Aquaporins (AQPs) are essential to coordinate the transit of water and ions through the cell membrane. In salivary glands (SGs), AQPs have been associated with saliva formation, facilitating water absorption through the epithelium during the formation of hypotonic saliva, which is then secreted into the oral cavity. Different members of the AQP family have been suggested to play distinct roles during embryonic development, highlighted by their specific expression patterns. Here, we have investigated the expression patterns of AQP-1, AQP-3 and AQP-5 by immunofluorescence at key stages of salivary gland development, utilising cultured mouse embryonic submandibular (SMG) and sublingual (SLG) glands. The expression of AQPs was compared to a mitotic marker, phospho-histone 3 (PH3), a myoepithelial marker, smooth muscle actin (SMA), and a vascular marker, CD31. Qualitative analysis revealed that AQP-1 and AQP-3 were primarily expressed during the earlier phases of SG morphogenesis and were associated with cells undergoing mitotic processes (PH3-positive). AQP-5, in contrast, was not associated to mitotic figures, but was predominantly expressed during late stages of SG morphogenesis. Our results highlight that AQPs are expressed from early stages of SG morphogenesis and exhibit complimentary expression patterns that may contribute to the morphogenesis of salivary glands.

Salivary gland aquaporins (AQPs) are essential for the control of saliva production and maintenance of glandular structure. However, little is known of their role in salivary gland neoplasia. Salivary gland tumors comprise a heterogeneous group of lesions, featuring variable histological characteristics and diverse clinical behaviors. Mucoepidermoid carcinoma (MEC) is the most common salivary gland malignancy. The aim of this study was to evaluate the expression of AQP1, AQP3, and AQP5 in 24 MEC samples by immunohistochemistry. AQP1 expression was observed in vascular endothelium throughout the tumor stroma. AQP3 was expressed in epidermoid and mucosal cells and AQP5 was expressed in mucosal cells of MEC. These proteins were expressed in the human MEC cell line UH-HMC-3A. Cellular ultrastructural aspects were analyzed by electron microscopy to certificate the tumor cell phenotype. In summary, our results show that, despite the fact that these molecules are important for salivary gland physiology, they may not play a distinct role in tumorigenesis in MEC. Additionally, the in vitro model may offer new possibilities to further investigate mechanisms of these molecules in tumor biology and their real significance in prognosis and possible target therapies.

Brewery spent grain represents approximately 85 % of total by-products generated in a brewery. Consisting of carbohydrates, fiber, minerals and low amounts of protein, the use of brewery spent grain is limited to the feeding of ruminants; however, its potential use should be investigated. The reuse of this by-product using microorganisms by solid-state fermentation process as the case of protein enrichment by single-cell protein incorporation is an alternative to ensure sustainability and generate commercially interesting products. In this context, the aim of this study was to grow Rhizopus oligosporus in brewery spent grain under different initial moisture contents and nitrogen sources to increase the protein content of the fermented material. After 7 days of fermentation, increase of 2-4 times in the crude protein and soluble protein content was verified, respectively, compared to unfermented brewery spent grain. The kinetics of protein enrichment demonstrated the possibility of application of this technique, which can be a great alternative for use in diets for animals.

INTRODUCTION: Hematopoietic Stem Cell Transplant (HSCT) has been successfully used as standard therapy for hematological disorders. After conditioning therapy, patients undergoing allogeneic HSCT, present three different phases of engraftment: early pre-engraftment, early post-engraftment, and late engraftment. Severe complications are associated with morbidity, mortality, and malignancies in these phases, which include effects on the oral cavity. OBJECTIVES: The changes in the salivary composition after HSCT may contribute to identifying relevant proteins that could map differences among the phases of diseases, driven for personalized diagnostics and therapy. METHODS: Unstimulated whole saliva was collected from patients submitted to HSCT. The samples were submitted to trypsin digestion for a Mass spectrometry analysis. MaxQuant processed the Data analysis, and the relevant expressed proteins were subjected to pathway and network analyses. RESULTS: Differences were observed in the most identified proteins, specifically in proteins involved with the regulation of body fluid levels and the mucosal immune response. The heatmap showed a list of proteins exclusively expressed during the different phases of HSCT: HBB, KNG1, HSPA, FGB, APOA1, PFN1, PRTN3, TMSB4X, YWHAZ, CAP1, ACTN1, CLU and ALDOA. Bioinformatics analysis implicated pathways involved in protein processing in the endoplasmic reticulum, complement and coagulation cascades, apoptosis signaling, and cholesterol metabolism. CONCLUSION: The compositional changes in saliva reflected the three phases of HSCT and demonstrated the usefulness of proteomics and computational approaches as a revolutionary field in diagnostic methods.

Differentiation between organic and conventional coffee has increased due to the growing demand and high consumption of healthy foods that contain compounds with antioxidant potential, which have been associated with the reduction of chronic diseases. We used organic and conventional coffee in powder 4% (w/w) and infusions 5%, 10% and 20% (w/v) incorporated in a commercial diet to test in vivo. The levels of chlorogenic acid, caffeine and trigonelline were determined by high performance liquid chromatography. The body weight, weight gain, food consumption, aberrant foci crypt, mucin depleted foci, stress biomarkers protein carbonyl and malondialdehyde, biochemical parameters and behavior of the rats were compared between the experimental and control groups within a framework of colon carcinogenesis. The organic coffee showed higher levels of chlorogenic acid, caffeine and trigonelline than conventional, however, this difference did not significantly affect behavior. The infusions had an antioxidant effect, reducing the levels of malondialdehyde; however, the biochemical parameters of the serum were not altered, and there was neither induction nor prevention of preneoplasic lesions.

INTRODUCTION: The aims of this study were to correlate the clinical and radiographic stability of titanium miniscrews when used as orthodontic anchorage for maxillary canine retraction and to assess bone quality. METHODS: Thirty titanium miniscrews were placed in 15 consecutive patients (8 male, 7 female; age range, 12 years 5 months-32 years 11 months) as orthodontic anchorage. Orthodontic loads were applied immediately after miniscrew placement (T1) with a nickel-titanium closing coil spring. The initial estimated load was 200 g. The bone quality in each region of interest was determined by multi-slice computed tomography. RESULTS: Bone mineral density (BMD) values ranged from 167 HU to 660.80 HU (mean, 420.63 HU). The specific regions had a mean close to the maximum value of a previously established scale for the posterior region of the maxilla (0-500 HU). The paired t test showed a statistically significant difference (P = 0.450) when the means of the differences between the right and left sides were compared. Twelve of the 15 subjects had significantly greater maxillary BMD on the right side. Linear regression also showed a low correlation between the 2 sides (P = 0.097). Clinically, the success index was 100%. Although 2 miniscrews were removed from 1 patient because of severe gingival inflammation with purulent secretion, none of the 28 remaining miniscrews showed any mobility after 90 days (T2). Comparisons of the means at T1 and T2 showed no statistically significant differences in these distances: between nasion and the miniscrew head, between orbitale and the miniscrew head, and between nasion and orbitale, assessed through cephalometric tracings made on lateral oblique radiographs (45 degrees ), pointing to the stability of the 28 miniscrews during the 90-day observation period. BMD values of all subjects were within the normal range of an established scale, and even close to maximum values. CONCLUSIONS: The regions between the maxillary second premolars and first molars, and mesial to the maxillary second premolars, are safe as far as bone quality is concerned for miniscrew placement during the first 90 days of canine distalization. A good surgical technique and appropriate planning for miniscrew placement, inflammation control, and adequate oral hygiene are fundamental to the success of this new anchorage system during maxillary canine distalization.

Some members of aquaporin family (AQP) plays crucial functions in salivary synthesis and secretion. These proteins expression has already been reported during salivary gland formation, however no previous studies in human developing glands have been performed. We evaluated AQP1, 3 and 5 expression through the stages of human salivary gland morphogenesis and discuss the possible role of AQP for glandular maturation. Human salivary glands derived from foetuses aged between 14 and 25 weeks were submitted to immunohistochemistry. At the bud stage, membrane expression of AQP1, 3 and 5 were observed within the epithelial bud cells presenting a similar apicolateral pattern, also found at the pseudoglandular stage, present within the terminal portions of future acini, while AQP5 was also particularly strong at the apical membrane of pre-acinar and pre-ductal cells. AQP5 was co-localised with Cytokeratin 7. Similar AQP1, 3 and 5 expression were observed at the following canalicular stage, where distinct and strongly luminal and acinar AQP5 expression is present. During the final terminal bud stage, AQP1 was only identified in serous acini, myoepithelial and endothelial cells, while differentiated mucous acinar cells and ducts were negative. AQP3 was detected at apicolateral membranes of both mucous and serous acini. AQP5 also showed a diffuse expression in mucous and serous acini, in addition to strong apical membrane expression within lumen of intercalated ductal cells. This topographic analysis of AQP1, 3 and 5 revealed differences in the expression pattern throughout salivary gland developmental stages, suggesting different roles for each protein in human glandular maturation.

Salivary glands are essential organs that produce and secrete saliva to the oral cavity. During gland morphogenesis, many developmental processes involve a series of coordinated movements and reciprocal interactions between the epithelium and mesenchyme that generate the ductal system and the secretory units. Recent studies have shown new findings about salivary gland development, particularly regarding lumen formation and expansion, with the involvement of apoptosis and cell polarization, respectively. Moreover, it has been observed that human minor salivary glands start forming earlier than previously published and that distinct apoptotic mediators can trigger duct lumen opening in humans. This review summarizes updated morphological and cellular features of human salivary glands and also explores new aspects of the human developmental process. Graft versus host disease (GVHD) occurs after bone marrow transplantation and is one of the most important causes of death worldwide. Reviews demonstrated GVHD patients with involvement of various tissues and organs, such as salivary glands. The diagnosis of acute GVHD has been the biopsies and the histopathologic evaluation of tissue from an involved organ. These procedures are invasive. Saliva proteins as possible biomarker for GVHD could facilitate the management and diagnosis accuracy. For support the proposed hypotheses, in this pilot study we collected whole saliva samples from patients with undergoing allogeneic hematopoietic cell transplantation (HCT) and from healthy subjects. Samples were collected prospectively between pre-transplant, thirty days, one hundred and, two hundred days after transplant. The proteomic profile was analyzed using SDS-PAGE and LCMS-ESI-IT-TOF mass spectrometry. The relevant personal data, past medical history were also recorded. The most relevant proteins found exclusively in GVHD patients were: CSF2RB, Protocadherin (Pcdh) Fat 2 precursor, protein capicua homolog isoform CIC-S, MUC16 and RGPD8_HUMAN RANBP2. This study aims to conduct an initial evaluation of the possible presence of such biomarkers in saliva from GVHD patients, and suggested a potential application of proteomics analysis as a alternative method to diagnose GVHD.

Patients with Down syndrome may not be cooperative during dental treatment and, in cases of complex treatment, it is necessary to use oral or inhaled sedation and/or general anesthesia, which represent safe and efficient treatment resources. There are few reports about oral rehabilitation with dental implants in patients with Down syndrome in the literature. This paper presents the case of a patient with Down syndrome and moderate mental retardation who suffered avulsion of the maxillary left central incisor after a fall, and was treated by means of a single dental implant with immediate load carried out under general anesthesia. After 4 years of follow up, there has been improvement in the sleep obstructive apnea syndrome due to the use of a tongue crib adapted for providing esthetics and preserving the implant placed.

A carreira médica pode desencadear alterações patológicas na saúde mental, que podem ter início já na graduação. O objetivo deste estudo foi identificar a prevalência de Transtornos Psiquiátricos Menores (TPMe) e a procura por ajuda em estudantes de um curso de Medicina. Trata-se de um estudo transversal, com uma população de 343 estudantes da primeira à quarta série, maiores de 18 anos. Foram realizadas entrevistas, em salas de aula, por meio de dois questionários estruturados, o Self Reporting Questionnaire (SRQ-20) e outro elaborado pelos autores. Análises descritiva, univariada, bivariada e estatística foram utilizadas mediante o programa Microsoft Excel. Os resultados evidenciaram que, entre os acadêmicos com TPMe, 41,6 por cento (a maioria) moravam sozinhos e 75 por cento eram mulheres. Entre os acometidos, 59,2 por cento não conheciam qualquer programa e apenas 9,1 por cento procuraram ajuda. O uso de medicamento foi duas vezes mais prevalente em mulheres com TPMe do que em homens, sendo antidepressivos e ansiolíticos os mais usados. A prevalência de 26,1 por cento de TPMe nos alunos, associada a baixa procura por cuidados e a relatos de automedicação, demonstra a inefetividade dos atuais programas de apoio.


The medical career can trigger pathological changes in mental health that may even begin during undergraduate medical training. The purpose of this study was to identify the prevalence of minor psychiatric disorders (MPD) and the search for counseling by medical students. This was a cross-sectional study of a sample of 343 1st to 4th-year medical students 18 years and older. Interviews were conducted in the classroom using two structured questionnaires, the Self Reporting Questionnaire (SRQ-20) and another prepared by the authors. Descriptive, univariate and bivariate analyses were performed using Microsoft Excel. Among students with MPD, 41.6 percent lived alone and 75.0 percent were women. Among the ill, 59.2 percent were unfamiliar with any counseling program, and only 9.1 percent sought help. Medication for PMD was twice as prevalent in females as compared to males (with antidepressants and anxiolytics as the most frequently used medicines). The 26.1 percent prevalence rate of students with MPD, plus the frequent self-medication and low demand for professional care, demonstrated the ineffectiveness of current psychological support programs.
