AIM: The aim of the current in vitro research was to evaluate the sealing capacity of three different agents employed for the repair of perforations at the furcation area. MATERIALS AND METHODS: Recently 60 extracted human mandibular permanent molars having well apart plus fully formed roots, and intact furcation were chosen. The 60 samples were allocated at random to three groups of 20 samples: Group I: Furcation perforation repair by means of mineral trioxide aggregate (MTA)-Angelus, Group II: Furcal perforation repair using Biodentine, Group III: Furcal perforation repair by EndoSequence. The specimens were subjected to sectioning with a hard tissue microtome and the sectioned parts of the samples were then examined. The specimens were subjected to gold sputtering and visualizing beneath scanning electron microscope (SEM) at 2000× magnification for assessing the sealing capacity of the agents. RESULTS: The highest sealing capacity was noted with the use of Biodentine at 0.96 ± 0.10, in pursuit by EndoSequence use at 1.18 ± 0.14 and MTA-Angelus use at 1.74 ± 0.08. The disparity amid the three groups was statistically significant with p < 0.001. CONCLUSION: In conclusion, it may be inferred that Biodentine exhibited the finest sealing capacity than EndoSequence and MTA- Angelus. It may thus be given consideration as a substance of preference for the repair of furcal perforation. CLINICAL SIGNIFICANCE: Using biologically compatible substances may be suggested to amend perforations thereby decreasing the occurrence of inflammatory response in the neighboring tissues. The sealing capacity is a significant feature in supporting the result of a root canal treatment of a tooth.

Blend films from chicken protein isolate (CPI) and fish skin gelatin (FSG) at various CPI/FSG ratios (100:0, 80:20, 70:30, 60:40, 0:100), prepared at pH 3 or 11 were characterized. At the same pH, tensile strength (TS) of CPI/FSG films was higher than CPI and FSG films, and CPI/FSG film (60:40) had highest TS. Moreover, elongation at break (EAB) of blend films increased as FSG content augmented. EAB of CPI film and CPI/FSG (80:20) film was similar for both pHs. CPI films generally possessed higher water vapor permeability (WVP), light barrier property and b*-value than FSG counterpart. CPI films prepared at both pHs were not sealable. Nevertheless, addition of FSG improved sealing ability of blend films. At the same CPI/FSG ratio, seal strength and seal efficiency were lower for films prepared at pH 11. Moreover, higher TS and b*-value were gained, compared to those of films prepared at pH 3. Less cracks on surface and cross-section appeared for CPI/FSG films as revealed by scanning electron microscopy images, compared to CPI and FSG films. Therefore, incorporation of FSG up to 40% into blend film was able to improve mechanical properties, WVP, and sealing ability of blend films.

BACKGROUND: Internal voids of materials can serve a hub for microorganism and affect the sealing ability. This study aimed to evaluate the sealing performance of calcium silicate-based cements in immature teeth treated with regenerative endodontics. METHODS: Twenty single root canals from immature permanent premolars were prepared using regenerative endodontic protocols. The root canals were randomly divided into two groups and sealed with mineral trioxide aggregate (MTA) and Biodentine (BD). The teeth were kept in humid environment for 7 days and scanned using micro-computed tomography. The voids within the cements were segmented and visualized using image processing, incorporating the modified Otsu algorithm. The porosity of each sample was also calculated as the ratio between the number of voxels of voids and the volume of the cements. Tortuosity was also calculated using the A-star algorithm. RESULTS: Voids larger than 70 µm were predominantly observed in the top and interfacial surface of cements. The others were evenly distributed. MTA and BD showed the same level of porosity and tortuosity at interfacial surfaces. In inner surfaces, MTA showed more less porosity and tortuosity compared to BD (p < 0.05). CONCLUSIONS: There were no differences in sealing performance between MTA and BD.

AIM: To assess the sealing ability of two calcium silicate-based materials in the treatment of iatrogenic furcal perforations using a dye-penetration leakage model. MATERIALS AND METHODS: Furcation perforations were performed using a size 12 round burr on the pulp chamber floor of 20 first mandibular molars. The teeth were then randomly divided into two groups, two additional molars served as negative controls. The defects were then filled with mineral trioxide aggregate (MTA) Angelus in the first group and Biodentine in the second group. Leakage at the repaired sites was then evaluated using the methylene blue dye penetration technique. RESULTS: Significant differences in microleakage were found between the two groups at 72 hours (p < 0.001). MTA Angelus had greater dye penetration than Biodentine with a statistically significant difference. Subsequently, the sealing ability of Biodentine was significantly better than MTA Angelus (p < 0.001). However, the mean values of leakage and inadequate adhesion were significantly different from the theoretical value for both the MTA Angelus (p < 0.001) and Biodentine (p < 0.001). CONCLUSION: The current results suggested that Biodentine possesses higher sealing quality than MTA Angelus. Yet, both materials are not ideal and still need improvement to ensure perfect adhesion in case of furcal perforation. CLINICAL SIGNIFICANCE: This article aims to compare the sealing ability of one dental repair material over another, after iatrogenically producing a furcal perforation. Leakage resistance and sealing ability are important factors in favoring the outcome of an endodontic treatment of a tooth that could otherwise be condemned for extraction.

OBJECTIVES: The aim of the present study was to evaluate the adaptation of a calcium silicate bioceramic (BC) sealer with either BC or conventional gutta-percha compared with that of AH Plus sealer in different root canal sections. MATERIALS AND METHODS: Seventy-two extracted mandibular premolars were divided randomly into six groups. After standardised chemomechanical preparation, four groups were obturated with the BC sealer and BC gutta-percha or conventional gutta-percha, and the other two groups were obturated with AH Plus sealer and conventional gutta-percha either in lateral compaction or in a single cone technique. Each root was sectioned into three sections. An impression was made from each section, and replicas were then made for scanning electron microscopy (SEM) analysis. Areas and interfacial gaps were identified using image analysis software. In addition to descriptive and explorative data analyses, linear regression analysis was performed. RESULTS: All specimens had measurable interfacial gaps. Significantly fewer gaps were found between conventional gutta-percha and sealer compared to those observed when using the BC gutta-percha (p < 0.001). However, minor interfacial gaps between sealer and dentin were observed with the BC sealer (p = 0.04). The technique of obturation in different root canal sections did not significantly affect the sealer adaptability. CONCLUSION: The type of gutta-percha as well as the sealer had a noticeable impact on the adaptability. CLINICAL RELEVANCE: Different obturation techniques will result in similar outcomes. However, within the limitations of the study, there seems to be no advantage in using the BC gutta-percha.

AIM: The aim of the present study was to compare the shear bond strength and marginal sealing ability of self-adhering flow-able composite and conventional fissure sealant. MATERIALS AND METHODS: The samples consisted of 30 healthy premolar teeth which were extracted due to orthodontic reasons and randomly divided into two groups of 15, i.e., group I (Fissurit F) and group II (Dyad Flow). Shear bond strength and marginal sealing ability of both the groups were evaluated in Statistical Package for the Social Sciences (SPSS) version 16. RESULTS: The mean shear bond strength of Dyad Flow (group II) was found to be 1.4 ± 0.87 MPa and in Fissurit F (group I), it was 1.3 ± 1.4 MPa. Differences between the groups were statistically significant. In group II, 53.3% of specimens demonstrated score 0; 33.3% showed score 1; and 13.3% showed score 2. In group I, scores 0 and 1 showed 33.3% of dye penetration respectively. Scores 2 and 3 demonstrated 26.6 and 6.6% of dye penetration respectively. But there was no significant difference between both the sealant groups. CONCLUSION: The present study concluded that self-adhering flowable composite was found to have better shear bond strength and marginal sealing ability than conventional fissure sealant. CLINICAL SIGNIFICANCE: Self-adhering flowable composite can be effectively used in pediatric patients in whom isolation is difficult and exclusion of bonding agent leads to decrease in time consumption.

OBJECTIVES: The aim of the present study is to evaluate the possible correlation between sealer penetration into dentinal tubules and sealing ability both in presence and absence of smear layer. MATERIALS AND METHODS: Fourteen maxillary central incisors were treated with 5.25 % NaOCl +10 % EDTA to remove the smear layer (SL-free group) or 5.25 % NaOCl without EDTA (SL group). Root canals were filled using #25 Thermafil Obturators with Topseal sealer labelled with 0.1 wt% rhodamine B. Sealing ability was measured as fluid filtration rate with a fluid-flow meter using water supplemented with 0.3 % calcein fluorescent dye. Specimens were sectioned, observed under confocal microscope to co-localize the presence of sealer (rhodamine B labelling) into dentinal tubules and gaps (calcein labelling) into the root canal. The depth of sealer penetration into dentinal tubules and the percentage of sealer penetration around the root canal were measured at 3, 5 and 8 mm from the apex. RESULTS: No significant differences between groups were observed in fluid filtration rate nor in depth of calcein penetration. Sealer penetration depth and percentage into dentinal tubules were not significantly different between groups, except at 8-mm level in absence of smear layer. CONCLUSION: Sealer penetration at 3- and 5-mm levels was not influenced by smear layer while it was significantly reduced at 8-mm level. Fluid filtration rate was not correlated either with depth of calcein penetration nor with sealer penetration into dentinal tubules. CLINICAL RELEVANCE: The sealing ability of Topseal sealer is not affected by presence or absence of smear layer.

PURPOSE: The micropercolation testing of dental materials is currently used to evaluate the sealing ability. The aim of the present study is to compare the sealing ability of distinct assembling materials for zirconium oxide peripheric crowns. MATERIALS AND METHODS: One hundred teeth were prepared to be thermocycled after being distributed at random in 10 groups each containing 10 specimens (group 1: Fuji One; group 2: Fuji Plus; group 3: Variolink II; group 4: RelyX Ultimate; group 5: Multilink; group 6: Panavia F2.0; group 7: Bifix QM; group 8: RelyX Unicem 2; group 9: Maxcem Elite; group 10: Bifix SE). RESULTS: RelyX Unicem 2 and RelyX Ultimate yielded high imperviousness percentages. The sealing ability in the vestibular area and lingual area is indeed best in the presence of these two materials. Our results show no difference whether the chamfer is situated in the enamel or in the dentin. CONCLUSION: A zirconium crown should be preferably cemented with a glass-ionomer cement modified by addition of resin material.

OBJECTIVES: The aim of this study was to evaluate the sealing ability and morphological microstructure of Biodentine in comparison to ProRoot mineral trioxide aggregate (MTA) after storage in an acidic environment. MATERIALS AND METHODS: Biodentine and ProRoot MTA were prepared and packed into the canal lumen of dentin disks. Twenty specimens of each material were further randomly divided into two groups according to the storage media: group A: materials with saline as storage medium; group B: materials with citric acid buffered at pH 5.4 as storage medium. The sealing ability was evaluated at 1, 3, 6, and 24 h and 1 or 3 months, using a fluid transport model for quantitative analysis of endodontic microleakage. The morphological microstructures of the materials were also evaluated using scanning electron microscopy. RESULTS: During the first 24 h, MTA showed greater fluid transport values than Biodentine in both environments. At the 3-month measurement, when the materials were stored in saline, MTA showed greater ability to prevent fluid movement than Biodentine (p < 0.0001). However, when the materials were stored in an acidic environment, no statistical significant difference was found after 3 months. After storage in saline, both materials showed an uneven crystalline surface with similar hexagonal crystals. The microstructure of Biodentine changed after exposure to citric acid, showing a relatively smooth surface with more spheroidal crystals. CONCLUSIONS: The exposure to an acidic environment, within the limits of this study, seems to result in morphological changes of Biodentine in a different manner than MTA. MTA shows good ability to prevent fluid movement over time, in both environments. The ability of Biodentine to prevent fluid movement over time was enhanced in the acidic environment. CLINICAL RELEVANCE: The findings of the present study could imply that both materials are indicated for use in an acidic environment.

AIM: To characterize three radiopaque Magnesium Phosphate Cements (MPCs) developed for endodontic purposes. METHODOLOGY: Three experimental MPCs containing Bi2 O3 were formulated. The experimental cements, which consisted of mixtures of magnesium oxide with different phosphate salts, were characterized for setting time, injectability, porosity, compressive strength and phase composition. The long-term sealing ability of the experimental MPCs applied in single-rooted teeth as root canal filling material or as sealer in combination with gutta-percha was also assessed using a highly sensitive fluid filtration system. A mineral trioxide aggregate (MTA) cement was used as control. Statistical analysis was performed with two- or three-way analysis of variance (anova) and Tukey's test was used for comparisons. RESULTS: The addition of 10 wt% Bi2 O3 within the composition of the MPCs provided an adequate radiopacity for endodontic applications according to ISO 6876 standard. The reaction products resulting from the MPCs were either struvite (MgNH4 PO4 ·6H2 O) or an amorphous sodium magnesium phosphate. The porosity of the three MPCs ranged between 4% and 11%. The initial setting time of the experimental cements was between 6 and 9 min, attaining high early compressive strength values (17-34 MPa within 2 h). All MPC formulations achieved greater sealing ability than MTA (P < 0.05) after 3 months, which was maintained after 6 months for two of the experimental cements (P < 0.05). CONCLUSIONS: These MPCs had adequate handling and mechanical properties and low degradation rates. Furthermore, a stable sealing ability was demonstrated up to 6 months when using the cement both as root filling material and as sealer in conjunction with gutta-percha.

Mineral trioxide aggregate (MTA) has been used successfully for the treatment of necrotic immature teeth as part of a 2-step apical barrier. Recently, some researchers have used MTA powder instead of wet-mix MTA. It has also been used in a single-step approach in order to decrease the number of patient appointments. The aim of this study was to evaluate the bacterial microleakage of MTA powder in comparison with wet-mix MTA, in single or 2-step barrier approaches. This ex vivo study was carried out on 70 extracted single-rooted teeth. The canals were enlarged to simulate immature teeth. The prepared specimens were assigned to 4 experimental (n = 15) and 2 control (n = 5) groups. In Group 1, a wet paper point was placed in the canal following placement of wet-mix MTA. In Group 2, a dry paper point was placed on wet-mix MTA. In Group 3, the samples were treated similar to Group 1, but MTA powder was used instead of wet-mix MTA. In Group 4, the same procedures were followed as Group 3 except that a dry paper point was used. Microleakage of the samples was compared using a 2-chamber bacterial leakage apparatus. Results showed that there were no significant differences in the number of leaked samples or rate of leakage over time (P > 0.05) among the experimental groups. It can be concluded that MTA used in a single step approach has the same resistance against bacterial penetration as in a 2-step approach, and MTA powder provides the same sealing ability as wet-mix MTA.

This in vitro study sought to evaluate the sealing ability of 3 root canal obturator systems after immersion in simulated body fluid for 1 year. The coronal sections of 30 single-rooted teeth were removed at the cementoenamel junction at 12 mm (±1 mm), and roots were instrumented with nickel titanium instruments. Specimens were divided into 3 groups (n = 10) according to the obturation manufacturer-specified system and immersed in simulated body fluid. A digital fluid flow-meter was used to detect the flow rate at 1 week, 1 month, and 12 months after immersion. This study demonstrated that the tested endodontic obturation systems were unable to keep their sealing ability stable during the first year. At 12 months, all root filling systems showed increased flow rates (P < 0.0001).

This study sought to investigate how pretreatment intracanal placement of calcium hydroxide (Ca(OH)2) affected the sealing ability of mineral trioxide aggregate (MTA) plugs in simulated open apices. Maxillary central incisor teeth were cleaned and shaped in an identical manner. After root-end resection, samples were divided randomly into 2 control (n = 5) and 2 experimental groups (n = 35). The specimens in Group 1 were pretreated using a Ca(OH)2 medicament, while the samples of Group 2 received no medicament. One week later, a 5 mm apical plug of MTA was applied into the canals. The remaining portions of the canals were obturated with gutta percha. Group 1 had a significantly higher frequency of apical leakage than Group 2 (P < 0.05).

This study investigated the wettability and consistency of various endodontic sealers, both inorganic and organic, and evaluated their sealing ability of root canals using the single-cone obturation technique, with and without ethylenediaminetetraacetic acid (EDTA) treatment. Bovine root canals were endodontically prepared and filled in preparation for the dye penetration test with toluidine blue solution. All sealers exhibited contact angles similar to or lower than dentin and displayed superior consistency. Among the sealers, organic sealers used without EDTA treatment showed reduced dye penetration compared to inorganic sealers. However, some inorganic and organic sealers showed dye penetration in the sealer and dentin of root canals subjected to EDTA treatment. In conclusion, the single-cone obturation technique, combined with these endodontic sealers, achieved close contact with root canal dentin due to their wettability and consistency. However, the sealing ability of certain sealers was influenced by EDTA treatment.

Background: Different obturation techniques are being used to improve canal sealing, thereby minimizing marginal leakage. Aims and Objectives: To evaluate the marginal leakage of three different obturating materials and techniques using stereomicroscope. Materials and Methods: Thirty freshly extracted maxillary anterior teeth were collected and were divided into three groups of 10 each for which the biomechanical preparation was done. The teeth were divided into three experimental groups according to the obturation material and technique used, Group I: gutta-percha with AH Plus root canal sealer (lateral condensation): Group II: thermoplasticized gutta-percha technique-non-carrier-based (calamus) with AH Plus sealer (backfill): Group III: C point (self-sealing root canal obturating system) with bioceramic sealer (single cone). Each specimen was subjected for testing apical sealing ability. The values of microleakage associated with different root canal sealers were evaluated using a stereomicroscope. Results: There was no statistically significant difference in the mean scores of apical dye penetration/leakage among all the three groups (P = 0.091), whereas Group I showed a higher degree of apical leakage than the other techniques tested. Conclusion: None of the tested filling materials provided a gap-free or void-free root canal filling; however, thermoplasticized gutta-percha is used with AH Plus sealer exhibited less voids and gaps when compared to other tested materials.

Background/purpose: Iontophoresis could enhance the delivery of chlorhexidine into oral tissue. This study aimed to determine the effect of 2% chlorhexidine iontophoresis (CHI) on the sealing ability of etch-and-rinse adhesive in human dentin using hydraulic conductance (HD) measurement, scanning electron microscopy and energy dispersive x-ray spectroscopy (SEM-EDS). Materials and methods: Thirty-nine sound dentin specimens were prepared from 39 extracted intact third molars. Thirty specimens were used for HD measurement and randomly divided into 3 equal-sized groups; (1) No chlorhexidine treatment (control), (2) passive chlorhexidine treatment (CHT) and (3) CHI on acid-etched dentin. Each dentin surface was treated with etch-and-rinse adhesive. HD of each specimen was measured before treatment, after immediate bonding and after 14 days. The other 9 specimens were subjected to SEM-EDS analysis of the acid-etched dentin and the dentin treated with CHT and CHI. ANOVA test and Student-Newman-Keuls method were used for statistical analysis. Results: After bonding, there was no significant difference in percentage decrease of HD among the treatment groups (P > 0.05). After 14 days, CHI and CHT groups had greater percentage decrease of HD than the control (P < 0.001 and P = 0.009, respectively). Under SEM-EDS analysis, acid-etched dentin with CHI presented opened dentinal tubule orifices and more chlorhexidine precipitates on dentin than the dentin with CHT, which strongly related to a higher percentage of chloride ions on the CHI dentin surface (P < 0.001). Conclusion: The use of CHI on acid-etched dentin had a positive effect on dentin sealing ability of etch-and-rinse adhesive.

This study aimed to compare the nanoleakage of retrograde fillings with premixed calcium silicate-based putty and mineral trioxide aggregate (MTA), using two different techniques (traditional and Lid). Sixty-four extracted human teeth were decoronated, then root canals and ends were instrumented for retrograde filling and divided into four groups according to the retrograde filling technique: the traditional and the Lid technique. Each group (n = 15) was filled with Ceraseal + Well-Root putty, Well-Root putty, Ceraseal + ProRoot MTA, and ProRoot MTA. The nanoleakage was evaluated using the Nanoflow device (IB Systems) on days 1, 3, 7, 15 and 30. Data were collected twice per second at the nanoscale (nL/s) and calculated after archiving the stabilization of fluid flow. The Kruskal-Wallis and Mann-Whitney U-tests were used for statistical analysis. All groups showed enhanced sealing ability over time. Regardless of filling materials, the Well-Root putty, Ceraseal+Well-Root putty, and Ceraseal+ProRoot MTA groups indicated less nanoleakage than the ProRoot MTA group in the first week of evaluation (p < 0.05). Although all groups did not show significant differences after 2 weeks, the Ceraseal+ProRoot MTA group leaked less than ProRoot MTA on Days 3 and 7 (p < 0.05). The scanning electron microscopic examined good adaptation to the cavity wall, which was similar to nanoleakage results. Premixed calcium silicate-based putty retrograde filling material alone and using the "lid technique" were shown to be faster and less prone to nanoleakage when compared to MTA.

Background: Root perforation repair presents a significant challenge in dentistry due to inherent limitations of existing materials. This study explored the potential of a novel polydopamine-based composite as a root repair material by evaluating its sealing efficacy, radiopacity, and surface topography. Methods: Confocal microscopy assessed sealing ability, comparing the polydopamine-based composite to the gold standard, mineral trioxide aggregate (MTA). Radiopacity was evaluated using the aluminium step wedge technique conforming to ISO standards. Surface roughness analysis utilized atomic force microscopy (AFM), while field emission scanning electron microscopy (FESEM) visualized morphology. Results: The polydopamine-based composite exhibited significantly superior sealing efficacy compared to MTA (P < 0.001). Radiopacity reached 3 mm aluminium equivalent, exceeding minimum clinical requirements. AFM analysis revealed a smooth surface topography, and FESEM confirmed successful composite synthesis. Conclusion: This study demonstrates promising properties of the polydopamine-based composite for root perforation repair, including superior sealing efficacy, clinically relevant radiopacity, and smooth surface topography. Further investigation is warranted to assess its clinical viability and potential translation to endodontic practice.

Background/purpose: Sealing ability in root canal obturation has always been a key concern for endodontic success. The purpose of this study was to analyze the percentage of voids in root canal space obturated by using single cone hydraulic condensation with different root canal sealers and to compare those with AH Plus sealer. Materials and methods: Experiments were conducted using twenty 3D-printed upper first premolars. After the buccal root canals were prepared using Ni-Ti rotary instruments, the teeth were divided into four groups: the AH Plus, BC sealer, BC sealer HiFlow, and Endoseal MTA groups. All buccal canals were obturated by single-cone hydraulic condensation. All specimens were scanned using micro-computed tomography and the percentage volume of the voids inside and outside the filled materials (Vin and Vout) at three different canal depth intervals were calculated by a Bruker micro-CT software. Differences according to root canal sealers were evaluated statistically using the Kruskal-Wallis test and the Wilcoxon Rank Sum test at a significance level of 0.05. Results: The results indicated that most of the voids were presented near the interface (Vout), the Vin is very small and not significant different between groups. The Vout decreased in the following order: AH Plus(1.837% ± 1.226%)≅BC sealer (1.225% ± 0.836%)>BC sealer Hiflow(0.349% ± 0.071%)>Endoseal MTA(0.203% ± 0.049%). Conclusion: For the percentage volume of voids between the root canal filling material and root canal surface, though the BC sealer Hiflow is slightly larger than Endoseal MTA, which is still much less than BC sealer and AH Plus.

(1) Background: Coronal microleakage can lead to endodontic treatment failure. This study aimed to compare the sealing ability of different temporary restorative materials used during endodontic treatment. (2) Methods: Eighty sheep incisors were collected, uniformized in length, and access cavities were performed, except for in the negative control group, where the teeth were left intact. The teeth were divided into six different groups. In the positive control group, the access cavity was made and left empty. In the experimental groups, access cavities were restored with three different temporary materials (IRM®, Ketac™ Silver, and Cavit™) and with a definitive restorative material (Filtek Supreme™). The teeth were submitted to thermocycling, and two and four weeks later, they were infiltrated with 99mTcNaO4, and nuclear medicine imaging was performed. (3) Results: Filtek Supreme™ obtained the lowest infiltration values. Regarding the temporary materials, at two weeks, Ketac™ Silver presented the lowest infiltration, followed by IRM®, whereas Cavit™ presented the highest infiltration. At four weeks, Ketac™ Silver remained with the lowest values, whereas Cavit™ decreased the infiltration, comparable to IRM®. (4) Conclusion: Regarding temporary materials, Ketac™ Silver had the lowest infiltration at 2 and 4 weeks, whereas the highest infiltration was found in the Cavit™ group at two weeks and in the IRM® group at 4 weeks.
