Thrombospondins (TSPs) are multidomain, calcium-binding glycoproteins that have wide-ranging roles in vertebrates in cell interactions, extracellular matrix (ECM) organisation, angiogenesis, tissue remodelling, synaptogenesis, and also in musculoskeletal and cardiovascular functions. Land animals encode five TSPs, which assembly co-translationally either as trimers (subgroup A) or pentamers (subgroup B). The vast majority of research has focused on this canonical TSP family, which evolved through the whole-genome duplications that took place early in the vertebrate lineage. With benefit of the growth in genome- and transcriptome-predicted proteomes of a much wider range of animal species, examination of TSPs throughout metazoan phyla has revealed extensive conservation of subgroup B-type TSPs in invertebrates. In addition, these searches established that canonical TSPs are, in fact, one branch within a TSP superfamily that includes other clades designated mega-TSPs, sushi-TSPs and poriferan-TSPs. Despite the apparent simplicity of poriferans and cnidarians as organisms, these phyla encode a greater diversity of TSP superfamily members than vertebrates. We discuss here the molecular characteristics of the TSP superfamily members, current knowledge of their expression profiles and functions in invertebrates, and models for the evolution of this complex ECM superfamily.

The current emphasis of schistosomiasis control is placed on preventive chemotherapy using praziquantel. However, reinfection may occur rapidly in the absence of complementary interventions. Recent studies from Senegal suggest that predatory prawns might feed on intermediate host snails and thus impact on schistosomiasis transmission. We designed a study with four repeated cross-sectional surveys pertaining to prawns and snails, coupled with a single cross-sectional parasitological survey among humans. We assessed for potential associations between the presence/density of prawns and snails and correlation with Schistosoma infection in a composite sample of school-aged children and adults. The study was carried out between October 2015 and December 2016 in 24 villages located near the Agnéby and Mé coastal river systems in south-eastern Côte d'Ivoire. At each site, snails and prawns were collected, and in each village, 150 individuals were subjected to stool and urine examination for the diagnosis of Schistosoma mansoni and S. haematobium. We found peaks of relative abundance of intermediate host snails in the villages of the Agnéby River system, while predatory prawns were predominantly recorded in the Mé River system. A negative association was observed between intermediate host snail densities and riverine prawns; however, no pattern was found between this trend in the predator-prey relationship and the prevalence of human schistosomiasis.

Ficolins have crucial functions in recognizing and eliminating pathogens in innate immunity. In this study, we identified two ficolin-like genes from the oriental river prawn, Macrobrachium nipponense. These genes were designated as MnFico1 and MnFico2. MnFico1 cDNA has 1600 bp, whereas MnFico2 has 1486 bp. In addition to a coiled-coil region or a low complexity region, the two ficolins both contained a signal peptide and a fibrinogen-related domain. qRT-PCR results showed that the highest expression level of MnFico1 expression was in the gills, whereas that of MnFico2 was in the heart. The transcripts of MnFico1 and MnFico2 could both respond to bacteria challenge. The transcription of antilipopolysaccharide factors ALFs (MnALF1 and MnALF2) and crustin genes (MnCru4, MnCru5, MnCru6, and MnCru7) was inhibited in the gills of MnFico1 or MnFico2 knockdown prawns at 24 h Vibrio parahaemolyticus challenge. Recombinant proteins of rMnFico1 and rMnFico2 could bind toward diverse bacteria and agglutinate Gram-negative and Gram-positive bacteria with the presence of calcium (Ca2+). rMnFico1 and rMnFico2 proteins also have lipopolysaccharide and peptidoglycan binding activity. Both recombinant ficolin proteins could help the prawn to facilitate the clearance of V. parahaemolyticus in vivo. Our results suggested that MnFico1 and MnFico2 might serve as pattern recognition receptors in M. nipponense.

Significant income was promised by giant freshwater prawn farming, which served as a key occupation for farmers. However, challenges were faced by traditional methods, including difficulties in selling prawns of incorrect sizes, limited market demand, low prices, and the risk of waterborne diseases. In pond-raised environments, these prawns were prone to diseases due to deteriorating environmental conditions, particularly poor pond bottoms, resulting in unsuitable water quality and vulnerability in prawn growth. Consequently, the application of water management technology in the prawn ponds becomes crucial to address these issues. This research aims to develop a prototype for a smart water management system, designed to regulate the water quality that significantly impacts the growth of Pak Phanang giant freshwater prawns within a pond. The equipment and tools utilized comprise ponds, water treatment systems, and smart water quality control systems. The investigation resulted in favorable findings, obtaining a total of 270 river prawns, each measuring an average length of 10-12 cm and weighing between 24 and 30 g. After a rearing period of 120 days in this system, 252 river prawns were successfully harvested, exhibiting a survival rate of 93.3 %. These prawns measured 20-25 cm in length and weighed between 190 and 268 g each, while remaining free from diseases. The results demonstrate that the developed system effectively manages water quality throughout the culture process, offering a valuable model to address the challenges faced by giant freshwater prawn farmers. The prawns obtained are of marketable size, and disease-free, enhancing the farmers' credibility with long-term benefits in the consumer market, in addition to their income generation potential.

Macrobrachiumlanchesteri (De Man, 1911), a translucent freshwater prawn has a wide distribution range throughout mainland Southeast Asia. A high morphological variation and genetic divergence between different geographical M.lanchesteri populations in Thailand have peculiarly extended the uncertainty of species boundaries and blended confusingly with several Macrobrachium species. To clarify these circumstances, broad sample examinations of the morphological variation, including topotype specimens, and phylogenetic reconstruction based on the concatenated mitochondrial dataset (16s rRNA and COI genes) were performed. Broad morphological examination of M.lanchesteri has shown congruency with phylogenetic analyses by revealing prominent lineages of M.lanchesteri sensu stricto and two new sibling lineages with interspecific variation between 6.48-8.76% for COI and 3.06-4.23% for 16S. Descriptions of two new species, named herein as M.panhai Chaowvieng & Siriwut, sp. nov. and M.rostrolevatus Chaowvieng & Siriwut, sp. nov. are provided. Morphological investigation of rostral form suggested plasticity in M.rostrolevatus populations showing the morphological trait associated with their habitat preferences. Furthermore, phylogenetic positions of the three taxa affirmed the hidden diversity of Thai freshwater Macrobrachium fauna correlated with the river network in the Mekong and Chao Phraya basins, Thailand. The genetic data and distribution records obtained in this study may also assist future river conservation plans as well as the sustainable management of freshwater prawn diversity.

The colour of seafood flesh is often not homogenous, hence measurement of colour requires repeat measurements to obtain a representative average. The aim of this study was to determine the optimal number of repeat colour measurements required for three different devices [machine vision (digital image using camera, and computer processing); Nix Pro; Minolta CR400 colorimeter] when measuring three species of seafood (Atlantic salmon, Salmo salar, n = 8; rockling, Genypterus tigerinus, n = 8; banana prawns, Penaeus merguiensis, n = 105) for raw and cooked samples. Two methods of analysis for number of repeat measurements required were compared. Method 1 was based on minimising the standard error of the mean and Method 2 was based on minimising the difference in colour over repeat measurements. Across species, using Method 1, machine vision required an average of four repeat measurements, whereas Nix Pro and Minolta required 13 and 12, respectively. For Method 2, machine vision required an average of one repeat measurement compared to nine for Nix Pro and Minolta. Machine vision required fewer repeat measurements due to its lower residual variance: 0.51 compared to 3.2 and 2.5 for Nix Pro and Minolta, respectively. In conclusion, machine vision requires fewer repeat measurements than colorimeters to precisely measure the colour of salmon, prawns, and rockling.

Use of agrochemicals, including insecticides, is vital to food production and predicted to increase 2-5 fold by 2050. Previous studies have shown a positive association between agriculture and the human infectious disease schistosomiasis, which is problematic as this parasitic disease infects approximately 250 million people worldwide. Certain insecticides might runoff fields and be highly toxic to invertebrates, such as prawns in the genus Macrobrachium, that are biocontrol agents for snails that transmit the parasites causing schistosomiasis. We used a laboratory dose-response experiment and an observational field study to determine the relative toxicities of three pyrethroid (esfenvalerate, λ-cyhalothrin, and permethrin) and three organophosphate (chlorpyrifos, malathion, and terbufos) insecticides to Macrobrachium prawns. In the lab, pyrethroids were consistently several orders of magnitude more toxic than organophosphate insecticides, and more likely to runoff fields at lethal levels according to modeling data. At 31 water contact sites in the lower basin of the Senegal River where schistosomiasis is endemic, we found that Macrobrachium prawn survival was associated with pyrethroid but not organophosphate application rates to nearby crop fields after controlling for abiotic and prawn-level factors. Our laboratory and field results suggest that widely used pyrethroid insecticides can have strong non-target effects on Macrobrachium prawns that are biocontrol agents where 400 million people are at risk of human schistosomiasis. Understanding the ecotoxicology of high-risk insecticides may help improve human health in schistosomiasis-endemic regions undergoing agricultural expansion.

This study detected two potential pathogens, Vibro parahaemolyticus, which causes acute hepatopancreatic necrosis disease (AHPND), and white spot syndrome virus (WSSV), in fishing bait in South Korea. However, their infectious nature was not confirmed, possibly due to the degradation caused by freezing/thawing or prolonged storage under frozen conditions. While infectivity was not confirmed in this study, there is still a significant risk of exposure to these aquatic products. Furthermore, fishing bait and feed should be handled with caution as they are directly exposed to water, increasing the risk of disease transmission. In Australia, cases of WSSV infection caused by imported shrimp intended for human consumption have occurred, highlighting the need for preventive measures. While freezing/thawing is a method for inactivating pathogens, there are still regulatory and realistic issues to be addressed.

Methods to induce analgesia and anesthesia for research purposes, handling, transport, or stunning have been used in 71 species of crustaceans. A non-systematic literature search was conducted on crustacean anesthetic methods. This review presents a comprehensive evaluation of drugs and non-chemical methods used to provide analgesia and anesthesia in many crustacean species rather than just decapod crustaceans. This information allows users to select an appropriate method or agent for their species of interest. We prepared an on-line tool based on datasette, a no-code open-source solution for simple web-based database frontends that allows exploration and downloading data by method, analgesic/anesthetic, species, life stage, or sex, as well as other data including environmental conditions (temperature, salinity, light), route of administration, dosage, and induction and recovery times. These values can be selected to filter the dataset and export it to CSV or JSON formats. Currently, several techniques and chemicals are, in our opinion, unsuitable for use as anesthetics in crustaceans, and the basis for these opinions are presented. Given the evidence of a pain-like experience in crustaceans, we propose that researchers should treat crustaceans humanely, applying the principles of good handling, care, and the management of stress and pain to safeguard their welfare.

Microplastic abundance and characteristics were assessed in five decapod crustaceans purchased from seafood markets and collected in coastal waters around Australia (South Australia, New South Wales, Queensland, Northern Territory, and Western Australia). Three species of prawns (king, banana and tiger prawns) and two species of crabs (blue-swimmer and mud crabs) were analysed. Muscle tissues and gastro-intestinal tracts in prawns, and gastro-intestinal tracts in crabs, were chemically digested, with microplastic identification verified using Fourier Transform Infrared spectroscopy. Forty-eight percent of crustaceans contained microplastics. Prawns and crabs had 0.8 ± 0.1 and 1.6 ± 0.1 pieces per individual, respectively, with spatial patterns evident. Microplastics were predominantly fibres (98%) of blue (58%) and black (24%) colours with polyolefin including polyester the most prevalent polymers. Overall, compared to a systematic review we performed of microplastics in decapod crustaceans worldwide, microplastic loads in crustaceans from Australia were in the lower range of plastic contamination.

Chlorine or chloric disinfectants are cost-effective disinfectants, which are widely used to disinfect domestic and industrial water. The residual chlorine levels in some of these waters have been proven toxic to several aquatic organisms; however, the molecular mechanisms of toxicity of residual chlorine to aquatic crustaceans, including Macrobrachium nipponense, an economically important freshwater prawn native to Asian countries, have not been investigated to date. Here, M. nipponense was exposed to 0.53 mg/L of residual chlorine, and comparative transcriptomics analyses were performed to determine their response mechanisms at the molecular level. Residual chlorine caused lethal effects on prawns. Furthermore, a total of 940 differentially expressed genes (DEGs), including 501 up-regulated and 439 down-regulated genes, were identified after 48 h of residual chlorine exposure compared to the control group. After enrichment analysis of GO (Gene Ontology) functions and KEGG (Kyoto Encyclopedia of Genes and Genomes) pathways, identified DEGs were demonstrated to be associated with a variety of functions including exerting "oxidoreductase activity", and participating in "oxidation-reduction process". In addition, cytochrome P450 family 1 subfamily A1 (CYP1A1), glutathione S-transferase (GST), and glucuronosyltransferase (UGT) were enriched in the pathway of metabolism of xenobiotics by cytochrome P450. Furthermore, protein-protein interaction (PPI) network analysis revealed interactions among actin beta/gamma 1 (ACTB_G1) gene encoding protein and a series of multiple functional DEGs (e.g., hexokinase (HK), fructose 1,6-biphosphate-aldolase A (ALDOA), cytochrome c (CYC), and elongation factor 1-alpha (EEF1A)) encoding proteins. This study laid a theoretical foundation for safety evaluation of chlorinated aquatic water and further investigation of the toxicity of chlorination to M. nipponense.

Acid sulfate soils are a major problem in modified coastal floodplains and are thought to have substantial impacts on estuarine species. In New South Wales, Australia, acid sulfate soils occur in every estuary and are thought to impact important fisheries species, such as Eastern School Prawn (Metapenaeus macleayi). These fisheries have experienced declining productivity over the last ten years and increasing occurrence of catchment-derived stressors in estuaries contribute to this problem. We evaluated the effect of pH 4-7.5 on School Prawn survival at two salinities (27 and 14.5), pH 5, 6 and 7.5 on the predation escape response (PER) speed at two salinities (27 and 14.5), and pH 4 and 7.5 on respiration rates. While mortality appeared to be greater in the high salinity treatment, there was no significant relationship between proportional survival and pH for either salinity treatment. Respiration was significantly slower under acidic conditions and the average PER was almost twice as fast at pH 7.5 compared to pH 5 (p < 0.05), indicating prawns may fall prey to predation more easily in acidic conditions. These findings confirm the hypothesised impacts of acidic water on penaeid prawns. Given that the conditions simulated in these experiments reflect those encountered in estuaries, acidic runoff may be contributing to bottlenecks for estuarine species and impacting fisheries productivity.

Biological synthesis of silver nanoparticles (AgNPs) by Cheatomorpha antennia and its in vitro and in vivo antibacterial activity against Vibrio harveyi in Macrobrachium rosenbergii was demonstrated in the study. In vitro growth curve analysis, cell viability and bacterial inhibitory assays were performed to test the efficacy of synthesised AgNPs against bacteria. Sodium caseinate was used as an encapsulating agent to deliver the antibacterial drugs and the commercial process of microencapsulation comprises the antibacterial bioelements for oral administration to improve the disease resistance of AgNPs against V. harveyi due to the eco-friendly for non-toxic behaviour of nanoparticle and their treatment. Characterisation of antibacterial silver was performed by UV spectroscopy, X-ray diffraction, Fourier Transform Infrared spectroscopy and Scanning Electron Microscopy. The peak at 420 nm showed the presence of nanoparticles in the solution and the crystal nature of the particle was identified by the XRD. FTIR characterised the functional harveyi biomolecules and further SEM confirmed the size of the nanoparticles around 24 ± 2.4 nm. Experimental pathogenicity of V. harveyi showed 100% mortality at the 120th hour. Treatment of encapsulated AgNPs was administered orally for the relative percentage of survival which acquired almost 90% of survival till 30 days of exposure. In conclusion, the microencapsulation of AgNPs in the biopolymer matrices promotes the health, growth responses, immunity and disease resistance of encapsulated AgNPs with an improved relative percentage of survival.

Aquatic ecosystems are used for extensive rice-shrimp culture where the available water alternates seasonally between fresh and saline. Poor water quality has been implicated as a risk factor for shrimp survival; however, links between shrimp, water quality and their main food source, the natural aquatic biota inhabiting these ponds, are less well understood. We examined the aquatic biota and water quality of three ponds over an entire year in the Mekong Delta, Vietnam, where the growing season for the marine shrimp Penaeus monodon has been extended into the wet season, when waters freshen. The survival (30-41%) and total areal biomass (350-531 kg ha-1) of shrimp was constrained by poor water quality, with water temperatures, salinity and dissolved oxygen concentrations falling outside known optimal ranges for several weeks. Declines in dissolved oxygen concentration were matched by declines in both shrimp growth rates and lipid content, the latter being indicative of nutritional condition. Furthermore, as the dry season transitioned into the wet, shifts in the taxonomic composition of phytoplankton and zooplankton were accompanied by declines in the biomass of benthic algae, an important basal food source in these systems. Densities of the benthic invertebrates directly consumed by shrimp also varied substantially throughout the year. Overall, our findings suggest that the survival, condition and growth of shrimp in extensive rice-shrimp ecosystems will be constrained when poor water quality and alternating high and low salinity negatively affect the physiology, growth and composition of the natural aquatic biota. Changes in management practices, such as restricting shrimp inhabiting ponds to the dry season, may help to address these issues and improve the sustainable productivity and overall condition of these important aquatic ecosystems.

Mass mortalities of the larval stage of the giant freshwater prawn, Macrobrachium rosenbergii, have been occurring in Bangladesh since 2011. Mortalities can reach 100% and have resulted in an 80% decline in the number of hatcheries actively producing M. rosenbergii. To investigate a causative agent for the mortalities, a disease challenge was carried out using infected material from a hatchery experiencing mortalities. Moribund larvae from the challenge were prepared for metatranscriptomic sequencing. De novo virus assembly revealed a 29 kb single­stranded positive-sense RNA virus with similarities in key protein motif sequences to yellow head virus (YHV), an RNA virus that causes mass mortalities in marine shrimp aquaculture, and other viruses in the Nidovirales order. Primers were designed against the novel virus and used to screen cDNA from larvae sampled from hatcheries in the South of Bangladesh from two consecutive years. Larvae from all hatcheries screened from both years were positive by PCR for the novel virus, including larvae from a hatchery that at the point of sampling appeared healthy, but later experienced mortalities. These screens suggest that the virus is widespread in M. rosenbergii hatchery culture in southern Bangladesh, and that early detection of the virus can be achieved by PCR. The hypothesised protein motifs of Macrobrachium rosenbergii golda virus (MrGV) suggest that it is likely to be a new species within the Nidovirales order. Biosecurity measures should be taken in order to mitigate global spread through the movement of post-larvae within and between countries, which has previously been linked to other virus outbreaks in crustacean aquaculture.

The influence of sedimentary metals on the cockle A. trapezia tissue was examined using a strong difference in sedimentary metal concentrations in an embayment (Hen and Chicken Bay) highly contaminated in Cu and an adjacent cove (Iron Cove), strongly enriched in Cd, Cu, Pb and Zn within the heavily-urbanised Sydney estuary catchment (Australia). Statistically significant differences were recorded for cockle tissue metal concentrations between the study locations reflecting differences in surficial sediment metal concentrations. Low metal uptake was apparent in A. trapezia tissue, which were considerably less than background sedimentary concentrations and was of low-risk for human consumption. Dissimilar bioconcentration of Cd, Cu, Pb and Zn was apparent in tissue of cockles (A. trapezia), prawns (Metapenaeus bennettae), mussels (Mytilus galloprovincialis) and oysters (Saccostrea glomerata), due possibly to different feeding patterns and biogeochemical conditions in bottom sediments.

Globally, vibrios represent an important and well-established group of bacterial foodborne pathogens. The European Commission (EC) mandated the Comite de European Normalisation (CEN) to undertake work to provide validation data for 15 methods in microbiology to support EC legislation. As part of this mandated work programme, merging of ISO/TS 21872-1:2007, which specifies a horizontal method for the detection of V. parahaemolyticus and V. cholerae, and ISO/TS 21872-2:2007, a similar horizontal method for the detection of potentially pathogenic vibrios other than V. cholerae and V. parahaemolyticus was proposed. Both parts of ISO/TS 21872 utilized classical culture-based isolation techniques coupled with biochemical confirmation steps. The work also considered simplification of the biochemical confirmation steps. In addition, because of advances in molecular based methods for identification of human pathogenic Vibrio spp. classical and real-time PCR options were also included within the scope of the validation. These considerations formed the basis of a multi-laboratory validation study with the aim of improving the precision of this ISO technical specification and providing a single ISO standard method to enable detection of these important foodborne Vibrio spp.. To achieve this aim, an international validation study involving 13 laboratories from 9 countries in Europe was conducted in 2013. The results of this validation have enabled integration of the two existing technical specifications targeting the detection of the major foodborne Vibrio spp., simplification of the suite of recommended biochemical identification tests and the introduction of molecular procedures that provide both species level identification and discrimination of putatively pathogenic strains of V. parahaemolyticus by the determination of the presence of theromostable direct and direct related haemolysins. The method performance characteristics generated in this have been included in revised international standard, ISO 21872:2017, published in July 2017.

Commercially-important species of fish and a crustacean from four sites in the Musa estuary and a site in the Persian Gulf have been analysed for the presence and location of microplastics (MPs). A total of 828 MPs were detected in the guts (gastrointestinal tracts), skin, muscle, gills and liver of demersal and pelagic fish (Platycephalus indicus, Saurida tumbil, Sillago sihama, Cynoglossus abbreviatus) from all five sites and in the exoskeleton and muscle of the tiger prawn, Penaeus semisulcatus, from three sites. On an individual basis, MPs were most abundant in P. indicus (mean = 21.8) and least frequently encountered in P. semisulcatus (mean = 7.8), but when normalized on a mass basis, MPs ranged from 0.16 g-1 for C. abbreviatus to 1.5 g-1 for P. semisulcatus. Microscopic analyses (polarized light, fluorescence, SEM/EDS) revealed that MPs were mainly fibrous fragments (with a few angular fragments) of various colour and size (<100 µm to > 1000 µm) and with strong C and O signatures. Additional particles detected that were distinctly different in colour, morphology, brittleness and elemental composition (part-metallic, and containing Cu) were suspected of being fragments of antifouling paint. The means of entry of MPs into tissues not involved in digestion are unclear but could be related to translocation or adherence. Regardless of the mode of accumulation, the presence of MPs in heavily fished species of fish and crustacean raises concerns about the potential transfer of synthetic materials into humans.

Calcium phosphate adsorbents, derived from prawns and crabs shell biomass wastes have been developed using wet chemistry and low temperature treatment. The adsorbents were characterized by X-ray diffractometry and Fourier transform infrared spectroscopy. Batch adsorption test were carried out to investigate their effectiveness in adsorption of fluoride from ground and surface waters. Adsorption capacities were compared with bone char and synthetic hydroxyapatite (CCHA). Results indicate that prawns derived adsorbent (PHA) formed hexagonal structure with phases identifiable with hydroxyapatite while crabs based adsorbent (CHA) formed predominantly monoclinic structure with crystalline phase characteristic of brushite. Vibrational analysis and kinetic studies predicted defluoridation occurred mainly by ion exchange and ion adsorption mechanisms. Defluoridation capacity of the adsorbents was found to be superior compared to bone char and CCHA. CHA was the most effective with efficiencies above 92% and highest capacity of 13.6 mg/g in field water with fluoride concentration of 5-70 mg/L. PHA had highest capacity of 8.5 mg/g which was still better than 2.6 mg/g recorded by CCHA and bone char. Adsorption was best described by pseudo 2nd order kinetics. The findings indicate that crustacean derived calcium phosphate systems have better potential for defluoridation than traditional bone char and synthetic systems.

Invertebrate neuropeptide F-I (NPF-I), much alike its mammalian homolog neuropeptide Y, influences several physiological processes, including circadian rhythms, cortical excitability, stress response, and food intake behavior. Given the role of autophagy in the metabolic stress response, we investigated the effect of NPF-1 on autophagy during fasting and feeding conditions in the hepatopancreas and muscle tissues of the male giant freshwater prawn Macrobrachium rosenbergii. Starvation up-regulated the expression of the autophagy marker LC3 in both tissues. Yet, based on the relative levels of the autophagosome-associated LC3-II isoform and of its precursor LC3-I, the hepatopancreas was more responsive than the muscle to starvation-induced autophagy. Injection of NPF-I inhibited the autophagosome formation in the hepatopancreas of fasting prawns. Relative to the body weight, the muscle weight was not affected, while that of the hepatopancreas decreased upon starvation and NPF-1 treatment could largely prevent such weight loss. Thus, the hepatopancreas is the reserve organ for the nutrient homeostasis during starvation and NPF-I plays a crucial role in the balancing of energy expenditure and energy intake during starvation by modulating autophagy.
