BACKGROUND: Environmental cadmium (Cd) exposure is linked to pulmonary function injury in the general population. But, the association between blood Cd concentration and pulmonary function has not been investigated thoroughly in chronic obstructive pulmonary disease (COPD) patients, and the potential mechanisms are unclear. METHODS: All eligible 789 COPD patients were enrolled from Anhui COPD cohort. Blood specimens and clinical information were collected. Pulmonary function test was conducted. The subunit of telomerase, telomerase reverse transcriptase (TERT), was determined through enzyme linked immunosorbent assay (ELISA). Blood Cd was measured via inductively coupled-mass spectrometer (ICP-MS). RESULTS: Blood Cd was negatively and dose-dependently associated with pulmonary function. Each 1-unit increase of blood Cd was associated with 0.861 L decline in FVC, 0.648 L decline in FEV1, 5.938 % decline in FEV1/FVC %, and 22.098 % decline in FEV1 % among COPD patients, respectively. Age, current-smoking, self-cooking and higher smoking amount aggravated Cd-evoked pulmonary function decrease. Additionally, there was an inversely dose-response association between Cd concentration and TERT in COPD patients. Elevated TERT obviously mediated 29.53 %, 37.50 % and 19.48 % of Cd-evoked FVC, FEV1, and FEV1 % declines in COPD patients, respectively. CONCLUSION: Blood Cd concentration is strongly associated with the decline of pulmonary function and telomerase activity among COPD patients. Telomere attrition partially mediates Cd-induced pulmonary function decline, suggesting an underlying mechanistic role of telomere attrition in pulmonary function decline from Cd exposure in COPD patients.

BACKGROUND: Oxidative stress is involved in the pathogenesis of chronic obstructive pulmonary disease (COPD). 8-Hydroxy-2'-deoxyguanosine (8-OHdG) is recognized as a biomarker of oxidative stress and is implicated in several pulmonary diseases. Nonetheless, the role of 8-OHdG remains unclear in COPD patients. This research aimed to evaluate the correlations between serum 8-OHdG on admission and the severity and prognosis of hospitalized COPD patients with acute exacerbation. METHODS: A total of 150 COPD hospitalized patients and 150 healthy individuals were recruited. Serum 8-OHdG was measured by ELISA and the length of hospital stay was calculated. The number of acute exacerbations of COPD was tracked within 1 year after this hospitalization. RESULTS: The levels of serum 8-OHdG were elevated in COPD patients compared with the control group. Serum 8-OHdG was gradually elevated with decreased pulmonary function in COPD patients. Furthermore, Pearson linear association found that the levels of serum 8-OHdG were inversely correlated with pulmonary function and positively correlated with inflammatory cytokines in COPD patients. In addition, logistic regression analysis revealed that serum 8-OHdG elevation was a risk factor for pulmonary function decline in COPD patients. The length of hospital stay was tracked at this time. Higher serum 8-OHdG on admission increased the length of hospital stay among COPD patients. CONCLUSION: Serum 8-OHdG on admission is positively correlated with the severity and adverse prognosis among COPD patients, suggesting that 8-OHdG may be involved in the pathogenesis of COPD. Serum 8-OHdG may be a biomarker to predict the progression of COPD.

The goal of this study was to analyze whether mitochondria-associated endoplasmic reticulum membrane (MAMs) dysfunction mediated arsenic (As)-evoked pulmonary ferroptosis and acute lung injury (ALI). As exposure led to alveolar structure damage, inflammatory cell infiltration and pulmonary function decline in mice. Ferritin, the marker of iron overload, was increased, GPX4, the index of lipid peroxidation, was decreased in As-exposed lungs and pulmonary epithelial cells (MLE-12). Pretreatment with ferrostatin-1 (Fer-1), the inhibitor of ferroptosis, alleviated As-evoked ALI. In addition, As-induced non-heme iron deposition was inhibited in Fer-1 pretreated-mice. Moreover, As-triggered mitochondria damage and ferroptosis were mitigated in Fer-1 pretreated-MLE-12 cells. Mechanistically, PERK phosphorylation and mitofusin-2 (Mfn-2) reduction was observed in As-exposed MLE-12 cells and mice lungs. Additionally, the interaction between PERK and Mfn-2 was downregulated and MAMs dysfunction was observed in As-exposed MLE-12 cells. Intriguingly, PERK inhibitor and Mfn-2-overexpression all mitigated As-induced ferroptosis in MLE-12 cells. Additionally, CLPP and mtHSP70, the markers of mitochondrial stress, were upregulated, mitochondrial ROS (mtROS) was elevated, mitochondrial membrane potential (MMP) and ATP were decreased in As-exposed MLE-12 cells. Mitoquinone mesylate (MitoQ), a novel mitochondrial-targeted antioxidant, alleviated As-induced excess mtROS, mitochondrial stress, MAMs dysfunction in pulmonary epithelial cells. Similarly, in vivo experiments indicated that MitoQ pretreatment countered As-induced pulmonary ferroptosis and ALI. These data indicated that mtROS-initiated MAMs dysfunction is, at least partially, implicated in As-evoked ferroptosis and ALI.

Polymerization at the liquid-liquid interface has attracted much attention for synthesizing ultrathin polymer films for molecular sieving. However, it remains a major challenge to conduct this process outside the alkane-water interface since it not only suffers water-caused side reactions but also is limited to water-soluble monomers. Here, we report the interfacial polymerization at the alkane/ionic liquid interface (IP@AILI) where the ionic liquid acts as the universal solvent for diversified amines to synthesize task-specific polyamide nanofilms. We propose that IP@AILI occurs when acyl chloride diffuses from the alkane into the ionic liquid instead of being triggered by the diffusion of amines as in the conventional alkane-water system, which is demonstrated by thermodynamic partitioning and kinetic monitoring. The prepared polyamide nanofilms with precisely adjustable pore sizes display unprecedented permeability and selectivity in various separation processes.

Manganese (Mn) is an essential micronutrient; however, few genes required for growth under low-Mn conditions have been identified. In this study, we isolated Arabidopsis thaliana mutants sensitive to low-Mn conditions from ethyl methanesulfonate-mutagenized seeds. Among them, we identified the causal genes of two mutants. One mutant (35-34) exhibited a short root phenotype and low Mn concentration in the shoots. The other mutant (30-11) exhibited a small shoot phenotype with Mn concentrations similar to the control. Genetic mapping, allelism tests, and gene complementation tests identified the causal genes as At1g80830 (NRAMP1) for 35-34 and At5g18480 (PGSIP6) for 30-11. NRAMP1 was previously reported to be essential for Mn uptake under low-Mn conditions, thus validating our screening method. PGSIP6 encodes inositol phosphorylceramide glucuronosyltransferase, which is involved in glycosyl inositol phosphorylceramide sphingolipid glycosylation. PGSIP6-green fluorescent protein was localized to the Golgi apparatus, which is consistent with its function in the glycosylation of sphingolipids. Our screening identified a novel gene required for low-Mn tolerance, and we also provide new insights towards understanding the physiological function of PGSIP6.

Greenhouse experiment was conducted to evaluate the potential effectiveness of a legume (Sesbania cannabina), arbuscular mycorrhizal fungi (AMF) (Glomus mosseae), and rhizobia (Ensifer sp.) symbiosis for remediation of Polycyclic aromatic hydrocarbons (PAHs) in spiked soil. AMF and rhizobia had a beneficial impact on each other in the triple symbiosis. AMF and/or rhizobia significantly increased plant biomass and PAHs accumulation in plants. The highest PAHs dissipation was observed in plant + AMF + rhizobia treated soil, in which >97 and 85-87% of phenanthrene and pyrene, respectively, had been degraded, whereas 81-85 and 72-75% had been degraded in plant-treated soil. During the experiment, a relatively large amount of water-soluble phenolic compounds was detected in soils of AMF and/or rhizobia treatment. It matches well with the high microbial activity and soil enzymes activity. These results suggest that the mutual interactions in the triple symbiosis enhanced PAHs degradation via stimulating both microbial development and soil enzyme activity. The mutual interactions between rhizobia and AMF help to improve phytoremediation efficiency of PAHs by S. cannabina.

Benzo[a]pyrene (BaP) and its metabolic end product benzo(a)pyren-7,8-dihydrodiol-9,10-epoxide (BPDE), are known toxic environmental pollutants. This study aimed to analyze whether sub-chronic BPDE exposure initiated pulmonary fibrosis and the potential mechanisms. In this work, male C57BL6/J mice were exposed to BPDE by dynamic inhalation exposure for 8 weeks. Our results indicated that sub-chronic BPDE exposure evoked pulmonary fibrosis and epithelial-mesenchymal transition (EMT) in mice. Both in vivo and in vitro, BPDE exposure promoted nuclear translocation of Snail. Further experiments indicated that nuclear factor erythroid 2-related factor 2 (Nrf2) and p62 were upregulated in BPDE-exposed alveolar epithelial cells. Moreover, Nrf2 siRNA transfection evidently attenuated BPDE-induced p62 upregulation. Besides, p62 shRNA inhibited BPDE-incurred Snail nuclear translocation and EMT. Mechanically, BPDE facilitated physical interaction between p62 and Snail in the nucleus, then repressed Snail protein degradation by p62-dependent autophagy-lysosome pathway, and finally upregulated transcriptional activity of Snail. Additionally, aryl hydrocarbon receptor (AhR) was activated in BPDE-treated alveolar epithelial cells. Dual-luciferase assay indicated activating AhR could bind to Nrf2 gene promoter. Moreover, pretreatment with CH223191 or α-naphthoflavone (α-NF), AhR antagonists, inhibited BPDE-activated Nrf2-p62 signaling, and alleviated BPDE-induced EMT and pulmonary fibrosis in mice. Taken together, AhR-mediated Nrf2-p62 signaling contributes to BaP-induced EMT and pulmonary fibrosis.

Porous substrates act as open "interfacial reactors" during the synthesis of polyamide composite membranes via interfacial polymerization. However, achieving a thin and dense polyamide nanofilm with high permeance and selectivity is challenging when using a conventional substrate with uniform wettability. To overcome this limitation, we propose the use of Janus porous substrates as confined interfacial reactors to decouple the local monomer concentration from the total monomer amount during interfacial polymerization. By manipulating the location of the hydrophilic/hydrophobic interface in a Janus porous substrate, we can precisely control the monomer solution confined within the hydrophilic layer without compromising its concentration. The hydrophilic surface ensures the uniform distribution of monomers, preventing the formation of defects. By employing Janus substrates fabricated through single-sided deposition of polydopamine/polyethyleneimine, we significantly reduce the thickness of the polyamide nanofilms from 88.4 to 3.8 nm by decreasing the thickness of the hydrophilic layer. This reduction leads to a remarkable enhancement in water permeance from 7.2 to 52.0 l/m2·h·bar while still maintaining ~96% Na2SO4 rejection. The overall performance of this membrane surpasses that of most reported membranes, including state-of-the-art commercial products. The presented strategy is both simple and effective, bringing ultrapermeable polyamide nanofilms one step closer to practical separation applications.

Fine design of surface charge properties of polyamide membranes is crucial for selective ionic and molecular sieving. Traditional membranes face limitations due to their inherent negative charge and limited charge modification range. Herein, we report a facile ionic liquid-decoupled bulk/interfacial diffusion strategy to elaborate the double charge flips of polyamide membranes, enabling on-demand transformation from inherently negative to highly positive and near-neutral charges. The key to these flips lies in the meticulous utilization of ionic liquid that decouples intertwined bulk/interfacial diffusion, enhancing interfacial while inhibiting bulk diffusion. These charge-tunable polyamide membranes can be customized for impressive separation performance, for example, profound Cl-/SO42- selectivity above 470 in sulfate recovery, ultrahigh Li+/Mg2+ selectivity up to 68 in lithium extraction, and effective divalent ion removal in pharmaceutical purification, surpassing many reported polyamide nanofiltration membranes. This advancement adds a new dimension to in the design of advanced polymer membranes via interfacial polymerization.

VeloPro integrates Ribo-seq data and AlphaFold2-predicted 3D protein structure information for characterization of the association patterns between translation velocity and many protein structure features in prokaryotic and eukaryotic organisms across different taxonomical clades such as bacteria, fungi, protozoa, nematode, plants, insect, and mammals. We illustrated that association patterns between translation velocity and protein structure features differ across organisms, partially reflecting their taxonomical relationship.

Interfacial polymerization is a well-known process to synthesize separation layers for thin film composite membranes at an immiscible organic liquid-aqueous liquid interface. The organic-aqueous interface determines the diffusion dynamics of monomers and the chemical environment for polymerization, exerting a critical influence on the formation of polymer thin films. This review summarizes recent advances in tailoring interfacial polymerization using interfaces beyond the conventional alkane-water interface to achieve high-performance separation films with designed structures. Diverse liquid-liquid interfaces are introduced for synthesizing separation films by adding co-solvents into the organic phase and/or the aqueous phase, respectively, or by replacing one of the liquid phases with other solvents. Innovative liquid-gel and liquid-gas interfaces are then summarized for the synthesis of polymer thin films for separation. Novel strategies to form reaction interfaces, such as spray-coating, are also presented and discussed. In addition, we discuss the details of how a physically or chemically patterned substrate affects interfacial polymerization. Finally, the potential of unconventional interfaces in interfacial polymerization is forecast with both challenges and opportunities.

It is difficult for beginners to learn and use amplicon analysis software because there are so many software tools to choose from, and all of them need multiple steps of operation. Herein, we provide a cross-platform, open-source, and community-supported analysis pipeline EasyAmplicon. EasyAmplicon has most of the modules needed for an amplicon analysis, including data quality control, merging of paired-end reads, dereplication, clustering or denoising, chimera detection, generation of feature tables, taxonomic diversity analysis, compositional analysis, biomarker discovery, and publication-quality visualization. EasyAmplicon includes more than 30 cross-platform modules and R packages commonly used in the field. All steps of the pipeline are integrated into RStudio, which reduces learning costs, keeps the flexibility of the analysis process, and facilitates personalized analysis. The pipeline is maintained and updated by the authors and editors of WeChat official account "Meta-genome." Our team will regularly release the latest tutorials both in Chinese and English, read the feedback from users, and provide help to them in the WeChat account and GitHub. The pipeline can be deployed on various platforms, and the installation time is less than half an hour. On an ordinary laptop, the whole analysis process for dozens of samples can be completed within 3 h. Benzo[a]pyrene (BaP), a representative polycyclic aromatic hydrocarbon, exists widely in automobile emissions and polluted atmosphere. The current study aimed to describe pulmonary inflammation during BaP-induced acute lung injury (ALI). All mice except controls were intratracheally instilled with a single dose of BaP (90 µg per mouse). The alveolar structure was damaged, accompanied by numerous inflammatory cell infiltration around pulmonary interstitium and small airway. Airway wall area and mean linear intercept were reduced in BaP-exposed mouse lungs. By contrast, airway wall thickness and destructive index were elevated in BaP-exposed mouse lungs. Several inflammatory genes, such as Tnf-α, Il-1ß, Il-6, Mip-2, Kc, and Mcp-1, were upregulated in mouse lungs. Phosphorylated IκBα was elevated in BaP-exposed mouse lungs. Nuclear translocation of NF-κB p65 and p50 was accordingly observed in BaP-exposed mouse lungs. Several molecules of the MAPK pathway, including JNK, ERK1/2, and p38, were activated in mouse lungs. Of interest, pretreatment with N-acetylcysteine (NAC), an antioxidant, alleviated BaP-induced ALI. Moreover, NAC attenuated BaP-induced inflammatory cell infiltration in mouse lungs and inflammatory gene upregulation in A549 cells. In addition, NAC attenuated BaP-induced NF-κB activation in A549 cells and mouse lungs. These results suggest that NAC alleviates pulmonary inflammatory response during BaP-evoked ALI.

Wild grapevines can show strong resistance to the downy mildew pathogen P. viticola, but the associated mechanisms are poorly described, especially at early stages of infection. Here, we performed comparative proteomic analyses of grapevine leaves from the resistant genotype V. davidii "LiuBa-8" (LB) and susceptible V. vinifera "Pinot Noir" (PN) 12 h after inoculation with P. viticola. By employing the iTRAQ technique, a total of 444 and 349 differentially expressed proteins (DEPs) were identified in LB and PN, respectively. The majority of these DEPs were related to photosynthesis, respiration, cell wall modification, protein metabolism, stress, and redox homeostasis. Compared with PN, LB showed fewer downregulated proteins associated with photosynthesis and more upregulated proteins associated with metabolism. At least a subset of PR proteins (PR10.2 and PR10.3) was upregulated upon inoculation in both genotypes, whereas HSP (HSP70.2 and HSP90.6) and cell wall-related XTH and BXL1 proteins were specifically upregulated in LB and PN, respectively. In the incompatible interaction, ROS signaling was evident by the accumulation of H2O2, and multiple APX and GST proteins were upregulated. These DEPs may play crucial roles in the grapevine response to downy mildew. Our results provide new insights into molecular events associated with downy mildew resistance in grapevine, which may be exploited to develop novel protection strategies against this disease.

Downy mildew of grapevine (Vitis vinifera L.), caused by the oomycete pathogen Plasmopara viticola, is one of the most serious concerns for grape production worldwide. It has been widely reported that the pathogenesis-related 4 (PR4) protein plays important roles in plant resistance to diseases. However, little is known about the role of PR4 in the defense of grapevine against P. viticola. In this study, we engineered loss-of-function mutations in the VvPR4b gene from the cultivar "Thompson Seedless" using the CRISPR/Cas9 system and evaluated the consequences for downy mildew resistance. Sequencing results showed that deletions were the main type of mutation introduced and that no off-target events occurred. Infection assays using leaf discs showed that, compared to wild-type plants, the VvPR4b knockout lines had increased susceptibility to P. viticola. This was accompanied by reduced accumulation of reactive oxygen species around stomata. Measurement of the relative genomic abundance of P. viticola in VvPR4b knockout lines also demonstrated that the mutants had increased susceptibility to the pathogen. Our results confirm that VvPR4b plays an active role in the defense of grapevine against downy mildew.

Internal browning (or brown heart) in radish is a physiological disorder, manifested as a reddish pigmentation in the central part of the tuberous root. Boron deficiency has been known to induce brown heart, but the relationship between B tissue concentration and the development of brown heart has not been tested. Here, we examined the relationship between these variables. Dissected root tissues of two inbred lines (i.e., cultivars) of East Asian big long radish exhibiting different severity of brown heart were submitted to inductively coupled plasma mass spectrometry (ICP-MS) analysis to reveal the spatial distribution of 19 chemical elements. Statistical analysis revealed that only B correlated negatively with the severity of brown heart. There was no significant difference in the average B concentration between the two cultivars, suggesting that differences in the efficient use of local B may be responsible for the variation in brown heart resistance between the two cultivars.

The Shiyang River Basin is an important ecological area of the Eastern Hexi Corridor, and is one of the most prominent areas of water conflict and ecological environment problems. An assessment of ecosystem quality in the Shiyang River Basin can provide a reference for ecological protection in arid inland basin. Based on the concept of ecosystem quality and the statistical yearbook, remotely sensed and land cover data, an evaluation index was established with consideration of three aspects of ecosystem (i.e., productivity, stability and bearing capacity). Kruskal-Wallis (Φ2) test and entropy method were applied to determine the weights of evaluation index. With the assistance of RS, GIS and SPSS software, a comprehensive evaluation and change analysis of ecosystem quality and corresponding index were conducted for various ecosystem types in the Shiyang River Basin in 2000, 2005, 2010 and 2015. Results showed that the average ecosystem quality of the Shiyang River Basin was 57.76, and presented an obvious decrease with a magnitude of 0.72 per year du-ring 2000-2015. The spatial pattern of ecosystem quality was that the upstream was better than the midstream, and the midstream was superior to the downstream. The mean values of production capacity, stability and carrying capacity of ecosystem were 67.52, 45.37, and 58.53, respectively. Production capacity and stability had increased slightly, while carrying capacity gradually decreased. Considering various ecosystem types, the highest quality was detected for forest ecosystem with average annual value of 78.12, and this ecosystem presented the lowest decreasing magnitude of 0.28 per year; for grassland, farmland and urban ecosystems, the average annual value was 62.45, 58.76 and 50.29, respectively; the quality of wetland ecosystem was the lowest, and suffered the largest decline with an average rate of 0.98 per year.

A thermocline characteristics contour on a spatial overlay map was plotted using data collected on a monthly basis from Argo buoys and data of monthly CPUE (catch per unit effort) bigeye tuna (Thunnus obesus) long-lines fishery from the Western and Central Pacific Fisheries Commission (WCPFC) to evaluate the relationship between fishing grounds temporal-spatial distribution of bigeye tuna and thermocline characteristics in the Western and Central Pacific Ocean (WCPO). In addition, Numerical methods were used to calculate the optimum ranges of thermocline characteristics of the central fishing grounds. The results showed that the central fishing grounds were mainly distributed between 10° N and 10° S. Seasonal fishing grounds in the south of equator were related to the seasonal variations in the upper boundary temperature, depth and thickness of thermocline. The fishing grounds were observed in areas where the upper boundary depth of thermocline was deep (70-100 m) and the thermocline thickness was more than 60 m. The CPUE tended to be low in area where the thermocline thickness was less than 40 m. The optimum upper boundary temperature range for distribution was 26-29 ℃, and the CPUE was mostly lower than the threshold value (Q3) of central fishing grounds when the temperature was higher than 29 ℃ or lower than 26 ℃. The temporal and spatial distribution of the fishing grounds was influenced by the seasonal variations in upper boundary depth and thermocline thickness. The central fishing grounds in the south of equator disappeared when the upper boundary depth of thermocline decreased and thermocline thickness became thinner. The lower boundary temperature and depth of thermocline and thermocline strength has little variation, but were strongly linked to the location of fishing grounds. The fishing grounds were mainly located between the two high-value zones of the lower boundary depth of thermocline, where the temperature was lower than 13 ℃ and the strength was high. When the depth was more than 300 m or less than 150 m, the lower boundary temperature was more than 17 ℃, or the strength was low, the CPUE tended to be low. The optimum range of thermocline characteristics was calculated using frequency analysis and empirical cumulative distribution function. The results showed that the optimum ranges for upper boundary thermocline temperature and depth were 26-29 ℃ and 70-110 m, the optimum lower boundary thermocline temperature and depth ranges were 11-13 ℃ and 200-280 m, the optimum ranges for thermocline thickness and thermocline strength were 50-90 m and 0.1-0.16 ℃·m-1, respectively. The paper documented the distribution interval of thermocline characteristics for central fishing ground of the bigeye tuna in WCPO. The results provided a reference for improving the efficiency of pelagic bigeye tuna fishing operation and tuna resource management in WCPO.
