The biomembrane natural (NRL-Natural Rubber Latex), manipulated from the latex obtained from the rubber tree Hevea brasiliensis, has shown great potential for application in biomedicine and biomaterials. Reflecting the biocompatibility and low bounce rate of this material, NRL has been used as a physical barrier to infectious agents and for the controlled release of drugs and extracts. The aim of the present study was to evaluate the incorporation and release of peptides using a latex biomembrane carrier. After incorporation, the release of material from the membrane was observed using spectrophotometry. Analyses using HPLC and mass spectroscopy did not confirm the release of the antimicrobial peptide [W6]Hylin a1 after 24 h. In addition, analysis of the release solution showed new compounds, indicating the degradation of the peptide by enzymes contained in the latex. Additionally, the release of a peptide with a shorter sequence (Ac-WAAAA) was evaluated, and degradation was not observed. These results showed that the use of NRL as solid matrices as delivery systems of peptide are sequence dependent and could to be evaluated for each sequence.

The present work aimed to evaluate the endectocide activity of a new injectable long-action formulation, containing ivermectin (IVM) and abamectin (ABA). In each one of the four experiments performed, the following groups were formed: group I: 2.25% IVM (450 microg/kg)+1.25% ABA (250 microg/kg), group II: 3.15% IVM (630 microg/kg) and group III: control. Eighteen bovine naturally infected by gastrointestinal nematoda were selected for anthelmintic evaluation and necropsied on posttreatment day (PTD) 14 to estimate the total parasitic burden. For the Rhipicephalus (Boophilus) microplus field trial, 30 bovine were selected by means of counts of semi-engorged R. (B.) microplus and the therapeutic and residual efficacy evaluated by tick counts on PTDs 1, 3, 7, 14, 21, 28, 35, 42, 49, 56, 63, 70, 77, 84 and 91. In the stall test, 15 calves were artificially infested with 5000 R. (B.) microplus (Mozzo strain) larvae three times a week and daily collections of all the engorged female ticks detached from each calf were performed until the PTD 80. Forty bovine naturally infected with Dermatobia hominis larvae were selected and the number of larvae was counted by visual and tactile inspection on PTDs 3, 7, 14, 28, 35, 49, 63, 77, 91 and 105. In this trial, a formulation containing 1% doramectin (200 microg/kg) was also used. IVM+ABA formulation and 3.15% IVM eliminated four of the eight species of nematode identified. The anthelmintic efficacy of the avermectins association against Haemonchus placei, Cooperia spatulata and C. punctata was 89.64%, 98.84% and 97.69%, while 3.15% IVM achieved 30.98%, 84.79% and 75.56%, respectively. The two formulations evaluated showed reduced acaricide action on the PTD 1 and 3, reaching high efficacy percentages from PTD 14 onward. The IVM+ABA showed efficacy above 95% in the period between PTDs 21 and 49. In the stall test, it observed no difference (P>0.05) between the two formulations regarding the R. (B.) microplus counts during the entire evaluation period. IVM+ABA reduced the number of ticks from the PTD 1 to 77 (P<0.05) and 3.15% IVM reduced (P<0.05) the tick number from PTD 4 up to PTD 80. The three endectocides showed no difference (P>0.05) regarding the number of D. hominis larvae and prevented this parasite reestablishment until PTD 105. These results indicate that the IVM+ABA association showed higher anthelmintic activity and similar efficacy against arthropods to the formulation containing 3.15% IVM.

Natural rubber latex biomedical (NRLb) obtained from the rubber tree Hevea brasiliensis has shown great potential in biomedicine and biomaterial applications. NRLb has been utilized as a physical barrier against infectious agents and in the controlled release of drugs and extracts. In the present work, NRLb was polymerized in a lyophilizer using different volumes of water to control the resultant membrane porosity and characterized regarding the surface morphology, water vapour permeability (WVP), mechanical properties, haemolytic activity and cytotoxicity. The release of bovine serum albumin protein from the latex membranes was evaluated. Drug release rates increased with porosity and membranes were able to control protein release up to 12 h. In addition, WVP increased with the quantity of pores. The cell viability observed for the porous membrane was higher than that noted for conventional membranes. In summary, the porosity control of natural latex membranes can be used to modulate properties and make them suitable for biomedical applications, such as wound dressings, modulated gas-exchange membranes and controlled drug delivery systems.

We evaluated the effect of different treatment protocols against gastrointestinal nematodes in Nelore beef cattle during the growing phase in the municipality of Terenos, MS, in central Brazil from May 2013 to April 2014 and from May 2014 to April 2015. Ninety-six Nelore calves were kept on Brachiaria brizantha grass during each trial period and were distributed into six experimental groups (replicate paddocks for each group) based on live weight and the number of eggs per gram of feces (EPG): T1 (control)-treated in May, July and September with a saline solution; T2-treated in May and November with 700 µg/kg doramectin; T3-treated in May (doramectin), July (4.7 mg/kg levamisole phosphate) and September (doramectin); T4-treated in May (doramectin), July (200 µg/kg moxidectin) and September (doramectin); T5-treated in May (doramectin), August (levamisole phosphate) and November (doramectin) and T6-treated in May (doramectin), August (moxidectin) and November (doramectin). The calves were weighed and feces were collected (for faecal culture and EPG counts) from calves every 28 days, concomitantly with the collection of forage samples. The efficacies of doramectin, moxidectin and levamisole were low, at 69.2, 65.9 and 69.4% in the first and 13.8, 92.6, and 76.5% in the second experimental periods, respectively, but only the untreated animals lost weight during the dry season. Final weight gains did not differ significantly (p>0.05) among the animals in T2 (120.8 kg), T3 (131.4 kg), T4 (131.2 kg) and T5 (134.4 kg). T6 was the only group with a significantly higher final weight gain (140.9 kg) compared to the protocol with two annual dosages (T2). The weight gain was 31.9% higher in T6 than in the untreated animals (T1). None of the protocols affected the number of larvae on the pasture. Body weight was significantly and negatively (r=-0.65) correlated with EPG counts, which were significantly lower in June (T2, T3, T4 and T6), August (T3), September (T5 and T6), October (T5) and November (T5 and T6). Haemonchus, Cooperia, Trichostrongylus and Oesophagostomum were identified. Treatments in May and November, the most common practice in Brazil, did not increase the final weight gain, so an additional and intermediate treatment during the dry season (August) is recommended.

Active infection by T. gondii was evaluated by immunoassay for soluble SAG-1 (p30), the major surface antigen from T. gondii, specific antibodies and immune complexes in human cerebrospinal fluid (CSF) samples. A total of 263 samples of CSF were collected from hospitalized patients presenting neurological disorders and analyzed for antibodies to HIV. Patients were divided into two groups: HIV positive (n = 96) or HIV negative (n =167). The results of the assays showed that 45% of all samples were positive for soluble SAG-1. Toxoplasma Ag/Ab immune complexes were detected in 19% of the CSF samples and 62% were positive for T. gondii- specific IgG. A combination of these assays in the presence of clinical findings consistent with active Toxoplasma infection may predict the presence of toxoplasmic encephalitis. Moreover, detection of soluble SAG-1 in the CSF of these individuals appears consistent with active infection.

Since its production in the 1980s, ivermectin (IVM) has been used indiscriminately and the selection pressure to which bovine gastrointestinal nematodes have been exposed has been intense, resulting in considerable economic losses due to parasitic resistance. One possibility for the control of resistant parasites is the use of P-glycoprotein (P-gp) modulators, because one of the main biochemical changes in ivermectin-resistant parasites is the increased activity of membrane proteins responsible for the efflux of drugs and xenobiotics. This study aimed to evaluate the in vitro effect of eight P-gp modulating drugs to potentiate IVM efficacy against an IVM-resistant field isolate of Haemonchus placei (Nematoda: Trichostrongylidae). The association of IVM with cyclosporin-A, ceftriaxone, dexamethasone, diminazene aceturate, quercetin, trifluoperazine, verapamil, or vinblastine resulted in increased IVM (10(-4)M) efficacy of 5.1%, 49.06%, 76.42%, 3.31%, 28.85%, 13.74%, 45.64% and 43.61%, respectively, and reduced the IVM half maximal effective concentration (EC50) from 4.381 × 10(-6)M to 9.877 × 10(-8), 2.739 × 10(-7), 1.240 × 10(-6), 1.651 × 10(-6), 2.710 × 10(-7), 1.159 × 10(-7), 1.026 × 10(-6) and 7.136 × 10(-7)M, respectively. Only diminazene aceturate did not significantly reduce the number of migrating larvae when associated with IVM (P > 0.05). The effect of P-gp modulating drugs depended on IVM concentration, with greater potentiating effect at lower IVM concentrations. The in vitro application of trifluoperazine, dexamethasone, quercetin, verapamil, cyclosporin A, vinblastine, and ceftriaxone potentiated IVM efficacy against an IVM-resistant field isolate of H. placei, resulting in higher efficacy and lower IVM EC50.

Ivermectin (IVM) resistance of Cooperia spp. in cattle has become an increasing and global problem. The early detection of anthelmintic resistance (AR) is important to propose strategies to slow down the development of resistance and requires sensitive, reliable, economic high-throughput and practical tests. The purpose of the present study was to apply a larval migration inhibition test (LMIT) for evaluating IVM and MOX efficacy against well-characterized field isolates of Cooperia spp. infecting cattle in Brazil. Eight isolates were used for IVM and seven for MOX. The following EC50 values of IVM were observed for the isolates: susceptible, 1.16 ηmol; Nova Alvorada do Sul I, 4.09 ηmol (RF=3.52); Campo Grande BNA, 3.57 ηmol (RF=3.07); Campo Grande TBR, 4.09 ηmol (RF=3,52); Nova Alvorada do Sul II, 2.50 ηmol (RF=2.15); Bandeirantes, 11.35 ηmol (RF=9.78); Campo Grande II, 6.03 ηmol (RF=5.20); and Porto Mortinho, 8.63 ηmol (RF=7.44). For MOX, the following EC50 values were observed: susceptible, 0.75 ηmol; Campo Grande BNA, 0.93 ηmol (RF=1.24); Campo Grande TBR, 0.36 ηmol (RF=0.48); Nova Alvorada do Sul II, 2.57 ηmol (RF=3.42); Bandeirantes, 1.43 ηmol (RF=1.90); Campo Grande II, 1.08 ηmol (RF=1.44); and Porto Mortinho, 0.49 ηmol (RF=0.65). The LMIT used in the present study can be a useful tool for in vitro evaluation of IVM, but not of MOX. However, such methodology cannot be used in large-scale studies yet. The isolates of Cooperia spp. showed various degrees of resistance to IVM, though remaining susceptible to MOX.

During the study of metazoan parasites of freshwater fish in the Pantanal wetland (state of Mato Grosso do Sul, Brazil), third-stage larvae of Brevimulticaecum sp. were collected from the abdominal cavity, liver parenchyma, mesentery, skeletal muscles and stomach wall of the following species: Gymnotus inaequilabiatus, Hemisorubim platyrhynchos, Hoplias aff. malabaricus, Myleus levis, Pseudoplatystoma corruscans, Pygocentrus nattereri, Serrasalmus marginatus and Sorubim lima. The larvae exhibited the main characteristics of the genus Brevimulticaecum, such as short ventricular appendices and the position of excretory pore and excretory nucleus. This is the first report and description of larval specimens of Brevimulticaecum parasitic in these hosts, which are fish species distributed throughout South America.

A membrana de látex natural (NRL - Natural Rubber Latex), manipulada a partir do látex extraído da seringueira Hevea brasiliensis, tem apresentado grande potencial de aplicação no campo da biomedicina e dos biomateriais. Graças a sua biocompatibilidade e baixa taxa de rejeição, ela tem sido utilizada para próteses e enxertos, atuando como estimulante da angiogênese, adesão celular e barreira física a agentes infecciosos. Além dessas aplicações, as membranas são utilizadas como matriz de sistemas de liberação para avaliar o comportamento da liberação de fármacos e extratos de origem vegetal que apresentam propriedades medicinais. O sistema extrato-membrana tem como objeto de estudo uma nova abordagem dessas substâncias no tratamento de feridas visando à cicatrização e regeneração do tecido envolvido. Casearia sylvestris, conhecida popularmente como guaçatonga, pertence à família Salicaceae, conhecida na medicina popular pelos seus efeitos antiulcerogênicos, cicatrizantes, antiofídicos, anti-inflamatórios e antissépticos, propriedades já comprovadas por estudos. Trabalhos recentes demonstraram que a liberação controlada de fármacos e extratos utilizando membranas de látex natural é uma alternativa interessante e promissora para aplicações biomédicas. Assim, o objetivo deste projeto foi estudar a liberação do extrato de Casearia sylvestris incorporado à membrana de látex em soluções com diferentes valores de pH, com o propósito de estudar seu comportamento e sua liberação de forma controlada. A taxa de liberação do extrato do sistema extrato-membrana foi monitorada e analisada utilizando-se o método de espectroscopia ótica (UV). O pH básico age desestabilizando a membrana indicando que o polímero possui a habilidade de proteger o extrato de ser liberado em valores de pH ácidos, direcionando sua aplicação para o sítio de melhor absorção. Natural Rubber Latex (NRL) membrane, made from latex extracted from the rubber tree Hevea brasiliensis, has shown great potential for use in the biomedicine and biomaterials area. Thanks to its biocompatibility and low rejection rate, it has been used for implants and grafts, acting as a stimulant of angiogenesis and cell adhesion and as a barrier against infectious agents. Besides these applications, the membranes are used as model release systems, to assess the release behavior of drugs and plant extracts that exhibit medicinal properties. The extract-membrane system represents a new approach to studying these substances, as aids to wound healing and tissue regeneration. Casearia sylvestris, popularly known as guaçatonga, belongs to the family Salicaceae, known in popular medicine for its anti-ulcerogenic, wound healing, anti-ophidian, antiinflammatory and antiseptic properties, all of which are proven by scientific studies. Recent studies have also shown that the controlled release of drugs and extracts from natural latex membranes is an interesting and promising process for biomedical applications. The objective of this project was thus to study the release of Casearia sylvestris extract incorporated into natural rubber membranes. The main concern was to study and optimize the controlled release of the extract at various pHs. The rate of release was monitored and analyzed by the method of optical spectroscopy (UV). The present study is part of an ongoing investigation into the characteristics of Myxozoan parasites of Brazilian freshwater fish and was carried out using morphology, histopathology and electron microscopy analysis. A new Myxosporea species (Henneguya pseudoplatystoma) is described causing an important reduction in gill function in the farmed pintado (a hybrid fish from a cross between Pseudoplatystoma corruscans and Pseudoplatystoma fasciatum), which is a commercially important South American catfish. From a total of 98 pintado juveniles from fish farms in the states of São Paulo and Mato Grosso do Sul (Brazil), 36 samples (36.7%) exhibited infection of the gill filaments. Infection was intense, with several plasmodia occurring on a same gill filament. The plasmodia were white and measured up to 0.5mm in length; mature spores were ellipsoidal in the frontal view, measuring 33.2+/-1.9 microm in total length, 10.4+/-0.6 microm in body length, 3.4+/-0.4 microm in width and 22.7+/-1.7 microm in the caudal process. The polar capsules were elongated, measuring 3.3+/-0.4 microm in length and 1.0+/-0.1 microm in width and the polar filaments had six to seven turns. Histopathological analysis revealed the parasite in the connective tissue of the gill filaments and lamella. No inflammatory process was observed, but the development of the plasmodia reduced the area of functional epithelium. Ultrastructural analyses revealed a single plasmodial wall, which was in direct contact with the host cells and had numerous projections in direction of the host cells as well as extensive pinocytotic canals. A thick layer (2-6 microm) of fibrous material and numerous mitochondria were found in the ectoplasm. Generative cells and the earliest stage of sporogenesis were seen more internally. Advanced spore developmental stages and mature spores were found in the central portion of the plasmodia.

Sheep and goat farming requires an efficient management program, due to losses caused by parasites in susceptible animals. Many factors may collaborate to improve infection tolerance in the herd, such as: genetics, nutrition, physiological status, and age. The problem caused by resistance to antihelmintic agents has led to the spread of alternative techniques for parasite controls. The latest strategies include selective treatment with the FAMACHA method, phytotherapy, biological control with predatory fungi, and strategies that still await scientific confirmation, such as homeopathy, the dilution of resistance with the introduction of susceptible parasites, and the combination of drugs without antihelmintic effect. The main objective of these methods is to reduce the usage of antiparasitic agents, thus slowing the development of resistance and promoting the better use of effective products and newly released products. The objective of this article is to describe techniques for controlling nematodes in small ruminants, and it is aimed at technicians interested in increasing their knowledge about the mechanisms of resistance to antihelmintic agents as well as alternatives to the use of these products.


A criação de ovinos e caprinos depende de um manejo eficiente, devido, principalmente, ao prejuízo causado por parasitos em animais susceptíveis. Vários fatores colaboram para que os animais tolerem as infecções parasitárias, como: genética, nutrição, estado fisiológico e idade. O grave problema causado pela resistência anti-helmíntica promoveu a difusão de técnicas alternativas de controle parasitário. Dentre essas novas estratégias incluem-se a adoção do tratamento parcial seletivo com o método FAMACHA, fitoterapia, controle biológico com o uso de fungos nematófagos e estratégias que ainda necessitam de mais dados como a homeopatia, a diluição da resistência com a introdução de parasitos sensíveis e a combinação de produtos químicos sem efeito antiparasitário. O objetivo central destes métodos é reduzir o uso de antiparasitários, retardando o desenvolvimento da resistência, e promover melhor utilização de produtos ainda eficazes na propriedade e de novos produtos que venham a ser lançados. Este artigo abordará as técnicas descritas acima para o controle de nematoides de pequenos ruminantes e é direcionado aos técnicos interessados em ampliar seu conhecimento sobre os mecanismos de resistência dos parasitos aos anti-helmínticos e de alternativas ao uso desses produtos.

The results for 2.25% ivermectin: Cmax (37.11 ng/mL +/- 7.42), Tmax (16 days +/- 5.29), T(1/2) (44.62 days +/- 53.89), AUC (928.2 ng x day/mL +/- 153.83) and MRT (36.73 days +/- 33.64), and for 1.25% abamectin: Cmax (28.70 ng/mL +/- 9.54), Tmax (14 days +/- 4.04), T(1/2) (15.40 days +/- 11.43), AUC (618.05 ng x day/mL +/- 80.27) and MRT (20.79 days +/- 8.43) suggest that this combination of 2.25% ivermectin + 1.25% abamectin possesses properties that give this pharmaceutical formula a longer activity time than two of the commercial products tested (1% ivermectin and 1% abamectin), and showed similarity to 3.15% ivermectin.
