RESUMO

PURPOSE: Presoaking the graft with vancomycin before implantation has been shown to reduce the risk of postoperative infection after anterior cruciate ligament reconstruction (ACLR). However, the effects of presoaking on the graft biomechanical properties remain unclear. This study aimed to determine whether presoaking the graft with vancomycin affects the graft biomechanical properties and length after cyclic loading. METHODS: Ten paired (20 specimens) gracilis and semitendinous tendons were harvested from fresh-frozen human cadaveric specimens. Two tendons were folded in half to make four strands, and the grafts were randomized into the vancomycin and control groups. The graft was exposed to the antibiotic solution for 15 min (5 mg/mL) and prepared by mixing 1 g of vancomycin with 200 mL of normal saline (NaCl 0.9%). The control group was soaked in normal saline for 15 min. The prepared grafts were attached to the actuator of a dynamic tensile-testing machine. All grafts were tested with 3000 cycles of cyclic loading followed by a pull-to-failure. The cyclic loading protocol consisted of position and load control blocks to simulate the graft in vivo in the postoperative phase after ACLR. RESULTS: Presoaking in vancomycin did not jeopardize the biomechanical properties of the graft. In addition, presoaking with vancomycin did not elongate the grafts. No significant differences were found in the mean Young's modulus and the mean total elongation of the graft of the specimen between the vancomycin group and the control group. CONCLUSION: Presoaking the graft with vancomycin jeopardized neither its biomechanical properties nor elongation even after cyclic loading in this in vitro study. It is suggested that vancomycin presoaking could be considered a safe and effective preventive measure for postoperative infections after ACLR. LEVEL OF EVIDENCE: Not applicable.

RESUMO

Investigating the effect of presoaking, as one of the most important physical factors affecting the adsorption behavior of biochar, on the adsorption of heavy metals by modified or non-modified biochar and presoaking mechanism is still an open issue. In this study, the water presoaking effect on the kinetics of cadmium (Cd) adsorption by rice husk biochar (produced at 450 °C, B1, and at 600 °C, B2) and the rice husk biochar modified with magnesium chloride (B1 modified with MgCl2, MB1, and B2 modified with MgCl2, MB2) was investigated. Furthermore, the effect of pH (2, 5, and 6), temperature (15, 25, and 35 °C), and biochar particle size (100 and 500 µm) on the kinetics of Cd adsorption was also investigated. Results revealed that the content of Cd adsorbed by the presoaked biochar was significantly higher than that by the non-presoaked biochar. The highest Cd adsorption capacity of MB2 and MB1 was 98.4 and 97.6 mg g-1, respectively, which was much better than that of B1 (7.6 mg g-1) and B2 (7.5 mg g-1). The modeling of kinetics results showed that in all cases pseudo-second-order model was well-fitted (R2>0.99) with Cd adsorption data. The results also indicated that the highest Cd adsorption values were observed at pH 6 in presoaked MB1 with size of 100 µm as well as at the temperature of 35 °C in presoaked MB2, indicating the optimum conditions for this process. The presoaking process was not affected by biochar size and pH, and the difference in adsorbed Cd content between presoaked biochars and non-presoaked ones was also similar. However, the temperature had a negative effect on presoaking. The presoaking process decreased micropores (<10 µm) in the biochars but had no effect on biochar hydrophobicity. Therefore, presoaking, which could significantly increase Cd adsorption and reduce equilibrium time by reducing the micropores of biochars, is suggested as an effective strategy for improving the efficiency of modified biochars or non-modified ones in the adsorption of contaminants (Cd) from aquatic media.

RESUMO

Foundry dust is the main refractory solid waste in the foundry industry, and its resource utilization is a top priority for realizing green and cleaner production. The massive amount of coal dust in foundry dust is a potential impediment to the recycling of foundry dust, and the efficient separation of coal dust is crucial to solving the above problems. In this paper, the flotation separation of coal dust from foundry dust enhanced by pre-soaking assisted mechanical stirring was reported. The influence of pre-soaking, stirring speed, and stirring time on the flotation results of foundry dust was systematically studied, and the enhancement mechanism was analyzed based on the microstructure and hydrophobicity of foundry dust. Flotation kinetics experiments with different stirring time were conducted to clarify the flotation process of foundry dust. The results indicate that the pre-soaking of foundry dust is beneficial for the water-absorbing swelling of clay minerals coated on the surface of coal dust, and the subsequent mechanical stirring pretreatment promotes the monomer dissociation of foundry dust, which increases the contact angle of foundry dust and considerably improves the flotation results. The optimal stirring speed and stirring time were 2400 rpm and 30 min, respectively. The classical first-order model presented the highest degree of fitting with the flotation data among the five flotation kinetics models. Therefore, the pre-soaking assisted mechanical stirring is a promising method for promoting flotation separation and the complete recycling of foundry dust.

RESUMO

PURPOSE: To determine and compare the incidence of postoperative septic arthritis following anterior cruciate ligament reconstruction (ACLR) with and without soaking of the graft in vancomycin solution prior to implantation in a large single-centre case series. METHODS: From 2004 to 2019, a total of 10,516 primary ACLR were performed and reviewed with regard to the occurrence of postoperative septic arthritis. From February 2017 onwards, all grafts were wrapped in a vancomycin-soaked (5 mg/ml) gauze swab between harvest and implantation (2294 patients, treatment group (2), prospectively followed). These were compared to 8222 patients before that date (control group (1), retrospectively evaluated). The technique of ACLR did not significantly change during the years of the study. There was no difference between the groups with regard to graft choice: Hamstring tendons were used in 99% and quadriceps tendons were used in 1% in both groups, respectively (n.s.). Routine follow-up examination was performed at 6 weeks (follow-up rate 97.1%) postoperatively. Patients with no treatment for septic arthritis at that time were classified as non-infected. RESULTS: There were 35 cases of postoperative septic arthritis in group 1 (incidence: 0.4%), and none in group 2 (incidence 0.0%), respectively. The difference was significant (p < 0.001). CONCLUSIONS: Soaking of the graft in vancomycin solution prior to implantation dramatically reduces the incidence of postoperative septic arthritis in primary ACLR and should, therefore, be used in prevention of this major complication. LEVEL OF EVIDENCE: III.

RESUMO

BACKGROUND: Soybeans are popularly known as a healthy food in many Asian countries and are mostly consumed as sprouts. The present study aimed to investigate the potential applications of chitosan as a natural growth regulator of soybean sprouts, as well as to determine the variation and composition of nutrients and anti-nutrients of soybean sprouts pre-soaked in different concentrations of chitosan solutions. RESULTS: The hypocotyl length and fresh weight of the soybean sprouts could be positively affected by chitosan treatment. The content changes of bioactive products in chitosan-soaked soybean sprouts were dependent on the concentration of chitosan and germination time. Additionally, the lowest phytic acid content was observed in 8 g kg-1 chitosan-treated soybean sprouts, and the phytic acid content values after 5 days of germination was 1.56 g kg-1 , which was decreased by 79.0% compared to the value in ungerminated soybean. CONCLUSION: Chitosan pre-soaking can increase the hypocotyl length and fresh weight of soybean sprouts and also positively effect the content of bioactive products in sprouts, whereas phytic acid can be partially removed, thereby leading to the enhancement of product quality. Chitosan pre-soaking is an alternative technology for the production of low-cost functional foods, where the dose of chitosan is a key factor. © 2018 Society of Chemical Industry.

RESUMO

Firefighters are exposed to hazardous chemicals at fire scenes, including polycyclic aromatic hydrocarbons (PAHs) among many others, which pose significant health risks. Current laundering practices are ineffective at removing persistent contaminants from turnout gear, necessitating further research to optimize cleaning methods. This study explores the impact of presoaking prior to the laundering process and the factors that can affect its effectiveness, including the presoaking duration and detergent concentration, in PAH removal when laundering. For this, contaminated fabric swatches were subjected to various presoaking durations (1, 3, and 12 h) and detergent concentrations (99:1 and 90:10 water-to-detergent ratios) before undergoing bench-scale washing. The cleaning efficacy was assessed for 16 PAH compounds, including both low-molecular-weight (LMW) PAHs and high-molecular-weight (HMW) PAHs. Moreover, the removal mechanisms of PAHs from turnout gear were fundamentally explained using partition coefficients and standard affinities with different parameters during washing. The results demonstrate that 3 h and 12 h of presoaking lead to 2.8 and 4.3 times greater HMW PAH removal, respectively. After 12 h of presoaking in a 90:10 water-to-detergent ratio, 97% of the LMW PAHs and 78% of the HMW PAHs were removed, compared to only an 11% removal of the HMW PAHs with a 99:1 ratio. Additionally, direct washing with a 90:10 ratio achieved comparable efficacy to that of presoaking with the same water-to-detergent ratio, indicating the crucial role of detergent concentration during laundering. These findings offer valuable insights for optimizing firefighter safety practices, emphasizing the role of presoaking and the appropriate methods to perform presoaking to mitigate firefighters' occupational exposure risks to toxic substances and ensure gear reliability.

RESUMO

Cooking is an important process before rice is consumed and constitutes the key process for rice flavor formation. In this paper, dynamic changes in aroma- and sweetness-related compounds were tracked during the entire cooking process (including washing with water, presoaking, and hydrothermal cooking). The volatiles, fatty acids, and soluble sugars in raw rice, washed rice, presoaked rice, and cooked rice were compared. After being washed with water, the total volatiles decreased while aldehydes and unsaturated fatty acids increased. Meanwhile, oligosaccharides decreased and monosaccharides increased. The changes in fatty acids and soluble sugars caused by the presoaking process were similar to those in the water-washing process. However, different changes were observed for volatiles, especially aldehydes and ketone. After hydrothermal cooking, furans, aldehydes, alcohols, and esters increased while hydrocarbons and aromatics decreased. Moreover, all fatty acids increased; among these, oleic acids and linoleic acid increased most. Unlike with washing and presoaking, all soluble sugars except fructose increased after hydrothermal cooking. Principal component analysis showed that cooked rice possessed a volatile profile that was quite different from that of uncooked rice, while washed rice and presoaked rice possessed similar volatile profiles. These results indicated that hydrothermal cooking is the pivotal process for rice flavor formation.

RESUMO

The main factor that alters the quality of recycled concrete aggregate (RCA) is the paste adhered to the natural aggregate (NA). Since it causes weakening of the interfacial transition zone (ITZ) between the aggregate and the cementitious paste, it becomes a determining factor for the mechanical behavior of concrete. It turns out that it is critical to enhance this interface by improving the surface of the aggregate or by removing the paste adhered to the NA. Considering the variety of methods for removing paste adhered to RCA-namely using acids such as hydrochloric acid (HCl), sulfuric acid (H2SO4), and phosphoric acid (H3PO4), among others-this paper presents a review of treatments for the removal of adhered paste using acidic solutions on the RCA, and their influence on the mechanical properties and durability of concrete produced with RCA. Pearson's correlation was used in the statistical analysis to determine the linear relationship of the main factors-for instance, immersion time, acidic solution, and aggregate size-involved in the removal of the paste in the RCA.

RESUMO

To investigate the effect of hemicellulose removal on subsequent choline chloride and lactic acid (ChCl-LA) based deep eutectic solvent (DES) extraction of wheat straw lignin, ChCL-LA of DES and hot water presoaking pretreatments were used for hemicellulose prehydrolysis. Both presoakings led to a significant hemicellulose removal and introduced morphological changes on fiber cell wall surface. DES presoaking also instigated ether bonds cleavage between lignin and hemicellulose and selectively removed lignin in compound middle lamella (CML) and cell corner (CC) leading to cell wall disruption and swelling which facilitated lignin extraction. Hot water presoaking removed more hemicellulose and caused a migration of lignin to fibers surface, but did not improve subsequent lignin extraction. This study demonstrated that a two-stage DES treatment method, presoaking at room temperature followed by extracting at an elevated temperature, is a viable process to produce high yield and purity of lignin.

RESUMO

Approximately 1% of anterior cruciate ligament reconstruction (ACLR) procedures develop septic arthritis despite intravenous antibiotic prophylaxis and other preventive measures. Infection is most commonly due to contamination during autograft harvest and preparation by introducing bacteria into the knee during graft insertion. Pre-soaking ACL grafts in 5 mg/mL vancomycin ("vancomycin wrap") has been utilised to eradicate such bacterial contamination. Many level III studies have reported a marked decrease in infection rates with no increase in graft failure rates. However, the lack of prospective randomised control trials and these studies' heterogeneity do not allow a universal recommendation for vancomycin pre-soaking of all grafts during ACLR. Randomised controlled trials are needed to confirm efficacy in reducing sepsis rates.

RESUMO

This study was conducted to investigate the yield and quality characteristics of red bean sprouts of three cultivars (Arari, Geomguseul, and Chungju) soaked in water for 0, 6, 12 or 24 h. The sprout yields of 'Arari' and 'Geomguseul' on day 7 were highest with the seeds soaked for 12 h. For 'Chungju', the yields from the seeds soaked for 12 and 24 h were not significantly (p > 0.05) different. Longer hypocotyls and shorter roots, which are also desirable characteristics of good sprouts, were also found in the sprouts with 12 h of seed soaking. The amounts of total minerals, thiamine, total free amino acids, and total phenols and DPPH radical scavenging potential of sprouts of all cultivars were higher than those of their seeds. This study showed that higher yield and better quality of red bean sprouts could be obtained with the seeds soaked for 12 h.

RESUMO

The continuous decline in regeneration of two important species of central Himalayan oak, namely Quercus glauca and Q. leucotrichophora, is of great concern. A study was therefore, carried out to improve germination ability of these species using various presoaking treatments. Seeds of both the species lost viability following storage; tetrazolium staining pattern and germination capacity of seeds following different period of storage at 4 °C and 20 °C indicated retainment of viability for a period of 12 months at 4 °C. Of the various physical, chemical and plant growth regulator treatments examined to improve seed germination, only KNO3 1.0 % was found to be effective. Seeds scarified at the chalazal end exhibited significant improvement in germination in both the species (94.4 % compared to 56.7 % in control in Q. glauca and 82.7 % compared to 64.0 % in control in Q. leucotrichophora). The results of this study impart simple methods to improve seed germination for developing nurseries for commercial purposes.

RESUMO

BACKGROUND: Postoperative septic arthritis is an uncommon but potentially devastating complication after anterior cruciate ligament (ACL) reconstruction. Our group started presoaking grafts with vancomycin to decrease this risk. PURPOSE: To compare the rate of septic arthritis in primary hamstring autograft ACL reconstruction with and without vancomycin-presoaked grafts. STUDY DESIGN: Cohort study; Level of evidence, 3. METHODS: Consecutive periods were studied, inclusive of April 2013 through October 2015 (pre-vancomycin protocol) and November 2015 through May 2018 (vancomycin protocol). A total of 490 patients were included in the study: 230 in the pre-vancomycin protocol and 260 in the vancomycin protocol. All patients who underwent a primary hamstring autograft ACL reconstruction by 2 senior surgeons during the periods studied were included. The final outcome studied was occurrence of highly probable postoperative septic arthritis in both groups. Diagnosis of probable septic arthritis was made by clinical diagnosis as well as cytological analysis of joint aspiration (cell count >50,000/µL and >90% neutrophils). Statistical analysis was performed with the Fisher exact test. Significance was set at P < .05. RESULTS: Four cases of probable postoperative septic arthritis were noted in the pre-vancomycin protocol (1.7%; 2 cases per surgeon), while no cases of septic arthritis were noted in the vancomycin protocol during the study period (P < .05). Diagnosis was made at a mean 21.7 days (range, 16-25 days). Staphylococcus epidermidis was isolated in 2 cases, and in the other 2 cases, no organism was isolated. CONCLUSION: Presoaking of hamstring autografts in vancomycin for primary ACL reconstruction prevented the occurrence of postoperative septic arthritis during the study period as compared with no soaking of the grafts.

RESUMO

BACKGROUND: The acid buffering capacity of high free ash in waste wheat straw (WWS) has been revealed to be a significant hindrance on the efficiency of autohydrolysis pretreatment. Previous researches have mainly relied on washing to eliminate the influence of ash, and the underlying mechanism of the ash influencing was not extensively investigated. Presently, studies have found that cations can destroy the acid buffering capacity of ash through cation exchange. Herein, different cations were applied to presoak WWS with the aim to overcome the negative effects of ash on autohydrolysis efficiency, further improving its enzymatic digestibility. RESULTS: Results showed that cations can be adsorbed on the surface of the material by electrostatic adsorption to change the acid buffering capacity of WWS. The acid buffering capacity of 120 mM Fe2+ presoaked WWS is reduced from 226.3 mmol/pH-kg of original WWS to 79.3 mmol/pH-kg. This reduced the autohydrolysis pretreatment medium pH from 5.7 to 3.8 and promoted the removal of xylan from 61.7 to 83.7%. In addition, the enzymatic digestibility of WWS was enhanced from 49.7 to 86.3% by presoaking with 120 mM Fe2+ solution. The relationship between enzymatic accessibility and hydrophobicity with enzymatic digestibility of the autohydrolyzed WWS was analyzed. CONCLUSIONS: The results showed that the acid buffering capacity of the high free ash was detrimental for the autohydrolysis efficiency of WWS. After WWS was presoaked with different cations, the acid buffering capacity of ash was weakened by cation exchange and electrostatic adsorption, which improved the autohydrolysis efficiency. The results expound that the enzymatic digestibility of WWS can be enhanced through presoaking to reduce the ash-influencing effect on autohydrolysis.

RESUMO

Functional properties of flours from pre-soaked and micronised (130°C) whole and dehulled bambara seeds (5, 10 and 15min) were determined. An increase in micronisation time significantly reduced the pasting viscosity of the flours. Significant reductions in the differential scanning calorimetry endothermic peak enthalpies and loss of birefringence in the starch were found, indicating starch pre-gelatinisation when micronised. The low viscous paste of resultant flours seems to be related to protein denaturation as shown by decrease in nitrogen solubility index. Starch was embedded in a protein matrix as shown by confocal laser scanning microscopy. This denatured protein matrixes could be in part preventing starch hydration and dispersion during pasting and thus reduced viscosity. Dehulling reduced the pasting viscosity suggesting higher effect of micronisation for dehulled than whole samples. Resulting flours can be useful ingredients in protein energy-dense foods due to low viscosity.

RESUMO

An efficient strategy was developed in current work for biochemical conversion of carbohydrates of corn stover into monosaccharides. Corn stover was first presoaked in mild alkaline solution (1% Na2S) under 40°C for 4h, after which about 35.3% of the lignin was successfully removed while the specific surface area was notably enlarged. Then the presoaked solids were subjected to organosolv pretreatment that employed 20% methanol with an addition of 0.2% HCl as catalyst at 160°C for 20min, and the maximum total sugar yield of the pretreated corn stover achieved was 98.6%. The intact structure of corn stover was disrupted by this two-step process, which resulted in a porous but crystalline structure of the regenerated solids that were mainly composed of cellulose. The enlarged specific surface area and increased accessibility made the regenerated solids highly digestible by a moderate enzyme loading.

RESUMO

Paricá seeds have dormancy and, even after mechanical scarification, these seeds show slow and uneven germination. Pre-germination treatments can be used to increase seed germination performance. Thus, the aimed to evaluate the physiological potential and initial growth of paricá seeds after pre-germination treatments, using different substances as plant regulators and nutrients, in addition to mechanical scarification. The experimental design was completely randomized, in a 2x7 factorial scheme, consisting of the following pre-germination treatments: mechanical scarification (10% and 50% of the seed coat) and seed pre-soaking [control-water, KNO3 0.2%,Ca(NO3)2 0.2%, gibberellin 0.02%, cytokinin 0.02%, and mixture of gibberellin + cytokinin (1:1)] besides a control group without pre-soaking, with four replicates. The study evaluated germination and emergence rates, germination and emergence speed indices, collar diameter, plant height, seedling dry mass, hypocotyl and seedling length, and electrical conductivity. It was observed that pre-soaking the seeds in gibberellin after mechanical scarification at 50% as a pre-germination treatment resulted in seeds with higher vigor expression and greater initial seedling growth.

The objective was to evaluate the effect of gibberellic acid (GA3) on seed germination and seedling vigor of passion fruit (Passiflora edulis Sims f. flavicarpa Deg). Seeds of passion fruit were pre-soaked in solutions of gibberellic acid to 4%, for six hours in the following concentrations: 0,5; 1,0; 2,0; 4,0 mL per liter of solution and distilled water as control. The seeds were spread on germination paper and kept in a germination chamber at a temperature of 25 °C with a photoperiod of 12 hours. The current test was conducted simultaneously with the pattern of germination and seedlings measured at 14 days after sowing (DAS). A completely randomized design with five treatments and four replicates with 50 seeds in each roll. It was evaluated the percentage of seed germination, seedling normal on the first count, abnormal seedlings, dead seeds, seed firm, root length, shoot and total seedling index and emergence rate (IVE). The data was subjected to analysis of variance and the average treatment were adjusted polynomial regression equations. The results indicate that pre-soaking the seeds of passion fruit with GA3, stimulates germination and reduces the percentage of dead seeds. The growth regulator provides an increase in the length of the tops of yellow passion fruit.

This study aimed to evaluate the effects of Stimulate® on germination, emergence and seedling vigor of sunflower. For the standard germination test, seeds of the variety Catissol 01 were pre-soaked at the following concentrations: 1,0, 2,5, 4,0, 5,5 and 7,0 mL of Stimulate® L-1 aqueous solution, taking control as pre-soaking seeds in water. Then, the seeds were subjected to three times of different presoaking: 4, 7 and 10 hours. After the treatments, the seeds were distributed in germination papers and kept in a germinator at a temperature of 25ºC with a photoperiod of 12 hours. The experimental design was completely randomized in a factorial of 3 x 6, with 4 replicates of 50 seeds in each paper roll germtest. A seedling vigor test was conducted simultaneously with the standard germination test. To study the rate of emergence and the percentage of emergence, seeds were genotyped: Catissol 01, Agrobel 962 and Agrobel 972. The seeds were subjected to two treatments: T1 = 4 mL solution containing Stimulate® L-1 solution in the pre-soaking for 4 hours and T2 = pre-soaking of sunflower seeds in water for 4 hours, being registered daily emergence seedling. The experimental design was completely randomized in a factorial of 3 x 2. The results indicated that pre-soaking of sunflower seeds with vegetable biostimulant Stimulate® (4 mL Stimulate® L-1 in pre-soaking for 4 hours) increased germination, promoting the formation of more vigorous seedlings and reducing the percentage of abnormal seedlings, besides Stimulate® na germinação... SANTOS, C. A. C. et al. Biosci. J., Uberlândia, v. 29, n. 2, p. 605-616, Mar./Abr. 2013 promoting a higher percentage of seedling emergence. However, prolonged periods of pre-soaking increased the percentage of abnormal seedlings.
