RESUMO

Indoor cases of Tetranychus cinnabarinus displaying resistance have been documented, but the resistance level in field populations remains unexplored in China. This study delves into the resistance dynamics of T. cinnabarinus to fenpropathrin in various field populations across China, a pressing concern in contemporary agricultural pest control. The conventional bioassay and amplicon sequencing reveal a notable absence of significant fenpropathrin resistance in field populations, contrasting with known resistance in indoor cases. Current study highlights the limitations of traditional bioassays in detecting early-stage resistance and underscores the nuanced capabilities and constraints of amplicon sequencing in resistance gene frequency analysis. By employing an integrated approach, we combined dose-response bioassays, amplicon sequencing, and statistical modeling to assess resistance levels and investigate underlying genetic factors. The model with empirical data indicates that a 5% mutation frequency represents the threshold before resistance emerges. However, the detection of the kdr mutation in certain populations ranging from 0 to 1.2%, signals an early looming threat of future resistance emergence. Additionally, we further assessed a specific dsRNA targeting VGSC genes at two concentrations (10 ng/µL and 100 ng/µL), both inducing substantial mortality by silencing target genes effectively. The exploration of RNA interference (RNAi) as a novel, more environmentally friendly pest control measure opens new avenues, despite the ongoing challenge of resistance evolution. Overall, this study underscores the necessity for evolving pest management strategies, integrating advanced biotechnological approaches with traditional methods, to effectively counter pesticide resistance and ensure sustainable agricultural productivity.

OBJECTIVE: In this study, C118P, a novel vascular disrupting agent (VDA), was evaluated for its ability in improving the ablative effect of high-intensity focused ultrasound (HIFU) on uterine fibroids by reducing blood perfusion. METHODS: Eighteen female rabbits were infused with isotonic sodium chloride solution (ISCS), C118P or oxytocin for 30 min, and an HIFU ablation of the leg muscles was performed within the last 2 min. Blood pressure, heart rate and laser speckle flow imaging (LSFI) of the auricular blood vessels were recorded during perfusion. Ears with vessels, uterus and muscle ablation sites were collected and sliced for hematoxylin-eosin (HE) staining to compare vascular size, as well as nicotinamide adenine dinucleotide-tetrazolium reductase (NADH-TR) staining to observe necrosis after ablation. RESULTS: Analyses revealed that the perfusion of C118P or oxytocin steadily reduced blood perfusion in the ears to approximately half by the end of the perfusion, constricted the blood vessels of the ears and uterus, and improved HIFU ablation in the muscle tissues. C118P increased blood pressure and decreased heart rate. The degree of contraction of the auricular and uterine blood vessels was positively correlated. CONCLUSION: This study confirmed that C118P could reduce blood perfusion in various tissues and had a better synergistic effect with HIFU ablation of muscle (the same tissue type as fibroids) than did oxytocin. C118P could therefore possibly replace oxytocin in facilitating HIFU ablation of uterine fibroids; however, electrocardiographic monitoring is required.

The fine and hyperfine interactions in PbF have been studied using the laser-induced fluorescence (LIF) spectroscopy method. Cold PbF molecular beam was produced by laser-ablating a Pb rod under jet-cooled conditions, followed by the reaction with SF6. The LIF excitation spectrum of the (0, 0) band in the B2Σ+-X2Π1/2 system of the 208PbF, 207PbF, and 206PbF isotopologues has been recorded with rotational, fine structure, and hyperfine-structure resolution. Transitions in the LIF spectrum were assigned and combined with the previous X2Π3/2-X2Π1/2 emission spectrum in the near-infrared region [Ziebarth et al., J. Mol. Spectrosc. 191, 108-116 (1998)] and the X2Π1/2 state pure rotational spectrum of PbF [Mawhorter et al., Phys. Rev. A 84, 022508 (2011)] in a global fit to derive the rotational, spin-orbit, spin-rotation, and hyperfine interaction parameters of the ground (X2Π1/2) and the excited (B2Σ+) electronic states. Molecular constants determined in the present work are compared with previously reported values. Particularly, the significance of the hyperfine parameters, A⊥ and A‖, of 207Pb is discussed.

Waste sorting is an important aspect of environmental protection, which is now being enforced on a large scale in China. More broadly, it is beneficial to improve the implementation strategies for waste sorting policy and programmes. This study analyses the psychology of consumers that influences the mechanisms of consumer waste sorting and the governmental policies influencing consumers' attitudes and behaviours. About 295 validated participants were collected from four regions of China. Regression analysis was used to examine the relationship among consumers' internal motivation, government's external publicity and consumer behaviour, and the Hayes' PROCESS macro was used to test mediating effect and moderating effect. The results show that consumers' internal motivation and government's external publicity have a positive effect on consumer behaviour. Consumer attitude played a complete mediating role between consumers' internal motivation and consumer behaviour. Between the government's external publicity and consumer behaviour, consumer attitude played a complementary mediating role. Consumers with self-transcendence values, positive motivation of waste sorting and the government's external publicity had a greater positive impact on their attitudes than consumers with self-enhancement values. The greater the material benefits and psychic income consumers received from waste sorting, the more it promoted a positive attitude to consumer behaviour. The article puts forward some management suggestions to improve the government's implementation of its waste sorting policy.

BACKGROUND: The risk of local recurrence after high-intensity focused ultrasound (HIFU) is relatively high, resulting in poor prognosis of malignant tumors. The combination of HIFU with traditional chemotherapy continues to have an unsatisfactory outcome because of off-site drug uptake. RESULTS: Herein, we propose a strategy of inflammation-tendency neutrophil-mediated clinical nanodrug targeted therapy for residual tumors after HIFU ablation. We selected neutrophils as carriers and PEGylated liposome doxorubicin (PLD) as a model chemotherapeutic nanodrug to form an innovative cell therapy drug (PLD@NEs). The produced PLD@NEs had a loading capacity of approximately 5 µg of PLD per 106 cells and maintained the natural characteristics of neutrophils. The targeting performance and therapeutic potential of PLD@NEs were evaluated using Hepa1-6 cells and a corresponding tumor-bearing mouse model. After HIFU ablation, PLD@NEs were recruited to the tumor site by inflammation (most in 4 h) and released PLD with inflammatory stimuli, leading to targeted and localized postoperative chemotherapy. CONCLUSIONS: This effective integrated method fully leverages the advantages of HIFU, chemotherapy and neutrophils to attract more focus on the practice of improving existing clinical therapies.

We examined our hypothesis that high-intensity focused ultrasound (HIFU) treatment of pancreatic ductal adenocarcinoma (PDAC) in nude mice models may lead to an increased occurrence of hematogenous metastasis. The human PDAC cell line BxPC-3 transfected with mCherry was implanted into nude mice to establish orthotopic and subcutaneous xenograft (OX and SX) tumor models. Mice were exposed to HIFU when tumor sizes reached approximately 200-300 mm3 . The OX and SX tumor models were monitored continuously for tumor growth characteristics and hematogenous metastasis using in vivo flow cytometric (IVFC) detection of circulating tumor cells (CTCs) from the pancreas. We chose an appropriate mouse model to further examine whether or not HIFU increases the potential risk of hematogenous metastasis, using IVFC detection. Our results showed that the CTC number was greater in the OX model than in the SX model. The CTC number in the OX model increased gradually over time, whereas the CTC number in the SX model remained low. Therefore, the OX model was better for studying tumor metastasis by IVFC detection. We found significantly decreased CTC numbers and tumor volume after HIFU ablation. Our results showed the applicability of the PDAC OX tumor model for studying the occurrence of tumor metastasis due to the generation of CTCs. HIFU ablation substantially restricted PDAC hematogenous metastasis and provided effective tumor control locally. © 2020 The Authors. Modern text hiding is an intelligent programming technique which embeds a secret message/watermark into a cover text message/file in a hidden way to protect confidential information. Recently, text hiding in the form of watermarking and steganography has found broad applications in, for instance, covert communication, copyright protection, content authentication, etc. In contrast to text hiding, text steganalysis is the process and science of identifying whether a given carrier text file/message has hidden information in it, and, if possible, extracting/detecting the embedded hidden information. This paper presents an overview of state of the art of the text hiding area, and provides a comparative analysis of recent techniques, especially those focused on marking structural characteristics of digital text message/file to hide secret bits. Also, we discuss different types of attacks and their effects to highlight the pros and cons of the recently introduced approaches. Finally, we recommend some directions and guidelines for future works.

BACKGROUND: Tetranychus cinnabarinus is one of the most common polyphagous arthropod herbivores, and is primarily controlled by the application of acaricides. The heavy use of acaricides has led to high levels of resistance to acaricides such as cyflumetofen, which poses a threat to global resistance management programs. Cyflumetofen resistance is caused by an increase in metabolic detoxification; however, the role of uridine diphosphate (UDP)-glycosyltransferase (UGT) genes in cyflumetofen resistance remains to be determined. RESULTS: Synergist 5-nitrouracil (5-Nul) significantly enhanced cyflumetofen toxicity in T. cinnabarinus, which indicated that UGTs are involved in the development of cyflumetofen resistance. Transcriptomic analysis and quantitative (q)PCR assays demonstrated that the UGT genes, especially UGT201H1, were highly expressed in the YN-CyR strain, compared to those of the YN-S strain. The RNA interference (RNAi)-mediated knockdown of UGT201H1 expression diminished the levels of cyflumetofen resistance in YN-CyR mites. The findings additionally revealed that the recombinant UGT201H1 protein plays a role in metabolizing cyflumetofen. Our results also suggested that the aromatic hydrocarbon receptor (AhR) probably regulates the overexpression of the UGT201H1 detoxification gene. CONCLUSION: UGT201H1 is involved in cyflumetofen resistance, and AhR may regulates the overexpression of UGT201H1. These findings provide deeper insights into the molecular mechanisms underlying UGT-mediated metabolic resistance to chemical insecticides. © 2024 Society of Chemical Industry.

Objective: The purpose of this study is to explore whether machine learning can be used to establish an effective model for the diagnosis of Parkinson's disease (PD) by using texture features extracted from cerebellar gray matter and white matter, so as to identify subtle changes that cannot be observed by the naked eye. Method: This study involved a data collection period from June 2010 to March 2023, including 374 subjects from two cohorts. The Parkinson's Progression Markers Initiative (PPMI) served as the training set, with control group and PD patients (HC: 102 and PD: 102) from 24 global sites. Our institution's data was utilized as the test set (HC: 91 and PD: 79). Machine learning was employed to establish multiple models for PD diagnosis based on texture features of the cerebellum's gray and white matter. Results underwent evaluation through 5-fold cross-validation analysis, calculating the area under the receiver operating characteristic curve (AUC) for each model. The performance of each model was compared using the Delong test, and the interpretability of the optimized model was further augmented by employing Shapley additive explanations (SHAP). Results: The AUCs for all pipelines in the validation dataset were compared using FeAture Explorer (FAE) software. Among the models established by Kruskal-Wallis (KW) and logistic regression via Lasso (LRLasso), the AUC was highest using the "one-standard error" rule. 'WM_original_glrlm_GrayLevelNonUniformity' was considered the most stable and predictive feature. Conclusion: The texture features of cerebellar gray matter and white matter combined with machine learning may have potential value in the diagnosis of Parkinson's disease, in which the heterogeneity of white matter may be a more valuable imaging marker.

With the development and the application popularization of artificial intelligence robot technology and 5G technology, a robotic arm is designed and developed for rinsing porcelain bushing in high voltage substation in this paper. Firstly, the components and implementation of robotic arm are presented, subsequently, a circular cleaning structure with a 120-degree split is proposed to rinse the porcelain bushing. Secondly, a two-stage simple and effective method to realize automatic orientation is proposed utilizing photoelectric switches. Moreover, a prototype of robotic arm with control system is developed based on the regime switching function, and the result of edge computing is transmitted by 5G technology. Finally, feasibility and effectiveness of the robotic arm are verified in the Nanjing power grid. The case study manifests that the robotic arm developed by the proposed method in the paper can achieve efficient rinsing and all the corresponding information can be transmitted preciously. The proposed method lays a foundation for wide application of cleaning robot in high voltage substation.

A three-dimensional (3D) anode is essential for high-performance microbial fuel cells (MFCs). In this study, 3D porous carbon monoliths from a wax gourd (WGCM) were obtained by freeze-drying and carbonization. Nano-TiO2 was further coated onto the surface of WGCM to obtain a nano-TiO2/WGCM anode. The WGCM anode enhanced the maximum power density of MFCs by 167.9% compared with the carbon felt anode, while nano-TiO2/WGCM anode additionally increased the value by 45.8% to achieve 1396.2 mW/m2. WGCM enhancement was due to the 3D porous structure, the good conductivity and the surface hydrophilicity, which enhanced electroactive biofilm formation and anodic electron transfer. In addition, nano-TiO2 modification enhanced the enrichment of Acinetobacter, an electricigen, by 31.0% on the anode to further improve the power production. The results demonstrated that the nano-TiO2/WGCM was an effective anode for power enhancement in MFCs.

A three-dimensional biofilm-electrode reactor (3D-BER) that combined heterotrophic and autotrophic denitrification (HAD) was developed to remove nitrate. The denitrification performance of the 3D-BER was evaluated under different experimental conditions, including current intensities (0-80 mA), COD/N ratios (0.5-5), and hydraulic retention times (2-12 h). The results showed that excessive current limited the nitrate removal efficiency. However, a longer hydraulic retention time was not required to achieve a better denitrification effect in the 3D-BER. Moreover, the nitrate could be effectively reduced over a broad range of COD/Ns (1-2.5), and its removal rate peaked at 89% at I = 40 mA, HRT = 8 h, and COD/N = 2. Although the current reduced the diversity of microorganisms in the system, it promoted the growth of dominant species. Nitrification microorganisms were enriched in the reactor, especially Thauera and Hydrogenophaga, which were crucial to the denitrification process. Thus, the combination of autotrophic denitrification and heterotrophic denitrification was promoted by the 3D-BER system to increase the efficiency of nitrogen removal.

Post-injury infection and wound healing are recurrent daily life problems. Therefore, the necessity of developing a biomaterial with antibacterial and wound-healing properties is paramount. Based on the special porous structure of hydrogel, this work modifies recombinant collagen and quaternary ammonium chitosan and fused them with silver nanoparticles (Ag@mental-organic framework (Ag@MOF)) with antibacterial properties, and asiaticoside-loaded liposomes (Lip@AS) with anti-inflammatory/vascularization effects to form the rColMA/QCSG/LIP@AS/Ag@MOF (RQLAg) hydrogel. The prepared hydrogel possesses good sustainable release capabilities of Ag+ and AS and exhibits concentration-dependent swelling properties, pore size, and compressive strength. Cellular experiments show that the hydrogel exhibits good cell compatibility and promote cell migration, angiogenesis, and M1 macrophage polarization. Additionally, the hydrogels exhibit excellent antibacterial activity against Escherichia coli and Staphylococcus aureus in vitro. In vivo, Sprague Dawley rats burn-wound infection model showed that the RQLAg hydrogel could efficiently promote wound healing and has stronger healing promoting abilities than those of Aquacel Ag. In summary, the RQLAg hydrogel is expected to be an excellent material for accelerating open wound healing and preventing bacterial infections.

Introduction: Kidney injury diagnosis is often delayed in patients with gout. We aimed to determine the characteristics of gout patients with CKD using musculoskeletal ultrasound (MSUS) and whether MSUS could be used as an auxiliary assessment to evaluate kidney injury and predict renal outcome in patients with gout. Methods: Clinical information, laboratory indicators, and MSUS findings were collected and compared between gout-only patients (gout - CKD) and gout patients with CKD (gout + CKD). Multivariate logistic regression was applied to identify risk factors for clinical and MSUS characteristics in both groups. Correlation analysis between MSUS signs and kidney-related indicators was performed, and the effects of MSUS characteristics on renal prognosis were evaluated. Results: In total, 176 patients with gout were included, namely, 89 gout - CKD and 87 gout + CKD cases. After adjusting for confounders, the gout patients with CKD showed more frequent episodes in the previous year, higher ultrasound semiquantitative scores, and more tophi than gout patients without CKD. Additionally, the number of tophi, bone erosion, and synovial hypertrophy measured by MSUS was found to be negatively correlated with the eGFR. The existence of tophi was independently associated with an increased risk of a ≥10% decline in eGFR in the first-year follow-up (OR, 3.56; 95% CI, 1.382-9.176). Conclusions: Ultrasound-detected tophi, bone erosion, and synovial hypertrophy were associated with kidney injury in gout patients. The existence of tophi was associated with faster renal function deterioration. MSUS could be a potential auxiliary diagnostic method to evaluate kidney injury and predict renal outcome in gout patients.

In this study, a heterotrophic/biofilm-electrode autotrophic denitrification reactor (HAD-BER) was constructed and nano-ɑ-Fe2 O3 was coated on granular activated carbon (GAC) as a third electrode to enhance the nitrate removal performance. The introduction of nano-ɑ-Fe2 O3 could stimulate microorganisms to secrete more extracellular polymeric substances (EPS), accelerating the electron transfer. Moreover, more denitrification bacteria were enriched on the particle electrodes, especially Pseudomonas and Thermomonas, which played a significant role in denitrification. The denitrification performance at different COD/N ratios (0.65-3.23) and current intensities (0-150 mA) was investigated in depth. When the nitrate concentration of the influent was 60 mg/L, nitrate was almost completely removed at the optimal current intensity (60 mA) and COD/N ratio (1.29). At the same time, there was almost no nitrite (<0.10 mg/L) and ammonia nitrogen (0 mg/L) accumulation in the effluent. This study provided a new direction for the advancement of HAD-BER and accelerated its implementation. PRACTITIONER POINTS: By introducing nano-a-Fe2O3 into HAD-BER, more denitrification bacteria were enriched on the particle electrodes. The increased contents of polysaccharide and protein content could accelerate the electron transfer. Almost completely denitrification could be achieved at current = 60 mA and COD/N = 1.29. The study provided a new direction for the further development of HAD-BERs.

Four fractions of water-extracted Sepia esculenta ink polysaccharides (SIP) were separated by dicthylaminoethy (DEAE) cellulose chromatography. The eluted fraction with the highest yield was characterized as a sulfate-rich glycosaminoglycan named SIP-IV. According to the analysis of laser scattering and refractive index signals, SIP-IV was determined to be 14.4 kDa and spherical molecular conformation in salt solution. SIP-IV is composed of fucose, galactosamine, glucosamine, mannose and glucuronic acid with a molar ratio of 5.1:7.3:3.8:1:4.4, which is obviously different from reported SIPs. SIP-IV promoted yeast proliferation and intercellular antioxidant level. Based on multi-omics strategy, data of transcriptome analysis suggested that growth promotion of SIP-IV on Saccharomyces cerevisiae might be attributed to regulation of Rho protein signal transduction, nuclear autophagy and nitrogen utilization. Combined with the metabolome results, SIP-IV also re-profiled metabolism of amino acids and phospholipids in yeast cells.

Diagnostic tools for the early detection of renal injury caused by hyperuricemia are still lacking. Here, we investigated whether contrast-enhanced ultrasound (CEUS) could be used as a diagnostic tool for hyperuricemic nephropathy (HN). In the HN rat model, CEUS detected a significant decline in renal cortical perfusion compared with that in control rats. Peak intensity (PI) values correlated significantly with serum KIM-1 levels and fibrosis scores in HN rats. An early decline in PI values was also observed in chronic kidney disease (CKD) stage 1 patients with HN compared with the controls (61.1±4.52 dB versus 65.80±7.10 dB) and correlated with renal function in the patients with HN. In contrast, an increase in time to reach PI values was detected in HN patients with stage 1 CKD (15.14±1.75 s versus 14.52±4.75 s) and was more pronounced in CKD stage 4 patients (67.32±3.29 s). CEUS was able to detect abnormal renal perfusion in early CKD with HN, which correlated with renal function decline, suggesting that CEUS could be used as a noninvasive tool for assessing renal function in patients with HN.

Resorption and loss of alveolar bone leads to oral dysfunction and loss of natural or implant teeth. Biomimetic delivery of growth factors based on stem cell recruitment and osteogenic differentiation, as the key steps in natural alveolar bone regenerative process, has been an area of intense research in recent years. A mesoporous self-healing hydrogel (DFH) with basic fibroblast growth factor (bFGF) entrapment and transforming growth factor ß3 (TGFß3) - loaded chitosan microspheres (CMs) was developed. The formulation was optimized by multiple tests of self-healing, in-bottle inversion, SEM, rheological, swelling rate and in vitro degradation. In vitro tubule formation assays, cell migration assays, and osteogenic differentiation assays confirmed the ability of DFH to promote blood vessels, recruit stem cells, and promote osteogenic differentiation. The optimum DFH formula is 0.05 ml 4Arm-PEG-DF (20%) added to 1 ml CsGlu (2%) containing bFGF (80 ng) and TGFß3-microspheres (5 mg). The results of in vitro release studied by Elisa kit, indicated an 95% release of bFGF in 7 d and long-term sustained release of TGFß3. For alveolar defects rat models, the expression levels of CD29 and CD45, the bone volume fraction, trabecular number, and trabecular thickness of new bone monitored by Micro-CT in DFH treatment groups were significantly higher than others (*P < 0.05, vs Model). HE and Masson staining show the same results. In conclusion, DFH is a design of bionic alveolar remodelling microenvironment, that is in early time microvessels formed by bFGF provide nutritious to recruited endogenous stem cells, then TGFß3 slowly released speed up the process of new bones formation to common facilitate rat alveolar defect repair. The DFH with higher regenerative efficiency dovetails nicely with great demand due to the requirement of complicated biological processes.

Tissue engineering, especially cell sheets-based engineering, offers a promising approach to tendon regeneration; however, obtaining a sufficient source of cells for tissue engineering applications is challenging. Adipose-derived stem cells (ASCs) are essential sources for tissue regeneration and have been shown to have the potential for tenogenic differentiation in vitro via induction by growth differentiation factor 5 (GDF-5). In this study, we explored the feasibility of ASCs cell sheets stimulated by GDF-5 for engineered tendon repair. As shown by quantitative polymerase chain reaction and western blotting, tenogenesis-related markers (Col I&III, TNMD, biglycan, and tenascin C) were significantly increased in GDF-5-induced ASCs cell sheets compared with the uninduced. Moreover, the levels of SMAD2/3 proteins and phospho-SMAD1/5/9 were significantly enhanced, demonstrating that GDF-5 may exert its functions through phosphorylation of SMAD1/5/9. Furthermore, the cell sheets were combined with P(LLA-CL)/Silk fibroin nanoyarn scaffolds to form constructs for tendon tissue engineering. Terminal deoxynucleotidyl transferase dUTP nick end labeling and immunofluorescence assays demonstrated favorable cell viability and tenogenesis-related marker expression in GDF-5-induced constructs. In addition, the constructs showed the potential for tendon repair in rabbit models, as demonstrated by histological, immunohistochemical, and biomechanical analyses. In our study, we successfully produced a new tissue-engineered tendon by the combination of GDF-5-induced ASCs cell sheets and nanoyarn scaffold which is valuable for tendon regeneration.

3D electrospun nanofibrous scaffolds have been developed for cartilage regeneration, however, there is no consensus on the preferable method for biocompatible scaffolds that enhance regeneration and attenuate inflammation. We designed a 3D porous electrospun polylactic acid (PLA) @gelatin-based scaffold by a novel method. Chondroitin sulfate (CS), commonly used in clinical cartilage treatment, is capable of regulating cartilage formation and inhibiting inflammation. Thus we further functionalized the 3D scaffold by crosslinking of CS, assuming that CS-functionalized scaffold (CSS) would promote cartilage regeneration and modulate inflammation. We confirmed that CSS exhibits not only appropriate reversible compressibility and mechanical property, but also appropriate biocompatibility, allowing cell proliferation. In vitro, the potential of CSS for chondrogenic differentiation was improved compared to control and PLA@gelatin scaffold as chondrogenic markers Collagen2 and Aggrecan was significantly increased. Meanwhile, significant reduction in two crucial inflammatory factors (NO and PGE2) in CSS group demonstrated inflammation inhibition. In vivo, rabbit cartilage defects were created and CSS effectively promoted cartilage repair. Additionally, superior anti-inflammation effect of CSS was demonstrated by reduction in iNOS and PGES, enzymes producing NO and PGE2, respectively by immunohistology. Our results indicated the preferable property of CSS for cartilage regeneration and its potential in immunoregulation.
