary lobectomy, and make more patients profit from such minimally invasive treatment.
