An increase in growth rates of children suffering from growth hormone deficiency (GHD) subjected to recombinant growth hormone treatment (rGHT) was shown to be accompanied by acceleration of metabolic processes that may stimulate oxygen consumption in various organs and tissues. Therefore, oxygen-transporting properties of RBC should undergo considerable changes during the rGHT. The aim of this study was to examine the effects of rGHT on erythrocyte shape and hemoglobin state in GHD children. The level of oxyhemoglobin (Oxy-Hb) in RBC was analyzed by Raman spectroscopy. The RBC count, mean corpuscular hemoglobin (MCH), mean corpuscular volume (MCV) and other parameters were calculated. The blood of eleven treatment-naive prepubertal children with GHD (aged 3-9, median 5.7 years) was examined and compared with control group (aged 5-7; median 6.0 years) at three time points: 0, 3 and 12 months of rGHT. Before rGHT, the MI in GHD children was higher (median 0.48 vs 0.14 p=0.0018) and the RBC count was lower (median 4.20 vs 4.96 1012 cells/L p=0.0022) than in control group. After the treatment, cell count in GHD patients did not differ significantly from the control group, but Oxy-Hb level became higher (median 0.64 vs 0.41 p=0.0075). During rGHT, MCV decreased (median 80.3 vs 83.2µm3 p=0.0231). Morphological and functional characteristics of erythrocytes in GHD children were shown to differ significantly from the healthy control group. A twelve-month rGHT partially improved some of the studied parameters but Oxy-Hb level and echinocyte count remained high.

The aim of this study was to optimize a set of technological parameters (travel speed, extruder temperature, and extrusion rate) for 3D printing with a PEEK-based composite reinforced with 30 wt.% glass fibers (GFs). For this purpose, both Taguchi and finite element methods (FEM) were utilized. The artificial neural networks (ANNs) were implemented for computer simulation of full-scale experiments. Computed tomography of the additively manufactured (AM) samples showed that the optimal 3D printing parameters were the extruder temperature of 460 °C, the travel speed of 20 mm/min, and the extrusion rate of 4 rpm (the microextruder screw rotation speed). These values correlated well with those obtained by computer simulation using the ANNs. In such cases, the homogeneous micro- and macro-structures were formed with minimal sample distortions and porosity levels within 10 vol.% of both structures. The most likely reason for porosity was the expansion of the molten polymer when it had been squeezed out from the microextruder nozzle. It was concluded that the mechanical properties of such samples can be improved both by changing the 3D printing strategy to ensure the preferential orientation of GFs along the building direction and by reducing porosity via post-printing treatment or ultrasonic compaction.

Low-temperature plasma treatment with atmospheric discharge with runaway electrons (DRE) was shown to be an efficient way to activate carbon fiber's (CF) surface and subsequently increase its interlayer shear strength (ILSS) values. It was demonstrated that an acceptable ILSS level was achieved after a DRE plasma treatment duration of 15 min. The treatment of CFs resulted in their surface roughness being increased and their functional groups grafting. The XPS data showed a change in the chemical composition and the formation of reactive oxygen-containing groups. SEM examinations of the PPS/CF laminates clearly demonstrated a difference in adhesive interaction at the PPS/CF interface. After the DRE plasma treatment, CFs were better wetted with the polymer, and the samples cohesively fractured predominantly through the matrix, but not along the PPS/CF interface, as was observed for the sample reinforced with the untreated CFs. The computer simulation results showed that raising the adhesive strength enhanced the ILSS values, but reduced resistance to transverse cracking under the loading pin. In general, higher flexural strength of the PPS/CF laminates was achieved with a greater interlayer adhesion level, which was consistent with the obtained experimental data.

(1) Background: The paper addresses the effect of carbon fibers (CFs) treatment by low-temperature plasma with runaway electrons on the deformation behavior of the polyetheretherketone (PEEK)-layered composites. (2) Methods: The effect of the interlayer adhesion on the mechanical response of the composites was assessed through the tensile and three-point bending tests. In addition, computer simulations of the three-point bending were carried out with the use of the finite element analysis (FEM) with varying conditions at the "PEEK-CF layers" interface. (3) Results: DRE-plasma treatment during the optimal time of t = 15 min led to formation of a rougher surface and partial desizing of a finishing agent. The shear strength of the layered composites increased by 54%, while the tensile strength and the flexural modulus (at three-point bending) increased by 16% (up to 893 MPa) and by 10% (up to 93 GPa), respectively. (4) Conclusions: The results of the numerical experiments showed that the increase in the stiffness, on the one hand, gave rise to enlarging the flexural modulus; on the other hand, a nonlinear decrease in the strength may occur. For this reason, the intention to maximize the level of the interlayer stiffness can result in lowering the fracture toughness, for example, at manufacturing high-strength composites.

Laminated composites based on polyetheretherketone (PEEK) and polyimide (PI) matrices were fabricated by hot compression. Reinforcing materials (unidirectional carbon-fiber (CF) tapes or carbon fabric) and their layout patterns were varied. Stress-strain diagrams after three-point flexural tests were analyzed, and both lateral faces of the fractured specimens and fractured surfaces (obtained by optical and scanning electron microscopy, respectively) were studied. It was shown that the laminated composites possessed the maximum mechanical properties (flexural elastic modulus and strength) in the case of the unidirectional CF (0°/0°) layout. These composites were also not subjected to catastrophic failure during the tests. The PEEK-based composites showed twice the flexural strength of the PI-based ones (0.4 and 0.2 GPa, respectively), while the flexural modulus was four times higher (60 and 15 GPa, correspondently). The reason was associated with different melt flowability of the used polymer matrices and varied inter- (intra)layer adhesion levels. The effect of adhesion was additionally studied by computer simulation using a developed two-dimensional FE-model. It considered initial defects between the binder and CF, as well as subsequent delamination and failure under loads. Based on the developed FE-model, the influence of defects and delamination on the strength properties of the composites was shown at different stress states, and the corresponding quantitative estimates were reported. Moreover, another model was developed to determine the three-point flexural properties of the composites reinforced with CF and carbon fabric, taking into account different fiber layouts. It was shown within this model framework that the flexural strength of the studied composites could be increased by an order of magnitude by enhancing the adhesion level (considered through the contact area between CF and the binder).

The aim of the paper was to improve the functional properties of composites based on ultra-high molecular weight polyethylene (UHMWPE) by loading with reinforcing fibers. It was achieved by designing the optimal composition for its subsequent use as a feedstock for 3D-printing of guides for roller and plate chains, conveyors, etc. As a result, it was experimentally determined that loading UHMWPE with 17% high density polyethylene grafted with VinylTriMethoxySilane (HDPE-g-VTMS) was able to bind 5% glass fillers of different aspect ratios, thereby determining the optimal mechanical and tribological properties of the composites. Further increasing the content of the glass fillers caused a deterioration in their tribological properties due to insufficient adhesion of the extrudable matrix due to the excessive filler loading. A multi-level approach was implemented to design the high-strength anti-friction 'UHMWPE+17%HDPE-g-VTMS+12%PP'-based composites using computer-aided algorithms. This resulted in the determination of the main parameters that provided their predefined mechanical and tribological properties and enabled the assessment of the possible load-speed conditions for their operation in friction units. The uniform distribution of the fillers in the matrix, the pattern of the formed supermolecular structure and, as a consequence, the mechanical and tribological properties of the composites were achieved by optimizing the values of the main control parameters (the number of processing passes in the extruder and the aspect ratio of the glass fillers).

The structure, mechanical and tribological properties of the polyimide-based composites reinforced with chopped carbon fibers (CCF) and loaded with solid-lubricant commercially available fillers of various natures were investigated. The metal- and ceramic counterparts were employed for tribological testing. Micron sized powders of PTFE, colloidal graphite and molybdenum disulfide were used for solid lubrication. It was shown that elastic modulus was enhanced by up to 2.5 times, while ultimate tensile strength was increased by up 1.5 times. The scheme and tribological loading conditions exerted the great effect on wear resistance of the composites. In the tribological tests by the 'pin-on-disk' scheme, wear rate decreased down to ~290 times for the metal-polymer tribological contact and to ~285 times for the ceramic-polymer one (compared to those for neat PI). In the tribological tests against the rougher counterpart (Ra~0.2 µm, the 'block-on-ring' scheme) three-component composites with both graphite and MoS2 exhibited high wear resistance. Under the "block-on-ring" scheme, the possibility of the transfer film formation was minimized, since the large-area counterpart slid against the 'non-renewable' surface of the polymer composite (at a 'shortage' of solid lubricant particles). On the other hand, graphite and MoS2 particles served as reinforcing inclusions. Finally, numerical simulation of the tribological test according to the 'block-on-ring' scheme was carried out. Within the framework of the implemented model, the counterpart roughness level exerted the significantly greater effect on wear rate in contrast to the porosity.

Abscess formation is a common complication of severe life-threatening infections caused by obligate anaerobes. Fusobacterium necrophorum is among the frequently detected anaerobic pathogens from clinical specimens associated with liver abscesses, skin and soft tissue infections, or oral abscesses. The antimicrobial therapy for this kind of infection needs to be optimized. Here, we examined the possibility of treating F. necrophorum-induced abscess wound infections with candidate therapeutics based on three endolysins with activity against a broad spectrum of aerobe Gram-negative pathogens. Antibacterial gel containing three Gram-negative bacteria-targeting endolysins, LysAm24, LysAp22, and LysECD7, was formulated for topical use. Abscess formation was induced in rabbits with F. necrophorum and caused systemic infection. The survival and lifespan of the animals, general parameters, and biochemical and hematological blood tests were analyzed to assess the effectiveness of the gel treatment for the wound infection. The administration of the investigated gel twice per day for 5 days resulted in less acute inflammation, with decreased leukocytes and segmented neutrophils in the blood, retardation of infection progression, and an almost two-fold increase in the lifespan of the animals compared to the placebo group. The results indicate that endolysin-based therapy is an effective approach to treat anaerobic bacterial infections. The use of endolysins as independent pharmaceuticals, or their combination with antibiotics, could significantly reduce the development of complications in infectious diseases caused by sensitive bacterial species.

The aim of the study was to develop a design methodology for the UltraHigh Molecular Weight Polyethylene (UHMWPE)-based composites used in friction units. To achieve this, stress-strain analysis was done using computer simulation of the triboloading processes. In addition, the effects of carbon fiber size used as reinforcing fillers on formation of the subsurface layer structures at the tribological contacts as well as composite wear resistance were evaluated. A structural analysis of the friction surfaces and the subsurface layers of UHMWPE as well as the UHMWPE-based composites loaded with the carbon fibers of various (nano-, micro-, millimeter) sizes in a wide range of tribological loading conditions was performed. It was shown that, under the "moderate" tribological loading conditions (60 N, 0.3 m/s), the carbon nanofibers (with a loading degree up to 0.5 wt.%) were the most efficient filler. The latter acted as a solid lubricant. As a result, wear resistance increased by 2.7 times. Under the "heavy" test conditions (140 N, 0.5 m/s), the chopped carbon fibers with a length of 2 mm and the optimal loading degree of 10 wt.% were more efficient. The mechanism is underlined by perceiving the action of compressive and shear loads from the counterpart and protecting the tribological contact surface from intense wear. In doing so, wear resistance had doubled, and other mechanical properties had also improved. It was found that simultaneous loading of UHMWPE with Carbon Nano Fibers (CNF) as a solid lubricant and Long Carbon Fibers (LCF) as reinforcing carbon fibers, provided the prescribed mechanical and tribological properties in the entire investigated range of the "load-sliding speed" conditions of tribological loading.

(1) Objective: Bacterial resistance to conventional antibiotic therapy is an increasingly significant worldwide challenge to human health. The objective is to evaluate whether bacteriophage therapy could complement or be a viable alternative to conventional antibiotic therapy in critical cases of bacterial infection related to cardiothoracic surgery. (2) Methods: Since September 2015, eight patients with multi-drug resistant or especially recalcitrant Staphylococcus aureus, Enterococcus faecium, Pseudomonas aeruginosa, Klebsiella pneumoniae, and Escherichia coli infections were treated with bacteriophage preparations as a therapy of last resort according to Article 37 of the Declaration of Helsinki. Patients had infections associated with immunosuppression after organ transplantation or had infections of vascular grafts, implanted medical devices, and surgical wounds. Individualized phage preparations were administered locally, orally, or via inhalation for different durations depending on the case. All patients remained on conventional antibiotics during bacteriophage treatment. (3) Results: Patients ranged in age from 13 to 66 years old (average 48.5 ± 16.7) with seven males and one female. Eradication of target bacteria was reached in seven of eight patients. No severe adverse side effects were observed. (4) Conclusions: Phage therapy can effectively treat bacterial infections related to cardiothoracic surgery when conventional antibiotic therapy fails.

The aim of this study was to assess the effect of adhesion between the non-polar, ultra-high molecular weight polyethylene (UHMWPE) matrix and the glass fiber fillers of various lengths treated with the commercially available "KH-550" agent, on the mechanical and tribological properties of the UHMWPE-based composites. The motivation was to find the optimal compositions of the polymer composite, for the compression sintering manufacturing of lining plates for the protection of marine venders and construction vehicles, as well as transport equipment. It was shown that the initial powder size at equal molecular weight determined the distribution patterns of the glass fibers in the matrix, and, as a consequence, the mechanical and tribological properties of the composites. Based on the obtained experimental data and the results of the calculation by a developed computer algorithm, control parameters were determined to give practical recommendations (polymer powder size and glass fiber length), for the production of the UHMWPE-composites having specified mechanical and tribological characteristics. The "GUR4022 + 10% LGF" composite, loaded with the chopped 3 mm glass fibers treated with the "KH-550", was recommended for severe operating conditions (high loads, including impact and abrasive wear). For mild operating conditions (including cases when the silane coupling agent could not be used), the "GUR2122 + 10% MGF" and "GUR2122 + 10% LGF" composites, based on the fine UHMWPE powder, were recommended. However, the cost and technological efficiency of the filler (flowability, dispersibility) and polymer powder processing should be taken into account, in addition to the specified mechanical and tribological properties.

The main goal of this paper is to design and justify optimized compositions of thermoplastic-matrix wear-resistant composites based on polyetheretherketone (PEEK) and polyphenylene sulfide (PPS). Their mechanical and tribological properties have been specified in the form of bilateral and unilateral limits. For this purpose, a material design methodology has been developed. It has enabled to determine the optimal degrees of filling of the PEEK- and PPS-based composites with carbon microfibers and polytetrafluoroethylene particles. According to the results of tribological tests, the PEEK-based composites have been less damaged on the metal counterpart than the PPS-based samples having the same degree of filling. Most likely, this was due to more uniform permolecular structure and greater elasticity of the matrix. The described methodology is versatile and can be used to design various composites. Its implementation does not impose any limits on the specified properties of the material matrix or the reinforcing inclusions. The initial data on the operational characteristics can be obtained experimentally or numerically. The methodology enables to design the high-strength wear-resistant composites which are able to efficiently operate both in metal-polymer and ceramic-polymer friction units.

Using a wide range of different physical and chemical methods, it was found that the oxidative stress caused by addition of hydrogen peroxide to the incubation medium has a significant effect on the conformation of haematoporphyrin, influencing the oxygen-binding properties of haemoglobin in red blood cells. Morphofunctional characteristics of red blood cells change; in particular, we have observed the transformation of erythrocytes, their transition into echinocytes. In erythrocytes, in response to increased lipid peroxidation (LPO) antioxidant enzymes become active. The use of natural antioxidants (ß-carotene and resveratrol) works towards reducting the level of oxidative processes. Resveratrol has the greatest antioxidant effect.

Fibrin glue has been used clinically for decades in a wide variety of surgical specialties and is now being investigated as a medium for local, prolonged drug delivery. Effective local delivery of antibacterial substances is important perioperatively in patients with implanted medical devices or postoperatively for deep wounds. However, prolonged local application of antibiotics is often not possible or simply inadequate. Biofilm formation and antibiotic resistance are also major obstacles to antibacterial therapy. In this paper we test the biocompatibility of bacteriophages incorporated within fibrin glue, track the release of bacteriophages from fibrin scaffolds, and measure the antibacterial activity of released bacteriophages. Fibrin glue polymerized in the presence of the PA5 bacteriophage released high titers of bacteriophages during 11 days of incubation in liquid medium. Released PA5 bacteriophages were effective in killing Pseudomonas aeruginosa PA01. Overall, our results show that fibrin glue can be used for sustained delivery of bacteriophages and this strategy holds promise for many antibacterial applications.

This study shows that membrane-associated cytoskeletal protein structures and the erythrocyte morphology undergo profound changes during hypoxia. Hypoxia also intensified oxidative processes in the lipid phase of the bilayer of red blood cell membranes. Sodium nitroprusside impaired the morphology of red blood cells and altered quantitative and qualitative composition of membrane-skeletal proteins. The findings suggest that hypoxia causes changes at all levels of red blood cell organization, which can cause the functional disorders of hemoglobin oxygen-transporting properties and, eventually, the complete degradation of red blood cells. The use of flavonoids has a protective effect against hypoxia.
