RESUMO

Three endophytic fungi isolated from Moquiniastrum polymorphum (Less.) G. Sancho (Asteraceae) were cultivated using the one strain many compounds (OSMAC) strategy to evaluate the production of griseofulvin derivatives. Extracts obtained were analyzed by HPLC-MS/MS and the chromatographic and spectrometric data used to elaborate a feature-based molecular network (FBMN) through the GNPS platform. This approach allowed the observation of differences such as medium-specific and strain-specific production of griseofulvin derivatives and variations of cytotoxic activity in most extracts. To evaluate the efficiency of the OSMAC approach allied with FBMN analysis in the prospection of compounds of biotechnological interest, griseofulvin and 7-dechlorogriseofulvin were isolated, and the relative concentrations were estimated in all culture media using HPLC-UV, allowing for the inference of the best strain-medium combinations to maximize its production. Malt extract-peptone broth and Wickerham broth media produced the highest concentrations of both secondary metabolites.

ETHNOPHARMACOLOGICAL RELEVANCE: Gochnatia polymorpha ssp. floccosa (Asteraceae) also known as ''Cambará'' is used as medicinal plant in Brazil to treat infections and inflammation. Previous studies showed that its ethanolic extract could be bioprospecting of a new anti-inflammatory phytotherapy for use during pregnancy. This work aimed to evaluate dichloromethane (DCM) and butanolic (BT) fractions from G. polymorpha on embryo-fetal development and DNA integrity in mice. MATERIALS AND METHODS: Female mice were treated with 50 and 20mg/kg of the DCM and BT fractions, respectively, during organogenesis and gestational period. RESULTS AND CONCLUSION: The present study shows that DCM and BT fractions from G. polymorpha possess mutagenic activity but are not teratogenic. Based on the fact that the reproductive indices are similar in control and treated animals, we may infer that the mutagenic effect was in somatic cell, at least in part, because the reabsorption number and reabsorption rates did not change in DCM and BT exposed groups.

BACKGROUND AND AIM: This study aimed to evaluate the in vivo antitumor actions and toxicity of the dichloromethane fraction (F1B) of Moquiniastrum polymorphum subsp. floccosum (formerly Gochnatia polymorpha ssp. floccosa), composed of sesquiterpene lactones, against Walker-256 carcinosarcoma in rats. METHODS: Male Wistar rats received 100 mg kg(-1) F1B per day orally for 16 days after subcutaneous inoculation of Walker-256 cells in the pelvic limb. The tumor progression was monitored, and after treatment, tumor weight, oxidative stress, plasma biochemistry, inflammatory parameters, gene expression and histology of tumor and/or liver were evaluated. The toxicity of F1B was analyzed through the relative weight of organs. Additionally, an LD50 test was performed in mice. RESULTS: F1B treatment significantly reduced tumor volume and weight. There was no difference in oxidative stress in tumor tissue after treatment. F1B treatment modified hepatic glutathione and superoxide dismutase, and normalized plasma glucose, alkaline phosphatase, and amylase. F1B did not affect the activity of myeloperoxidase and N-acetylglucosaminidase or the nitric oxide levels in tumor tissue. However, F1B decreased the tumor necrosis factor (TNF)-α levels. Additionally, F1B increased apoptosis in the tumor, mediated by up-regulation of the p53 and Bax gene expression. No clinical signs of toxicity or death were observed in the rats treated with F1B. The LD50 calculated for mice was 1209 mg kg(-1). CONCLUSIONS: F1B, which is rich in sesquiterpene lactones, showed antitumor activity against Walker-256 carcinosarcoma. This effect may be, at least in part, related to the induction of apoptosis and inhibition of TNF-α signaling.

Abstract This study aimed to analyze the physical properties of the seeds of Moquiniastrum polymorphum when processed by mini-SAS, X-ray and seed blower systems. The seeds were purchased from Seeds Caiçara, located in Penápolis-SP. After purchase, the seed lot was characterized by standard tests. In the X-ray test the seeds were classified according to their internal morphology as filled, unfilled or malformed. The seed blower was set at six opening settings, resulting in light and heavy fractions. The evaluations were performed by weighing the seeds that remained in the blower (heavy fraction) and by the amount of filled seeds indicated by the X-ray test. The germination tests were composed of four replicates of 50 seeds. The mini-SAS was used for external evaluation. It was concluded that the combined use of the seed blower and the X-ray test was efficient for the evaluation and determination of the physical quality of the seeds of M. polymorphum. The seed blower at opening settings 5 resulted in the best lot quality. The external parameters were not able to qualitatively evaluate the seeds of M. polymorphum.


Resumo O presente estudo objetivou analisar as propriedades físicas das sementes de Moquiniastrum polymorphum por meio dos equipamentos raios-X, mini-SAS e soprador de sementes. As sementes foram adquiridas da empresa Sementes Caiçara em matrizes localizadas no município de Penápolis-SP. Após adquirido e beneficiado, o lote de sementes foi caracterizado por meio de testes padrões. No teste de raios-X as sementes foram classificadas de acordo com sua morfologia interna como cheias, vazias ou mal formadas. O soprador de sementes foi ajustado em seis aberturas, resultando em frações leves e pesadas. As avaliações foram realizadas pesando-se as sementes que permaneceram no soprador (fração pesada) e pela quantidade de sementes cheias pela análise de raios-X. Posteriormente foram realizados testes de germinação compostos por quatro repetições de 50 sementes. Na avaliação externa das sementes utilizou-se o equipamento mini-SAS. Após análise dos dados, concluiu-se que a utilização em conjunto dos equipamentos soprador de sementes e raios-X foi eficiente para avaliação e determinação da qualidade física das sementes de M. polymorphum. O soprador de sementes na regulagem da abertura 5 resultou na melhor qualidade do lote. Os parâmetros externos não foram capazes de avaliar qualitativamente as sementes de M. polymorphum.

Gochnatia polymorpha (Less.) Cabrera, Asteraceae, is popularly known as cambará and cambara-de-folha-grande in Brazil. It is used in traditional medicine to treat respiratory and gastrointestinal disorders. Pharmacological studies revealed anti-inflammatory, antispasmodic, antibacterial and antiviral activities. The goal of this paper was to carry out morphological and anatomical studies in order to describe the aerial parts of G. polymorpha. The botanical material was collected, fixed, and prepared according to usual light and scanning electron microtechniques. The leaves are simple, oblong-lanceolate to elliptical-lanceolate in form with mucronate acute apex, rounded base, entire or slightly toothed margin, and short petiole. In transection, the epidermis is uniseriate along the leaf blade. A subepidermal layer next to the adaxial side is present. Anomocytic stomata are seen only on the abaxial surface. Capitate glandular trichomes and T-shaped non-glandular trichomes occur on the leaves. The mesophyll is dorsiventral and minor collateral vascular bundles are enclosed by a sheath of thick walled parenchymatic cells. The midrib is biconvex and the petiole has a circular shape. The epidermis of the stem consists of a single layer of cells with glandular and nonglandular trichomes. The vascular cylinder shows typical structure and perivascular fiber caps are next to the phloem.

Phytochemical study of the flowers of Gochnatia polymorpha subsp. floccosa, Asteraceae, yielded eleven known triterpenes identified as lupeol, lupeyl acetate, lupeyl palmitate, taraxasterol, taraxasteryl acetate, pseudotaraxasterol, pseudotaraxasterol acetate, α-amyrin, α-amyryl palmitate, β-amyrin and β-amyryl palmitate, along with sitosterol, stigmasterol, palmitic and stearic acids. These compounds are been reported for the first time in the species. The compounds were identified by analysis of NMR spectra (¹H, 13C and DEPT), GC-MS and comparison with literature data. Previous work have reported the isolation of triterpenes, diterpenes, sesquiterpenes, flavonoids, coumarins and phenolic compounds from aerial parts and roots from G. polymorpha.
