The natriuretic peptide (NP) family consists of cardiac NPs (ANP, BNP, and VNP) and brain NPs (CNPs) in teleosts. In addition to CNP1-4, a paralogue of CNP4 (named CNP4b) was recently discovered in basal teleosts including Japanese eel. Mammals have lost most Cnps during the evolution, but teleost cnps were conserved and diversified, suggesting that CNPs are important hormones for maintaining brain functions in teleost. The present study evaluated the potency of each Japanese eel CNP to their NP receptors (NPR-A, NPR-B, NPR-C, and NPR-D) overexpressed in CHO cells. A comprehensive brain map of cnps- and nprs-expressing neurons in Japanese eel was constructed by integrating the localization results obtained by in situ hybridization. The result showed that CHO cells expressing NPR-A and NPR-B induced strong cGMP productions after stimulation by cardiac and brain NPs, respectively. Regarding brain distribution of cnps, cnp1 is engaged in the ventral telencephalic area and periventricular area including the parvocellular preoptic nucleus (Pp), anterior/posterior tuberal nuclei, and periventricular gray zone of the optic tectum. cnp3 is found in the habenular nucleus and prolactin cells in the pituitary. cnp4 is expressed in the ventral telencephalic area, while cnp4b is expressed in the motoneurons in the medullary area. Such CNP isoform-specific localizations suggest that function of each CNP has diverged in the eel brain. Furthermore, the Pp lacking the blood-brain barrier expressed both npra and nprb, suggesting that endocrine and paracrine NPs interplay for regulating the Pp functions in Japanese eels.

The objective of this study was to evaluate the effectiveness of foot and mouth disease (FMD) control strategies using multi-criteria decision analysis (MCDA). The FMD control strategies were evaluated for epidemiological, economic and social-environmental criteria, accounting for the preferences of Chief Veterinary Officers (CVOs) from the Asia-Oceania region, with the preferences quantified using a questionnaire study. Previously reported simulation results of an FMD epidemic were used to measure the epidemiological effectiveness of FMD control strategies. The simulated FMD epidemic results, such as the number of infected farms, the duration of the epidemic and the outbreak area, were compared for a number of FMD control strategies. Based on previous simulation and economic modelling results and incorporating a literature search to quantify the social and environmental outcomes of an FMD outbreak, MCDA analysis was conducted to evaluate control strategies. According to the overall score in the MCDA results, an FMD control strategy which consisted of a 3-km radius of pre-emptive slaughter, 100 days of movement restriction and vaccinating all FMD-susceptible animals (baseline strategy) was the preferred strategy, with the highest overall score of 62.99. In contrast, the same duration of movement restriction and vaccination strategy with a 0.5-km radius of pre-emptive slaughter was least preferred, with the lowest overall score of 37.05. The evaluation of the FMD control strategies presented here using MCDA has provided scientific evidence for selecting an appropriate baseline FMD control strategy, based on the preferences of CVOs.
Les auteurs présentent les résultats d'une étude qui visait à évaluer l'efficacité d'un certain nombre de stratégies de lutte contre la fièvre aphteuse en recourant à une analyse décisionnelle multicritères (ADMC). Diverses stratégies de lutte contre la fièvre aphteuse ont été évaluées au regard de critères épidémiologiques, économiques et socio-environnementaux, en prenant en compte les préférences exprimées par les chefs des Services vétérinaires de la région Asie-Océanie, recueillies et quantifiées lors d'une étude par questionnaire. L'efficacité des stratégies de lutte contre la fièvre aphteuse au plan épidémiologique a été évaluée à partir des résultats d'une simulation d'épidémie de fièvre aphteuse effectuée au préalable. Les résultats de la simulation, notamment le nombre d'exploitations infectées, la durée de l'épidémie et l'extension de la zone focale, ont été comparés au regard de chacune des stratégies de lutte pouvant être appliquées contre la fièvre aphteuse. Une ADMC a été réalisée pour évaluer les stratégies de lutte en s'appuyant sur les résultats de simulations antérieures et de modélisations économiques, ainsi que sur les données réunies lors d'une recherche documentaire visant à quantifier les répercussions sociales et environnementales d'un foyer de fièvre aphteuse. Il ressort du classement des notes globales attribuées par l'ADMC que la stratégie préférée de lutte contre la fièvre aphteuse (note globale de 62,99) était celle associant l'abattage préventif dans un rayon de 3 km à une restriction des déplacements d'animaux pendant une période de 100 jours et à la vaccination de toutes les espèces sensibles au virus de fièvre aphteuse (stratégie de référence). En revanche, la stratégie associant une restriction des déplacements d'animaux de même durée, la vaccination et l'abattage préventif dans un rayon de 0,5 km recueillait le score le plus bas dans l'ordre de préférence (note globale de 37,05). Les évaluations des stratégies de lutte contre la fièvre aphteuse présentées par les auteurs à partir des résultats de l'ADMC fournissent des éléments scientifiques probants pour choisir une stratégie de référence appropriée afin de lutter contre la fièvre aphteuse, fondée sur les préférences des chefs des Services vétérinaires.
Los autores describen un estudio encaminado a evaluar la eficacia de distintas estrategias de control de la fiebre aftosa, para lo cual se siguió un proceso de análisis de decisiones con empleo de múltiples criterios de tipo epidemiológico, económico y socioambiental que recogían las preferencias de los jefes de los Servicios Veterinarios de la región Asia-Oceanía (preferencias cuantificadas con empleo de un cuestionario). Para medir la eficacia epidemiológica de las estrategias de lucha se utilizaron los resultados previamente descritos de simulaciones de una epidemia de fiebre aftosa. Después se compararon entre sí los resultados (por ejemplo, número de explotaciones infectadas, duración de la epidemia o territorio afectado por el brote) que deparaban distintas estrategias de lucha en una epidemia simulada de fiebre aftosa. A partir de los resultados de simulaciones y modelizaciones económicas anteriores e incorporando una búsqueda bibliográfica para cuantificar los efectos sociales y ambientales de un brote de fiebre aftosa, se procedió a evaluar las estrategias de lucha empleando el análisis de decisiones por criterios múltiples. A tenor de la puntuación global que arrojaban los resultados de este análisis, se determinó que la estrategia preferible de lucha contra la fiebre aftosa era la consistente en sacrificios sanitarios preventivos en un radio de 3 kilómetros, restricciones de desplazamientos durante 100 días y vacunación de todos los animales sensibles a la enfermedad (estrategia básica), pues era la que deparaba la puntuación global más elevada (62,99). La menos eficaz, es decir, la que deparaba la puntuación global más baja (37,05), preveía la aplicación de restricciones de desplazamientos durante el mismo período e idéntica estrategia de vacunación, pero con sacrificios sanitarios preventivos en un radio de solo 0,5 kilómetros. La evaluación de estrategias de lucha mediante un análisis de decisiones por criterios múltiples que aquí se presenta ha aportado sólidos datos científicos con los que seleccionar, partiendo de las preferencias de los jefes de los Servicios Veterinarios, una estrategia básica adecuada para combatir la fiebre aftosa.

Resistance to treatments with Temephos or plant derived oil, Pb-CVO, between a field collected Wild Strain (WS) and a susceptible Laboratory Strain (LS) of Ae. aegypti were measured. The Temephos (0.1mg/L) showed the greatest percentage of mosquito mortality compared to Pb-CVO (1.5mg/L) in LS Ae. aegypti. However, WS Ae. aegypti was not significantly affected by Temephos (0.1mg/L) treatment compare to the Pb-CVO (1.5mg/L). However, both strains (LS and WS) when treated with Pb-CVO (1.5mg/L) displayed steady larval mortality rate across all instars. The LC50 of Temephos was 0.027mg in LS, but increased in WS to 0.081mg/L. The LC50 of Pb-CVO treatment was observed at concentrations of 0.72 and 0.64mg/L for LS and WS strains respectively. The enzyme level of α- and ß-carboxylesterase was reduced significantly in both mosquito strains treated with Pb-CVO. Whereas, there was a prominent deviation in the enzyme ratio observed between LS and WS treated with Temephos. The GST and CYP450 levels were upregulated in the LS, but decreased in WS, after treatment with Temephos. However, treatment with Pb-CVO caused both enzyme levels to increase significantly in both the strains. Visual observations of the midgut revealed cytotoxicity from sub-lethal concentrations of Temephos (0.04mg/L) and Pb-CVO (1.0mg/L) in both strains of Ae. aegypti compared to the control. The damage caused by Temephos was slightly less in WS compared to LS mosquito strains.

Previous studies have established the subventricular (SVZ) and subgranular (SGZ) zones as sites of neurogenesis in the adult forebrain (Doetsch et al., 1999a; Doetsch, 2003a). Work from our laboratory further indicated that midline structures known as circumventricular organs (CVOs) also serve as adult neural stem cell (NSC) niches (Bennett et al., 2009, 2010). In the quiescent rat brain, NSC proliferation remains low in all of these sites. Therefore, we recently examined whether ischemic stroke injury (MCAO) or sustained intraventricular infusion of the mitogen bFGF could trigger an up-regulation in NSC proliferation, inducing neurogenesis and gliogenesis. Our data show that both stroke and bFGF induce a dramatic and long-lasting (14day) rise in the proliferation (BrdU+) of nestin+Sox2+GFAP+ NSCs capable of differentiating into Olig2+ glial progenitors, GFAP+nestin-astrocyte progenitors and Dcx+ neurons in the SVZ and CVOs. Moreover, because of the upsurge in NSC number, it was possible to detect for the first time several novel stem cell niches along the third (3V) and fourth (4V) ventricles. Importantly, a common feature of all brain niches was a rich vasculature with a blood-brain-barrier (BBB) that was highly permeable to systemically injected sodium fluorescein. These data indicate that stem cell niches are more extensive than once believed and exist at multiple sites along the entire ventricular system, consistent with the potential for widespread neurogenesis and gliogenesis in the adult brain, particularly after injury. We further suggest that because of their leaky BBB, stem cell niches are well-positioned to respond to systemic injury-related cues which may be important for stem-cell mediated brain repair.

BACKGROUND: Patients on hemodialysis are particularly vulnerable to central venous occlusion (CVO). Endovascular treatment has gained wide acceptance for the treatment of CVO. However, difficulties in crossing the occluded segment can be encountered during conventional endovascular management. Sharp recanalization has been adopted when conventional endovascular methods could not recanalize the obstructed region. This study aimed to assess the outcome of the sharp venous recanalization technique with angioplasty and stenting in the treatment of CVO in Egyptian patients undergoing hemodialysis. METHODS: This retrospective study is based on data from a prospectively maintained department database of patients under regular hemodialysis who underwent the sharp venous recanalization technique for CVO. Routinely, the patients were followed up at 3, 6, and 12 months with a clinical examination. The primary outcomes were technical success and primary patency. Secondary outcomes included complication rates and clinical success. RESULTS: This study included 40 patients. Thirty-six patients (90%) achieved technical and clinical success. Seven patients (17.5%) had immediate postoperative complications. Four cases had minor complications (10%) and three patients had major complications (7.5%): hemothorax in two patients (5.1%) and pneumothorax in one patient (2.6%). At the 1-year follow-up, reintervention was required in nine patients (22.5%), with primary patency rate of 77.5% and a secondary patency rate of 100%. CONCLUSIONS: Sharp recanalization offers a solution for patients undergoing hemodialysis who developed CVO and failed to be recanalized using the conventional endovascular method. It offered promising technical success, clinical improvement, and good primary patency rates.

Vanadium pentoxide (V2O5) is considered a promising material for electrochromic supercapacitors due to its rich color transitions and excellent electrochemical capacity. However, V2O5 exhibits low electrical conductivity, and its volume changes dramatically during charge-discharge cycles, leading to structural collapse and poor long-term cyclability. These issues have hindered the development and application of V2O5. In this study, copper vanadium oxide yolk-shell microspheres (CVO) were synthesized through a one-step solvent heat treatment with an annealing process. With the doping of copper element, the capacitance, conductivity, and cyclic stability of CVO microspheres were significantly enhanced. Subsequently, the sphere-wire network structure was formed by blending Na2V6O16·3H2O nanowires (NVO), resulting in the formation of CVO/NVO composites. The three-dimensional sphere-wire network efficiently facilitates the acquisition of additional redox sites and strengthens the material-to-substrate bonding. Under the combined influence of these favorable factors, CVO/NVO achieved a high specific capacitance of 39.2 mF cm-2, with a capacitance retention of 84% after 7500 cycles at a current density of 0.7 mA cm-2. The fully inorganic solid-state electrochromic supercapacitor (ECSC), assembled on the basis of CVO/NVO, demonstrates a vivid and clearly distinguishable color change (ΔE* = 37). Even more impressive is the energy storage capacity (18.4 mF·cm-2) and the cycling stability (up to 89% retention after 10,000 cycles) exhibited by the devices. These key performances are superior to those of most of the previously reported V2O5-based ECSCs, opening a promising avenue for the development of V2O5-based electrochromic energy storage devices.

Purpose: To investigate a correlation between the central venous minus arterial CO2 pressure to arterial minus central venous O2 content ratio (Pcv-aCO2/Ca-cvO2) combined with optic nerve sheath diameter (ONSD) in predicting prehospital elevated intracranial pressure (ICP) in traumatic brain injury (TBI) patients. Patients and Methods: This was a prospective observational study of all adult TBI patients from the surgical intensive care unit who underwent invasive ICP monitoring between January 2023 and December 2023. Using a Delica MVU-6300 machine with 14-5 MHz linear probe to measure ONSD. We drew blood samples for arterial and central venous blood gases to measure and calculate the following indicators such as Pcv-aCO2, Ca-cvO2, and Pcv-aCO2/Ca-cvO2 ratio. ONSD and Pcv-aCO2/Ca-cvO2 were recorded during the first 3 days after admission. Simultaneous ICP values were gained from the invasive monitoring. Associations between ONSD, Pcv-aCO2/Ca-cvO2 and simultaneous ICP were explored by Spearman correlation analysis. We constructed an ROC curve to identify the ONSD and Pcv-aCO2/Ca-cvO2 cutoff for the evaluation of elevated ICP. Results: We included 54 patients aged mean 57.13 (standard deviation 4.02) years and 24 (44%) were male. A significant correlation was observed between ONSD and ICP (r = 0.74, P < 0.01). The AUC was 0.861 (95% CI: 0.727-0.951), with a best cutoff value of 5.62 mm. Using a cutoff of 5.62mm, ONSD had a sensitivity of 92.8%, specificity of 80.4%. The Pcv-aCO2/Ca-cvO2 ratio also significantly correlated with ICP (r = 0.70, P < 0.01). The AUC was 0.791 (95% CI: 0.673-0.889). The optimal Pcv-aCO2/Ca-cvO2 value for predicting elevated ICP was 1.98 mmHg/mL. Using a cutoff of 1.98 mmHg/mL, Pcv-aCO2/Ca-cvO2 had a sensitivity of 87.3%, specificity of 77.2%. The AUC for ONSD combined with Pcv-aCO2/Ca-cvO2 was 0.952 (95% CI: 0.869-0.971), which had a sensitivity of 95.1%, specificity of 93.9%. Conclusion: Pcv-aCO2/Ca-cvO2 combined with ONSD performed best in predicting elevated intracranial pressure of patients with TBI in a prehospital setting. Our findings provide a crucial tool to improve earlier management of these patients in prehospital care, where the availability and utilization of invasive monitoring is limited. It could lead to significant changes in how TBI patients are monitored and treated before reaching a hospital.

Every year, millions of adult sockeye salmon (Oncorhynchus nerka) perform an arduous, once-in-a-lifetime migration up the Fraser River (BC, Canada) to return to their natal stream to spawn. The changes in heart rate, stroke volume, and arterio-venous oxygen extraction (i.e., factors determining rates of oxygen delivery to the tissues by the cardiovascular system) have never been directly and simultaneously measured along with whole animal oxygen uptake in a maximally swimming fish. Here, such measurements were made using three sockeye salmon populations (Early Stuart, Chilko and Quesnel), which each performed two consecutive critical swimming speed (Ucrit) challenges to provide a comprehensive quantification of cardiovascular physiology, oxygen status and blood chemistry associated with swimming and recovery. Swim performance, oxygen uptake, cardiac output, heart rate and stroke volume did not significantly vary at rest, during swimming or during recovery between populations or sexes. Despite incomplete metabolic recovery between swim challenges, all fish repeated their swim performance and similar quantitative changes in the cardiorespiratory variables were observed for each swim challenge. The high maximum cardiorespiratory performance and excellent repeat swim performance are clearly beneficial in allowing the salmon to maintain steady ground speeds and reach the distant spawning grounds in a timely manner.

Background: To determine whether Pcv-aCO2/Ca-cvO2 combined with Pcv-aCO2 could predict the outcomes in patients complicated with abdominal infection and sepsis after abdominal tumor operation. Methods: Total 92 patients admitted to our hospital from January 2017 to December 2020 who underwent abdominal tumor operation were enrolled. Blood gas analysis of artery and central vein, various laboratory indexes, SOFA score, hemodynamic parameters at different time points and treatment outcome were recorded. Results: ROC curve analysis showed that hemodynamic parameter alone could not predict ICU treatment outcome and mortality of patients, but 72-hour SOFA score could predict treatment outcome of patients (AUC = 0.930, 95% CI: 0.803-1.000, p = 0.019). The significant hemodynamic parameter for evaluating treatment outcome and prognosis of patients was Pcv-aCO2 + Ratio of T3. Kaplan-Meier univariate survival curve and Log-rank suggested that patients who had higher combined predictive parameter of T3 Ratio + T3 Pcv-aCO2 still had ischemia and hypoxia of tissues and organs after standard fluid resuscitation, and treatment outcome was not good. In subgroup analysis, patients with higher Ratio had higher lactate, higher T72 SOFA score, and poor treatment outcome. Conclusion: The combination of Ratio and Pcv-aCO2 could evaluate clinical treatment outcome of patients complicated with abdominal infection and sepsis after abdominal tumor operation.

BACKGROUND: Multiple complete central venous occlusion (CVO) is rare complication among the hemodialysis population. Percutaneous transluminal angioplasty (PTA) is the recommended treatment for CVO; however, cases with long-segment occlusion remain challenging. CASE DESCRIPTION: We reported a patient who complained of a swollen right arm for 1 month. On admission, his vital signs were within normal limits. The 76-year-old man had been on hemodialysis with a right forearm arteriovenous fistula (AVF) for 4 years with a history of temporizing catheterization and left forearm AVF failure. One year ago, he gradually developed a slight swelling in his right arm and the swelling in his arm was significantly worse one month ago. Digital subtraction angiography (DSA) revealed occlusion in his right innominate vein (IV), proximal subclavian vein (SV), and external and internal jugular veins, as well as stenosis of the ipsilateral cephalic arch and axillary vein (AV). The operation was performed with a pioneered bidirectional approach via ipsilateral superior vena cava (SVC) and AV puncture. The occluded lesions were successfully recanalized, and the patient's symptoms resolved after the operation. The patency of his vascular access was well maintained at the 4-month follow-up. CONCLUSIONS: To the best of our knowledge, this is the first report regarding the application of SVC puncture in PTA for CVO. This technique could be a possible approach when performed by appropriately qualified operators in patients with limited or no other options.

We describe an 81-year-old man with end-stage renal disease and central venous occlusion who was referred for dialysis access creation. This case illustrates a novel percutaneous image fusion-guided recanalization of an occluded right subclavian vein and brachiocephalic vein stent in a patient with limited remaining dialysis access sites. Co3V2O8 (CVO) with high theoretical specific capacity derived from the multiple oxidation states of V and Co is regarded as a potential electrode material for lithium-ion batteries (LIBs). Herein, reduced graphene oxide (rGO)-supported ultrafine CVO (rGO@CVO) nanoparticles are successfully prepared via the hydrothermal and subsequent annealing processes. The CVO supported on 2D rGO nanosheets possess excellent structural compatibility for the accommodation of volume variation to maintain the structural integrity of an electrode during the repeated lithiation/delithiation process. On the other hand, the rGO, as a highly-conductive network in the rGO@CVO composite, facilitates rapid charge transfer to ensure fast reaction kinetics. Moreover, the CV kinetic analysis indicates that the capacity of rGO@CVO is mainly dominated by a pseudocapacitive process with favorable rate capability. As a result, the rGO@CVO composite exhibits improved specific capacity (1132 mAh g-1, 0.1 A g-1) and promising rate capability (482 mAh g-1, 10 A g-1).

Introduction: la crise vaso-occlusive (CVO) est la plus fréquente manifestation de la drépanocytose et la première cause d´hospitalisation des enfants atteints. L´objectif de cette étude est de décrire les aspects cliniques des CVO sévères, de déterminer les étiologies des syndromes infectieux qui les accompagnent et de décrire leur prise en charge. Méthodes: il s'agit d'une étude transversale descriptive portant sur 137 drépanocytaires majeurs hospitalisés pour CVO sévères du 1er janvier 2009 au 31 décembre 2011 dans le service de pédiatrie du CHU Sylvanus Olympio. Résultats: les drépanocytaires homozygotes SS étaient les plus nombreux (n=98; 71,5%), suivis des doubles hétérozygotes SC (n=28; 20,5). Le délai moyen de consultation était de 4,7 ± 4,4 jours. Le traitement avant l´admission comportait des antibiotiques (28,5%). Les CVO étaient surtout ostéo-articulaires (70,8%). Dans 98,5% des cas, une infection bactérienne associée a été confirmée (48,9%) ou présumée (49,6%). Les principales étiologies étaient le syndrome thoracique aigu (26,3%), l´ostéomyélite aiguë (10,9%), l´infection urinaire (6,6%), la septicémie (3,6%). Un germe a été isolé chez 14,6% des patients, Escherichia coli (30%) étaient en tête suivi de Klebsiella pneumoniae (25%), Staphylococcus aureus (15%), Salmonella typhi (10%), Streptococcus pneumoniae (5%), le Streptocoque D (5%), l´Enterobacter (5%) et l´Acinetobacter (5%). Le taux de mortalité était de 2,2%. La durée moyenne d´hospitalisation était de 11,4 ± 8,8 jours. Conclusion: les CVO drépanocytaires sévères sont en majorité associées aux infections bactériennes en milieu tropical. Une antibiothérapie adaptée et précoce constitue le moyen thérapeutique indispensable pour prévenir ou traiter ces patients.
Introduction: vaso-occlusive crisis (VOC) is the most common manifestation of sickle cell disease and the leading cause of hospitalization among affected children. The purpose of this study is to describe the clinical features of severe VOCs, to determine the etiologies of infectious syndromes that accompany them and to describe their management. Methods: we conducted a descriptive cross-sectional study of 137 adult patients with sickle cell disease hospitalised for severe VOC in the Paediatric Department of the Sylvanus Olympio University Hospital from 1 January 2009 to 31 December 2011. Results: the majority of patients (n=98; 71.5%) had homozygous sickle cell (SS), followed by double heterozygous SC disease (n=28; 20.5). The median of consultation time was 4.7 ± 4.4 days. Treatment before admission was based on antibiotics (28.5%). VOCs were mainly osteoarticular (70.8%). In 98.5% of cases, an associated bacterial infection was confirmed (48.9%) or suspected (49.6%). The main etiologies included acute chest syndrome (26.3%), acute osteomyelitis (10.9%), urinary tract infection (6.6%) and septicaemia (3.6%). One germ was isolated from 14.6% of patients: Escherichia coli (30%), followed by Klebsiella pneumoniae (25%), Staphylococcus aureus (15%), Salmonella typhi (10%), Streptococcus pneumoniae (5%), Streptococcus D (5%), Enterobacter (5%) and Acinetobacter (5%). Mortality rate was 2.2%. The average length of stay in hospital was 11.4 ± 8.8 days. Conclusion: severe sickle cell related vaso-occlusive crisis is mainly associated with bacterial infections in tropical environments. Appropriate and early antibiotic therapy is the essential therapeutic means to prevent or treat these patients.

Designing composite structures of active materials is critical for high-performance lithium-ion batteries, as it determines the reversibility of lithium-ion insertion and extraction of the electrodes. The V2O3 anode has a high specific capacity but presents poor cycling stability due to a large volume change. Herein, a novel C@V2O3-Li4Ti5O12 composite with ultrastable cycling stability is constructed. In this composite structure, the interconnected ultrasmall V2O3 and Li4Ti5O12 nanoparticles (5-10 nm) construct robust interfaces in the carbon matrix. The Li4Ti5O12 nanoparticles with excellent cycling stability and a minor volume change act as fixtures that effectively restrict the volume change of V2O3 nanoparticles and improve the cycling stability of the C@V2O3-Li4Ti5O12 composite. The C@V2O3-Li4Ti5O12 composite maintains no degradation during 500 cycles under a current density of 100 mA g-1. The results demonstrate that constructing a highly stable interface between the active nanoparticles with smaller and larger volume changes is of great significance to suppress their pulverization and achieve high reversibility. This work contributes to a new strategy to design the structure of long-cycling anode materials for highly stable lithium-ion batteries.

Dengue is the most rapidly spreading mosquito-borne viral disease in the world. The mosquito, Aedes aegypti, also spreads Yellow fever, Chikungunya, and Zika virus. As the primary vector for dengue, Ae. aegypti now occurs in over 20 countries and is a serious concern with reports of increasing insecticide resistance. Developing new treatments to manage mosquitoes are needed. Formulation of crude volatile oil from Piper betle leaves (Pb-CVO) was evaluated as a potential treatment which showed larvicidal, ovipositional, and repellency effects. Gut-histology and enzyme profiles were analyzed post treatment under in-vitro conditions. The Pb-CVO from leaves of field collected plants was obtained by steam distillation and separated through rotary evaporation. The Pb-CVO were evaluated for chemical constituents through GC-MS analyses revealed 20 vital compounds. The peak area was establish to be superior in Eudesm-7(11)-en-4-ol (14.95%). Pb-CVO were determined and tested as four different concentrations (0.25, 0.5, 1.0, and 1.5 mg/L) of Pb-CVO towards Ae. aegypti. The larvicidal effects exhibited dose dependent mortality being greatest at 1.5 mg Pb-CVO/10 g leaves. The LC50 occurred at 0.63 mg Pb-CVO/L. Larva of Ae. aegypti exposed to Pb-CVO showed significantly reduced digestive enzyme actions of α- and ß-carboxylesterases. In contrast, GST and CYP450 enzyme levels increased significantly as concentration increased. Correspondingly, oviposition deterrence index and egg hatch of Ae. aegypti exposed to sub-lethal doses of Pb-CVO demonstrated a strong effect suitable for population suppression. Repellency at 0.6 mg Pb-CVO applied as oil had a protection time of 15-210 min. Mid-gut histological of Ae. aegypti larvae showed severe damage when treated with 0.6 mg of Pb-CVO treatment compared to the control. Non-toxic effects against aquatic beneficial insects, such as Anisops bouvieri and Toxorhynchites splendens, were observed at the highest concentrations, exposed for 3 h. These results suggest that the Pb-CVO may contain effective constituents suitable for development of new vector control agents against Ae. aegypti.

OBJECTIVE: Adult neurogenesis in the subgranular zone and subventricular zone is generally accepted, but its existence in other brain areas is still controversial. Circumventricular organs, such as the area postrema (AP) have recently been described as potential neurogenic niches in the adult brain. The AP is the major site of action of the satiating hormone amylin. Amylin has been shown to promote the formation of neuronal projections originating from the AP in neonatal rodents but the role of amylin in adult neurogenesis remains unknown. METHODS: To test this, we first performed an RNA-sequencing of the AP of adult rats acutely injected with either amylin (20 µg/kg), amylin plus the amylin receptor antagonist AC187 (500 µg/kg) or vehicle. Second, animals were subcutaneously equipped with minipumps releasing either amylin (50 µg/kg/day) or vehicle for 3 weeks to assess cell proliferation and differentiation with the 5'-bromo-2-deoxyuridine (BrdU) technique. RESULTS: Acute amylin injections affected genes involved in pathways and processes that control adult neurogenesis. Amylin consistently upregulated NeuroD1 transcript and protein in the adult AP, and this effect was blocked by the co-administration of AC187. Further, chronic amylin treatment increased the number of newly proliferated AP-cells and significantly promoted their differentiation into neurons rather than astrocytes. CONCLUSION: Our findings revealed a novel role of the satiating hormone amylin in promoting neurogenesis in the AP of adult rats.

Diverse transmitter systems (e.g. acetylcholine, dopamine, endocannabinoids, endorphins, glutamate, histamine, 5-hydroxytryptamine, substance P) have been implicated in the pathways by which nausea and vomiting are induced and are targets for anti-emetic drugs (e.g. 5-hydroxytryptamine3 and tachykinin NK1 antagonists). The involvement of TRPV1 in emesis was discovered in the early 1990s and may have been overlooked previously as TRPV1 pharmacology was studied in rodents (mice, rats) lacking an emetic reflex. Acute subcutaneous administration of resiniferatoxin in the ferret, dog and Suncus murinus revealed that it had "broad-spectrum" anti-emetic effects against stimuli acting via both central (vestibular system, area postrema) and peripheral (abdominal vagal afferents) inputs. One of several hypotheses discussed here is that the anti-emetic effect is due to acute depletion of substance P (or another peptide) at a critical site (e.g. nucleus tractus solitarius) in the central emetic pathway. Studies in Suncus murinus revealed a potential for a long lasting (one month) effect against the chemotherapeutic agent cisplatin. Subsequent studies using telemetry in the conscious ferret compared the anti-emetic, hypothermic and hypertensive effects of resiniferatoxin (pungent) and olvanil (non-pungent) and showed that the anti-emetic effect was present (but reduced) with olvanil which although inducing hypothermia it did not have the marked hypertensive effects of resiniferatoxin. The review concludes by discussing general insights into emetic pathways and their pharmacology revealed by these relatively overlooked studies with TRPV1 activators (pungent an non-pungent; high and low lipophilicity) and antagonists and the potential clinical utility of agents targeted at the TRPV1 system.

This study examined which brain regions are influenced by an inhibitory lateral parabrachial nucleus (LPBN) mechanism that affects water intake. Controls and rats with bilateral LPBN lesions were administered angiotensin II (AngII) (0.5mg/kg subcutaneous - SC), drinking responses measured, and brains processed for Fos-immunohistochemistry. A separate group of LPBN-lesioned and non-lesioned animals were denied water for 90 min prior to perfusion to remove any confounding factor of water intake. LPBN-lesioned rats drank a cumulative volume of 9 mL compared with <4 mL by controls (p<0.01). Compared with sham-lesioned animals, Fos expression was attenuated in overdrinking LPBN-lesioned rats in the median preoptic nucleus (MnPO), paraventricular nucleus of the hypothalamus (PVN), supraoptic nucleus (SON) (p<0.001), bed nucleus of the stria terminalis and central nucleus of the amygdala (p<0.01). In LPBN-lesioned rats that did not drink, greater numbers of activated neurons were detected in the PVN (p<0.001), SON (p<0.01), MnPO, nucleus of the solitary tract (NTS) and area postrema (p<0.05) in response to SC AngII, compared with non-lesioned rats. These data suggest that the direct effects of LPBN lesions caused an increase in AngII-induced water intake and in rats that did not drink an increase in Fos expression, while indirect secondary effects of LPBN lesions caused a reduction in Fos expression possibly related to excessive ingestion of water. An inhibitory mechanism, likely related to arterial baroreceptor stimulation, relayed by neurons located in the LPBN influences the responses of the MnPO, PVN and SON to increases in peripheral AngII.

Using a double immunofluorescence procedure, we report the discovery of a novel group of fibrous astrocytes that co-express epithelial sodium channel (ENaC) γ-subunit protein along with glial acidic fibrillary protein (GFAP). These cells are concentrated along the borders of the sensory circumventricular organs (CVOs), embedded in the white matter (e.g., optic nerve/chiasm, anterior commissure, corpus callosum, pyramidal tract) and are components of the pia mater. In the CVOs, a compact collection of ENaC γ-immunoreactive glial fibers form the lamina terminalis immediately rostral to the organum vasculosum of the lamina terminalis (OVLT). Astrocyte processes can be traced into the median preoptic nucleus - a region implicated in regulation of sodium homeostasis. In the subfornical organ (SFO), ENaC γ-GFAP astrocytes lie in its lateral border, but not in the ventromedial core. In the area postrema (AP), a dense ENaC γ-GFAP glial fibers form the interface between the AP and nucleus tractus solitarius; this area is termed the subpostremal region. Antibodies against the ENaC α- or ß-subunit proteins do not immunostain these regions. In contrast, the antibodies against the ENaC γ-subunit protein react weakly with neuronal cell bodies in the CVOs. Besides affecting glial-neural functions in the CVOs, the astrocytes found in the white matter may affect saltatory nerve conduction, serving as a sodium buffer. The ENaC γ-expressing astrocytes of the ventral medulla send processes into the raphe pallidus which intermingle with the serotoninergic (5-HT) neurons found in this region as well as with the other nearby 5-HT neurons distributed along ventral medullary surface.

Insulin resistance is a hallmark of obesity, the cardiorenal metabolic syndrome and type 2 diabetes mellitus (T2DM). The progression of insulin resistance increases the risk for cardiovascular disease (CVD). The significance of insulin resistance is underscored by the alarming rise in the prevalence of obesity and its associated comorbidities in the Unites States and worldwide over the last 40-50 years. The incidence of obesity is also on the rise in adolescents. Furthermore, premenopausal women have lower CVD risk compared to men, but this protection is lost in the setting of obesity and insulin resistance. Although systemic and cardiovascular insulin resistance is associated with impaired insulin metabolic signaling and cardiovascular dysfunction, the mechanisms underlying insulin resistance and cardiovascular dysfunction remain poorly understood. Recent studies show that insulin resistance in obesity and diabetes is linked to a metabolic inflammatory response, a state of systemic and tissue specific chronic low grade inflammation. Evidence is also emerging that there is polarization of macrophages and lymphocytes towards a pro-inflammatory phenotype that contributes to progression of insulin resistance in obesity, cardiorenal metabolic syndrome and diabetes. In this review, we provide new insights into factors, such as, the renin-angiotensin-aldosterone system, sympathetic activation and incretin modulators (e.g., DPP-4) and immune responses that mediate this inflammatory state in obesity and other conditions characterized by insulin resistance.
