RESUMO

COVID-19 has accounted for more than 6 million deaths worldwide. Bacillus Calmette-Guérin (BCG), the existing tuberculosis vaccine, is known to induce heterologous effects over other infections due to trained immunity and has been proposed to be a potential strategy against SARS-CoV-2 infection. In this report, we constructed a recombinant BCG (rBCG) expressing domains of the SARS-CoV-2 nucleocapsid and spike proteins (termed rBCG-ChD6), recognized as major candidates for vaccine development. We investigated whether rBCG-ChD6 immunization followed by a boost with the recombinant nucleocapsid and spike chimera (rChimera), together with alum, provided protection against SARS-CoV-2 infection in K18-hACE2 mice. A single dose of rBCG-ChD6 boosted with rChimera associated with alum elicited the highest anti-Chimera total IgG and IgG2c Ab titers with neutralizing activity against SARS-CoV-2 Wuhan strain when compared with control groups. Importantly, following SARS-CoV-2 challenge, this vaccination regimen induced IFN-γ and IL-6 production in spleen cells and reduced viral load in the lungs. In addition, no viable virus was detected in mice immunized with rBCG-ChD6 boosted with rChimera, which was associated with decreased lung pathology when compared with BCG WT-rChimera/alum or rChimera/alum control groups. Overall, our study demonstrates the potential of a prime-boost immunization system based on an rBCG expressing a chimeric protein derived from SARS-CoV-2 to protect mice against viral challenge.

Telomeres are the physical ends of eukaryotic linear chromosomes that play critical roles in cell division, chromosome maintenance, and genome stability. In many plants, telomeres are comprised of TTTAGGG tandem repeat that is widely found in plants. We refer to this repeat as canonical plant telomeric repeat (CPTR). Peanut (Arachis hypogaea L.) is a spontaneously formed allotetraploid and an important food and oil crop worldwide. In this study, we analyzed the peanut genome sequences and identified a new type of tandem repeat with 10-bp basic motif TTTT(C/T)TAGGG named TAndem Repeat (TAR) 30. TAR30 showed significant sequence identity to TTTAGGG repeat in 112 plant genomes suggesting that TAR30 is a homolog of CPTR. It also is nearly identical to the telomeric tandem repeat in Cestrum elegans. Fluorescence in situ hybridization (FISH) analysis revealed interstitial locations of TAR30 in peanut chromosomes but we did not detect visible signals in the terminal ends of chromosomes as expected for telomeric repeats. Interestingly, different TAR30 hybridization patterns were found between the newly induced allotetraploid ValSten and its diploid wild progenitors. The canonical telomeric repeat TTTAGGG is also present in the peanut genomes and some of these repeats are closely adjacent to TAR30 from both cultivated peanut and its wild relatives. Overall, our work identifies a new homolog of CPTR and reveals the unique distributions of TAR30 in cultivated peanuts and wild species. Our results provide new insights into the evolution of tandem repeats during peanut polyploidization and domestication.

Plant expansins are structural cell wall-loosening proteins implicated in several developmental processes and responses to environmental constraints and pathogen infection. To date, there is limited information about the biological function of expansins-like B (EXLBs), one of the smallest and less-studied subfamilies of plant expansins. In the present study, we conducted a functional analysis of the wild Arachis AdEXLB8 gene in transgenic tobacco (Nicotiana tabacum) plants to clarify its putative role in mediating defense responses to abiotic and biotic stresses. First, its cell wall localization was confirmed in plants expressing an AdEXLB8:eGFP fusion protein, while nanomechanical assays indicated cell wall reorganization and reassembly due to AdEXLB8 overexpression without compromising the phenotype. We further demonstrated that AdEXLB8 increased tolerance not only to isolated abiotic (drought) and biotic (Sclerotinia sclerotiorum and Meloidogyne incognita) stresses but also to their combination. The jasmonate and abscisic acid signaling pathways were clearly favored in transgenic plants, showing an activated antioxidative defense system. In addition to modifications in the biomechanical properties of the cell wall, we propose that AdEXLB8 overexpression interferes with phytohormone dynamics leading to a defense primed state, which culminates in plant defense responses against isolated and combined abiotic and biotic stresses.

The lectins are carbohydrate-binding proteins that are highly specific to sugar groups associated to other molecules. In addition to interacting with carbohydrates, a number of studies have reported the ability of these proteins to modulate the activity of several antibiotics against multidrug-resistant (MDR) strains. In this study, we report the enhanced antibacterial activity of the gentamicin against MDR strains when complexed with a lectin from Canavalia ensiformis seeds (ConA). Hemagglutination activity test and intrinsic fluorescence spectroscopy revealed that the gentamicin can interact with ConA most likely via the carbohydrate recognition domain (CRD) with binding constant (Kb) value estimated of (0.44 ± 0.04) x 104 M-1. Furthermore, the minimum inhibitory concentrations (MIC) obtained for ConA against all strains studied were not clinically relevant (MIC ≥ 1024 µg/mL). However, when ConA was combined with gentamicin, a significant increase in antibiotic activity was observed against Staphylococcus aureus and Escherichia coli. The present study showed that ConA has an affinity for gentamicin and modulates its activity against MDR strains. These results indicate that ConA improves gentamicin performance and is a promising candidate for structure/function analyses.

This study aimed to evaluate the intrinsic antibacterial activity and antibiotic-enhancing effect of an arylamino methylene derivative (MAD) in association with fluoroquinolones. The antibacterial activity against multiresistant Pseudomonas aeruginosa, Staphylococcus aureus and Escherichia coli was analyzed by determining the minimum inhibitory concentration (MIC) using the broth micro dilution method. A reduction in the MIC of the fluoroquinolones against strains treated simultaneously with the MAD was interpreted as an enhanced antibiotic activity. While the MAD exhibited no clinically effective action (MIC ≥ 1.024 µg/mL), it was found to significantly potentiate the activity of norfloxacin, ofloxacin and lomefloxacin against all the strains, which may be related to structural similarities between the MAD and quinolones. Our findings suggest that Meldrum's acid arylamino derivatives may represent promising molecules in the elaboration of new drugs to reverse resistance to fluoroquinolones.

The currently used hemostatic agents are highly effective in stopping hemorrhages but have a limited role in the modulation of the wound-healing environment. Herein, we propose an intrinsically bioactive hemostatic cryogel based on platelet lysate (PL) and aldehyde-functionalized cellulose nanocrystals (a-CNCs). PL has attracted great attention as an inexpensive milieu of therapeutically relevant proteins; however, its application as a hemostatic agent exhibits serious constraints (e.g., structural integrity and short shelf-life). The incorporation of a-CNCs reinforced the low-strength PL matrix by covalent cross-linking its amine groups that exhibit an elastic interconnected porous network after full cryogelation. Upon blood immersion, the PL-CNC cryogels absorbed higher volumes of blood at a faster rate than commercial hemostatic porcine gelatin sponges. Simultaneously, the cryogels released biomolecules that increased stem cell proliferation, metabolic activity, and migration as well as downregulated the expression of markers of the fibrinolytic process. In an in vivo liver defect model, PL-CNC cryogels showed similar hemostatic performance in comparison with gelatin sponges and normal material-induced tissue response upon subcutaneous implantation. Overall, owing to their structure and bioactive composition, the proposed PL-CNC cryogels provide an alternative off-the-shelf hemostatic and antibacterial biomaterial with the potential to deliver therapeutically relevant proteins in situ.

Thin-walled drip tapes with continuous labyrinth have been used for irrigation of vegetables and other short-cycle crops, especially due to their low cost. The continuous labyrinths welded into the pipe inner wall affect the head loss along such emitting pipes. In addition, the flow cross section of thin-walled pipes may change due to the effects of the operating pressure, which also has consequences for the head loss. The objective of this work was to investigate experimentally the friction factor and the head loss on thin-walled drip tapes with continuous labyrinths operated under various pressures. Two models of commercial thin-walled drip tapes with continuous labyrinths were evaluated. Nonperforated samples were used to determine the head-loss equations. The equations were adjusted as a function of flow rate and pressure head at the pipe inlet. Alternatively, the diameter in the Darcy-Weisbach equation was adjusted as a function of the pressure head by a power-law model. The possibility of using a mean diameter in the Darcy-Weisbach equation was also analyzed. Experimental investigation indicated that the friction factor in the Darcy-Weisbach equation can be accurately described using a power-law model, like the Blasius equation, but characterized by a coefficient a=0.3442 for the Turbo Tape and a=0.3225 for the Silver Tape. The obtained values of a are larger than those generally used and available in the literature. The influence of the operating pressure on the pipe diameter can be neglected for the purpose of calculating the head loss. The two approaches, considering the variation of the diameter with the pressure head and considering an optimum average diameter for the calculation of head loss by the Darcy-Weisbach equation, produce similar results, allowing accurate prediction of head loss. Evaluating the proposed mathematical models, 95% of predictions presented relative errors of head loss smaller than 5%. For the Turbo Tape, the optimum diameter for the purpose of calculating the head loss is 16.01 mm, which is very close to the value indicated by its manufacturer (15.9 mm). For the Silver Drip, the optimum diameter is 15.71 mm, while the manufacturer gives a value of 16.22 mm, which produces considerable error in the calculation of head loss.

Hunting is a problem to animal conservation in different parts of the world and it has caused the local extinction of several species. The aim of this study was to characterize the poaching activities in one of the main tabuleiro forest remnants of Brazil, the Linhares-Sooretama Block (LSB). Poaching records from 2010 to 2013 were gathered from the agencies responsible for monitoring and combating environmental crimes in the LSB. A total of 693 records (mean = 173 events/year) were collected involving direct (hunted animals, firearms, handmade firearms, traps, poachers, and various hunting supplies) and indirect (tree stands, baits, and poacher signs) evidences of poaching. No differences in the monthly cumulative number of records were found among years, but the distribution of records differed according to the type of evidence. A total of 40 animal seizure events were recorded involving a total of at least 15 taxa directly affected by poaching (reptiles = 2, birds = 6, mammals = 7) and 75 individuals seized (19 individuals/year). Five of the poached species are threatened. Lowland paca (Cuniculus paca) and armadillos were the most poached mammals in the region. Most of the poachers conduct such activities for fun (entertainment) and/or professionally (commercial hunting). The collected data show an approximately 32% increase in the number of poaching events in the region compared with the historical data available for LSB. It may have resulted from a gradual decrease in protection, both in terms of the number of agents deployed and the levels of effort of the teams, which began in 2009. The data demonstrate that poaching is a significant threat to the conservation of the LSB fauna, as it is in other Atlantic Forest remnants and in other regions of the world. Protection activities must be intensified to effectively combat the impacts of poaching in the LSB region, thereby contributing to the conservation of species in one of the few Atlantic Forest remnants whose original species composition still remains intact.

BACKGROUND AND AIMS: Arachis batizocoi is a wild relative of cultivated peanut (A. hypogaea), an allotetraploid with an AABB genome. Arachis batizocoi was once considered the ancestral donor of the peanut B genome, but cytogenetics and DNA phylogenies have indicated a new genome classification, 'K'. These observations seem inconsistent with genetic studies and breeding that have shown that A. batizocoi can behave as a B genome. METHODS: The genetic behaviour, genome composition and phylogenetic position of A. batizocoi were studied using controlled hybridizations, induced tetraploidy, whole-genome in situ fluorescent hybridization (GISH) and molecular phylogenetics. KEY RESULTS: Sterile diploid hybrids containing AK genomes were obtained using A. batizocoi and the A genome species A. duranensis, A. stenosperma, A. correntina or A. villosa. From these, three types of AAKK allotetraploids were obtained, each in multiple independent polyploidy events. Induced allotetraploids were vigorous and fertile, and were hybridized to A. hypogaea to produce F1 hybrids. Even with the same parental combination, fertility of these F1 hybrids varied greatly, suggesting the influence of stochastic genetic or epigenetic events. Interestingly, hybrids with A. hypogaea ssp. hypogaea were significantly more fertile than those with the subspecies fastigiata. GISH in cultivated × induced allotetraploids hybrids (harbouring AABK genomes) and a molecular phylogeny using 16 intron sequences showed that the K genome is distinct, but more closely related to the B than to the A genome. CONCLUSIONS: The K genome of A. batizocoi is more related to B than to the A genome, but is distinct. As such, when incorporated in an induced allotetraploid (AAKK) it can behave as a B genome in crosses with peanut. However, the fertility of hybrids and their progeny depends upon the compatibility of the A genome interactions. The genetic distinctness of A. batizocoi makes it an important source of allelic diversity in itself, especially in crosses involving A. hypogaea ssp. hypogaea.

Peanut (Arachis hypogaea L.) is an important legume cultivated mostly in drought-prone areas where its productivity can be limited by water scarcity. The development of more drought-tolerant varieties is, therefore, a priority for peanut breeding programs worldwide. In contrast to cultivated peanut, wild relatives have a broader genetic diversity and constitute a rich source of resistance/tolerance alleles to biotic and abiotic stresses. The present study takes advantage of this diversity to identify drought-responsive genes by analyzing the expression profile of two wild species, Arachis duranensis and Arachis magna (AA and BB genomes, respectively), in response to progressive water deficit in soil. Data analysis from leaves and roots of A. duranensis (454 sequencing) and A. magna (suppression subtractive hybridization (SSH)) stressed and control complementary DNA (cDNA) libraries revealed several differentially expressed genes in silico, and 44 of them were selected for further validation by quantitative RT-PCR (qRT-PCR). This allowed the identification of drought-responsive candidate genes, such as Expansin, Nitrilase, NAC, and bZIP transcription factors, displaying significant levels of differential expression during stress imposition in both species. This is the first report on identification of differentially expressed genes under drought stress and recovery in wild Arachis species. The generated transcriptome data, besides being a valuable resource for gene discovery, will allow the characterization of new alleles and development of molecular markers associated with drought responses in peanut. These together constitute important tools for the peanut breeding program and also contribute to a better comprehension of gene modulation in response to water deficit and rehydration.

INTRODUCTION: The purpose of this study was to measure cell density, average area, and morphology of corneal endothelium of chinchillas in different age groups. MATERIALS AND METHODS: The corneal endothelium was studied with a contact specular microscope. Thirty chinchillas (Chinchilla lanigera) were evaluated, and 30 male chinchillas were divided into three groups of 10 chinchillas each, by age: Group 1 (2-4-month-old), Group 2 (48-month-old), and Group 3 (10 years of age). Data are presented as endothelial cell density (ECD), average cell area, and pleomorphism. RESULTS: Endothelial cell density decreases and an increase in endothelial cell area and pleomorphism were observed with age, in the corneas of normal chinchillas. For Group 1 chinchillas, the mean cell density was 3.423 cells per mm(2) . The mean ECD for Group 2 chinchillas was 2.650 cells per mm(2) . The mean ECD for Group 3 chinchillas was 2.124 cells per mm(2) . The average area for Group 1 chinchillas was 350.5 µm(2) . The average area for Group 2 chinchillas was 442.15 µm(2) . The average area for Group 3 chinchillas was 583.64 µm(2) . The pleomorphism for Group 1 was 70.05%. The pleomorphism for Group 2 was 65.18%, and the pleomorphism for Group 3 was 62.28%. CONCLUSIONS: The results suggested that chinchilla corneal endothelium undergoes age-related changes. Moreover, with advancing age, the mean cell area increased and cell density decreased.

BACKGROUND AND AIMS: Peanut (Arachis hypogaea) is an allotetraploid (AABB-type genome) of recent origin, with a genome of about 2·8 Gb and a high repetitive content. This study reports an analysis of the repetitive component of the peanut A genome using bacterial artificial chromosome (BAC) clones from A. duranensis, the most probable A genome donor, and the probable consequences of the activity of these elements since the divergence of the peanut A and B genomes. METHODS: The repetitive content of the A genome was analysed by using A. duranensis BAC clones as probes for fluorescence in situ hybridization (BAC-FISH), and by sequencing and characterization of 12 genomic regions. For the analysis of the evolutionary dynamics, two A genome regions are compared with their B genome homeologues. KEY RESULTS: BAC-FISH using 27 A. duranensis BAC clones as probes gave dispersed and repetitive DNA characteristic signals, predominantly in interstitial regions of the peanut A chromosomes. The sequences of 14 BAC clones showed complete and truncated copies of ten abundant long terminal repeat (LTR) retrotransposons, characterized here. Almost all dateable transposition events occurred <3·5 million years ago, the estimated date of the divergence of A and B genomes. The most abundant retrotransposon is Feral, apparently parasitic on the retrotransposon FIDEL, followed by Pipa, also non-autonomous and probably parasitic on a retrotransposon we named Pipoka. The comparison of the A and B genome homeologous regions showed conserved segments of high sequence identity, punctuated by predominantly indel regions without significant similarity. CONCLUSIONS: A substantial proportion of the highly repetitive component of the peanut A genome appears to be accounted for by relatively few LTR retrotransposons and their truncated copies or solo LTRs. The most abundant of the retrotransposons are non-autonomous. The activity of these retrotransposons has been a very significant driver of genome evolution since the evolutionary divergence of the A and B genomes.

Although the boll weevil (BW), Anthonomus grandis grandis (Coleoptera: Curculionidae) has been attributed to the significant losses caused to cotton yield in the Americas, the categorization as a quarentenary pest in places where it is still not occurring has increased its relevance worldwide. In areas where it is widespread, pest suppression relies on many broad-spectrum insecticide applications. However, other control tactics are sought. Considering that early-flowering cultivars escape from boll weevil infestation, we investigated if three different planting dates (November, December, and January) could alter the plant life cycle, allowing the plants to escape from boll weevil infestation. Field trials were run in two seasons (2014/2015 and 2017/2018), and variables (days required to reach each flowering stage, fruiting plant structures-undamaged and damaged by the BW, and totals-number of boll weevils on plants and that had emerged from fallen structures on the ground) were assessed over 29 and 33 weeks, respectively. Based on the number of days required to initiate and terminate the flowering stage, the time to reach the economic threshold (ET), the number of undamaged, damaged, and the total reproductive structures, we concluded that planting dates in December for the Central Cerrado of Brazil should be preferred over the other two tested dates. Cultivations run at this planting date, anticipating the flowering period initiation and termination, reduced infested flowering structures, and delayed the decision making to control the pest, when compared to the other two planting dates.

Cholesterol plays a crucial role in biomembranes by regulating various properties, such as fluidity, rigidity, permeability, and organization of lipid bilayers. The latest version of the Martini model, Martini 3, offers significant improvements in interaction balance, molecular packing, and inclusion of new bead types and sizes. However, the release of the new model resulted in the need to reparameterize many core molecules, including cholesterol. Here, we describe the development and validation of a Martini 3 cholesterol model, addressing issues related to its bonded setup, shape, volume, and hydrophobicity. The proposed model mitigates some limitations of its Martini 2 predecessor while maintaining or improving the overall behavior.

BACKGROUND: Cultivated peanut (Arachis hypogaea) is one of the most widely grown grain legumes in the world, being valued for its high protein and unsaturated oil contents. Worldwide, the major constraints to peanut production are drought and fungal diseases. Wild Arachis species, which are exclusively South American in origin, have high genetic diversity and have been selected during evolution in a range of environments and biotic stresses, constituting a rich source of allele diversity. Arachis stenosperma harbors resistances to a number of pests, including fungal diseases, whilst A. duranensis has shown improved tolerance to water limited stress. In this study, these species were used for the creation of an extensive databank of wild Arachis transcripts under stress which will constitute a rich source for gene discovery and molecular markers development. RESULTS: Transcriptome analysis of cDNA collections from A. stenosperma challenged with Cercosporidium personatum (Berk. and M.A. Curtis) Deighton, and A. duranensis submitted to gradual water limited stress was conducted using 454 GS FLX Titanium generating a total of 7.4 x 10(5) raw sequence reads covering 211 Mbp of both genomes. High quality reads were assembled to 7,723 contigs for A. stenosperma and 12,792 for A. duranensis and functional annotation indicated that 95% of the contigs in both species could be appointed to GO annotation categories. A number of transcription factors families and defense related genes were identified in both species. Additionally, the expression of five A. stenosperma Resistance Gene Analogs (RGAs) and four retrotransposon (FIDEL-related) sequences were analyzed by qRT-PCR. This data set was used to design a total of 2,325 EST-SSRs, of which a subset of 584 amplified in both species and 214 were shown to be polymorphic using ePCR. CONCLUSIONS: This study comprises one of the largest unigene dataset for wild Arachis species and will help to elucidate genes involved in responses to biological processes such as fungal diseases and water limited stress. Moreover, it will also facilitate basic and applied research on the genetics of peanut through the development of new molecular markers and the study of adaptive variation across the genus.

The environmental quality of the Jacuípe River's estuary (very important in northeastern Brazil) was assessed during 2007 and 2008. In water, concentrations (mg L⁻¹) of NO2⁻ (<0.004 to 0.016), NO3⁻ (0.01 to 0.33), soluble PO4³â» (<0.02 to 0.22), dissolved oxygen (3.9 to 9.6), total contents (mg L⁻¹) of Cd (<0.001), Cu (<0.01), Pb (<0.01), and Zn (<0.1), pH (5.60 to 8.00), and electrical conductivity (0.12 to 48.60 mS cm⁻¹) agreed with environmental standards. In sediments, clay and total organic matter (%, m/m) varied, respectively, from 8.8 to 12.0 and from 1.1 to 8.8, while infrared, thermogravimetric profile, electronic micrograph, as well as X-Ray analyses showed desirable adsorptive characteristics. However, maximum exchangeable levels (mg kg⁻¹) of Cd (1.3), Cu (44.6), Pb (35.7), and Zn (43.7) and their respective maximum pseudototal concentrations (mg kg⁻¹): 19.4, 95.1, 68.2, and 30.3 were below the recommended limits. In this sense, it was possible to demonstrate good environmental preservation even with the growing number of industries and touristic activities in the evaluated estuarine area.

BACKGROUND: Knowledge of the spatio-temporal distribution of pests is important for the development of accurate management approaches. The boll weevil, Anthonomus grandis grandis Boh., is a deleterious cotton, Gossypium hirsutum L., pest in the western hemisphere. The spread of boll weevils across cotton fields remains poorly understood. We assessed the dispersal pattern of adult weevils through cotton fields cultivated in a tropical area during dry and wet seasons using geostatistics for the number of adults and infested reproductive structures (buds, bolls and total). RESULTS: Adult weevils and infested reproductive structures increased across both seasons despite the prevailing climatic variables. In both seasons, boll weevil adults and infested reproductive structures followed an aggregated distribution. The distances over which samples maintained spatial dependence varied from 0.7 to 43.4 m in the dry season and from 6.0 to 614.4 m in the wet season. Boll weevil infestations started at field borders and the infested reproductive structures (oviposition and/or feeding punctured) were greater than the adults regardless of cotton growth stage. CONCLUSION: Sampling for boll weevils in cotton fields should start at the field borders and focus on total infested reproductive structures (buds + bolls) and as cotton plants develop, sampling should focus on the field as a whole. Distances among samples will vary from 6 to 470 m. Thus, despite the cotton phenological stage or growing season, monitoring of boll weevil should be done by sampling total infested reproductive structures with a minimum distance of 6 m among samples. © 2022 Society of Chemical Industry.

Polyphenols have been extensively exploited in the biomedical field because of their wide range of bioactive properties and historical use as traditional medicines. They typically present antioxidant, antimicrobial, antiamyloidogenic, and/or antitumor activities. In particular, cork water extracts and their components, have been previously reported to present antioxidant and antiamyloidogenic properties. On the basis of this knowledge, we tested cork water extract (CWE), cork water enriched extract (CWE-E), vescalagin/castalagin (two of the main polyphenols present in CWE and CWE-E) for their antibacterial activity against four bacterial strains, namely, methicillin-resistant Staphylococcus epidermidis (MRSE), Staphylococcus aureus (SA), methicillin-resistant Staphylococcus aureus (MRSA), and Pseudomonas aeruginosa (PA). Vescalagin and castalagin presented bactericidal activity against all the tested bacterial strains, in particular toward the methicillin-resistant ones, i.e., MRSA and MRSE, as well as the ability to inhibit the formation of biofilms and to disrupt preformed ones. Moreover, vescalagin/castalagin seem to modulate the normal assembly of the peptidoglycans at the bacteria surface, promoting the disruption of their cell wall, leading to bacterial cell death. We also demonstrate that vescalagin/castalagin can be loaded into alginate hydrogels to generate antibacterial biomaterials that are not toxic to eukaryotic cells.

AIMS: This study aimed to investigate the potential of limonene as an efflux pump (EP) inhibitor in Staphylococcus aureus strains, RN-4220 and IS-58, which carry EPs for erythromycin (MrsA) and tetracycline (TetK), respectively. BACKGROUND: The evolution of bacterial resistance mechanisms over time has impaired the action of most classes of antibiotics. Staphylococcus aureus is a notable bacterium, with high pathogenic potential and demonstrated resistance to conventional antibiotics. Considering the importance of discovering novel compounds to combat antibiotic resistance, our group previously demonstrated the antibacterial properties of limonene, a compound present in the essential oils of several plant species. OBJECTIVE: This study aimed to investigate the potential of limonene as an efflux pump (EP) inhibitor in Staphylococcus aureus strains RN-4220 and IS-58, which carry EPs for erythromycin (MrsA) and tetracycline (TetK), respectively. METHODS: The minimum inhibitory concentrations (MIC) of limonene and other efflux pump inhibitors were determined through the broth microdilution method. A reduction in the MIC of ethidium bromide was used as a parameter of EP inhibition. RESULT: While limonene was not shown to exhibit direct antibacterial effects against EP-carrying strains, in association with ethidium bromide and antibiotics, this compound demonstrated enhanced antibacterial activity, indicating the inhibition of the MrsA and TetK pumps. CONCLUSION: In conclusion, this pioneering study demonstrated the effectiveness of limonene as an EP inhibitor in S. aureus strains, RN-4220 and IS-58. Nevertheless, further studies are required to characterize the molecular mechanisms associated with limonene-mediated EP inhibition.

Arachis stenosperma is a wild peanut relative exclusive to South America that harbors high levels of resistance against several pathogens, including the peanut root-knot nematode (RKN) Meloidogyne arenaria. In this study, a proteomic survey of A. stenosperma-M. arenaria interaction using 2-DE and LC-MS/MS identified approximately 1400 proteins, out of which 222 were differentially abundant (DAPs) when RKN inoculated root samples were compared to the control. Most of these DAPs were assigned to functional categories related to plant responses to pathogens including stress, glycolysis, redox and tricarboxylic acid cycle. The comparison between the transcriptome (RNA-Seq) and proteome expression changes, showed that almost 55% of these DAPs encode genes with a similar expression trend to their protein counterparts. Most of these genes were induced during RKN infection and some were related to plant defense, such as MLP-like protein 34 (MLP34), cinnamoyl-CoA reductase 1 (CCR1), enolase (ENO), alcohol dehydrogenase (ADH) and eukaryotic translation initiation factor 5A (eIF5A). The overexpression of AsMLP34 in Agrobacterium rhizogenes transgenic roots in a susceptible peanut cultivar showed a reduction in the number of M. arenaria galls and egg masses, indicating that AsMLP34 is a promising candidate gene to be exploited in breeding programs for RKN control in peanut. SIGNIFICANCE: The use of an integrated approach to compare plant-nematode transcriptional and translational data enabled the identification of a new gene, AsMLP34, for Meloidogyne resistance.
