RESUMO

Doublesex (Dsx) is a crucial member of the Dmrt gene family and plays a vital role in sex determination and differentiation among the animal kingdom. In the present study, a doublesex (designated as Spdsx) gene was identified and characterized for the first time in the mud crab, Scylla paramamosain. The Spdsx cDNA contains an 801 bp open reading frame (ORF) encoding 266 amino acids with a conserved DM domain. Meanwhile, to elucidate the conservation of Dsx, its orthologus were identified in several crustacean species as well. In addition, the expression pattern of Spdsx in various adult tissues and during embryo development was analyzed with qRT-PCR technology. Finally, the roles of Spdsx might play in the testis, androgenic gland, and ovary were analyzed by RNAi technology. The main results are as follows: (1) the Spdsx gene widely existed in analyzed crustacean species, and the multiple sequences alignment result indicated the conservation of Dsx was low except for the DM domain; (2) only one dsx gene was identified in analyzed crab and lobster, while 2 dsx genes (dsx-1 and dsx-2) were identified in shrimps; (3) the Spdsx gene was widely expressed in analyzed tissues, and the expression level in androgenic gland was obviously higher than that in other tissues. Interestingly, the expression level of Spdsx in the ovary was significantly higher than that in testis (p < 0.05); (4) The expression pattern of Spdsx during embryo development was divided into two groups: remained stable from blastula stage to 5 pairs of appendages stage; after 5 pairs of appendages stage, the expression level increased and remained stable from 7 pairs of appendages stage to hatching stage; (5) After the silencing of Spdsx, the expression level of marker genes in testis, ovary, and androgenic gland significantly changed, among which the expression level of vtg and vtgR in ovary down-regulated, the dmrt-like and dmrt-1a (exclusively expressed in testis) in testis up-regulated and the IAG in androgenic gland down-regulated. All the results above demonstrated that the Spdsx play crucial roles in regulating the reproduction system development of mud crab.

RESUMO

Exosomes (Exos), belonging to extracellular vesicles, are cell-derived nano-sized vesicles with the potential to carry different kinds of biological molecules. Many studies have proved the impacts of exosomal cargo on several biological processes in female and male reproductive systems. It is also hypothesized that changes in exosomal cargo are integral to the promotion of certain pathological conditions, thus Exos can be used as valid biomarkers for the diagnosis of infertility and other abnormal conditions. Here, efforts are made to collect some recent data related to the physiological significance of Exos in the reproductive system, and their potential therapeutic effects. It is anticipated that the current review article will lay the groundwork for elucidating the source and mechanisms by which Exos control the reproductive system additionally supplying fresh methods and concepts for the detection and treatment of disorders associated with fertility for future studies.

Aspongopus chinensis is a Chinese traditional edible and medicinal insect, which is in great demand in the society. This insect reproduces once a year which is caused by reproductive diapause resulting in insufficient production in wild resources. However, the mechanism of diapause in A. chinensis is still unclear. In this study, we focus on the relationship between juvenile hormones (JHs) and A. chinensis diapause. The results showed that JHIII concentration in diapause adult individuals was significantly lower than that in diapause termination adult individuals. When exogenous JHⅢ was injected into diapause adults, the rate of mating was increased significantly, development of the reproductive systems was accelerated, consumption of fat intensified, the expression of juvenile hormone acid o-methyl-transferase (JHAMT) was upregulated, and juvenile hormone epoxide hydrolase (JHEH) and fatty acid synthase (FAS) gene expressions were downregulated. In addition, RNAi of JHAMT decreased JH concentration, delayed the development of reproductive systems, slowed down fat consumption, and delayed the mean mating occurrence time significantly. Conversely, RNAi of JHEH resulted in an increased concentration of JH, development of reproductive systems was accelerated, consumption of fat was intensified, and mean mating occurrence time advanced significantly. Taken together, these findings uncovered that JH plays an important role in regulating reproductive diapause in A. chinensis and, thus, could provide a theoretical basis for further research on the diapause of A. chinensis.

Rhesus macaque (Macaca mulatta) is widely applied in animal model construction of infertility, spermatogonia stem cell transplantation and male reproductive diseases. In this review, we describe the seasonal changes of the reproductive system in rhesus macaques, the regular pattern of spermatogenesis and spermatozoa maturation, and the differentiation of spermatogonia and spermatocytes. The duration of the M. mulatta spermatogenesis is approximately 10 days and seminiferous epithelium cycles mainly consist of 12 stages, which provide a suitable model for reproductive studies in non-human primates. Here, we summarize the features of gonadal development and sperm maturation in the rhesus monkeys, which provide important information in the studies of reproductive biology. Rhesus macaque is an excellent animal model in spermatogonia stem cell transplantation. We discuss the applications and progresses of assisted reproductive technologies in sperm liquefaction, semen cryopreservation and spermatogonia stem cell transplantation of rhesus macaques. Besides, we sort out recent proteomic analyses of male reproductive systems and semen samples in rhesus macaques. This review mainly focuses on male reproductive biology and application studies using M. mulatta, which would promote the development of new therapeutic interventions on assisted reproduction and reproductive disease studies in the future.

Purpose: The role of transposable elements in formation of radiobiological effects is understudied and contradictory. The aim of this study was to investigate the response of Drosophila melanogaster to irradiation depending on the level of activity hobo transposons and the role of hobo transposons in formation of ionizing radiation late effects.Materials and methods: The individuals of Drosophila melanogaster with different level activity of hobo-elements were exposed to acute irradiation in doses of 1-100 Gy at early ontogenesis stages. The reaction of individuals to exposure was studied using the larvae survival rate, morphological parameters of reproduction system, DNA damage rate, and mutability of mini-white locus.Results: We found the pronounced linear deferred effects of irradiation for animals with a high activity level of full-size hobo copies. The radiosensitivity of individuals with a mean level of activity transposon was whether higher or did not differ from the radiosensitivity of animals with a low activity hobo.Conclusion: The obtained results suggest that full-size hobo-elements with a high activity level (less often with a mean activity level) are responsible for delayed deleterious irradiation effects.

This study developed an in vitro diaphragm motion reproduction system (IVDMRS) based on noninvasive and real-time ultrasound imaging to track the internal displacement of the human diaphragm and diaphragm phantoms with a respiration simulation system (RSS). An ultrasound image tracking algorithm (UITA) was used to retrieve the displacement data of the tracking target and reproduce the diaphragm motion in real time using a red laser to irradiate the diaphragm phantom in vitro. This study also recorded the respiration patterns in 10 volunteers. Both simulated and the respiration patterns in 10 human volunteers signals were input to the RSS for conducting experiments involving the reproduction of diaphragm motion in vitro using the IVDMRS. The reproduction accuracy of the IVDMRS was calculated and analyzed. The results indicate that the respiration frequency substantially affects the correlation between ultrasound and kV images, as well as the reproduction accuracy of the IVDMRS due to the system delay time (0.35s) of ultrasound imaging and signal transmission. The utilization of a phase lead compensator (PLC) reduced the error caused by this delay, thereby improving the reproduction accuracy of the IVDMRS by 14.09-46.98%. Applying the IVDMRS in clinical treatments will allow medical staff to monitor the target displacements in real time by observing the movement of the laser beam. If the target displacement moves outside the planning target volume (PTV), the treatment can be immediately stopped to ensure that healthy tissues do not receive high doses of radiation.

Environmental factors, such as electromagnetic waves, induce biological and genetic effects. One of the most important physiological systems involved with electromagnetic fields (EMFs) is the genital system. This paper reviews the effects of EMFs on human reproductive organs, female animals, fetus development and the importance of two types of natural antioxidants, i.e., vitamin E and fennel. The studies presented in this review referred to the effects of different exposures to EMFs on the reproductive system, and we tried to show the role of natural antioxidants in reducing the effects of the exposures. Many studies have been done on the effects of ionizing and non-ionizing electromagnetic waves on the cell line of spermatogenesis, sexual hormones, and the structure of the testes. Also, about the hormonal cycle, folliculogenesis and female infertility related to EMF have been given more consideration. In particular, attention is directed to pregnant women due to the importance of their fetuses. However, in addition to the studies conducted on animals, further epidemiological research should be conducted.

Paca is a wild rodent typical of the tropical region. In order to study this species with focus on the proper management and preservation, this study aims to characterize the histochemistry of the accessory glands of the reproductive tract of the male paca. For this purpose, histological sections were stained with Periodic Acid-Schiff (PAS), Alcian Blue (AB), PAS + AB e PAS + Amilase. In the histochemical reaction of the bulbouretral gland was found that the epithelium produces secretion rich in neutral and acid glycoproteins, glycosaminoglycans, and in some regions produces more than one type of secretion. The epithelium did not contain any glycogen. In the vesicular gland the epithelium had bush border stained by neutral glycoproteins, but no acid glycoproteins, glycogen or glycosaminoglycan. The coagulating gland showed small amount of neutral glycoproteins in the brush border of the epithelium, without glycogen. It is concludes that the accessory glands of paca have histochemical characteristics that share some similarity with other species of the Order Rodentia, with some glands with neutral and acid glycoproteins, mainly in the epithelium of the bulbouretral gland and in the brush border of the other glands.

Conclui-se que as glândulas anexas do trato reprodutor da paca apresentam características histoquímicas que compartilham certa similaridade com outras espécies da ordem Rodentia, com a presença de glicoproteínas neutras e ácidas em algumas glândulas, principalmente no epitélio da glândula bulbouretal e na borda em escova do epitélio das demais glândulas.

Biological invasions are generally thought to occur after human aided migration to a new range. However, human activities prior to migration may also play a role. We studied here the evolutionary genetics of introduced populations of the invasive ant Wasmannia auropunctata at a worldwide scale. Using microsatellite markers, we reconstructed the main routes of introduction of the species. We found three main routes of introduction, each of them strongly associated to human history and trading routes. We also demonstrate the overwhelming occurrence of male and female clonality in introduced populations of W. auropunctata, and suggest that this particular reproduction system is under selection in human-modified habitats. Together with previous researches focused on native populations, our results suggest that invasive clonal populations may have evolved within human modified habitats in the native range, and spread further from there. The evolutionarily most parsimonious scenario for the emergence of invasive populations of the little fire ant might thus be a two-step process. The W. auropunctata case illustrates the central role of humans in biological change, not only due to changes in migration patterns, but also in selective pressures over species.

Eriocnema fulva Naudin is a perennial herb, endemic to Minas Gerais state, SE Brazil, found on humid, shaded rocky riverbanks in montane semideciduous seasonal forests. The species is threatened, but information regarding its biology is still lacking, although such information is fundamental to any management plan. We aimed to evaluate the reproductive system of Eriocnema fulva in the Jambreiro Forest (19° 58'-59' S and 43° 55'-52' W, 800-1100 m altitude), municipality of Nova Lima, by experiments carried out in 1997 and 1998. The flowers are white, and flowering is of the steady state type, occurring once a year from November to December. Anthers are poricidal, and pollen is the only resource for visitors. The chromosome number is n = 17 during meiosis. The species is self-compatible, but does not produce fruits by spontaneous self-pollination or agamospermy; it requires pollen vectors and buzz pollination in order to produce fruits. Cross-pollination is the main reproductive strategy of E. fulva, and is accentuated by the small number of flowers (one or two in each plant) opened per day. Although the population studied was shaded by forest canopy, the seeds needed light to germinate. Germination ratio was lower in germination cabinet on filter paper (14 percent after 30 days) than in greenhouse on soil brought from the forest (47 percent after 25 days). Although the fruit is a capsule and the seeds are small, dispersion (anemochory or hydrochory) does not seem to occur at long distance, as it is the case for other Melastomataceae species with similar syndrome.


Eriocnema fulva Naudin é uma planta herbácea, perene, endêmica no estado de Minas Gerais, sendo encontrada em paredões rochosos úmidos, ao longo de riachos sombreados pela Floresta Estacional Semidecídua Montana. A espécie é ameaçada de extinção e não existem informações sobre a sua biologia, embora sejam fundamentais para um plano de manejo. Com o objetivo de avaliar o sistema de reprodução, foram feitos experimentos em 1997 e 1998 em uma população na Mata do Jambreiro (19° 58'-59' S e 43° 55'-52' W, 800-1100 m de altitude), no município de Nova Lima. As flores são brancas e a floração é do tipo steady state, ocorrendo uma vez ao ano durante novembro e dezembro. As anteras são poricidas e o pólen é a única recompensa para os visitantes. O número cromossômico encontrado na meiose foi n = 17. A espécie é autocompatível, não produz frutos por autopolinização espontânea nem por agamospermia, mas requer obrigatoriamente vetores de pólen e polinização vibrátil. A polinização cruzada é a principal estratégia reprodutiva de Eriocnema fulva, sendo acentuada devido ao pequeno número de flores abertas por dia, apenas uma ou duas em cada planta. Embora as populações estejam localizadas em ambientes sombreados e úmidos, as sementes precisam de luz para germinar, sendo fotoblásticas positivas. A taxa de germinação em papel de filtro na câmara de germinação foi menor (14 por cento após 30 dias) que em solo da floresta na casa de vegetação (47 por cento após 25 dias). Embora o fruto seja do tipo cápsula com sementes pequenas, a dispersão (anemocoria ou hidrocoria) parece não ocorrer a distâncias longas, como é observada em outras espécies de Melastomataceae com síndrome de dispersão semelhante.
