Olive leaves are a rich source of polyphenols with healthful properties and represent one of the most abundant waste products of olive oil production. The aims of this study were to explore the phenolic composition of olive leaves from the three main Tuscan cultivars (Leccino, Moraiolo and Frantoio) collected in Siena and Grosseto provinces and to investigate the possible use of these compounds as varietal and geographic origin markers. Discriminant factorial analysis (DFA) was used for distinguishing between different cultivars and locations. Apigenin and caffeoyl-secologanoside showed significant differences between cultivars. DFA showed that ligstroside, apigenin and luteolin have the most influence in determining the differences between sites, whereas total polyphenols, olacein and hydroxytyrosol acetate allowed for separation between leaves from the same province. The results of the present study indicate that concentrations of phenolic compounds, measured through high-resolution mass spectrometry, can be used as a marker for both the cultivar and of geographical origin of olive leaves, and possibly of olive-related products, as well as across small geographic scales (less than 50 km distance between sites).

BACKGROUND: We performed a secondary exploratory cluster analysis on the data collected from the validation phase of the study leading to the development of the model care pathway (CP) for Myasthenia Gravis (MG), in which a panel of 85 international experts were asked some characteristics about themselves and their opinion about the model CP. Our aim was to identify which characteristics of the experts play a role in the genesis of their opinion. METHODS: We extracted the questions probing an opinion and those describing a characteristic of the expert from the original questionnaire. We performed a multiple correspondence analysis (MCA) and a subsequent hierarchical clustering on principal component (HCPC) on the opinion variables, integrating the characteristic variables as supplementary (predicted). RESULTS: After reducing the dimensionality of the questionnaire to three dimensions we noticed that the not-appropriateness judgement of the clinical activities may overlap with the completeness one. From the HCPC it seems that the working setting of the expert may play a crucial role in determining the opinion about the setting of the sub-processes of MG: shifting from a cluster where the experts do not work in sub-specialist settings to one where the experts are working in them, the opinion changes accordingly from a mono-disciplinary setting to a multi-disciplinary one. Another interesting result is that the experience in neuromuscular diseases (NMD) measured in years and the expert typology (whether general neurologist or NMD expert) seem not to contribute significantly to the opinions. CONCLUSIONS: These findings might indicate a poor ability of the expert to discriminate what is not appropriate from what is not complete. Also, the opinion of the expert might be influenced by the working setting, but not by the experience in NMD (as measured in years).

BACKGROUND: Morse taper junction tribocorrosion is recognized as an important failure mode in total hip arthroplasty. Although taper junctions are used in almost all shoulder arthroplasty systems currently available in the United States, with large variation in design, limited literature has described comparable analyses of taper damage in these implants. In this study, taper junction damage in retrieved reverse total shoulder arthroplasty (RTSA) implants was assessed and analyzed. METHODS: Fifty-seven retrieved RTSAs with paired baseplate and glenosphere components with Morse taper junctions were identified via database query; 19 of these also included paired humeral stems and trays or spacers with taper junctions. Components were graded for standard damage modes and for fretting and corrosion with a modified Goldberg-Cusick classification system. Medical records and preoperative radiographs were reviewed. Comparative analyses were performed assessing the impact of various implant, radiographic, and patient factors on taper damage. RESULTS: Standard damage modes were commonly found at the evaluated trunnion junctions, with scratching and edge deformation damage on 76% and 46% of all components, respectively. Fretting and corrosion damage was also common, observed on 86% and 72% of baseplates, respectively, and 23% and 40% of glenospheres, respectively. Baseplates showed greater moderate to severe (grade ≥ 3) fretting (43%) and corrosion (27%) damage than matched glenospheres (fretting, 9%; corrosion, 13%). Humeral stems showed moderate to severe fretting and corrosion on 28% and 30% of implants, respectively; matched humeral trays or spacers showed both less fretting (14%) and less corrosion (17%). On subgroup analysis, large-tapered implants had significantly lower summed fretting and corrosion grades than small-tapered implants (P < .001 for both) on glenospheres; paired baseplate corrosion grades were also significantly lower (P = .031) on large-tapered implants. Factorial analysis showed that bolt reinforcement of the taper junction was also associated with less fretting and corrosion damage on both baseplates and glenospheres. Summed fretting and corrosion grades on glenospheres with trunnions (male) were significantly greater than on glenospheres with bores (female) (P < .001 for both). CONCLUSIONS: Damage to the taper junction is commonly found in retrieved RTSAs and can occur after only months of being implanted. In this study, tribocorrosion predominantly occurred on the taper surface of the baseplate (vs. glenosphere) and on the humeral stem (vs. tray or spacer), which may relate to the flexural rigidity difference between the titanium and cobalt-chrome components. Bolt reinforcement and the use of large-diameter trunnions led to less tribocorrosion of the taper junction. The findings of this study provide evidence for the improved design of RTSA prostheses to decrease tribocorrosion.

Triglyceride (TG)-lowering LPL variants in combination with genetic LDL-C-lowering variants are associated with reduced risk of coronary artery disease (CAD). Genetic variation in the APOA5 gene encoding apolipoprotein A-V also strongly affects TG levels, but the potential clinical impact and underlying mechanisms are yet to be resolved. Here, we aimed to study the effects of APOA5 genetic variation on CAD risk and plasma lipoproteins through factorial genetic association analyses. Using data from 309,780 European-ancestry participants from the UK Biobank, we evaluated the effects of lower TG levels as a result of genetic variation in APOA5 and/or LPL on CAD risk with or without a background of reduced LDL-C. Next, we compared lower TG levels via APOA5 and LPL variation with over 100 lipoprotein measurements in a combined sample from the Netherlands Epidemiology of Obesity study (N = 4,838) and the Oxford Biobank (N = 6,999). We found that lower TG levels due to combined APOA5 and LPL variation and genetically-influenced lower LDL-C levels afforded the largest reduction in CAD risk (odds ratio: 0.78 (0.73-0.82)). Compared to patients with genetically-influenced lower TG via LPL, genetically-influenced lower TG via APOA5 had similar and independent, but notably larger, effects on the lipoprotein profile. Our results suggest that lower TG levels as a result of APOA5 variation have strong beneficial effects on CAD risk and the lipoprotein profile, which suggest apo A-V may be a potential novel therapeutic target for CAD prevention.

BACKGROUND: To date, no validated assessment of motor imagery (MI) ability with temporomandibular disorders (TMD) exists preventing identification of good imagers and appropriate MI use during TMD rehabilitation. OBJECTIVE: To assess the reliability and construct validity of the previously developed Tongue and Mouth Imagery Questionnaire (TMIQ) compared with the gold-standard Kinaesthetic and Visual Imagery Questionnaire (KVIQ-10). METHODS: Both KVIQ-10 and TMIQ assess MI ability using vividness (i.e. clarity/brightness for visual MI, V MI; or intensity for kinesthetic MI, K MI) of MI using a 5-point Likert scale (1: no image/sensation, 5: clear/intense image/sensation). The KVIQ-10 was administered once (test) and the TMIQ twice (test-retest) to heathy participants and patients with TMD. Questionnaire validity was investigated using concurrent validity (Pearson correlation and paired t test); TMIQ-test-retest reliability (intraclass correlation coefficients, ICCs); internal consistency (Cronbach âº) and the factorial structure (principal factor extraction). RESULTS: A total of 94 participants were included (n = 47 per group). The mean vividness scores of the KVIQ-10 and the TMIQ were significantly correlated, and not significantly different for both groups indicating concurrent validity. ICCs in the control group (range: 0.82-0.90), and in the TMD group (range: 0.75-0.82) indicated good reproducibility. The Cronbach ⺠values were all above 0.94, indicating excellent reliability. Two factors were extracted corresponding to V MI and K MI, and explained 66% of total variance. CONCLUSION: The TMIQ is a valid and reproducible MI questionnaire showing excellent internal consistency and, therefore, can be used to assess imagined movements of the TM region in healthy individuals and patients with TMD.

For a number of years, the decline of honeybee (Apis mellifera) in North America and Europe has been the subject of much debate. Among the many factors proposed by hundreds of studies to explain this phenomenon is the hypothesis that agricultural activities using pesticides contribute to the weakness of bee colonies. Moreover, while urban beekeeping is presently booming in several cities, we do not know if this environment is more beneficial for bees than the typical, rural area. In the summer of 2018, we sampled honeybees (foragers and larvae) in rural (Laurentians) and urban (city of Montreal) areas and compared them using the following biomarkers: carotenoids, retinoids, α-tocopherol, metallothionein-like proteins (MTLPs), lipid peroxidation, triglycerides, acetylcholinesterase activity (AChE) and proteins. Pesticides, pharmaceuticals and personal care products (PPCPs) and metals were also quantified in honeybees' tissues. Our result revealed that, globally, urban foragers had higher levels of insecticides and PPCPs and that metals were in greater concentrations in urban larvae. Compared to rural foragers, urban foragers had higher concentrations of MTLPs, triglycerides, protein and AChE activity. The multifactorial analysis confirmed that insecticides, some metals and PPCPs were the most influential components in the contaminantâbiomarker relationships for both foragers and larvae.

Water scarcity causes a series of eco-environmental problems, such as land salinization, biodiversity reduction and food crisis, which seriously restricts the sustainable development of the Aral Sea basin. In this study, a stepwise-cluster factorial analysis (SCFA) approach is proposed for assessing the effects of natural condition and human activity on the outflow of Syr Darya River (abbreviated as OSR) that has significant effects on the eco-environmental restoration of the Aral Sea. SCFA coupled stepwise cluster analysis and factorial analysis cannot only reflect the variability of outflow, but also identify the driving factors quantitatively. The results disclose that, in 1960-1991, the dominant factors (affecting the OSR) are upstream inflow (25.77%) > agricultural water use of Uzbekistan (7.21%) > industrial water use of Uzbekistan (4.53%) > agricultural water use of Kazakhstan (3.81%) > Precipitation (3.66%); interactions between upstream inflow and agricultural water use of Uzbekistan, Kazakhstan and Tajikistan and interactions between reservoir and evapotranspiration have the significant effects on the OSR. Results also indicate that, in 1992-2015, the dominant factors that affect the OSR are agricultural water use of Uzbekistan (23.31%) > agricultural water use of Kazakhstan (22.15%) > industrial water use of Uzbekistan (8.31%) > domestic water use of Kazakhstan (4.68%) > agricultural water use of Tajikistan (4.54%) > domestic water use of Uzbekistan (4.41%); the interactions between industrial water use and agricultural water use of Uzbekistan, Kazakhstan and Tajikistan and the interactions between reservoir and upstream inflow have the pivotal effects on OSR. In the future, when the agricultural water use of the basin decrease as 4% and the industry water use of Uzbekistan decrease as 2%, the OSR may recover to the middle level of 1970s. The results help identify the major factors affecting the outflow of Syr Darya River as well as seek an effective approach to restore the eco-environment of Aral Sea basin.

Climate change has significant impacts on the Pearl River Basin, and the regional ecological environment and human production may face severe challenges in the future due to changes in temperature and precipitation, as well as their derivative disasters (e.g., drought). Therefore, a full understanding of the possible impacts of climate change on Pearl River Basin is desired. In this study, the potential changes in temperature, precipitation, and drought conditions were projected through a stepwise clustering projection (SCP) model driven by multiple GCMs under two different RCPs. The developed model could facilitate specifying the inherently complex relationship between predictors and predictands, and its performance was proven to be great by comparing the observations and model simulations. A multi-level factorial analysis was employed to explore the major contributing factors to the variations in projecting drought conditions. The results suggested that the Pearl River Basin would suffer significant increasing trends in Tmean (i.e., 0.25-0.34 °C per decade under RCP4.5 and 0.42-0.60 °C per decade under RCP8.5), and the annual mean precipitation would increase under both RCPs. The drought events lasting for 1-2 months would be decreased by 7.7%, lasting for 3-4 months would be increased by 4.3%, and lasting for more than five months would be increased by 3.4% under RCP4.5, respectively. While they changed to 6.1%, 1.4%, and 4.7% under RCP8.5, respectively. More medium and long-term drought events with higher drought severity would occur. GCM has dominant influences on four different responses of drought duration, accounting for 50.20%, 52.61%, 56.71%, and 56.24% of total variabilities, respectively. Meanwhile, the effects explained by GCM*RCP interactions cannot be neglected, with an average contribution rate of 44.37%, 37.86%, 37.66%, and 35.83%, respectively.

Virtual water is an important indicator measuring the amount of water needed from the perspective of consumption, which can help decision makers to identify desired system design and optimal management strategy against water resources shortage. In this study, a novel model named as factorial ecologically-extended input-output model (abbreviated as FEIOM) is developed for virtual water management. FEIOM integrates techniques of input-output model (IOM), ecological network analysis (ENA) and factorial analysis (FA) into a general framework. It is effective to evaluate the virtual water flows, reveal ecological inter-connections in virtual water system (VWS), and identify key water consumption sectors that have significant individual and interactive effects on VWS's performance. FEIOM is then applied to identifying optimal virtual water management strategies for Kazakhstan in Central Asia. The main findings are: (i) Kazakhstan is a net importer of virtual water (reaching up to 46.0 × 109 m3), demonstrating that the national economic structure is reasonable, which can abate the national water scarcity and improve its eco-environmental protection; (ii) the virtual water of agricultural sector is net exporter, where vegetables, fruits and nuts occupy 86% of the total agricultural exports; the massive export of water-intensive products further squeezes the water for other users; (iii) the key factors affecting the national VWS are agriculture > primary manufacturing > advanced manufacturing > services. Therefore, from solving water resources shortage and facilitating sustainable development perspectives, Kazakhstan should stimulate the domestic primary manufacturing productions and improve agriculture and advanced manufacturing water-use efficiencies.

Urban carbon emission is one of the hotspots of global change research. The interactive relationships among the carbon emissions of various sectors within the social-economic system are often overlooked. The goal of this research is to take Northeast Industrial District (NID) including Liaoning, Jilin and Heilongjiang provinces as a special case study to explore the impact of different emission sources on the internal interaction of carbon emission system. To do so, a carbon emission interactive network (CEIN) model is developed to demonstrate the interactions of embodied carbon emissions within the system using factorial analysis and input output analysis. The control situation and mutual interactions between sectors within the system of NID were examined, and the impacts of different emission sources on the internal interaction of carbon emission system were discussed. Moreover, the utility indicators were revised to distinguish the impact of system boundaries on changes in system ecological relationships and evaluate the comprehensive properties of the system. It is found that the indirect emissions of NID were huge. Liaoning Province had the largest total emissions during the study period, with indirect emissions accounting for more than 60% of the total emissions. The pulling weights of the agricultural sector in three provinces of the system were significantly insufficient, with all weights less than 3%. The driving weights of the agricultural, mining and primary manufacture sectors in three provinces were insufficient, while the driving weight of the advanced manufacture sector was excessively strong. The most important transmission department did not have the most important effect on the network mutualism. The definition of system boundary had a huge impact on the accounting of system emissions. The results will provide a scientific basis for guiding the reform of the carbon emission system and achieving regional coordinated development.

BACKGROUND: Inflammatory bowel disease comprises two conditions: ulcerative colitis and Crohn's disease. Inflammatory Bowel Disease Questionnaire 32 (IBDQ-32) is a specific questionnaire which has been translated from English into Spanish and validated. In the Spanish-speaking countries of America it has not been validated. The aim was to determine the psychometric properties, validity and reliability of the Mexican version of the IBDQ-32 questionnaire. METHODS: A total of 316 patients with inflammatory bowel disease and 100 healthy controls participated in the study. The questionnaires IBDQ-32 and SF-36 were issued on two occasions (separated by 15 days). The psychometric properties of the Mexican version of the IBDQ-32 questionnaire were determined. RESULTS: Patients with inflammatory bowel disease had an impaired quality of life compared to healthy controls. There were no differences between ulcerative colitis and Crohn's disease in the total scores of IBDQ-32 and its domains. The internal consistency reliability was good. The intraclass coefficient showed good reliability (repeated measurement) for total scale and all four subscales. Factor analysis explained variance is higher than 50% therefore is considered adequate/acceptable. The correlation between IBDQ-32 and SF-36 showed a satisfactory association. The social domain is the only one that presented a ceiling effect. CONCLUSIONS: The Mexican version of the IBDQ-32 quality of life questionnaire is valid and reliable. This sample included the entire spectrum of inflammatory disease (remission and activity) and was comparable when assessing quality of life with the SF-36 generic questionnaire.

PURPOSE: Eating self-efficacy (ESE) is the belief in one's ability to self-regulate eating. Social and emotional situations may be differently challenging depending on the individual eating habits, body mass index and affects. Several ESE scales have been developed but most of them focus on weight management, dieting or healthy eating. The aim of the study was to validate a new brief scale assessing ESE in situations in which people face social or emotional pressures for excessive food intake. METHODS: Study 1: A sample of 412 volunteer females (age M = 25.44 ± 5.03) completed a first 25-item version of the scale. Exploratory factor analysis (EFA) was conducted for selecting a subgroup of items composing the ESE brief scale (ESEBS). Study 2 assessed its psychometric properties through a Confirmatory Factor Analysis (CFA), analyzing the responses of 410 volunteer adults (273 females, 137 males). RESULTS: EFA of Study 1 evidenced a bifactorial structure. Four items for each factor were selected, explaining 63% of the variance. Study 2 confirmed the good fit of the bifactorial model (CFI = 0.9589; χ2 (19) = 62.852, p < 0.001; RMSEA = 0.075; SRMR = 0.040) and provided support for the measurement invariance of the scale across gender. The internal consistency was as follows: Social (α = 0.786), Emotional (α = 0.820). The concurrent validity of the subscales was demonstrated by significant latent negative correlations with measures of eating disorders and emotional eating. CONCLUSIONS: The 8-items ESEBS appears as a valid and reliable instrument to assess eating self-efficacy related to social and emotional situations. Future studies should evaluate its potential use in non-clinical and clinical research and interventions. LEVEL OF EVIDENCE: Level V, descriptive cross-sectional study.

OBJECTIVES: This study investigated the psychometric properties of the Cognitive Emotion Regulation Questionnaire in a Tunisian Arabic-Speaking population. METHOD: The CERQ original version was translated and back-translated and then administrated to 360 participants. Afterwards, using the explanatory and confirmatory factor analysis, we studied the latent factor structure for CERQ. The internal consistency of the subscales was assessed by Cronbach's alfa coefficients, and the test-retest and interscale reliability were assessed with Pearson correlations. The criterion validity was also examined using correlations between the CERQ subscales and both the Beck Depression Inventory (BDI) and State-Trait Anxiety Inventory (STAI) scales at first evaluation and at a one year follow-up. RESULTS: The Tunisian version was quite satisfactory with the nine-factor structure, as in the original CERQ. Moreover, all validity and reliability measures were comparable to the original CERQ. CONCLUSION: The present study is the first work devoted to a Tunisian adaptation of the CERQ. Our findings highlight that the Tunisian version is both reliable and valid for the measurement of cognitive emotion regulation strategies.

In this study, a factorial Bayesian copula (FBC) method is proposed to quantify parameter uncertainties in copula-based models and then reveal their impacts on hydrologic risk inferences within a multivariate context. In detail, Bayesian inference and factorial analysis are integrated into copula-based multivariate risk models to (1) quantify parameter uncertainties, (ii) reveal their individual and interactive effects, and (iii) identify their detailed contributions to uncertain risk inferences. Streamflow observations at Xiangxi and Wei River basins is China are used to illustrate the applicability of FBC. The results indicate that imprecise parameters in marginal distributions and the dependence structure would lead to extensive uncertainties in predictive joint return periods and failure probabilities. Also, individual and interactive effects of parameters are well revealed through multilevel factorial analysis, and the detailed contributions of one parameter to different failure probabilities under different service time scenarios are identified.

Independent reports describe the structural differences between the human male and female brains and the differential gender-susceptibility to mood disorders. Nutrition is one of the modifiable risk factors that has been reported to impact brain chemistry and affect mental health. Objectives: To study dietary patterns in adult men and women in relation to mental distress. Another aim was to develop evidence-based prototypes using System Dynamic Modeling methodology to better describe our findings. Methods: An anonymous internet-based survey was sent through social media platforms to different social and professional networks. Multivariate analyses were used for data mining. Data were stratified by gender and further by tertiles to capture the latent variables within the patterns of interest. Results: Mental distress in men associated with a consumption of a Western-like diet. In women, mental wellbeing associated with a Mediterranean-like diet and lifestyle. No other patterns in both genders were linked to mental distress. Based on the generated prototypes, men are more likely to experience mental wellbeing until nutritional deficiencies arise. However, women are less likely to experience mental wellbeing until a balanced diet and a healthy lifestyle are followed. In men, dietary deficiencies may have a profound effect on the limbic system; whereas dietary sufficiency in women may potentiate the mesocortical regulation of the limbic system. Discussion and conclusion: Our results may explain the several reports in the literature that women are at a greater risk for mental distress when compared to men and emphasize the role of a nutrient-dense diet in mental wellbeing.

INTRODUCTION: We investigated the association between atrophy subtypes of Alzheimer's disease (AD), the ATN classification scheme, and key demographic and clinical factors in 2 cohorts with different source characteristics (a highly selective research-oriented cohort, the Alzheimer's Disease Neuroimaging Initiative [ADNI]; and a naturalistic heterogeneous clinically oriented cohort, Karolinska Imaging Dementia Study [KIDS]). METHODS: A total of 382 AD patients were included. Factorial analysis of mixed data was used to investigate associations between AD subtypes based on brain atrophy patterns, ATN profiles based on cerebrospinal fluid biomarkers, and age, sex, Mini Mental State Examination (MMSE), cerebrovascular disease (burden of white matter signal abnormalities, WMSAs), and APOE genotype. RESULTS: Older patients with high WMSA burden, belonging to the typical AD subtype and showing A+T+N+ or A+T+N- profiles clustered together and were mainly from ADNI. Younger patients with low WMSA burden, limbic-predominant or minimal atrophy AD subtypes, and A+T-N- or A+T-N+ profiles clustered together and were mainly from KIDS. APOE ε4 carriers more frequently showed the A+T-N- and A+T+N- profiles. CONCLUSIONS: Our findings align with the recent framework for biological subtypes of AD: the combination of risk factors, protective factors, and brain pathologies determines belonging of AD patients to distinct subtypes.

Spilled oil frequently reaches the shorelines and affects coastal biota and communities. The application of surface washing agents is an important shoreline cleanup technique that can help remove stranded oil from substrate surfaces with the advantages of high removal efficiency, low toxicity, and strong economic viability. In this study, the investigation into the oil removal from contaminated sand using a surface washing agent under variable environmental conditions was conducted. A preliminary test was conducted to obtain the optimal combination of operating factors of surface washing agent-to-oil ratio (SOR) 2:1, mixing speed 150 rpm, and mixing time 30 min. The results of single-factor experiments showed that high temperature and humic acid concentration of flush water contributed to the performance of a surface washing agent, while salinity and kaolinite concentration could inhibit its performance. The factorial analysis revealed the main effects of temperature and salinity, and the interactive effects of temperature and salinity as well as salinity and humic acid concentration that were significant to the washing efficiency of the surface washing agent. In addition, the comprehensive assessment of a surface washing agent from the aspects of toxicity, detergency, dispersion properties, and field trials was conducted. The results have significant implications for future application of surface washing agents in the shoreline cleanup.

In this study, a factorial fuzzy chance-constrained programming (FFCP) method is developed for planning urban agglomeration ecosystem under uncertainty. FFCP cannot only address uncertainties presented as possibility distributions and random variables, but also quantitatively evaluate the individual and the interactive effects of multiple eco-environmental factors on urban agglomeration ecosystem. The FFCP method is applied to planning the Guangzhou-Foshan-Zhaoqing (GFZ) urban agglomeration, one of the most important economic circles of the Pearl River Delta region. With rapid industrialization, urbanization and population growth, a number of eco-environmental issues (e.g., water contamination, air pollution, and ecological deterioration) are becoming more and more serious. Results reveal that (i) the main environmental problem of Guangzhou is water pollution due to excessive chemical oxygen demand (COD) discharge (with a contribution of 53.1%), (ii) Foshan would suffer air pollution due to sulfur dioxide (SO2) emitted from industrial processing and production, and (iii) solid waste disposal would continue to be challenge faced by Zhaoqing. Results also disclose that multiple uncertainties in system components have sound effects on the urban agglomeration ecosystem management as well as eco-environmental constraints have individual and/or joint effects on the system benefit. Different probability levels would lead to changed land-use patterns. Decisions at a lower violation level would lead to an increased reliability in fulfilling ecosystem requirement but with a lower system benefit. The findings can help decision makers to gain insights into tradeoff between economic development and eco-environmental protection as well as generate synergetic development strategies of urban agglomeration in an economic-effective and ecological-harmonious way.

BACKGROUND: Traditional toxicological studies focus on individual compounds. However, this single-compound approach neglects the fact that the mixture exposed to human may act additively or synergistically to induce greater toxicity than the single compounds exposure due to their similarities in the mode of action and targets. Mixture effects can occur even when all mixture components are present at levels that individually do not produce observable effects. So the individual chemical effect thresholds do not necessarily protect against combination effects, an understanding of the rules governing the interactive effects in mixtures is needed. The aim of the study was to test and analyze the individual and combined estrogenic effects of a mixture of three endocrine disrupting chemicals (EDCs), bisphenol A (BPA), nonylphenol (NP) and diethylstilbestrol (DES) in immature rats with mathematical models. METHOD: In the present study, the data of individual estrogenic effects of BPA, NP and DES were obtained in uterotrophic bioassay respectively, the reference points for BPA, NP and DES were derived from the dose-response ralationship by using the traditional no observed adverse effect (NOAEL) or lowest observed adverse effect level (LOAEL) methods, and the benchmark dose (BMD) method. Then LOAEL values and the benchmark dose lower confidence limit (BMDL10) of single EDCs as the dose design basis for the study of the combined action pattern. Mixed prediction models, the 3 × 2 factorial design model and the concentration addition (CA) model, were employed to analyze the combined estrogenic effect of the three EDCs. RESULTS: From the dose-response relationship of estrogenic effects of BPA, NP and DES in the model of the prepuberty rats, the BMDL10(NOAEL) of the estrogenic effects of BPA, NP and DES were 90(120) mg/kg body weight, 6 mg/kg body weight and 0.10(0.25) µg/kg body weight, and the LOAEL of the the estrogenic effects of three EDCs were 240 mg/kg body weight, 15 mg/kg body weight and 0.50 µg/kg body weight, respectively. At BMDL10 doses based on the CA concept and the factorial analysis, the mode of combined effects of the three EDCs were dose addition. Mixtures in LOAEL doses, NP and DES combined effects on rat uterine/body weight ratio indicates antagonistic based on the CA concept but additive based on the factorial analysis. Combined effects of other mixtures are all additive by using the two models. CONCLUSION: Our results showed that CA model provide more accurate results than the factorial analysis, the mode of combined effects of the three EDCs were dose addition, except mixtures in LOAEL doses, NP and DES combined effects indicates antagonistic effects based on the CA model but additive based on the factorial analysis. In particular, BPA and NP produced combination effects that are larger than the effect of each mixture component applied separately at BMDL doses, which show that additivity is important in the assessment of chemicals with estrogenic effects. The use of BMDL as point of departure in risk assessment may lead to underestimation of risk, and a more balanced approach should be considered in risk assessment.

Multi-soil-layering (MSL) system was developed as an attractive alternative to traditional land-based treatment techniques. Within MSL system, the environmental cleanup capability of soil is maximized, while the soil microbial communities may also change during operation. This study aimed to reveal the nature of biophysiological changes in MSL systems during operation. The species diversity in soil mixture blocks was analyzed using Illumina HiSeq sequencing of the 16S rRNA gene. The interactive effects of operating factors on species richness, community diversity and bacteria abundance correlated with COD, N and P removal were revealed through factorial analysis. The results indicated the main factors, aeration, bottom submersion and microbial amendment, had different significant effects on microbial responses. The surface area and porosity of zeolites in permeable layers decreased due to the absorption of extracellular polymeric substances. The findings were applied for the design and building of a full-size MSL system in field and satisfied removal efficiency was achieved. The results of this study can help better understand the mechanisms of pollutant reduction within MSL systems from microbial insights. It will have important implications for developing appropriate strategies for operating MSL systems with high efficiency and less risks.
