RESUMO

Heading date is one of the important agronomic traits that affects rice yield. In this study, we cloned a new rice B3 family gene, OsL1, which regulates rice heading date. Importantly, osl1-1 and osl1-2, two different types of mutants of OsL1 were created using the gene editing technology CRISPR/Cas9 system and exhibited 4 days earlier heading date than that of the wild type under short-day conditions. Subsequently, the plants overexpressing OsL1, OE-OsL1, showed a 2-day later heading date than the wild type in Changsha and a 5-day later heading date in Lingshui, but there was no significant difference in other yield traits. Moreover, the results of subcellular localization study indicated that OsL1 protein was located in the nucleus and the expression pattern analysis showed that OsL1 gene was expressed in rice roots, stems, leaves, and panicles, and the expression level was higher at the root and weak green panicle. In addition, the OsL1 gene was mainly expressed at night time under short-light conditions. The transcriptomic analysis indicated that OsL1 might be involved in the Hd1-Hd3a pathway function. Together, our results revealed that the cloning and functional analysis of OsL1 can provide new strategy for molecular design breeding of rice with suitable fertility period. The ultimate goal of this work is the study of the effect of luminescence stimulations and signals reading modes combinations on the thermoluminescence intensity and glow curve behaviour for the same X-ray irradiation dose. Three interesting stimulating and reading modes are considered, namely, infrared stimulated luminescence (IRSL), blue light-emitting diode stimulated luminescence (BLSL) and thermally stimulated luminescence (TSL). The studied stimulation and reading modes combination protocols are (Protocol 1) IRSL-TSL, (Protocol 2) IRSL-BLSL-TSL and (Protocol 3) BLSL-IRSL-TSL. Experiments are performed on beryllium oxide (BeO) dosimeter. Results demonstrate well that the combination of reading modes have direct impact on the TL signal in terms of intensity and glow curve shape. It was also found that when reading modes are correctly combined, particularly when IRSL is applied first, then BLSL and TL, it is possible to collect two or more exploitable signals of different stimulation types for the same irradiation that can be used for different purposes and final applications.

Chuandong Cave is an important Late Paleolithic site because it documents the early appearance of bone tools in southern China. We used the single-aliquot regenerative-dose protocol for optically stimulated luminescence dating to improve the precision of the chronology for the Chuandong Cave sedimentary sequence. The age of each layer was determined using a Bayesian modeling approach which combined optically stimulated luminescence ages with published AMS 14C dates. The results showed that Layer 10 began accumulating since 56 ± 14 ka and provides the upper age limit for all artifacts from the sequence. Bone awl tools from Layer 8, the earliest grinding bone tools in this site, were recovered within sediments between 40 ± 7 ka and 30 ± 4 ka. Layer 8 also indicates the appearance of modern humans in the Chuandong Cave sequence. Layers 4-2, ranging from 15 ± 3 ka until 11 ± 1 ka and including the Younger Dryas period, contain a few bone awls and an eyed bone needle. The shift from bone awls to eyed bone needles in the Chuandong Cave sequence indicates that modern humans adapted to the changing climate of southern China. We conclude that modern human behavior in bone tools appeared in southern China as early as 40 ± 7 ka, became more sophisticated during the Last Glacial Maximum, and spread more widely across southern China during the Younger Dryas.

Renewed research at Amanzi Springs has increased resolution on the timing and technology of the Acheulian industry in South Africa. The archeology from the Area 1 spring eye has recently been dated to MIS 11 (∼404-390 ka), and analyses revealed significant technological variability when compared to other southern African Acheulian assemblages. We expand on these results in presenting new luminescence dating and technological analyses of Acheulian stone tools from three artifact-bearing surfaces exposed within the White Sands unit of the Deep Sounding excavation in the Area 2 spring eye. The two lowest surfaces (Surfaces 3 and 2) are sealed within the White Sands and dated between ∼534 to 496 ka and ∼496 to 481 ka (MIS 13), respectively. Surface 1 represents materials deflated onto an erosional surface that cut the upper part of the White Sands (∼481 ka; late MIS 13), which occurred before the deposition of younger Cutting 5 sediments (<408-<290 ka; MIS 11-8). Archaeological comparisons reveal that the older Surface 3 and 2 assemblages are predominated by unifacial and bifacial core reduction and relatively thick, cobble-reduced large cutting tools. In contrast, the younger Surface 1 assemblage is characterized by discoidal core reduction and thinner large cutting tools, mostly made from flake blanks. Typological similarities between the older Area 2 White Sands and younger Area 1 (404-390 ka; MIS 11) assemblages further suggest long-term continuity in site function. We hypothesize Amanzi Springs represent a workshop locality that Acheulian hominins repeatedly visited to access unique floral, faunal, and raw material resources from at least ∼534 to 390 ka.

Soil mixing over long (>102 y) timescales enhances nutrient fluxes that support soil ecology, contributes to dispersion of sediment and contaminated material, and modulates fluxes of carbon through Earth's largest terrestrial carbon reservoir. Despite its foundational importance, we lack robust understanding of the rates and patterns of soil mixing, largely due to a lack of long-timescale data. Here we demonstrate that luminescence, a light-sensitive property of minerals used for geologic dating, can be used as a long-timescale sediment tracer in soils to reveal the structure of soil mixing. We develop a probabilistic model of transport and mixing of tracer particles and associated luminescence in soils and compare with a global compilation of luminescence versus depth in various locations. The model-data comparison reveals that soil mixing rate varies over the soil depth, with this depth dependency persisting across climate and ecological zones. The depth dependency is consistent with a model in which mixing intensity decreases linearly or exponentially with depth, although our data do not resolve between these cases. Our findings support the long-suspected idea that depth-dependent mixing is a spatially and temporally persistent feature of soils. Evidence for a climate control on the patterns and intensities of soil mixing with depth remains elusive and requires the further study of soil mixing processes.

The Aral Sea basin in Central Asia and its major rivers, the Amu Darya and Syr Darya, were the center of advanced river civilizations, and a principal hub of the Silk Roads over a period of more than 2,000 y. The region's decline has been traditionally attributed to the devastating Mongol invasion of the early-13th century CE. However, the role of changing hydroclimatic conditions on the development of these culturally influential potamic societies has not been the subject of modern geoarchaeological investigations. In this paper we report the findings of an interdisciplinary investigation of archaeological sites and associated irrigation canals of the Otrar oasis, a United Nations Educational, Scientific and Cultural Organization (UNESCO) World Heritage site located at the confluence of the Syr Darya and Arys rivers in southern Kazakhstan. This includes radiometric dating of irrigation canal abandonment and an investigation of Arys river channel dynamics. Major phases of fluvial aggradation, between the seventh and early ninth century CE and between 1350 and 1550 CE coincide with economic flourishing of the oasis, facilitated by wet climatic conditions and higher river flows that favored floodwater farming. Periods of abandonment of the irrigation network and cultural decline primarily correlate with fluvial entrenchment during periods of drought, instead of being related to destructive invasions. Therefore, it seems the great rivers of Central Asia were not just static "stage sets" for some of the turning points of world history, but in many instances, inadvertently or directly shaped the final outcomes and legacies of imperial ambitions in the region.

INTRODUCTION: To investigate the impact of partial lateral scatter (LS), backscatter (BS) and presence of air gaps on optically stimulated luminescence dosimeter (OSLD) measurements in an acrylic miniphantom used for dosimetry audit on the 1.5 T magnetic resonance-linear accelerator (MR-linac) system. METHODS: The following irradiation geometries were investigated using OSLDs, A26 MR/A12 MR ion chamber (IC), and Monaco Monte Carlo system: (a) IC/OSLD in an acrylic miniphantom (partial LS, partial BS), (b) IC/OSLD in a miniphantom placed on a solid water (SW) stack at a depth of 1.5 cm (partial LS, full BS), (c) IC/OSLD placed at a depth of 1.5 cm inside a 3 cm slab of SW/buildup (full LS, partial BS), and (d) IC/OSLD centered inside a 3 cm slab of SW/buildup at a depth of 1.5 cm placed on top of a SW stack (full LS, full BS). Average of two irradiated OSLDs with and without water was used at each setup. An air gap of 1 and 2 mm, mimicking presence of potential air gap around the OSLDs in the miniphantom geometry was also simulated. The calibration condition of the machine was 1 cGy/MU at SAD = 143.5 cm, d = 5 cm, G90, and 10 × 10 cm2 . RESULTS: The Monaco calculation (0.5% uncertainty and 1.0 mm voxel size) for the four setups at the measurement point were 108.2, 108.1, 109.4, and 110.0 cGy. The corresponding IC measurements were 109.0 ± 0.03, 109.5 ± 0.06, 110.2 ± 0.02, and 109.8 ± 0.03 cGy. Without water, OSLDs measurements were ∼10% higher than the expected. With added water to minimize air gaps, the measurements were significantly improved to within 2.2%. The dosimetric impacts of 1 and 2 mm air gaps were also verified with Monaco to be 13.3% and 27.9% higher, respectively, due to the electron return effect. CONCLUSIONS: A minimal amount of air around or within the OSLDs can cause measurement discrepancies of 10% or higher when placed in a high b-field MR-linac system. Care must be taken to eliminate the air from within and around the OSLD.

The source-sink relationship determines the overall agronomic performance of rice. Cloning and characterizing key genes involved in the regulation of source and sink dynamics is imperative for improving rice yield. However, few source genes with potential application in rice have been identified. Glucan, Water-Dikinase 1 (GWD1) is an essential enzyme that plays a pivotal role in the first step of transitory starch degradation in source tissues. In the present study, we successfully generated gwd1 weak mutants by promoter editing using CRISPR/Cas9 system, and also leaf-dominant overexpression lines of GWD1 driven by Osl2 promoter. Analysis of the gwd1 plants indicated that promoter editing mediated down-regulation of GWD1 caused no observable effects on rice growth and development, but only mildly modified its grain transparency and seed germination. However, the transgenic pOsl2::GWD1 overexpression lines showed improvements in multiple key traits, including rice yield, grain shape, rice quality, seed germination and stress tolerance. Therefore, our study shows that GWD1 is not only involved in transitory starch degradation in source tissues, but also plays key roles in the seeds, which is a sink tissue. In conclusion, we find that GWD1 is an ideal biotechnological target with promising potential for the breeding of elite rice cultivars via genetic engineering.

In this study, the validity of using the optically stimulated luminescence (OSL) technique as a scanning method for the detection of irradiated foodstuffs was investigated. Gamma-irradiated thyme, mint and clove samples were stimulated with a continuous wave blue light (CW-OSL) and their OSL signal sensitivity and stability properties were measured. While no OSL signal was observed for all non-irradiated spices, a significant increase in the intensity of OSL signals was noted for all samples after irradiation with 5 kGy, 10 kGy and 20 kGy. It was also observed that the measured OSL signals were well above the background level even after one year of irradiation. It is therefore concluded that irradiated thyme, mint and clove samples can clearly be identified using the CW-OSL technique without any pre-treatment even one year after irradiation. This result indicates that, where spices contain sufficient silicate minerals, the CW-OSL technique could be used as a method for detecting irradiation.

Optically stimulated luminescence (OSL) is used for dating in archaeology and geology, and in dosimetry of ionizing radiation. Some crystalline wide band-gap insulators may be used as OSL radiation detectors. Recently it was found that cerium-doped potassium sulfate crystals have promising luminescence properties. This paper presents some OSL properties of K2 SO4 :Ce crystals that were prepared in the form of pellets. The crystals exhibited a strong luminescence signal, good measurement repeatability, and high sensitivity for ionizing radiation. Best results were obtained after annealing at 1000°C. Dose-dependence characteristics were determined in the range 10 mGy to 11.2 Gy. OSL decay curves consisted of several components. Fast and slow OSL components of the phosphor had different properties. The slow component was quite stable and obeyed power law dependence. It decayed in the 55 days after irradiation. Its luminescence properties depended on sample preparation and the storage time after irradiation.

A direct verification of the three-dimensional (3D) proton clinical treatment plan prepared for tumor in the eyeball, using the Eclipse Ocular Proton Planning system (by Varian Medical Systems), has been presented. To achieve this, a prototype of the innovative two-dimensional (2D) circular silicone foils, made of a polymer with the embedded optically stimulated luminescence (OSL) material in powder form (LiMgPO4), and a self-developed optical imaging system, consisting of an illuminating light source and a high-sensitive CCD camera has been applied. A specially designed lifelike eyeball phantom has been used, constructed from 40 flat sheet LMP-based silicone foils stacked and placed together behind a spherical phantom made by polystyrene, all to reflect the curvature of the real eyeball. Two-dimensional OSL signals were captured and further analyzed from each single silicone foil after irradiation using a dedicated patient collimator and a 58.8 MeV modulated proton beam. The reconstructed 3D proton depth dose distribution matches very well with the clinical treatment plan, allowing for the consideration of the new OSL system for further 3D dosimetry applications within the proton radiotherapy area.

Optically stimulated luminescence (OSL) signal properties of pellets from three types of NaCl (two household salts and one analytical grade salt) were investigated for their use in prospective dosimetry. Special attention was given to the OSL signal behaviour with time. The readout protocol was optimised in terms of preheat temperature, and the OSL signal yield of the NaCl pellet with time as well as the fading of the OSL signal with time was investigated. The effects of acute and chronic irradiations were compared. Irradiations and readout were performed using a Risø TL/OSL reader (TL/OSL-DA-15, DTU Nutech, Denmark). The optimal preheat temperature was determined to be 100 ºC, yielding OSL signals similar to a 1 h pause before OSL signal readout. There was no OSL signal fading observed as a function of time, but a decrease in the OSL signal yield of the NaCl pellets with time resulted in an apparent inverse fading when converting the OSL signal to an absorbed dose. For chronic radiation exposures of up to five weeks, the sensitivity of the NaCl pellets was found to be stable. The results of this study show that the use of NaCl pellets for prospective dosimetry is a promising, cost-effective, and accessible complement to commercially available alternatives for accurate absorbed dose determinations.

The relative energy responses of three indigenously developed optically stimulated luminescence (OSL) phosphors in the disc form were studied in therapeutic photon and electron beams. Calibration in terms of absorbed dose was carried out in the dose range 5-500 cGy in 60 Co gamma rays, high energy X-rays, and electron beams used in radiotherapy. The combined standard uncertainty in the estimation of absorbed dose using these OSL discs (OSLDs) was 3.3%. Dose-response curves of these OSLDs in 60 Co gamma rays, 6 and 10 MV (flat and unflat), 15 MV and 6 and 15 MeV electron beams were found to be linear. Furthermore, these OSLDs exhibited a relative energy-dependent response for both photon and electron beams. The relative energy response correction factor for photon and electron beams were in the range 1.01-1.05 and 1.03-1.06, respectively.

This study investigated the feasibility of dosimetric measurements using Al2O3:C optically stimulated luminescence (OSL) dosimeters during fluoroscopy-guided procedures. The linearity and energy dependence of Al2O3:C OSL dosimeters were evaluated, and the air kerma rate at the operator's position was measured. The response of Al2O3:C OSL dosimeters to short, repetitive irradiations was compared to that of long uninterrupted irradiation. The change in response of the Al2O3:C OSL dosimeter under automatic exposure rate control (AERC) was evaluated with the use of various thicknesses of polymethyl-methacrylate (PMMA) plates (15-30 cm). The Al2O3:C OSL dosimeters could detect 5µGy and showed good linearity in doses of ≥10µGy (R2: 0.997-0.999,p< 0.001). The relative response of the Al2O3:C OSL dosimeter normalised to that of 36.8 keV was 0.828-1.101 at the energies investigated (30.6-46.0 keV). The air kerma rate at the operator's position was estimated to be 2.61-7.17µGy min-1depending on the heights representing different body parts. Repetitive short irradiations had no significant impact on the relative response of the Al2O3:C OSL dosimeters (p> 0.05). Despite a high energy dependence on the low energy beam used in fluoroscopy, the change in relative response of the Al2O3:C OSL dosimeter under AERC was within 5.7% depending on the thickness of the PMMA plates. Dosimetric measurement using Al2O3:C OSL dosimeters for patients and operators is feasible. However, one should be cautious about high standard deviations when measuring small doses of ≤20µGy using Al2O3:C OSL dosimeters. It is essential to perform intensive bleaching before measuring very small doses to minimise pre-irradiation counts.

Ordinary salt, NaCl, has many properties suitable for dosimetry and has been suggested for both retrospective and prospective optically stimulated luminescence (OSL) dosimetry. Lately, the focus has been on NaCl that is compressed into solid pellets, as this improves both its handling and dosimetric properties. In this project, the energy dependence of NaCl pellets produced in-house was investigated for photon energies between 30 and 1.25 MeV. The NaCl pellets were first exposed to free-in-air conditions, and the estimated absorbed dose to the NaCl pellets was compared to the air kerma,Kair, at the point of exposure. Second, a backscatter medium of polymethyl methacrylate (PMMA) was added, and NaCl pellets were exposed when positioned on a ISO slab phantom to relate the response in the NaCl to the personal dose equivalent,Hp(10). The results show a significant energy dependence for exposure to low-energy photons with a peak over-response compared toKairandHp(10) of up to 18. Comparisons with Monte Carlo simulations show good agreement, even though the simulations cannot account for properties related to the intrinsic luminescence effects of the NaCl pellets or the readout and calibration process. The finite thickness of the NaCl pellet makes it an imperfect Bragg-Grey cavity, which complicates the behaviour of the energy dependence. The results presented here may serve as an important basis for further experimental and theoretical modelling of a build-up layer and filters in efforts to develop a passive personal dosemeter based on NaCl.

The Initial Upper Paleolithic (IUP) temporally overlaps with the range expansion of Homo sapiens populations in various parts of Eurasia and is often considered a key archaeological phase for investigating behavioral changes from the Middle Paleolithic. This paper reports upon new data from IUP occupations at Wadi Aghar, a rock shelter site in the southern Levant. In combining the results of radiometric dates and lithic analyses, we clarify the chronological and cultural position of Wadi Aghar assemblages in the Levantine IUP. As for the records about mobility, on-site activities, and resource procurement behaviors, we present analyses of lithic use-wear, tool-type composition, soil micromorphology, and marine shells. The lithic analyses and the optically stimulated luminescence (and subsidiary radiocarbon) dating of the Wadi Aghar materials suggest their chronocultural position in the IUP (45-40 ka for Layers C-D1; 39-36 ka for Layer B; possibly 50 ka for Layer D2), providing the southernmost location for the IUP in Eurasia. In the Levant, Wadi Aghar represents one of the few IUP sites in the inland areas. The results also indicate that the timing and technological sequences from the IUP to the following bladelet industries differed between the inland and coastal zones, likely reflecting geographically variable adaptive behaviors and/or cultural transmissions. One of the behavioral characteristics of IUP foragers at Wadi Aghar is the procurement of remote resources, represented by the transportation of marine shells from the Red Sea: Canarium fusiforme and Canarium cf. mutabile. Whether it was a direct procurement with increased mobility or a result of intergroup exchanges, it was not part of behavioral repertoires during the late MP in the same area. This can be understood as the expansion of resource procurement range, functioning as additional buffers from risk in the semiarid environments in the inland Levant.

In this study the dosimetric properties of alumina (Al2O3) substrates found in resistors retrieved from mobile phones were investigated. Measurements of the decline of optically stimulated luminescence (OSL) generated following exposure of these substrates to ionising radiation showed that 16% of the signal could still be detected after 2 years (735 days). Further, the magnitude of the regenerative dose (calibration dose; D i) had no impact on the accuracy of dose estimates. Therefore, it is recommended that the D i be set as low as is practicable, so as to accelerate data retrieval. The critical dose, D CL, and dose limit of detection, D DL, taking into account the uncertainty in the dose-response relation as well as the uncertainty in the background signal, was estimated to be 7 and 13 mGy, respectively, 1 h after exposure. It is concluded that given the significant long-term component of fading, an absorbed dose of 0.5 Gy might still be detectable up to 6 years after the exposure. Thus, OSL from alumina substrates can be used for dosimetry for time periods far in excess of those previously thought.

RESUMO

Scintillation materials and detectors that are used in many applications, such as medical imaging, security, oil-logging, high energy physics and non-destructive inspection, are reviewed. The fundamental physics understood today is explained, and common scintillators and scintillation detectors are introduced. The properties explained here are light yield, energy non-proportionality, emission wavelength, energy resolution, decay time, effective atomic number and timing resolution. For further understanding, the emission mechanisms of scintillator materials are also introduced. Furthermore, unresolved problems in scintillation phenomenon are considered, and my recent interpretations are discussed. These topics include positive hysteresis, the co-doping of non-luminescent ions, the introduction of an aimed impurity phase, the excitation density effect and the complementary relationship between scintillators and storage phosphors.

The paper presents the results of optical dating of potassium-rich feldspar grains obtained from the Haua Fteah cave in Cyrenaica, northeast Libya, focussing on the chronology of the Deep Sounding excavated by Charles McBurney in the 1950s and re-excavated recently. Samples were also collected from a 1.25 m-deep trench (Trench S) excavated during the present project below the basal level of the Deep Sounding. Optically stimulated luminescence (OSL) data sets for multi-grain, single aliquots of quartz for samples from the Middle Trench were previously published. Re-analyses of these OSL data confirm significant variation in the dose saturation levels of the quartz signal, but allow the most robust OSL ages to be determined for comparison with previous age estimates and with those obtained in this study for potassium-rich feldspars from the Deep Sounding. The latter indicate that humans may have started to visit the cave as early as ∼150 ka ago, but that major use of the cave occurred during MIS 5, with the accumulation of the Deep Sounding sediments. Correlations between optical ages and episodes of "Pre-Aurignacian" artefact discard indicate that human use of the cave during MIS 5 was highly intermittent. The earliest phases of human activity appear to have occurred during interstadial conditions (5e and 5c), with a later phase of lithic discard associated with more stadial conditions, possibly MIS 5b. We argue that the "Pre-Aurignacian" assemblage can probably be linked with modern humans, like the succeeding "Levalloiso-Mousterian" assemblage; two modern human mandibles associated with the latter are associated with a modelled age of 73-65 ka. If this attribution is correct, then the new chronology implies that modern humans using "Pre-Aurignacian" technologies were in Cyrenaica as early as modern humans equipped with "Aterian" technologies were in the Maghreb, raising new questions about variability among lithic technologies during the initial phases of modern human dispersals into North Africa.

Mammalian extinction worldwide during the Late Pleistocene has been a major focus for Quaternary biochronology and paleoecology. These extinctions have been variably attributed to the impacts of climate change and human interference. However, until relatively recently, research has been largely restricted to the Americas, Europe, and Australasia. We present the oldest Middle-Late Pleistocene stratified and numerically dated faunal succession for the Indian subcontinent from the Billasurgam cave complex. Our data demonstrate continuity of 20 of 21 identified mammalian taxa from at least 100,000 y ago to the present, and in some cases up to 200,000 y ago. Comparison of this fossil record to contemporary faunal ranges indicates some geographical redistribution of mammalian taxa within India. We suggest that, although local extirpations occurred, the majority of taxa survived or adapted to substantial ecological pressures in fragmented habitats. Comparison of the Indian record with faunal records from Southeast and Southwest Asia demonstrates the importance of interconnected mosaic habitats to long-term faunal persistence across the Asian tropics. The data presented here have implications for mammalian conservation in India today, where increasing ecological circumscription may leave certain taxa increasingly endangered in the most densely populated region of the world.
