Previously, we found that the ultraviolet filter benzophenone-3 (BP3) causes fetal growth restriction in mice when is applied when implantation occurs (first week of gestation). However, whether BP3 can affect gestation and fertility after implantation period is unknown. We aimed to study the effects on reproductive physiology of the offspring caused by perinatal exposure to BP3. C57BL/6 pregnant mice were dermally exposed to 50 mg BP3/kg bw.day or olive oil (vehicle) from gestation day 9 (gd9) to postnatal day 21 (pnd1). We observed no differences in mother's weights, duration of gestation, number of pups per mother, onset of puberty or sex ratio. The weights of the pups exposed to benzophenone-3 were transiently lower than those of the control. Estrous cycle was not affected by perinatal exposure to BP3. Besides, we performed a fertility assessment by continuous breeding protocol: at 10 weeks of age, one F1 female and one F1 male mouse from each group was randomly chosen from each litter and housed together for a period of 6 months. We noticed a reduction in the number of deliveries per mother among dams exposed to BP3 during the perinatal period. To see if this decreased fertility could be associated to an early onset of oocytes depletion, we estimated the ovarian reserve of germ cells. We found reduced number of oocytes and primordial follicles in BP3. In conclusion, perinatal exposure to BP3 leads to a decline in the reproductive capacity of female mice in a continuous breeding protocol linked to oocyte depletion.

The chemical UV filter 2-ethylhexyl salicylate (EHS) is used in various personal-care products. The dermal and oral metabolism of EHS have already been targeted by different studies. However, toxicokinetic data after a single dermal exposure to EHS was missing. In our study, three volunteers were dermally exposed to a commercial EHS-containing sunscreen for 9 h with an application dose of 2 mg sunscreen per cm2 body surface area. The exposure was performed indoors, and sunscreen was applied on about 75% of the total skin area. Complete urine voids were collected over 72 h and eight blood samples were drawn from each subject. Urine samples were analyzed for EHS and seven known metabolites (5OH-EHS, 4OH-EHS, 2OH-EHS, 6OH-EHS, 4oxo-EHS, 5oxo-EHS, and 5cx-EPS) by online-SPE UPLC MS/MS. The peaks of urinary elimination occurred 10-11 h after application. The elimination half-lives (Phase 1) were between 6.6 and 9.7 h. The dominant urinary biomarkers were EHS itself, followed by 5OH-EHS, 5cx-EPS, 5oxo-EHS, and 4OH-EHS. 2OH-EHS, 6OH-EHS, and 4oxo-EHS were detected only in minor amounts. An enhanced analysis of conjugation species revealed marginal amounts of unconjugated metabolites and up to 40% share of sulfate conjugates for 5OH-EHS, 5oxo-EHS, and 5cx-EPS. The results demonstrated a delayed systemic resorption of EHS via the dermal route. Despite an extensive metabolism, the parent compound occurred as main urinary parameter. The delayed dermal resorption as well as the slow elimination of EHS indicate an accumulation up to toxicological relevant doses during daily repeated dermal application to large skin areas.

BACKGROUND: Evidence from animal models suggests a role for the organic ultraviolet filter benzophenone-3's (BP-3) on white blood cells (WBCs). However, BP-3's effect on WBCs in humans is unknown. MATERIALS AND METHODS: We used National Health and Nutrition Examination Survey data from 2003 to 2016. We included participants >6 years with data on urinary BP-3, urinary creatinine, and WBC count. Quintiles of urinary creatinine-normalized BP-3 (CnBP-3) levels were used in linear regression models adjusting for age, gender, race, body mass index (BMI), smoking status, education level, family income to poverty threshold ratio, survey cycle, and season. RESULTS: Of the 16 959 participants, 8564 (50.5%) were females, 6602 (38.9%) were White, and 3870 (22.8%) were Black. The mean (standard deviation) age was 37.6 (22.7) years, BMI was 26.8 (7.40) kg/m2, WBC count was 7.22 (2.53) × 109/L, neutrophil count was 4.15 (1.86) × 109/L, and lymphocyte count was 2.25 (1.33) × 109/L and median (interquartile range) of CnBP-3 was 12.1 (44.9) µg/gm. The highest quintile of CnBP-3 was associated with significantly lower WBC and neutrophil counts compared to the lowest quintile of CnBP-3 (Δ quintiles = -137 × 106/L, 95% CI: -249 to -24, p = 0.02 and = -177 × 106/L, 95% CI: -323 to -30, p = 0.02, respectively). In contrast, we did not observe a difference in lymphocyte count between the lowest and highest quintiles of CnBP-3 in unadjusted or adjusted analyses. CONCLUSION: We found an inverse relationship between BP-3 levels and WBC and neutrophil counts, and not with lymphocyte count. Further research is needed to confirm our findings.

Organic ultraviolet filters (OUVFs) are extensively released into aquatic environments, where they undergo complex phototransformation. However, there is little knowledge regarding their transformation products (TPs) and associated endocrine disruption potentials. In the present study, we characterized the chemical and toxicological profiles of TPs for two common OUVFs, oxybenzone (BP3) and ethylhexyl methoxycinnamate (EHMC), by photooxidation under environmentally relevant conditions. It is hypothesized that TPs of the tested OUVFs will show varied estrogenicity at different reaction times. High-resolution liquid chromatography quadrupole time-of-flight mass spectrometry (LC-QTOF-MS) identified 17 TPs of 7 m/z for BP-3 and 13 TPs of 8 m/z for EHMC at confidence levels ≤2. Five novel TPs of 2 m/z were reported for the first time with structure-diagnostic MS/MS spectra. Estrogenicity assessment using the MCF-7-luc cell line showed discrepant estrogenic activities exhibited by OUVF-TPs over time. Specifically, BP3-TPs exhibited significantly greater estrogenicity than the parent at several reaction times, whereas EHMC-TPs displayed fluctuating estrogenicity with a declining trend. Correlation analysis coupled with molecular docking simulations further suggested several TPs of BP3 as potential endocrine disruptive compounds. These findings underscore the necessity of considering mixtures during chemical testing and risk assessment and highlight the potentially greater risks associated with post-transformation cocktails.

We present a case study that provides a practical step-by-step example of how the internal Threshold of Toxicological Concern (iTTC) can be used as a tool to refine a TTC-based assessment for dermal exposures to consumer products. The case study uses a theoretical scenario where there are no systemic toxicity data for the case study chemicals (avobenzone, oxybenzone, octocrylene, homosalate, octisalate, octinoxate, and ecamsule). Human dermal pharmacokinetic data following single and repeat dermal exposure to products containing the case study chemicals were obtained from data published by the US FDA. The clinical studies utilized an application procedure that followed maximal use conditions (product applied as 2 mg/cm2 to 75% of the body surface area, 4 times a day). The case study chemicals were first reviewed to determine if they were in the applicability domain of the iTTC, and then, the human plasma concentrations were compared to an iTTC limit of 1 µM. When assessed under maximum usage, the external exposure of all chemicals exceeded the external dose TTC limits. By contrast, the internal exposure to all chemicals, except oxybenzone, was an order of magnitude lower than the 1 µM interim iTTC threshold. This work highlights the importance of understanding internal exposure relative to external dose and how the iTTC can be a valuable tool for assessing low-level internal exposures; additionally, the work demonstrates how to use an iTTC, and highlights considerations and refinement opportunities for the approach.

Protection against sunburn, skin damage and the carcinogenic effects of ultraviolet light are the primary health benefits associated with UV filters used in topical sunscreen drug products. Countries such as Europe have 30+ UV filters approved for sunscreen products while the US has about 10, greatly reducing the options to provide diverse, effective sun protection products. Bemotrizinol (BEMT) is the first new sunscreen active ingredient to be evaluated for inclusion in the Over-The-Counter (OTC) sunscreen monograph using FDA's new Generally Recognized as Safe and Effective (GRASE) testing guidelines. An in vitro skin permeation test (IVPT) and clinical pilot pharmacokinetic Maximum Usage Trial (MUsT) were completed to support the GRASE determination for 6% BEMT. IVPT results indicated an oil +10% ethanol as the model sunscreen intervention for the pilot MUsT. The open-label trial revealed: BEMT concentrations rarely exceeded FDA's defined threshold (0.5 ng/mL) in plasma; no evidence for BEMT accumulation or steady-state concentrations above threshold; only one moderate and few mild treatment emergent adverse events (TEAEs). Therefore, maximal topical applications of 6% BEMT in a model sunscreen formulation did not contribute to meaningful systemic exposure. These results support the safety of BEMT 6% for human sunscreen use.

Due to recent global warming threats, the changes in the atmosphere have caused significant ultraviolet (UV) radiation exposure, primarily emitted by the sun, which creates more awareness of photoprotection. Sunscreen development has been a convenient and crucial approach to photoprotection against ultraviolet radiation. Due to high demand, upgrading the quality of sunscreen products and certifying methods are necessary to guarantee the safety of commercial sunscreen products for use. Sunscreen products should have a satisfactory amount of sun protection factor (SPF), ultraviolet A protection factor, as well as the photostability of the sunscreens for them to be considered effective and safe for use. A rigorous study on the effectiveness of the sunscreen components and their safety standards is essential for the productive use and further improvement of the available sunscreen materials. This article summarizes the effects and issues, protective measures of sunscreen usage, and its components, mainly ultraviolet filters.

Skin cancer continues to increase in incidence year-on-year and represents the most common form of cancer across the globe. Every human undergoes premature ageing, particularly on the face, neck and hands. Both phenomena are driven primarily by chronic, daily exposure to solar ultraviolet radiation (UVR). While sunscreen products play a primary role in the prevention of UVR skin damage, the active ingredients, i.e., UVR filters, are facing unprecedented challenges in the coming 10 years and their future is by no means certain. This article, therefore, reviews afresh the facts around photoprotection and the role of sunscreen products in the prevention of acute (sunburn) and chronic (cancer, photoageing) skin damage and compares/contrasts these with various emerging questions and opinions around UVR filter technology. We present a passionate defence of this remarkable technology, but also attempt to imagine a world without it.
L'incidence du cancer de la peau continue d'augmenter année après année, et ce cancer est le plus fréquent dans le monde. Tous les êtres humains connaissent un vieillissement prématuré, en particulier au niveau du visage, du cou et des mains. Les deux phénomènes sont causés principalement par une exposition quotidienne chronique aux rayons ultraviolets (UVR) du soleil. Bien que les protections solaires jouent un rôle essentiel dans la prévention des lésions cutanées dues aux UVR, les principes actifs, c'est-à-dire les filtres UVR, seront confrontés à des difficultés sans précédent dans les 10 prochaines années, et leur avenir n'est en aucun cas certain. Cet article examine donc les faits concernant la photoprotection et le rôle des produits de protection solaire dans la prévention des lésions cutanées aiguës (coups de soleil) et chroniques (cancer, photovieillissement) ; et les compare/oppose aux différentes questions et opinions émergentes concernant la technologie des filtres UVR. Nous présentons une défense passionnée de cette technologie remarquable, mais nous essayons également d'imaginer un monde sans elle.

OBJECTIVE: A 2-year sampling campaign was realized on French Mediterranean beach (Palavas-les-Flots Hérault) in order to measure the concentration of UV filters released from the sunscreen used by bathers. Multiple factors suspected of playing determining roles in the UV filter pattern in water were explored, such as the seasonal and daily time evolutions, or the vertical and horizontal distributions, and they were regarded through the UV filter characteristics. METHODS: The beach was monitored during periods of high and low tourist attendance, typically before, during and after the summer peak. The beachgoers attendance was counted. Bathing water was sampled distinctly from the bulk column and from the top surface layer, testing different sampling tools. Sediments and mussels were also sampled and analysed as potential UV filter sinks. Three organic UV filters (octocrylene OCR, avobenzone BMDBM and octyl methoxycinnamate OMC) and one mineral (titanium dioxide TiO2 ) were studied here as representatives of the current cosmetic market. RESULTS: Summer peak attendance on the beach was confirmed associated with peak levels of UV filter concentration in the bathing water, even more pronounced during a heat wave period. This relation was also observed at day scale with an afternoon peak, suggesting a rapid evolution of the UV filter pattern in water. Contrasted fates were measured between the four studied UV filters, that could be mainly explained by their respective characteristics, i.e. particulate or dissolved, hydrophilic or lipophilic, lifetime. Generally, this resulted in a concentration ranking TiO2 > OCR > OMC > BMDBM, ranging from 0.5 to 500 µg/L. The most lipophilic and recalcitrant OCR was found most vertically differentiated and over concentrated in the top surface layer of water. Finally, a large horizontal heterogeneity was also observed in the UV filter concentration pattern, raising the need for sample replicates that cover a significant area. CONCLUSION: This work fulfils some knowledge gaps on the issue of UV filter release in coastal environments, not only by providing original field data and methodological recommendations but also importantly in the comparison made of organic and mineral UV filters, which are often considered separately and rarely evaluated at the same time.
OBJECTIF: Une campagne d'échantillonnage de deux ans a eu lieu sur une plage de la Méditerranée en France (Palavas-les-Flots dans l'Hérault) afin de mesurer la concentration de filtres UV libérés par la protection solaire utilisé par les baigneurs. Plusieurs facteurs suspectés de jouer des rôles déterminants dans le modèle de filtre UV dans l'eau ont été étudiés, comme les évolutions saisonnières et quotidiennes, ou les distributions verticales et horizontales, et ils ont été examinés à travers les caractéristiques du filtre UV. MÉTHODES: La plage a été surveillée pendant les périodes de fréquentation touristique élevée et faible, généralement avant, pendant et après le pic estival. La présence des baigneurs a été comptabilisée. L'eau de baignade a été prélevée distinctement de la colonne principale et de la couche superficielle supérieure, en testant différents outils de prélèvement d'échantillons. Des sédiments et des moules ont également été prélevés et analysés comme réservoirs de filtre UV potentiels. En l'occurrence, trois filtres UV organiques (octocrylène OCR, avobenzone BMDBM et octyl méthoxycinnamate OMC) et un minéral (dioxyde de titane TiO2 ) ont été étudiés comme représentants du marché cosmétique actuel. RÉSULTATS: Les pics estivaux de présence de baigneurs sur la plage ont été confirmés comme étant associés à des pics de concentration du filtre UV dans l'eau de baignade, encore plus prononcés pendant une période de vague de chaleur. Cette relation a également été observée à l'échelle d'une journée, avec un pic l'après-midi, suggérant une évolution rapide du profil de filtre UV dans l'eau. Les effets de contraste ont été mesurés entre les quatre filtres UV étudiés, ce qui pourrait s'expliquer principalement par leurs caractéristiques respectives, c'est-à-dire particulaires ou dissous, hydrophiles ou lipophiles, tout au long de la vie. En général, cela a donné lieu à un classement de la concentration : TiO2 > OCR > OMC > BMDBM, comprise entre 0,5 et 500 ug/L. Il est apparu que c'est l'OCR le plus lipophile et le plus récalcitrant qui est le plus différencié verticalement et sur-concentré dans la couche supérieure de l'eau. Enfin, on a observé une grande hétérogénéité horizontale dans le profil de concentration du filtre UV, ce qui a nécessité des réplicats d'échantillons couvrant une zone significative. CONCLUSION: Ce travail comble certaines lacunes en matière de connaissances sur la libération des filtres UV dans les environnements côtiers, non seulement en fournissant des données originales sur le terrain et des recommandations méthodologiques, mais également en comparant des filtres UV organiques et minéraux, qui sont souvent pris en compte séparément et rarement évalués en même temps.

Present in an increasing number of products, UV-filters are continuously discharged into aquatic environments. Despite potential risks for inhabiting organisms are recognized, the effects of UV-filter 4-methylbenzylidenecamphor (4-MBC) on marine invertebrates are poorly investigated. By combining in vitro/in vivo exposures through a multi-biomarker approach on sperms and adults, the present study evaluated how 4-MBC affect the mussel species Mytilus galloprovincialis, providing ecologically relevant information on organisms' responses. From the obtained results, considering mortality as endpoint, sperms revealed a greater sensitivity (EC50:347 µg/L) than adults (EC50: not calculable). From an ecotoxicological perspective, this resulted in a derived threshold concentration (LOEC) of 100 µg/L and 72 µg/L, respectively. Effects at the cell/molecular level were provided by general redox-status imbalance and oxidative stress. Sperms showed functional and structural impairments, hyperactivation and DNA damage, while adults showed physiological, metabolic/energetic dysfunctions, DNA damage and activation of oxidative and biotransformation enzymes. High 4-MBC bioaccumulation was also observed in exposed mussels (BCFs:14.0-32.0 L/kg). These findings suggest that 4-MBC may impair fitness and survival of the broadcast spawning mussel M. galloprovincialis, affecting reproduction success and population growth.

Tyrosinase is a key metalloenzyme for the biosynthesis of melanin that plays a critical role in the prevention of skin damage caused by ultraviolet (UV) radiation. However, the overproduction of melanin may cause a variety of skin diseases. Due to the toxicity and inefficiency of existing tyrosinase inhibitors, it is urgent to identify safe and potent alternatives from natural sources. Theaflavin, a single-component extracted from black tea, has been found to possess a variety of pharmacological activities. Herein, the inhibition kinetics of theaflavin on tyrosinase and inhibitory mechanism were determined using spectroscopy, molecular docking, and zebrafish model. The results showed that theaflavin inhibited the diphenolase activity of tyrosinase in a reversible mixed type manner with IC50 of 229.75 µmol/L and hindered the synthesis of melanin in zebrafish. This may be due to the formation of eight hydrogen bonds and hydrophobic effects between theaflavin and tyrosinase according to the results of molecular docking. To study the possible effects on the prevention of free radical-mediated skin cancer and photoaging caused by UV radiation, the antioxidation and UV filter properties of theaflavin were further verified. This study demonstrates that theaflavin is a potential multifunctional compound that can be used in cosmetic and medicinal products.

A current trend of research in the health field is toward the discovery of multifunctional compounds, capable of interacting with multiple biological targets, thus simplifying multidrug therapies and improving patient compliance. The aim of this work was to synthesize new multifunctional chemical entities bearing a benzothiazole nucleus, a structure that has attracted increasing interest for the great variety of biological actions that it can perform, and already used as a scaffold in several multifunctional drugs. Compounds are reported, divided into two distinct series, synthetized and tested in vitro for the antioxidant, and include UV-filtering and antitumor activities. DPPH and FRAP tests were chosen to outline an antioxidant activity profile against different radical species. The UV-filtering activity was investigated, pre- and post-irradiation, through evaluation of a O/W sunscreen standard formulation containing 3% of the synthetic compounds. The antitumor activity was investigated both on human melanoma cells (Colo-38) and on immortalized human keratinocytes as a control (HaCat). A good antiproliferative profile in terms of IC50 was chosen as a mandatory condition to further investigate apoptosis induction as a possible cytotoxicity mechanism through the Annexin V test. Compound BZTcin4 was endowed with excellent activity and a selectivity profile towards Colo-38, supported by a good antioxidant capacity and an excellent broad-spectrum photoprotective profile.

The cassava root is an important global agro-industrial crop that yields cassava leaf as a left-over co-product of interest for further development as a sustainable resource of health and cosmeceutical active compounds. This work aimed to investigate the cosmeceutical potential and chemical composition of an ethanolic cassava leaf extract (BM). rutin, apigenin, and kaempferol were found to be major constituents via HPLC-DAD UV analysis. Interestingly, the multiple beneficial bioactivities of BM for cosmeceutical applications were manifested in a dose-dependent manner, including anti-oxidation in a 2,2-diphenyl-1-picrylhydrazyl assay, anti-melanogenesis in B16 melanoma cells, collagen synthesis enhancement in human fibroblasts, and anti-adipogenesis in 3T3-L1 adipocytes. Furthermore, the potential of the collagen synthesis enhancement of BM and rutin was significant when compared to ascorbic acid. Additionally, a UV filter property comparable to BEMT with characteristics of board spectral absorption and constant high absorptivity throughout all UV wavelength ranges was exhibited by UV-visible spectrophotometric analysis. In conclusion, the cassava leaf was found to be a potential natural cosmeceutical active agent with multiple cosmeceutical-related bioactivities with respect to a substantial composition of bioactive flavonols. These obtained data will support and encourage the further study and development of cassava leaves as potential economic and sustainable sources of bioactive agents for health and cosmeceutical applications.

Sunscreen products containing inorganic micronized titanium dioxide (TiO2) and zinc oxide (ZnO) have been available since the 1950s. Their cosmetic acceptance remained limited as they persist as a white paste on the skin. By reducing the size of the particles into the nano-range below 100â¯nm, their optical property of reflecting visible light is reduced. After the year 2000, organic filters of this size range were developed. The enthusiasm for nanotechnology that prevailed at the time did not transfer to sunscreen products with nanoparticulate filters. Consumers suspect that the particles permeate the skin, are absorbed by the blood, and spread throughout the body causing illness. Not least due to public pressure, cosmetics-which include sunscreen products-became the first product segment in which accordingly manufactured substances were subjected to strict regulations. Despite advanced regulation and strict approval procedures for nanoparticulate filters, public reservations remained. Possible reasons for this are lack of knowledge or mistrust of the applicable legislation, unclear perception of the behavior of nanoparticles in sunscreen products and as a result unclear perceptions of hazard, risk, and exposure. Against this background, the nature and behavior of nanoparticulate filters in sunscreens on the skin and potentially in the skin, as well as the regulatory framework that ensure that nanoparticulate filters and the products containing them are safe to use are discussed.

The UV filter 4-methylbenzylidene camphor (4-MBC), used in cosmetics, the antioxidant butylated hydroxytoluene (BHT), used inter alia as a food additive and in cosmetics, and the plasticizer tris(2-ethylhexyl) trimellitate (TOTM), used mainly in medical devices as substitute for di-(2-ethylhexyl) phthalate (DEHP), are suspected to have endocrine disrupting effects. Human biomonitoring methods that allow for assessing the internal exposure of the general population to these substances were recently developed in a German cooperation to enhance the use of human biomonitoring. First-morning void urine samples from 3- to 17-year-old children and adolescents living in Germany were analysed for metabolites of 4-MBC (N = 447), BHT (N = 2091), and TOTM (N = 431) in the population-representative German Environmental Survey on Children and Adolescents 2014-2017 (GerES V). 4-MBC metabolites were found in quantifiable amounts only in single cases and exposure levels remained well below health-based guidance values. In contrast, ubiquitous exposure to BHT became evident with a geometric mean (GM) urinary concentration of the metabolite BHT acid of 2.346 µg/L (1.989 µg/gcreatinine) and a maximum concentration of 248 µg/L (269 µg/gcrea). The highest GM concentration was found in young children aged 3-5 years, yet no specific sources of exposure could be identified. Also, TOTM metabolites were found in quantifiable amounts only in very few samples. None of these findings could be related to previous hospital treatment or exposure via house dust. The presented results will be the basis to derive reference values for exposure of children and adolescents in Germany to BHT and will facilitate to identify changing exposure levels in the general population.

Plants, as with humans, require photoprotection against the potentially damaging effects of overexposure to ultraviolet (UV) radiation. Previously, sinapoyl malate (SM) was identified as the photoprotective agent in thale cress. Here, we seek to identify the photoprotective agent in a similar plant, garden cress, which is currently used in the skincare product Detoxophane nc. To achieve this, we explore the photodynamics of both the garden cress sprout extract and Detoxophane nc with femtosecond transient electronic absorption spectroscopy. With the assistance of liquid chromatography-mass spectrometry, we determine that the main UV-absorbing compound in garden cress sprout extract is SM. Importantly, our studies reveal that the photoprotection properties of the SM in the garden cress sprout extract present in Detoxophane nc are not compromised by the formulation environment. The result suggests that Detoxophane nc containing the garden cress sprout extract may offer additional photoprotection to the end user in the form of a UV filter booster.

UV-filters are widely used in cosmetics and personal care products to protect users' skin from redamage caused by ultraviolet (UV) radiation from the sun. Globally, an estimated 16,000 to 25,000 tonnes of products containing UV-filters were used in 2014 with modern consumption likely to be much higher. Beyond this use in cosmetics and personal care products, UV-filters are also widely used to provide UV-stability in industrial products such as paints and plastics. This review discusses the main routes by which UV-filters enter aquatic environments and summarises the conclusions of studies from the past 10 years that have investigated the effects of UV-filters on environmentally relevant species including corals, microalgae, fish, and marine mammals. Safety data regarding the potential impact of UV-filters on human health are also discussed. Finally, we explore the challenges surrounding UV-filter removal and research on more environmentally friendly alternatives to current UV-filters.

Oxidative stress is the product or aetiology of various multifactorial diseases; on the other hand, the development of multifunctional compounds is a recognized strategy for the control of complex diseases. To this end, a series of benzothiazole derivatives was synthesized and evaluated for their multifunctional effectiveness as antioxidant, sunscreen (filter), antifungal and antiproliferative agents. Compounds were easily synthesized via condensation reaction between 2-aminothiophenols and different benzaldehydes. SAR study, particularly in position 2 and 6 of benzothiazoles, led to the identification of 4g and 4k as very interesting potential compounds for the design of multifunctional drugs. In particular, compound 4g is the best blocker of hERG potassium channels expressed in HEK 293 cells exhibiting 60.32% inhibition with IC50 = 4.79 µM.

Emerging contaminants such as synthetic musks and UV-filters as ingredients personal care products were widely used in human daily life in Thailand. The occurrence and fate of four synthetic musks and nine UV-filters were investigated in eight full-scale sewage treatment plants (STPs) and their receiving aquatic environments in Bangkok and Pattaya, Thailand. All target compounds were detected in every single sample from STPs and surface water with magnitude from ng/L to µg/L. HHCB-lactone and HMS were found as the predominant musk and UV filter in influent and effluent of STPs, respectively. HHCB-lactone was also found with the highest concentration up to 79501 ng/g (dw) in the sludge. Low removal efficiency range from -37% (HHCB-lactone) to 58% (AHTN) were found for four musks in the STPs. The total emission of Σ4musks and Σ9UV-filters were estimated to be up to 16.7 mg/person/day and 0.28 mg/person/day by the STPs. Three synthetic musks and seven UV-filters were detected in fish from the receiving river. Concentration and emission of musks and UV filters found in this study from Thailand were much higher than those reported in many other countries worldwide. The preliminary ecological risk assessment showed that Musk xylene, 4-MBC and OC may pose high risk to aquatic organisms in the riverine and estuarine environment in Thailand.

In the search for scaffolds for multifunctional compounds we investigated the structure activity relationship of a class of benzimidazole derivatives bearing 5-membered ring. The newly synthesized and the already known compounds were divided into three classes that present different substituent at 5 position of the benzimidazole ring (-H, -COOH or -SO3H) and different heterocycle at position 2 (thiophene, furan or pyrrole). All the derivatives were synthesized and tested to determine their photoprotective profile against UV rays, in vitro antioxidant capacity against different radicals (DPPH and FRAP test), antifungal inhibitory activity (dermatophytes and Candida albicans), antiviral and antiproliferative activity. A Structure-Activity Relationship study indicated compound 10, bearing a pyrrole heterocycle on the benzimidazole ring, as the best multifunctional derivative of the series and as potential candidate for the development of drugs especially in case of melanoma.
