Porcine epidemic diarrhea (PED) is a serious disease in piglets that leads to high mortality. An effective measure that provides higher IgA levels in the intestine and milk is required to decrease losses. Porcine epidemic diarrhea virus (PEDV) was dissolved in calcium alginate (Alg) and combined with chitosan (CS) via electrostatic interactions between cationic chitosan and anionic alginate to create a porous gel (Alg-CS+PEDV). The gel was used to immunize mice orally or in combination with subcutaneous injections of inactivated PEDV vaccine. At 12 and 24 days after immunization, levels of IgA and IgG in Alg-CS+PEDV were higher than with normal PEDV oral administration. At 24 days after immunization, the concentration of IFN-γ in Alg-CS+PEDV was higher than with normal PEDV oral administration. Furthermore, oral administration combining subcutaneous immunization induced higher levels of IgG and IgA than oral administration alone. Our study provides a new method for the preparation and administration of oral vaccines to achieve enhanced mucosal immunity against PEDV.

PCV2 belongs to the genus Circovirus in the family Circoviridae, whose genome is replicated by rolling circle replication (RCR). PCV2 Rep is a multifunctional enzyme that performs essential functions at multiple stages of viral replication. Rep is responsible for nicking and ligating single-stranded DNA and unwinding double-stranded DNA (dsDNA). However, the structure and function of the Rep are still poorly understood, which significantly impedes viral replication research. This study successfully resolved the structure of the PCV2 Rep ATPase domain (PRAD) using X-ray crystallography. Homologous structure search revealed that Rep belonged to the superfamily 3 (SF3) helicase, and multiple conserved residues were identified during sequence alignment with SF3 family members. Simultaneously, a hexameric PRAD model was generated for analysing characteristic structures and sites. Mutation of the conserved site and measurement of its activity showed that the hallmark motifs of the SF3 family influenced helicase activity by affecting ATPase activity and ß-hairpin just caused the loss of helicase activity. The structural and functional analyses of the PRAD provide valuable insights for future research on PCV2 replication and antiviral strategies.

BACKGROUND: Yaks are able to utilize the gastrointestinal microbiota to digest plant materials. Although the cellulolytic bacteria in the yak rumen have been reported, there is still limited information on the diversity of the major microorganisms and putative carbohydrate-metabolizing enzymes for the degradation of complex lignocellulosic biomass in its gut ecosystem. RESULTS: Here, this study aimed to decode biomass-degrading genes and genomes in the yak fecal microbiota using deep metagenome sequencing. A comprehensive catalog comprising 4.5 million microbial genes from the yak feces were established based on metagenomic assemblies from 92 Gb sequencing data. We identified a full spectrum of genes encoding carbohydrate-active enzymes, three-quarters of which were assigned to highly diversified enzyme families involved in the breakdown of complex dietary carbohydrates, including 120 families of glycoside hydrolases, 25 families of polysaccharide lyases, and 15 families of carbohydrate esterases. Inference of taxonomic assignments to the carbohydrate-degrading genes revealed the major microbial contributors were Bacteroidaceae, Ruminococcaceae, Rikenellaceae, Clostridiaceae, and Prevotellaceae. Furthermore, 68 prokaryotic genomes were reconstructed and the genes encoding glycoside hydrolases involved in plant-derived polysaccharide degradation were identified in these uncultured genomes, many of which were novel species with lignocellulolytic capability. CONCLUSIONS: Our findings shed light on a great diversity of carbohydrate-degrading enzymes in the yak gut microbial community and uncultured species, which provides a useful genetic resource for future studies on the discovery of novel enzymes for industrial applications.

A label-free fluorescence assay has been developed for sensitive and selective detection of adenosine triphosphate (ATP) by using poly(thymine) (poly T)-templated copper nanoparticles (CuNPs) as fluorescent indicator. In our design, ATP aptamer was split into two fragments, both of which were elongated with poly T strands that can be utilized as efficient template for the formation of copper nanoparticles through the reduction of copper ions by sodium ascorbate. In the presence of ATP, the two split aptamers could be dragged to form aptamer-ATP aptamer complex, which drew the poly T strands close to each other and induced a remarkable fluorescence enhancement of poly T-templated CuNPs. Thus, an elevated fluorescence enhancement of poly T-templated CuNPs was obtained with the increase in ATP concentration. Under optimized conditions, a good linear range for ATP detection was realized from 100 nM to 100 µM with a detection limit of 10.29 nM. In addition, the application of this biosensing system in complex biological matrix was demonstrated with satisfactory results. This assay provided a simple, label-free, cost-effective, and sensitive platform for the detection of ATP.

The misuse of antibiotics in veterinary medicine presents significant challenges, highlighting the need for alternative therapeutic approaches such as antibody drugs. Therefore, it is necessary to explore the application of antibody drugs in veterinary settings to reduce economic losses and health risks. This study focused on targeting the F4ac subtype of the FaeG protein, a key adhesion factor in enterotoxigenic Escherichia coli (ETEC) infections in piglets. By utilizing formaldehyde-inactivated ETEC and a soluble recombinant FaeG (rFaeG) protein, an antibody library against the FaeG protein was established. The integration of fluorescence-activated cell sorting (FACS) and a eukaryotic expression vector containing murine IgG Fc fragments facilitated the screening of anti-rFaeG IgG monoclonal antibodies (mAbs). The results demonstrate that the variable regions of the screened antibodies could inhibit K88-type ETEC adhesion to IPEC-J2 cells. Furthermore, in vivo neutralization assays in mice showed a significant increase in survival rates and a reduction in intestinal inflammation. This research underscores the potential of antibody-based interventions in veterinary medicine, emphasizing the importance of further exploration in this field to address antibiotic resistance and improve animal health outcomes.

Foot-and-mouth disease virus (FMDV) 2C protein is a conserved non-structural protein and crucial for replication of the virus. In this study, FMDV 2C protein was prepared and the enzymatic activities were investigated in detail. The protein could digest ssDNA or ssRNA into a small fragment at about 10 nt, indicating that the protein has nuclease activity. But it did not show digestion to blunt-end dsDNA or dsRNA. The nuclease activity of 2C protein could be inhibited in 2 mM Zn2+ or Ca2+ while enhanced by Mg2+ or Mn2+. FMDV 2C protein exhibited unwinding activity to all the three kinds of dsDNA and dsRNA (5' protruded, 3' protruded, and blunt-end). The unwinding velocity to 5' protruded dsRNA was higher than to the blunt-end dsRNA. 2C protein only showed unwinding activity in high concentration of Mg2+, but no unwinding activity in physiological concentrations of Mg2+ and Ca2+, as well as in cell lysate. The 2C protein could catalyze two structured ssRNA to form double strand, thus it was proved to have RNA chaperone activity. The Mg2+ and ATP in different concentrations did not show promotion to the RNA chaperone activity. Finally, six mutant proteins (K116A, D160A, D170A, N207A, R226A, and F316A) were constructed and the enzymatic activities were analyzed. All the six mutations reduced the ATPase activity, D170A and F361A could inactivate the nuclease activity, while the N207A and F316A could inactivate the helicase activity. Our study provides a comprehensive understanding of the enzymatic activities of FMDV 2C protein.

The damping properties of polybutyl methacrylate (PBMA)/aromatic petroleum resin (C9) composite were investigated in this work. In particular, a trace of styrene (St) was introduced to copolymerize with PBMA to improve the compatibility between C9 and matrix. The structure of the copolymer, P(BMA-co-St), was characterized by FTIR and 1HNMR. The P(BMA-co-St)/C9 composites were tested by differencial scanning calorimetry (DSC), scanning electron microscopy (SEM) and dynamical mechanical analysis (DMA). DSC curves of all P(BMA-co-1wt%St)/C9 composites expressed only one glass transition temperature (Tg). SEM images showed that C9 had good compatibility with the matrix after St was introduced. DMA curves indicated that the addition of C9 had a positive effect on the damping properties of PBMA. The loss tangent (tanδ) peak moved to a higher temperature with the increment content of C9, and the effective damping temperature range increased significantly. The influence of aromatic resin C9 and aliphatic resin (C5) on PBMA damping performance was compared. It was further shown that C9 with benzene ring effectively improved the damping performance of PBMA.

Tibetan pig is an important domestic mammal, providing products of high nutritional value for millions of people living in the Qinghai-Tibet Plateau. The genomes of mammalian gut microbiota encode a large number of carbohydrate-active enzymes, which are essential for the digestion of complex polysaccharides through fermentation. However, the current understanding of microbial degradation of dietary carbohydrates in the Tibetan pig gut is limited. In this study, we produced approximately 145 gigabases of metagenomic sequence data for the fecal samples from 11 Tibetan pigs. De novo assembly and binning recovered 322 metagenome-assembled genomes taxonomically assigned to 11 bacterial phyla and two archaeal phyla. Of these genomes, 191 represented the uncultivated microbes derived from novel prokaryotic taxa. Twenty-three genomes were identified as metagenomic biomarkers that were significantly abundant in the gut ecosystem of Tibetan pigs compared to the other low-altitude relatives. Further, over 13,000 carbohydrate-degrading genes were identified, and these genes were more abundant in some of the genomes within the five principal phyla: Firmicutes, Bacteroidetes, Spirochaetota, Verrucomicrobiota, and Fibrobacterota. Particularly, three genomes representing the uncultivated Verrucomicrobiota encode the most abundant degradative enzymes in the fecal microbiota of Tibetan pigs. These findings should substantially increase the phylogenetic diversity of specific taxonomic clades in the microbial tree of life and provide an expanded repertoire of biomass-degrading genes for future application to microbial production of industrial enzymes.

In this paper, the rational design and synthesis of ZIF-8-derived ternary ZnO/ZnCo2O4/NiO wrapped by nanosheets is introduced. Polyhedral ternary ZnO/ZnCo2O4/NiO composites surrounded by nanosheets with different compositions are successfully fabricated through in situ growth on ZIF-8 templates and subsequent thermal annealing in air. Electrochemical investigation reveals that when the molar ratio of nickel nitrate to cobalt nitrate is 1, the composite material is more outstanding, which shows a high specific capacitance of 1136.4 F g-1 at 1 A g-1 and excellent cycling stability of 86.54% after 5000 cycles. Moreover, the excellent performance of this material is also confirmed by assembling an asymmetric supercapacitor. The assembled hybrid device can reach a large potential range of 0-1.6 V and deliver a high energy density of 46.04 W h kg-1 as well as the maximum power density of 7987.5 W kg-1.

In recent years, the electrochemical properties of supercapacitors have been greatly improved due to continuous improvement in their composite materials. In this study, an urchin-like MgCo2O4@PPy/NF (MgCo2O4@polypyrrole/Ni foam) core-shell structure composite material was successfully developed as an electrode for supercapacitors. The MCP-2 composite material, obtained by a hydrothermal method and in situ chemical oxidative polymerization, shows a high specific capacitance of 1079.6 F g-1 at a current density of 1 A g-1, which is much higher than that of MC (783.6 F g-1) under the same conditions. Simultaneously, it has low resistance and an excellent cycling stability of 97.4% after 1000 cycles. Furthermore, an all-solid-state asymmetric supercapacitor (ASC) was assembled using MCP-2 as the positive electrode and activated carbon (AC) as the negative electrode. The MCP-2//AC ASC exhibits high specific capacitance (94 F g-1 at a current density of 0.4 A g-1), high energy density (33.4 W h kg-1 at a power density of 320 W kg-1), high volumetric energy density (17.18 mW h cm-3 at a volumetric power density of 0.16 W cm-3) and excellent cycling stability (retaining 91% of the initial value after 10 000 cycles). Simultaneously, the device has low leakage current and excellent self-discharge characteristics. All these results indicate that the MCP-2//AC ASC is a good energy storage device; it can support the function of two LEDs for 20 minutes. These results indicate that the MCP-2//AC ASC will play an important role in energy structures in the future.

Viscoelastic polymers can be used as damping materials to control unexpected vibration and noise through energy dissipation. To investigate the effect of an inorganic filler on damping property, a series of inorganic particles, Ferriferous oxide(Fe3O4), Graphene/Fe3O4(GF), and Fe3O4 of demagnetization(α-Fe2O3) were incorporated into poly(butyl-methacrylate) (PBMA). The effects of the dispersion of particles, as well as the interaction between particles and the PBMA matrix on the damping property of composites, were systematically studied. Results revealed that the addition of three types of particles can effectively improve the damping properties and broaden the effective damping temperature range. Dispersion of α-Fe2O3 in the PBMA matrix is better than that of Fe3O4. As a result, the damping peak can be increased more. The interaction between GF and the PBMA matrix is stronger than that between Fe3O4 and the PBMA. The damping peak of the composites can be suppressed by GF, which is opposite to Fe3O4 and α-Fe2O3. In addition, glass transition temperature (Tg) of all composites in the study shifted to low temperatures.

Zeolitic imidazolate frameworks have stimulated great attention due to their potential applications in energy storage, catalysis, gas sensing, drug delivery etc. In this paper, the three-dimensional porous nanomaterial Co3O4/ZnCo2O4/CuO with hollow polyhedral nanocage structures and highly enhanced electrochemical performances was synthesized successfully by a zeolitic imidazolate framework-67 route. The composites hold the shape of the ZIF-67 templates well and the shell has multiple compositions. In the process, we first synthesized the nanostructure hydroxide precursors and then transformed them into the corresponding metal oxide composites by thermal annealing in air. In addition, the mass ratio of Zn to Cu in this material is discussed and optimized. We found that when the mass ratio is 3, the composite material has better electrochemical properties. When applied as an electrode material, Co3O4/ZnCo2O4/CuO-1 shows enhanced pseudocapacitive properties and good cycling stability compared with Co3O4/ZnCo2O4, Co3O4/CuO and Co3O4/ZnCo2O4/CuO-2, and Co3O4/ZnCo2O4/CuO-3. The assembled Co3O4/ZnCo2O4/CuO-1//AC hybrid device can be reversibly cycled in a large potential range of 0-1.6 V and can deliver a high energy density of 35.82 W h kg-1 as well as the maximum power density of 4799.25 W kg-1.
