BACKGROUND: Medicinal plants are rich source of traditional herbal medicine around the globe. Most of the plant's therapeutic properties are due to the presence of secondary bioactive compounds. METHODS: The present study analyzed the High Pressure Liquid Chromatography (HPLC) fractions of Puncia granatum (peel) extracts (aqueous, chloroform, ethanol and hexane) against multidrug resistant bacterial pathogens (Acinetobacter baumannii, Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus). All the fractions having antibacterial activity was processed for bioactive compounds identification using LC MS/MS analysis. RESULTS: Among total HPLC fractions (n = 30), 4 HPLC fractions of P. granatum (peel) showed potential activity against MDR pathogens. Fraction 1 (F1) and fraction 4 (F4) collected from aqueous extract showed maximum activity against P. aeruginosa. Fraction 2 (F2) of hexane showed antibacterial activity against three pathogens, while ethanol F4 exhibited antibacterial activity against A. baumannii. The active fractions were processed for LC MS/MS analysis to identify bioactive compounds. Valoneic acid dilactone (aqueous F1 and F4), Hexoside (ethanol F4) and Coumaric acid (hexane F2) were identified as bioactive compounds in HPLC fractions. CONCLUSION: Puncia granatum peel extracts HPLC fractions exhibited potential inhibitory activity against MDR bacterial human pathogens. Several bioactive compounds were identified from the HPLC fractions. Further characterization of these compounds may be helpful to conclude it as therapeutic lead molecules against MDR pathogens.

Medicated jelly formulations are patient friendly dosage form for pediatric, geriatric and dysphagic patients. These formulations offer rapid dissolution and absorption of drugs through oral mucosa therefore show the early onset of action. The objective of the study was to develop and evaluate oral jelly formulations of vitamin C. Slurry method was adopted using glucose 103gm, sugar 67gm, gelatin 10gm and sorbitol 6.56gm. Preformulation studies were performed including the organoleptic profile, pH, and solubility of both drugs. The medicated jelly of Vitamin C was prepared and evaluated for physical characteristics, weight variation, syneresis, pH, taste and palatability, drug content, release rate characteristics and stability studies. All the jellies were found to have patient welcoming taste and were palatable. All formulations showed more than 50% drug release within 15 minutes, while 93% drug was released in 30 minutes. The results of release kinetics showed that the formulation followed the zero order release kinetics. Thus the drug was released at constant rate independent of the drug concentration involved in the process. All the medicated jellies were found to remain stable stored for 60 days at different temperatures. The present study revealed that medicated jellies of vitamin C could be employed orally in an effective form as an alternative solid oral dosage form for special population such as pediatrics, geriatrics and patients with dysphagia.

In the third millennium, developing countries will confront significant environmental problems such as ozone depletion, global warming, the shortage of fossil resources, and greenhouse gas emissions. This research looked at a multigenerational system that can generate clean hydrogen, fresh water, electricity, heat, and cooling. The system's components include Rankine and Brayton cycles, an Organic Rankine Cycle (ORC), flash desalination, an Alkaline electrolyzer, and a solar heliostat. The proposed process has been compared for two different start-up modes with a combustion chamber and solar heliostat to compare renewable and fossil fuel sources. This research evaluated various characteristics, including turbine pressure, system efficiency, solar radiation, and isentropic efficiency. The energy and exergy efficiency of the proposed system were obtained at around 78.93% and 47.56%, respectively. Exergy study revealed that heat exchangers and alkaline electrolyzers had the greatest exergy destruction rates, at 78.93% and 47.56%, respectively. The suggested system produces 0.04663 kg/s of hydrogen. Results indicate that at the best operational conditions, the exergetic efficiency, power, and hydrogen generation of 56%, 6000 kW, and 1.28 kg/s is reached, respectively. Also, With a 15% improvement in the Brayton cycle's isentropic efficacy, the quantity of hydrogen produced increases from 0.040 kg/s to 0.0520 kg/s.

Microbial fuel cells (MFCs) offer a dual solution of generating electrical energy from organic pollutants-laden wastewater while treating it. This study focuses on enhancing MFC performance through innovative electrode design. Three-dimensional (3D) anodes, created from corncobs and mango seeds via controlled graphitization, achieved remarkable power densities. The newly developed electrode configurations were evaluated within sewage wastewater-driven MFCs without the introduction of external microorganisms or prior treatment of the wastewater. At 1,000°C and 1,100°C graphitization temperatures, corncob and mango seed anodes produced 1,963 and 2,171 mW/m2, respectively, nearly 20 times higher than conventional carbon cloth and paper anodes. An advanced cathode composed of an activated carbon-carbon nanotube composite was introduced, rivaling expensive platinum-based cathodes. By optimizing the thermal treatment temperature and carbon nanotube content of the proposed cathode, comparable or superior performance to standard Pt/C commercial cathodes was achieved. Specifically, MFCs assembled with corncob anode with the proposed and standard Pt/C cathodes reached power densities of 1,963.1 and 2,178.6 mW/m2, respectively. Similarly, when utilizing graphitized mango seeds at 1,100°C, power densities of 2,171 and 2,151 mW/m2 were achieved for the new and standard cathodes, respectively. Furthermore, in continuous operation with a flow rate of 2 L/h, impressive chemical oxygen demand (COD) removal rates of 77% and 85% were achieved with corncob and mango seed anodes, respectively. This work highlights the significance of electrode design for enhancing MFC efficiency in electricity generation and wastewater treatment.

New bio-composite nanofibers composed of polyethylene oxide (PEO)/silk fibroin (SF)/barium titanate (BaTiO3) are introduced in this study. The SF solution was added to the PEO solution to form a PEO/SF blend with different weight percentages (5, 10, 15, 20 wt.%). The PEO/15 wt.% SF blend was selected to continue the experimental plan based on the optimum nanofiber morphology. Different wt.% of BaTiO3 particles (0.2, 0.4, 0.8, 1 wt.%) were added to the PEO/15 wt.% SF blend solution, and the suspensions obtained were introduced to an electrospinning device. The fabricated tissue was characterized by scanning electron microscope (SEM), X-ray diffraction (XRD) and Fourier-transform infrared (FTIR) spectroscopy. The zeta potential of the solution and the piezoelectric performance of the fabricated tissue were characterized. A newly designed pizoTester was used to investigate piezoelectric properties. The results showed that a well-organized, smooth PEO/15 wt.% SF/0.2 wt.% BaTiO3 nanofiber composite with low bead contents was obtained. Improved properties and electrical coupling were achieved in the newly introduced material. Electrospun PEO/15 wt.% SF/0.2 wt.% BaTiO3 mats increased the output voltage (1150 mV) compared to pristine PEO and PEO/SF composite fibers (410 and 290 mV, respectively) upon applying 20 N force at 5 Hz frequency. The observed enhancement in piezoelectric properties suggests that the prepared composite could be a promising material in cardiac tissue engineering (CTE).

RESUMO

Leishmania tropica was isolated from the clinical patients of cutaneous leishmaniasis in rural community of Kohat district in Khyber Pakhtunkhwa province and was identified through PCR, microscopy, and culture techniques. A total of 113 samples from the clinical patients were examined through PCR, microscopy, and culture which showed 87.61% (99/113), 53.98% (61/113), and 46.90% (53/113) prevalence. During the study, 186 bp Leishmania tropica was identified through PCR. Thus the sensitivity of PCR is very high as compared to the conventional techniques.
