RESUMO

The differentiation of αßT cells from thymic precursors is a complex process essential for adaptive immunity. Here we exploited the breadth of expression data sets from the Immunological Genome Project to analyze how the differentiation of thymic precursors gives rise to mature T cell transcriptomes. We found that early T cell commitment was driven by unexpectedly gradual changes. In contrast, transit through the CD4(+)CD8(+) stage involved a global shutdown of housekeeping genes that is rare among cells of the immune system and correlated tightly with expression of the transcription factor c-Myc. Selection driven by major histocompatibility complex (MHC) molecules promoted a large-scale transcriptional reactivation. We identified distinct signatures that marked cells destined for positive selection versus apoptotic deletion. Differences in the expression of unexpectedly few genes accompanied commitment to the CD4(+) or CD8(+) lineage, a similarity that carried through to peripheral T cells and their activation, demonstrated by mass cytometry phosphoproteomics. The transcripts newly identified as encoding candidate mediators of key transitions help define the 'known unknowns' of thymocyte differentiation.

BACKGROUND: Infection of suckling mice with influenza virus expands a CD4-CD8- double-negative (DN) natural killer T (NKT) cell subpopulation that protects the mice as adults against allergen-induced airway hyperreactivity (AHR). However, this NKT cell subset has not been characterized, and the underlying mechanisms of protection remain unknown. OBJECTIVE: We characterized this specific NKT cell subpopulation that developed during influenza infection in neonatal mice and that suppressed the subsequent development of AHR. METHODS: A cell-surface marker was identified by comparing the mRNA expression profile of wild-type CD4+ NKT cells with that of suppressive Vα14 DN NKT cells. The marker-enriched NKT cell subset was then analyzed for its cytokine profile and its suppressive in vitro and in vivo abilities. RESULTS: We showed that DN NKT cells with high CD38 expression produced IFN-γ, but not IL-17, IL-4, or IL-13, and inhibited development of AHR through contact-dependent suppression of helper CD4 T-cell proliferation. The NKT subset expanded in the lungs of neonatal mice after infection with influenza and also after treatment of neonatal mice with Nu-α-GalCer, which effectively increased DN CD38hi NKT cell numbers. CONCLUSION: These results suggest that early/neonatal exposure to infection or antigen challenge affects subsequent lung immunity by altering the cellular composition of cells in the lung and that some subsets of NKT cells suppress AHR. These results provide a possible mechanism by which prior infections can protect against the development of allergic asthma and might be further explored as a protective measure for young children.

Twelve small watersheds in central Iowa were used to evaluate the effectiveness of prairie filter strips (PFS) in trapping sediment from agricultural runoff. Four treatments with PFS of different size and location (100% rowcrop, 10% PFS of total watershed area at footslope, 10% PFS at footslope and in contour strips, 20% PFS at footslope and in contour strips) arranged in a balanced incomplete block design were seeded in July 2007. All watersheds were in bromegrass ( L.) for at least 10 yr before treatment establishment. Cropped areas were managed under a no-till, 2-yr corn ( L.)-soybean [ (L.) Merr.] rotation beginning in 2007. About 38 to 85% of the total sediment export from cropland occurred during the early growth stage of rowcrop due to wet field conditions and poor ground cover. The greatest sediment load was observed in 2008 due to the initial soil disturbance and gradually decreased thereafter. The mean annual sediment yield through 2010 was 0.36 and 8.30 Mg ha for the watersheds with and without PFS, respectively, a 96% sediment trapping efficiency for the 4-yr study period. The amount and distribution of PFS had no significant impact on runoff and sediment yield, probably due to the relatively large width (37-78 m) of footslope PFS. The findings suggest that incorporation of PFS at the footslope position of annual rowcrop systems provides an effective approach to reducing sediment loss in runoff from agricultural watersheds under a no-till system.

We evaluated the relationships between landscape characteristics and lake water quality in receiving waters by regressing four water quality responses on landscape variables that were measured for whole watersheds and three different buffer distances (30, 60, and 120 m). Classical percolation theory was used to conceptualize nutrient pathways and to explain nonlinear responses. The response variables were total nitrogen (TN), total phosphorus (TP), chlorophyll-a (Chl-a), and Secchi transparency (SD). Landscape data were obtained from satellite image-derived maps of 130 watersheds in Iowa using geographic information systems software. We developed regression models with a stepwise protocol selecting the optimal number of significant explanatory variables. Configuration variables such as contagion, the cohesion of cropland and urban land, and the aggregation index of forest were very important and more important than variables assessing landscape composition (e.g., percentage farmland). Whole watershed models predicted between 15 and 67% of the variability in TN, TP, Chl-a, and SD. Proximity-explicit data offered only slightly improved statistical power over land cover data derived from the entire watershed for variables TN, Chl-a. and SD, but not for TP.

Information on the water quality impact of perennial warm-season grasses (WSGs) when grown in marginal lands as dedicated energy crops is limited. We studied how WSGs affected runoff, sediment, and nutrient losses and related near-surface soil properties to those of no-till corn ( L.) on an eroded soil in southwestern Iowa and a center pivot corner in east-central Nebraska. The experiment at the eroded soil was established in 2012, and treatments included 'Liberty' switchgrass ( L.) and no-till continuous corn. The experiment at the pivot corner was established in 2013 with 'Liberty' switchgrass, 'Shawnee' switchgrass, low-diversity grass mixture, and corn. We simulated rainfall at 63.5 ± 2.8 mm h for 1 h to portray 5-yr return periods and measured water erosion in spring 2017. Time to runoff start and runoff depth did not differ between WSGs and corn. On the eroded soil, sediment and nutrient losses did not differ between treatments. At the pivot corner, sediment (0.71 vs. 0.15 Mg ha) and PO-P (0.037 vs. 0.006 kg ha) losses were five times higher in corn than in WSGs. Near-surface soil properties did not differ on the eroded soil, but at the pivot corner, wet aggregate stability was four times higher and residue cover was 34% higher in WSGs than in corn. Water-stable aggregates were negatively correlated with NO-N and PO-P losses. Overall, WSGs can improve water quality in marginally productive croplands, but their effectiveness appears to be site specific.
