RESUMO

eIF4G is an important eukaryotic translation initiation factor. In this study, eIF4G1, one of the eIF4G isoforms, was shown to directly participate in biogenesis of the large (60S) ribosomal subunit in Saccharomyces cerevisiae cells. Mutation of eIF4G1 decreased the amount 60S ribosomal subunits significantly. The C-terminal fragment of eIF4G1 could complement the function in 60S biogenesis. Analyses of its purified complex with mass spectrometry indicated that eIF4G1 associated with the pre-60S form directly. Strong genetic and direct protein-protein interactions were observed between eIF4G1 and Ssf1 protein. Upon deletion of eIF4G1, Ssf1, Rrp15, Rrp14 and Mak16 were abnormally retained on the pre-60S complex. This purturbed the loading of Arx1 and eL31 at the polypeptide exit tunnel (PET) site and the transition to a Nog2 complex. Our data indicate that eIF4G1 is important in facilitating PET maturation and 27S processing correctly. This article has an associated First Person interview with the first author of the paper.

INTRODUCTION: The aim of this study was to compare the biological effects of four chemical caries removal materials and to assess their cytotoxicity using human dental pulp stem cells (hDPSCs). METHODS: The products evaluated are: 1 - papain-based product (BRIX 3000®); 2 - papain/chloramine based products (NATURAL-CARE and Papacárie Duo®); and 3 - chloramine based product (Cariesolut). The following in vitro experiments were carried out: IC50 measurement, cell metabolic activity (MTT) assay, cell migration, immunofluorescence experiment, cell apoptosis analysis, and reactive oxygen species (ROS) production analysis. Statistical analyses were performed using one-way ANOVA followed by Tukey's post hoc test (p < 0.05). RESULTS: The IC50 values were: Brix 3000: 0.596%; Papacárie Duo: 0.052%; NATURAL CARE: 1.034%; and Cariesolut: 0.020%. The MTT assays showed non-adequate cell viability of all chemical-mechanical caries removal tested at 2% at 24, 48, and 72 h (p < 0.001). The same behaviour was observed at 0.1% in the Papacárie Duo and Cariesolut groups. In contrast, 0.1% of Brix 3000 at all times and NATURAL CARE at 24 h treated cells showed cell viability rates similar to the control group. At 0.01% only Brix 3000 did not show statistically significant differences at any time. Delayed cell migration was observed in all hDPSCs treated with Papacárie Duo and Cariesolut (p < 0.01 and p < 0.001). Phalloidin staining images showed a high confluence of cells in the presence of NATURAL CARE, similar to the control group. On the contrary, no cells were observed in Brix 3000 and Cariesolut at 2% and 0.1% concentrations. Papacárie Duo showed cells at all concentrations, but hDPSCs treated at 0.01% concentration exhibited better proliferation and spreading than those in the control group. Apoptosis essay showed that Brix 3000 at both 0.1% and 0.01% had a percentage of live cells higher than 99%, with 68.4% live cells at 2%, 3.69% early apoptotic cells, and 27.9% late apoptotic cells. Conversely, the rest of the materials showed an abundance of apoptotic cells, even at low concentrations. 0.1% and 0.01% of BRIX 3000 did not affect the ROS production levels, while 2% of BRIX 3000 counterpart very significantly increased the percentage of CM-H2DCFDA positive cells. Again, all concentrations of Cariesolut showed significantly higher levels of ROS production than those observed in control cells. CONCLUSION: Our results suggest that Brix 3000 would be the most suitable material for chemical caries removal, with Papacárie Duo and NATURAL CARE also being good options, and discourage the use of Cariesolut due to its low cytocompatibility on dental pulp stem cells.

The prepartum diet as well as individual metabolic status of the cow influences colostrum parameters. The objectives of this study were to 1) investigate the effect of increasing prepartum dietary MP supply on colostrum yield, composition, and immunoglobulin G (IgG) concentration, and 2) identify prepartum metabolic indicators associated with these outcomes. Multiparous Holstein cows (n = 96) were blocked by expected calving date and randomly assigned to 1 of 2 prepartum diets formulated to contain a control (CON; 85 g of MP/kg DM; 1,175 g of MP/d) or high (HI; 113 g of MP/kg DM; 1,603 g of MP/d) level of MP starting at 28 d before expected calving. Both prepartum diets were formulated to supply Met and Lys at an equal amount of 1.24 and 3.84 g/Mcal of metabolizable energy (ME), respectively. Metabolic indicators were determined in serum (albumin, glutamate dehydrogenase, cholesterol, aspartate transaminase, total protein, total bilirubin, and IgG) or plasma (Ca, glucose, fatty acids, BHB, and urea nitrogen) twice weekly in a subset of cows (n = 60). Colostrum was harvested at 3.6 ± 2.4 h from calving and yield as well as concentrations of IgG, fat, protein, and Ca were determined. Cows were retrospectively grouped based on the typical volume of colostrum needed for 2 colostrum meals (<6 or ≥ 6 kg), IgG concentration (<100 or ≥ 100 g/L), as well as the median concentrations of fat (<4.4 or ≥ 4.4%), protein (<16.5 or ≥ 16.5%), Ca (<0.21 or ≥ 0.21%), and total colostrum ME (<8.65 or ≥ 8.65 Mcal). Data were analyzed using mixed effects ANOVA, with repeated measures where applicable. Feeding HI tended to increase colostrum yield in cows entering parity 2 (9.4 vs. 7.2 ± 0.9 kg), but treatment did not affect yield from cows entering parity ≥3 (5.1 vs. 6.4 ± 1.0 kg). Supply of MP did not affect concentrations of IgG, fat, protein, or Ca. Cows that produced ≥ 6 kg vs. those producing <6 kg of colostrum had lower plasma concentrations of glucose. Metabolic indicators were not associated with IgG group. Colostrum fat ≥4.4% was associated with cows having lower prepartum concentrations of glucose, total protein, albumin, and aspartate transaminase activity. Colostrum protein ≥ 16.5% was associated with lower circulating serum IgG and elevated cholesterol. Elevated glucose as well as lower cholesterol and BHB concentrations were associated with colostrum Ca ≥ 0.21%. Further, higher albumin and fatty acids as well as lower glucose concentrations were associated with a greater colostrum energy output. In conclusion, increasing prepartum MP supply tended to increase colostrum yield in cows entering parity 2, but did not affect the composition or IgG concentration. The observed associations between metabolic indicators and colostrum parameters suggest that slight adjustment in metabolism during late gestation might be necessary to support colostrogenesis, but the causality of these relationships should be considered.

BACKGROUND: Industrial starch hydrolysis allows the production of syrups with varying functionality depending on their Brix value and dextrose equivalent (DE). As the current methods for evaluating these products are labor-intensive and time-consuming, the objective of this study was to investigate the potential of near-infrared (NIR) spectroscopy for classifying the different tapioca starch hydrolysis products. RESULTS: NIR spectra of samples of seven products (n = 410) were recorded in transflectance mode in the 12 000-4000 cm-1 range. Next, orthogonal partial least squares (OPLS) regression models were built to predict the Brix and DE values of the different samples. To classify the different starch hydrolysis products, support vector machines (SVM) were trained using either the raw spectra or latent variables (LVs) obtained from the OPLS models. The best classification accuracy was obtained by the SVM classifier based on the LVs from the OPLS model for DE prediction, resulting in 95% correct classification over all classes. CONCLUSION: These results show the potential of NIR spectroscopy for classifying tapioca starch hydrolysis products with respect to their functional properties related to the Brix and DE values. © 2024 Society of Chemical Industry.

The strategy for controlling the existence of radionuclides in drinking water depends upon an individual dose criterion (IDC) of 0.1 mSv/y, which represents a very low level of risk that is not expected to cause any identified adverse health effects. Radon gas, considered a carcinogenic radionuclide, can dissolve and accumulate in drinking water. Non-alcoholic carbonated beverages (NACBs), which mainly contain drinking water, phosphoric acid, citric acid, caffeine, and sugar, represent one of the most consumed groups worldwide and in Türkiye. In this study, the radon activity concentration and some physicochemical characteristics of 45 NACB samples from 24 most preferred commercial brands in Türkiye were determined to assess the radiological health risk associated with the ingestion of these samples. Radon activity concentrations measured in NACB samples using the AlphaGUARD radon analyzer ranged from 22.8 ± 0.7 to 54.9 ± 1.7 mBq/L. The pH, conductivity, total dissolved solids, and brix values in NACB samples ranged from 2.31 to 7.29, 401 to 3281 µSv/cm, 355 to 2453 mg/L, and 0.10 to 12.95%, respectively. Total (ingestion and inhalation) annual effective doses and the corresponding excess lifetime cancer risks estimated for adults to assess the radiological health risk are significantly below the IDC and advised safety limit (10-3), respectively.

AIMS: To investigate the effect of preservation by addition of yoghurt starter, potassium sorbate and citric acid on counts of aerobic bacteria, Lactobacillus spp., Streptococcus thermophilus and coliforms, Brix percentage, pH, protein, fat and anhydrous lactose concentrations at 0, 7 and 14 days after collection for colostrum stored at ambient temperature. METHOD: Approximately 2 L of first milking colostrum was collected from 10 farms in the Waikato region. Following mixing, it was split into five 400-mL sub-samples and allocated randomly to a control (two sub-samples), or treatment with yoghurt, potassium sorbate, or citric acid preservative. Throughout the trial samples remained in the laboratory at ambient temperature with the lids slightly ajar, and were stirred daily for 15-30 seconds using a sterile spatula. Sub-samples were tested on Days 0, 7 and 14. On Days 0 and 14 aerobic bacteria (by aerobic plate count (APC)), Lactobacillus spp., coliforms and Streptococcus thermophilus counts, pH, Brix percentage, protein, fat and anhydrous lactose were measured. On Day 7 only bacterial counts were completed.The data were analysed using non-parametric clustered bootstrap sampling to estimate the effect of treatment, time, and their interaction on the outcome variables. RESULTS: Compared to control samples, on Day 7 the APC for potassium sorbate (1.0 (90% CI = 0.6-1.6) × 108 cfu/mL) was approximately seven-fold lower than for yoghurt (7.3 (90% CI = 4.1-11) × 108 cfu/mL), and approximately three-fold lower than citric acid (3.2 (90% CI = 0.2-4.3) × 108 cfu/mL) remaining low to Day 14. All preservatives reduced coliform growth compared to control samples at Day 7 but growth was lower for potassium sorbate than the other preservatives. For Lactobacillus spp., at Day 7, samples with yoghurt preservative had greater counts than the other two preservatives. Potassium sorbate reduced growth of S. thermophilus compared to the other treatments, especially at Day 7, with 7-10 times fewer S. thermophilus per mL compared to the other three groups. All groups showed an obvious acidification over time, with very little variation within days and treatment groups. There was no evidence for change in fat or protein percentage over time regardless of treatment. CONCLUSION AND CLINICAL RELEVANCE: Aerobic and coliform bacteria proliferate extensively in unpreserved colostrum. All preservatives decreased coliform counts compared to un-preserved colostrum, but potassium sorbate was more effective at decreasing both coliforms and aerobic bacteria than either yoghurt or citric acid.

In this study, we aimed to determine the quality of colostrum in sheep by using Brix refractometer. The research included 100 sheep of Merino X Kivircik crossbred. From each, we collected 15 mL of colostrum samples in falcon tubes within the first 8 h after delivery. Mean colostral IgG level of sheep was 156.68 ± 7.23 g L-1, optical and digital Brix refractometer values (%) were determined as 27.43 ± 0.53 and 27.69 ± 0.60, respectively. Ewes carrying twin lambs produced significantly higher quality colostrum than those carrying single lambs. However, parity did not affect the colostrum quality. Optical and digital Brix values were correlated with gold standard radial immunodiffusion (RID) colostral IgG level (r = 0.70 and r = 0.64, respectively). Also, optical and digital Brix refractometers were found to be highly correlated (r = 0.98, P < 0.001). While the optimal Brix value was 22% for the 50, 60 and 70 g L-1 IgG threshold values (by means of RID as the potential good quality threshold value for ewe colostrum quality), this value was 23% for 80 g L-1. We can conclude that Brix refractometers is a valuable tool for determining ewe colostrum quality. A cut point of 22% Brix for defining good quality colostrum in ewes was most appropriate for our data.

Colostrum yield and quality are influenced by prepartum nutrition and the metabolic status of the cow; however, data considering these associations on multiple dairy farms are limited. Our objective was to identify cow-level prepartum metabolic indicators, as well as farm-level nutritional strategies associated with colostrum yield and the indicator of colostrum quality, Brix %. A convenience sample of 19 New York Holstein dairies (median: 1,325 cows; range: 620 to 4,600 cows) were enrolled in this observational study. Records for individual colostrum yield and Brix % were collected by farm personnel between October 2019 and February 2021. Farms were visited 4 times, approximately 3 mo apart, to obtain feed samples of the prepartum diets, collect blood samples from 24 pre- and postpartum cows, respectively, and determine prepartum body condition score. Feed samples were submitted for analysis of chemical composition, and particle size was determined on-farm using a particle separator. Prepartum serum samples (n = 762) were analyzed for glucose and nonesterified fatty acid concentrations. Whole blood from postpartum cows was analyzed for herd prevalence of hyperketonemia (proportion of samples with ß-hydroxybutyrate ≥1.2 mmol/L). A cohort of primiparous (PP; n = 1,337) and multiparous (MPS; n = 3,059) cows calving ± 14 d of each farm visit were included in the statistical analysis. Animals calving in this period were assigned results for the close-up diet composition and herd prevalence of hyperketonemia collected from the respective farm visit. Greatest colostrum yield from PP and MPS cows was associated with moderate starch [18.6-22.5% of dry matter (DM)] and a moderate herd prevalence of hyperketonemia (10.1-15.0%). Greatest colostrum yield from MPS cows was associated with moderate crude protein (13.6-15.5% of DM) and a less severe negative dietary cation-anion difference (DCAD; >-8 mEq/100 g), whereas greatest colostrum yield from PP cows was associated with low crude protein (≤13.5% of DM). In addition, a moderate proportion of the diet with particle length ≥19 mm (15.3-19.1%) was associated with lowest colostrum yield from PP and MPS cows. Highest colostrum Brix % was associated with prepartum dietary factors of low neutral detergent fiber (≤39.0% of DM) and high proportion of the diet with particle length ≥19 mm (>19.1%). In addition, low starch (≤18.5% of DM) and low and moderate DCAD level (≥-15.9 mEq/100 g) were associated with greatest Brix % from PP cows, whereas moderate DCAD (-15.9 to -8.0 mEq/100 g) was associated with greatest Brix % from MPS cows. Prepartum serum nonesterified fatty acid concentration ≥290 µEq/L was associated with increased colostrum yield, but prepartum serum glucose concentration and body condition score were not associated with colostrum yield or Brix %. These data provide nutritional and metabolic variables to consider when troubleshooting colostrum production on farms.

Adequate supply of high-quality colostrum is essential for calf health. Colostrum production, at first milking, varies between animals and seasons, but herd-level and management associations with colostrum production have not been well described. Our objectives were to (1) describe colostrum production and colostrum handling practices and (2) to identify individual cow, herd management, and environmental factors associated with colostrum production. A convenience sample of 19 New York Holstein dairy farms (620 to 4,600 cows) were enrolled in this observational study to describe colostrum production and to evaluate cow, management, and prepartum environmental factors associated with colostrum yield and Brix %. Herd owners or managers were given a colostrum management questionnaire, and farm personnel recorded individual colostrum yield and Brix % for primiparous (PP; n = 5,978) and multiparous (MPS; n = 13,228) cows between October 2019 and February 2021. Temperature, relative humidity, and light intensity were measured by sensors placed in each farm's close-up dry cow pens for the entire length of the study. Median colostrum yield for each farm ranged from 2.5 to 7.6 kg for PP and 4.0 to 7.7 kg for MPS cows. Mean Brix % from each farm ranged from 22.2 to 27.9% for PP and 22.0 to 28.8% for MPS cows. Lowest colostrum yield from PP animals was associated with calf sex (female) and colostrum Brix % (≤22%). Greatest colostrum yield from MPS cows was associated with colostrum Brix % (≤22%), calf sex (twin), dry period length (>67 d), gestation length (283-293 d), an alive calf, second parity, previous lactation length (>344 d) and previous lactation 305-d mature equivalent milk yield (>13,091 kg), heat and humidity exposure area under the curve (AUC) 7 d before calving (>69.2 average temperature-humidity index per 30-min interval), and light intensity AUC 14 d before calving (>154.2 average lux per 15-min interval). Greatest colostrum Brix % from PP animals was associated with calf sex (male), an alive calf, and light intensity AUC 14 d before calving (≤64.0 average lux per 15-min interval). Greatest colostrum Brix % from MPS cows was associated with dry period length (>67 d), an alive calf, 305-d mature equivalent milk yield of previous lactation (≤15,862 kg), gestation length (274-282 d), colostrum yield (<6 kg), fifth or greater parity, and heat and humidity exposure AUC 7 d before calving (≤50.1 average temperature-humidity index per 30-min interval). Dairy producers can use this information to recognize the variation in colostrum production and alter colostrum management programs in anticipation of periods of low production or quality.

In eukaryotes, three of the four ribosomal RNAs (rRNAs)­the 5.8S, 18S, and 25S/28S rRNAs­are processed from a single pre-rRNA transcript and assembled into ribosomes. The fourth rRNA, the 5S rRNA, is transcribed by RNA polymerase III and is assembled into the 5S ribonucleoprotein particle (RNP), containing ribosomal proteins Rpl5/uL18 and Rpl11/uL5, prior to its incorporation into preribosomes. In mammals, the 5S RNP is also a central regulator of the homeostasis of the tumor suppressor p53. The nucleolar localization of the 5S RNP and its assembly into preribosomes are performed by a specialized complex composed of Rpf2 and Rrs1 in yeast or Bxdc1 and hRrs1 in humans. Here we report the structural and functional characterization of the Rpf2-Rrs1 complex alone, in complex with the 5S RNA, and within pre-60S ribosomes. We show that the Rpf2-Rrs1 complex contains a specialized 5S RNA E-loop-binding module, contacts the Rpl5 protein, and also contacts the ribosome assembly factor Rsa4 and the 25S RNA. We propose that the Rpf2-Rrs1 complex establishes a network of interactions that guide the incorporation of the 5S RNP in preribosomes in the initial conformation prior to its rotation to form the central protuberance found in the mature large ribosomal subunit.

Matcha is a powdered green tea obtained from the Camellia sinensis L. plant intended for both "hot" and "cold" consumption. It is a rich source of bioactive ingredients, thanks to which it has strong antioxidant properties. In this research, an organoleptic evaluation was carried out, and the physical characteristics (i.e., instrumental color measurement (L*a*b*), water activity, water solubility index (WSI), water holding capacity (WHC) of 10 powdered Matcha green teas, and in the 2.5% Matcha water solutions, pH, °Brix and osmolality were tested. Also, the content of phenolic ingredients, i.e., selected phenolic acids, flavonoids and total polyphenols, was assessed. The content of chlorophyll, vitamin C and antioxidant potential were also examined. Matcha M-4 was used to design two functional model beverages, in the form of ready-to-use powdered drinks, consisting of Matcha green tea, protein preparations, inulin, maltodextrin and sugar. The obtained powdered drink, when dissolved in the preferred liquid (water, milk, juice), is regenerative, high-protein and rich in bioactive ingredients from the Matcha drink, with prebiotic properties derived from the added inulin. The beverage is also characterized by low osmolality. It can be recommended as a regenerating beverage for a wide group of consumers, athletes and people with deficiencies, among others protein, and elderly people, as well as in the prevention and supportive treatment of bone and joint tissue diseases.

The intensification of ewe production, focusing on increasing prolificity, results in a higher incidence of metabolic disorders, especially in the transition period, which can lead to production losses with consequences for the health of the animals. The objective of this study was to evaluate the metabolic profile in transition period Lacaune ewes according to the parity and litter size born and its influence on the transference of passive immunity. Nineteen Lacaune ewes, with initial mean body weight of 76.36 ± 10.37 kg (SD), kept in a semi-extensive system and receiving the same diet, were used. Ewes were classified according to parity (primiparous and multiparous) and the litter size born at parturition (1, 2, and 3). On days - 14, - 7, and - 4 of the expected date of parturition and on days 1, 4, 7, and 14 days postpartum, blood sampling, the evaluation of the body condition score (BCS), and the weighing of animals were performed. Blood samples were taken from the lambs 48 h after birth. Colostrum was collected up to 6 h postpartum to determine the brix percentage. Serum concentrations of total plasma proteins (TPP), albumin, aspartate aminotransferase (AST), gamma-glutamyl transferase (GGT), beta-hydroxybutyrate (BHB), non-esterified fatty acids (NEFA), paraoxonase (PON1), calcium (Ca), and magnesium (Mg) were evaluated. In lambs, the blood parameters evaluated were TPP, albumin, GGT, and PON1. Lamb plasma was used to estimate colostrum efficiency, brix percentage immunity, and total plasma protein (PPT). Productive data, pre- and postpartum metabolic variables for ewes, lambs, and those related to colostrum brix were analyzed. The BCS had a reduction (p < 0.0001) in the postpartum period, which is evidenced on days 7 and 14 (2.88 and 2.73, respectively). Serum concentration of BHB presented the highest concentrations in the prepartum period for ewes that gave birth to triplets, from day - 14, and for ewes that gave birth to twins on day - 4 (p = 0.0245). Serum concentration of NEFA demonstrated the highest value on day 14 in primiparous (1.07; p = 0.0402). Calcium had the lowest concentrations on day 1 (8.85 mg/dl, p < 0.0001) and magnesium on day 3 (1.79 mg/dl, p < 0.0001) postpartum. The multiparous with two lambs had higher brix percentages (30.5%), and the lowest value was observed in primiparous ewes that gave birth to a single lamb (27.06%; p = 0.0395). Triplet lambs had the lowest weight (3.73 kg, p = 0.0007), and the best brix percentage in plasma was observed in twins (10.29%, p = 0.0174). Regardless of the parity, the ewes that presented the greatest metabolic challenge were those that gave birth to triplets, and these lambs presented the worst immunity and the lowest live weight. The quality of colostrum was influenced by the parity, and multiparous ewes that gave birth to triplets had the lowest brix percentage in the colostrum.

BACKGROUND: Sweet sorghum is an important crop with sugary stem that can mainly be used for syrup, fodder and bio-fuel. Many sugar content QTLs have been discovered from different sources through breeding worldwide. Most of these QTLs are detected using exotic germplasm as a mapping population. This study aimed to detect and map QTLs for stem sugar content and stem diameter targeting Ethiopian recombinant inbred lines of sorghum using genotyping-by-sequencing. METHODS AND RESULT: Genotyping-by-sequencing and phenotyping using 139 recombinant inbred lines of sorghum as mapping populations were conducted. A total of 1082 polymorphic and high quality SNP markers that are evenly distributed across the ten linkage groups of sorghum were selected to detect and map the trait of interest. A genetic linkage map using 1082 SNP markers was constructed and several QTLs associated with stem sugar content and stem diameter were identified. Phenotypic variation explained by qBrix4-1 and qBrix2-1 ranged from 6.33 to 14%, respectively. Over two seasons, four QTLs for stem sugar content (qBrix1-1, qBrix2-1, qBrix4-1 and qBrix4-2) and three QTLs for stem diameter (qSD1-1, qSD8-1 and qSD9-1) were detected. CONCLUSION: QTLs that significantly associated with stem sugar content and stem diameter have been detected and mapped. This will help sorghum breeding program to develop superior sweet sorghum varieties through the use of appropriate crop improvement approaches like marker assisted breeding. This ultimately contributes to the current development plan to considerably improve food, feed and bio-fuel supply in developing countries like Ethiopia.

This paper presents a Brix sensor based on the differential pressure measurement principle. Two piezoresistive silicon pressure sensors were applied to measure the specific gravity of the liquid, which was used to calculate the Brix level. The pressure sensors were mounted inside custom-built water-tight housings connected together by fixed length metallic tubes containing the power and signal cables. Two designs of the sensor were prepared; one for the basic laboratory testing and validation of the proposed system and the other for a fermentation experiment. For lab tests, a sugar solution with different Brix levels was used and readings from the proposed sensor were compared with a commercially available hydrometer called Tilt. During the fermentation experiments, fermentation was carried out in a 1000 L tank over 7 days and data was recorded and analysed. In the lab experiments, a good linear relationship between the sugar content and the corresponding Brix levels was observed. In the fermentation experiment, the sensor performed as expected but some problems such as residue build up were encountered. Overall, the proposed sensing solution carries a great potential for continuous monitoring of the Brix level in liquids. Due to the usage of low-cost pressure sensors and the interface electronics, the cost of the system is considered suitable for large scale deployment at wineries or juice processing industries.

Quality assessment of fruits, vegetables, or beverages involves classifying the products according to the quality traits such as, appearance, texture, flavor, sugar content. The measurement of sugar content, or Brix, as it is commonly known, is an essential part of the quality analysis of the agricultural products and alcoholic beverages. The Brix monitoring of fruit and vegetables by destructive methods includes sensory assessment involving sensory panels, instruments such as refractometer, hydrometer, and liquid chromatography. However, these techniques are manual, time-consuming, and most importantly, the fruits or vegetables are damaged during testing. On the other hand, the traditional sample-based methods involve manual sample collection of the liquid from the tank in fruit/vegetable juice making and in wineries or breweries. Labour ineffectiveness can be a significant drawback of such methods. This review presents recent developments in different destructive and nondestructive Brix measurement techniques focused on fruits, vegetables, and beverages. It is concluded that while there exist a variety of methods and instruments for Brix measurement, traits such as promptness and low cost of analysis, minimal sample preparation, and environmental friendliness are still among the prime requirements of the industry.

Using the framework of aquaphotomics, we have sought to understand the changes within the water structure of kiwifruit juice occurring with changes in temperature. The study focuses on the first (1300-1600 nm) and second (870-1100 nm) overtone regions of the OH stretch of water and examines temperature differences between 20, 25, and 30 °C. Spectral data were collected using a Fourier transform-near-infrared spectrometer with 1 mm and 10 mm transmission cells for measurements in the first and second overtone region, respectively. Water wavelengths affected by temperature variation were identified. Aquagrams (water spectral patterns) highlight slightly different responses in the first and second overtone regions. The influence of increasing temperature on the peak absorbance of the juice was largely a lateral wavelength shift in the first overtone region and a vertical amplitude shift in the second overtone region of water. With the same data set, we investigated the use of external parameter orthogonalisation (EPO) and extended multiple scatter correction (EMSC) pre-processing to assist in building temperature-independent partial least square regression models for predicting soluble solids concentration (SSC) of kiwifruit juice. The interference component selected for correction was the first principal component loading measured using pure water samples taken at the same three temperatures (20, 25, and 30 °C). The results show that the EMSC method reduced SSC prediction bias from 0.77 to 0.1 °Brix in the first overtone region of water. Using the EPO method significantly reduced the prediction bias from 0.51 to 0.04 °Brix, when applying a model made at one temperature (30 °C) to measurements made at another temperature (20 °C) in the second overtone region of water.

Reports in the literature have proposed and analyzed several minimally invasive techniques for caries removal in recent decades. In light of recent events surrounding the ongoing epidemiological context, concerns have been raised regarding the generation of aerosols during dental procedures. The aim of our research was to provide an overview of the scientific literature on the topic of chemo-mechanical caries removal (CMCR) methods, focusing on two products (Carisolv, BRIX3000), commercially available in Europe. A bibliometric analysis was used to investigate the scientific articles included in Web of Science (WoS) Core Collection database, published from January 2010 to December 2020. We analyzed the co-occurrence of all keywords (Author Keywords and the KeyWords Plus section), co-authorship and co-citation, using the free software VOSviewer. Our bibliometric analysis revealed a worldwide interest in the subject of chemo-mechanical methods of caries removal, which has transcended the area of pediatric dentistry. The analyzed studies have been conducted mainly in high-income countries that have developed sanitary policies regarding prevention and early treatment of carious lesions as a health priority.

AIM: The aim of the present study was to compare and evaluate the efficacy of two minimally invasive methods in the removal of infected carious dentin in primary molars. MATERIALS AND METHODS: Eighty primary molars with class I occlusal caries involving dentin were selected and allocated to either of the two groups using convenient sampling. Group A (n = 40) caries were excavated using BRIX 3000 and in Group B (n = 40) atraumatic restorative treatment (ART) caries were excavated using hand instruments only. In both groups, caries removal efficacy was evaluated clinically using caries detection dye (Sable and Sleek), and the patient's discomfort level was noted using the Wong-Baker faces pain rating scale (WBFPS). RESULTS: The results indicated that BRIX 3000 was effective in the removal of caries from primary molars with a significant difference. While there was no statistically significant difference seen in pain perception on caries excavation using both the methods. Although, a mild discomfort was noted in Group B. CONCLUSION: Comparing the two minimally invasive techniques, the new chemomechanical solution (BRIX 3000) was an effective method for the removal of caries from primary molars. So, it can be considered an alternative to the conventional caries removal method in treating pediatric dental patients. CLINICAL SIGNIFICANCE: BRIX 3000 gel is effective in the removal of caries in children who are anxious and apprehensive as it is a noninvasive method of treatment in the pediatric age group.

This paper reports a coherent method for ranking fruit juice products in the Iranian food industry by the best-worst method by examining the factors affecting product quality. To achieve the aim, a combination of laboratory methods and mathematical calculations was used. The statistical population in this study included samples of products of several food companies (SH, S, P) in Iran. Some factors affecting the quality of fruit juice products including alcohol, sugar, degrees Brix, and pH were considered and several samples of apple and grape juice from different brands were prepared and analyzed. In the first stage, to determine the value of the parameters of each sample was determined by the standard analysis methods. Then, the best-worst method was applied to measure the selection criteria of juice producers, and then the function of linear values of the piece was used to rank the Juice products of the studied companies (SH, S, P). The results showed the following order for the studied brands: P-Apple > S-Grape > SH-Apple > SH-Grape > S-Apple. This study showed that the multi-criteria decision theory is a promising method to rank food products of different companies.

The objective of this study was to identify the calf-level colostrum management practices associated with an adequate transfer of passive immunity (TPI; defined as serum Brix refractance ≥8.4% in the first week of life) in small-sized herds. A total of 818 calves from 61 commercial Holstein dairy farms were included in this observational cross-sectional study. For each calf, sex, colostrum delivery method, colostrum volume fed at first meal, and time to first feeding (delay between birth and first colostrum meal) were noted. Blood and colostrum samples were collected to estimate the serum and colostrum quality using Brix refractometry. To quantify the level of bacterial contamination in colostrum samples, total bacteria count and total coliform count (TCC) were measured using the Petrifilm (3M, St. Paul, MN) culture system. In this study, 68% of calves had an adequate TPI (≥8.4%). For data distribution, the 25th, 50th, and 75th percentiles were 1.3, 2.8, and 3.3 L for the colostrum volume fed at the first meal; 20.9, 23.5, and 26.5% Brix; and 1.1, 3.1, and 6.5 h for the time to first feeding of colostrum, respectively. The odds of adequate TPI were 2.6 times higher in calves receiving ≥2.5 L colostrum at their first meal, 2.9 times higher in calves receiving colostrum with ≥24.5% Brix, and 1.6 times higher in calves receiving colostrum within 3 h after birth, than in calves not meeting these criteria. In the present study, median bacterial contamination distribution (interquartile range) in the first colostrum meal was 14,000 cfu/mL (3,000-83,000 cfu/mL) for total bacteria count, and 0 cfu/mL (0-1,000 cfu/mL) for TCC. Total bacteria count and TCC were not associated with the odds of adequate TPI in the final model. Overall, these results suggest that specific calf-level colostrum management practices are associated with adequate TPI in small- to medium-sized dairy herds.
