RESUMO

Long-read sequencing technologies have now reached a level of accuracy and yield that allows their application to variant detection at a scale of tens to thousands of samples. Concomitant with the development of new computational tools, the first population-scale studies involving long-read sequencing have emerged over the past 2 years and, given the continuous advancement of the field, many more are likely to follow. In this Review, we survey recent developments in population-scale long-read sequencing, highlight potential challenges of a scaled-up approach and provide guidance regarding experimental design. We provide an overview of current long-read sequencing platforms, variant calling methodologies and approaches for de novo assemblies and reference-based mapping approaches. Furthermore, we summarize strategies for variant validation, genotyping and predicting functional impact and emphasize challenges remaining in achieving long-read sequencing at a population scale.

Loss of gut microbial diversity1-6 in industrial populations is associated with chronic diseases7, underscoring the importance of studying our ancestral gut microbiome. However, relatively little is known about the composition of pre-industrial gut microbiomes. Here we performed a large-scale de novo assembly of microbial genomes from palaeofaeces. From eight authenticated human palaeofaeces samples (1,000-2,000 years old) with well-preserved DNA from southwestern USA and Mexico, we reconstructed 498 medium- and high-quality microbial genomes. Among the 181 genomes with the strongest evidence of being ancient and of human gut origin, 39% represent previously undescribed species-level genome bins. Tip dating suggests an approximate diversification timeline for the key human symbiont Methanobrevibacter smithii. In comparison to 789 present-day human gut microbiome samples from eight countries, the palaeofaeces samples are more similar to non-industrialized than industrialized human gut microbiomes. Functional profiling of the palaeofaeces samples reveals a markedly lower abundance of antibiotic-resistance and mucin-degrading genes, as well as enrichment of mobile genetic elements relative to industrial gut microbiomes. This study facilitates the discovery and characterization of previously undescribed gut microorganisms from ancient microbiomes and the investigation of the evolutionary history of the human gut microbiota through genome reconstruction from palaeofaeces.

PURPOSE OF REVIEW: Modernization and Westernization in industrialized and developing nations is associated with a substantial increase in chronic noncommunicable diseases. This transformation has far-reaching effects on lifestyles, impacting areas such as economics, politics, social life, and culture, all of which, in turn, have diverse influences on public health. Loss of contact with nature, alternations in the microbiota, processed food consumption, exposure to environmental pollutants including chemicals, increased stress and decreased physical activity jointly result in increases in the frequency of inflammatory disorders including allergies and many autoimmune and neuropsychiatric diseases. This review aims to investigate the relationship between Western lifestyle and inflammatory disorders. RECENT FINDINGS: Several hypotheses have been put forth trying to explain the observed increases in these diseases, such as 'Hygiene Hypothesis', 'Old Friends', and 'Biodiversity and Dysbiosis'. The recently introduced 'Epithelial Barrier Theory' incorporates these former hypotheses and suggests that toxic substances in cleaning agents, laundry and dishwasher detergents, shampoos, toothpastes, as well as microplastic, packaged food and air pollution damage the epithelium of our skin, lungs and gastrointestinal system. Epithelial barrier disruption leads to decreased biodiversity of the microbiome and the development of opportunistic pathogen colonization, which upon interaction with the immune system, initiates local and systemic inflammation. Gaining a deeper comprehension of the interplay between the environment, microbiome and the immune system provides the data to assist with legally regulating the usage of toxic substances, to enable nontoxic alternatives and to mitigate these environmental challenges essential for fostering a harmonious and healthy global environment.

Microplastics (MPs), plastic particles smaller than 5 mm, are now a growing environmental and public health issue, as they are detected pervasively in freshwater and marine environments, ingested by organisms, and then enter the human body. Industrial development drives this environmental burden caused by MP formation and human uptake by elevating plastic pollution levels and shaping the domestic dietary structure. We map the MP human uptake across 109 global countries on five continents from 1990 to 2018, focusing on the world's major coastlines that are affected by plastic pollution that affects the United Nations' Sustainable Development Goals (SDGs): SDG 6 (Clean Water and Sanitation), SDG 14 (Life Below Water), and SDG 15 (Life on Land). Amid rapid industrial growth, Indonesia tops the global per capita MP dietary intake at 15 g monthly. In Asian, African, and American countries, including China and the United States, airborne and dietary MP uptake increased over 6-fold from 1990 to 2018. Eradicating 90% of global aquatic plastic debris can help decrease MP uptake by more than 48% in Southeast Asian countries that peak MP uptake. To reduce MP uptake and potential public health risks, governments in developing and industrialized countries in Asia, Europe, Africa, and North and South America should incentivize the removal of free plastic debris from freshwater and saltwater environments through advanced water treatment and effective solid waste management practices.

China's dairy farming is undergoing a critical transition from extensive to industrial systems. To achieve sustainable milk production within China's dual-carbon goals, understanding the multidimensional impacts of industrialization on greenhouse gas (GHG) emissions is imperative. This study comprehensively analyzed the implications of China's dairy industrialization on GHG emissions and explored future mitigation potential. Results indicated that industrial systems exhibited lower methane but higher carbon dioxide intensities, with net GHG intensity lower than other systems. During 2002-2020, China's milk production increased by 165%, while GHG emissions increased by 105% to 50.27 Tg CO2eq, accompanying an industrialization rate increased from 16% to 75%. The industrialization progress played a mitigating effect on GHG primarily through intensification within individual production systems before 2008 and transformation between systems post-2008. However, the industrialization's effect was relatively modest compared to other socio-economic factors. By 2030, 11.8 Tg CO2eq will be triggered by predicted milk production growth, but only 0.6 Tg can be offset by system transformation. Integrating measures to improve feed, herd, and manure management on industrial farms could decouple GHG emissions from milk production and achieve a carbon peak before 2030. We suggest transforming to improved industrial systems as a necessary step toward sustainable livestock production.

Approximately 74 thousand years ago (ka), the Toba caldera erupted in Sumatra. Since the magnitude of this eruption was first established, its effects on climate, environment and humans have been debated. Here we describe the discovery of microscopic glass shards characteristic of the Youngest Toba Tuff-ashfall from the Toba eruption-in two archaeological sites on the south coast of South Africa, a region in which there is evidence for early human behavioural complexity. An independently derived dating model supports a date of approximately 74 ka for the sediments containing the Youngest Toba Tuff glass shards. By defining the input of shards at both sites, which are located nine kilometres apart, we are able to establish a close temporal correlation between them. Our high-resolution excavation and sampling technique enable exact comparisons between the input of Youngest Toba Tuff glass shards and the evidence for human occupation. Humans in this region thrived through the Toba event and the ensuing full glacial conditions, perhaps as a combined result of the uniquely rich resource base of the region and fully evolved modern human adaptation.

Industrial development has resulted in economic progress and the well-being of the society. At the same time, the impact of the industrial complex has disrupted the environment and resulted in climate change related impacts. The purpose of this study was to carry out an exploratory diagnosis and propose a technological change and sustainable industrial development index at the international level. Therefore, a network study was conducted to identify the main nodes and thematic clusters associated with cleaner production. A patent analysis was applied to technologies related three selected/relevant areas of cleaner production, i.e. carbon footprint, wastewater treatment, and renewable energy. Additionally, based on factor analysis, an index including different indicators related to scientific, technological, economic, environmental, and social issues was developed and proposed in this study.

The Discovery to Innovation in Animal Health Conference (DIAH) was organised to bridge the gap between early developers, including academia, regulators, research organizations, and spin-offs/start-ups on one side, and medium-to large-sized companies on the other. The DIAH Conference confronted and aligned vision from academia, industry and regulators, emphasizing the need for early collaboration, careful IP management, and strategic planning for successful product development and partnerships. Recent breakthroughs in vaccinology have not only accelerated the vaccine production process but have also improved antigen quality significantly. These novel technologies are likely to transform vaccine development and play a crucial role in addressing both immediate health challenges (such as cancer vaccines) and ensuring preparedness for future pandemics. The potential and pitfalls of leveraging AI to drive forward R&I activities in the field of animal health were also discussed. Researchers and entrepreneurs looking for collaboration or investment presented a series of new technologies and start-ups, respectively. A market analysis showed that the animal health industry, while highly consolidated, also shows great diversity, ranging from big pharma to companies offering diagnostics, nutritional health services, wearables, feed additives, animal feed and genetic analyses. An analysis of the investment landscape, although subject to external factors, showed that the chances for success are high when good science, a well established regulatory pathways, with a clearly defined market need can be combined with experienced management and a strong investor consortium.

BACKGROUND: Health tourism is an important component that may influence the direction of development in a region. Mazandarn, Iran, is recognized as a highly sustainable market in the region because of its abundant natural resources, temperate temperature, and strategic geographical location. Therefore, considering the importance of health tourism and its existing potential in Mazandaran, Iran, this study was conducted with the aim of exploring a comprehensive perspective on local drivers in community-based health tourism industry development and factors affecting the attraction of health tourism. METHODS: We conducted this study in Mazandarn, Iran, using a qualitative approach. Participants included a sample of Iranian people, aged 34-54 years, with previous history of health tourism or expert in it. Participants were selected from three different categories of the community: academic professionals in health tourism, managers in health tourism, and health tourists. Data were obtained via semi-structured in-depth interviews and focus group discussions. Inductive qualitative content analysis was used to converge and compare themes through participant data. The interviews kept going until data saturation was achieved. RESULTS: Based on our findings, we distilled local drivers in community-based health tourism industry development into five main categories and 30 subcategories: (Sharifabadi AM, Ardakani FA. The findings of the study showed that, from the participants' point of view, although there are several strategies, such as the development of public service and tourism infrastructure, increasing tourist attractions, and formulating appropriate policies and procedures for the development of health tourism, they are also faced with many challenges, especially political, economic, and cultural challenges. CONCLUSION: This study showed that improving infrastructure and resources, promoting tourist attractions, informing socio-cultural contexts, improving economic and financial capacity, and developing political and communicative contexts might increase the attraction of health tourists. The suggested components are not contextually driven, although empirical outcomes may differ based on the level of service offerings in health tourism locations.

The so-called "hard-to-transfect cells" are well-known to present great challenges to intracellular delivery, but detailed understandings of the delivery behaviors are lacking. Recently, we discovered that vesicle trapping is a likely bottleneck of delivery into a type of hard-to-transfect cells, namely, bone-marrow-derived mesenchymal stem cells (BMSCs). Driven by this insight, herein, we screened various vesicle trapping-reducing methods on BMSCs. Most of these methods failed in BMSCs, although they worked well in HeLa cells. In stark contrast, coating nanoparticles with a specific form of poly(disulfide) (called PDS1) nearly completely circumvented vesicle trapping in BMSCs, by direct cell membrane penetration mediated by thiol-disulfide exchange. Further, in BMSCs, PDS1-coated nanoparticles dramatically enhanced the transfection efficiency of plasmids of fluorescent proteins and substantially improved osteoblastic differentiation. In addition, mechanistic studies suggested that higher cholesterol content in plasma membranes of BMSCs might be a molecular-level reason for the greater difficulty of vesicle escape in BMSCs.

This research investigates the intricate relationships between economic variables and how they affect South Asian nation's ability to develop sustainably. Given the growing concerns about climate change and global warming brought on by emissions of greenhouse gases, this study looks into the connection between emissions of CO2, green energy, industrialization, foreign direct investment, economic globalization, and financial development from 1995 to 2022. Second-generation panel techniques were employed in this study to look at the relationship between variables because of the potential of residual cross-sectional dependency and heterogeneity. The empirical outcomes display that green energy, economic globalization, and financial development reduce CO2 emissions by 1.839%, 1.223%, and 3.902% respectively. Industrialization and foreign direct investment degrade the environment by 4.302% and 1.893% respectively. A bidirectional causality link between green energy, industrialization, economic globalization, and CO2 emissions was found by Dumitrescu and Hurlin (D-H). Based on our findings, we recommend legislative support for renewable energy, cleaner technologies, and strict environmental regulations, aligning with the Sustainable Development Goals (SDGs). Encouraging FDI, sustainable practices, and financial development can drive economic growth while preserving the environment. As we approach COP28, this holistic approach to sustainable development becomes increasingly vital for South Asian countries to achieve their SDG targets and combat climate change.

Digital industrialization represented by big data provides substantial support for the high-quality development of the digital economy, but its impact on urban energy conservation development requires further research. To this end, based on the panel data of Chinese cities from 2010 to 2019 and taking the establishment of the national big data comprehensive pilot zone (NBDCPZ) as a quasi-natural experiment, this paper explores the impact, mechanism, and spatial spillover effect of digital industrialization represented by big data on urban energy conservation development using the Difference-in-Differences (DID) method. The results show that digital industrialization can help achieve urban energy conservation development, which still holds after a series of robustness tests. Mechanism analysis reveals that digital industrialization impacts urban energy conservation development by driving industrial sector output growth, promoting industrial upgrading, stimulating green technology innovation, and alleviating resource misallocation. Heterogeneity analysis indicates that the energy conservation effect of digital industrialization is more significant in the central region, intra-regional demonstration comprehensive pilot zones, large cities, non-resource-based cities, and high-level digital infrastructure cities. Additionally, digital industrialization can promote energy conservation development in neighboring areas through spatial spillover effect. This paper enriches the theoretical framework concerning the relationship between digital industrialization and energy conservation development. The findings have significant implications for achieving the coordinated development of digitalization and conservation.

The eastern coastline of Gresik, located in East Java, Indonesia, experienced significant industrialization, leading to the development of numerous diverse sectors. These diverse industrial activities, in addition to other human activities, result in the contamination of sediment across the eastern coast of Gresik with a variety of metals. Metals like arsenic (As), cadmium (Cd), copper (Cu), and zinc (Zn) have exceeded the international standards for sediment quality, potentially causing significant harm to the aquatic ecosystem in this coastal region. The results of the multivariate analysis indicate that the metals found in the sediment are related to a combination of anthropogenic inputs, specifically those originating from industrial effluents in the area under study. Based on the assessment of enrichment factor, contamination factor, geo-accumulation index, degree of contamination, ecological risk index, and pollution load index, it can be concluded that the metals examined displayed different degrees of sediment contamination, ranging from minimal to severely contaminated.

RESUMO

Scholarship on Latin America's history of technology has expanded significantly in recent years. By reviewing articles in English- and Spanish-language journals from 2012 to the first half of 2023, we illustrate the emerging themes, geographies, and methodologies in this literature. The four main themes we identify are industrialization, institutions and policies, infrastructure, and moving beyond technological adaptation. We also highlight two emerging themes: Indigenous technologies and the circulation of knowledge. We conclude that the scholarship has generally moved in three directions: the study of technologies associated with traditional economic activities in the region (e.g., monocrop agriculture), national industrialization and modernization processes, and cases that demonstrate alternative ways of knowing the world and how communities use these types of knowledge. We suggest that deepening the connections between these three lines of research could be fruitful for future work.

Replacing fossil fuels with energy sources and carriers that are sustainable, environmentally benign, and affordable is amongst the most pressing challenges for future socio-economic development. To that goal, hydrogen is presumed to be the most promising energy carrier. Electrocatalytic water splitting, if driven by green electricity, would provide hydrogen with minimal CO2 footprint. The viability of water electrolysis still hinges on the availability of durable earth-abundant electrocatalyst materials and the overall process efficiency. This review spans from the fundamentals of electrocatalytically initiated water splitting to the very latest scientific findings from university and institutional research, also covering specifications and special features of the current industrial processes and those processes currently being tested in large-scale applications. Recently developed strategies are described for the optimisation and discovery of active and durable materials for electrodes that ever-increasingly harness first-principles calculations and machine learning. In addition, a technoeconomic analysis of water electrolysis is included that allows an assessment of the extent to which a large-scale implementation of water splitting can help to combat climate change. This review article is intended to cross-pollinate and strengthen efforts from fundamental understanding to technical implementation and to improve the 'junctions' between the field's physical chemists, materials scientists and engineers, as well as stimulate much-needed exchange among these groups on challenges encountered in the different domains.

The current important limitations to the implementation of Evidence-Based Practice (EBP) in the rehabilitation field are related to the validation process of interventions. Indeed, most of the strict guidelines that have been developed for the validation of new drugs (i.e., double or triple blinded, strict control of the doses and intensity) cannot-or can only partially-be applied in rehabilitation. Well-powered, high-quality randomized controlled trials are more difficult to organize in rehabilitation (e.g., longer duration of the intervention in rehabilitation, more difficult to standardize the intervention compared to drug validation studies, limited funding since not sponsored by big pharma companies), which reduces the possibility of conducting systematic reviews and meta-analyses, as currently high levels of evidence are sparse. The current limitations of EBP in rehabilitation are presented in this narrative review, and innovative solutions are suggested, such as technology-supported rehabilitation systems, continuous assessment, pragmatic trials, rehabilitation treatment specification systems, and advanced statistical methods, to tackle the current limitations. The development and implementation of new technologies can increase the quality of research and the level of evidence supporting rehabilitation, provided some adaptations are made to our research methodology.

Combating climate change and reducing carbon dioxide emissions are serious challenges shared by countries around the world. In the current era, digitalization has a significant impact on CO2 emissions. However, prior studies have not assessed the synergy between digitalization and industrialization on carbon emission performance. The principal component analysis and non-radial directional distance function (NDDF) are used to measure the digitalization and total factor carbon emission performance of Chinese 245 prefecture-level cities from 2003 to 2019. This study establishes a fixed effects model to study the panel data. The findings are as follows: (1) Digitalization can significantly promote Chinese cities' CO2 emission reduction. This result still holds after several robustness checks. (2) The heterogeneity results indicate that digitalization mainly improves central cities' carbon emission performance. Meanwhile, the impact of digitalization is more obvious after 2011. (3) Digitalization improves urban carbon emission performance through energy efficiency, industrial transformation, and technological innovation. (4) It is worth noting that digitalization synergizes with industrialization to improve carbon emission performance in Chinese cities. This study provides empirical evidence and some constructive policy recommendations for the government to push the collaborative development of the digitalization and low-carbon economy.

RESUMO

Objectification is a real problem in medicine that can lead to bad medical practice or, in the worst case, dehumanization of the patient. Nevertheless, objectification also plays a major and necessary role in medicine: the patient's body should be viewed as a biological organism in order to find diseases and be able to cure them. Listening to the patient's illness story should not be replaced, but, indeed, developed by the physical examination of his body searching for the causes of his complaints. Whereas phenomenologists have so far mainly been identifying the back sides of objectification in medicine, in this paper the aim is to analyze differences between detrimental objectifications and objectifications that do not deprive the patient of his subjectivity but, rather, at least in some cases, may lead the patient to feel more at home with his body.

RESUMO

Currently, many research and technology development projects with older people are described as "participatory." This literature-based article highlights patterns in current approaches to participatory research and technology development with older and old people. To this end, various strands of argumentation and historical lines of development are examined as well as the democracy-oriented model and the consumption-oriented approach to participation. This article goes on to set out the opportunities, challenges and criticisms associated with participatory projects. Finally, topics for future research and technology developments with older and old people are named and the questions which should be critically reflected upon when designing a participatory project are described.
