RESUMO

The limited availability of phosphate, nitrogen and silicon in the growth media affects the growth, cellular processes, and metabolism of diatoms. Silicon deficiency primarily affects diatom morphology, while phosphate deficiency reduces the production of nucleic acids and phospholipids. Differences in pigment and protein composition are mainly due to nitrogen deficiency. In this study, Chaetoceros socialis and Chaetoceros costatus were cultured under phosphate, nitrogen, and silicon deprivation conditions. The diatom biomass was collected during the stationary growth phase and extracted with 70% ethanol under ultrasonication. The chemical profiles of the extracts were analyzed by high-performance liquid chromatography with high-resolution mass spectrometry with electrospray ionisation (UHPLC-ESI-HRMS), while the antioxidant capacity was determined by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and oxygen radical absorbance capacity (ORAC) assays. Pigments, fatty acids, sterols, and derivatives were detected in both species. The total phenolic content in the extracts ranged from 46.25 ± 1.08 to 89.38 ± 6.21 mg of gallic acid equivalent (GAE)/L and from 29.58 ± 1.08 to 54.17 ± 1.18 mg GAE/L. for C. costatus and C. socialis, respectively. Antioxidant activity was higher in C. costatus extracts, especially those obtained from nitrogen-deprived media. The results of this study contribute to the existing knowledge and the ongoing efforts to overcome application and commercialization barriers of microalgae for wide-ranging potential in different industries.

Even when fresh, non-alcoholic, and low-alcoholic beers (NABLABs) exhibit significant staling defects due to premature oxidation. In this study, the antioxidant power of eleven fresh commercial NABLABs was assessed by means of three different assays: the oxygen radical absorbance capacity (ORAC), the linoleic acid-induced oxidation (TINH), and the indicator time test (ITT). Only the first two assays, both involving radicalar degradations initiated by AAPH, were found to correlate with each other. NABLABs displayed lower ORAC values than conventional beers (on average, 6127 µmol eq. Trolox/L), except for three samples made with special-colored malts or dry-hopped. Dealcoholization was the step with the greatest impact on the ORAC value (up to a 95% loss) and on flavan-3-ols, sotolon, and polyfunctional thiols, while pasteurization strongly affected color, TBA, and Strecker aldehydes. ORAC assays applied to hop, alternative cereals, and various botanical ingredients indicated that mashing with red sorghum, dry hopping/spicing, and wood maturation could bring the antioxidant power of a NABLAB close to those of conventional beers. With an ORAC value not reached by any other tested botanical ingredient (5234 µmol eq. Trolox/g), African Vernonia amygdalina leaves (traditionally used for Rwandan Ikigage beers) emerged here as the best candidate.

The study aimed to optimize ultrasonic (US: 40 kHz/200 W for 10, 20, 30, 40, and 50 min), and microwave (MW: 160 W for 45, 90, 125, 180, and 225 s) pretreatment conditions on protein extraction yield and degree of protein hydrolysis (DH) from almond de-oiled meal, an industrial by-product. First order model was used to describe the kinetics of almond protein hydrolysates obtained with Alcalase. The highest DH, 10.95% was recorded for the US-50 min and 8.87% for MW-45 s; while it was 5.76% for the untreated/control sample. At these optimized pretreatment conditions, a 1.16- and 1.18-fold increment in protein recovery was observed for the US and MW pretreatments, respectively in comparison to the conventional alkaline extraction. The molecular weight distribution recorded for pretreated samples disclosed a significant reduction in the band thickness in comparison with control. Both the pretreatments resulted in a significant increase (P < 0.05) in the antioxidant activity, and TCA solubility index when compared with the control. Results evinced that US and/or MW pretreatments before enzymatic hydrolysis can be a promising approach for the valorization of almond meal for its subsequent use as an ingredient for functional foods/nutraceuticals which otherwise fetches low value as an animal feed.

Jeriva (Syagrus romanzoffiana) is a fruit from palm trees of the Arecaceae family, widely distributed in tropical and subtropical areas of Latin America. It has low production costs and high productivity throughout the year; however, its consumption is very low, and the production goes almost entirely to feed animals or to waste. To improve its consumption, a good characterization of the whole fruit is necessary. The objective of this work was to evaluate the jeriva pulp, peel and seeds according to carotenoids, phenolic compounds, vitamin C, tocopherols and antioxidant potential using HPLC, microplate readers and spectrophotometric methods. Every part of the fruit exhibited antioxidant capacity in the ORAC and TEAC tests, which can be attributed to its high concentration of polyphenols. Carotenoids were more present in the pulp and peel and almost absent in the seeds. Vitamin C ranged from 12 ± 1 for the seeds up to 92 ± 3 mg/100 g for the pulp. The total phenolic content was quantified between 473 ± 39 for the seeds and 1089 ± 32 mg of gallic acid equivalents (GAEs)/100 g for the pulp. These results demonstrate that all parts of this fruit have important bioactive nutrients, with promising perspectives for further scientific approaches and for composing formulations of food products to enhance functional properties.

Pretreatment of grape pomace seeds with a pulsed electric field (PEF) was applied to improve the extraction yield of cold-pressed grape seed oil. The effects of different PEF conditions, electric field intensities (12.5, 14.0 and 15.6 kV/cm), and durations (15 and 30 min) on the oil chemical composition were also studied. All PEF pretreatments significantly increased the oil yield, flow rate and concentration of total sterols (p < 0.05). In addition, similar trends were observed for total tocochromanols and phenolic compounds, except for PEF pretreatment under the mildest conditions (12.5 kV/cm, 15 min) (p < 0.05). Notably, the application of 15.6 kV/cm for 30 min resulted in the highest relative increase in oil yield and flow rate (29.6% and 56.5%, respectively) and in the concentrations of total tocochromanols, nonflavonoids, and flavonoids (22.1%, 60.2% and 81.5%, respectively). In addition, the highest relative increase in the concentration of total sterols (25.4%) was achieved by applying 12.5 kV/cm for 30 min. The fatty acid composition of the grape seed oil remained largely unaffected by the PEF pretreatments. These results show that PEF pretreatment effectively improves both the yield and the bioactive properties of cold-pressed grape seed oil.

Atypical aging (ATA) is a well-known wine defect that leads to unacceptable off-odors of wet mop, soap, waxy, furniture varnish, and dish rag. The main compound that is responsible for these undesired scents, 2-aminoacetophenone (AAP), results from the oxidative degradation of 3-indole acetic acid. This study aimed to investigate different technological factors for a possible effect on the oxygen radical absorption capacity (ORAC) and ATA development in white wine. Oxygenation during fermentation did not result in a significant effect on the development of ORAC or ATA development. Instead, the addition of ascorbic acid after vinification led to increased ORAC and decreased AAP formation. The first evidence of a tendency toward a correlation between ORAC and potential ATA development was found, with r = 0.365. This study showed the potential of the ORAC assay for an early assessment of the possible formation of ATA during wine storage.

Extracts from Veronica species (speedwells) are known for the various biological activities they show, such as cytotoxic, antimicrobial, anti-inflammatory, and antioxidant activities. Also, the plants from this genus are known as medicinal plants used in traditional medicine worldwide. Phenolic compounds are specialized metabolites that contribute to biological activity the most. Therefore, the aim of this research is identification and quantification of phenolic compounds present in three Veronica species (Veronica anagallis-aquatica L., Veronica persica Poir., and Veronica polita Fr.) using the liquid chromatography-mass spectrometry (LC-MS/MS) technique. All extracts were tested for antioxidant activity with two methods: DPPH (2,2-diphenyl-1-picrylhydrazyl) and ORAC (oxygen radical absorbance capacity). Also, standards for compounds that were detected in the highest amount in all species were also tested for antioxidant activity. Three different solvents (pure methanol, 80% ethanol, and water) were used for the extraction of phenolic components and their comparison in order to test their antioxidant activity as a final goal. The main compounds present in the tested Veronica extracts were: p-hydroxybenzoic acid, vanillic acid, caffeic acid, gentisic acid, and apigenin. V. anagallis-aquatica contained the highest amount of phenolic components in comparison with the two other tested species, V. persica and V. polita. Caffeic acid showed the highest antioxidant activity in both studied methods with an IC50 value for DPPH activity of 1.99 µg/mL. For the plant extracts, in general, methanolic/ethanolic extracts showed higher activity than water extracts in both methods which was expected, as organic solutions extract more phenolic compounds. This research points to the potential application of extracts of different Veronica species for antioxidant activity.

This work relates to the design and synthesis of a series of novel multi-target directed ligands (MTDLs), i.e., compounds 4a-l, via a convenient one-pot three-component Hantzsch reaction. This approach targeted calcium channel antagonism, antioxidant capacity, cathepsin S inhibition, and interference with Nrf2 transcriptional activation. Of these MTDLs, 4i emerged as a promising compound, demonstrating robust antioxidant activity, the ability to activate Nrf2-ARE pathways, as well as calcium channel blockade and cathepsin S inhibition. Dihydropyridine 4i represents the first example of an MTDL that combines these biological activities.

RESUMO

Endophytic fungi have emerged as a significant source of natural products with remarkable bioactivities. Recent research has identified numerous antioxidant molecules among the secondary metabolites of endophytic fungi. These organisms, whether unicellular or micro-multicellular, offer the potential for genetic manipulation to enhance the production of these valuable antioxidant compounds, which hold promise for promoting health, vitality, and various biotechnological applications. In this study, we provide a critical review of methods for extracting, purifying, characterizing, and estimating the total antioxidant capacity (TAC) of endophytic fungi metabolites. While many endophytes produce metabolites similar to those found in plants with established symbiotic associations, we also highlight the existence of novel metabolites with potential scientific interest. Additionally, we discuss how advancements in nanotechnology have opened new avenues for exploring nanoformulations of endophytic metabolites in future studies, offering opportunities for diverse biological and industrial applications.

The ORAC (Oxygen Radical Absorbance Capacity) assay is commonly employed for determining the antioxidant capacity of bioactive peptides. To gain insights into the meaning of this index for peptides containing a single Trp, we studied the consumption of this residue and fluorescein (FLH, the probe of ORAC method), induced by radicals generated by AAPH (2,2'-Azo-bis(2-amidinopropane) dihydrochloride) thermolysis. ORAC values were rationalized from kinetics and computational calculations of bond dissociation energies (BDE) of the N-H bond (indole ring of Trp). Free Trp, di- and tri- peptides, and three larger peptides were studied. Solutions containing 70 nM FLH, 1-5 µM free Trp or peptides, and 10 mM AAPH were incubated at 37 °C in phosphate buffer. Kinetic studies showed that FLH minimally affected Trp consumption. However, a clear protection of FLH, characterized by pseudo-lag times, was evidenced, reflecting radical-radical reactions and FLH repairing. Peptides showed similar ORAC values (~1.9-2.8 Trolox equivalents), while BDE varied between 91.9 and 103.5 kcal. These results, added to the protection of FLH observed after total consumption of Trp, indicate a lack of discrimination of the assay for the chemical structure of peptides and the contribution of oxidation products to the index.

Acetylcholinesterase (AChE), hydrolyzes acetylcholine to choline and acetate, thereby terminating this neurotransmitter effect at cholinergic synapses. Therefore, AChE inhibition is used for counterbalance the cholinergic deficit in Alzheimer's disease (AD) patients. In the present work, in order to find new plant acetylcholinesterase inhibitors, the hydroalcoholic extracts from seventeen medicinal plant species were screened for their acetylcholinesterase inhibition activity, as well as total phenolic (TPC) and flavonoids contents (TFC) and antioxidant activity using ORAC (Oxygen Radical Absorbance Capacity) assay, and their ability to inhibit lipid peroxidation. The results revealed that Rumex acetosa, Taraxacum officinale and Hypericum perforatum extracts possessing the highest TPC and TFC, were the most effective in terms of ORAC antioxidant activity, and acetylcholinesterase inhibition, in addition to their ability to inhibit liposomes peroxidation, suggesting that those plant species may provide a substantial source of secondary metabolites, which act as natural antioxidants and acetylcholinesterase inhibitors, and may be beneficial in the treatment of AD.


La acetilcolinesterasa (AChE) hidroliza la acetilcolina se hidroliza en colina y acetato, terminando así este efecto neurotransmisor en las sinapsis colinérgicas. Por lo tanto, la inhibición de la AChE se utiliza para contrarrestar el déficit colinérgico en pacientes con enfermedad de Alzheimer (EA). En el presente trabajo, con el fin de encontrar nuevos inhibidores de la acetilcolinesterasa vegetal, se analizaron los extractos hidroalcohólicos de diecisiete especies de plantas medicinales para determinar su actividad inhibidora de la acetilcolinesterasa, así como el contenido total de fenólicos (TPC) y flavonoides (TFC) y la actividad antioxidante utilizando ORAC (Capacidad de absorbancia de radicales de oxígeno) y su capacidad para inhibir la peroxidación de lípidos. Los resultados revelaron que los extractos de Rumexacetosa, Taraxacum officinale e Hypericum perforatum que poseen los más altos TPC y TFC, fueron los más efectivos en términos de actividad antioxidante ORAC e inhibición de acetilcolinesterasa, además de su capacidad para inhibir la peroxidación de los liposomas, sugiriendo que esas especies de plantas puede proporcionar una fuente sustancial de metabolitos secundarios, que actúan como antioxidantes naturales e inhibidores de la acetilcolinesterasa, y puede ser beneficioso en el tratamiento de la EA.

RESUMEN Introducción: La especie Spondias mombin L. es una planta que crece en Cuba y a la que se le reporta un número considerables de aplicaciones etnofarmacológicas y elevado poder antioxidante. Sin embargo, la mayor parte de estos reportes está orientada al fruto, el cual resalta por su valor nutricional. Objetivo: Evaluar la actividad antioxidante de un extracto de hojas de Spondias mombin L. en etanol al 70 %. Método: La actividad antioxidante fue evaluada por seis métodos diferentes: determinación del índice de oxidación, poder reductor, inhibición de los radicales DPPH y ABTS, capacidad reductora del peróxido de hidrógeno y capacidad de neutralización del radical oxígeno. Se evaluaron concentraciones desde los 1 000 hasta los 62,5 µg/mL preparadas a partir del valor de sólidos totales determinados. Resultados: Para el índice de oxidación, concentraciones de 250 µg/mL resultaron suficientes para decolorar la solución de permanganato en igual período de tiempo que el patrón de quercetina. Sin embargo, los IC50 calculados para el poder reductor, la inhibición del radical DPPH y del ABTS resultaron mayores que el estándar de ácido ascórbico con valores de 362,33; 190,34 y 241,88 µg/mL, respectivamente. Conclusiones: El extracto de hojas de Spondias mombin L. en etanol al 70 % presenta una elevada acción antioxidante en los seis modelos experimentales ensayados, aun cuando en la mayor parte de casos, con valores inferiores al de la sustancia de referencia empleada. Estos resultados conjuntamente con la diversidad estructural de metabolitos presentes en el mismo resaltan su valor como potencial antioxidante natural.


ABSTRACT Introduction: The species Spondias mombin L. is a plant that grows in Cuba, and to which a considerable number of ethnopharmacological applications and high antioxidant power are reported. However, most of these reports are oriented to the fruit, which stands out for its nutritional value. Objective: To evaluate the antioxidant activity of an extract of Spondias mombin L. leaves in 70% ethanol. Method: The antioxidant activity was evaluated by six different methods: determination of the oxidation index, reducing power, inhibition of DPPH and ABTS radicals, hydrogen peroxide reducing capacity and oxygen radical neutralization capacity. Concentrations from 1000 to 62.5 µg/mL were evaluated, prepared from the value of total solids determined. Results: For the oxidation index, concentrations of 250 µg/mL were sufficient to decolorize the permanganate solution in the same period of time as the quercetin standard. However, the IC50 calculated for the reducing power, the inhibition of the DPPH radical and ABTS were higher than the ascorbic acid standard with values of 362.33; 190.34 and 241.88 µg/mL, respectively. Conclusions: The extract of leaves of Spondias mombin L. in 70% ethanol presents a high antioxidant action in the six experimental models tested, even though in most cases, with lower values than the reference substance used. These results, together with the structural diversity of metabolites present in it, highlight its value as a potential natural antioxidant.

No entanto, a maioria desses relatos é voltada para o fruto, que se destaca pelo seu valor nutricional. Objetivo: Avaliar a atividade antioxidante de um extrato de folhas de Spondias mombin L. em etanol 70%. Método: A atividade antioxidante foi avaliada por seis diferentes métodos: determinação do índice de oxidação, poder redutor, inibição dos radicais DPPH e ABTS, capacidade redutora do peróxido de hidrogênio e capacidade de neutralização do radical oxigênio. Foram avaliadas concentrações de 1000 a 62,5 µg/mL, preparadas a partir do valor de sólidos totais determinado. Resultados: Para o índice de oxidação, concentrações de 250 µg/mL foram suficientes para descolorir a solução de permanganato no mesmo período de tempo do padrão de quercetina. No entanto, o IC50 calculado para o poder redutor, a inibição do radical DPPH e ABTS foram superiores ao padrão de ácido ascórbico com valores de 362,33; 190,34 e 241,88 µg/mL, respectivamente. Conclusões: O extrato de folhas de Spondias mombin L. em etanol 70% apresenta alta ação antioxidante nos seis modelos experimentais testados, embora na maioria dos casos com valores inferiores à substância de referência utilizada. Esses resultados, juntamente com a diversidade estrutural de metabólitos presentes nele, destacam seu valor como potencial antioxidante natural.

Background: antioxidants can eliminate excess free radicals present in our bodies, and it has been shown that eating a diet rich in polyphenols and anthocyanins, present in vegetables, fruits and legumes, has a protective effect against cancer, heart disease, and some neurological diseases. Objectives: the objective of the present work was to evaluate the eating habits of the university community in Murcia. Methods: the research was carried out by means of surveys on the choice of dishes at university canteens. In addition, the antioxidant activity of the most widely consumed dishes (pork Milanese style, pasta Bolognese, lentil and chorizo stew, Spanish omelette, and vegetable paella) were evaluated using the DPPH, FRAP, and ORAC methods. Results: the results obtained indicated that canteen users preferred consumption of pasta, meat, and fried potatoes rather than vegetables, pulses, and fish as main courses. The most consumed dessert was yogurt, closely followed by fruits. The consumption of water also stands out, and a majority of subjects accompanied their meals with white bread and salad. With regard to antioxidant capacity, it was found that the dish with the highest antioxidant capacity was lentil and chorizo stew, with Spanish omelette being the less antioxidant dish. Conclusion: in conclusion, consumption of fruit, vegetables, and legumes should be encouraged, as they contribute decisively to the antioxidant capacity of the diet. (AU)


Introducción: los antioxidantes pueden eliminar el exceso de radicales libres presentes en nuestro organismo, y se ha demostrado que consumir una dieta rica en polifenoles y antocianinas, presentes en verduras, frutas y legumbres, tiene un efecto protector frente al cáncer, las enfermedades cardíacas y algunas enfermedades neurológicas. Objetivos: el objetivo del presente trabajo fue evaluar los hábitos alimentarios de la comunidad universitaria de Murcia. Métodos: la investigación se llevó a cabo mediante encuestas sobre la elección de platos en los comedores universitarios. Además, se evaluó la actividad antioxidante de los platos más consumidos (cerdo a la milanesa, pasta boloñesa, lentejas con chorizo, tortilla española y paella de verduras) a través de los métodos DPPH, FRAP y ORAC. Resultados: los resultados obtenidos indicaron que los usuarios del comedor prefirieron el consumo de pasta, carne y patatas fritas en lugar del de verduras, legumbres y pescado. El postre más consumido fue el yogur, seguido de cerca por la fruta. También destaca el consumo de agua, y la mayoría acompañan sus comidas con pan blanco y ensalada. En cuanto a la capacidad antioxidante, se encontró que el plato con mayor capacidad antioxidante fueron las lentejas con chorizo, siendo la tortilla española el plato menos antioxidante. Conclusión: en conclusión, conviene fomentar el consumo de frutas, verduras y legumbres, ya que contribuyen de forma decisiva a la capacidad antioxidante de la dieta. (AU)

Background: Mango (Mangifera indica L.) is one of the world's most consumed fruit, and it is also a rich source of antioxidants that may prevent oxidative stress. Objectives: This study aimed to determine if mango (cv. Azúcar) juice can improve the antioxidant status of healthy individuals with low consumption of vegetables and fruit. Methods: This was a cross-over single-blind study carried out with 16 healthy individuals for 73 days. Participants were randomly assigned to either a mango juice period or a placebo period. Total phenolic content, antioxidant capacity, mangiferin, thiobarbituric acid reactive substances (TBARS), total glutathione, and 8 hydroxydeoxyguanosine levels were determined in plasma. Results: Plasma antioxidant activity was significantly higher in the juice consumption period than the placebo consumption period; however, total phenolic content, total glutathione, TBARS, and 8-hydroxydeoxyguanosine levels did not show significant differences between juice period and placebo period. Mangiferin was detected in every participant after juice consumption. Conclusions: Mango (cv. Azúcar) juice daily consumption improves plasma antioxidant capacity.


Antecedentes: El mango (Mangifera indica L.) es una de las frutas más consumidas en el mundo y también es una fuente rica en antioxidantes los cuales pueden prevenir el estrés oxidativo. Objetivos: El objetivo de este estudio fue determinar si el mango (c.v Azúcar) puede mejorar el estado antioxidante de individuos sanos con un bajo consumo de frutas y vegetales. Métodos: Se llevó a cabo un estudio cruzado, simple-ciego en 16 individuos sanos durante 73 días. Los participantes fueron asignados aleatoriamente al período del consumo del jugo o del placebo. Se determinó el contenido fenólico total, la capacidad antioxidante y los niveles de sustancias reactivas al ácido tiobarbiturico (TBARS), mangiferina, glutatión total y 8-hidroxi-guanosina, en el plasma obtenido de los participantes. Resultados: La capacidad antioxidante en plasma fue mayor en el período del consumo del jugo en comparación con el período del consumo del placebo; sin embargo, el contenido fenólico total, y los niveles de glutation total, 8-hidroxideoxiguanosina y TBARS no mostraron diferencias significativas entre el período del jugo y el período del placebo. La mangiferina se detectó en todos los individuos después del consumo del jugo. Conclusiones: El consumo diario de jugo de mango variedad Azúcar mejora la capacidad antioxidante en plasma.

ABSTRACT Edible films are an alternative to synthetic materials used for packaging foods. The aim of this study was to develop and characterize films made from cassava starch, freeze-dried acai and glycerol. The films were characterized for thickness, water vapor permeability, water solubility, anthocyanins, antioxidant capacity and mechanical properties. The results indicated a rich edible film in anthocyanins with considerable antioxidant capacity (150.70 micromol Trolox), which can extend the commercial validity of the packaged food and meets the growing demand for biodegradable packaging that tends to reduce the environmental impact. The addition results in a significant effect on the polymer matrix reducing the water solubility and water activity. The elongation test at rupture reported to be 17.738%, indicating that addition of pulp increased film flexibility.

In the body, the reactions by which energy is created produce reactive oxygen molecules (ROS). ROS can damage biomolecules such as lipids, proteins, carbohydrates, proteins and DNA, which has been linked to chronic diseases such as cancer, cardiovascular diseases, inflammatory processes, diabetes and aging. Antioxidants present in foods can counteract the harmful effect of ROS. The aim of this study was to quantify the average intake of antioxidant capacity in the Spanish diet. For this purpose, average daily food intake published in the National Survey of Spanish Dietetics in 2011 and the database that has been published by the USDA (based on the ORAC) was used. The Spanish population surpassed the estimated daily recommendation of antioxidants and was superior to other countries that have similar diets.


En el organismo, las reacciones mediante las cuales se obtienen la energía producen moléculas reactivas de oxígeno (ROS). Los ROS pueden dañar biomoléculas como los lípidos, proteínas, hidratos de carbono, proteínas y el ADN; y esto han sido relacionados con enfermedades crónicas como el cáncer, enfermedades cardiovasculares, procesos inflamatorios, diabetes y el envejecimiento Los antioxidantes presentes en los alimentos pueden contrarrestar el efecto nocivo de los ROS. El objetivo de este estudio fue cuantificar mediante la ingesta media de los españoles de índice de capacidad antioxidante. Para ello se empleó la ingesta media de alimentos por día de los españoles publicada en la Encuesta Nacional de Dietética Española del año 2011 y la base de datos que ha publicado la USDA (basada en el ORAC). La población española superó la recomendación diaria estimada de antioxidantes, siendo incluso superior a otros países que siguen dietas muy similares.

In order to prepare antioxidant peptide through hydrolyzing low-value protein resources with bacterial extracellular proteases and to discover novel proteases, crude extracellular protease from Pseudoalteromonas sp. SHK1-2 was obtained through fermentation which was used to hydrolyze collagen extracted from Cirrhinus molitorella skin. Small peptide fraction was isolated from hydrolysate by ultrafiltration and Sephadex LH-20 size exclusion chromatography and showed 1, 1-diphenyl-2-picrylhydrazyl radical scavenging activity (35.6%±7%), oxygen radical absorbance capacity and inhibition of DNA oxidation damage. The molecule weight was 776.2 Da, and amino acid sequence was Thr-Ala-Gly-His-Pro- Gly-Thr-His through liquid chromatography mass spectrum. Our findings suggest that peptide obtained from low-value protein of fish waste by hydrolysis with bacterial protease has antioxidant activity.

Phytochemical investigation of the aerial components of Ducrosia ismaelis Asch. led to the isolation of six known compounds, psoralen (1), isopsoralen (2), cnidioside A (3), (-)-syringaresinol-O-beta-D-glucopyranoside (4), (E)-plicatin B (5), trilinolein (6). The chemical structures of these compounds were elucidated from spectroscopic data and by comparison of these data with previously published results. The antioxidant, anti-osteoporotic and cardiovascular related activities of the isolated compounds were assessed using oxygen radical absorbance capacity (ORAC), reducing capacity, tartrate-resistant acid phosphatase (TRAP), and soluble epoxide hydrolase (sEH) inhibitory activity assays. Compounds (3-5) showed potent peroxyl radical-scavenging capacities with ORAC values of 11.06 +/- 0.39, 7.98 +/- 0.10, and 13.99 +/- 0.06 Trolox equivalent (TE) at concentrations of 10 microM, respectively. Only compounds 4 and 5 was able to significantly reduce Cu2+ ions, with a reduction value of 9.06 +/- 0.32 and 4.61 +/- 0.00 microM Trolox Equivalent (TE) at a concentration of 10 microM. Compound 5 at 10 microM exhibited a potent inhibitory effect on osteoclastic TRAP activity with a TRAP value of 86.05 +/- 6.55% of the control. Compounds 1, 3 and 5 potently inhibited sEH activity with IC50 values of 41.6 4.9, 16.0 1.1, and 49.0 5.7 microM, respectively.

Antecedentes: La mora contiene compuestos fenólicos que contribuyen a su capacidad antioxidante. El método más común para aislar estas sustancias es la extracción con solventes, que puede asistirse con ultrasonido para aumentar el contenido de analitos extraídos. Objetivos: Determinar las condiciones de tipo y concentración de solvente en agua, y de tiempo de extracción asistida con ultrasonido, con las cuales se obtiene el mayor contenido de compuestos fenólicos y el valor de capacidad antioxidante más alto en extractos de mora andina (Rubus glaucus Benth). Comparar la capacidad antioxidante de los extractos de mora con la de antioxidantes de referencia como el di-terc-butilhidroxitolueno (BHT) y el a-tocoferol. Métodos: Se hicieron tres diseños de experimentos de dos factores con tres niveles cada uno. Las variables fueron la concentración de acetona, etanol o metanol en agua y el tiempo de extracción asistida con ultrasonido. Los observables-respuesta fueron el contenido total de compuestos fenólicos, que se determinó utilizando el reactivo de Folin-Ciocalteu, y la capacidad antioxidante, determinada mediante el ensayo ORAC (capacidad de absorción de radicales de oxígeno). Se calculó el coeficiente de correlación entre las dos variables de respuesta. Se comparó la capacidad antioxidante de los extractos de mora, obtenidos bajo las condiciones inferidas como óptimas, con las del BHT y del a-tocoferol. Resultados: Los extractos adquiridos con soluciones acuosas de acetona al 31% (v/v), asistiendo la extracción con ultrasonido durante 100 min, mostraron los mayores contenidos de compuestos fenólicos (30 ± 1 mg ácido gálico/g mora seca), y los valores de capacidad antioxidante más altos (273 ± 6 mmol Trolox®/g mora seca). El coeficiente de correlación entre las dos variables de respuesta fue 0,92. Los extractos de mora presentaron mejor capacidad antioxidante que el BHT y el a-tocoferol. Conclusiones: Bajo las condiciones óptimas de extracción, con las soluciones acuosas de acetona se obtuvieron el mayor contenido de compuestos fenólicos y el valor de capacidad antioxidante más alto. Se observó una relación lineal entre las dos variables de respuesta. Los extractos de mora, obtenidos bajo las mejores condiciones halladas, presentaron mayor capacidad antioxidante en comparación con la de los compuestos de referencia analizados.

This study compared the regular and decaffeinated coffees in relation to antioxidant capacity, levels of some antioxidant molecules and stability of these parameters over a six-month period under different storage conditions. The regular coffee samples analyzed right after the industrial production showed higher antioxidant capacity (ORAC and DPPH), the same levels of phenolic compounds and higher levels of phenolic acids than decaffeinated coffee. After six months, the closed packs of both the grades kept under vacuum at 20°C did not show significant changes from the initial results; the open packs stored at 4°C showed small but statistically significant reductions and the open packs stored at 20°C showed higher and statistically significant losses (p < 0.05). Oxygen was the most important factor for these losses but temperature also played an important role. Tests showed that the storage conditions were very important to preserve the quality of coffees regarding the analyzed parameters.
