BACKGROUND: The Epidemiologic Study of Xolair (omalizumab): Evaluating Clinical Effectiveness and Long-term Safety in Patients with Moderate-to-Severe Asthma (EXCELS) assessed the long-term safety of omalizumab in a clinical practice setting as part of a phase IV US Food and Drug Administration postmarketing commitment. OBJECTIVE: We sought to evaluate long-term safety in omalizumab-treated and nonomalizumab-treated patients. Primary outcome measures focused on assessment of malignancies. METHODS: EXCELS was a prospective observational cohort study in patients (≥12 years of age) with moderate-to-severe allergic asthma. There were 2 cohorts: omalizumab (taking omalizumab at baseline) and nonomalizumab (no history of omalizumab treatment). Primary outcomes included all confirmed, incident, study-emergent primary malignancies (malignancies), including and excluding nonmelanoma skin cancer (NMSC); all malignancies were externally adjudicated. RESULTS: The omalizumab cohort had a higher proportion of patients with severe asthma compared with the nonomalizumab cohort (50.0% vs 23.0%). Median follow-up was approximately 5 years for both cohorts. Crude malignancy rates were similar in the omalizumab and nonomalizumab cohorts, with a rate ratio of 0.84 (95% CI, 0.62-1.13) for all malignancies and 0.98 (95% CI, 0.71-1.36) for all malignancies excluding NMSC. Kaplan-Meier plots of time to first confirmed study-emergent primary malignancy were similar for the 2 treatment cohorts. Cox proportional hazards modeling, adjusting for confounders and risk factors, resulted in a hazard ratio (omalizumab vs nonomalizumab) of 1.09 (95% CI, 0.87-1.38) for all malignancies and 1.15 (95% CI, 0.83-1.59) for all malignancies excluding NMSC. CONCLUSION: Results from EXCELS suggest that omalizumab therapy is not associated with an increased risk of malignancy.

Bertholletia excelsa is native to the Amazon Rainforest and is popularly known as the Brazil nut. It has socioeconomic importance due its nuts being a great export product. There are few studies in the literature regarding the biotechnological potential of its bark, although it is used in folk medicine. The aim of this study was to determine the chemical constituents, anti-inflammatory and antioxidant properties of B. excelsa bark extract (BEB). Twelve substances were identified by LC/MS/MS, and cytotoxicity tests were carried out, as well as analyses of nitric oxide production and elimination of free radicals. BEB caused cytoprotection against oxidative stress in macrophages, increased HMOX-1 expression, overcame the antioxidant effects of GPx-1 and reduced its expression and was able to inhibit leukocyte migration in use peritonitis. BEB efficiently attenuated oxidative stress due to its antioxidant and anti-inflammatory properties and, as such, can be used as a safe and effective source of a natural herbal medicine.


Bertholletia excelsa es originaria de la selva amazónica y se la conoce popularmente como nuez de Brasil. Tiene importancia socioeconómica debido a que sus frutos secos son un gran producto de exportación. Existen pocos estudios en la literatura sobre el potencial biotecnológico de su corteza, aunque se utiliza en medicina popular. El objetivo de este estudio fue determinar los componentes químicos, las propiedades antiinflamatorias y antioxidantes del extracto de corteza de B. excelsa (BEB). Se identificaron 12 sustancias mediante LC/MS/MS y se realizaron pruebas de citotoxicidad, así como análisis de producción de óxido nítrico y eliminación de radicales libres. BEB causó citoprotección contra el estrés oxidativo en macrófagos, aumentó la expresión de HMOX-1, superó los efectos antioxidantes de GPx-1 y redujo su expresión y fue capaz de inhibir la migración de leucocitos en la peritonitis de uso. BEB atenuó eficazmente el estrés oxidativo debido a sus propiedades antioxidantes y antiinflamatorias y, como tal, puede utilizarse como una fuente segura y eficaz de un medicamento a base de hierbas naturales.

The official methods adopted by different worldwide agencies for determination of water content of Brazil nut is the dissication in drying oven at 105 ºC during 3 or 24 hours and dissication until constant height of samples. However, applying these methods for Brazil nut, may result in inconsistent values, possibly due to lipid oxidation. Thus, the objective of this study was to evaluate the accuracy of the oven-drying methods, recommended by the official agencies, and to determine the oven-drying correct parameters, such as temperature and exposure time. For this purpose, samples were placed in drying ovens set at 85, 90, 95 and 105 °C and weighed hourly, between 3 and 12 hours and after 24 hours of exposure, and the results were compared to Karl Fisher titration, considered as a reference method in this study. The temperature of 105 °C, for any exposure time, resulted in overestimated water content compared to reference method. However, there was no difference between the water content values obtained by oven-drying assay at 90 °C for 6 hours and by the reference method, allowing to conclude that the determination of water content in Brazil nut samples, in drying oven under these conditions, can be performed with the same accuracy and precision of the Karl Fischer method.


Os métodos oficiais adotados por diferentes órgãos ao redor do mundo para a determinação do teor de água da castanha-do-brasil são a dessecação em estufa a 105 °C por 3 ou 24 horas e a dessecação até peso constante. Contudo, a aplicação destes métodos para a castanha-do-brasil pode resultar em valores inconsistentes, possivelmente pela oxidação dos lipídeos. Assim, o objetivo deste estudo foi o de avaliar a acurácia dos métodos de estufa, recomendados pelas agências oficiais, bem como determinar os parâmetros adequados do método, como temperatura e tempo de exposição. Para isto, amostras foram colocadas em estufas ajustadas em 85, 90, 95 e 105 °C e pesadas a cada hora, entre 3 e 12 horas e ao final de 24 horas de exposição, e os resultados foram comparados com aqueles obtidos por titulação de Karl Fisher, considerado como método de referência neste estudo. A temperatura de 105 °C, para quaisquer tempos de exposição, resultou na superestimação do teor de água comparado ao método de referência. Não houve diferença entre os valores para o teor de água obtidos em estufa a 90 °C por 6 horas e o método de referência, permitindo concluir que a determinação do teor de água em amostras de castanha-do-brasil, em estufas nestas condições, pode ser executada com a mesma acurácia e precisão do método de Karl Fisher.

The essential oils of branchlets and fruits of Juniperus excelsa subsp. excelsa and Juniperus excelsa subsp. polycarpos were examined for their antioxidant activity. The compositions of the essential oils were studied by GC and GC-MS. To evaluation the antioxidants activity of the volatile oils, pure components and positive controls at different concentrations, thin-layer chromatography (TLC) screening methods, diphenylpicrylhydrazyl (DPPH) assay, deoxyribose degradation test and modified deoxyribose degradation test were employed. The results of the present study demonstrate some antioxidant activity for the tested essential oils obtained from various parts of both plants. It indicates that the use of these essential oils, in very low concentrations, may be useful as a natural preservative. However before any final conclusion, it is suggested that the antioxidant activity of these oils should also be evaluated by using lipid solvent system methods.

ABSTRACT Objective: To assess and compare the fatty acid composition of edible seeds and a nut native to the Cerrado (Brazilian savannah) to that of traditional oilseeds. Methods: Baru almonds, Cerrado cashew nuts, and pequi almonds were extracted from the fruits using appropriate equipment. All edible seeds and nuts were roasted, except for the Brazil nut. The sample lipids were extracted via cold pressing. The fatty acids were esterified, and the fatty acid esters were analyzed by gas chromatography. Results: The native and traditional edible seeds and nuts contain mostly monounsaturated fatty acids (42.72 g to 63.44 g/100 g), except for the Brazil nut, which showed predominance of polyunsaturated fatty acids (45.48 g/100 g). Pequi almond had the highest saturated fatty acid content (36.14 g/100 g). The fatty acids with the highest concentration were oleic and linoleic acids, and palmitic acid was also found in considerable concentration in the oilseeds studied. The Cerrado cashew nut and the traditional cashew nut have similar fatty acid profiles. As for the ratio of ω-6 to ω-3, the baru almond showed the highest ratio, 9:1, which was the closest to the recommended intake of these fatty acids. Conclusion: The fatty acid profile of the edible seeds and nuts native to the cerrado is similar to those of traditional oilseeds. We suggest the inclusion of native oilseeds in the diet aiming at reducing the risk of cardiovascular disease, especially the baru almond and the cerrado cashew nut, due to the fact they have high ratio of monounsaturated fatty acids to saturated fatty acids.


RESUMO Objetivo: Avaliar a composição de nozes e sementes comestíveis nativas do cerrado, no que diz respeito aos ácidos graxos, e comparar com oleaginosas tradicionais. Métodos: A amêndoa de baru, a castanha-de-caju-do-cerrado e a amêndoa de pequi foram extraídas dos frutos com equipamentos apropriados. Todas as nozes e sementes comestíveis foram torradas, exceto a castanha-do-brasil. Os lipídeos das amostras foram extraídos a frio, os ácidos graxos foram esterificados e os ésteres de ácidos graxos foram analisados por cromatografia gasosa. Resultados: As nozes e sementes comestíveis tradicionais e nativas possuem, predominantemente, ácidos graxos monoinsaturados (42,72 g a 63,44 g/100 g), exceto a castanha-do-brasil, que apresentou predominância de ácidos graxos poli-insaturados (45,48 g/100 g). A amêndoa de pequi apresentou o maior teor de ácidos graxos saturados (36,14 g/100 g). Os ácidos graxos encontrados em maior concentração nas oleaginosas foram o ácido oleico e o linoleico; o ácido palmítico foi também detectado em proporções consideráveis. O perfil de ácidos graxos da castanha-de-caju-do-cerrado é comparável ao da castanha-de-caju tradicional. Quanto à relação entre ácidos graxos w-6 e w-3, a amêndoa de baru apresentou o valor de 9:1, mais próximo ao recomendado para consumo. Conclusão: O perfil de ácidos graxos das nozes e sementes comestíveis nativas do cerrado se assemelha ao das tradicionais. Sugere-se, portanto, a inclusão das oleaginosas nativas em planos alimentares que visem a redução do risco de doenças cardiovasculares, sobretudo a amêndoa de baru e a castanha-de-caju-do-cerrado, por suas elevadas concentrações de ácidos graxos monoinsaturados em relação aos ácidos graxos saturados.

RESUMO

This is the first record in Brazil of an infestation of the Indian meal moth, Plodia interpunctella (Hubner 1813), for Brazil nut. P. interpunctella, is a cosmopolitan microlepidoptera of the family Pyralidae, considered an important insect pest of many different stored products. The record was for Brazil nut stored in bags in Itacoatiara county, Amazonas state.


Este é o primeiro registro no Brasil da infestação da traça-das-farinhas, Plodia interpunctella (Hubner 1813), em castanha-do-brasil. Plodia interpunctella é um microlepidóptero pertencente à família Pyralidae, cosmopolita e considerado importante inseto-praga dos mais variados produtos armazenados. O registro foi realizado em castanhas armazenadas em sacarias no município de Itacoatiara, estado do Amazonas.
