This study was aimed to determine the prevalence of Toxocara canis/cati, Strongyloides stercoralis, Giardia spp., and Cryptosporidium spp., which occur and are potentially zoonotic to humans in domestic dogs and cats in Moscow (Russia). The fecal flotation method and larvae detection by microscopy of a direct feces smear were performed to detect Toxocara, Giardia spp., and Cryptosporidium spp. The total parasitic prevalence in dogs was as follows: Giardia spp.: 10.2 % (226/2208), Cryptosporidium spp.: 2.7 % (60/2208), T. canis: 2 % (45/2208), S. stercoralis larvae: 1.1 % (25/2208). The younger animals under were infected more than those over 12 months of age (p<0.001). The preva lence rates were along these lines: Giardia spp. (18.2 %), Cryptosporidium spp. (5.7 %), T.canis (3 %), S. stercoralis larvae (2.3 %). The overall prevalence in cats was as follows: Giardia spp. - 5.2 % (71/1350), Cryptosporidium spp. - 4.8 % (65/1350), T. cati - 4.1 % (56/1350). Similarly to dogs, the infection rates were higher in cats under 12 months of age Giardia spp. (8.2 %), Cryptosporidium spp. (8.6 %), T. cati (7.5 %. Analysis of combined infections in dogs revealed the following combinations: Giardia spp. and Cryptosporidium spp. (35.5 %) larvae of S. stercoralis sp. and Giardia spp. (32.3 %), T.canis and Giardia spp. (22.6 %), T.canis and Cryptosporidium spp. (6.6 %), T.canis and S.stercoralis and (3.2 %), respectively. In cats, only two coinfections by Giardia spp. and Cryptosporidium spp. (58.3 %), and T.cati with Giardia spp. (41.7 %) were noticed. Further research is needed to study the spread of parasitic diseases in pet animals. The data will improve countermeasures to prevent these diseases' spread among animals and humans.

Giardia spp. is a common gastrointestinal (GI) parasite of multiple host species, including dogs and humans, with the potential for zoonotic transmission. The risk of GI parasitism in dogs (including Giardia spp.) may increase with park use in urban areas. This study aimed to (1) determine whether park attendance is a risk factor for Giardia spp. infection in dogs and (2) characterize the behavioural and demographic risk factors for Giardia spp. infection in park-attending and non-park-attending dogs. From August to September 2012, a total of 1293 dog owners completed a survey and 860 corresponding dog faecal samples were collected. Dog faeces were screened for Giardia spp. using a direct immunofluorescence assay and associations assessed among behaviours, demographics, and Giardia spp. infection. Main results included off-leash and swimming frequencies within parks as significantly positively associated with Giardia spp. infection in dogs. Dog-owner age was negatively associated with off-leash and swimming frequencies in parks. The results suggest some recreational behaviours in parks and certain demographics are risk factors for parasitism in pet dogs.

The order Testudines comprises some of the most endangered groups of vertebrates. Under specific circumstances, infectious and parasitic diseases may affect the survival and fecundity of these animals, potentially threatening Testudines populations. In Sardinia, besides the three species of tortoises present in the wild (Testudo hermanni, Testudo graeca and Testudo marginata), many others are kept as pets. However, epidemiological studies on these animals have not been conducted so far. Thus, the aim of the work was to investigate the presence of gastrointestinal parasites in captive and wild tortoises of Sardinia, Italy, with particular regard to those of zoonotic importance. For the 215 animals examined (n = 36 wild caught and n = 179 private-owned), fecal samples were collected and processed by flotation and modified Ziehl-Neelsen technique. An overall prevalence of 81.4% for endoparasites was detected, with oxyurids being the most prevalent (74.4%), followed by Nyctotherus spp. (18.6%), Cryptosporidium spp. (12.6%), Angusticaecum spp. (2.8%), strongyles (0.9%), Balantidium spp. (0.9%), coccidia (0.9%), cestodes (0.5%), and Giardia spp. (0.5%). Data suggest that tortoises are affected by a great variety of endoparasites, and further molecular analysis are required to assess the impact of Cryptosporidium and Giardia species in these hosts. Therefore, regular health screenings are of importance for the management of these animals and for preventing emerging infectious diseases.

Parasitic infections of the gastrointestinal tract of domestic animals play a major role in the transmission of disease, which in turn may result in financial and productive losses. Notwithstanding, studies on the burden and distribution of diarrheagenic protists in zoological gardens are still insufficient. Given the close animal-animal and animal-human interaction in these settings, Public Health concerns under the One Health context are raised. Using molecular detection tools and phylogenetic analysis, the goal of this study was to assess the occurrence of four potentially zoonotic protists-Balantioides coli, Blastocystis sp., Cryptosporidium spp. and Giardia spp.-in animals residing in zoological parks in Portugal. Occurrence of Eimeria spp. was also assessed because of its veterinary relevance. Although Blastocystis sp. represents most of the positive samples obtained (11.6%; 95% CI: 0.08-0.17), all parasites were detected (B. coli (2.9%; 95% CI: 0.01-0.06), and Cryptosporidium spp., Eimeria spp. and Giardia spp. presented the same prevalence (0.5%; 95% CI: 0.00-0.03)). We also describe the first molecular detection of B. coli in a collared peccary (Tayassu tajacu), of Blastocystis sp. in three different python species, and G. muris in a central bearded dragon (Pogona vitticeps), suggesting the broadening of the host range for these parasites.

Multi-host communities are perfect scenarios for the emergence and spread of pathogens, threatening the recovery of endangered, isolated, or inbred populations, such as the brown bear (Ursus arctos) in northwestern Spain. The population recovery in recent years has forced bears to occupy highly anthropized areas, increasing their interaction with human and domestic animals, with potential consequences for global health. During 2022-2023 a survey of parasites, bacteria and viruses shared between wildlife, domestic animals and humans was performed in this population using non-invasive surveillance, i.e., bear fecal samples (n = 73) and sponge-based sampling of trees (n = 42; 14 rubbed trees and 28 control trees). Pathogen detection rates were defined as the percentage of qPCR or culture-positive samples. Generalized linear models were fitted to assess their relationship with environmental variables including dispersion of the human population, and percentage of agricultural and periurban habitats in a 6 km-buffer around each sample. Canine Adenovirus type 1 (45.2%), Giardia spp. (15.1%), Salmonella spp. (12.3%), and extended-spectrum-beta-lactamases (ESBL) Escherichia coli (1.4%) were identified in fecal samples. In contrast, only five sponges from three rubbed and two control trees resulted positive to E. coli (14.3%). The results suggest that several pathogens are common in the Cantabrian brown bear population and that anthropization of the territory modulates their prevalence and richness. The effective design of management programs for bear conservation will require a one-health approach, in which genetic analysis of non-invasive samples can be key tools for the sanitary surveillance at the wildlife-livestock-human interface.

This study examined the correlation between intestinal protozoans and the bacterial microbiome in faecal samples collected from 463 patients in New Zealand who were diagnosed with gastroenteritis. In comparison to traditional microscopic diagnosis methods, Multiplexed-tandem PCR proved to be more effective in detecting intestinal parasites. Among the identified protozoans, Blastocystis sp. and Dientamoeba fragilis were the most prevalent. Notably, D. fragilis was significantly associated with an increase in the alpha-diversity of host prokaryotic microbes. Although the exact role of Blastocystis sp. and D. fragilis as the primary cause of gastroenteritis remains debatable, our data indicates a substantial correlation between these protozoans and the prokaryote microbiome of their hosts, particularly when compared to other protists or patients with gastroenteritis but no detectable parasitic cause. These findings underscore the significance of comprehending the contributions of intestinal protozoans, specifically D. fragilis, to the development of gastroenteritis and their potential implications for disease management.

Objective: Enteric diseases remain a serious health problem globally. High prevalence is evident in regions with poor socioeconomic conditions, poor sanitation, and inadequate clean water supply, such as South Africa. Designing an effective strategy, however, requires local knowledge, which can be particularly challenging to acquire in low-and middle-income countries. As the first step in this process, we investigated the prevalence and distribution of protozoan parasites Cryptosporidium and Giardia in the rural and urban gastrointestinal clinics of South Africa. Methods: A cross-sectional study was conducted to assess the prevalence of enteric parasites Cryptosporidium and G. lamblia in rural and urban communities of South Africa. Stool samples were collected from November 2013 to June 2015 from patients with diarrhea (n=227) and without diarrhea (n=257). DNA was extracted and a diagnostic Taqman qPCR assay was performed to detect these protozoan parasites, which was further confirmed by the Sanger sequencing of a few samples. Results: Of the 484 stool specimens collected, 34% (166/484) were positive for either Cryptosporidium spp. or Giardia lamblia parasites, with only 5% containing both parasites (22/484). In both study populations, Cryptosporidium was the most prevalent parasite (overall 25%) followed by Giardia (19%). Conclusion: This study discovered that both Giardia and Cryptosporidium parasites might contribute to diarrheal disease in South Africa and are more prevalent in rural communities. Future studies are needed to identify the source of the infection and design appropriate interventions to reduce the burden of the disease. Objective: The presence of protozoan parasites in agricultural irrigation and drinking water resources at Denizli city center was investigated in detail for the first time. Methods: The research was carried out between October 2017 and October 2018 and 84 water samples were taken from 7 different stations identified from the Denizli city center. After examining the samples by direct visualization (Native-Lugol), they were stained with quinton's acid fast, giemsa and trichrome dyes. The preparations were evaluated parasitologically under a light microscope. Results: Cryptosporidium spp. was detected in 21 (25%) of 36 agricultural irrigation water samples collected during the study, Cyclospora cayetanensis in 5 samples (5.95%) and Giardia spp. in 12 samples (14.28%). No parasite findings were found in any of the 48 drinking water samples collected. Conclusion: The widespread use of animal husbandry and agriculture as grazing land in the sampling stations, the mixing of domestic wastewater into these waters without any treatment, and seasonal conditions cause the protozoan parasites to be seen more in certain periods. It is thought that waterborne protozoan infections that may occur in the future can be significantly prevented by taking the necessary precautions in terms of public health and environmental animal husbandry.

Intestinal protozoan infection is a persisting public health problem affecting the populations of developing countries in the tropical and subtropical regions. The diagnosis of intestinal protozoa remains a challenge especially in developing countries due to a shortage of laboratory facilities, limited health funding, and the remoteness of communities. Despite still being widely used, conventional diagnoses using microscopy and staining methods pose important limitations, particularly due to their low sensitivities and specificities. The selection of diagnostic methods needs to be carefully considered based on the objective of examination, availability of resources, and the expected parasite to be found. In this review, we describe various immunodiagnosis and molecular diagnostic methods for intestinal protozoa infection, including their advantages, disadvantages, and suitability for different settings, with a focus on Entamoeba histolytica, Giardia duodenalis, and Cryptosporidium spp.

The efficiency of microalgae harvesting on the removal of Giardia spp. cysts, Cryptosporidium spp. oocysts, total coliforms, Escherichia coli, Enterococcus spp. and Clostridium spp. was assessed in lab-scale experiments (Jartest and Flotatest) using effluent from a flat panel photobioreactor used for Chlorella sorokiniana cultivation. Three harvesting methods were evaluated: (1) flocculation induced by pH modulation followed by sedimentation (pH-SED), (2) flocculation induced by pH modulation followed by dissolved air flotation (pH-DAF), and (3) coagulation using an organic coagulant (Tanfloc SG) followed by dissolved air flotation (Coag-DAF). The results indicated that the three harvesting methods were efficient in removing protozoan (oo)cysts and bacteria, achieving percentages of removal higher than 97% for all the analyzed pathogens. Among the three methods, pH-SED showed the best removal performance: 99.60% (2.5 log) for Giardia spp. cysts, 100% (>6.3 log) for total coliforms, 100% (>4.6 log) for Escherichia coli, 100% (>5.8 log) for Enterococcus spp. and 99.96% (3.6 log) for Clostridium spp. Clostridium spp. seemed to be more tolerant to the harvesting methods than the other groups of bacteria analyzed in the study, and its presence was positively correlated to the presence of Giardia spp. cysts.

Disease may limit recovery of endangered species. We surveyed parasites in the federally endangered salt marsh harvest mouse (SMHM; Reithrodontomys raviventris halicoetes) and sympatric rodents in Suisun Marsh (Solano County, California, USA) from April 2018 through March 2019. We investigated individual SMHM risk factors (age, sex, reproductive status, and body condition) for infection and relationships among the estimated parasite prevalence and season and habitat management (natural tidal habitats versus diked, nontidal habitats). We captured 625 individual rodents, including 439 SMHM, and tested these for infection with Bartonella spp., Borrelia spp., Rickettsia spp., Francisella tularensis, Leptospira spp., Cryptosporidium spp., Giardia spp., and Toxoplasma gondii by PCR. Over one-third (34.6%, confidence interval [CI], 30.2-39.3%) of SMHM tested positive for at least one parasite. Four percent (CI, 2.8-6.3%) of SMHM were infected with F. tularensis holarctica, a virulent bacterium that causes mortality in rodents shortly after infection. Additionally, we detected three species of Bartonella (B. henselae, B. rochalimae, B. vinsonii arupensis), Leptospira borgpetersenii serovar Ballum, Cryptosporidium sp. (deer mouse [Peromyscus maniculatus] genotype), Cryptosporidium parvum, Giardia intestinalis, and an unidentified Borrelia sp. The only parasite that was associated with habitat management was Bartonella spp., which was more prevalent in diked than tidal areas. Male SMHM were more likely to be parasitized than females, and individuals in modestly poor body condition were most likely to be infected with Bartonella spp. The estimated sample prevalence of multiple parasites varied by season and by host species. This is the first major parasite assessment in a long-endangered species, and these results will assist managers to incorporate parasitic disease into recovery planning and provide a critical baseline for future investigations, including how climatically induced habitat and species composition changes could alter disease dynamics.

Cats (Felis catus) have the potential to spread and infect humans and other animals with a variety of endoparasites. This study was conducted to investigate the prevalence of parasites in community, outdoor, and strictly indoor domestic cats in Rockland County, New York. Between November 2020 and June 2021, fecal samples were collected from 3 populations (community = 65, outdoor domestic = 28, strictly indoor domestic = 35). A minimum of 2 g of feces were collected per sample and examined with centrifugal fecal floatation tests with 33% zinc sulfate solution and microscopy. Overall, community cats had a significantly higher prevalence of parasites compared to outdoor and strictly indoor cats (chi-square test, X2 = 24.943, df = 2, P = 3.835 × 10-6). The most prevalent endoparasite in all 3 populations was Toxocara cati. There was a significant difference between the 3 populations and parasite richness (chi-square test, X2 = 25.452, df = 4, P = 4.081 × 10-5). Polyparasitism was frequently observed in the community population, followed by the outdoor population. This study demonstrates that because community cats spend their entire lives outdoors, they are more at risk of being exposed to parasites and contribute to the contamination of the environment with these parasites.

Giardiasis in humans is a gastrointestinal disease transmitted by the potentially zoonotic Giardia duodenalis genotypes (assemblages) A and B. Small wild rodents such as mice and voles are discussed as potential reservoirs for G. duodenalis but are predominantly populated by the two rodent species Giardia microti and Giardia muris. Currently, the detection of zoonotic and non-zoonotic Giardia species and genotypes in these animals relies on cumbersome PCR and sequencing approaches of genetic marker genes. This hampers the risk assessment of potential zoonotic Giardia transmissions by these animals. Here, we provide a workflow based on newly developed real-time PCR schemes targeting the small ribosomal RNA multi-copy gene locus to distinguish G. muris, G. microti and G. duodenalis infections. For the identification of potentially zoonotic G. duodenalis assemblage types A and B, an established protocol targeting the single-copy gene 4E1-HP was used. The assays were specific for the distinct Giardia species or genotypes and revealed an analytical sensitivity of approximately one or below genome equivalent for the multi-copy gene and of about 10 genome equivalents for the single-copy gene. Retesting a biobank of small rodent samples confirmed the specificity. It further identified the underlying Giardia species in four out of 11 samples that could not be typed before by PCR and sequencing. The newly developed workflow has the potential to facilitate the detection of potentially zoonotic and non-zoonotic Giardia species in wild rodents.

Parasitic infections are a public health concern as they can cause aggression, growth retardation, weight loss, anemia, and other health problems. In this study, a systematic review and meta-analysis were performed to find an association between the probable social-environmental risk factors including lack of safe drinking water, no hand washing, sex, age, no access to education, no access to toilet, and Giardia spp infection. We conducted literature research among international databases including Scopus, PubMed, Web of Science and Cochrane from1 January 1995 to March 15, 2020, including 23 articles with 102 studies while the odds ratio (OR) was calculated using 2 × 2 tables or extracted. The pooled effect size with 95% confidence intervals (CIs) was estimated using a random or fixed-effects model. The results show a significant association between intestinal Giardia spp infection risk and lack of safe drinking water (OR: 1.14; 95%CI: 1.02-1.25); no access to toilet (OR: 1.22; 95%CI: 1.07-1.37); and age (2-10 vs 10-30 year) (OR: 1.42; 95%CI: 1.09-1.78). An insignificant association was observed between intestinal Giardia spp infection risk and age (<2 vs 2-10 year) (OR: 0.89; 95%CI: 0.75-1.02); no access to education (OR: 1.10; 95%CI: 0.80-1.40); Sex (male vs female) (OR: 1.04; 95%CI: 0.74-1.34); and no handwashing (OR: 1.18; 95%CI: 0.87-1.49). The results of meta-regression also indicate that intestinal Giardia spp infection has decreased over time (C= -0.024, p-value = 0.03) significantly, and also by growth of human development index (HDI) (C= - 0.42, p-value = 0.29) insignificantly. Improvement of personal hygiene, sanitation and raising public awareness can be effective in reducing Giardia spp infections.

Lab-scale studies were carried out to investigate the efficiency of dissolved air flotation (DAF) for the removal of Giardia spp. cysts and Cryptosporidium spp. oocysts from anaerobic effluent from the pilot UASB reactor. Raw wastewater, UASB and DAF effluent samples were collected weekly and protozoan (oo)cysts were concentrated using IMS followed by protozoa detection using immunofluorescense assay (IFA). The number of Cryptosporidium spp. oocysts in the raw wastewater was always lower than that of Giardia spp. cysts with 28-33 oocystsâ L-1 and 3177-4267 cystsâ L-1, respectively. Log10 removal of Giardia cysts utilising polyaluminium chloride (PACl) was higher than that with FeCl3, but no statistically significant difference between the two coagulants was observed. Cryptosporidium was absent in most of the treated effluent samples. The results indicate that DAF reached more than 2 log of cyst removal. In addition, the results demonstrated that these parasites are prevalent in the study area and E. coli and total coliforms were not good indicator microorganisms in terms of cyst and oocysts numbers.

Giardia spp. is detected frequently in humans and animals. Although many studies have been conducted on the epidemiology of giardiasis, there is a scarcity of information on the genetic diversity and the dynamics of transmission of Giardia spp. in Vietnam. The zoonotic potential of Giardia spp. remains elusive. The objective of this study was to determine the genetic diversity of Giardia spp. in both humans and livestock to assess the existence of a route of infection between livestock and humans. Our goal was to assess the role animals play in the epidemiology of human infection in northern Vietnam. In Hien Khanh commune in northern Vietnam, 311 households with 1508 residents were randomly selected for a diarrheal cohort study. Of these, 2120 human diarrheal samples were collected from 1508 residents in 2014 and 2017. Of these, non-diarrheal samples were cross-sectionally collected from 471 residents. At the same site, livestock samples from buffalo, dairy and beef cattle, pigs, and dogs were collected. All stool samples were examined for Giardia spp. by Direct Immunofluorescence Assay (DFA) using fluorescent microscope. DNA extraction, PCR analysis of the 3 genes (bg, gdh, tpi), and sequencing analysis were continuously carried out. A total of 23 animal stool samples, 8 human non-diarrheal samples, and 36 human diarrheal samples were Giardia spp. were positive by PCR using the bg and gdh genes. Giardia spp. assemblage AII and E were detected in both animal samples and human samples in this study site. The detection of assemblage E in human stool samples suggests the first human case report in Vietnam. We assume that the unexpected human infection of all Giardia assemblages including A, B, and E may be due to an environment contaminated with animal and human feces in this village.

BACKGROUND: Infections by protozoans of the genus Giardia are a common cause of diarrhea in dogs. Canine giardiosis constitutes a disease with a zoonotic potential; however, it is often underestimated due to its challenging diagnosis. The objective of the study was to assess the diagnostic performance of an immunochromatographic strip test (SpeedTM Giardia, Virbac, France) comparing it with microscopy (zinc sulfate flotation) by utilizing the combination of an enzyme immunoassay (ProSpecTTM Giardia EZ Microplate Assay, Oxoid Ltd., UK) and the PCR as the gold standard. A positive result in both ELISA and PCR was set as the gold standard. METHODS: Initially, fecal samples from dogs with clinical signs compatible with giardiosis were tested with the SpeedTM Giardia test and separated into two groups of 50 samples each: group A (positive) and group B (negative). Thereafter, all samples were examined by zinc sulfate centrifugal flotation technique and assayed by the ProSpecTTM Giardia Microplate Assay and PCR. The performance of the SpeedTM Giardia and zinc sulfate centrifugal flotation tests were calculated estimating sensitivity, specificity, and positive and negative likelihood ratio; the chi-square and McNemar tests were used for the comparison of the two methods. RESULTS: Giardia cysts were not detected by microscopy in 16 out of the 50 samples (32%) of group A and in none of group B samples. Eight out of 50 samples in group B (16%) were tested positive both with the ProSpecTTM Giardia Microplate Assay and PCR. Fecal examination with the SpeedTM Giardia test was more sensitive (86.2%) than the parasitological method (58.6%, P < 0.001) while the specificity of both methods was 100%. CONCLUSIONS: The SpeedTM Giardia test is an easy-to-perform diagnostic method for the detection of Giardia spp., which can increase laboratory efficiency by reducing time and cost and decrease underdiagnosis of Giardia spp. infections. This immunochromatographic strip test may be routinely exploited when a rapid and reliable diagnosis is required, other diagnostic techniques are unavailable and microscopy expertise is inefficient. In negative dogs with compatible clinical signs of giardiosis, it is recommended either to repeat the exam or proceed with further ELISA and PCR testing.

Giardia spp. and Cryptosporidium spp. are common gastrointestinal parasites with the potential for zoonotic transmission. This study aimed to (1) determine the genotypes occurring in dogs and coyotes occupying a similar urban area; (2) determine if these hosts were infected with potentially zoonotic genotypes; (3) provide baseline molecular data. In August and September 2012, 860 dog owners living in neighborhoods bordering six urban parks in Calgary, Alberta, Canada, provided faecal samples from their dogs. From March 2012 through July 2013, 193 coyote faeces were also collected from five of six of the same parks. Direct immunofluorescence microscopy (DFA) indicated that Giardia spp. and Cryptosporidium spp. infected a total of 64 (7.4%) and 21 (2.4%) dogs, as well as 15 (7.8%) and three (1.6%) coyotes, respectively. Semi-nested, polymerase chain reactions targeting the 16S small-subunit ribosomal ribonucleic acid (SSU rRNA) and 18S SSU rRNA genes of Giardia spp. and Cryptosporidium spp., respectively, were conducted on samples that screened positive by DFA, and products were sequenced and genotyped. Dogs were infected with Giardia intestinalis canid-associated assemblages C (nâ¯=â¯14), D (nâ¯=â¯13), and Cryptosporidium canis (nâ¯=â¯3). Similarly, G. intestinalis assemblages C (nâ¯=â¯1), D (nâ¯=â¯1) and C. canis (nâ¯=â¯1), were detected in coyotes, as well as G. intestinalis assemblage A (nâ¯=â¯1) and Cryptosporidium vole genotype (nâ¯=â¯1). Dogs and coyotes were predominantly infected with host-specific genotypes and few potentially zoonotic genotypes, suggesting that they may not represent a significant risk for zoonotic transmission of these parasites in urban areas where these hosts are sympatric.

OBJECTIVE: In this study, our objective was to compare direct microscopic examination and direct fluorescence antibody (DFA) method for Giardia diagnosis in stool samples and to evaluate the possible risk factors related to Giardia infections. METHODS: Stool samples of 185 patients with diarrhoea collected between June 2019 and July 2019 in Erzurum Yakutiye Research Hospital were included in the study. Microscopic examination of the samples was performed with native-lugol, and they were subsequently scanned by the indirect fluorescent assay microscope using the DFA method at 100-200X magnification. In addition, all patients filled a questionnaire prepared to determine the possible risk factors related to Giardia infection. RESULTS: The age of the 185 participating patients who belonged to different groups was between 0 and 94 years. Giardia spp. cysts were detected in five stool samples (2.7%) using direct microscopic examination. Nine samples (4.9%) were DFA-positive. The incidence of giardiasis was noted to be 7.5% in children, 3.8% in adults, 7.3% in people living in rural areas, 2.9% in people living in urban areas, 10% in people having pets and 4.2% in people who do not have pets. CONCLUSION: By taking the DFA method as a reference, the sensitivity and specificity of the microscopic examination were found to be 44.4% and 99.4%, respectively. The Giardia positivity rate was higher in children, those living in rural areas, those having pets and those using well water as drinking water.

BACKGROUND: Rattus norvegicus are reservoirs for transmission of various zoonotic parasites, and they have become a threat to public health worldwide. Given the large number and the significant presence of R. norvegicus throughout the city of Tehran, this study aims to assess the frequency of zoonotic parasites carried by commensal rodents wandering in Tehran, Iran. The study considered the north, south, west, east, and center regions of Tehran for the purposes of this study. The serological tests were applied in order to detect effective antibodies against Trichomonas vaginalis (T. vaginalis), Babesia spp., and Cryptosporidium spp. using a commercial qualitative rat ELISA kit. The frequency of Toxoplasma gondii (T. gondii) was surveyed by using the conventional PCR method. Furthermore, nested PCR was employed to detect the presence of Giardia spp. and Leishmania spp. in commensal R. norvegicus dispersed in Tehran. RESULTS: Approximately, 76% of the 100 R. norvegicus tested were infected with at least one zoonotic parasite, indicating the significant frequency of parasites within the study areas. Seroreactivity against T. vaginalis, Babesia spp., and Cryptosporidium spp. was detected in 5%, 0%, and 1% of the R. norvegicus tested, respectively. T. gondii DNA was detected in 32 out of 100 (32%) R. norvegicus. In addition, Leishmania spp. and Giardia spp. DNA were found in 18 out of 100 (18%) and 76 out of 100 (76%) R. norvegicus investigated, respectively. T. vaginalis with 15% and T. gondii with 70% had the highest frequency of parasites among the R. norvegicus collected from the western and northeastern regions of Tehran, respectively. Moreover, Giardia spp. with 95% and Leishmania spp. with 30% had the highest frequency in the east and center districts, respectively. CONCLUSION: The findings showed a wide geographical dissemination of Giardia spp., Toxoplasma gondii, and Leishmania spp. in R. norvegicus within five districts of Tehran. In contrast, other parasites such as Cryptosporidium spp. infection were rarely detected in Rattus populations. No evidence for the circulation of Babesia spp. was found in this study.
