OBJECTIVE: To determine the effect of Radix Angelicae Sinensis and Radix Hedysari (RAS-RH) on radiosensitivity of human liver cancer H22 cells to heavy ion ¹²C6⁺and its possible mechanism. METHODS: The experiment involved a comparison of proliferation of H22 cells (detected by CCK-8 assay) between four groups: control,drug (RAS-RH),radiation,and combination (RAS-RH+radiation). H22 cells were treated with different doses of radiation alone or radiation followed by RAS-RH. The radiation enhancement effect of RAS-RH on H22 was detected by Colony forming assay. The effect of RAS-RH on the apoptosis of H22 cells was detected by flow cytometry. The influence of RAS-RH on the expression levels of related protein Survivin and Caspase-9 was investigated by Western blot. RESULTS: RAS-RH inhibited the proliferation of H22 cells,with a time and dose dependency [inhibitory concentration 20% (IC20)=(117.60±2.15) mg/L]. The survival rate of H22 cells decreased significantly with the increase of heavy ion ¹²C6⁺. The two survival curves produced by the Graph Pad Prism 5.0 software were clearly separated. The combination group demonstrated smaller shoulder area at low dosage and lower survival rate of cells compared with radiation group,with a sensitization enhancement ratio (SER) of 1.39±0.07. The combination group (100 mg/L RAS-RH+2 Gy) had higher apoptosis rate and Caspase-9 protein expression level,and lower Survivin protein expression level,compared with other 3 groups ( P<0.01). CONCLUSION: RAS-RH has radiation sensitization effect on human hepatocellular carcinoma H22 cells. The mechanism may be related to down-regulation of Survivin protein expression and up regulation of Caspase-9 protein expression.

Radix Hedysari, a well-known traditional Chinese herbal medicine, has a long history as a medicinal plant in China based on its wide spectrum of biological and pharmacological activities. Until now, many chemical constituents have been isolated and identified from Radix Hedysari, such as polysaccharides, flavonoids, phenylpropanoids, trace elements and so on. Of these, Radix Hedysari polysaccharides are one of the most important active compounds of the Radix Hedysari and have various biological activities, including anti-tumor activity, antioxidant activity, anti-diabetic activity, immunity enhancement effect and regulation of intestinal flora. These beneficial biological activities are related to the chemical structure of the Radix Hedysari polysaccharides. The chemical structure of HPS is the basis of its biological activity, which is affected by many factors, such as the composition of monosaccharide, the size of relative molecular weight, the way of glycoside bond connection, the three-dimensional structure of polysaccharide, and so on. Different extraction and separation methods lead to different configurations of polysaccharides and different biological activities of polysaccharides. In general, the bioactivity of polysaccharides showed a certain dose-response or structure-activity relationship. At present, few studies of regarding the structure-function relationships of these polysaccharides have been reported, and it is not easy to relate the structures of HPS to their biological activities. Nevertheless, some relationships can be inferred as follows. This article is aimed to provide a systematic and up-to-date review on the extraction, purification, structural characterization, and biological activities of the Radix Hedysari polysaccharides to support its further therapeutic potentials and sanitarian functions. Furthermore, the possible development and a perspective for future research of Radix Hedysari polysaccharides are also discussed.

Ulcerative colitis (UC) is a chronic inflammatory disease with an unknown precise etiology. This study proves that Radix Hedysari (RH) ameliorates UC. Four RH extracts were used to ameliorate UC induced by 2,4-Dinitrobenzenesulfonic acid by 7 days intervention in agreement to preliminary studies. Compared to treatment with RH extracts, the RH ethanol extract (EE) was found to be more effective in ameliorating UC. With EE, the DAI were significantly decreased. Macroscopic and histopathological assessments suggest that the colon mucosa was repaired, the organizational structure of the colon had been rebuilt. The levels of MPO, TNF-α, IL-1ß, and MDA were significantly decreased (p < .01), the levels of T-SOD and CAT were significantly increased (p < .01). Moreover, the compounds in EE were analyzed by HPLC. The results show that EE can ameliorate UC, and its anti-inflammatory capability probably plays an important role. RH can act as a functional food and ameliorate UC. PRACTICAL APPLICATIONS: In this work, the ameliorative effect of RH on UC was evaluated from multiple angles. There are two practical applications of this work. On the one hand, a new approach to ameliorating UC is provided by this work. In addition, UC patients have a new option for improving their symptoms. On the other hand, this work also provides information on how best to process RH for therapeutic use. In addition, we can utilize some compounds of RH that were once considered useless and reduce the waste of natural resources.

Aim To explore the possible mechanism of "component-target-pathway" of Radix Hedysari against target organ damage caused by radiotherapy and chemotherapy, and to verify the " dose-effect" relationship of the main active components. Methods TCMSP, Uniprot, Swiss Target Prediction, GeneCards, Cytoscape, Omicshare and other platforms were used for network pharmacology analysis. Autodock, Pymol and Ligplot were used for molecular docking. The water extract of Radix Hedysari was used for animal experiment verification. The contents of eight main components were determined by HPLC. Results Four active components, eight key targets and four key pathways of Radix Hedysari were identified to resist the damage of target organs caused by radiotherapy and chemotherapy. Molecular docking showed that formononetin and quercetin had good binding activity with HSP90AA1, naringenin and MAPK3, and ursolic acid and TP53. Animal experiments showed that gastrointestinal factors MTL and VIP increased significantly, liver and kidney factors Cr, BUN, AST and ALT decreased significantly, inflammatory factor IL-10 increased significantly and TNF-a decreased significantly. The traditional Chinese medicine of Radix Hedysari plays an important role in invigorating gas for ascending, benefiting blood for promoting production of fluid, and promoting circulation for removing obstruction in collaterals, which is consistent with the principle of treatment for osteoporosis. This study is designed to investigate the bioactive components on increasing peak bone mass (PBM) by exploring the spectrum-effect relationship between chromatography fingerprints and effect. Multiple indicators are selected to evaluate the pharmacological activity. In fingerprints, 21 common peaks are obtained, five of which are identified. Furthermore, gray relational analysis (GRA) is a quantitative method of gray system theory and is used to describe the correlation degree of common peaks and pharmacological activities with relational value. 21 components are then divided into three different regions, of which ononin and calycosin play an extremely significant role in increasing PBM. In addition, factor analysis and hierarchical cluster analysis (HCA) are used to screen the optimal producing area for Radix Hedysari. This provides a comprehensive and efficient method to improve the quality evaluation of Radix Hedysari, confirming the bioactive components for PBM-enhancement and further develop its medicinal value.

Radix Angelica Sinensis and Radix Hedysari are traditional Chinese medicines that are used for preventing and treating various diseases. This study aimed to investigate the effect and possible underlying mechanisms of Radix Angelica Sinensis and Radix Hedysari ultrafiltration extract (RAS-RH) on X-irradiation-induced cardiac fibrosis in rats. Our data demonstrated that (a) a single dose of total body irradiation (TBI) at 8 Gy resulted in cardiac fibrosis, whereas the control hearts exhibited less collagen and fibrosis. RAS-RH mitigated these morphological injuries. (b) TBI resulted in an increase in the serum levels of transforming growth factor ß1 (TGF-ß1) and troponin-I (TnI). RAS-RH inhibited the release of TBI-induced serum TGF-ß1 and the TnI levels. (c) TBI inhibited the apoptosis of primary rat cardiac fibroblasts, whereas RAS-RH induced the apoptosis of primary rat cardiac fibroblasts after X- irradiation. (d) TBI resulted in an increase in the expression of osteopontin (OPN), c-fos, c-jun, miRNA-21 and collagen1α (COL1α) in primary rat cardiac fibroblasts, and RAS-RH mitigated the TBI-induced increased expression of OPN, c-jun, miRNA-21 and COL1α. In conclusion, these results demonstrate that RAS-RH exerts antifibrotic effects possibly through inducing the apoptosis of fibroblasts, inhibiting the release of serum TGF-ß1, reducing the levels of serum TnI and reducing the expression of OPN, c-jun, miRNA-21 and COL1α. Therefore, RAS-RH may potentially be developed as a medical countermeasure for the mitigation of radiation-induced myocardial fibrosis.

OBJECTIVE: To study the intervention effects of Radix Hedysari, Radix Astragalus and compatibility of Angelica Sinensis on blood deficiency model mice induced by cyclophosphamide (CTX). METHODS: The mice were randomly divided into 7 groups, 10 mice each group. The blood deficiency model was established by CTX. The blank group and model group were treated with saline by gavage, while mice in positive group were administered with Lvjiaobuxue granule. Four dosage group were administered with Radix Hedysari, Radix Hedysari-Radix Angelica Sinensis(5:1), Radix Astragalus and Radix Astragalus-Radix Angelica Sinensis(5:1) water decoction. All the drugs were administered to mice for consecutive 7 d. The contents of red blood cell (RBC), lymphocyte(LYM), hematocrit (HCT), white blood cell (WBC), platelet (PLT) were detected by hematology analyzer, while thymus index(TI), spleen index(SI), reticulocyte (RC), marrow karyocyte (MK) were calculated, and the femur by pathological section were observed by microscope. RESULTS: Compared with blank group, the contents of RBC, WBC, HCT, PLT, LYM were decreased in model group (P<0.05). Compared with model group, the contents of RBC, WBC, HCT, PLT, LYM, RC and marrow karyocyte were increased in Hedysari-Angelica Sinensis(5:1) and Astragalus Angelica Sinensis(5:1) (P<0.05), at the same time, the pathological damage of femur could be improved. CONCLUSIONS: The effect of enrichment blood on blood deficiency model mice in Hedysari-Angelica Sinensis (5:1) and Astragalus-Angelica Sinensis(5:1) were superior to Hedysari and Astragalus.

Beta-amyloid (Aß) peptide, the hallmark of Alzheimer's disease, invokes oxidative damage to neurons and eventually leads to neuronal death. Selenylation modification of polysaccharide obtained from Radix hedysari (RHP) was studied to access antioxidant activities and neuroprotective effects against oxidative stress and apoptosis induced by Aß25-35 in vitro. A series of the selenylation derivatives of RHP (Se-RHP) was synthesized using nitric acid-sodium selenite (HNO3-Na2SeO3) method. The organic selenium content of Se-RHP increased from 1.04 to 3.29 mg/g. However, compared with the weight-average molecular mass (Mw) of RHP, Mw of Se-RHP showed a significant decrease, and varied from 27.7 kDa to 62.7 kDa. FT-IR spectra and (13)C NMR spectra indicated the selenite groups had been introduced mainly at the C-6 positions of RHP. Compared with RHP, Se-RHP showed greater antioxidant activities in vitro. Furthermore, both RHP and Se-RHP3 had neuroprotective effects against Aß25-35-induced oxidative stress and apoptosis in SH-SY5Y human neuroblastoma cells, which might be a potential therapeutic agent for preventing or treating neurodegenerative diseases.

PURPOSE: The aim of this study is to investigate the anti-inflammatory effect of Radix Hedysari Polysaccharide (HPS) on clinical indicators, the expression of Toll-like receptor-4 (TLR4) and its downstream transduction molecules during endotoxin-induced uveitis in rats. METHODS: EIU was induced through the intraperitoneal injection of male Wistar rats with lipopolysaccharide (LPS 200 µg). HPS (400mg/kg), DXM (1mg/kg) or an equivalent volume of normal saline was injected intraperitoneally 1h before the LPS induction. The clinical manifestation was observed and scored at 2-h intervals using a slit microscope. The degree of inflammatory reaction was determined by routine histological examinations, and the expression of TLR4 and MyD88 in the iris-ciliary body complex was detected through a double-labeled immunofluorescence study. Real-time RT-PCR was used to assess the effects of HPS on the expression of the TLR4 complex, MyD88 and NF-κB p65 mRNA. The protein expression levels of TLR4, MyD88 and NF-κB p65 were examined by western blot. RESULTS: HPS treatment produced similar therapeutic results with dexamethasone by significantly reducing the clinical severity of EIU as well as fibrin exudations and inflammatory cell infiltration in the eye. Correspondingly, according to the immunofluorescence results, HPS treatment significantly suppressed the expression of TLR4 and MyD88 in the iris-ciliary body complex. HPS treatment could also remarkably reduce the mRNA and protein expression of the TLR4 complex, MyD88 and NF-κB p65. CONCLUSION: HPS can suppress the intraocular inflammation observed in EIU by inhibiting TLR4 and its downstream signal transduction pathway.

Radix Hedysari polysaccharides (HPS) is the principal active fraction of Radix Hedysari (RH). The information about HPS3d, the main fraction of HPS3, and its effect on bone is still unknown. In the present study, the purified HPS3d was obtained by anion-exchange column. It consisted of 94.38% polysaccharide, 3.40% protein and 13.30% uronic acid. The molecular weight was measured to be 84.6kDa. The backbone consisted of galactopyranose and galacturonopyranose, and the side chains were composed of glucopyranose, rhamnopyranose and arabinofuranose. The FT-IR and elemental analysis showed that HPS3d was the sulfated polysaccharide. HPS3d upregulated alkaline phosphatase (ALP) activity and the expression of other osteogenic marker genes in osteoblast. In addition, HPS3d increased the expression and transcriptional activity of Runt-related transcription factor 2 (Runx-2) and Osterix, the two master genes of osteoblast differentiation. These findings suggest that HPS3d stimulates osteoblast differentiation by activation of Runx-2 and Osterix.

A new sulfated acetamido-heteropolysaccharide, HPS4-2A, was obtained by aqueous extraction followed by precipitation with ethanol and fractionation with DEAE column chromatography from Radix Hedysari. It was composed of rhamnose, arabinose, glucose, galactose and 2-acetamido-2-deoxy-d-galactose in the molar ratio of 10.09%:25.90%:25.90%:25.0%:12.30%. Elemental analysis indicated that HPS4-2A was a sulfated polysaccharide containing small amount of sulfate groups (1.87%). Partial acid hydrolysis, GC, GC-MS, 1D and 2D NMR spectroscopy analysis of the HPS4-2A revealed a predominance of glucose, galactose and 2-acetamido-2-deoxy-D-galactose linked in a highly-branched structure. The molecular weight of HPS4-2A was determined by HPSEC and HPSEC-MALLS. AFM study indicated that HPS4-2A took a highly branched conformation, which in consistent with the result studied by SEC-MALLS. Structural features of HPS4-2A were also investigated by SEM and TEM. Antioxidant assays demonstrated that HPS4-2A possessed of strong DPPH and hydroxyl radicals scavenging activities, suggesting that HPS4-2A could potentially be used as natural antioxidant.

Objective To observe the effects of Radix Hedysari on collagen area, hyaluronic acid (HA) and laminin (LN) of lung tissues in rat with pulmonary interstitial fibrosis; To investigate the mechanism and screen the best active ingredients in Radix Hedysari.Methods Totally 144 SPF Wistar rats were randomly divided into control group, model group, metacortandracin group, and Radix Hedysari polysaccharide, flavone and saponin groups were set as high-, medium-, and low-dose groups. Pulmonary interstitial fibrosis models were set up by dropping bleomycin into air tube of rats. All groups were taken corresponding medicine with appropriate dose daily on day 2 after models were established for 28 days. Collagen fibrils in lung tissue were observed by Masson dying and contents of HA and LN in lung tissue of rats in each group were detected by radioimmunoassay.Results Compared with the model group, pulmonary alveoli in Radix Hedysari flavone high-, medium-, and low-dose groups and Radix Hedysari polysaccharide low-dose group were relieved obviously; collagenous fiber area in Radix Hedysari flavone low-dose group, Radix Hedysari polysaccharide medium-dose group, and Radix Hedysari saponin low-dose group decreased obviously. Compared with the model group, the content of HA in Radix Hedysari polysaccharide high-dose group, Radix Hedysari flavone high-, medium-, and low-dose groups, and Radix Hedysari saponin high-dose group decreased obviously; the content of LN in Radix Hedysari polysaccharide high-dose group, Radix Hedysari flavone medium- and low-dose groups, and Radix Hedysari saponin medium- and low-dose groups decreased obviously. Conclusion Polysaccharide, flavone and saponin of Radix Hedysari have the abilities to alleviate inflammation of pulmonary alveoli, inhibit proliferation and deposition of collagen fibrils, and reduce the contents of HA and LA in lung tissue. Among these active ingredients, flavone has the best effect.

An HPLC method was established for the determination of adenosine, γ-aminobutyric acid (GABA) and six flavonoids (calycosin-7-glucoside, ononin, calycosin, isoliquiritigenin, formononetin and medicarpin) in Radix Hedysari. The samples were extracted with methanol by refluxing for 4 h. The HPLCDAD was performed on a Diamonsil C18 column (250 mm×4.6 mm, 5 μm) with acetonitrile-water as the mobile phase. The column temperature was at 40℃ and the flow rate was 1.0 mL·min-1, while the temperature of drift tube was 110.5℃ and the nebulizing gas flow was 3.1 L·min-1 for the ELSD system. The results showed all the eight components had good linear relationships (r2=0.992 8-1.000 0) in the range of the test concentration. The RSD of precision, stability and repeatability were less than 2%. The average recovery rates were 96.78%-103.45%, and RSD were 0.29%-1.61%. The index component contents of Radix Hedysari form 24 different origins were determined and used as variable factors in clustering analysis. The results were classified into 2 groups basically in accordance with the regional cluster. And the consequence was in consistent with the results of principal component analysis. This HPLC method is simple, shows good sensitive and accurate, and provides the experimental basis for multi-index control of Radix Hedysari. Clustering analysis for Radix Hedysari quality control has a certain reliability and objectivity.

Objective:To establish an HPLC method for the determination of calycosin-7-glucoside, genistein, formononetin and medicarpin in Radix Hedysari. Methods: The sample was refluxed with methanol in a water bath. The HPLC was performed on an SunFire C18 column(250 mm × 4. 6 mm,5 μm) with acetonitrile-0. 2% H3 PO4 as the mobile phase by gradient elution. The flow rate was 1. 0 ml·min-1 and the column temperature was at 35℃. The detection wavelength was 240 nm and the sample size was 20 μl. Results:The linear range of calycosin-7-glucoside was 0.035-1.042 μg(r=0.999 6), that of genistein was 0.027-0.821 μg(r=0. 999 7), that of formononetin was 0. 031-0. 941 μg(r=0. 999 9) and that of medicarpin was 0. 025-0. 745 μg(r=0. 999 6). The average recovery of calycosin-7-glucoside, genistein, formononetin and medicarpin was 100. 32%(RSD=1. 87%), 99. 3%(RSD=1. 76%), 100. 5%(RSD=1. 48%) and 99. 2%(RSD=1. 45%)(n=6), respectively. Conclusion:The method shows short analyt-ic time, good stability and promising operation accuracy, which provides the reference for the quality control of Radix Hedysari.

RESUMO

Objective To discuss radix hedysari flavonoids mechanism of preventing pulmonary fibrosis.Methods Totally 216 SPF Wistar rats were randomized into normal group, model group, prednisone group, radix hedysari flavonoids high, medium, and low dose groups, and then pulmonary fibrosis model was established by intratracheal dripping of bleomycin. From the second day after modeling, every treatment group received gavage with the corresponding dose of medicine at the planned time for 7, 14, and 28 continuous days. Expressions of MMP-2 and TIMP-1 protein were detected by immunohistochemical method.Results When radix hedysari flavonoids high dose group was at the 7th, 14th, 28th day, and medium dose group was at the 14th day, MMP-2 protein expression was lower than model group, similar to prednisone group. When radix hedysari flavonoids high dose group was at the 14th, 28th day and medium dose group was at 14th day, TIMP-1 protein expression was lower than model group.Conclusion Radix hedysari flavonoids can adjust the expressions of MMP-2 and TIMP-1 protein at different phases, and tend to make the balance of MMPs/TIMPs, which may be an effective mechanism for the inhibition of fibrosis process.

Objective To discuss the effects of Radix Hedysari flavonoids on microvessel density (MVD) of pulmonary interstitial fibrosis model rats, and provide evidence for its development. Methods SPF Wistar rats were divided into blank group, model group, prednisone group, and Radix Hedysari flavonoids high-, medium- and low-dosage group. Pulmonary interstitial fibrosis model was established by intratracheal instillation of bleomycin. From the second day after model established, each drug treatment group was administered with corresponding drugs intragastrically at different points in time for 14 and 28 days. MVD was detected by immunohistochemistry. Results Neomicrovessel was increased significantly in pulmonary tissue of model rats of 14 days treatment, but slightly decreased in 28 days. MVD in 14, 28 days model group was significantly more than blank group (P<0.01). MVD in 14, 28 days Radix Hedysari flavonoids high dosage group was significantly less than model group (P<0.01). MVD in Radix Hedysari flavonoids medium-dosage group was between high-and low-dosage group, and MVD in medium-dosage group was similar with that in model group. Conclusion Radix Hedysari flavonoids could inhibit the formation of neomicrovessel in a dose dependent manner.

OBJECTIVE: To isolate and purify radix hedysari polysaccharide 3 (HPS-3) and determine the monosaccharide composition and the contents of polysaccharide in its four components. METHODS: Radix Hedysari polysaccharide was extracted by water, precipitated with ethanol, treated with Sevag method, decolored with H2O2, then precipitated with ethanol and dialyzed. By using DEAE-cellulose 52 and Sephadex G-100, HPS-3-A, HPS-3-B, HPS-3-C and HPS-3-D were purified. Their purity were determined by high performance gel permeation chromatography (HPGPC). By TLC and GC, the monosaccharide composition of four components was analyzed, and then the modified phenol sulfuric acid method was used to determine the content of polysaccharide of the four fractions. RESULTS: HPS-3-A was composed mainly of glucose, and the content of polysaccharide was 99.2%. But HPS-3-B, HPS-3-C and HPS-3-D were all composed mainly of arabinose and galactose. The contents of polysaccharide of them were 96.5%, 95.3% and 95.2%, respectively. CONCLUSION: This study provides the foundation for the research of structure-function relationship of Radix Hedysari polysaccharide 3. Copyright 2012 by the Chinese Pharmaceutical Association.
