Analysis of glucocorticoid profiles serves as a valuable, multi-faceted tool for insight into the behavior and physiology of wild populations. Recently, the measurement of fecal glucocorticoid metabolites (FCMs) has exploded in popularity due to its compatibility with noninvasive techniques and remote environments A critical first step is to perform a biological validation to ensure that the assay accurately reflect changes in FCM levels. We use an enzyme immunoassay (EIA) to perform a biological validation on samples collected from two males and six females in a wild population of Colobus vellerosus in response to three naturally occurring potential stressors. We also describe the FCM response pattern in the week following parturition in three females and examine the influence of sex, reproductive state, and time of day on the concentrations of baseline samples collected daily from 13 adult individuals over a period of four months. We validated the assay: FCM levels increase in response to natural stressors with a two-day lag. In the two days surrounding parturition, FMC levels increased. Baseline concentrations were affected by collection time and female reproductive state, with lactating females having lower concentrations than pregnant or cycling females. Thus, we successfully carried out the first validation and characterization of FCMs in a wild African colobine. This will serve as an essential foundation for future studies of C. vellerosus and similar wild primates whose objective is to investigate the role glucocorticoids play in responses to social and ecological challenges.

OBJECTIVES: The function of the browridge in primates is a subject of enduring debate. Early studies argued for a role in resisting masticatory stresses, but recent studies have suggested sexual signaling as a biological role. We tested associations between circumorbital form, diet, oral processing, and social behavior in two species of colobus monkey-the king colobus (Colobus polykomos) and western red or bay colobus (Piliocolobus badius). MATERIALS AND METHODS: We quantified circumorbital size and dimorphism in a sample of 98 crania. Controlling for age and facial size, we tested whether variation in circumorbital morphology can be explained by variation in diet, oral processing behavior, masticatory muscle size, and mating system. To contextualize our results, we included a broader sample of facial dimorphism for 67 anthropoid species. RESULTS: Greater circumorbital thickness is unrelated to the stresses of food processing. King colobus engages in longer bouts of anterior tooth use, chews more per ingestive event, and processes a tougher diet, yet circumorbital thickness of C. polykomos is reduced compared to P. badius. Differences in circumorbital development do not vary with wear or facial size. Greater sexual dimorphism is present in P. badius; comparisons across anthropoids indicated patterns of circumorbital dimorphism were decoupled from overall size dimorphism. CONCLUSIONS: The expanded circumorbits of male red colobus monkeys evolved in response to intense male-male competition. This hypothesis is consistent with the pattern across anthropoid primates and highlights the underappreciated role of sexual selection in shaping the primate face.

Here, we report on a new collection of mostly isolated molars of a colobine monkey from near Hasnot on the Potwar Plateau of Pakistan. The specimens are from three late Miocene localities, with ages constrained to between 7.9 and 7.1 Ma. Morphological and metrical comparisons of the new Hasnot molars with those of previously recognized Mesopithecus species and living Asian colobines lead to the conclusion that the Hasnot colobine is most probably Mesopithecus, but not Mesopithecus pentelicus. The most morphologically distinctive aspect of the Hasnot specimens is the lower third molars, which exhibit large and bulbous protoconids set off by deeply incised mesial buccal and median buccal clefts and large, broad distobuccally placed hypoconulids. Colobine specimens previously recovered from the Potwar Plateau have been assigned to Mesopithecus sivalensis, but because these specimens have not yet been fully described, a detailed comparison with the new Hasnot specimens is not yet possible. For these reasons, we assign the new Hasnot colobine fossils to cf. Mesopithecus sp. Mesopithecus was one of the most widespread and successful of late Miocene primates. As a colobine equipped with features of the molar teeth, limbs, and, presumably, gut enabling it to succeed in more highly seasonal woodland environments, Mesopithecus was able to rapidly disperse into and adapt to the conditions in South Asia brought about by profound climatic and concomitant environmental change during the latest Miocene.

RESUMO

We identified a novel Kaposi's sarcoma herpesvirus-related rhadinovirus (Colobine gammaherpesvirus 1) in a mantled guereza (Colobus guereza kikuyensis). The animal had multiple oral tumors characterized by proliferation of latent nuclear antigen 1-positive spindle cells and was not co-infected with immunosuppressive simian viruses, suggesting that it had Kaposi sarcoma caused by this novel rhadinovirus.

RESUMO

We determined the complete genome sequence of a virus isolated from a mantled guereza that died of primary effusion lymphoma. The virus is closely related to Kaposi's sarcoma-associated herpesvirus (KSHV) but lacks some genes implicated in KSHV pathogenesis. This finding may help determine how KSHV causes primary effusion lymphoma in humans.

The mammalian order primates contains wide species diversity. Members of the subfamily Colobinae are unique amongst extant primates in that their gastrointestinal systems more closely resemble those of ruminants than other members of the primate order. In the growing literature surrounding nonhuman primate microbiomes, analysis of microbial communities has been limited to the hindgut, since few studies have captured data on other gut sites, including the foregut of colobine primates. In this study, we used the red-shanked douc (Pygathrix nemaeus) as a model for colobine primates to study the relationship between gastrointestinal bacterial community structure and gut site within and between subjects. We analyzed fecal and pregastric stomach content samples, representative of the hindgut and foregut respectively, using 16S recombinant DNA (rDNA) sequencing and identified microbiota using closed-reference operational taxonomic unit (OTU) picking against the GreenGenes database. Our results show divergent bacterial communities clearly distinguish the foregut and hindgut microbiomes. We found higher bacterial biodiversity and a higher Firmicutes:Bacteroides ratio in the hindgut as opposed to the foregut. These gut sites showed strong associations with bacterial function. Specifically, energy metabolism was upregulated in the hindgut, whereas detoxification was increased in the foregut. Our results suggest a red-shanked douc's foregut microbiome is no more concordant with its own hindgut than it is with any other red-shanked douc's hindgut microbiome, thus reinforcing the notion that the bacterial communities of the foregut and hindgut are distinctly unique. Detailed analyses and comparisons of postcranial specimens of Plio-Pleistocene cercopithecids provide an opportunity to examine the recent evolutionary history and locomotor diversity in Old World monkeys. Studies examining the positional behavior and substrate preferences of fossil cercopithecids are also important for reconstructing the paleoenvironments of Plio-Pleistocene hominin sites. Here we describe a new fossil cercopithecid tibia (EP 1100/12) from the Australopithecus afarensis-bearing Upper Laetolil Beds (∼3.7 Ma) of Laetoli in northern Tanzania. The fossil tibia is attributed to cf. Rhinocolobus sp., which is the most common colobine at Laetoli. In addition to qualitative comparisons, the tibial shape of EP 1100/12 was compared to that of 190 extant cercopithecids using three-dimensional landmarks. Discriminant function analyses of the shape data were used to assess taxonomic affinity and shape variation relating to positional behavior. EP 1100/12 clustered with extant colobines, particularly the large-bodied genera Nasalis and Rhinopithecus. Comparisons reveal that EP 1100/12 belongs to a large-bodied monkey that engaged in arboreal pronograde quadrupedalism. These findings add further support to previous inferences that woodland and forest environments dominated the paleoenvironment of the Upper Laetolil Beds, which supported the diverse community of cercopithecids at Laetoli. The inferred paleoecology and the presence of large-bodied arboreally-adapted monkeys at Laetoli show that A. afarensis had access to a range of diverse habitats, including woodlands and forests. This supports the possibility that A. afarensis, with its potential range of positional capabilities, was able to utilize arboreal settings for food acquisition and refuge from predators.

Both diet and host phylogeny shape the gut microbial community, and separating out the effects of these variables can be challenging. In this study, high-throughput sequencing was used to evaluate the impact of diet and phylogeny on the gut microbiota of nine colobine monkey species (N = 64 individuals). Colobines are leaf-eating monkeys that fare poorly in captivity-often exhibiting gastrointestinal (GI) problems. This study included eight Asian colobines (Rhinopithecus brelichi, Rhinopithecus roxellana, Rhinopithecus bieti, Pygathrix nemaeus, Nasalis larvatus, Trachypithecus francoisi, Trachypithecus auratus, and Trachypithecus vetulus) and one African colobine (Colobus guereza). Monkeys were housed at five different captive institutes: Panxi Wildlife Rescue Center (Guizhou, China), Beijing Zoo, Beijing Zoo Breeding Center, Singapore Zoo, and Singapore Zoo Primate Conservation Breeding Center. Captive diets varied widely between institutions, but within an institution, all colobine monkey species were fed nearly identical or identical diets. In addition, four monkey species were present at multiple captive institutes. This allowed us to parse the effects of diet and phylogeny in these captive colobines. Gut microbial communities clustered weakly by host species and strongly by diet, and overall, colobine phylogenetic relationships were not reflected in gut microbiota analyses. Core microbiota analyses also identified several key taxa-including microbes within the Ruminococcaceae and Lachnospiraceae families-that were shared by over 90% of the monkeys in this study. Microbial species within these families include many butyrate producers that are important for GI health. These results highlight the importance of diet in captive colobines.

The social organization of natural groups of Rhinopithecus bieti (Yunnan snub-nosed monkey) is virtually unknown. We studied the demography and social structure of a free-ranging group at Samage Forest, China, for nearly 2 years. This study confirmed that R. bieti exhibits a multilevel social organization of core 1-male units (OMUs) that congregate in a band of >400 members. Even though the band appeared to be unified for the most part, we also witnessed occasional fission-fusion. OMUs were cohesive entities, and their members were spatially and socially isolated from members of other OMUs. Large all-male units associated with the band, and when they closely followed OMUs there was a tendency for elevated male aggression. Within OMUs, females associated preferentially with males and vice versa, resulting in a bisexually bonded society. Contrary to other Asian colobines, R. bieti were comparatively social, with grooming occupying 7.3% of the time. Social grooming was primarily a female affair, but males also participated in grooming networks. The integration of males into the social network of the OMU is thought to help to maintain OMU integrity and cohesion with other social units being in close proximity.

Body mass plays an important role in primate positional behavior and in sexually dimorphic arboreal primate species may influence how immature and adult individuals travel through the forest canopy and access food resources. In this study, we examined age- and sex-based patterns of positional behavior and substrate utilization in wild golden snub-nosed monkeys (Rhinopithecus roxellana), an endangered species of Asian colobine. Our results indicated that among all age and sex classes, sitting was the most common feeding and resting posture and during travel, quadrupedal walking was the dominant locomotor behavior. Despite the fact that adult male R. roxellana are reported to exhibit a body mass nearly two times that of adult females, we found no significant sex differences in the positional repertoire during feeding and traveling. In addition, we found that while infants and juveniles used similar postural and locomotor behaviors as their adult counterparts, younger golden snub-nosed monkeys more frequently engaged in risky or escape-oriented behaviors such as climbing, running, leaping, and forelimb suspension. With increasing age, the use of quadrupedal walking and dropping (downward in-air displacement of body mass that does not require hindlimb propulsion) increased and the use of leaping, suspensory postures, and bridging decreased. Finally, given differences in the positional repertoire of adult and immature golden snub-nosed monkeys, we argue that studies of ontogenetic patterns of positional behavior should emphasize what it takes to survive at each life stage rather than what it takes to match an adult repertoire.

Primate females often inspect, touch and groom others' infants (natal attraction) and they may hold and carry these infants in a manner resembling maternal care (infant handling). While natal attraction and infant handling occur in most wild colobines, little is known about the factors influencing the expression of these behaviors. We examined the effects of female parity, kinship, and dominance rank, as well as infant age and sex in wild Colobus vellerosus at Boabeng-Fiema Monkey Sanctuary, Ghana. We collected data via focal sampling of females in 2008 and 2009 (N = 61) and of infants in 2010 (N = 12). Accounting for the individuals who interacted with our focal subjects, this study includes 74 females and 66 infants in 8 groups. We recorded female agonistic interactions ad libitum to determine dominance ranks. We used partial pedigree information and genotypes at 17 short tandem repeat loci to determine kinship. We knew female parity, infant age and sex from demographic records. Nulliparous females showed more natal attraction and infant handling than parous females, which may suggest that interactions with infants are more adaptive for nulliparous females because they learn mothering skills through these behaviors. Compared to non-kin, maternal kin were more likely to handle infants. Maternal kin may be permitted greater access to infants because mothers are most familiar with them. Handlers may incur inclusive fitness benefits from infant handling. Dominance rank did not affect female interactions with infants. The youngest infants received the most natal attraction and infant handling, and male infants were handled more than female infants. The potential benefits of learning to mother and inclusive fitness, in combination with the relatively low costs of natal attraction and infant handling, may explain the high rates of these behaviors in many colobines.

Exceptional species and phenotypic diversity commonly are attributed to ecological opportunity (EO). The conventional EO model predicts that rates of lineage diversification and phenotypic evolution are elevated early in a radiation only to decline later in response to niche availability. Foregut fermentation is hypothesized to be a key innovation that allowed colobine monkeys (subfamily Colobinae), the only primates with this trait, to successfully colonize folivore adaptive zones unavailable to other herbivorous species. Therefore, diversification rates also are expected to be strongly linked with the evolution of traits related to folivory in these monkeys. Using dated molecular phylogenies and a dataset of feeding morphology, I test predictions of the EO model to evaluate the role of EO conferred by foregut fermentation in shaping the African and Asian colobine radiations. Findings from diversification methods coupled with colobine biogeographic history provide compelling evidence that decreasing availability of new adaptive zones during colonization of Asia together with constraints presented by dietary specialization underlie temporal changes in diversification in the Asian but not African clade. Additionally, departures from the EO model likely reflect iterative diversification events in Asia.

The Bornean banded langur (Presbytischrysomelaschrysomelas) is critically endangered species primarily found in Sarawak, Malaysia. Albeit this species is in peril, the ecology knowledge of this endemic species of Borneo is still scarce. Thus, a rapid survey employing total count and scan sampling method was conducted between July to August 2023 at Tanjung Datu National Park (TDNP), Sarawak to observe the social interaction of species with the environment. The behaviour of langur was recorded by employing scanning sampling method at 10 minutes intervals. This study sought to provide preliminary data on behavioural ecology of the Bornean banded langur within the national park. During the survey, three groups (consisting two to seven individuals) and a solitary male Bornean banded langur were recorded. The langurs were observed in both dipterocarp forests and coastal forests within the park. The daily activities of the Bornean banded langurs in TDNP were predominantly resting (31%), moving (29%), feeding (26%), vocalizing (14%), but not engaging in other social activities such as grooming, playing and mating. Knowing the behavioural ecological status as well as understanding ecology by identifying the activity pattern of langur is essential to government authorities and pertinent stakeholders to implement conservation strategies for the Bornean banded langur and their habitats.

The Guizhou golden monkey (Rhinopithecus brelichi) is a critically endangered wildlife species, and understanding its diet composition may be useful for assessing its feeding strategies. DNA metabarcoding was used to determine the dietary diversity of R. brelichi. DNA was extracted from 31 faecal samples and amplified chloroplast rbcL and mitochondrial COI DNA was sequenced using the Illumina NovaSeq platform. A comparative analysis of the sequences revealed that the five most abundant plant genera were Magnolia, Morinda, Viburnum, Tetradium and Eurya. In winter, R. brelichi mostly consumed shrubs, herbs and shrubs/trees according to the habit of plant genera with higher abundances comparatively. The five most abundant families in animal diet were Psychodidae, Trichinellidae, Staphylinidae, Scarabaeidae and Trichoceridae. This study is the first to show the composition of the winter animal diets of R. brelichi based on DNA metabarcoding. These results provide an important basis for understanding the diet of wild R. brelichi, which inhabits only the Fanjingshan National Nature Reserve, China.

Folivorous primates are typically considered time minimizers because the constraints of their unique digestive systems require them to spend a large portion of their activity budgets resting. However, inter- and intraspecific behavioral variation in their activity budgets may be influenced by local geographic and climatic conditions and evolutionary history. We compiled 48 studies representing ten genera, 31 species, and 50 populations to assess geographic (elevation, latitude), climatic (precipitation, temperature), and phylogenetic correlates of colobine activity budgets. Time spent resting negatively correlated with time spent feeding, moving, and socializing. Except for time spent socializing, activity budgets were independent of phylogeny, with more time spent feeding and less time resting in higher-elevation habitats and at lower temperatures. Among the four most common genera in our sample, only in Rhinopithecus did time spent feeding increase with higher elevations (range, 950-3950 m above mean sea level) and lower temperatures (range, 0.9-25 ℃). Only in Trachypithecus did time spent resting decrease with lower temperatures (range, 19-25 ℃). Our findings suggest that there are no apparent effects of geographic or climatic gradients on colobine activity budgets except for Rhinopithecus and Trachypithecus, whose activities are biased in favor of energy maximization rather than time minimization. Compared with other colobines, the ability of Rhinopithecus to adapt their activity budget at high elevations may make them less vulnerable to climate change, while the greater sensitivity of Trachypithecus to heat may make them more vulnerable to climate change.

Given the current rate of habitat degradation and loss in the tropics, data on primate population densities and habitat use are indispensable for assessing conservation status and designing feasible management plans for primates. The Omo River guereza (Colobus guereza guereza) is a subspecies of the eastern black-and-white colobus monkey endemic to the western Rift Valley forests of Ethiopia. Their restricted distribution along with habitat loss and hunting within their range render them vulnerable to local extirpation and extinction. Furthermore, there are no published data available on the population status and habitat use patterns of the Omo River guereza. We therefore aimed to assess the population size of Omo River guerezas in different habitats (Erica-Juniperus mixed forest, mixed plantation forest, undisturbed natural forest, disturbed natural forest) using transect surveys at Wof-Washa Natural State Forest (WWNSF) in central Ethiopia. Our surveys covered a cumulative distance of 88.5 km in four different habitats, during which we recorded a total of 140 Omo River guereza groups. The average group density was 14.3 groups/km2, average individual density was 94.4 individuals/km2, and we estimated the total population size within WWNSF to be 2549 individuals. The sex ratio of the population was split evenly between males and females, though the age classes skewed strongly towards adults. Of the habitats surveyed, the highest group encounter rate (1.83 groups/km) occurred in the disturbed natural forest. However, the highest individual density (110.1 individuals/km2) was recorded in undisturbed natural forest. Still, sizable densities (group and individual) were recorded in three of the disturbed habitats (disturbed natural forest, mixed plantation forest, and to a lesser extent Erica-Juniperus mixed forest). Our study offers the first baseline information with which to compare future population density estimates and habitat use in the range of Omo River guerezas.

One male and three female captive proboscis monkeys (Nasalis larvatus) were used as test animals to compare fecal nutrients after being fed seasonal foliage. Foliage and fecal samples were collected during three seasons (spring, summer, and winter). We analyzed crude ash (CA), ether extract (EE), crude protein (CP), neutral detergent fiber (NDF), acid detergent fiber (ADF), and acid detergent lignin (ADL). In addition to the above components, for the fecal samples, we examined total fecal nitrogen (TFN), nitrogen in fecal NDF (NDF-N), metabolic fecal nitrogen (MFN), and particle size distribution (MPS). Seasonal differences in foliage components were observed, with NDF and ADF being lowest in spring (p < 0.05). Fecal NDF and ADF also tended to be lowest in spring. Further, the distribution of fecal particle size indicated that MPS tended to increase in winter. The mean fecal NDF content (47.4%) for all three seasons measured in this study was as much as 1.3 times lower than previously reported values in free-ranging specimens. These data provide useful information that can be used to find some suitable food items for the endangered highly folivorous monkeys in captivity.

Right-handedness in humans reflects the functional brain specialisation of the left hemisphere. To better understand the origins of this population-level tendency, it is crucial to understand manual lateralisation in other non-human primate species. The aim of this article is to present a first approach to the hand preference of two primates from Vietnam, the endangered hatinh langur (Trachypithecus hatinhensis) and the critically endangered grey-shanked douc langur (Pygathrix cinerea). Eighteen individuals from each species (N = 36) were evaluated by means of the bimanual coordinated tube task and their responses were recorded in terms of manual events and bouts. Our results showed that subjects presented strong individual-level preferences but not lateralisation at the group-level. No sex differences were detected within species. The index finger was used in all of the extractions during this bimanual task, alone (86 %) or in combination with other fingers (14 %). In addition, hatinh langurs exhibited a greater strength of hand preferences than grey-shanked douc langurs, pointing to a possible higher manual specialisation during the leaf-eating process. These findings help to broaden our scarce knowledge of manual laterality in Asian colobine monkeys and confirm the bimanual tube task as a sensitive measure for assessing manual laterality in non-human primates.

We studied endangered golden langurs in fragmented and altered habitats to understand the consequences of habitat conditions on group size, social organization, and birth seasonality. We selected 12 groups inhabiting forest edge and forest core of Chakrashila Wildlife Sanctuary (henceforth Chakrashila WLS) and adjoining the Abhaya rubber plantation. Each group was monitored every month from May 2013 to September 2016 and recorded the age-sex of individuals in the group. The births were recorded with the individual identity of females in five focal groups. The overall group size of golden langur was 11.3 ± 3.5SD, and ranged between 5 and 18. The mean group size in forest core, forest edge, and rubber plantation differed significantly. We recorded a total of 46 births in 12 groups across the three different habitats. The number of infants correlates positively with adult females and group size across all the 12 groups for all the years. The number of births that occurred in all the months varied significantly across the months. Births occurred in all the months but peaked between May and September (82.6%). The mean number of births positively correlated with mean monthly rainfall. Mean inter-birth interval was 24.5 ± 1.6SD months that did not vary between the females. It therefore appears that group size is sensitive to forest type, and births are positively related to social and environmental factors. The behavioral parameters may influence life-history traits if continuous habitat alteration persists.

Bitter taste perception enables the detection of potentially toxic molecules and thus evokes avoidance behavior in vertebrates. It is mediated by bitter taste receptors, TAS2Rs. One of the best-studied TAS2R is TAS2R38. Phenylthiocarbamide (PTC) perception and TAS2R38 receptors vary across primate species, and this variation may be related to variation in dietary preferences. In particular, we previously found that the low sensitivity of TAS2R38s in Asian colobines likely evolved as an adaptation to their leaf-eating behavior. However, it remains unclear whether this low PTC sensitivity is a general characteristic of the subfamily Colobinae, a primate group that feeds predominantly on leaves. We performed genetic analyses, functional assays with mutant proteins, and behavioral analyses to evaluate the general characteristics of TAS2R38 in colobines. We found that PTC sensitivity is lower in TAS2R38s of African colobines than in TAS2R38s of omnivorous macaques. Furthermore, two amino acids shared between Asian and African colobines were responsible for low sensitivity to PTC, suggesting that the last common ancestor of extant colobines had this phenotype. We also detected amino acid differences between TAS2R38s in Asian and African colobines, indicating that they evolved independently after the separation of these groups.
